Abstract:
The presented case study illustrates the possibility of adding value to the biological surplus remaining from the mushroom cultivation industry. In essence, the unused mushroom parts were submitted to UV-C irradiation, with the purpose of increasing the vitamin D2 content and validating its extraction. Vitamin D2 concentration in three different mushroom species (Agaricus bisporus, A. bisporus Portobello, and Pleurotus ostreatus) was obtained by high-performance liquid chromatography (HPLC), by means of an ultraviolet (UV) detector. The method was validated using an A. bisporus Portobello sample, and its reproducibility and accuracy were confirmed. Independently of the UV-C irradiation dose, the effect on vitamin D2 concentration was significant, allowing it to increase from less than 4 µg/g dry weight (dw) to more than 100 µg/g dw in all mushroom species. These results are good indicators of the feasibility of industrial surplus mushrooms as sustainable vitamin D2 food sources, besides contributing to strengthen the circularity principals associated to the mushroom production chain.

This work aimed to identify the essential competence to take competitive advantage of a ceramic tiles company concerning its performance in a market with a high competitiveness based on differentiation and sophistication, identified as the Belgian market. The factors that generated competitive advantage were collected according to the perspective of the participants of the distribution chain of the company products in the referred market. Those factors were analyzed to identify the resources and abilities which originated them to check if their combination leads to an essential competence, as well as what that would be. The design was highlighted as one of the possible abilities which generate competitive advantage because of its appropriateness among the main competitors of the studied market. The use of the concept of essential competence results from the view of strategy as a collective learning that generates unique combinations of resources and abilities allowing higher performances. The main theoretical references that constitute the development of the school of dynamic competences were collected, since the beginning of the view based on resources until the application of the concepts of essential competence. The concepts that give the design the differential characteristics which can turn it into an essential competence were also studied. This work results from a qualitative and descriptive survey which was obtained through a case study. Data was collected by indirect observation and interviews. Data analysis was inductive. The investigation led to the identification of three competences, and the design culture seemed to be the essential competence of the studied company in the Belgian market according to the perspective of the key participants of the distribution chain

Al-Timimi, Iman Abdullah Jaaffer. "Metal and oxide nanoparticles : green biosynthesis using Portobello Mushroom Spores (PMS) for nanocomposites and replicas, properties and applications." Thesis, Brunel University, 2018. http://bura.brunel.ac.uk/handle/2438/17164.

Full text
Abstract:
This thesis describes novel research using Portobello mushroom spores (PMS) as a biotemplate and reducing agent that may be used to produce nanocomposites and replicas with societal benefits. First, the use of PMS as a green eco-friendly bioreagent for the biosynthesis of Ag, Au and TiOx nanoparticles (NPs) and their hybrids is described. These have been characterised using SEM, TEM, XRD, FTIR, UV, SIMS, DLS and TGA (where a full list of acronyms is to be found on page iv). The spherical shape, location and mean diameter of the PMS-held (5-30nm) Ag and Au NPs were confirmed. PMS functional groups are comparable with those in plant sources and allow them to act as reducing/capping/stabilising agents. Second, the sequential biosynthesis of PMS-held Ca2+, CaCO3 and nanohydroxyapatite (nHAp) is described where surface nucleation and growth are facilitated by biotemplate surface hydrophilic polar groups (-OH and -COO-). XRD and HRTEM suggest that the nHAp so formed has lower crystallinity and greater directed growth in the (002) direction than commercial nHAp (e.g. Bio-Oss), making PMS-routes useful for the delivery of artificial bone (after subsequent PMS removal). Furthermore, PMS-held nHAp showed more rapid in-vitro mineralization in simulated body fluid (SBF). Third, it is shown that ascorbic acid (AA), haemoglobin (Hb) and insulin (In) can be loaded into/released by PMS through its cell walls faster than with synthetic hydrogels (PVA); the transdermal use of PMS nanocomposites is considered in the context of drug delivery nanotechnology, avoiding size-dependent toxicity. Fourth, it is shown by traditional diffusion and novel methods that the antimicrobial activity especially of Ag/PMS, but also TiOx/PMS and Ag-TiOx/PMS (but not Au/PMS) is good in the inhibition of the growth of E.coli and S.aureus bacteria. The highest activity of Ag/PMS is attributed to Agx+ release. Fifth, the synthesis of NPs/PMS and biomimetic TiOx/PMS has shown to lead to good photocatalysis for the removal of coloured organic pollutants (e.g. methyl orange) from water, with faster rates of removal in molecules/mg/s and turnover numbers (TON) (s-1) than commercial anatase-rutile TiO2 (P25) possibly as the texture of the titania replicas raises the number of multiple reflections of light.

Abstract:
In the sixteenth century, the Panamanian isthmus became the Spanish Crown’s most strategic overseas territory. The isthmus’s two main cities, Panama, in the Pacific, and Nombre de Dios, later Portobelo, in the Atlantic, were connected by the Camino Real, a road that allowed the gold and silver of the Americas to cross en route to Europe. This vital roadway was key to the economy of the Crown, but it was never safe. Paradoxically, the greatest threat to the coffers of the realm arose from black slavery, which had been introduced into America as the main driver of the economy. The attacks by the maroons (escaped slaves) on the Camino and on the main cities, as well as the alliances they built up with the corsairs, posed a real threat to the stability of the colonial order. The concept of marronage became one of the links of union between the metropolis and its colony, since letters, reports and royal decrees crossed the Atlantic in both directions as the colonial authorities sought ways of bringing it to an end. When their military methods proved ineffective, they were obliged to negotiate with the main maroon communities of the isthmus and, as a result, to plan and found villages of free blacks. In view of the importance of marronage in the development of the colonial history of the isthmus, this thesis aims to analyse the phenomenon during the sixteenth century through a study of the group known as the negros de Portobelo from an Atlantic perspective, and to present a preliminary study of the little known history of Santiago del Príncipe, which could be considered the first village of free blacks in America.

Agaricus blazei Murill, almond portobello, known as Himematsutake in Japan, was found in Brazil. In vitro and in vivo A. blazei had shown immunomodulatory and antimutagenic properties. It was the subject of scientific research which had shown its important nutritional and medicinal properties. Therefore, the research and development of cultivation techniques of A. blazei was necessarily for an industry value. Mushroom culture involves several different operations, each of which must be carefully performed. This study will select the suitable culture media and spawn materials and improve techniques for spawn preparation, and experiment the suitable amount of spawn inoculum, amount of compost substrate and inoculation methods for mycelia growth in compost and mushroom yield of A. blazei. Yeast malt agar medium and potato dextrose agar medium were suitable for mycelia growth of A. blazei. An experiment was conducted to evaluate the efficacy of five different types of spawn materials, viz., wheat, unhusked rice, jasmine rice, brown rice, and brown long rice, and get the results that the best spawn materials for mycelia growth was jasmine rice, and brown rice was second. In nutrient supplementation on jasmine rice experiments, check, wheat bran, calcium carbonate, and corn starch were better for mycelia growth then rice bran and soybean flour. The spawn preparation of A. blazei was recommended that 450 g spawn material inoculated with 6 g master brown millet spawn basing on experimental results. In experiment of the mycelial growth and mushroom production of A. blazei on rice straw compost in culture box (58.8×41.8×33cm), the results showed that mycelial growth of A. blazei were not significantly different on the different inoculation amount of sawdust spawn, the biological efficiency were not significantly different on inoculation 150g, 200g, and 250g. In experiment of the mycelial growth and mushroom production of A. blazei on pangolagrass compost in culture box(58.8×41.8×33cm), the results showed that mycelial growth of A. blazei were not significantly different on inoculation 100g, 200g, and 250g;the biological efficiency were not significantly different on inoculation 100g, 150g, 200g, and 250g. Effect of different weight of rice straw compost on the mycelial growth of A. blazei in culture box, the results showed that mycelial growth of A. blazei were not significantly different;the biological efficiency were not significantly different between 10kg and 12kg of rice straw compost. Effect of different weight of pangolagrass compost on the mycelial growth of A. blazei in culture box, the results showed that mycelial growth of A. blazei were not significantly different;the biological efficiency were not significantly different on 8kg, 10kg, and 12kg of pangolagrass compost, but the 8kg of pangolagrass compost was the best. Effect of different inoculation method of rice straw compost on the yields of A. blazei, the results showed that inoculation on surface layer and middle layer, surface layer and mixing in compost were better than inoculation only on surface layer;the biological efficiency was 17.4% (surface layer and middle) and 17.8% (surface layer and mixing), respectively. Effect of different inoculum materials of A. blazei inoculated on rice straw compost, the results showed that the yields, biological efficiency, and day required after casing to harvesting were not significantly different among wheat spawn, brown rice spawn, and jasmine rice;but the brown rice spawn can get the best weight of mushroom. Effect of casing soil on the yields of A. blazei cultured on rice straw compost, the results showed that the yields, biological efficiency, and weight of mushroom were not significantly different between new peat moss and recycle peat moss. Effect of myceial cutting method on the yields of A. blazei cultured on rice straw compost, the results showed that the yields, biological efficiency, weight of mushroom, and day required after casing to harvesting were not significantly different between two cutting methods. Effect of casing soil with different myceial cutting method on the yields of A. blazei cultured on rice straw compost, the results showed that the yields and biological efficiency, was not significantly different on new peat moss + cutting, new peat moss + scraping and recycle peat moss + scraping.

Marques, Pimentel Sarah-Leah. "Two encounters with the cultural other: Paulo Coelho's A Bruxa de Portobello and its English translation." Thesis, 2009. http://hdl.handle.net/10539/7336.

Full text
Abstract:
ABSTRACT This study recognises the need for greater cultural interaction in a globalised world in which cultural identities are no longer contained within national borders. The aim of this dissertation is, therefore, to address concerns arising from the role played by literary translation as a cultural mediator within contemporary societies. The theoretical framework is drawn from the concept of an “in-between space” in the work of the post-colonial theorist, Homi Bhabha (1990, 2005), and Emmanuel Levinas‟s (1991, 1993) understanding of the relationship between the self (the Same) and an external other (the Other). The relevance of these theoretical concepts to the field of translation studies is explored and these are then practically applied to A Bruxa de Portobello (2006), a novel written by a contemporary Brazilian author, Paulo Coelho, and its English translation (2007). The analysis of the two novels considers the ways in which otherness is presented in both texts, from which conclusions are drawn showing that literary translation can be used as an instrument to promote greater cultural understanding.

DuBard, Bryana. "The Key to All the Indies: Defense of the Isthmus of Panama." Thesis, 2013. http://hdl.handle.net/1969.1/149481.

Full text
Abstract:
Beginning in the 16th century, the Isthmus of Panama was identified as a region of strategic importance. Although mountainous and prone to adverse weather, it provided the most direct route between the Atlantic and Pacific oceans. During this period the Isthmus served as the primary route for the shipments of silver and gold extracted from the mines of Peru. The bullion was transported via coastal armadas from Peru to Panamá la Vieja, where it was loaded onto pack mules and hauled across the Isthmus to Nombre de Dios until 1597, and after that to Portobelo. Once the bullion arrived at the Caribbean port cities it was transferred to the ships of the Armada de la Guardia de la Carrera de Indias and shipped across the Atlantic to the royal coffers of Spain. Because of the Isthmus’s role in the transportation of valuable commodities, it quickly became a region prone to attacks by pirates and privateers looking to profit from the plunder of Spanish assets. Thus the Spanish crown began a campaign to defend the Isthmus early on and repeatedly adapted its defensive strategy in order to meet the ever-changing tactics of the pirates and privateers. This thesis investigates the history of the Isthmus of Panama and the ways in which Spain defended this strategically significant locale during the 16th century. It incorporates an historical analysis of the tactics planned, ordered, and executed by the crown; an overview of the most relevant structural remains of the fortifications built during this period; and a synopsis of previous archaeological investigations, as well as the prospects of future archaeological research. The thesis begins with a brief history of the three main cities located on the Isthmus as well as a general description of the geography and climate in order to better explain the challenges faced by the inhabitants, soldiers, and attackers in this region. It then discusses the strategic importance of the Isthmus as it was perceived in the 16th century. Since defense would not have been necessary had it not been for the presence of pirates and privateers, the history of attacks on the Isthmus is discussed, and a general overview of piracy in the Spanish Main during the period under analysis is presented. Lastly, the archaeological work previously undertaken in the region is examined and summarized, and recommendations for further research are provided in an effort to provide a basis for future study of the ships and infrastructure used during this era for the defense of the Isthmus of Panama.

Abstract:
After much delay, Bernardo Macaya finally arrived in Cartagena de Indias in late October 1675. The chief constable of the city of Portobelo in Panama had hastily arranged for Bernardo’s departure to Cartagena shortly after taking office some weeks before. The magistrate had heard worrisome notices about Bernardo during his time in Portobelo and had discovered equally disturbing accounts about him in paperwork left by the previous constable. Bernardo, a thirty-four-year-old West Central African slave of “Congo caste,” was a feared and renowned ritual practitioner operating around Portobelo. The chief constable was worried that he would not be able to keep Bernardo in prison or, worse, that he would end up like his immediate predecessor in office: dead. His fears were eventually realized by the events surrounding Bernardo’s departure from Portobelo....

The walls of Cartagena de Indias through methodological experimentation and survey systems protocols. Digital tools comparison for the study of the Antonelli’s defense systemCartagena de Indias, one of the main Spanish commercial ports in the Caribbean Sea, was strategically built on a system of islands and peninsulas that formed a lacustrine system along the coast of Tierra Firme, known today as Colombia. For several centuries, Cartagena fortifications have been at the fore-front of Spanish military technologies. This site became the scene of action of the main military engineers at the service of the Spanish crown. In 1586 Battista Antonelli received from King Philipe II the task to design this monumental defensive system. The first project for the Cartagena wall enclosure (1595) is due to Battista and it was continued and modified by his nephew Cristoforo Roda. Nowadays, Antonelli walls still fit into the urban fabric of the city and delineate the perimeter of the historic city. The research project follows the previous research experiments conducted by the Lab DAda-LAB of the University of Pavia in the territory of Panama for the study of the Antonelli fortifications systems of Portobello and San Lorenzo del Chagres. It concerned an extensive action aimed at the documentation and to the study of the entire fortified system of the historic center of Cartagena. The perimeter walls of the old city and the fort of San Felipe de Barajas have been documented through the use of a mobile laser scanner that uses SLAM technology, evaluating the most effective performed strategies for fast survey activities. In parallel, a more specific action was conducted on the portion of the Baluarte of Santa Catalina walls, where it was possible to give a comparison between different methods and instruments, in order to verify the reliability of the 3D databases. Analysis protocols have been developed for the documentation and study of the defensive system. The paper will highlight the construction technologies that qualify the fortresses of Cartagena de Indias and the results obtained by the comparison between different data acquisition technologies to evaluate the quality of the models for the development of documentation strategies for heritage enhancement and protection.
