Abstract:

The order Trypanosomatida has been well studied due to its pathogenicity and the unique biology of the mitochondrion. In Trypanosoma brucei, four DNA polymerases, namely PolIA, PolIB, PolIC, and PolID, related to bacterial DNA polymerase I (PolI), were shown to be localized in mitochondria experimentally. These mitochondrion-localized DNA polymerases are phylogenetically distinct from other family A DNA polymerases, such as bacterial PolI, DNA polymerase gamma (Polγ) in human and yeasts, “plant and protist organellar DNA polymerase (POP)” in diverse eukaryotes. However, the diversity of mitochondrion-localized DNA polymerases in Euglenozoa other than Trypanosomatida is poorly understood. In this study, we discovered putative mitochondrion-localized DNA polymerases in broad members of three major classes of Euglenozoa—Kinetoplastea, Diplonemea, and Euglenida—to explore the origin and evolution of trypanosomatid PolIA-D. We unveiled distinct inventories of mitochondrion-localized DNA polymerases in the three classes: (1) PolIA is ubiquitous across the three euglenozoan classes, (2) PolIB, C, and D are restricted in kinetoplastids, (3) new types of mitochondrion-localized DNA polymerases were identified in a prokinetoplastid and diplonemids, and (4) evolutionarily distinct types of POP were found in euglenids. We finally propose scenarios to explain the inventories of mitochondrion-localized DNA polymerases in Kinetoplastea, Diplonemea, and Euglenida.

Abstract:

Komunikasi dengan bahasa menjadikan setiap orang dapat menyesuaikan diri dengan lingkungannya. Tujuan spesifik dari penelitian ini adalah menemukan fungsi implikatur percakapan pada Vlog Jerome Polin di YouTube. penelitian ini juga bertujuan untuk menganalisis relevansi implikatur percakapan Vlog Jerome Polin di YouTube terhadap pembelajaran teks anekdot di Sekolah Menengah Atas. Jenis penelitian ini adalah penelitian kualitatif deskriptif dengan dasar penelitian studi kasus. Subjek penelitiannya adalah dialog percakapan pada Vlog Jerome Polin. Langkah-langkah yang dilakukan dalam penelitian ini adalah mencari dan mengumpulkan Vlog Jerome Polin, mentranskrip Vlog Jerome Polin yang sudah dikumpulkan, melakukan pendokumentasian tuturan pada Vlog Jerome Polin yang mengandung praanggapan, menganalisis data, membuat simpulan dari seluruh analisis. Hasil penelitian alih kode dan campur kode yang terjadi dalam video Youtube Jerome Polin akan dikaitkan dalam KD 3.6 menganalisis struktur dan kebahasaan teks anekdot dan 4.6 menciptakan kembali teks anekdot dengan memerhatikan struktur teks anekdot dan struktur kebahasaan teks anekdot. Handout ini dengan menambahkan pengertian, ciri-ciri, struktur dan kebahasaan teks anekdot yang dapat membantu dalam kegiatan pembelajaran. Implikasi penelitian ini untuk menyelesaikan fungsi implikatur percakapan pada sebuah tayangan harus menganalisis tuturan terlebih dahulu dan menyelesaikannya.

Berkembanganya dunia teknologi dengan penggunaan media baru yaitu internet membuat banyak khalayak melek terhadap kemajuan dunia digital dan penggunaan media baru seperti media sosial menjadi salah satu media yang disukai untuk mengekspresikan diri. Salah satu aplikasi media social yang banyak unduh masyarakat Indonesia adalah Instagram. Instagram yang memiliki fitur menarik secara sengaja dimanfaatkan bagi pelaku konten kreator dalam membangun Personal Branding. Dengan kata lain, Personal Branding adalah proses pembentukan persepsi publik terhadap berbagai aspek seseorang, termasuk kepribadian, kemampuan, atau nilai, sehingga menghasilkan persepsi positif dari publik yang pada akhirnya dapat digunakan sebagai alat pemasaran. Dalam penelitian ini penulis tertarik dengan Personal Branding yang dibangun oleh Jerome Polin Pemilik akun Instagram @jeromepolin dalam membangun citranya sebagai mahasiswa dan konten kreator. Metode penelitian ini menggunakan deskriptif kualitatif dalam melihat 8 konsep Personal Branding dalam Instagram Jerome Polin. Hasil penelitian ini diketahui bahwa Personal Branding Jerome poline memiliki ciri khas citra diri sebagai mahasiswa, kepribadian yang menarik dengan pola pikir positif dan terus semangat belajar, dan konsisten terhadap konten yang mengedukasi para pengikutnya. Abstract:

ABSTRACT Kinetoplast DNA (kDNA), the mitochondrial genome of trypanosomes, is a catenated network containing thousands of minicircles and tens of maxicircles. The topological complexity dictates some unusual features including a topoisomerase-mediated release-and-reattachment mechanism for minicircle replication and at least six mitochondrial DNA polymerases (Pols) for kDNA transactions. Previously, we identified four family A DNA Pols from Trypanosoma brucei with similarity to bacterial DNA Pol I and demonstrated that two (POLIB and POLIC) were essential for maintaining the kDNA network, while POLIA was not. Here, we used RNA interference to investigate the function of POLID in procyclic T. brucei. Stem-loop silencing of POLID resulted in growth arrest and the progressive loss of the kDNA network. Additional defects in kDNA replication included a rapid decline in minicircle and maxicircle abundance and a transient accumulation of minicircle replication intermediates before loss of the kDNA network. These results demonstrate that POLID is a third essential DNA Pol required for kDNA replication. While other eukaryotes utilize a single DNA Pol (Pol γ) for replication of mitochondrial DNA, T. brucei requires at least three to maintain the complex kDNA network.

Wystawę tworzy siedem galerii historycznych (od X w. do czasów powojennych) oraz symboliczny „Las”, w których przedstawiono tysiąc lat dziejów polskich Żydów. Koncepcja galerii „Zagłada” została opracowana przez dwoje wybitnych polskich naukowców – prof. Barbarę Engelking i prof. Jacka Leociaka. Muzeum Polin zostało stworzone jako miejsce otwarte na dialog, refleksję i krytyczną analizę przeszłości. Zgodnie ze słowami Konstantego Geberta „jest to Muzeum pytań”. Według klasyfikacji Piotra Piotrowskiego Muzeum Polin należy do muzeów-forów, budujących społeczeństwo otwarte.

Jerome Polin merupakan seseorang terkenal akan kecerdasannya dan seorang content creator, Ia juga mendapatkan beasiswa untuk melanjutkan studinya di Jepang. Berdasarkan penjelasan di atas, penelitian dilakukan untuk menentukan apakah konten YouTube Jerome Polin sudah memenuhi kebutuhan hiburan para remaja di Kota Bandung. Penelitian ini menggunakan teori uses and gratifications atau yang disebut dengan teori kegunaan dan kepuasan. Metode penelitian dilakukan secara kuantitatif dengan Teknik probability sampling dengan jenis simple random sampling. Pada penelitian ini mendapatkan pengaruh yang signifikan antar variabel konten media Youtube Jerome Polin terhadap pemenuhan kebutuhan remaja di Kota Bandung. Hasil uji T yang telah dilakukan, memperoleh t hitung sebesar 11.340 hal ini berarti nilai t hitung lebih besar dari t tabel, Artinya Ho ditolak dan Ha diterima. Dari hasil uji koefisien determinasi mendapatkan kesimpulan bahwa konten media YouTube Jerome Polin berpengaruh sebesar 39,4% dalam pemenuhan kebutuhan remaja di Kota Bandung.

The background of this study is that researchers want to examine how strong Jerome Polin's influence is on Menantea's business development and the strategies undertaken by Jerome Polin as an influencer and brand ambassador due to the rampant development of modern information technology as evidenced by social media influencers who have a major influence in increasing marketing. a brand (brand ambassador). The purpose of this study was to determine Jerome Polin's strategy as an influencer and also a brand ambassador for the Menantea brand. This study uses the brand ambassador theory proposed by Lea Greenwood and the influencer theory according to Kadekova & Holiencinova with descriptive qualitative research methods through case studies where data collection is carried out by interviews, observations, documentation, and literature studies. The results of this study indicate that social media engagement is very important because it can form a good and original personal branding, and the main strategy that Jerome Polin as a brand ambassador does is to build good relationships with consumers and also develop promotional strategies that can highlight brand image. So, it can be concluded that Jerome Polin can become a brand ambassador who meets the criteria by using his strategy as a content creator and influencer as well as the Menantea brand ambassador mentioned above. Latar belakang dari penelitian ini adalah peneliti ingin meneliti seberapa kuat pengaruh Jerome Polin pada perkembangan bisnis Menantea serta strategi yang dilakukan oleh Jerome Polin sebagai influencer dan brand ambassador dikarenakan perkembangan teknologi informasi modern yang sudah merajalela dibuktikan dengan influencer media sosial yang memiliki pengaruh besar dalam peningkatan pemasaran suatu brand (brand ambassador). Tujuan penelitian ini ialah untuk mengetahui strategi Jerome Polin sebagai influencer dan juga brand ambassador pada brand Menantea. Penelitian ini menggunakan teori brand ambassador yang dikemukakan oleh Lea Greenwood serta teori influencer menurut Kadekova & Holiencinova dengan metode penelitian kualitatif deskriptif melalui studi kasus yang pengambilan datanya dilakukan dengan wawancara, observasi, dokumentasi, dan studi kepustakaan. Hasil dari penelitian ini menunjukan bahwa engagement media sosial merupakan hal yang sangat penting karena dapat membentuk personal branding yang bagus dan orisinal, serta strategi yang utama yang dilakukan Jerome Polin sebagai brand ambassador adalah membangun hubungan baik dengan para konsumen dan juga menyusun strategi promosi yang dapat menonjolkan brand image. Jadi, dapat ditarik kesimpulan bahwa Jerome Polin dapat menjadi brand ambassador yang memenuhi kriteria dengan menggunakan strateginya sebagai seorang content creator dan influencer serta sebagai brand ambassador Menantea yang telah disebutkan di atas.

Abstract:

Advertising is a tool used as a communication process to convey information to audiences that can generate attention, interest and decisions on actions to buy the products offered. The use of celebrity endorsers in advertisements is the main attraction of a brand to the intended target market, as well as building a strong emotional relationship with consumers. The purpose of this study was to determine 1) Jerome Polin as a celebrity endorser for Sedaap Tasty Beef Yakiniku noodle products, 2) Purchase decisions and 3) The influence of celebrity endorser Jerome Polin on purchasing decisions for Sedaap Tasty Beef Yakiniku Noodles. This study uses a quantitative approach with a survey method with a questionnaire as the data collection instrument. The population in this study were followers of the Instagram account @miesedaaptasty with a population of 120 and a sample of 55 people. The sampling technique used simple random sampling technique. The results showed that celebrity endorser Jerome Polin had t-count (10.796) > t-table (2.398), it was stated that there was an influence between Celebrity Endorser Jerome Polin on the Purchase Decision of Sedaap Tasty Beef Yakiniku Noodles. Meanwhile, the R square result is 0.687, which means that Celebrity Endorser Jerome Polin influences the Purchase Decision of Mie Sedaap Tasty Beef Yakiniku by 68.7%, and the remaining 31.3% is influenced by other factors outside of this study.

Abstrak:Analisis ini bertujuan untuk mendeskripsikan tindak tutur, yaitu tindak tutur lokusi dalam konten Jerome Polin. Metode yang digunakan dalam analisis ini adalah deskriptif kualitatif. Sumber data yang digunakan adalah video-video yang diunggah oleh Jerome Polin dalam channel YouTubenya. Pengumpulan data dilakukan dengan cara menonton video-video Jerome Polin, kemudian mencatat hal-hal penting untuk dianalisis. Hasil analisis ini menunjukkan bahwa Jerome Polin sering menggunakan tindak tutur lokusi dalam kontennya. Tindak tutur lokusi merupakan tindak tutur yang disampaikan untuk mengatakan sesuatu. Dari hasil analisis terdapat 30 data berupa tindak tutur lokusi. Tindak tutur tersebut masuk ke dalam dua kategori yakni tindak tutur lokusi kategori berita dan tindak tutur lokusi kategori tanya. 14 tindak tutur lokusi kategori berita dan 16 tindak tutur lokusi kategori tanya. Dengan adanya penelitian ini, diharapkan pembaca mampu memahami mengenai tindak tutur lokusi. Penulis berharap penelitian ini dapat membawa manfaat bagi pembaca dan penulis lain sebagai sumber referensi mengenai tindak tutur lokusi.Abstract: This analysis aims to describe speech acts, namely locutionary speech acts in Jerome Polin's content. The method used in this analysis is descriptive qualitative. The data sources used are videos uploaded by Jerome Polin on his YouTube channel. Data collection was done by watching Jerome Polin's videos, then recording important things for analysis. The results of this analysis indicate that Jerome Polin often uses locutionary speech acts in his content. Locutionary speech acts are speech acts that are conveyed to say something. From the results of the analysis there are 30 data in the form of locutionary speech acts. These speech acts fall into two categories, namely locutionary speech acts in the news category and locutionary speech acts in the question category. 14 locutionary speech acts in news category and 16 locutionary speech acts in question category. With this research, it is hoped that the reader will be able to understand the locutionary speech act. The author hopes that this research can bring benefits to readers and other writers as a reference source regarding locutionary speech acts.

Abstract:

Jerome Polin Sijabat is an Indonesian YouTuber. Jerome Polin is known after starting a YouTube channel called Nihongo Mantappu, which shares his personal life in Japan. Apart from speaking Indonesian, Jerome Polin also uses other languages, such as English and Japanese. Jerome Polin's mastery of the language causes code-mixing in the video. This study describes the forms of code-mixing and the factors that cause code-mixing in videos on Jerome Polin's YouTube channel. This study uses a qualitative descriptive method with a sample of conversational quotations. The data collection technique used is the listening method using note-taking techniques and free-involved-talk listening techniques. The results showed that the forms of code-mixing insertion in Jerome Polin's YouTube video include elements of words, phrases, and clauses. The types of code-mixing in Jerome Polin's YouTube videos are outer code-mixing.

Ocenianie wystawy głównej Muzeum Polin w odniesieniu do jej założeń jako multimedialnej wystawy narracyjnej jest czymś całkiem innym niż zarzucanie jej, że nie jest wystawą innego typu, opartą na obiektach oryginalnych. Rzekoma analiza krytyczna naszej wystawy głównej tak naprawdę oznacza odrzucenie jej założeń; brakuje jednak krytycznej oceny założeń owej analizy, a mianowicie że na skutek braku obiektów oryginalnych multimedialna wystawa narracyjna to de facto „simulakrum”. Punkt wyjścia wystawy głównej Muzeum Polin stanowi opowieść, a wybrane przez nas podejście to „teatr historii” – opowieść snuta w trójwymiarowej przestrzeni, której wątki rozwijają się wraz z kolejnymi krokami zwiedzających. Dążymy do stworzenia Gesamtkunstwerk – totalnego dzieła sztuki, całości większej niż suma jej elementów.

Pada artikel ini penulis mengkaji bentuk tindak tutur ilokusi yang terdapat dalam vlog di YouTube Nihongo Mantappu milik Jerome Polin yang berjudul “JIKA AKU MENJADI MENTERI PENDIDIKAN…”. Penelitian ini bertujuan untuk menganalisis tindak tutur ilokusi menurut teori Austin yang dikembangkan menjadi lima bentuk, antara lain representatif atau asertif, komisif, direktif, ekspresif, dan deklaratif yang ada dalam video tersebut. Alasan lain yang mendasari penelitian ini adalah untuk menjelaskan maksud dan tujuan tuturan yang diucapkan Jerome Polin dalam video berbentuk opini yang membahas topik pendidikan. Jenis riset ini menggunakan pendekatan pragmatik dengan metode deskriptif kualitatif yang dilakukan dengan teknik studi dokumen, teknik simak, dan teknik catat. Hasil penelitian menyimpulkan bahwa tindak tutur ilokusi dalam vlog YouTube Nihongo Mantappu milik Jerome polin yang berjudul “JIKA AKU MENJADI MENTERI PENDIDIKAN INDONESIA…” meliputi (1) 30 tuturan representatif atau asertif, (2) 3 tuturan komisif (3) 4 tuturan direktif, (4) 1 tuturan ekspresif, dan (5) 1 tuturan deklaratif.

Penelitian ini dibuat dengan tujuan untuk mendeskripsikan bentuk alih kode dan campur kode yang terjadi dalam video Youtube Jerome Polin bersama chef Arnold, faktor yang menyebabkan terjadinya alih kode dan campur kode, dan relevansinya sebagai bahan ajar teks anekdot di Sekolah Menengah Atas. Peneliti menggunakan jenis penelitian kualitatif, sedangkan metode yang digunakan peneliti adalah metode deskriptif. Subjek dalam penelitian ini bersumber dari video Youtube Jerome Polin bersama chef Arnold. Peneliti mengumpulkan data menggunakan teknik simak dan catat. Peneliti menyimak bentuk alih kode dan campur kode yang terdapat dalam video Youtube Jerome Polin bersama chef Arnold, kemudian dari hasil menyimak tersebut ditulis dalam bentuk catatan. Adapun bentuk alih kode yaitu intern dan ekstern. Sedangkan campur kode berupa kata, frasa, baster, perulangan kata, ungkapan, dan klausa. Faktor yang menyebabkan terjadinya alih kode dan campur kode yaitu penutur, mitra tutur, hadirnya orang ketiga, istilah yang lebih populer, kaum terpelajar, menjelaskan sesuatu, dan keterbatasaan kode.

Banyak bermacam macam jenis video yang berkembang di media YouTube salah satunya adalah vlog. Tujuan penelitian ini untuk mengetahui adakah pengaruh vlog YouTube Jerome Polin terhadap minat siswa kelas 12 SMANU M.H Thamrin melanjutkan studi ke Jepang. Responden penelitian ini adalah siswa kelas 12 dikarenakan siswa kelas 12 akan melanjutkan studinya ke jenjang yang lebih tinggi serta umur responden termasuk kedalam kategori umur terbanyak yang menggunakan YouTube di Indonesia 15-34 tahun. Teori pada penelitian ini yaitu menggunakan teori S-O-R (Stimulus, Organism, Respons) yang dimana stimulus yaitu vlog youtube Jerome Polin, organisme yaitu siswa kelas 12 SMANU M.H Thamrin dan respon yaitu minat untuk melanjutkan studi ke Jepang. Hasil penelitian yang diperoleh adalah terdapat pengaruh positif terhadap minat siswa kelas 12 SMANU M.H Thamrin melanjutkan studi ke Jepang dan hasil uji hipotesis menyatakan bahwa ho ditolak dan ha diterima yang artinya terdapat pengaruh antara vlog YouTube Jerome Polin terhadap minat siswa kelas 12 SMANU M.H Thamrin melanjutkan studi ke Jepang.

Saat ini, berbagai informasi, karya, dan ide dapat dilihat di media sosial. Media sosial merupakan tempat bagi pengguna untuk berinteraksi sosial, berbagi, berkomunikasi, dan bekerja. Ini tentu memungkinkan pengguna untuk berkomunikasi dengan idola seperti penyanyi, aktris, penulis, dan komedian. Jerome Polin memiliki 6,9 juta pengikut di media sosial Instagram . Tujuan dari penelitian ini adalah mendeskripsikan bentuk tindak tutur lokusi dan ilokusi serta fungsinya pada akun Instagram Jerome Polin. Penelitian ini menggunakan metode deskriptif kualitatif. Sumber data dalam penelitian ini adalah caption pada akun Instagram Jerome Polin pada bulan Desember hingga April. Teknik pengumpulan data dilakukan dengan metode dokumentasi. Hasil penelitian menemukan 121 data berupa tindak tutur lokusi dan 87 data berupa tindak tutur ilokusi. Tindak tutur lokusi dan ilokusi digunakan sebagai tempat untuk menunjukkan apa yang ingin disampaikan penutur kepada pengikutnya di Instagram , mulai dari menginformasikan, menceritakan, berterima kasih, berterima kasih, senang, sedih, memesan, bertanya, dan menawarkan. Penelitian ini diharapkan dapat mengembangkan bidang pragmatik dan masyarakat dapat memahami makna tindak tutur lokusi dan ilokusi yang berguna dalam kehidupan.

CEL/TEZA: Artykuł przedstawia wyniki badań, których celem było ustalenie zakresu i sposobów wykorzystywania technologii cyfrowej przez dwa znane polskie muzea narracyjne: Muzeum Powstania Warszawskiego (MPW) i Muzeum Historii Żydów Polskich POLIN (Muzeum POLIN). KONCEPCJA/METODY BADAŃ: W przedstawionych studiach wykorzystano trzy techniki badawcze: analizę źródeł, która pozwoliła na zorientowanie się w zmieniającej się roli muzeów oraz wykorzystywanie przez te placówki technologii cyfrowej; wizję lokalną i wywiad z pracownikami Muzeum Powstania Warszawskiego i Muzeum Historii Żydów Polskich POLIN, które pozwoliły na zapoznanie się z zastosowanymi w tych placówkach elementami technologii cyfrowej; analizę stron internetowych wspomnianych dwóch muzeów. Triangulacja tych technik umożliwiła przedstawienie systematycznego przeglądu zastosowań różnych narzędzi i metod technologii cyfrowej w różnych obszarach i formach działalności wybranych muzeów. WYNIKI I WNIOSKI: Badania pozwoliły stwierdzić, że wdrażanie w muzeach technologii cyfrowej jest procesem długotrwałym i dalekim od zakończenia, choćby z uwagi na pojawiające się ciągle nowe rozwiązania technologiczne. Polskie muzea obecnie nie wykorzystują wszystkich możliwości technologii cyfrowej, ale z uwagi na rosnące wymagania zwiedzających będą zapewne zmuszone do wdrożenia najnowocześniejszych rozwiązań. ORYGINALNOŚĆ/WARTOŚĆ POZNAWCZA: Artykuł prezentuje dziewięć obszarów działalności muzeów, w których można wykorzystać technologię cyfrową. Stanowi skrócone kompendium informacji z tej dziedziny wiedzy i praktyki, zilustrowane przykładami zastosowań poszczególnych technologii i narzędzi w dwóch muzeach poddanych analizie.

Tindak tutur direktif dapat ditemukan dalam tuturan influencer ketika mengiklankan suatu produk atau jasa perusahaan tertentu di media sosial miliknya. Penggunaan bahasa influencer tersebut tentu didominasi tuturan yang bertujuan mengajak, memerintah, atau bahkan merayu pendengar agar tertarik pada produk yang diulas. Dalam penelitian ini, penulis menganalisis tuturan influencer Jerome Polin. Hal ini dikarenakan (1) dia seorang influencer di bidang pendidikan yang memiliki jutaan pengikut di media sosialnya, (2) dia banyak melakukan promosi barang atau jasa perusahaan di akunnya. Even though a gender perspective, in reference to various aspects of museums and their exhibits, permeates the reflection on museums, gender is not explicitly taken up as a category of knowledge within the self-reflective narratives about the core exhibition or the conceptualization of the Holocaust gallery in POLIN Museum of the History of Polish Jewish, which opened in Warsaw, Poland in 2014. Building upon the research gendering the memory of the Holocaust, especially with regard to historical exhibitions, and using a cultural studies framework to the study of representation, this article asks how femininities are framed by the representation of masculinities and how museum technologies work to produce gendered meanings. It concludes that most of the Holocaust gallery in POLIN problematically instrumentalizes gender relations to underpin a chronological historical narrative. In a dialogue with queer research on temporality, underscoring the coincidence of normative gender/sexuality and linear progressive narrative, it analyses this strategy as gender chronotechnology.
