Abstract:
Dyslipidemia is one of the high-risk factors that cause cardiovascular disease and the prevalence is increasing all the time. Garlic (Alii sativi) has been suggested to reduce serum lipid level. Poly-herbal is herbal medicine that has been widely used in Indonesia, and it contains Alii sativi, Belericae fructus, Curcumae aeroginosae, and Amomi fructus extract. This study aimed to evaluate the effects of poly-herbal tablet on lipid levels in dylipidemia subjects. The study was conducted for 6 weeks on 58 dyslipidemic subjects. The subject was divided into 2 groups. Group 1 was given Poly-herbal with the dose 2x1200 mg, and group 2 was not given anti-dyslipidemic drugs (control group). Serum lipid level (total cholesterol, LDL, HDL, and triglycerides) were checked before the treatment (H0) and after the treatment (M6). There was a significant difference on the reduction of serum lipids level between the group that given Poly-herbal vs control group, respectively total cholesterol (-12.04±3.2 vs 8.38±4.3; p=.000), LDL (-4.42±2.8 vs 6.93±4.7; p=.049) and HDL (-2.62+1.0 vs 3.31+1.4; p=.005). Triglyceride level on Poly-herbal group is not significantly decreased. In Poly-herbal group there was also a decrease in HDL levels which was not statistically significant (-2.62±1, p=.015). This is probably due to the HDL baseline in the control group included in the normal HDL category (57.38 ± 14.9) so that the HDL levels after being given treatment did not increase. This study suggests that using Poly-herbal in dyslipidemia subjects can reduce the level of total cholesterol and LDL.

Abstract:
3C-SiC layers of different microstructures (monocrystalline (100) and (111) oriented and polycrystalline) were implanted with high energy (800 keV) 129Xe++ ions. Implantations were performed at room temperature (RT) and at 500 °C using two different fluences of Φ1 = 1x1016 and Φ2 = 1x1017 at/cm2. Surface blistering was only observed for RT and Φ2 implantations into poly-SiC material while mono-SiC kept rather smooth surface. This was due to more homogeneous Xe bubbles distribution (200 nm deep) in the mono-SiC than in the poly-SiC. Xe retention was found to be almost complete for all samples. Some Xe enhanced diffusion was detected in the poly-SiC material which was attributed to grain boundaries. Some irradiation-induced oxidation effect was evidenced, O element being located at the depth where Xe bubbles are accumulating. This was more pronounced for poly than for mono-SiC. These results demonstrate that SiC microstructure affects many aspects of its behavior upon Xe irradiation.

Abstract:

Abstract:
High-valent metal-oxo, -peroxo and -nitrene cores of late transition metals have been proposed as reactive intermediates in transformations that are both technologically attractive and fundamental for the functioning of biological systems, like the activation of C−H bonds in hydrocarbons and the reduction of dioxygen. The isolation and characterization of such species, despite being challenging, provides valuable information about the mechanisms of the reactions performed and can help in the design of improved catalysts. In this work, three different kinds of intermediates were studied, namely iron-oxo, cobalt-peroxo and copper-nitrenes. Several new reactive species were isolated and spectroscopically characterized, and their reactivity was thoroughly investigated. It was shown that the reactivity patterns of the high-valent reactive intermediates here studied depend greatly on subtle changes in the ligand system, the metal center and the oxidant employed.

Abstract:

An objective of animal carcinogenicity studies is to identify a tumorigenic potential in animals and to assess relevant risks in humans. Without using the cause-of-death information, the Cochran-Armitage test is applied for detecting a linear trend in the incidence of a tumor of interest across dose groups. The survival-adjusted Cochran–Armitage test, known as the Poly-k test, is investigated for the animals not at equal risk of tumor development by reflecting the shapes of the tumor onset distributions. In this thesis, we will validate Poly-k test through a Monte Carlo simulation study. We will design the simulation study to assess the size and power of the poly-k test using a wide range of k values for various tumor onset rates, for various competing risks rates, and for various tumor lethality rates. In this thesis, the Poly-k testing approach will be investigated to evaluate a dose-related linear trend of a test subject on the incidence of tumor and will be implemented in R package to be used widely amongst toxicologists.

Damodaran, Vinod Babu. "Solid-phase protein PEGylation: Achieving mono-PEGylation through molecular tethering." Thesis, University of Canterbury. Chemical and Process Engineering, 2009. http://hdl.handle.net/10092/3757.

Full text
Abstract:
Protein PEGylation (covalent attachment of poly(ethylene glycol) or PEG to proteins) is an excellent example of a drug delivery system that improves pharmacokinetics and pharmacodynamic properties of therapeutics. However, although PEGylation is clinically proven and attracts both scientific and commercial interest, the technique is associated with many process constraints, in particular related to controlling the number of conjugated PEG chains. A novel, solid-phase PEGylation methodology was attempted to overcome the drawbacks of the commonly used solution-phase methods for preparing PEGylated products. The solid-phase PEGylation methodology involved conjugating protein onto a tethered PEG derivative attached onto a solid matrix, followed by hydrolytic cleavage of the PEG chain from the solid matrix under mild conditions to yield PEGylated protein in free solution. PEGs with molecular weights (MWs) 2000 and 4000 Da were used and a heterobifunctional PEG derivative, α-(β-alanine)-ω-carboxy PEG, with a cleavable β-alanine ester terminal was prepared for surface grafting and protein conjugation. The amine terminal of this PEG derivative was used for grafting PEG onto carboxy functionalized hydrophilic Sephadex and hydrophobic polystyrene derivatives. The free carboxyl terminal was used for protein conjugation via amine coupling. A kinetic study of PEG-surface grafting was performed to understand the influence of a number of parameters on the PEG surface concentration and its conformation, including temperature, reaction time, nature of the matrix, solvent and base, and MW of PEG. PEG grafted matrices were characterized using various surface characterization tools including Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and X-ray photoelectron spectroscopy (XPS). Higher PEG grafting was observed with polystyrene matrices (up to 0.3 mmol/g) than either of the Sephadex derivatives (less than 0.15 mmol/g) using both molecular weights. Detailed surface characterization using XPS studies showed a layer thickness of 11.87 nm was achieved with polystyrene matrices using 4000 Da PEG derivatives after a grafting period of 72 hours at 40°C, indicating the presence of brush conformations for the grafted PEGs. In contrast, mushroom conformations were observed for PEG molecules grafted on both carboxymethyl and carboxypentyl Sephadex derivatives after the same reaction period, with a layer thickness of 2.62 nm and 4.14 nm respectively. Optimized PEG grafting and hydrolysis conditions were developed for solid-phase protein PEGylation using Cytochrome c as a model protein. The presence of PEGylated species were detected by size exclusion chromatography (SEC) from Sephadex derivatives but were absent when using polystyrene matrices. Both Sephadex derivatives gave mainly multi-PEGylated species with poor yields, in place of the expected mono-PEGylated products. A solution-phase PEGylation using the same PEG derivatives was performed successfully and various PEGylated species were identified and characterized using SEC and gel electrophoresis, based on their viscosity radius. An examination of the surface characteristics of the PEG-grafted was carried out by XPS, showing that protein conjugation was greatly influenced by surface force interactions, which depended on the PEG grafting densities and the nature of the solid matrices. Finally, fluorescent images obtained using confocal microscope with fluorescein isothiocyanate labelled Cytochrome c provided supporting evidence regarding the factors that constrained the solid-phase PEGylation process.

Abstract:

"From mono- to poly-nuclear late-transition metal catalysts for the polymerisation of ethylene." Thesis, University of Leicester, 2006. http://hdl.handle.net/2381/29986.

Full text
Abstract:
In Chapter One, an introduction to the field of metal-mediated olefin polymerisation catalysis is undertaken. The area of non-metallocene monometallic systems and, in particular, imino-based chelated (NN-, NNN-) late transition metal systems is briefly documented with a view to introducing the emerging field of olefin polymerisation mediated by multi-component catalysts Concurrent Tandem Catalysis (CTC) and Encapsulated Bimetallic (EB). The Chapter concludes with a description of the objectives set for the thesis. Chapters Two to Five discuss the synthesis and characterisation of mono- and bi metallic complexes (cobalt, iron and nickel) containing mono- and bi-nucleating ligands that incorporate bi- and tri-dentate binding domains (viz., iminophenanthroline, iminobipyridine, iminopyridine, a-diimine). Each Chapter is dedicated to the preparation of a specific type of precatalysts monometallic (Chapter Two), fused EB (Chapter Three), remote EB (Chapter Four) and functionalised remote EB (Chapter Five). In Chapter Two, the syntheses of monometallic complexes are described. Chapter Three reports the preparation of fused EB precatalysts, in which the two catalytic sites share a central aryl moiety. In Chapter Four, remote EB precatalysts have been developed, each possessing two discrete binding domains tethered by a methylene linker. Chapter Five is concerned with the functionalisation of the linker in remote EB iminopyridine-based nickel precatalysts/catalysts All complexes incorporate electronically active groups on the ligand manifold. Chapter Six compares the performances of the complexes from all four chapters by our complexes for the polymerisation of ethylene. Both the oligomer and the polymer portions were analysed through a range of techniques (e.g., GC, GPC, DSC, *H and C NMR). Regardless of the metal employed, encapsulated bimetallic catalysts produced higher molecular weights PE compared to their monometallic counterparts. In Chapter Seven, the details of the experimental procedures are disclosed.

At the Place Louis XV the three young people separated,—that is to say, Morrel went to the Boulevards, Château-Renaud to the Pont de la Révolution, and Debray to the Quai. Most probably Morrel and Château-Renaud returned to their ‘domestic hearths,’ as they say in...

Abstract:
In this paper, the passive optical network (PON) evolution trend has been discussed. The literature of PON standards which are ATM PON (APON), Broadband PON (BPON), Gigabit PON (GPON), 10-Gigabit-capable passive optical networks (XG-PON) and next-generation PON2 (NG-PON2) has been reviewed extensively. From the literature review, a comparison of the PON technologies is made. The GPON, XG-PON and NG-PON2 are the most recent PON generations, and these are simulated to find the split per wavelength supported. The Q-factor below 6 is considered unacceptable in the analysis. The results show that the GPON can support 32 ONU when transmitted power set to 3dB. In the case of XG-PON, the Q-factor remains more than 6 for a split ratio up to 16. For the NG-PON2, the observed Q-factor is more than 6 for split up to 32. However, as the split increases above 32, the Q-factor falls below the acceptable level of 6.

Abstract:
Conjugated polymers are materials that have attracted much attention from the research community because of their charge and energy transport properties. In this sense, it is necessary to understand the mechanism behind exciton transfer in this particular class of systems. However, direct application of procedures done for different organic compounds is not straightforward for long polymeric chains, because such procedures would be computationally impracticable. In that matter, alternative treatments are required. In this work, we perform spectrum simulations for poly-thiophene (PTH) and poly(p-phenylene vinylene) (PPV) chains by analyzing the evolution of electronic properties with oligomer sizes and its effects on exciton diffusion. Furthermore, employing a kinetic Monte Carlo model, we also investigate the efficiency of intrachain exciton diffusion. Our results show a reliable description of the optical properties for long polymeric chains, and a comparison is made between the different approaches in describing the optical properties of such polymers. This study may be useful in the development of more sophisticated optoelectronic devices that use conjugated polymers as its active materials.
