Full textHigh-valent metal-oxo, -peroxo and -nitrene cores of late transition metals have been proposed as reactive intermediates in transformations that are both technologically attractive and fundamental for the functioning of biological systems, like the activation of C−H bonds in hydrocarbons and the reduction of dioxygen. The isolation and characterization of such species, despite being challenging, provides valuable information about the mechanisms of the reactions performed and can help in the design of improved catalysts. In this work, three different kinds of intermediates were studied, namely iron-oxo, cobalt-peroxo and copper-nitrenes. Several new reactive species were isolated and spectroscopically characterized, and their reactivity was thoroughly investigated. It was shown that the reactivity patterns of the high-valent reactive intermediates here studied depend greatly on subtle changes in the ligand system, the metal center and the oxidant employed.

Full textAn objective of animal carcinogenicity studies is to identify a tumorigenic potential in animals and to assess relevant risks in humans. Without using the cause-of-death information, the Cochran-Armitage test is applied for detecting a linear trend in the incidence of a tumor of interest across dose groups. The survival-adjusted Cochran–Armitage test, known as the Poly-k test, is investigated for the animals not at equal risk of tumor development by reflecting the shapes of the tumor onset distributions. In this thesis, we will validate Poly-k test through a Monte Carlo simulation study. We will design the simulation study to assess the size and power of the poly-k test using a wide range of k values for various tumor onset rates, for various competing risks rates, and for various tumor lethality rates. In this thesis, the Poly-k testing approach will be investigated to evaluate a dose-related linear trend of a test subject on the incidence of tumor and will be implemented in R package to be used widely amongst toxicologists.

Billions of individuals across the world lack clean, affordable drinking water, and the unavailability of fresh drinking water can be attributed to both physical and economic reasons. Several techniques have been utilized to produce potable water for human consumption that include both water desalination and recycling procedures. Water desalination is a process that allows for purifying salt contaminated water into drinking water. The two major desalination processes involve either distillation or passage through polymer membranes. Distillation separates water from salt by heating liquid water to form a gas, and collecting the vapor as condensate while impurities remain in the heated bulk material. Polymer membranes separate impurities through filtration where membranes allow water to pass through a physical barrier while rejecting the unwanted contaminants, including salt. Reverse osmosis desalination is the most common membrane separation process. Reverse osmosis membranes are comprised of either short-chain crosslinked oligomers or long-chain linear polymers. Commercial reverse osmosis membranes are largely poly(amide)s where a thin film is formed in an interfacial reaction. The membranes allow for almost quantitative salt rejection with high water fluxes. But, these membranes degrade over time from periodic cleaning with chlorine disinfectants. This dissertation primarily focuses on the implementation of an alternative polymer membrane material known as a mono-sulfonated polysulfone that strategically distributes the fixed sulfonate charged groups along the polymer backbone. Theses reverse osmosis mono-sulfonated polysulfones display comparable salt rejection with better chemical resistance than commercial poly(amide)-based membranes, and could potentially offer a replacement in the market.

Full textAbstractIntroductionIPS e.max® Press is a polychromatic, lithium disilicate glass-ceramic designed for thermal pressing. It is produced in various degrees of opacity, translucency, color and saturation. A polychromatic layered variant, the IPS e.max® Press Multi, has been developed to show gradual shifts of opacity, translucency, color and saturation similar to those of natural teeth.PurposeThe purpose of present study is to compare the strength of polychromatic, layered, standardized norm crowns made of multi ingots and standardized norm crowns made of the conventional monochromatic ingots.Material and methodFor the study, 30 specimens of standardized norm crowns were generated and divided into three spci- men groups: ten IPS e.max® Press LT A2, ten IPS e.max® Press Multi A2 and ten IPS e.max® Press Multi BL2. 30 dies were produced in a light-curing 3D material.The crowns were thermocycled, whereupon they were cemented on the printed dies, followed by cyclic mechanical pre-load before load to fracture in a universal testing machine. The values at fracture were recorded in Newtons (N). Analysis of the fracture strength was performed using one- way ANOVA, Tukey’s test. The confidence interval was 95% and the significance level was α= 0.05.ResultsA statistically significant difference in breaking strength was detected between all three groups. The group with the lowest average result was A Multi (IPS e.max® Press Multi A2), with 866 N, followed by group Multi B (IPS e.max® Press Multi BL2) with 987 N. The highest average was detected in group Mono C (IPS e.max® Press LT A2) with 1108 N.ConclusionWithin the scope of present study, the following conclusions can be drawn:• The monochromatic, lithium disilicate glass-ceramic IPS e.max® Press LT A2 has a higher fracture strength than the polychromatic glass-ceramics IPS e.max® Press Multi A2 and BL2.• The hypothesis is confirmed, but further studies on the subject are necessary to secure sufficient results.

Full textThe constant evolution of the data storage needs over the last decades have led the technological landscape to completely change and reinvent itself. From the early stage of magnetic storage to the most recent solid state devices, the bit density keeps increasing toward what seems from a consumer point of view infinite storage capacity and performances. However, behind each storage technology transition stand density and performances limitations that required strong research work to overcome. This manuscript revolves around one of the promising emerging technology aiming to revolutionize data storage landscape: the Conductive Bridge Random Access Memory (CBRAM). This technology based on the reversible formation and dissolution of a conductive path in a solid electrolyte matrix offers great advantages in term of power consumption, performances, density and the possibility to be integrated in the back end of line. However, for this technology to be competitive some roadblocks still have to be overcome especially regarding the technology variability, reliability and thermal stability. This manuscript proposes a comprehensive understanding of the CBRAM operations based on experimental results and a specially developed Kinetic Monte Carlo model. This understanding creates bridges between the physical properties of the materials involved in the devices and the devices performances (Forming, SET and RESET time and voltage, retention, endurance, variability). A strong emphasis is placed on the current limitations of the technology previously stated and how to overcome these limitations. Improvement of the thermal stability and device reliability are demonstrated with optimized operating conditions and proper devices engineering

Full textCellular senescence is a complex stress response that arrests cell proliferation and is accompanied by widespread changes in metabolism, chromatin structure, and gene-expression, including the overexpression and secretion of inflammatory factors. Cellular senescence is health-promoting as a tumor-suppressive mechanism, facilitating embryonic development and tissue regeneration. However, it is also considered a major contributor to aging and age-related diseases, mostly through its inflammatory phenotype, the so-called SASP (senescence-associated secretory phenotype). Current research supports the role of PARP1 (Poly (ADP-ribose) polymerase 1) in the transcriptional regulation of inflammatory processes and modulation chromatin structure. However, the exact mechanisms by which PARP1 exerts its regulatory functions, and its roles in the context of regulating senescence gene-expression are underexplored. In my thesis, I set out to define the functional role of PARP1 catalytic and chromatin binding activities in gene regulation and chromatin structure in cells undergoing senescence. I performed time-resolved transcriptomics, chromatin-accessibility studies, and mapping of the genome-wide locations of PARP1 using ChIP-seq and ADP-ribosylated chromatin using a novel technique CRAP-seq (Chromatin-Ribosylation-Affinity-Pulldown). Together, I identified a dichotomy of PARP1 function – one related to its enzymatic ADP-ribosylation activity and the other related to its non-enzymatic chromatin binding activity – with distinct impacts on the senescence transcriptional program. Based on these findings, I can define a novel and global role for PARP1 in and chromatin structure modulation by stabilizing nucleosomes positioning at gene promoters and ADP-ribosylation of cis-regulatory modules to fine-tune transcription of lowly expressed genes. Indeed, based on my investigations, the role of PARP-inhibitors in senescence targeting therapies (senolytic therapies) for the treatment of age-related pathologies can be envisioned

Full textBesides exerting cytostatic or cytotoxic effects on tumor cells, some anti-cancer therapies (anthracyclines, oxaliplatin, X-Rays) could trigger an immunogenic cell death modality, releasing danger signals to alert immune system. We have shown that tumor-specific IFN- producing CD8+ T cells (Tc1) are mandatory for the success of chemotherapy to prevent tumor outgrowth. Priming of Tc1 response depends on IL-1β secretion by DC confronted with anthracycline-treated tumor cells releasing ATP. To identify the inflammatory components which link innate and cognate immune responses, we analyzed the influence of immunogenic chemotherapy on tumor microenvironment. We found an upregulated Th1- and Th17-related gene expression pattern in growth-retarded tumor after anthracycline treatment. By interfering with IFN- or IL-17A pathways, therapeutic effect of doxorubicin and oxaliplatin was abolished and dying tumor cell-based vaccine lost its efficacy to protect mice from live tumor cell rechallenge. Interestingly, we discovered that distinct subsets of T lymphocytes (V4+ and V6+) colonized tumors shortly after chemotherapy, where they proliferated and became the dominant IL-17 producers within tumor beds. In three tumor models treated with chemotherapy or radiotherapy, a strong correlation between the presence of IL-17-producing T ( T17) and IFN--producing CD8+ TIL (Tc1) was discovered. IL-17A signaling acts as upstream of IFN- since defect in IL-17RA led to complete loss of antigen specific Tc1 priming. The contribution of T17 cells (V4+ and V6+) to chemotherapy is critical as V4/6-/- mice showed reduced sensitivity to chemotherapy and vaccination. Also, tumor infiltrating T17 and Tc1 cells were reduced to basal level in this strain. IL-1β/IL-1R, but not IL-23/IL-23R, is pivotal for IL-17 production by T cells and the success of chemotherapy. Importantly, adoptive transfer of T cells could restore the efficacy of chemotherapy in IL-17A-/- mice and ameliorate the effect of chemotherapy in wild type host, provided that they retain the expression of IL-1R and IL-17A. Our research suggest a DC (IL-1β) → T cells (IL-17) → Tc1 (IFN-) immune axis triggered by chemotherapy-induced dying tumor cells, which is critical for the favorable therapeutic response. To boost the immune system, we try to combine immunogenic chemotherapy with tumor vaccine in the presence of TLR3 agonist Poly (A:U). This sequential combined therapy, which we named VCT, could significantly retard tumor growth or even completely eradicate tumor and establish long-term protection against rechallenge in highly tumorigenic models. To dissect the effect of Poly (A:U) on immune system and that on TLR3 expressing-tumor cells, we performed VCT treatment in nude mice, TRIF-/- mice and with TRIF-silencing tumors. Interestingly, our results suggested that anti-tumor effect of VCT required T cells and intact TRIF signaling pathway at the level of the host and that of tumor cells. Poly (A:U) treatment could induce high level of CCL5 and CXCL10 production from tumor cells both in vitro and in vivo, which could negatively and positively influence the therapeutic outcome. By uncoupling the effect of CCL5 from that of CXCL10, the VCT treatment can be ameliorated. Our study emphasizes that both tumor and host derived inflammatory factors participate in regulating anti-tumor response. We also highlight that therapeutic application of TLR agonists can be optimized through regulating the profile of chemokines and their downstream signaling events

Sub-micron patterning is an integral part of making many modern technologies such as memory storage devices or integrated circuits. As this technology becomes smaller and smaller, the limiting factor for making these devices has become the ability to manufacture effectively at the appropriate scale. Traditional commercial manufacturing methods lack the resolution needed to attain small enough features. Manufacturing methods that can make small enough features are often either extremely expensive or offer incomplete control of the feature morphology. Imprint lithography is a high-throughput, inexpensive alternative to making sub-micron features that can be tailored to work with a variety of materials. Imprint lithography is simple process in which a patterned stamp is pressed into a softened film of material in order to transfer the pattern of the stamp onto that material. Traditionally, imprint lithography works best with polymers and researchers have struggled to pattern nanoparticle-based materials. This work has achieved high-fidelity pattern transfer onto polymer-nanoparticle hybrid films with feature sizes on the same order as the polymer films found reported in literature. The polymer-nanoparticle hybrid was realized by creating a liquid suspension of functionalized ZnO nanoparticles and poly(methyl methacrylate) (PMMA) in a solvent. The ZnO particles were functionalized by adding nonanoic acid, allowing the normally polar particles to disperse in the non-polar solvent needed to dissolve the PMMA. This suspension was spread onto a glass substrate, imprinted with a patterned stamp, allowed to dry, and was demolded. The final result was the successful transfer of features ranging from 250 nm to 1 μm in size with good fidelity. The effect of the ZnO content as well as the method of combining the suspension components on the feature fidelity was studied. To help prove the broad applicability of this imprint method, it was adapted for use with polymer-derived ceramics. Additionally, a computer simulation was developed to help understand the behavior of the nanoparticle-polymer suspension during the imprint process.

The question of the world as phenomenological problem opens up the field of research to the body and to others. The world is the place where corporeity and otherness come together. Analyses of the body-subject refer consistently to the body of the other, be they in the studies of touch, sexuality, or speech. Full textPoly, to live in several loving and intimate relationships at the same time, is one alternative to the mono norm. In Sweden polygamy is illegal yet in recent years poly has slowly started to enter the hetero normative political agenda and raise debate in the media. But what does poly mean, and how do those who identify with this type of relationship describe it in contrast to mono? The theoretical framework is based on a structural viewpoint of the changes in the organisation of the family and the growing de-traditionalism of society. Amongst other I use a Foucauldian perspective to explain the structural shift (yet not replacement) between an Alliance pattern and a Sexuality pattern. Other theories in use are that of the emerging ideal of The Pure Relationship and Queer Tendencies. The essay draws its empirical results from seven e-mail and/or face interviews with individuals who identify with poly. The results show that if poly and mono are both understood as expression of the pure relationship they are potentially not so different. Yet the stigma and the lack of role models show that there is much needed public debate about it to unleash it from old discourses that still seem to uphold the equation that love equals two partners. Drawing attention to queer mechanism and the initiated process of the decentralisation of the heterosexual norm, I speculate that in the future polygamy must not be a foreign concept in Swedish society.

Full textThe aim of this work was to evaluate if poly(ADP-ribose)polymerase inhibition could be a therapeutic strategy for traumatic brain injury (TBI), as excessive activation of this enzyme by oxidative stress (OS) induces cell death by energetic depletion. A cerebral OS in vivo leads to nitrosative stress and PARP activation. 3?aminobenzamide (3AB), a PARP inhibitor, reduces the lesion and PARP activation, showing the deleterious role of this enzyme in cell death induced by cerebral OS. TBI caused by fluid percussion leads to nitrosative stress and PARP activation. 3AB reduces the neurological deficit, the lesion and PARP activation. Two PARP inhibitors more potent than 3AB, PJ34 and INO-1001 reduce the deficit, the PARP activation without affecting the brain lesion. In conclusion, our work shows that PARP is involved in the pathogenesis of TBI and may be a promising therapeutic target for brain trauma. However, complementary studies are needed to evaluate which PARP isoforms to inhibit

Full textOptical access networks are essential to meet overgrowing demands of bandwidth and to facilitate the future telecom services. Lt presents an interesting solution for broadband access networks compared to DSL technology which has a limited data rate as weil as a short geographical reach eligibility. Actual optical access networks solutions are based on classical and simple NRZ (Non Return to Zero) modulation technique of the signal in the optical domain. Recently, Orthogonal Frequency Division Multiplexing (OFDM) modulation has been proposed to better exploit the capacity of the optical channel. Coupled with resources allocation techniques, OFDM transmission takes into consideration the channel quality in order to maximize the overall data rate. This thesis experimentally implements OFDM coupled with bit and power allocation algorithms in a downstream Next Generation PON (NG-PON) architectures. First, OFDM coupled with Levin-Campello algorithm so called Adaptively Modulated Optical OFDM (AMOOFDM) is presented for single user OFDM-based systems. Then, another technique Optimal Spectrum Balancing (OSB) is used for resource allocation in multi-user OFDM-based systems. Lmplementing OSB, Multi-Band OFDM (MB-OFDM) has been experimentally demonstrated, then a comparison is made between MB-OFDM and Orthogonal Frequency Division Multiple Access (OFDMA) both using OSB algorithm. The impact of the laser chirp and optical fibre chromatic dispersion on transmission performance is studied as weil as a comparison is made between APD and PIN photodiodes both using AMOOFDM signal

Full textDietary lipids are able to affect fasting and postprandial lipoprotein metabolism in a significant way, thereby modifying the risk of cardiovascular disease, a major cause of morbidity and mortality. The aims of the present study were firstly to evaluate in normolipidemic healthy men, the qualitative effect of dietary fatty acids on fasting and postprandial lipid parameters, in order to define an optimal range of monounsaturated fat supply and secondly to determine the role of postprandial lipoprotein modifications in the mechanisms involved in the reverse cholesterol transport. Our results showed that in conditions of balanced and stabilized dietary fat supply comprising 11% to 16% monounsaturated, associated with 5-6% polyunsaturated fatty acids ( of total energy intake),plasma lipid parameters

Full textThis dissertation examines the universal phenomenon of poly-divergence in archaic Chinese with five subtypes: the polygrammaticalization with the case of huò 或; the polylexicalization and the hybridization of these two with the case of rán 然; the poly-divergence under the influence of cult-culture-philosophical with the case of yī 一; the poly-divergence of the multifunctional structure with the case of "Reference + Comment"; the poly-divergence of the phrase with the case of "numeral +month". These divergences have the effect of reducing and avoiding the opacity caused by the non-obligatory usage of function word or the lack of exclusive function word. And these types of poly-divergence in Chinese are realized through mechanisms that are distinct from those of Western languages. Finally, a conclusion is drawn and it is argued that the poly-divergence is the essential model of evolution in archaic Chinese

Full textWhat is the « milieu »? What more is there to the milieu than to space, to the environment, and to the world? The etymology that connects the "between" to the "around" is significant, as is problematic the history of a word that turns against its own self, against its physical origins, in the name of the irreducibility of the living being in its constituent relation to its milieu. A relation, as opposed to a link, presupposes that the terms brought into relation do not preexist to the interaction. It is therefore useless to seek who, of man or his milieu, determines the other. What is a living individual? What does it mean "being in relation"? What does it mean "being-to-the milieu"! This thesis assumes that these three questions are one. The "mi-lieu", designating the ambivalence of the "in-between", does not only speak of the living being, it also speaks of philosophy. Against the impatient souls without history rushing to put an end to metaphysical dualisms, we would like to show that there is no way out of the Two, other than to put oneself in between, in the mi-lieu (medium). At first, we will appreciate the pregnancy of the mi-lieu in some canonical texts. Then, we will propose a history of the milieu, given that the concept is not the word, and the word is not the concept. The core of the analysis lies in the transition, which became a breakdown, from physics to biology. Special attention will be given to what has been called the Great Dualism of the World and of Life. Do we have to surrender to it?

Full textThe purpose of this thesis is to define and identify the effects of price discrimination policies on the use and organization of urban public transports. Price discrimination is the possibility to price a service with different level of tariff. At first, we will define the issues around public transport fares, whose complexity is now mitigated by existing smart cards and other payment tools. Then, the spatial dimension of the fare will be given special attention through the study of the relationships between fee structure and spatial morphology of 10 cities around the world. The relationships between urban morphology and pricing will be confirmed. Knowing that usage of public transport are diverse, it is essential to have a good knowledge of users habits. Smard Card data reconstruction provides geographical and statistical information that can be re used for urban planning and for pricing policy. How ?Grand Paris orbital line 15, will be launched in 2020. It is an example of introducing a new metro line inducing significant time savings for commuters. Passengers will be given a real alternative between their usual route and the new one. The new route can offer several advantages: gain of time spent during the trip, gain in terms of number of transfers, comfort gain, security gain, price advantages. This could be an opportunity for an agency to review its fare policy. Our model is based on existing travel flows derived from the itinerary reconstruction from Navigo Smart Card pass. It establishes the principles of a fare grid based on travelers’ preferences and current travel patterns. For this research, we used data from public transport pass users in the Paris’ Metropolitan area, for one month. Finally, we show that price discrimination is related to "mobility" and not to “public transport”. If we consider the mobility needs of the user, pricing is no longer that of “transportation” but a package of services enabling access to different forms of mobility. Therefore it is transport governance that can be a problem. How to federate all mobility stakeholders in France? This dimension is essential for public transport authorities. It is a key success factor for achieving ambitious and sustainable urban planning policies

Full textPoly(ADP-ribose) polymerase inhibitors (PARPi) selectively target cancer cells with DNA repair deficiencies such as BRCA1/2 mutations or ERCC1 defects. Clinically, several PARPi are currently approved for the treatment of BRCA-mutant or platinum-sensitive advanced ovarian and breast cancers, and ongoing clinical trials are investigating the efficacy of PARPi in platinum-sensitive Non-Small Cell Lung Cancer (NSCLC). While PARPi constitute potent targeted therapies for the treatment of DNA repair-deficient malignancies, an increasing number of clinical trials are also evaluating their efficacy in combination with immune checkpoint inhibitor (ICI) in various populations. In this context, it is of critical importance to better understand how PARPi might modulate immune responses against cancer, and to investigate the inherent immunological potential of these agents.In this study, we show that ERCC1-defective NSCLC cells exhibit an enhanced type I interferon (IFN) transcriptomic signature and that low ERCC1 expression correlates with increased lymphocytic infiltration in human NSCLC tumours. Using isogenic cell lines and patient-derived xenografts, we further demonstrate that several clinical PARPi, including olaparib and rucaparib, display cell-autonomous immunomodulatory properties in ERCC1-defective NSCLC and BRCA1-mutant triple-negative breast cancer (TNBC) models. Mechanistically, PARPi generate cytoplasmic chromatin fragments with micronuclei characteristics; this activates the cGAS/STING pathway and elicits downstream type I IFN signalling and CCL5 secretion. Importantly, these effects are suppressed in BRCA1-reverted TNBC cells and ERCC1-rescued NSCLC cells, suggesting that DNA repair defects exacerbate the innate immunity-related phenotypes triggered by PARPi. Similarly, these effects are totally abrogated in PARP1-null TNBC cells, supporting the on-target effect of PARPi in mediating such phenotypes. Besides this potential to activate tumour cell-autonomous immunity through cGAS/STING and type I IFN signalling, we also observed that PARPi synergize with type II IFN to induce PD-L1 expression in NSCLC cell lines and fresh patient tumour cells, especially in the ERCC1-deficient setting. Moreover, we show that lethal concentrations of some PARPi independently activate the key damage-associated molecular patterns dictating the immunogenicity of cancer cell death, including calreticulin exposure at the tumour cell surface, ATP secretion and HMGB1 release in the extracellular compartment.Together, these preclinical data suggest that PARPi have intrinsic immunomodulatory properties that activate anti-cancer immune responses; this could be exploited clinically in combination with ICI in appropriately molecularly-selected populations

Abstract In our discussion of light induced degradation for normal mono/Poly modules and passivated emitter and rear cell mono/poly crystalline silicon solar photovoltaic modules, we found that two different process technology for the p-type solar module on load and unload system had different light induced degradation. When the modules were exposured to the sun over 80 KWh/m² , the power was no clear attenuation and gradually stabilized. According to the measurement of maximum power and electroluminescent fluorescent, we found that the passivated emitter and rear cell mono/poly modules of light induced degradation is more clear than the normal mono/Poly modules. It should be a relationship with boron and oxygen atom content of the passivated emitter and rear cell mono/poly modules. Sunlight will result in the boron-oxygen complexes increasing and the maximum power decreasing. As the decreasing of energies such as fossil fuel, the energy price is getting higher herewith serious issues of pollution, the greenhouse effect, and climatic anomalies. On March, 2011, the Fukushima Daiichi nuclear disaster in Japan injured thousands of people to increase the safety concern of nuclear. Besides, the treatment of nuclear waste is still a big problem. Renewable energies such as solar power, wind power, and geothermal power, are good solutions to substitute those non-renewable resources. For the lack of natural resources in Taiwan, the renewable resources are more important. Solar power is the best solution since it exits everywhere. The fabrication technology of solar cells is similar with the one of semiconductors, which is mutual in Taiwan. In this research, a mono-crystalline silicon photovoltaic system and a poly-crystalline silicon photovoltaic system were build to analyze their performances and connected to commercial DC-to-AC (D/A) inverters in real environment. These D/A inverters were developed with maximum power point tracking (MPPT) function. The AC power generated from the photovoltaic systems was transferred from DC powers to the on-grid system directly. A LabView program was developed to record the generated powers and the temperatures of solar modules. The final results were calculated including the data of solar irradiance provided by the Department of Atmospheric Science of National Central University. In the analysis we can understand the relationships between photovoltaic module temperature, solar irradiance, and efficiency.
