ENGLISH ABSTRACT: Red colour development on bi-coloured apples and pears ensures better prices for producers. The use of postharvest irradiation to improve colour has been successful on apples, and the objectives of this thesis were to optimise conditions during irradiation for apples and to evaluate the effects of irradiation on pears. 'Cripp's Pink' apples responded to postharvest irradiation with high-pressure sodium (HPS) lights by developing a red blush, whereas the response to irradiation with UV 8 plus incandescent lights was less effective. '8raeburn' apples held at -0.5°C for 4 or 8 weeks prior to irradiation showed a decrease in hue angle and an increase in anthocyanin concentration after 72 hours of irradiation with HPS lamps. In 'Forelle' pears treated in the same way, neither colour development nor anthocyanin synthesis was affected by irradiation. '8raeburn' and 'Cripp's Pink' apples picked weekly for 5 weeks until the optimum harvest date were irradiated with HPS lights. A significant increase in fruit colour was only measured in mature fruit of both cultivars. 'Cripp's Pink' apples were harvested from two production areas with different microclimates, namely, Ceres and Grabouw, and stored for 0, 2 or 5 days at -0.5 °C before irradiation for 120 hours at either 6°C or 20°C. Fruit from Ceres that were irradiated immediately after harvest developed better colour at 6 °C than at 20°C. The differences between fruit irradiated at the two temperatures were no longer significant after 5 days of cold storage prior to irradiation. Fruit from Grabouw consistently developed better colour when irradiated at 6°C than at 20°C. Colour development slightly after 5 days of cold storage prior to irradiation. In another experiment, fruit from both areas were stored at -0.5°C for 20 days before irradiation at either 6°C or 6/20°C. The fluctuating temperature regime resulted in decreases in hue angle of 70° and 65° for the fruit from Grabouw and Ceres, respectively. The decreases were smaller (±200) when fruit were irradiated at 6°C. The hue angle value of well-coloured 'Cripp's Pink' apples held at 3rC under HPS lights for 144 hours increased from 29.3° to 48.3°, and anthocyanin concentration decreased from 739.9 IJg·g·1to 283.6 IJg·g·1. Control fruit held at the same temperature in the dark did not show any change in hue angle value or anthocyanin concentration. 'Bon Rouge' and 'Red d' Anjou', two full red pear cultivars, irradiated with HPS lights for 72 hours, showed no significant changes in hue angle. 'Forelle' pears, harvested with or without attached leaves, were irradiated with HPS at two temperature regimes, 20°C and 200/6°C. The resulting decreases in hue angle were attributed to yellowing and not red colour formation. In conclusion, the response of apples to postharvest irradiation was affected by maturity and temperature, while pears failed to respond at all.

Abstract:
The sensing needs for the fresh produce industry can be split into two primary stages: during maturation in the field, also referred to as Precision Farming, and during storage and transport of the produce, or Postharvest Storage. This work seeks to improve the accuracy and reliability of commercially available electrochemical and spectroscopic sensors tailored to the sensing needs of the fresh produce industry. For electrochemical sensing, this study proposes the use of an inline filter to remove polar organic compounds, which can interfere with the readings of a platinum-based electrochemical sensor. A 50% improvement in measurement accuracy was achieved when monitoring the storage headspace of a container of apples. For portable spectroscopy instruments, this study suggests improvements for the alignment of the optical bench and the spectral collect protocol. Methods to reduce the influence of environmental noise, such as variability of background light (sunlight in the field) and thermal effects on hardware performance, are presented. This study also presents the first report of the calibration transfer of spectral regressions developed with Karl Norris's Derivative Quotient Method. The motivation for this aspect of research was to develop methods to collect stable and accurate data in the field, which can be used to improve the quality of fresh produce reaching the customer and reduce premature food spoilage.

Abstract:
The purpose of this study was to determine the quality and microbial load of blueberries at different maturity stages, to develop the effective microbial recovery method and to compare sanitation postharvest treatments on microbial counts of blueberries. The soluble solids and the sugar to acid ratio increased, while pH and TA decreased with maturity. Ripe berries had lower yeast and mold counts (YMC) at other maturity stages, but there were no differences on aerobic plate counts (APC). The medium pH was lower for stomaching and blending than hand massaged samples. This leads to higher recovery of microorganisms by massaging. Sodium hypochlorite at 400 ppm was effective in reducing APC but not YMC. Acidified sodium chloride was very effective, lowering APC and YMC below detectable level. All sanitation treatment did not influence sensory attributes of blueberries.

ENGLISH ABSTRACT: By world standards South Africa’s contribution to fresh blueberry production is very small, a mere 0.25 %. This seemingly insignificant and unimpressive statistic is, however, by no means indicative of the impending impressive potential the South African blueberry industry holds. South Africa is encountering a rapid growth phase off a very small base and the current 100-150 ton export crop is expected to increase to a range of 500 to 600 tons or more in the next five years. Consequently, export by sea must be considered as a logistic alternative. Seafreight success relies on controlled atmosphere storage which in turn relies on berries of exceptional quality. To achieve this, fundamental differences between blueberry cultivars must be examined and blueberry handling and storage regimes must be assessed and optimised. This study investigates the storage potential of certain rabbiteye blueberries relative to controlled atmospheres and short pre-cooling delays. Quality assessments were based on external colour, soluble solids, titratible acidity, firmness and decay development. Furthermore, the respiration and ethylene production rates as well as moisture loss, of various cultivars were determined to characterize fundamental differences amongst cultivars and to gain insight into their postharvest behaviour. Lastly the effect of infection on pigment levels of blueberry flesh and peel was examined for the purpose of laboratory experience. Controlled atmosphere storage had the greatest impact on berry firmness and percentage decay. No off flavours were detected during sensory analysis at the CA levels tested. CAstored berries were firmer and incidence of decay was reduced significantly. Significant decay reduction was challenged by the market tolerance of no more than 2 %. Decay suppression and storage extension was insufficient (for the set period), particularly after the shelf life period. Cultivar and seasonal differences as well as picking maturity and inoculum pressure determine blueberry CA storability. CA may be a valuable technology, however advances must first be made in ensuring optimal blueberry quality to warrant export by sea. Short pre-cooling delays (0, 2, 4, 6 h) at 20 ºC had no effect on colour (L, C, H), total soluble solids, titratable acidity and firmness of blueberry cultivars, ‘Centurion’, ‘Premier’ and ‘Tifblue’. Results for percentage decay indicate that prompt cooling becomes more critical as the duration of storage increases, particularly when market specification of no more than 2 % decay must be met. Berries subjected to longer delays appear to be more susceptible to decay. Differences exist among cultivars and knowledge gained from this research can be used to adapt handling and cooling regimes. Information generated will benefit cultivar selection for marketing (local vs. export), storage (short vs. long term) and logistics (air vs. seafreight). Rabbiteye blueberry cultivars differ considerably in terms of their respiration rate, ethylene production and moisture loss. These quality parameters were evaluated for various cultivars at 15 ºC. ‘Climax’ appears to have the highest respiration rate, ethylene production and moisture loss, whilst ‘Premier’ has the lowest respiration rate and ethylene production was too low to be detected by gas chromatography. The lowest moisture loss proportionality constant was found for ‘Tifblue’ and ‘Centurion’. A change in blueberry flesh colour from white to dark red was noted in infected berries. Artificial inoculation with Botrytis cinerea showed a similar result. The colour change was evaluated by comparing anthocyanin pigment levels (determined by spectrophotometry and HPLC) of the peel and flesh. Anthocyanin content of infected flesh doubled, whilst that of infected peel was less than 50 % of the control. Pigment leakage has been reported in blueberries. It would appear that cell disruption, induced by infection may facilitate migration of anthocyanin from the peel to the flesh. The storage potential of blueberries depends on cultivar, maturity at harvest, climatic conditions as well as the inoculum pressure. Under favourable conditions storage life of blueberries can be manipulated effectively. Further cultivar specific research that targets relevant quality issues should be investigated. Reliable techniques should be developed to quantify blueberry firmness changes. Research on inoculum levels may give insight into the reduction of decay pressure and decay suppressing alternatives such as antibacterial pads can be examined. Peel integrity relative to decay susceptibility can also be researched on a cellular level.
AFRIKAANSE OPSOMMING: Suid-Afrika se bydrae tot wêreld bloubessie produksie is baie klein, ‘n skrale 0.25 %. Hierdie onindrukwekkende statistiek weerspieel egter glad nie die indrukwekkende potensiaal van die Suid-Afrikaanse bloubessie bedryf nie. Die bedryf ondervind tans ‘n dramatiese groei met ‘n klein begin vanaf die huidige 100-150 ton opbrengs tot wat volgens verwagting binne die volgende vyf jaar sal styg tot in die orde van 500-600 ton. Na aanleiding van die verwagte toename in produksie, sal seevrag verskeping oorweeg moet word as alternatief vir uitvoer eerder as lugvrag. Die sukses van seevrag lê in die gebruik van beheerde atmosfeer opberging (BA), waarvan die sukses op sy beurt bepaal word deur bessies van hoogstaande kwaliteit. Om steeds aan die verwagtings van die oorsese mark te voldoen moet grondliggende verskille tussen bloubessie kultivars ondersoek word en daaropvolgend moet hantering en opbergingstelsels aangepas en geoptimeer word. Die navorsing bestudeer die opbergingspotensiaal van verskeie rabbiteye bloubessies relatief tot beheerde atmosfere en kort vertragings in verkoeling. Kwaliteit, gebasseer op eksterne kleur, oplosbare deeltjies, titreerbare sure, fermheid en persentasie bederf, is ondersoek. Die respirasie en etileenproduksie tempos sowel as vogverlies van verskeie kultivars was verder bepaal om die fundamentele verskille tussen kultivars te bepaal, sowel as insig te verwerf in na-oes gedrag. Die effek van infeksie op pigment vlakke van bloubessie vlees en skil was ondersoek vir die doel van laboratorium ondervinding. Beheerde atmosfeer opberging het die grootste effek getoon op bessie fermheid en persentasie bederf. Na aanleiding van sensoriese toetse is daar geen wansmaake aangeteken ten op sigte van die BA vlakke wat getoets is nie. Bessies onderworpe aan beheerde atmosfeeriese toestande was fermer en bederfonderdrukking was beduidend. Die markverdraagsaamheid ten opsigte van bessiebederf, is egter 2 %. Hiervolgens was onderdrukking van verval asook opbergingsvermoë, onvoldoende (spesifiek na die rakleeftyd). Kultivar en seisoenale verskille asook plukrypheid en inokulum druk, bepaal ook die sukses van BA opberging. BA word beskou as ‘n waardevolle tegnologie vir verlenging van opberging. Dit blyk egter dat daar eers ‘n vooruitgang gemaak moet word in die versekering van optimale bessie kwaliteit voordat verskeping oorweeg kan word. Kort vertragings (0, 2, 4, 6 uur by 20 ºC) in verkoeling, het geen invloed gehad op kleur, (L, C, H), totale oplosbare vastestowwe, titreerbare suur en fermheid van bloubessie kultivars ‘Centurion’, ‘Premier’ en ‘Tifblue’ nie. Bederfresultate dui daarop dat onmiddelike verkoeling krities is, veral by verlengde opberging, om aan die 2 % bederf drempelwaarde te voldoen. Bessies onderhewig aan langer vertragings voor verkoeling, blyk meer vatbaar te wees vir bederf. Verskille bestaan tussen kultivars en kennis ingewin tydens hierdie navorsing kan toegepas word om hanteringsprosedures en verkoeling aan te pas. Ingeligte besluitneming kan dus ten opsigte van kultivarspesifieke bemarking (plaaslik en uitvoer), opberging (kort- en langtermyn) en vervoer (lug- en seevrag) gemaak word. Rabbiteye bloubessie kultivars verskil aansienlik ten opsigte van hul respirasietempo, etileenproduksie en vogverlies. Laasgenoemde kwaliteitsbepalers is by verskillende kultivars ondersoek by 15 ºC. ‘Climax’ het die hoogste respirasietempo, etileenproduksie en vogverlies getoon, terwyl ‘Premier’ die laagste respirasietempo gehad het, en die etileenproduksie te laag was om met die gaschromatograaf te bepaal. Die laagste vogverlies proporsionaliteitskonstante is waargeneem tussen ‘Tifblue’ en ‘Centurion’. ‘n Verandering is waargeneem in bloubessie vrugvleeskleur van wit tot donkerrooi by besmette bessies. ‘n Soortgelyke resultaat is verkry met kunsmatige inokulering met Botrytis cinerea. Die kleurverskil is geëvalueer deur vergelyking te tref met die antosianienpigment vlakke tussen die skil en die vlees (bepaal deur spektrofotometrie en HPLC). Antosianieninhoud van besmette vrugvlees het verdubbel, terwyl die van besmette vrugskil minder as 50 % van die kontrole groep was. Pigmentlekkasie is al vermeld by bloubessies. Dit wil voorkom of selskade veroorsaak deur infeksie beweging van antosianien vanaf die skil na die vlees kan aanhelp. Die opbergingspotensiaal van bloubessies hang af van kultivar, rypheid by oes, omgewings toestande sowel as inokulum druk. Onder gunstige toestande kan bloubessies se opbergingsvermoë effektief gemanipuleer word. Verder kultivar-spesifieke navorsing wat relevante kwaliteits kwessies aanspreek, kan gedoen word. Verder kan betroubare tegnieke in die meting van bloubessie fermheid veranderinge ondersoek word. Navorsing ten opsigte van inokulum vlakke mag insig gee in terme van bederf-druk vermindering en die gebruik van bederf-onderdrukking alternatiewe soos anti-bakteriese lappies. Skil integriteit relatief tot bederf-vatbaarheid kan ook op sellulere vlak ondersoek word.

ENGLISH ABSTRACT: The aim of this study was to develop non-destructive methods to predict external and internal quality of citrus fruit. A critical review of the literature identified presymptomatic biochemical markers associated with non-chilling rind physiological disorders. The prospects for the use of visible to near infrared spectroscopy (Vis/NIRS) as non-destructive technology to sort affected fruit were also reviewed. Initial studies were conducted to determine the optimum condition for NIRS measurements and to evaluate the accuracy of this technique and associated chemometric analysis. It was found that the emission head spectroscopy in diffuse reflectance mode could predict fruit mass, colour index, total soluble solids, and vitamin C with high accuracy. Vis/NIRS was used to predict postharvest rind physico-chemical properties related to rind quality and susceptibility of ‘Nules Clementine’ to RBD. Partial least squares (PLS) statistics demonstrated that rind colour index, dry matter (DM) content, total carbohydrates, and water loss were predicted accurately. Chemometric analysis showed that optimal PLS model performances for DM, sucrose, glucose, and fructose were obtained using models based on multiple scatter correction (MSC) spectral pre-processing. The critical step in evaluating the feasibility of Vis/NIRS was to test the robustness of the calibration models across orchards from four growing regions in South Africa over two seasons. Studies on the effects of microclimatic conditions predisposing fruit to RBD showed that fruit inside the canopy, especially artificially bagged fruit, had lower DM, higher mass loss, and were more susceptible to RBD. The study suggested that variations in microclimatic conditions between seasons, as well as within the tree canopy, affect the biochemical profile of the rind, which in turn influences fruit response to postharvest stresses associated with senescence and susceptibility to RBD. Principal component analysis (PCA) and PLS discriminant analysis (PLS-DA) models were applied to distinguish between fruit from respectively, inside and outside tree canopy, using Vis/NIRS signal, suggesting the possibility of using this technology to discriminate between fruit based on their susceptibility to RBD. Results from the application of optical coherence tomography (OCT), a novel non-destructive technology for imaging histological changes in biological tissues, showed promise as a potential technique for immediate, real-time acquisition of images of rind anatomical features of citrus fruit. The study also demonstrated the potential of Vis/NIRS as a non-destructive tool for sorting citrus fruit based on external and internal quality.
AFRIKAANSE OPSOMMING: Die studie het ten doel gestaan om nie-destruktiewe meeting metodes te toets en ontwikkel wat die interne en eksterne-kwaliteit van sitrusvrugte kan voorspel. In ʼn litratuuroorsig is biochemies verandering in die skil en wat geassosieer word met die ontwikkeling van fisiologies skildefekte geïdentifiseer, asook is die moontlikheid ondersoek om Naby Infrarooi spektroskopie (NIRS) as ‘n nie-destruktiewe tegnologie te gebruik om vrugte te sorteer. Eerstens was die optimale toestande waarby NIRS meetings van sitrusvrugte geneem moet word asook die akkuraatheid van die toerusting en chemometrika data-ontleding beproef. Daar is gevind dat die uitstralings-kop spektrofotometer in diffusie-weerkaatsings modus vrugmassa, skilkleur, totale opgeloste stowwe asook vitamien C akkuraat kan voorspel. Daarna van NIRS gebruik om na-oes fisies-chemiese eienskappe wat verband hou met skilkwaliteit en vatbaarheid vir skilafbraak van ‘Nules Clementine’ mandaryn. Deur gebruik te maak van “Partial least squares” (PLS) statistieke was gedemonstreer dat die skilkleur, droë massa (DM), totale koolhidrate en waterverlies akkuraat voorspel kon word. Chemometriese analises het ook getoon dat optimale PLS modelle vir DM, sukrose, glukose en fruktose verkry kan word deur modelle te skep wat gebaseer is op “Multiple scatter correction” (MSC) spektrale voor-verwerking. ʼn Belangrike stap in die ontwikkeling van NIRS gebaseerde indeling is om die robuustheid van die kalibrasiemodelle te toets en was gedoen deur vrugte te meet en sorteer van vier boorde en oor twee seisoene. ʼn Verder eksperiment om die impak van mikroklimaat op die skil se vatbaarheid vir fisiologiese defekte te ontwikkel het getoon dat vrugte wat binne in die blaardak ontwikkel (lae vlakke van sonlig) ʼn laer DM, hoër gewigsverlies het en was ook meer vatbaar vir skilafbraak. Die resultate dui daarop dat verskille in mikroklimaat oor die seisoen asook in die blaardak die skil se biochemiese profiel beïnvloed, wat lei tot ʼn negatiewe reaksie op na-oes stres en verhoogde voorkoms van fisiologiese skilafbraak. Die ontwikkelde “Principal component analysis” (PCA) en PLS-diskriminant analise modelle was daarna suksesvol toegepas om vrugte te skei na NIRS meetings, op die basis van vrugpossies in die blaardak. Nuwe, nie-destruktiewe tegniek, nl. “Optical coherence tomography” (OCT) was suksesvol getoets as manier om ʼn fotografiese beeld te skep van histologiese veranderinge in die skil. Die resultate dui op die potensiaal van die onontginde tegnologie om intak biologiese-materiaal te analiseer. ENGLISH ABSTRACT: Grapes are lost annually due to spoilage by Botrytis cinerea. Botrytis cinerea is currently controlled using fungicides and SO2 fumigation in storage rooms but with limited success. Although these techniques have been used for decades they have been associated with numerous of disadvantages. Fungicides are restricted from being used during storage due to legislation and toxicity of the fungicides present on the surface of the berries. SO2 fumigation does not kill the dormant infections present inside the grape tissue and if the SO2 concentration is too high the berries are damaged. During this study two different polymer nanofibrous platforms was synthesized in an attempt to prevent the rotting effect of B. cinerea. The first polymer was modified to yield a polymer with a positively charged quaternized nitrogen moiety, which was subsequently reacted with sodium metabisulfite through an ion exchange process. The modified polymer was electrospun into nanofibrous mats for the benefit of the nanofibers’ high available surface area. A further functionalization was done to increase the quantity of sodium metabisulfite on the surface of the polymer nanofibers. Sodium metabisulfite salt released SO2 gas upon reaction with water vapour present in the atmosphere which resulted in the inhibition of conidial germination of B. cinerea. The second polymer was synthesized and then electrospun into polymer nanofibrous mats followed by chemical modification of the electrospun polymer nanofibers. This post-electrospun modification resulted in the covalent attachment of a fungicide-derivative to the surface of the polymer nanofibers. The fungicide-derivative showed excellent inhibition of B. cinerea mycelium growth. Anti-fungal studies were conducted using the two modified polymer nanofibrous mats against B. cinerea to evaluate these nanofibrous surfaces as B. cinerea inhibiting membranes. Results indicated that B. cinerea conidial germination and mycelium growth were successfully inhibited.
AFRIKAANSE OPSOMMING: Druiwe bederf jaarliks weens die verrottende effek van die fungi Botrytis cinerea. Botrytis cinerea word tans beheer deur gebruik te maak van swamdoders en swaweldioksied (SO2) besproeiings in stoorkamers, maar met beperkte sukses. Alhoewel hierdie tegnieke al vir dekades in gebruik is, word dit geassosieer met verskeie probleme. Die gebruik van swamdoders word in stoorkamers verbied, weens die moontlike skadelike inname daarvan deur die mens aangesien die swamdoders op die druiwe se oppervlakte teenwoordig is. SO2-gasbesproeiings maak nie dormante infeksies binne-in die druiwe self dood nie en indien SO2 -konsentrasies te hoog is kan dit die druiwe beskadig. Tydens hierdie studie is twee verskillende polimeer-nanoveselplatforms gesintetiseer met die doel om die verrottende effek van B. cinerea te voorkom. Die eerste polimeer is aangepas met ’n primêre amienverbinding met die doel dat dit deur ‘n opvolgende modifikasiereaksie verander om sodoende ’n polimeer met ‘n kwatenêre ammoniumgroep te verkry. Die doel van die kwatenêre eienskap is dat ioniese uitruiling plaasvind tussen die anioon van die polimeer en die natruimmetabisulfiet-anioon van die sout. Die voorbereide kopolimeer is geëlektrospin in nanoveselmatte deur middel van die enkelnaald-en-enkelbal-elektrospintegniek om SMI-qC12 nanovesels te lewer wat gefunksionaliseer is. Die nanoveselmatte is na die elektrospinproses verder aangepas om die hoeveelheid natruimmetabisulfiet op die oppervlak van die nanovesels te verhoog. Die natruimmetabisulfiet stel SO2-gas vry sodra dit in aanraking kom met waterdamp in die lug wat ’n beperkende effek op spoorontkieming van B. cinerea tot gevolg het. Die tweede polimeer is voor en na die elektrospinproses gefunksionaliseer met gesintetiseerde organiese verbindings (swamdoder). Die aanpassing van die polimeer met die organiese verbindings het plaasgevind op ‘n kovalente wyse om te verhoed dat die organiese verbinding vrygelaat word tydens gebruik. Die organiese verbindings het effektiewe beperking van miselium-groei getoon. Anti-swamstudies is uitgevoer tussen die twee gefunksionaliseerde polimeer platforms en B. cinerea om die oppervlaktes van die gefunksionaliseerde polimeer/nanovesels te evalueer as B. cinerea beperkende platforms. Resultate het aangetoon dat spoorontkieming en miselium groei van B. cinerea suksesvol beperk is.

Advanced research in Controlled/Modified Atmosphere storage systems has shown that open channels of different lengths and cross sectional areas, connected to an air tight storage chamber, are able to maintain variable stable gas concentrations which could be near optimal concentrations for CA/MA storage of various commodities.
This study was geared towards assessing the suitability of the diffusion channels in maintaining a desired gas concentration for CA storage of spinach. Initially, the respiratory behaviour of spinach was studied in gas sealed chambers (replicated four times) stored at four different temperatures, 2$ sp circ$C, 8$ sp circ$C, 15$ sp circ$C and 23$ sp circ$C. The respiration rate of spinach was 20 mgCO$ sb2$/kg.h, 66 mgCO$ sb2$/kg.h, 163 mgCO$ sb2$/kg.h and 271 mgCO$ sb2$/kg.h for 2$ sp circ$C, 8$ sp circ$C, 15$ sp circ$C and 23$ sp circ$C, respectively. A model was developed based on principles of enzymatic kinetics which could reliably predict the respiration rate of spinach at any given storage temperature.

ENGLISH ABSTRACT: The development of science-based management tools and appropriate postharvest handling protocols are required for the determination of optimal storage performance of pomegranate fruit. The South African pomegranate industry experiences considerable fruit quality losses due to the lack of knowledge on optimal storage and handling practices. The cultivar ‘Wonderful’ is the widely grown in South Africa; however, to date there is currently limited scientific knowledge on the storage requirements. To develop quality standards for the export market, knowledge of optimum storage conditions are required to provide an understanding of postharvest quality attributes and consumer organoleptic perceptions. The overall aim of this research was to provide science-based management tools for the storage performance of pomegranate fruit (cv. Wonderful). The research reported in Chapter 3 focused on the physiological responses of pomegranate fruit at different storage temperatures. Commercially harvested fruit were stored at 5±0.7°C, 7.5±0.3°C and 10±0.5°C with 92±2% RH and at room temperature (21±3°C, 65±6% RH) for 5 months. Fruit respiration and physiological disorders during long term storage were investigated. During storage, low temperatures evidently resulted in lower respiration rates; however, respiration rate increased gradually after 2 months resulting in higher respiration rates at 5°C than 7.5°C after 3 month storage period. Overall, fruit became more susceptible to internal and external disorders as storage period progressed. Storage of fruit longer than 2 months at 5°C resulted in chilling injury and this was observed over the 5 month storage period. Fruit stored at 21°C and 10°C were discarded after 1 and 4 months, respectively, due to complete fruit loss to decay and peel shrinkage. Furthermore, the severity of browning increased with storage temperatures, although this became more severe at 5°C after 3 months. Therefore, to maintain a relatively low respiration rate and minimize physiological disorders, the cv. Wonderful should be stored at 5°C and >92% RH for storage period up to 3 months. In Chapter 4, the effects of temperature and storage duration on pomegranate fruit quality and mechanical properties were conducted. This study revealed that weight loss increased with rise in temperature and storage duration with the primary source of moisture loss being the fruit skin (peel), which resulted in significant reduction in peel thickness with prolonged storage period. The CIE (L*, a*, b* and C*) colour parameters of fruit and arils decreased during storage. However, the hue (hº) for whole fruit increased as a result of browning incidence, and decreased in arils suggesting an increase in redness. Significant increases in total soluble solids (TSS), pH, TSS:TA and BrimA were observed with significant decreases in titratable acidity (TA) occurring throughout the storage period. Storage temperature and duration significantly affected majority of the investigated mechanical properties. Puncture resistance, fruit and aril compression strength decreased with storage temperature and duration. These findings showed that fruit may be stored between 2 to 3 months at 5°C to ensure the best internal and external quality attributes. The studies in Chapter 5 investigated the effects of storage temperature and duration on phytochemical and antioxidant properties. Fresh pomegranate juice was assessed for concentrations of total phenolic compounds, total anthocyanin and ascorbic acid. The antioxidant property of the fruit juice was tested against 2, 2-diphenyl–1–picryl hydrazyl (DPPH). The results showed that total phenolic and total anthocyanin concentration increased up to 3 months of storage at 5°C, 7.5°C, 10°C and 21°C and decreased gradually over time. For antioxidant activity, storage of fruit at 5°C, 7.5°C and 10°C significantly (p< 0.05) reduced the radical scavenging activity of juice by more than 56% when stored beyond 2 months. Furthermore, ascorbic acid concentration gradually declined with increasing storage duration, resulting in reduced juice antioxidant capacity. These findings are beneficial to pomegranate export industries, especially where fruit are stored for long for use in health-promoting purposes. The research conducted in Chapter 6 focused on determining suitable storage conditions based on the combination of instrumental measurements and sensory attributes. During storage, individual fruit were evaluated by trained sensory panel based on the overall appearance, taste and aril texture. Discriminant analysis at different storage temperatures was used to distinguish fruit from each other at 2 months of storage with sensory attributes such as overall pomegranate flavour (R2 = 0.56), total anthocyanin (R2 = 0.46) and Chroma (C*) colour index (R2 = 0.37). Discriminant analysis further showed that storage time rather than storage temperature led to the reduction in overall quality when storing fruit beyond 2 months. Based on sensory attributes, suitable storage temperature and duration were found to be 5°C and 2 months when overall flavor were highly rated; thereafter, significant reductions in overall appearance, aril and kernel texture were observed. Furthermore, the proposed storage conditions were supported with instrumental measurements, which revealed a decline in important fruit attributes such as total phenolics, total anthocyanin, aril colour and aril texture after 2 months of storage. Overall, this study provides science-based tools required for developing cold chain handling protocols needed to manage the long supply chain of ‘Wonderful’ pomegranate fruit grown in South Africa.
AFRIKAANSE OPSOMMING: Die ontwikkeling van wetenskap-baseerde beheerinstrumente en toepaslike na-oes hanteringsmetodes is nodig vir die vasstelling van die optimale stoorprestasie van granate. Die Suid-Afrikaanse granaatindustrie ondervind groot vrug kwaliteit verliese as gevolg van die gebrek aan kennis oor optimale stoor en hantering praktyke. Die kultivar Wonderful is die wyd gegroei in Suid-Afrika, maar tot hede daar is tans beperk wetenskaplike kennis oor die stoor vereistes. Om gehaltestandaarde vir die uitvoermark te ontwikkel word kennis van die optimale stoortoestande benodig sodat ’n begrip van die na-oes gehalte-kenmerke en verbruiker se organoleptiese persepsies gevorm kan word. Die oorhoofse doelwit van die navorsing is om wetenskap-baseerde beheerinstrumente vir die stoor van granate (bv. Wonderful) te verskaf. Die navorsing wat in Hoofstuk 3 beskryf word is gerig op die fisiologiese respons van granate op verskillende bergingtemperatuur. Kommersieel-gekweekte vrugte is by 5±0.7°C, 7.5±0.3°C en 10±0.5°C met 92±2% RH en by kamertemperatuur by (21±3°C, 65±6% RH) vir 5 maande gestoor. Die respirasie van die vrugte en die fisiologiese ongesteldhede gedurende langtermyn stoor word ondersoek. Gedurende stoor het die laer temperature gelei tot laer respirasie koerse; maar respirasie koers het geleidelik na 2 maande verhoog wat lei tot hoër respirasie koerse by 5°C as teen 7.5°C na ’n 3-maande stoorperiode. Algehele, vrugte het egter meer vatbaar geword vir interne en eksterne ongesteldhede hoe langer die stoortydperk geduur het. Die stoor van vrugte langer as 2 maande teen 5°C lei tot skade as gevolg van verkoeling en dit is oor die 5 maande stoor tydperk waargeneem. Vrugte wat teen 21°C en 10°C gestoor is moes na onderskeidelik 1 tot 4 maande as gevolg van verlies wat die gevolg was van swam skade en skil krimping, weggegooi word. Die erns van die verbruining het verhoog toe die stoortemperature verhoog, alhoewel dit meer geraak het teen 5°C na 3 maande. Om dus ’n betreklik lae respirasie koers en min fisiologiese probleme te verseker, moet die kultivaar Wonderful teen 5°C en >92% RH vir 3 maande gestoor word. In Hoofstuk 4 word die effek van temperatuur en die duur van stoor op die gehalte van die granate en die meganiese eienskappe gemeet. Daar is bevind dat gewigsverlies met verhoogte toename in temperatuur en langer stoorperiodes toeneem en dat die hoofbron van verlies aan vog die skil van die vrug is. Die gevolg hiervan is ’n betekenisvolle reduksie in die dikte van die skil na ’n lang stoorperiode. Die CIE (L*, a*, b* and C*) kleur parameters van vrugte en granaatpitte het tydens stoor verminder. Die tint, (hº) van die hele vrug het as gevolg van verbruining, verhoog en het verminder in granaatpitte wat daarop dui vermeerdering in rooiheid. Daar was betekenisvolle verhogings in die totale oplosbare vaste stowwe (TSS), pH, TSS:TA en BrimA is opgemerk met betekenisvolle vermindering in asiditeit waarvan die waarde bepaal kan word (TA) en wat tydens die stoortydperk plaasvind. Stoortemperatuur en die duur van die stoor het ’n groot invloed gehad op die meganiese kenmerke wat ondersoek is. Weerstand teen priken die kompressie krag van die vrugte en die granaatpitte het met verhoogde temperatuur en duur van stoor afgeneem. Hierdie bevindinge het getoon dat vrugte kan gestoor word tussen 2 tot 3 maande by 5°C die beste interne en eksterne kwaliteit eienskappe om te verseker. In hoofstuk 5 is die effek van stoortemperatuur en duur op die fitochemiese en antioksidant kenmerke ondersoek. Vars granaatsap is ondersoek en ramings is gemaak t.o.v. totale konsentrasies van fenoliese samestellings, totale antosianiene en askorbinesuur. Die antioksidant kenmerke van die vrugtesap is getoets vir met 2, 2-diphenyl–1–picryl hydrazyl (DPPH). Daar is bevind dat die totale fenoliese en totale antosianiene konsentrasies tot by 3 maandemaande van stoor teen 5°C, 7.5°C, 10°C and 21°C toegeneem het en toe mettertyd afgeneem het. Wat betref antioksidant aktiwiteit, is daar gevind dat die stoor van vrugte teen 5°C, 7.5°C en 10°C die radikale reinigingsaktiviteite van die sap betekenisvol (p< 0.05) met meer as 56% verminder as dit vir meer as 2 maande gestoor word. Verder, askorbiensuur konsentrasie geleidelik afgeneem met toenemende stoor duur, wat lei tot verlaagde sap antioksidant kapasiteit. Hierdie bevindings is van belang vir die granaatuitvoerindustrie, veral waar vrugte vir 'n lang tydperk gestoor vir gebruik in gesondheids-bevordering doeleindes. Die navorsing wat in hoofstuk 6 beskryf is, het gefokus op die vasstelling van geskikte stoortoestande baseer op ’n kombinasie van instrumentale meting en sensoriese kenmerke. Gedurende stoor word individuele vrugte deur ’n opgeleide panel evalueer t.o.v. voorkoms, smaak en tekstuur van die granaatpitte. Diskriminantontleding teen verskillende stoor temperature is gebruik om vrugte na 2 maande stoor vrugte t.o.v sensoriese kenmerke soos algehele granaat smaak. (R2 = 0.56), totale antosianiene (R2 = 0.46) en Chroma (C*) kleur indeks (R2 = 0.37) te onderskei. Diskriminantontleding het verder getoon dat die duur van die stoor en nie die stoortemperatuur nie, gelei het tot die reduksie in algehele gehalte as die vrugte vir langer as 2 maande gestoor word. Gegrond op sensoriese eienskappe is geskik stoor temperatuur en duur gevind word by 5°C en 2 maande wanneer algehele geur was as hoog beoordeel; en daarna, is aansienlike vermindering in die algehele voorkoms, en die tekstuur van die granaatpitte afgeneem. Hierdie voorgestelde stoortoestande word ook ondersteun deur instrumentele meting, wat ’n afname in belangrike kenmerke soos totale fenologie, totale antosianiene en die kleur en tekstuur van die granaatpitte na ’n 2 maande stoorperiode toon. In die geheel verskaf die bevindinge van hierdie studie wetenskap-baseerde instrumente vir die ontwikkel van koue-ketting hantering protokol vir die bestuur van die lang verskaffingsketting van Wonderful granate wat in Suid-Afrika gekweek word.

[EN] In recent years, persimmon crop has become very relevant in Mediterranean Spain, where the production of this fruit centres on only one variety, persimmon cv. Rojo Brillante, majorly located in the Valencian Community. The main postharvest disorders manifested by 'Rojo Brillante' persimmons are flesh browning, which is associated with mechanical damage and chilling injury displayed after low-temperature storage. Previous research has determined the postharvest conditions that lead fruit to develop such alterations. However, the biochemical process behind flesh browning and chilling injury disorders is still unknown. Currently, there is special interest in introducing cultivars from other countries to broaden the varietal range. Besides, prolonging the fruit storage period to supply the markets according to the demand is one of the main challenges. In this context, the present Thesis approached three main objectives: 1) Studying the biochemical process implied in the main physiological postharvest disorders manifested in persimmon fruits by focusing on changes in the fruit redox state; 2) Evaluating postharvest treatments to preserve fruit quality during cold storage; 3) Assessing the physico-chemical and nutritional quality of persimmon cultivars introduced from other countries to increase the varietal range. Biochemical, chromatographic and microstructural studies have revealed that flesh browning manifested by fruits submitted to mechanical damage after removing astringency is associated with a tannins oxidation process caused by a stress oxidative situation. A new flesh disorder, "pinkish bruising", has been described on fruits submitted to mechanical impacts while showing high astringency levels. Sensitiveness to the flesh browning disorder has also been evaluated on different cultivars introduced from other countries. The implication of the redox system in the chilling injury manifestation on 'Rojo Brillante' persimmon has been determined. Moreover, we described the changes in this system associated with chilling injury alleviation by 1-MCP treatment. The effect of a controlled atmosphere based on 4-5% O2 + N2 to prolong the storage of cultivars 'Rojo Brillante' and 'Triumph' has been seen to strongly depend on variety. The results were highly positive on cultivar 'Triumph', in which the evaluated atmosphere extended the storage period up to 3 months. The use of short-term high CO2 treatments was another technology assayed to alleviate chilling injury in non-astringent cultivar 'Fuyu'. This treatment significantly reduced the main chilling injury symptom manifested by this cultivar, which is flesh gelling. This effect was related to cell structure preservation. Recent studies have shown that ethyl formate treatment is highly effective for pest control of persimmon 'Fuyu'. However, this treatment induces fruit softening, which causes quality loss. This Thesis revealed that ethyl formate treatment induces the activity of ethylene synthesis-related genes and that flesh softening is mediated by this hormone. It also demonstrated that by applying 1-MPC pretreatment, fruit softening associated with ethyl formate can be controlled. Therefore, the combined used of both treatments is seen as a potential treatment to disinfect persimmon fruits while preserving quality. Finally, the study of ten cultivars introduced from other countries helped select the most interesting cultivars to broaden the varietal range according to their maturation date and their response to deastringency treatment. The main nutritional compounds of persimmon and how they are affected by CO2 deastringency treatment are described.
[ES] El caqui es un cultivo de gran relevancia en el área mediterránea de España, estando la producción centrada en una única variedad, el cv. Rojo Brillante, y localizada principalmente en la Comunidad Valenciana. Las principales alteraciones presentadas por el caqui 'Rojo Brillante' durante el periodo postcosecha son el pardeamiento de la pulpa asociado a los daños mecánicos y los daños por frío manifestados tras la conservación a bajas temperaturas. Las investigaciones previas han determinado las condiciones de manejo bajo las cuales se desarrollan dichas alteraciones, sin embargo los procesos bioquímicos involucrados en la manifestación de estos desórdenes no se conocen en profundidad. Actualmente uno de los principales retos es la introducción de nuevas variedades que permitan ampliar la gama varietal, así como prolongar los periodos de conservación del caqui con el fin de poder escalonar la puesta en el mercado en función de la demanda. En la presente Tesis se han abordado tres objetivos principales: 1) Estudiar los procesos bioquímicos implicados en los principales desórdenes postcosecha del caqui, prestando especial atención a los cambios en el sistema redox del fruto; 2) Evaluar diferentes tratamientos postcosecha para preservar la calidad del fruto durante la conservación frigorífica; 3) Evaluar la calidad físico-química y nutricional de diferentes variedades de caqui introducidas desde otros países para ampliar la gama varietal. Estudios bioquímicos, cromatográficos y microestructurales, han revelado que el pardeamiento de la pulpa o "browning", manifestado por la fruta que ha sufrido daños mecánicos tras la eliminación de la astringencia está asociado a un proceso de oxidación de taninos motivado por una situación de estrés oxidativo. Además se ha descrito una nueva alteración de la pulpa, "pinkish bruising", manifestada por los frutos sometidos a daño mecánico con alto nivel de astringencia. También se ha evaluado la sensibilidad al pardeamiento de diferentes variedades introducidas desde otros países. Con el objetivo de prolongar la conservación del fruto se ha ensayado el uso de la atmósfera controlada basada en 4-5% O2 + N2 sobre las variedades 'Rojo Brillante' y 'Triumph', observándose que la respuesta a esta tecnología depende de manera muy importante de la variedad. Los resultados fueron especialmente positivos en el caso del cultivar 'Triumph', en el que la atmósfera ensayada permitió prolongar la conservación frigorífica hasta tres meses. Otra tecnología ensayada para el control de los daños por frío fue la aplicación de choques con altas concentraciones de CO2 en el cultivar no astringente 'Fuyu'. Este tratamiento alivió el principal síntoma de daño por frío manifestado por esta variedad, la gelificación de la pulpa. Este efecto se ha relacionado con la preservación de la estructura celular de la pulpa del fruto. Estudios recientes han mostrado que el tratamiento con etil formato es altamente efectivo en el control de insectos durante la conservación y comercialización del caqui 'Fuyu', sin embargo este tratamiento provoca un ablandamiento del fruto mermando su calidad. En esta Tesis se ha demostrado que las aplicaciones de etil formato activan los genes de síntesis de etileno, siendo está hormona mediadora del ablandamiento. Los estudios también revelaron que la aplicación de un tratamiento previo con 1-MCP reduce el ablandamiento del fruto asociado al etil formato, por lo que el uso combinado de ambos tratamientos puede ser considerado una alternativa para la desinfección de los frutos sin detrimento de su calidad. Por último, el estudio de diez variedades introducidas desde otros países permitió seleccionar en base a su momento de maduración y respuesta al tratamiento de desastringencia aquellas variedades de mayor interés para ampliar la gama varietal. [CAT] El caqui s'ha convertit en els últims anys en un cultiu de gran rellevància en l'àrea mediterrània d'Espanya, estant la producció centrada en una única varietat, el cv. Rojo Brillante, i localitzada principalment a la Comunitat Valenciana. Les principals alteracions presentades pel caqui 'Rojo Brillante' durant el període postcollita són l'enfosquiment de la polpa associat als danys mecànics i els danys per fred manifestats després de la conservació a baixes temperatures. Les investigacions prèvies han determinat les condicions de maneig sota les quals es desenvolupen estes alteracions, però els processos bioquímics involucrats en la manifestació d'estos desordres no es coneixen en profunditat. D'altra banda, actualment un dels principals reptes és la introducció de noves varietats que permetin ampliar la gamma varietal, així com prolongar els períodes de conservació del caqui per tal de poder escalonar la posada en el mercat en funció de la demanda. En este context, en la present Tesi s'han abordat tres objectius principals: 1) Estudiar els processos bioquímics implicats en els principals desordres postcollita del caqui, posant especial atenció als canvis en el sistema redox del fruit; 2) Avaluar diferents tractaments postcollita per preservar la qualitat del fruit durant la conservació frigorífica; 3) Avaluar la qualitat fisicoquímica i nutricional de diferents varietats de caqui introduïdes des d'altres països per ampliar la gamma varietal. Estudis bioquímics, cromatogràfics i microestructurals, han revelat que l'enfosquiment de la polpa o "Browning", manifestat per fruita que ha patit danys mecànics després de l'eliminació de l'astringència està associat a un procés d'oxidació de tanins motivat per una situació d'estrès oxidatiu. A més s'ha descrit una nova alteració de la polpa, "pinkish bruising", manifestada pels fruits sotmesos a dany mecànic amb alt nivell d'astringència. També s'ha avaluat la sensibilitat a l'enfosquiment de diferents varietats introduïdes des d'altres països. Amb l'objectiu de perllongar la conservació del fruit s'ha assajat l'ús de l'atmosfera controlada basada en 4-5% O2 + N2 sobre les varietats 'Rojo Brillante' i 'Triumph', observant-se que la resposta a esta tecnologia depèn de manera molt important de la varietat. Els resultats van ser especialment positius en el cas del cultivar 'Triumph', en què l'atmosfera assajada va permetre prolongar la conservació frigorífica fins a tres mesos. Una altra tecnologia assajada per al control dels danys per fred va ser l'aplicació de xocs amb altes concentracions de CO2 en el cultivar no astringent 'Fuyu'. Este tractament va alleujar el principal símptoma de dany per fred manifestat per esta varietat, la gelificació de la polpa. Este efecte s'ha relacionat amb la preservació de l'estructura cel¿lular de la polpa del fruit. Estudis recents han mostrat que el tractament amb etil format és altament efectiu en el control d'insectes durant la conservació i comercialització del caqui 'Fuyu', però este tractament provoca un estovament del fruit minvant la seua qualitat. En esta Tesi s'ha demostrat que les aplicacions d'etil format activen els gens de síntesi d'etilè, sent esta hormona mediadora de l'estovament. Els estudis també van revelar que l'aplicació d'un tractament previ amb 1-MCP redueix l'estovament del fruit associat a l'etil format, per la qual cosa l'ús combinat de tots dos tractaments pot ser considerat una alternativa per a la desinfecció dels fruits sense detriment de la seua qualitat. ENGLISH ABSTRACT: Rind colour is one of the most important external quality characteristics of citrus fruit and plays an important role in purchasing decisions by consumers. Consumers perceive brightlycoloured fruit to be sweet and mature, whereas citrus with a green rind is perceived to be sour and immature. However, there is a poor correlation between rind colour and internal quality, contradicting what is generally assumed by the fruit-buying public. In general, a bright orange rind colour improves consumer acceptance. Thus, it is important to ensure that the rind of citrus fruit is well-coloured on arrival at the market. Various pre-harvest cultural practices and postharvest techniques can be applied to improve rind colour. Degreening with ethylene gas is the most commonly used postharvest technology to improve rind colour, but has various negative side-effects. Degreened fruit are more prone to decay, have rinds which appear dull and flaccid, have been reported to develop off-flavours and have a shorter shelf-life period. Therefore, it is necessary to find alternatives to ethylene degreening and to extend shelf-life of citrus fruit. Under normal orchard conditions, rind colour development is associated with low night temperatures, usually experienced during autumn or following the passing of a cold front. To simulate cold front conditions, a hydrocooler and cold room were used to rapidly drop fruit temperature to 4 ºC for 6 hours, and then fruit were incubated at 20 to 22 ºC for 72 hours. This “cold shock” treatment of ‘Nules Clementine’ mandarin improved rind colour to a level similar to that of degreened fruit in the 2002 season due to a decrease in chlorophyll content and increase in carotenoid content. However, this result could not be repeated. Storage temperature is one of the most important postharvest factors affecting rind colour. Citrus fruit shipped to export markets requiring low temperatures (-0.6 ºC) for pest disinfestations purposes have been reported to arrive with poor rind colour. Shipping under low temperatures results in poor rind colour of fruit on arrival in the market. To comply with the USA’s phytosanitary requirement for imported citrus, fruit is held at -0.6 ºC for a minimum of 22 days. The effect of shipping at various temperatures (-0.6 ºC or 4.5 ºC), durations and the influence of initial rind colour, “orange” or “yellow”, on fruit colour upon arrival in the market was evaluated. Fruit shipped at a higher temperature (4.5 ºC) had a marginally better rind colour than fruit shipped at -0.6 ºC. The perceived loss of rind colour following shipping at sub-zero temperatures is probably due to carotenoid degradation. Therefore, initial rind colour plays a critical role in final product quality. Depending on market destination and shipping temperature, pale-coloured fruit should not be packed for markets sensitive to rind colour. Holding temperature after shipping can be effectively used to improve the rind colour of fruit arriving in the market with undesirable rind colour. An intermediate holding temperature of between 11 and 15 ºC resulted in the greatest improvement in rind colour after 2 weeks. A high holding temperature (20 ºC) caused colour degradation, whereas a low holding temperature (4.5 ºC) resulted in the maintenance of rind colour. By selecting the correct holding temperature, even after shipping at sub-zero temperatures, final colour can be improved.
AFRIKAANSE OPSOMMING: Tyd-temperatuur interaksie op na-oes skilkleur ontwikkeling by sitrus Skilkleur is een van die belangrikste eksterne kwaliteitseienskappe van die sitrusvrug en spëel ʼn belangrikke rol in wat verbruikers koop. Verbruikers verwag dat heldergekleurde vrugte soet en ryp sal wees, terwyl sitrus met ʼn groen skil geassosieer word met onrypheid en ʼn suur smaak. In teenstelling hiermee is daar egter ʼn swak korrelasie tussen skilkleur en interne kwaliteit. Aangesien ʼn heldergekleurde oranje skil verbruikersaanvaarding verbeter, is dit dus belangrik om te verseker dat die sitrusvrug ʼn goeie skilkleur het teen die tyd wat dit die mark bereik. Verskeie voor-oes bestuurspraktyke en na-oes tegnieke kan toegepas word om die skilkleur te verbeter. Ontgroening met etileen gas is die tegnologie wat mees algemeen gebruik word om skilkleur na oes te verbeter, maar dit het egter verskeie newe effekte tot gevolg. Ontgroende vrugte is meer vatbaar vir bederf en verwelkde skille met ʼn dowwe voorkoms. Afsmaake kan voorkom en ʼn verkorte rakleeftyd is al gerapporteer. Matige temperature tussen 11 en 15 °C het na 2 weke die beste verbetering in skilkleur gelewer. Hoër temperature (20 °C) het skilkleur nadelig beinvloed, terwyl lae temperature skilkleur behou het. Deur die korrekte temperatuur te kies, selfs na verskeping by temperature onder vriespunt, kan uiteindelike skilkleur steeds verbeter word.

[ES] En las últimas dos décadas, la producción de caqui en España ha aumentado exponencialmente y la superficie de cultivo se ha multiplicado casi por ocho. En la actualidad, la producción de caqui está centralizada en el cultivar 'Rojo Brillante', que es mundialmente apreciado. Sin embargo, esta centralización supone una importante limitación comercial. En la presente Tesis se abordan dos objetivos principales: 1) Optimización de la tecnología pre y postcosecha con el fin de garantizar la calidad del caqui 'Rojo Brillante' durante la frigoconservación y el transporte a mercados internacionales. 2) Evaluación del comportamiento bajo condiciones mediterráneas de variedades introducidas de otras regiones productoras con el fin de ampliar el rango varietal de caqui. El primer objetivo de esta Tesis se logró a través de los estudios presentados en los Capítulos I al V. El segundo objetivo se abordó en los Capítulos VI y VII. En el Capítulo I se esclarecieron las causas del pardeamiento interno del fruto en los envíos a los mercados internacionales. La temperatura a la que se expone la fruta inmediatamente tras la aplicación del tratamiento de desastringencia con altas concentraciones de CO2 se identificó como el principal factor implicado en esta alteración. Los resultados obtenidos mostraron que esta alteración se puede evitar mediante la implementación de un período de atemperado de 24 h después del tratamiento con CO2, antes de conservar las frutas a bajas temperaturas. En el Capítulo II se evaluó la eficacia de un tratamiento de desastringencia recientemente patentado basado en la aplicación de una nueva cera que contiene etanol en su formulación. Los resultados mostraron que esta cera podría ser una alternativa al tratamiento convencional de desastringencia con altas concentraciones de CO2 cuando la fruta se envía a mercados lejanos a bajas temperaturas. La aplicación de la nueva cera antes de conservación en frío provocó la pérdida de astringencia de la fruta tras 30 días a 0 °C, preservando la firmeza y la calidad interna. Los resultados del Capítulo III demostraron que la realización de dos aplicaciones de ácido giberélico (AG) en precosecha permite retrasar el proceso de maduración de la fruta en el árbol con respecto a una sola aplicación. La combinación de AG en precosecha y 1-metilciclopropeno (1-MCP) en postcosecha resultó en una mayor calidad del fruto durante la frigoconservación en comparación con la aplicación únicamente de 1-MCP. Además, la fruta tratada con múltiples aplicaciones de AG mostró una firmeza ligeramente mayor tras de conservación a baja temperatura. En el Capítulo IV se demostró que, paralelamente al retraso de la maduración, el tratamiento con AG también retrasa la senescencia del cáliz, incrementando la calidad externa del fruto. La senescencia del cáliz durante la maduración del fruto se relacionó con una disminución de los parámetros de fluorescencia de clorofila (Fo, Fm y Fv/Fm). En el Capítulo V se estudiaron las condiciones óptimas de almacenamiento del caqui producido bajo cultivo ecológico. Según los resultados, no se deben almacenar los frutos cosechados con firmeza inferior a 30 N. La calidad del fruto durante la conservación dependió de las condiciones de almacenamiento, momento de aplicación del tratamiento de deastringencia y estado de madurez inicial. Por último, en los Capítulos VI y VII se abordó la evaluación bajo condiciones mediterráneas de 14 variedades de caquis no-astringentes procedentes de otros países. Se identificaron las variedades tempranas y tardías que pueden ser potencialmente interesantes para ampliar el período de cosecha en nuestra región. Los estudios postcosecha mostraron que todas las variedades estudiadas presentan una buena aptitud para ser comercializadas en los mercados nacionales y europeos. Sin embargo, la sensibilidad a los daños por frío que presentaron algunas de las variedades estudiadas podría limitar su comercialización a países de ultramar. Además, se observó que la respuesta al tratamiento con 1-MCP, para el control de los daños por frío, dependió en gran medida de la variedad.
[CA] En les últimes dues dècades, la producció de caqui a Espanya ha augmentat exponencialment i la superfície de cultiu s'ha multiplicat quasi per huit vegades. En l'actualitat, la producció de caqui està centralitzada en el la cultivar 'Rojo Brillante', que és mundialment apreciat. Malgrat això, aquesta centralització suposa d'una banda una important limitació comercial, ja que la producció es concentra en un període de collita relativament curt i, d'altra banda implica un alt risc fitosanitari. En la present Tesi s'aborden dos objectius principals: 1) Optimització de la tecnologia pre i postcollita amb la finalitat de garantir la qualitat del caqui 'Rojo brillante' durant la frigoconservació i el transport a mercats internacionals. 2) Avaluació del comportament de varietats introduïdes d'altres regions productores sota condicions mediterrànies amb la finalitat d'ampliar el rang varietal de caqui. En el Capítol I es van esclarir les causes de l l'enfosquiment intern. La temperatura a la qual s'exposa la fruita immediatament després de l'aplicació del tractament de desastringència amb altes concentracions de CO¿ es va identificar com el principal factor implicat en aquesta alteració. Els resultats obtinguts van mostrar que aquesta alteració es pot evitar mitjançant la implementació d'un període de temperat de 24 h després del tractament amb CO¿, abans de transferir la fruita a baixes temperatures. En el Capítol II es va avaluar l'eficàcia d'un tractament de desastringència recentment patentat basat en l'aplicació d'una nova cera que conté etanol en la seua formulació en caquis 'Rojo Brillante' i 'Triumph'. Els resultats van mostrar que aquesta cera podria ser una alternativa al tractament convencional de desastringència amb altes concentracions de CO¿ quan la fruita s'envia a mercats llunyans a baixes temperatures. L'aplicació de la nova cera abans de conservació en fred va provocar la pèrdua d'astringència de la fruita després de 30 dies a 0 °C, preservant la fermesa i la qualitat interna. Els resultats del Capítol III van demostrar que l'aplicació de dos tractaments d'àcid giberèlic (AG) en precollita permet retardar el procés de maduració de la fruita en l'arbre respecte a una sola aplicació. La combinació de AG en precollita i 1-metilciclopropeno (1-MCP) en postcollita va incrementar la qualitat del fruit durant la frigoconservació en comparació amb l'aplicació únicament de 1-MCP. A més, la fruita tractada amb múltiples aplicacions de AG va mostrar una fermesa lleugerament major després de conservació a baixa temperatura; no obstant això, una sola aplicació de AG també va ser eficaç per a previndre la manifestació de danys per fred en la fruita. En el Capítol IV es va demostrar que, paral·lelament al retard de la maduració, el tractament amb AG també retarda la senescència del calze, incrementant la qualitat externa del fruit. La senescència del calze durant la maduració del fruit es va relacionar amb una disminució dels paràmetres de fluorescència de clorofil·la (Fo, Fm i Fv/Fm). En el Capítol V es van estudiar les condicions òptimes d'emmagatzematge del caqui produït baix cultiu ecològic. D'acord amb els resultats obtinguts, no s'han d'emmagatzemar els fruits collits amb fermesa inferior a 30 N. La conservació del fruit va dependre de les condicions d'emmagatzematge, moment d'aplicació del tractament de desastringència i estat de maduresa de la fruita. Finalment, en els Capítols VI i VII es va abordar l'avaluació de 14 varietats de caquis no-astringents. Es van identificar les varietats primerenques i tardanes que poden ser potencialment interessants per a ampliar el període de collita. D'altra banda, els estudis postcollita van mostrar que totes les varietats estudiades presenten una bona aptitud per a ser comercialitzades en els mercats nacionals i europeus. No obstant això, la sensibilitat als danys per fred que van presentar algunes de les varietats estudiades podria limitar la seua comercialització a països d'ultramar. A més, es va observar que la resposta al tractament amb 1-MCP, per al control dels danys per fred, va dependre en gran manera de la varietat.
[EN] In the last two decades, persimmon production in Spain has increased exponentially and the cultivation area has expanded almost eight-fold. The current production of persimmon is focused on the cultivar 'Rojo Brillante', which is appreciated worldwide due to the high quality of the fruit. Nevertheless, centralized production of this single cultivar implies a major commercial limitation as its high volume of production is concentrated in a relative short harvesting period. Furthermore, it also implies a high phytosanitary risk. In this context, this Thesis addresses two main objectives: 1) Optimization of pre- and postharvest technology in order to guarantee the quality of 'Rojo Brillante' persimmon after cold storage and transportation to overseas markets. 2) Evaluation of the behavior of foreign varieties of persimmon under Mediterranean conditions in order to extend the varietal range. The first objective of this Thesis was tackled through the studies presented in Chapters I to V. The second objective was covered in Chapters VI and VII. Chapter I dealt with the causes of internal browning disorder manifested after cold shipment of 'Rojo Brillante' persimmons to overseas markets. The temperature to which fruit is exposed immediately after CO2 deastringency treatment was identified as the main factor involved in this alteration. Our results showed that this disorder can be avoided by implementing an attemperation period of 24 h after the CO2 treatment, before transferring fruit to cold storage. In Chapter II, the effectiveness of a recently patented deastringency treatment based on applying a new wax containing ethanol was assayed in 'Rojo Brillante' and 'Triumph' persimmons. The use of this new ethanol-based wax could be a potential alternative to the conventional CO2 deastringency treatment when the fruit are sent to distant overseas markets at low temperatures. Application of the new wax before cold storage led to loss of fruit astringency after 30 days at 0 °C while preserving fruit firmness and internal quality. The results of Chapter III demonstrated that two applications of gibberellic acid (GA) made it possible to delay the fruit maturity process on the tree more than with just a single application. The combination of GA and 1-methylciclopropene (1-MCP) preserved the fruit quality better than with the application of 1-MCP alone during cold storage. Moreover, the fruit treated by multiple GA applications showed a slightly higher firmness after cold storage. However, a single GA application was also effective in preventing the manifestation of chilling injury (CI) in fruit. Chapter IV demonstrated that in parallel to delaying maturity, the GA treatment also delayed calyx senescence, which meant that the fruit preserved a good appearance. The calyx lobe senescence during fruit ripening was linked to a decrease in all Chlorophyll Fluorescence Imaging (CFI) parameters (Fo, Fm, and Fv/Fm). Chapter V addressed the optimization of storage conditions for organic persimmons. According to our results, fruit harvested with a firmness lower than 30 N must not be stored. Fruit harvested with a higher level of firmness could be stored for up to three weeks depending on the storage conditions, the moment of application of the CO2 deastringency treatment, and the stage of fruit maturity. The longest storage period (3 weeks) was achieved when fruit were harvested with a firmness of around 45 N, submitted to CO2 treatment and then stored at 15 °C. Finally, 14 foreign varieties grown under Mediterranean conditions were evaluated to identify early- and late-season varieties with potential to extend the harvesting period. This is presented in Chapters VI and VII. In general, all the evaluated varieties fulfilled the quality criteria to be commercialized in domestic and European markets. However, sensitivity to CI was variety dependent and response to 1-MCP treatment was also observed to depend on the variety.

Agriculture has emerged as a major economic activity in the African continent. Therefore, fruits and vegetables are considered as essential source of vitamins, minerals and proteins. However, fruits and vegetables are perishing rapidly. Thus, the adequate handling starts from the field, by applying appropriate method of storage and preservation in order to reduce post-harvest losses and extend its shelf life. Preservation by removing heat from the products is the most common method of preservation. In rural areas, the access to the grid is expensive or in some cases impossible. Therefore, researchers have been paying more attention to find alternative sources of power to run the cooling units. This study provides proof of a concept for the use of solar energy to cool down harvested fruits and vegetables at the farm level to an adequate farm storage temperature. The target storage temperature range is between 5 and 15 oC. A model refrigerator was designed and constructed in the mechanical engineering workshop of Cape Peninsula University of Technology. It was installed and tested in outdoor conditions to get the effect of different weather conditions. The model consists of a typical vapour compression system powered by a 12 V solar PV system. A DC compressor was used, and therefore, there is no inverter. The model was first tested without a product and then with 20 kg batches of different fruits for a period of two weeks in April 2016. Wind speed, ambient temperature and solar radiation intensity data were monitored and collected from a Campbell Scientific weather station mounted on the roof adjacent to the model. By monitoring the temperatures and compressor current hourly, the refrigeration effect, power consumption and coefficient of performance were determined. The overall COP based on input solar energy was 2.8. It was thus proved that a suitably sized PV system could be designed and implemented at farm level to cool harvested fruits from ambient to storage temperature without the use of an inverter. This could go some way to helping retard deterioration of fruits and vegetables before delivery to a marketplace or to storage.

ENGLISH ABSTRACT: The table grape industry uses rapid cooling and packaging to protect grapes from desiccation and decay. Numerous packaging methods and combinations are used in the industry with each having their own advantages and disadvantages. Inferior postharvest grape quality can usually be ascribed to either deficient or excessive moisture in the carton. Berry split, decay and S02 damage are all disorders that are either caused or aggravated by wet berries in conjunction with elevated temperature. On the other hand, grapes that are exposed to desiccating conditions will develop brown stems and cause ineffective control by S02 gas generators. Moisture management is governed by perforated or non-perforated liners and/or by placing moisture absorbing materials inside the liners. To find the optimum liner perforation or moisture sheet combination, 'Thompson Seedless' and 'Red Globe' (Vitis vinifera Linnaeus) table grape quality was evaluated in various trials. The investigation of non-perforated liners compared to liners with different degrees of perforation concluded the following: Perforated liners benefit grape quality by decreasing S02 damage and berry split due to less moisture in the carton. These benefits, however, also lead to loss in quality due to increased stem desiccation and a lower S02 concentration in the packaging. The lower moisture content in the carton compensates for the lower S02 concentration, creating an environment less favourable for decay development. S02 damage and berry split decreased with an increase in degree of liner perforation, irrespective of the cultivars sensitivity to the disorder. Optimum level of perforation depends on the specific sensitivity of a cultivar to certain quality disorders and the characteristics of the quality disorders associated with a cultivar. Additionally, packing conditions such as product temperature and humidity should be considered. The specific costs associated with the advantages and disadvantages influenced by the degree of liner perforation will be the deciding factor in liner selection. The investigation of a clay-containing, moisture absorbing sheet emphasized the benefits and risks of absorbing large amounts of water within the packaging. Irrespective of using a perforated or non-perforated liner the influence of the desiccant sheet was evident throughout the trials. It benefited grape quality by lowering the incidence of berry split and S02 damage. However, decay control was impaired by the desiccant sheet, and stem desiccation was aggravated. The comparison of non-perforated liners with liners of various degrees of perforation showed the benefit of faster cooling rates of perforated liners. The various perforated liners showed little variation in airflow and cooling times. Morphological studies of various cultivars could not ascribe differences in stem condition to anatomical dissimilarities between various cultivars. It was found that 'Red Globe' had a much larger berry volume to stem weight ratio contributing to a high rate of water loss and stem dehydration. Stem visibility is high in 'Red Globe' due to the straggly, loose nature of the bunches. This heightens the perception of dry, brown stems and overemphasizes the actual severity of the disorder.
AFRIKAANSE OPSOMMING: Die tafeldruifbedryf gebruik versnelde verkoeling en verpakking om druiwe te beskerm teen uitdroging en bederf. Verskeie verpakkingsmetodes word gebruik in die industrie waarvan elkeen sy eie voor- en nadele het. Ondergeskikte na-oes kwaliteit kan gewoonlik toegeskryf word aan óf te min óf te veel vog in die karton. Korrelbars, S02 skade en bederf is almal kwaliteitsdefekte wat óf veroorsaak word, óf vererger word deur nat korrels, saam met 'n verhoging in temperatuur. In teenstelling hiermee sal druiwe wat blootgestel word aan droë toestande, bruin stingels ontwikkel en S02 beheer salook ondoeltreffend wees. Vog in verpakking word beheer deur geperforeerde of nie-geperforeerde binnesakke en/of deur vogabsorberende materiaal binne die binnesak te plaas. Om die optimum binnesak perforasie of vogabsorberende vel kombinasie te vind is 'Thompson Seedless' en 'Red Globe' (Vitis vinifera Linnaeus) tafeldruif kwaliteit ge-evalueer in verskeie proewe. Die bestudering van nie-geperforeerde binnesakke teenoor binnesakke met verskillende grade van perforasies het die volgende resultate gelewer: Geperforeerde binnesakke bevoordeel druif kwaliteit deur die vermindering van S02 skade en korrelbars weens minder vog in die karton. Hierdie voordele sal egter lei tot verlies in kwaliteit weens die vinniger uitdroging van stingels en die verlaging van S02 konsentrasie in die verpakking. Resumo: A hortaliça folhosa mais consumida e produzida no Brasil é a alface (Lactuca sativa L.) e seu consumo vem crescendo a cada ano devido a uma tendência da população mundial em adquirir hábitos de vida mais saudáveis. Existem diferentes métodos de resfriamento para conservação pós-colheita, já utilizados e aplicados em olerícolas, porém não se tem conhecimento sobre seus efeitos e benefícios para a alface. O objetivo deste estudo foi avaliar a aplicação de diferentes métodos de resfriamento e sua adequação para diferentes variedades de alface. Os tipos de alfaces analisadas foram a repolhuda crespa ou americana, a solta crespa, a repolhuda lisa e a solta lisa, adquiridas de um produtor da região de Campinas-SP. Foram avaliados três métodos de resfriamento seguidos de acondicionamento em câmara fria a 5ºC: resfriamento com ar forçado (com controle da umidade relativa do ar e sem controle da umidade relativa do ar), resfriamento evaporativo em túnel (nebulização e ventilador-meio poroso) e resfriamento com água fria (imersão e aspersão), sendo que o controle consistiu apenas no acondicionamento do produto em câmara fria a 5ºC, e os parâmetros físico-químicos (pH, ácido ascórbico, clorofila e perda de massa). O delineamento experimental foi inteiramente casualizado (DIC) com cinco repetições por tratamento, observando-se a interação entre os métodos de resfriamento e tempo de armazenamento, e estes parâmetros foram submetidos à análise estatística multivariada aplicando-se a análise de componentes principais. Conclui-se que o melhor método para o resfriamento de alface é o do ar forçado com umidificação, pois equilibra eficiência de resfriamento e menor degradação do produto. O resfriamento da alface pelo método evaporativo apresenta uma lenta retirada de calor especifico, desfavorecendo a manutenção da vida útil do produto, assim como o método por água fria que causa acúmulo de água no interior do produto facilitando as reações de deterioração. Com relação à análise de custo energético, o resfriamento de alface pelo método ar forçado se torna mais interessante em termos de aplicabilidade e manutenção da qualidade do produto, mesmo apresentando um maior custo se comparado ao resfriamento de alface pelo método evaporativo
Abstract: The leafy vegetable most consumed and produced in Brazil is the lettuce (Lactuca sativa L.) and its consumption is growing up every year due a tendency of world population to acquire healthier lifestyle. There are different cooling methods for post-harvest quality, already used and applied in vegetable crops, but there is no knowledge about yours effects and benefits for lettuce. The objective of this study was evaluating the application of different cooling methods and their suitability for different varieties of lettuce. The types of lettuce analyzed were "repolhuda crespa" ou "americana", a "solta crespa", a "repolhuda lisa" e a "solta lisa", acquired from a region producer in Campinas-SP. We evaluated three cooling methods followed to a storage in cold chamber at 5 ° C: forced air cooling (with relative humidity control and no relative humidity control), evaporative cooling tunnel (misting and porous-fan) and hydroccoling (immersion and spray), and control consisted in the product cold storage at 5°C, was evaluate the physic-chemical parameters (pH, ascorbic acid, chlorophyll and weight loss). The experimental design was completely randomized, with five replicates per treatment, observing the interaction between cooling methods and storage time, and these parameters were subjected to multivariate statistical analysis with applying the principal component analysis. It is concluded that the best method for cooling lettuce is forced air with humidification it balances cooling efficiency and less degradation of the product. The cooling of the lettuce by evaporative method has a slower specific heat removed, which does not favor the maintenance product shelf-life, as well as the hydrocooling causes water accumulation in the product facilitates the degradation reactions. Regarding the analysis of cost energy, the cooling lettuce by forced air method becomes more interesting in terms of applicability and maintenance at product quality, even with a higher cost compared to the cooling of lettuce evaporative method

ENGLISH ABSTRACT: Knowledge of postharvest quality attributes of minimally processed packaged fruit is essential in order to establish the optimum shelf life period. The aim of this study was to investigate the effects of Passive-modified atmosphere packaging (MAP) on the quality of minimally processed pomegranate (cv. Bhagwa) arils and aril-sacs. These began by understanding the physiological processes i.e. respiration and transpiration rates of the whole fruit, arils and aril-sacs. The respiration rates (RR) of whole fruit, aril-sacs and arils were studied at 5, 10, 15 and 22°C, and comparisons were made among these fruit fractions. A high RR was observed in aril-sacs compared to whole fruit and arils across all storage temperatures. A 74.5% decrease in RR was observed when storage temperature was reduced from 22°C to 5°C. A significant increase in RR occurred from day 3 of storage across all fruit fractions and storage temperatures. The transpiration rates (TR) of arils and aril-sacs were studied at storage conditions of 5, 10 and 15°C and 76, 86 and 96% relative humidity (RH), and was found to increase with increase in temperature and decrease in relative humidity, with lowest TR occurring in fruit fractions stored at 5°C and 96% RH showing lower TR. Arils had high TR compared to aril-sacs, and this may be related to high surface area to volume ratio of exposed arils. The effects of modified atmosphere packaging and application of anti-browning agents on quality of arils and aril-sacs stored at 5°C were studied. Compared to clamshell packaging, Passive-MAP using POLYLID® 107 polyethylene (PE) polymeric film showed greater positive effects in maintaining the quality and extends the shelf life of the arils and aril-sacs. Furthermore, the anti-browning agents used controlled browning on the cut-surfaces of the peel of the aril-sacs and reduced microbial growth in both arils and aril-sacs. When the effects of MAP and anti-browning were combined, aril-sacs stored better than arils. These treatments extended the shelf life of aril-sacs to 12 days while arils lasted up to 9 days. The water vapour transmission rate (WVTR) of pomegranate fruit membrane was evaluated at cold storage (5°C, 90% RH) and room condition (18.7°C, 70% RH). A high WVTR occurred in membranes stored at room condition, compared to those stored at cold storage. Further studies are warranted to improve our understanding of the biophysical properties of pomegranate membranes in relation to possible exchange of water vapour and gases between the aril-sacs. In summary, the use of MAP in combination with anti-browning agents showed a high potential in maintaining the quality of pomegranate arils and aril-sacs and consequently increase their shelf-life.
AFRIKAANSE OPSOMMING: Kennis van naoes- gehalte-eienskappe van minimaal geprosesseerde verpakte vrugte is essensieel ten einde optimum rakleeftyd te bepaal. Die doel van hierdie studie was om die gevolge van passiewe gemodifiseerde atmosfeerverpakking (GAV) op die gehalte van arils en arilsakkies van minimaal geprosesseerde granaat (kv. Bhagwa) te ondersoek. ʼn Aanvang is gemaak deur die fisiologiese prosesse, m.a.w. respirasie- en transpirasietempo’s van die hele vrugte, arils en arilsakkies, te begryp. Die respirasietempo’s (RT) van hele vrugte, arilsakkies en arils is by 5, 10, 15 en 22°C bestudeer, en vergelykings is getref tussen hierdie vrugdele. ʼn Hoë RT is waargeneem by arilsakkies in vergelyking met hele vrugte en arils oor alle bergingstemperature heen. ʼn Afname van 74.5% RT is waargeneem toe bergingstemperatuur van 22°C na 5°C verminder is. ʼn Beduidende toename in RT het van dag 3 van berging af oor alle vrugdele en bergingstemperature heen voorgekom. Die transpirasietempo’s (TR) van arils en arilsakkies is by bergingstoestande van 5, 10 en 15°C en 76, 86 en 96% relatiewe humiditeit (RH) bestudeer, en daar is bevind dat dit verhoog met ’n toename in temperatuur en ʼn afname in relatiewe humiditeit, met die laagste TR wat voorkom by vrugdele geberg by 5°C en 96% RH wat dus laer TR toon. Arils het hoë TR gehad in vergelyking met arilsakkies, en dit kan verband hou met die verhouding van hoë oppervlakarea tot volume blootgestelde arils. Die gevolge van gemodifiseerde atmosfeerverpakking en aanwending van middels vir die voorkoming van verbruining op gehalte van arils en arilsakkies geberg teen 5°C is bestudeer. In vergelyking met verpakking in toeknipbakkies (clamshell packaging), het passiewe GAV waarby POLYLID® 107 poliëtileen- (PE) polimeriese film gebruik is, groter positiewe gevolge by die behoud van gehalte getoon, en die rakleeftyd van die arils en arilsakkies is verleng. Daarbenewens het die middels vir die voorkoming van verbruining beheerde verbruining op die sny-oppervlakke van die skil van die arilsakkies gebruik en mikrobiese groei in beide arils en arilsakkies verminder. Toe die gevolge van GAV en die voorkoming van verbruining gekombineer is, het arilsakkies beter as arils geberg. Hierdie behandelings het die rakleeftyd van arilsakkies tot 12 dae verleng terwyl arils tot 9 dae gehou het. Die waterdamptransmissiespoed (WDTS) van granaatvrugtemembraan is geëvalueer by koel berging (5°C, 90% RH) en kamertoestande (18.7°C, 70% RH). ʼn Hoë WDTS het voorgekom by membrane wat by kamertoestande geberg is in vergelyking met dié wat in koelbewaring geberg is. Verdere studies is geregverdig vir verbetering van ons begrip van die biofisiese eienskappe van granaatmembrane in verhouding met moontlike uitruiling van waterdamp en atmosfere tussen die arilsakkies.

[ES] El objetivo de esta tesis doctoral es evaluar la técnica de imagen hiperespectral en el rango visible e infrarrojo cercano, en combinación con técnicas quimiométricas para la evaluación de la calidad de la fruta en poscosecha de manera eficaz y sostenible. Con este fin, se presentan diferentes estudios en los que se evalúa la calidad de algunas frutas que por su valor económico, estratégico o social, son de especial importancia en la Comunidad Valenciana como son el caqui 'Rojo Brillante', la granada 'Mollar de Elche', el níspero 'Algerie' o diferentes cultivares de nectarina. En primer lugar se llevó a cabo la monitorización de la calidad poscosecha de nectarinas 'Big Top' y 'Magique' usando imagen hiperespectral en reflectancia y transmitancia. Al mismo tiempo se evaluó la transmitancia para la detección de huesos abiertos. Se llevó a cabo también un estudio para distinguir los cultivares 'Big Top' y "Diamond Ray", los cuales poseen un aspecto muy similar pero sabor diferente. En cuanto al caqui 'Rojo Brillante', la imagen hiperespectral fue estudiada por una parte para monitorear su madurez, y por otra parte para evaluar la astringencia de esta fruta, que debe ser completamente eliminada antes de su comercialización. Las propiedades físico-químicas de la granada 'Mollar de Elche' fueron evaluadas usando imagen de color e hiperespectral durante su madurez usando la información de la fruta intacta y de los arilos. Finalmente, esta técnica se usó para caracterizar e identificar los defectos internos y externos del níspero 'Algerie'. En la predicción de los índices de calidad IQI y RPI usando imagen en reflectancia y transmitancia se obtuvieron valores de R2 alrededor de 0,90 y en la discriminación por firmeza, una precisión entorno al 95 % usando longitudes de onda seleccionadas. En cuanto a la detección de huesos abiertos, el uso de la imagen hiperespectral en transmitancia obtuvo un 93,5 % de clasificación correcta de frutas con hueso normal y 100 % con hueso abierto usando modelos PLS-DA y 7 longitudes de onda. Los resultados obtenidos en la clasificación de los cultivares 'Big Top' y 'Diamond Ray' mostraron una fiabilidad superior al 96,0 % mediante el uso de modelos PLS-DA y 14 longitudes de onda seleccionadas, superando a la imagen de color (56,9 %) y a un panel entrenado (54,5 %). Con respecto al caqui, los resultados obtenidos indicaron que es posible distinguir entre tres estados de madurez con una precisión del 96,0 % usando modelos QDA y se predijo su firmeza obteniendo un valor de R2 de 0,80 usando PLS-R. En cuanto a la astringencia, se llevaron a cabo dos estudios similares en los que en el primero se discriminó la fruta de acuerdo al tiempo de tratamiento con altas concentraciones de CO2 con una precisión entorno al 95,0 % usando QDA. En el segundo se discriminó la fruta de acuerdo a un valor de contenido en taninos (0,04 %) y se determinó qué área de la fruta era mejor para realizar esta discriminación. Así se obtuvo una precisión del 86,9 % usando la zona media y 23 longitudes de onda. Los resultados obtenidos para la granada indicaron que la imagen de color e hiperespectral poseen una precisión similar en la predicción de las propiedades fisicoquímicas usando PLS-R y la información de la fruta intacta. Sin embargo, cuando se usó la información de los arilos, la imagen hiperespectral fue más precisa. En cuanto a la discriminación del estado de madurez usando PLS-DA, la imagen hiperespectral ofreció mayor precisión, 95,0 %, usando la información de la fruta intacta y del 100 % usando la de los arilos. Finalmente, los resultados obtenidos para el níspero indicaron que la imagen hiperespectral junto con el método de clasificación XGBOOST pudo discriminar entre muestras con y sin defectos con una precisión del 97,5 % y entre muestras sin defectos o con defectos internos o externos con una precisión del 96,7 %. [CAT] L'objectiu de la present tesi doctoral se centra en avaluar la capacitat de la imatge hiperespectral en el rang visible i infraroig pròxim, en combinació amb mètodes quimiomètrics, per a l'avaluació de la qualitat de la fruita en post collita de manera eficaç i sostenible. A aquest efecte, es presenten diferents estudis en els quals s'avalua la qualitat d'algunes fruites que pel seu valor econòmic, estratègic o social, són d'especial importància a la Comunitat Valenciana com són el caqui 'Rojo Brillante', la magrana 'Mollar de Elche', el nispro 'Algerie' o diferents cultivares de nectarina. En primer lloc es va dur a terme la monitorització de la qualitat post collita de nectarines 'Big Top' i 'Magique' per mitjà d'imatge hiperespectral en reflectància i trasnmitancia. Així mateix es va avaluar la transmitància per a la detecció d'ossos oberts. Es va dur a terme també un estudi per distingir els cultivares 'Big Top' i 'Diamond Ray', els quals posseeixen un aspecte molt semblant però sabor diferent. Pel que fa al caqui 'Rojo Brillante', la imatge hiperespectral va ser estudiada d'una banda per a monitoritzar la seua maduresa, i per un altre costat per avaluar l'astringència, que ha de ser completament eliminada abans de la seua comercialització. Les propietats fisicoquímiques de la magrana 'Mollar de Elche' van ser avaluades per la imatge de color i hiperespectral durant la seua maduresa usant la informació de la fruita intacta i els arils. Finalment, aquesta tècnica es va fer servir per caracteritzar i identificar els defectes interns i externs del nispro 'Algerie'. En la predicció dels índexs de qualitat IQI i RPI usant imatge en reflectància com en trasnmitancia es van obtindre valors de R2 al voltant de 0,90 i en la discriminació per fermesa una precisió entorn del 95,0 % utilitzant longituds d'ona seleccionades. Pel que fa a la detecció d'ossos oberts, l'ús de la imatge hiperespectral en transmitància va obtindre un 93,5 % classificació correcta de fruites amb os normal i 100 % amb os obert usant models PLS-DA i 7 longituds d'ona. Els resultats obtinguts en la classificació dels cultivares 'Big Top' i 'Diamond Ray' van mostrar una fiabilitat superior al 96,0 % per mitjà de l'ús de models PLS-DA i 14 longituds d'ona, superant a la imatge de color (56,9 %) i a un panell sensorial entrenat (54,5 %). Quant al caqui, els resultats obtinguts van indicar que és possible distingir entre tres estats de maduresa amb una precisió del 96,0 % usant models QDA i es va predir la seua fermesa obtenint un valor de R2 de 0,80 usant PLS-R. Pel que fa a l'astringència, es van dur a terme dos estudis similars en què el primer es va discriminar la fruita d'acord al temps de tractament amb altes concentracions de CO2 amb una precisió al voltant del 95,0 % usant QDA. En el segon, es va discriminar la fruita d'acord a un valor de contingut en tanins (0,04 %) i es va determinar quina part de la fruita era millor per a realitzar aquesta discriminació. Així es va obtindre una precisió del 86,9 % usant la zona mitjana i 23 longituds d'ona. Els resultats obtinguts per la magrana van indicar que la imatge de color i hiperespectral posseïxen una precisió semblant a la predicció de les propietats fisicoquímiques usant PLS-R i la informació de la fruita intacta. No obstant això, quan es va usar la informació dels arils, la imatge hiperespectral va ser més precisa. Quant a la discriminació de l'estat de maduresa usant PLS-DA, la imatge hiperespectral va oferir major precisió (95,0 %) usant la informació de la fruita intacta i del 100 % usant la dels arils. Finalment, els resultats obtinguts pel nispro indiquen que la imatge hiperespectral juntament amb el mètode de classificació XGBOOST va poder discriminar entre mostres amb i sense defectes amb una precisió del 97,5 % i entre mostres sense defectes o amb defectes interns o externs amb una precisió del 96,7 %. [EN] The objective of this doctoral thesis is to evaluate the potential of the hyperspectral imaging in the visible and near infrared range in combination with chemometrics for the assessment of the postharvest quality of fruit in a non-destructive, efficient and sustainable manner. To this end, different studies are presented in which the quality of some fruits is evaluated. Due to their economic, strategic or social value, the selected fruits are of special importance in the Valencian Community, such as Persimmon 'Rojo Brillante', the pomegranate 'Mollar de Elche', the loquat 'Algerie' or different nectarine cultivars. First, the quality monitoring of 'Big Top' and 'Magique' nectarines was carried out using reflectance and transmittance images. At the same time, transmittance was evaluated for the detection of split pit. In addition, a classification was performed to distinguish the 'Big Top' and 'Diamond Ray' cultivars, which look very similar but have different flavour. Whereas that for the 'Rojo Brillante' persimmon, the hyperspectral imaging was studied on the one hand to monitor its maturity, and on the other hand to evaluate the astringency of this fruit, which must be completely eliminated before its commercialization. The physicochemical properties of the 'Mollar de Elche' pomegranate were evaluated by means of hyperspectral and colour imaging during its maturity using the information from the intact fruit and arils. Finally, this technique was used to characterise and identify the internal and external defects of the 'Algerie' loquat. In the prediction of the IQI and RPI quality indexes using reflectance and transmittance images, R2 values around 0.90 were obtained and in the discrimination according to firmness, accuracy around 95.0 % using selected wavelengths was obtained. Regarding the split pit detection, the use of the hyperspectral image in transmittance mode obtained a 93.5 % of fruits with normal bone correctly classified and 100% with split pit using PLS-DA models and 7 wavelengths. The results obtained in the classification of 'Big Top' and 'Diamond Ray' fruits show accuracy higher than 96.0 % by using PLS-DA models and 14 selected wavelengths, higher than the obtained with colour image (56.9 %) and a trained panel (54.5 %). According to persimmon, the results obtained indicated that it is possible to distinguish between three states of maturity with an accuracy of 96.0 % using QDA models and its firmness was predicted obtaining a R2 value of 0.80 using PLS-R. Regarding astringency, two similar studies were carried out. In the first study, the fruit was classified according to the time of treatment with high concentrations of CO2 with a precision of around 95.0 % using QDA. In the second, the fruit was discriminated according to a threshold value of soluble tannins (0.04 %) and was determined what fruit area was better to perform this discrimination. Thus, an accuracy of 86.9 % was obtained using the middle area and 23 wavelengths. The results obtained for the pomegranate indicated that the use of colour and hyperspectral images have a similar precision in the prediction of physicochemical properties using PLS-R and the intact fruit information. However, when the information from the arils was used, the hyperspectral image was more accurate. Regarding the discrimination by the state of maturity using PLS-DA, the hyperspectral image offered greater precision, of 95.0 % using the information from the intact fruit and 100 % using that from the arils. Finally, the results obtained for the 'Algerie' loquat indicated that the hyperspectral image with the XGBOOST classification method could discriminate between sound samples and samples with defects with accuracy of 97.5 % and between sound samples or samples with internal or external defects with an accuracy of 96.7 %. It was also possible to distinguish between the different defects with an accuracy of 95.9 %.

Resumo: No Brasil, as perdas na colheita e pós-colheita estão estimadas em 30 a 40% da produção. As hortícolas são as espécies vegetais cultivadas economicamente que mais contribuem para a manutenção deste elevado índice de perdas. Dentre elas, destacam-se as frutas, e em especial a banana, com perdas totais estimadas em 40%. O conhecimento das propriedades mecânicas dos materiais biológicos contribui não só para o projeto e desenvolvimento de equipamentos de colheita e pós-colheita, como também para a melhor gestão da qualidade e vida útil dos produtos. Neste trabalho avaliou-se o efeito do transporte manual quanto aos danos produzidos nos frutos da bananeira ((Musa cavendishii), determinaram-se as áreas de contato entre ombro do carregador e frutos, mediram-se as acelerações produzidas nos frutos durante o transporte manual, para topografias plana, acidentada e em superfícies gramada e de concreto. Realizaram-se ensaios mecânicos de penetração, de compressão entre pratos planos e paralelos e ensaios cíclicos de compressão. Resultados evidenciaram a sensibilidade dos frutos aos danos mecânicos mesmo no estádio verde. Danos no pedúnculo não foram observados devido a ação do transporte. Observaram-se acelerações entre 0,15 a 1,35g (9,8ms-2) indicando cargas significativas de compressão, adicionais ao peso próprio, geradas durante o caminhamento. Compressões com cargas crescentes não produziram manchas de tamanhos correlacionados com a intensidade da carga revelando que o tamanho dos danos constitui bom indicador da severidade da solicitação mecânica. Os ensaios de compressão do fruto inteiro, entre prato planos e paralelos, revelaram escoamento biológico e diferentes níveis de resistência entre frutos localizados nas regiões superior, média e inferior do cacho. Os ensaios cíclicos de compressão revelaram que solicitações repetidas de pequenas amplitudes podem promover modificações permanentes no fruto com grande potencial de danos. De uma forma geral concluiu-se que danos mecânicos estão presentes no transporte manual e que soluções para proteção do cacho ou dispositivos de auxílio ao transporte sejam necessários para a redução desses danos
Abstract: Around 30% to 40% of total losses of agricultural production in Brazil occurs during harvesting and post harvesting processes. Fruits and vegetables are the major contributor to these high losses. Among them, banana can reach losses up to 40% of total production. The knowledge of mechanical properties of fruits and vegetables is important not only for better designing of handling equipment but also to improve quality management as well as shelf life. This work reports the development of a methodology to evaluate the mechanical damage of banana fruit during manual transportation. It was measured the contact area between shoulder and fruit, accelerations during manual transportation on irregular topography on grass and concrete surfaces. Mechanical tests of penetration, compression and cyclic compression between parallel rigid plates were also performed. Results brought into evidence the high sensitivity of green banana fruits to mechanical loads. Damages in the peduncle were not observed after manual transportation. Values of acceleration between 0,15 and 1,35 g (g=9,8ms-2) were measured indicating significant additional load to the weight of the bunch during transportation. Compressions using increasing loads did not produce bruise areas correlated to load values revealing that bruise area is not a good indicator of the magnitude of the applied load. Simple compression between flat plates reveled the presence of bio-yielding and different mechanical resistance in whole fruits grown in the upper, medium and lower part of the bunch. Cyclic compression showed that repeated loads of even of small amplitudes may promote permanent changes in mechanical behavior of the tissues with great possibility of damaging them. It was concluded that mechanical damages are present during manual transportation and solutions for protection of the bunch and development of transportation aids are desirable to reduce those damages

O abacate é um fruto climatérico altamente perecível, e para sua comercialização é necessário que se faça uso de métodos de conservação pós colheita. O presente trabalho teve como objetivo a conservação do abacate ‘Hass” com a utilização de 1- metilciclopropeno (1-MCP). Os frutos após a colheita foram selecionados e expostos a diferentes concentrações de 1-MCP (200 ppm, 300 ppm e 400 ppm) e após o procedimento foram armazenados sob refrigeração a 10ºC±1 e 90±5% de UR. Foram realizadas análises de perda de massa fresca, atividade respiratória, acidez titulável (AT), pH, sólidos solúveis (SS), índice de maturação, firmeza, compostos fenólicos totais, capacidade antioxidante determinada por DPPH, atividade das enzimas polifenoloxidase (PFO), poligalacturonase (PG) e pectinametilesterase (PME). As análises foram realizadas nos frutos a cada 3 dias, durante 15 dias, depois desse período os mesmos foram transferidos para condições ambiente ( 23±4ºC e 70±5% de UR) para simulação da comercialização. Após 3 dias em condições ambientes foi realizada a última avaliação. O delineamento utilizado foi inteiramente casualizado, com 3 repetições por tratamento, utilizando-se o teste de Tukey a 5% de probabilidade. Nas condições em que o trabalho foi realizado pode-se concluir que a aplicação de 1-MCP associada ao armazenamento refrigerado foi eficiente no controle do amadurecimento dos abacates ‘Hass’. Os frutos sob aplicação de altas doses de 1-MCP apresentaram menor perda de massa ao longo do tempo. Quanto maior a dose de 1-MCP aplicada, menor o pico respiratório dos frutos, sendo que os frutos com aplicação de 400ppm de 1-MCP não apresentaram pico respiratório. O 1- MCP foi eficiente na manutenção da firmeza dos frutos porém não demonstrou efeitos sobre as enzimas poligalacturonase, pectinametilesterase e polifenoloxidase

The present study aimed at the conservation of avocado 'Hass' with the use of 1-methylcyclopropene (1-MCP). The fruits after harvest were selected and exposed to different concentrations of 1-MCP (200 ppm, 300 ppm and 400 ppm) and after the procedure they were stored at 10 º C ± 1 to 90 ± 5% RH. Analyses of weight loss, respiratory activity, titratable acidity (TA), pH, soluble solids (SS), maturation index, firmness, total phenolics, antioxidant capacity determined by DPPH, enzyme activity of polyphenoloxidase (PPO) polygalacturonase (PG) and pectin (SMEs). The analyzes were performed on fruit every 3 days for 15 days, after this period they were transferred to ambient conditions (23 ± 4 ° C and 70 ± 5% RH) for simulation of marketing. ENGLISH ABSTRACT: Stone fruit is susceptible to chilling injury and intermittent warming has been shown to alleviate chilling injury during cold storage. A dual temperature storage regime was developed in South Africa for plums based on the principles of intermittent warming. The regime consists of an initial period at -0.5°C, a variable duration warming period at 7.5°C, followed by -0.5°C. Refrigerated integral containers were designed to maintain product temperature and not to reduce product temperature, per se. Considering that dual temperature shipment requires significant refrigeration and effective distribution of cool air to remove sensible- and respiratory heat, the capacity of integral containers to ship plums successfully at dual temperature is questioned. The objectives of this study were, firstly, to analyse pulp temperature data and possibly identify different temperature zones within containers shipping plums at dual temperature. Secondly, to understand the underlying processes differentiating the temperature zones and thirdly, to determine the effect of container performance on fruit quality. Three processes were identified as important characteristics of pulp temperature data sets recorded during dual temperature shipping, namely cooling down, heating up and over heating in the container. The order of importance differed according to the cultivar shipped and the container’s performance. Three temperature zones were identified in dual temperature containers, where the average pulp temperature, time to heat up and time to cool down for each temperature zone increased along the length, across the width from the left to the right and up the container system. The variable temperature conditions were possibly due to a variation in delivery air temperature, poor airflow and the effect of increased respiration and, therefore, production of vital heat by the fruit. The cooling down process was identified as the most important process discriminating the temperature zones. With the exception of ‘Fortune’, variable temperature conditions found within integral containers shipping plums at dual temperature had a significant influence on the fruit firmness post-shipment, where deterioration levels increased from the front to the door end of the container due to an increase in pulp temperature. However, it was also shown that fruit firmness prior to shipment could have a determining effect on differences found. It could not be proven that variable temperature conditions resulted in significantly higher levels of internal defects within the integral container. Temperature zones could not be identified within refrigerated integral containers shipping plums at single temperature, suggesting that the containers are able to maintain the temperature well throughout the container area. A constant 2°C storage temperature could possibly replace the commercial dual temperature regime in the case of ‘Pioneer’ plums due to improved fruit firmness, similar colour development to the control and less sensible heat produced in the container resulting in a more stable container environment. However, unacceptably high levels of shrivel and internal browning were found.

Verkoelde integrale houers is ontwerp om produktemperature te handhaaf en nie soseer om produktemperatuur te verlaag nie. Die kapasiteit van integrale houers om pruime suksesvol teen dubbeltemperatuur te verskeep word dus bevraagteken, in ag geneem dat dubbeltemperatuurverskeping betekenisvolle verkoeling en effektiewe verspreiding van koue lug vereis om die waarneembare- en respiratoriese hitte te verwyder. Die doelwitte van die studie was eerstens om die pulptemperatuurdata te analiseer en moontlik verskillende temperatuursones binne houers wat pruime teen dubbeltemperatuur verskeep te identifiseer. ENGLISH ABSTRACT: Quality of certain Leucospermum and Leucadendron cultivars after approximately 21 days shipping has been reported to be substandard due to ‘drying out’ of leaves and, in the case of Leucadendrons, involucral leaves. The nature of the symptoms of this ‘drying out’ and the conditions under which they form, viz. long exposures to low temperatures, has led us to hypothesize that these are symptoms of chilling injury (CI). Chilling injury, as far as we are aware, has not been documented on Leucospermums or Leucadendrons. Typical CI symptomology is discussed and shown for Leucospermum ‘Gold Dust’, ‘High Gold’ and ‘Succession’ and for Leucadendron ‘Chameleon’, ‘Laurel Yellow’ and ‘Safari Sunset’. The nature of CI symptoms for Leucospermums and Leucadendrons was generally membranous breakdown that manifested in some cases as a ‘water soaked’ appearance which, at a more advanced stage, was generally visible as ‘dried out’ patches on the leaves. In the case of the Leucadendrons CI was also visible on the immature involucral leaves which are more sensitive to chilling conditions than mature leaves. Dark discoloration of especially immature involucral leaves is also a symptom of CI. As water uptake of shoots with chilling injury is hindered the styles of the Leucospermums wilt. As can be expected, the lower the temperature below the threshold temperature and the longer the exposure the more severe the symptoms. CI was recorded on cut flower shoots of Leucospermum ‘Gold Dust’, ‘High Gold’, ‘Rigoletto’, ‘Succession’ and ‘Vlam’ after 21 and 24 days storage at 1ºC. After 24 days storage the chilling injury was more severe than after 21 days storage in most cases. Each cultivar was pulsed with 5 ml per stem of a 2% (w/v) sugar solution of either lactulose, sucrose, glucose, fructose or mannose before storage. After storage, CI was recorded on day 0, 3, 7 and 10 of the vase phase. Of the cultivars tested ‘Vlam’ and especially ‘Rigoletto’ were more prone to chilling injury development. ‘High Gold’ and ‘Vlam’ shoots were pulsed with 0 (control), 1.5, 3 or 4% (w/v) solutions of either mannose or fructose. The best control of CI for both cultivars was achieved with 1.5% (w/v) solution. Lower concentrations of mannose and fructose were tested on ‘High Gold’ shoots, with a 1% (w/v) solution giving the best control for both. At high concentrations signs of toxicity became evident directly after pulsing. ‘High Gold’ shoots were pulsed with 1% (w/v) solutions of mannose and fructose and sugar analyses were performed on shoots at different stages of storage and after 10 days in the vase. A slight increase in mannose and fructose was detectable in the stems of the shoots directly after pulsing but not in the leaves or the inflorescences. This is due to the low concentrations being used. The levels of all the carbohydrates decreased during the 21 days storage and more so during the vase phase of the flowering shoots. The fact that such low concentrations were effective in controlling chilling injury suggests that the sugars may have an effect other than on the osmotic potential. Cut ‘flower’ shoots of Leucadendron ‘Chameleon’, ‘Laurel Yellow’ and ‘Safari Sunset’ were stored for 14, 21 and 28 days, at 1º, 3º and 5ºC and CI development recorded during the subsequent 10 day vase phase. ‘Laurel Yellow’ and ‘Safari Sunset’ showed signs of chilling injury on the leaves after 28 days storage at 3ºC or lower and ‘Safari Sunset’ stored for 21 days developed chilling injury during the vase phase. Immature involucral leaves were more sensitive to chilling injury than leaves. CI increased with longer exposure times and lower storage temperatures for all three cultivars evaluated. ‘Chameleon’ was the most chilling tolerant of the cultivars up to 21 days. At 5ºC chilling injury was low irrespective of cold storage duration but longer exposures to 1º and 3ºC resulted in increased chilling injury development during the vase phase. All three cultivars were pulsed with 5 ml per stem of a 1% (w/v) solution of lactulose, sucrose, glucose, fructose or mannose and stored for 14, 21 and 28 days at 1ºC. The sugars reduced chilling injury on the leaves for ‘Safari Sunset’ when stored for 28 days and, to a lesser extent, in ‘Chameleon’. The sugars failed to reduce chilling injury of the involucral leaves of ‘Chameleon’ and ‘Laurel Yellow’ whereas there was some control especially after 28 days for ‘Safari Sunset’. In some cases the sugar pulse exacerbated chilling injury. Chilling injury generally increased rapidly after storage during the first three days in the vase and then at a lower rate for the next seven days. Leucadendron ‘Chameleon’, ‘Laurel Yellow’ and ‘Safari Sunset’ ‘cut flower’ shoots were pulsed with a 1% (w/v) glucose solution. Expressed on a dry weight basis, an increase in glucose concentration was not detected. The reduction in chilling injury of leaves by a sugar pulse is speculated, as for the Leucospermums, to be as a result of their presence in the apoplast and not the symplast and that their presence there protects the membranes against chilling conditions in some way.
AFRIKAANSE OPSOMMING: Die kwaliteit van sekere Leucospermum en Leucadendron kultivars na ongeveer 21 dae verskeping is waargeneem as substandaard as gevolg van die uitdroog van blare en, in die geval van Leucadendrons, die ‘involucral’ blare. Die aard van die simptome van hierdie uitdroging en die toestande waaronder dit plaasvind nl. lang periodes van blootstelling aan lae temperature, het ons tot die hipotese gebring dat hierdie simptome van koueskade is. Sover as wat ons bewus is, is koueskade nog nie gedokumenteer op Leucospermums of Leucadendrons nie. Tipiese koueskade simptomologie word bespreek en gewys vir Leucospermum ‘Gold Dust’, ‘High Gold’ en ‘Succession’ en vir Leucadendron ‘Chameleon’, ‘Laurel Yellow’ en ‘Safari Sunset’. Die beste beheer van koueskade vir beide kultivars is deur die 1.5 (g/v) oplossing behaal. Laer konsentrasies van mannose en fruktose is getoets op ‘High Gold’ stele met ‘n 1% (g/v) mannose oplossing wat die beste beheer gegee het. Met hoë konsentrasies het tekens van toksisiteit sigbaar geword direk na opneem van die oplossing. ‘High Gold’ stele is geplaas in 1% (g/v) oplossings van mannose of fruktose en suiker analises is uitgevoer op stele by verskillende stadiums van opberging en na 10 dae in die vaas. ‘n Effense toename in mannose en fruktose is waargeneem in die stele van die blomme direk na opname van die oplossing, maar nie in die blare of die blomme nie. Dit is as gevolg van die lae konsentrasies wat gebruik is. Die vlakke van al die koolhidrate het afgeneem gedurende die 21 dae opberging en nog meer so gedurende die vaas periode van die blommende stele. Die feit dat sulke lae konsentrasies effektief is in die beheer van koueskade dui daarop dat die suikers ‘n effek het anders as op die osmotiese potensiaal. Snyblomme van Leucadendron ‘Chameleon’, ‘Laurel Yellow’ en ‘Safari Sunset’ is opgeberg vir 14, 21 en 28 dae, by 1º, 3º en 5°C en koueskade ontwikkeling is aangeteken gedurende die opvolgende 10 dae vaas periode. ‘Laurel Yellow’ en ‘Safari Sunset’ het tekens gewys van koueskade op die blare na 28 dae opberging by 3°C of laer en ‘Safari Sunset’ opgeberg vir 21 dae het koueskade ontwikkel gedurende die vaas periode. Al drie kultivars is voorsien met 5ml per steel van ‘n 1% (g/v) oplossing van lactulose, sucrose, glucose, fruktose of mannose en opgeberg vir 14, 21 en 28 dae by 1°C. Die suikers het koueskade verminder op die blare van ‘Safari Sunset’ wanneer opgeberg vir 28 dae en, tot ‘n mindere mate, in ‘Chameleon’. Die suikers het egter nie koueskade verminder van die ‘involucral’ blare van ‘Chameleon’ en ‘Laurel Yellow’ nie, waar daar egter wel in ‘n mate beheer was veral na 28 dae vir ‘Safari Sunset’. In sommige gevalle het die voorsiening van suiker die koueskade vererger. Koueskade het oor die algemeen vinnig toegeneem na opberging gedurende die eerste drie dae in die vaas en dan teen ‘n laer tempo vir die volgende sewe dae. Leucadendron ‘Chameleon’, ‘Laurel Yellow’ en ‘Safari Sunset’ snyblom stele is voorsien van ‘n 1% (g/v) glukose oplossing. Uitgedruk op ‘n droëmassa basis is ‘n toename in glukose konsentrasie nie waargeneem nie. Die afname in koueskade van blare deur die voorsiening van ‘n suiker oplossing is gespekuleer vir die Leucospermums, om ‘n resultaat te wees van hulle teenwoordigheid in die apoplas en nie die simplas nie, en dat die teenwoordigheid daar die membrane op ‘n manier beskerm teen koue toestande.

Objetivou-se avaliar as melhores condições pós-colheita para frutos de mangaba. Os experimentos foram conduzidos no laboratório de Tecnologia de Alimentos da Universidade Estadual Paulista (UNESP) Campus de Ilha Solteira. No primeiro experimento, as mangabas nos três estádios de maturação foram acondicionadas em polietileno tereftalato, bandeja de Poliestireno expandido recoberto com filme de policloreto de vinila e saco plástico com fechamento. Os tratamentos foram constituídos das combinações entre os graus de maturação e embalagens e foram avaliadas: perda de massa fresca, firmeza, vitamina C, pH, sólidos solúveis, acidez titulável, índice de maturação, açúcares redutores e aparência visual. A melhor conservação foi nos frutos de vez acondicionados em bandeja de isopor, mantendo classificação ótima em todo período de armazenamento. No segundo experimento, mangabas no estádio de vez foram acondicionadas em bandejas de poliestireno expandido recobertas com filme de policloreto de vinila. Receberam os tratamentos com diferentes concentrações de quitosana (0; 0,25; 0,5; 1 e 2%). As avaliações realizadas foram: perda de massa fresca, firmeza, vitamina C, sólidos solúveis, acidez titulável, índice de maturação, pH, açúcar redutor e aparência visual. Os tratamentos com quitosana nas concentrações de 1% e 2% mostraram-se efetivos com notas máximas até o final do armazenamento. ENGLISH ABSTRACT: Fruit quality is the most important factor that determines prices for the fruit in the international markets. Although different consumers perceive quality differently there are quality variables that are always associated with poor quality by all consumers. In table grapes (Vitis vinifera L.) these variables may include overall appearance, stem condition, SO2 damage, decay, berry browning and shatter. The presence of these quality defects negatively affects prices and most often results in quality claims. Cooling is the most widely used method to reduce the postharvest loss of fruit quality. In South Africa, most deciduous fruits including table grapes are forced air cooled to a statutory pulp temperature of –0.5°C prior to shipping in an effort to preserve quality, thus ensuring good market prices for the fruits. Despite these efforts, there are still quality claims from the markets and this reduces the returns to the growers. The objectives of this research were to: (i) see if cooling time can be reduced by cooling to higher pulp temperatures of 1.5°C and 3°C without causing quality losses, thus improving the throughput of the cold rooms; (ii) see if the problem of berry browning can be alleviated by cooling grapes to higher pulp temperature, and (iii) see whether pallet positioning in the cooling tunnels and reefer container affect quality. The trends showed better quality when ‘Victoria’ and ‘Regal Seedless’ were forced air cooled (FAC) to pulp temperatures of 1.5 °C and 3 °C as opposed to –0.5 °C. There were no economic losses associated with pre-cooling grapes to pulp temperatures of 1.5 °C and 3 °C. There were no significant differences in berry browning related to pre-cooling treatments. However, cooling time was reduced significantly. In most of the cooling tunnels and reefer containers used in this trial, grape quality results showed no significant differences between the positions in the stack and in reefer containers. However, in cases where there were significant differences, the middle and the rear positions showed better grape quality in terms of stem condition (dry and brown stems) than the front position (near fan) in both the pre-cooling stack and reefer containers. The trends showed that the front is cooler than the back of the pre-cooling stack. The pulp temperature differences between the front and rear positions in the reefer container were as high as 1.23 °C. The trends also showed that the bottom layers of the pallets were cooler than the top layers in the reefer container. FAC to 3°C resulted in a constant reduction in percentage electrolyte leakage after 4 weeks of storage at –0.5°C, while FAC to 1.5°C, -0.5°C and static room cooling (control) in some cases showed an initially low electrolyte leakage followed by an increase in leakage after 4 weeks of storage. FAC grapes to higher pulp temperatures of 3°C and 1.5°C could reduce the cooling time, thereby improving the throughput of cold rooms. There was no clear evidence to suggest that browning was due to pre-cooling practices. Both preharvest and postharvest conditions need to be further investigated to better understand the problems of browning in white table grapes.
AFRIKAANSE OPSOMMING: Vrugkwaliteit is ʼn kritiese faktor in die bepaling van pryse op die internasionale markte. Alhoewel daar variasie voorkom tussen verbruikers in wat vrugkwaliteit is, bly sekere aspekte altyd onveranderd. Ononderhandelbare kwaliteit aspekte in tafeldruiwe (Vitis vinifera L.) sluit die algemene voorkoms, toestand van die trosstingels, SO2 skade, bederf, korrel verbruining en los-korrels in. Indien enige van die kwaliteit-defekte voorkom het dit ʼn negatiewe impak op die prys en lei gewoonlik tot gehalte eise. Verkoeling word algemeen gebruik om die verlies van na-oes kwaliteit te verminder. Die meeste sagtevrugte geproduseer in Suid Afrika (insluitend tafeldruiwe) ondergaan geforseerde verkoeling tot ʼn statutêre pulptemperatuur van -0.5°C, voor verskeping. Ondanks hierdie maatreëls om hoë pryse te verseker, is daar steeds kwaliteiteise in die mark wat lei tot ‘n laer inkomste vir produsente. Die navorsing het dus ten doel gehad om : (i) te bepaal of die tyd van verkoeling verminder kan word, indien na hoër pulptemperature van 1.5°C en 3°C verkoel kan word, sonder ‘n verlies in kwaliteit en sodoende die deurvloeitempo van die koelkamers verhoog; (ii) om te bepaal of die voorkoms van korrelverbruining verlaag kan word indien tot hoër pulp-temperature verkoel word, en (iii) laastens om te bepaal of posisie van die palet in die verkoelingstonnel en verskepingshouer ʼn invloed het op vrugkwaliteit. Tendense toon dat ‘Victoria’ en ‘Regal Seedless’ kwaliteit beter was indien verkoel tot pulptemperature van 1.5°C en 3°C in vergelyking met -0.5°C. Daar was geen ekonomiese verliese waargeneem indien die hoër verkoelingstemperature gebruik is nie. Alhoewel daar geen betekenisvolle verskille in korrelverbruining voorgekom het tussen temperatuur behandelings nie is die verkoelingsperiode verkort. In die meeste van die verskepingshouers, asook in posisies tydens geforseerde verkoeling is daar geen betekenisvolle verskille waargeneem nie. In die gevalle waar daar egter wel betekenisvolle verskille voorgekom het, het die middel en agter posisies beter vrugkwaliteit gehad as die voorste posisie tydens verkoeling asook houerverskeping. Die palette aan die voorkant (naby die waaier) het as ʼn algemene tendens laer temperature as in die agterkant van die verkoelingstonnel. Verskille in pulptemperature tussen palette in die voor en agterkant van verskepingshouers was so hoog as 1.23°C. Die temperatuurdata het uitgewys dat die onderste laag kartonne neig om by ‘n laer temperatuur te wees as die boonste lae kartonne tydens houerverskeping. Geforseerde verkoeling teen 3°C het gelei tot ‘n afname in persentasie elektrolietlekkasie na 4 weke van verkoeling teen -0.5°C. Terselfdertyd het geforseerde verkoeling tot 1.5°C en -0.5°C asook statiese verkoeling (kontrole) in sekere gevalle gelei tot ‘n laer aanvanklike uitlek van elektrolietlekkasie, gevolg deur ʼn verhoging na 4 weke opberging. Geforseerde verkoeling van tafeldruiwe tot pulptemperature van 1.5°C en 3°C verkort die verkoelingstyd en verhoog dus die deurvloeitempo in die verkoelingskamers. Daar was gedurende die studie geen duidelike bewyse gevind dat korrelverbruining voorkom as gevolg van verkoelingspraktyke nie. Beide voor en na-oes praktyke sal verder ondersoek moet word om die invloed daarvan te bepaal op die verbruining van wit tafeldruiwe.

Resumo: Botrytis cinerea Pers, causador da doença conhecida como mofo cinzento, é o principal problema para a conservação pós-colheita de uvas de mesa. A utilização do dióxido de enxofre (SO2) é a prática pós-colheita mais comum para o controle desta doença. Pesquisas buscam alternativas a este produto devido às reações que causa em pessoas alérgicas, danos que pode causar nos frutos e às restrições ao seu uso em sistemas de produção orgânico. Foram avaliados os efeitos da aplicação de uma atmosfera de 40% de CO2 por 24 ou 48 horas (pré-armazenagem) combinado com armazenagem em atmosfera controlada (AC) (12% O2 + 12% CO2) no controle de B. cinerea, e nos atributos de qualidade de uvas 'Flame Seedless' e 'Crimson Seedless'. Também foram avaliados, em uvas 'Crimson Seedless', e os efeitos da associação deste tratamentos com aplicações pré-colheita de cloreto de cálcio (CaCl2) ou dióxido de cloro (ClO2). Os tratamentos foram aplicados em uvas orgânicas infectadas de três formas: infectadas naturalmente, superficialmente inoculadas com conídios e inoculadas com uma baga coberta de micélio. Uvas 'Flame Seedless', naturalmente infectadas, tratadas com 40% de CO2 por 48 horas + AC apresentaram redução da podridão pós-colheita, de 22% para 0,6%, após 4 semanas, e de 100% para 7,4%, após 7 semanas. O pré-armazenamento em 40% de CO2 sozinho também limitou a incidência de mofo cinzento em frutos infectados naturalmente e em uvas inoculadas artificialmente, porém foi menos eficaz do que quando seguido pelo armazenamento em AC. A aplicação de CaCl2 ou ClO2 em pré-colheita reduziu a incidência do mofo cinzento em uvas 'Crimson Seedless' inoculadas com uma solução de conídios, porém não houve controle quando os cachos de uva foram inoculados com micélio. A aplicação de CaCl2 e ClO2 reduziram o mofo cinzento de 45% para 23,2% e 15.6%, respectivamente, em cachos inoculados com conídios e avaliados após 6 semanas armazenadas 0oC. O pré-tratamento com 40% CO2 + CA não afetou as características físico-químicas e sensoriais de uvas 'Crimson Seedless' ou 'Flame Seedless'. Em experimentos in vitro os tratamentos com 40% CO2 por 24 ou 48 h limitaram o crescimento micelial até 72 horas após o tratamento. A germinação dos conídios de B. cinerea foi retardada por 12h. Os resultados mostram que o pré-tratamento com 40% CO2 + CA possui grande potencial para ser adotado como prática comercial para conservação de uvas orgânicas
Abstract: Gray mold, caused by Botrytis cinerea Pers, is the main postharvest decay of table grapes. The use of sulfur dioxide (SO2) is the common post-harvest practice for its control. Several researchers are looking for alternative methods of control, because SO2 can cause allergic reactions, damage fruits and also it cannot be applied in organic production system. In this thesis, it was evaluated the effects of applying an atmosphere of 40% CO2 for 24 or 48 hours (pre-storage) combined with controlled atmosphere storage (CA = 12% O2 + 12% CO2) in the control of B. cinerea, and the effects in the quality and sensory attributes of 'Flame Seedless' and 'Crimson Seedless' table grapes. In addition, it was evaluated the efficacy of CaCl2 or ClO2 application in pre-harvest alone or in combination with pre-storage of 40% CO2 for 24 h + CA, to control gray mold on 'Crimson Seedless' table grapes, and the determination of the impact of these treatments on fruit quality. The treatments were applied in certified organic table grapes naturally infected, surface inoculated and nesting inoculated (inoculated with an infected berry). After 4 weeks of storage, the pre-storage in 40% CO2 for 48 hours + CA reduced postharvest rot from 22% to 0.6%, and after 7 weeks, the decay was reduced from 100% to 7.4% in 'Flame Seedless' naturally infected. The pre-storage in 40% CO2 alone also reduced the incidence of gray mold in fruits naturally infected and in artificially inoculated, but it was less effective than combined treatment. The application of CaCl2 or ClO2 pre-harvest reduced the incidence of gray mold on grapes 'Crimson Seedless' inoculated with a spore solution, but there was no control when fruits were nesting inoculated. After 6 weeks at 0oC, the application of CaCl2, and the ClO2 in fruits surface inoculated, reduced the gray mold from 45% to 23.2% and 15.6%, respectively. The pretreatment with 40% CO2 + CA did not affect quality and nor sensory attributes for both varieties tested. In vitro experiments, the treatment with 40% CO2 for 24 or 48 h limited mycelial growth for at least 72 hours after treatment. Conidial germination of B. cinerea was delayed for 12 hours. Our results showed the potential that pre-treatment with 40% CO2 associated with CA has to be adopted in commercial practice for preservation of organic grapes

Resumo: O morango é produzido e apreciado nas mais variadas regiões do mundo, sendo a espécie de maior expressão econômica entre as pequenas frutas. O Estado de São Paulo é um dos seus maiores produtores no Brasil. De todo o ciclo produtivo do morango, a comercialização tem se mostrado a fase mais complexa e problemática, podendo afetar mais o lucro do que todas as outras etapas de produção. Isso ocorre devido às características do produto, como fragilidade, manuseio inadequado, alta perecibilidade e falta de embalagens e de refrigeração adequados. Por possuir elevado valor comercial, principalmente no mercado in natura, a tecnologia do frio para sua melhor conservação pós-colheita, pode ser utilizado para manter as boas características de qualidade do fruto. Este trabalho teve por objetivo geral definir qual a melhor condição pós-colheita para conservação de morangos quanto a embalagem cumbucas de tereftalato de polietileno (PET) ou em meia cumbuca de tereftalato de polietileno, envoltas por filme de policloreto de vinila (PVC), resfriamento rápido e armazenamento refrigerado, visando seu consumo in natura. Realizaram-se dois experimentos, sendo um para monitoramento da taxa respiratória dos pseudofrutos a 0°C, 5°C, 10°C e 15°C em dois sistemas (sistema de fluxo contínuo e sistema estático). Outro experimento foi feito para avaliar a resposta da fruta expressa pelos parâmetros respiração, sólidos solúveis, pH e acidez titulável, quando estes foram submetidos a processos de resfriamento rápido e armazenagem. As variedades avaliadas foram Camarosa, Sweet Charlie, Dover e Oso Grande. Neste experimento utilizou-se câmaras de armazenamento com regulagem de 0 e 10°C, e os frutos foram embalados em cumbucas PET e PVC. As frutas foram analisadas a cada dois dias, até ficarem impróprias para o consumo. Verificou-se que o melhor método para avaliação da taxa respiratória é o método do sistema de fluxo contínuo, por possuir um fluxo constante durante todo o experimento, assim como melhor reprodutibilidade dos tempos de retenção e da quantificação do CO2 dentro dos frascos. A temperatura 0°C, foi a que possibilitou a maior vida de prateleira dos frutos. Não se observou diferença entre as embalagens PET e PVC no armazenamento a 0°C e a 10°C. Das variedades avaliadas a Sweet Charlie é a que se mostrou mais indicada para consumo in natura, devido suas características fisico-químicas. Enquanto a variedade Camarosa a mais indicada para indústria.
Abstract: The strawberry is cultivated and appreciated in several regions around the world, being the specie with the most economical value among the small fruits. The São Paulo state is one of the biggest producers in Brazil. From all productive cycle of strawberry the commercialization has shown the most complex and problematic phase, and it can affect more the profit than all the others. This happens due to the product characteristics, such as fragility, improper handling, high perishable, inadequate packaging, and use of cold chain. Since strawberry has high commercial value, mainly on in natura maket, the cold chain technology may be used to keep the good characteristics of fruit quality. The objectives were to define which are the best conditions postharvest for the strawberry fruits in terms of packing PET or PVC, precooling and refrigerated storage aiming the consumption in natura. Two experiments were realized, being one to monitoring and evaluating the respiratory rate of the fruits at the temperatures of 0, 5, 10 and 15°C in two different systems: continuous flow system, and static flow system, in BOD's. The other experiment was to evaluate the answer of biochemical parameters (respiration rate, soluble solids, pH and titratable acidity) submitted to the precooling and refrigerated processes using the Camarosa, Sweet Charlie, Dover, and Oso Grande varieties. For this experiment was used chambers at 0 and 10oC, and the fruits had been packed in packs of tereftalate of polyethylene (PET) clam shell type, and packs of tereftalate of polyethylene (PET) wrap with PVC film. Physical-chemical analyses were carried every two days until the fruits got improper for consumption. The experiment show that the optimum method for evaluation respiratory rate is the one using the continuous flow system since presented a constant flow during all the experiments, and allows better accuracy for the retention time and CO2 quantification inside of the bottles. The temperature of 0°C was the one that shows longest fruits shelf life time. There were not differences between packs of tereftalate of polyethylene (PET) clam shell type and packs of tereftalate of polyethylene (PET) wrap with PVC film storaged at 0 and 10°C. Among the varieties, the Sweet Charlie was the most indicated for in natura consumption, due to its physical-chemical characteristics. Meanwhile the variety Camarosa was the most appropriate for industry use.

Abstract:
Limes (Citrus latifolia Tanaka) are an attractive fruit crop but generally suffer a loss in value as their colour changes from green to yellow. Various approaches were taken to slow degreening including low temperature storage, use of controlled atmosphere (CA) environments, and treatment of fruit with physiologically active agents such as gibberellic acid (GA3). However, the cold storage life of lime fruit can also be restricted by a number of factors including chilling injury (CI) and rots. Various pretreatments such as the use of fungicide (thiabendazole, TBZ) and hot water dipping (HWD) and several postharvest regimes based on temperature conditioning (step down technique) and intermittent warming (IW) regimes were further investigated to protect the fruit against rots and CI during cold storage. The objective of this study was to determine what storage conditions and pretreatments would permit long term storage of NZ limes with minimal loss of quality. CA storage (10% O2 with 0 or 3% CO2) was compared to regular air storage (RA) and IW (varying durations) treatments across a range of temperatures. Although some CA storage regimes could assist in delaying degreening, none of the treatments provided protection against CI. CA storage at 3% CO2 delayed yellowing and gave better fruit quality than the low CO2 treatment. High CO2 CA treatments at 5 or 7°C decreased the rate of colour change compared to other constant temperature treatments but did not protect against CI. CI limited storage of fruit under all conditions at constant low temperatures. Including fungicide (TBZ) in the dip water reduced the incidence of rots and had a secondary effect on protection against CI of lime fruit. However, fungicide use may sometimes exacerbate stresses such as heat injury on lime peel. Hot water dipping has been shown previously to hold potential as a storage pretreatment, but this technique may give risk of damage on produce if it is dipped at too high a temperature. Some HWD treatments did delay degreening, but there was no major effect on CI. HWD at > 47°C for = 4 min caused heat injury to NZ limes. All HWD treatments showed severe CI (>15%) after 10 weeks of cold storage; and HWD fruit stored under RA at 13°C did not show any CI but showed some pitting (= 10%) and degreened rapidly. Overall no suitable HWD treatment for limes was identified in this trial. This project identified the critical periods and temperature conditions for successful IW of limes. The IW conditions successfully delayed losses in quality of lime fruit provided the first warming period was applied within the first 20 days of storage. At least 2-cycle IW was required to maintain lime quality during long term storage. Some benefits were found after just one cycle of IW treatment but there were not enough to extend storage life. IW storage benefited fruit quality and provided the highest overall fruit quality of all postharvest treatments tested. The degreening of lime during cold storage at 5°C could be delayed by IW treatments in which the fruit were stored at 5°C for 12, 16 or 20 days then moved to 15°C for 2 days. Both 2- and 6-cycle IW treatments proved satisfactory for maintaining colour on the green and yellow side of lime for 12 weeks of storage. IW treatments in which fruit were warmed within 20 day of cold storage did not show significant CI symptoms after 12 weeks of storage, and the 2-cycle IW treatment showed only a low percentage of CI fruit at this time. A 2-cycle IW treatment was almost as effective as 6 cycles, and a step down treatment also showed some promising results, indicating that it may be possible to further optimize the time and duration of variable temperature storage regimes to meet both quality requirements and the constraints of temperature management in commercial coolstores. The application of these regimes to other citrus species may also be beneficial. There are a number of physiological explanations that may account for the effectiveness of IW including positive effects on heat shock protein (HSP) and cell membranes. Nutritional factors such as vitamin C and flavonoid compositions were also investigated and fruit that did not show visible CI were found to retain at-harvest levels of these factors. Practical ways of implementing IW are discussed. In order to understand the effectiveness of IW on degreening, I used a logistic model to describe degreening of lime peel. This modelling approach demonstrated that IW did not change the mechanism of lime degreening based on the similarity between the hue values predicted by the model and the actual hue values measured during lime storage. The activation energy (Ea) for degreening based on either hue angle (H°) or colour score (CS) during air storage was estimated to be ~53 and ~86 KJ.mol-1, respectively. Relationship between colour (H° and CS) and chlorophyll content, relationship between reflectance spectra (%), chlorophyll content and H° of lime fruit stored under different conditions are presented and discussed. This data allowed deduction to be made about the changes in individual pigments that are driving colour change during “good” and “bad” storage.

ENGLISH ABSTRACT: Prototypes of a new polymer S02 gas-generating sheet for the control of Botrytis cinerea during the post-harvest storage of table grapes, were developed and manufactured for evaluation using a pilot scale production plant. Attention was paid to the appearance of the sheet, in order to make it technologically efficient as well as aesthetically acceptable to both industry and consumers. The storage quality of semi-commercial export consignments of various cultivars table grapes packed with the monolithic thin-film polymer S02 slow release sheet, was evaluated and compared to results obtained using the locally manufactured Uvasys S02 sheet. The following were investigated: the efficacy of the new polymer sheets in controlling storage decay, the stage at which S02 damage is manifested on table grapes, the level of S02 damage associated with different S02 concentrations, whether S02 damage is manifested more readily at a particular position on the bunch, and the possible effect of an increase in storage temperature, from an initial storage at -O.5°C to 10°C, on the levels of S02 bleaching. Results showed that the new polymer S02 sheet compared favourably with the existing, commercially available Uvasys S02 sheets. The exact S02 concentration required for effective decay control varied for different cultivars, as well as for the different types of grape packages. The S02 concentration incorporated within the sheet was shown to be lower for grapes packed in non-perforated bags, and slightly higher for those in perforated bags. Differences between cultivars occurred with regard to the level of control and the levels of S02 damage. Levels of S02 damage were also significantly affected by the storage period and temperature fluctuations. No significant differences in the levels of decay development and S02 damage were observed in relation to the orientation of the bunches in the carton. The extent of damage incurred to grape tissue by the absorption of S02 gas was determined by low-temperature scanning (LTSEM) and transmission electron microscopy (TEM) techniques. LTSEM and TEM micrographs of areas damaged by S02 gas revealed that exposure to S02 gas may lead to plasmolysis and the loss of cellular fluids. Although damage to the cell walls, cell wall structures and cell membranes, caused by S02 gas, was more prominent in the tissue layers nearer to the fruit surface, damage also occurred to a lesser extent in deeper tissue layers. S02 gas release-rate studies of polymer S02 sheets containing various concentrations Na2S205 revealed that levels of S02 gas emitted depended largely on the levels of Na2S205 incorporated into the sheets. Higher levels of S02 gas were released with the polymer sheets of higher concentrations Na2S205. The release curve for the commercial Uvasys S02 sheet was very different to that of the polymer sheets, with much higher levels of S02 gas emitted initially by the Uvasys S02 sheet compared to the polymer sheets, while the polymer sheets emitted low levels of S02 gas for longer periods compared to the Uvasys S02 sheet. The manufacturing process and the pilot scale production plant that was developed and constructed was successfully used to manufacture polymer S02 generating sheets that are technically sound and efficient, and aesthetically acceptable to industry. The efficacy of such sheets, regarding levels of decay control and S02 damage, was similar to that obtained with the presently available, commercially used Uvasys S02 sheet.

'n Nuwe loodsaanleg is spesiaal vir hierdie doel ontwerp en gebou. Aandag is geskenk aan die voorkoms van die velle aangesien dit belangrik is dat die nuwe velle beide tegnologies effektief en esteties aanvaarbaar moet wees vir die sagtevrugtebedryf en verbruikers. Die opbergingskwaliteit van semi-kommersiële uitvoerbesendings van verskeie kultivars tafeldruiwe, verpak met die nuwe monolitiese S02- gasvrystellingsvelle, is bepaal. Alhoewel S02-skade aan die selwande en membrane meer prominent in die weefsel naby die oppervlak van die vrug was, het skade ook in die onderliggende lae plaasgevind. Die vlakke van vrygestelde S02-gas het grootendeels afgehang van die konsentrasie natriummetabisulfiet in die velle. Die S02-vrystellingskurwe van die nuut ontwikkelde polimeriese S02-velle het baie verskil van dié van die Uvasys vel. Laasgenoemde lewer aanvanklik 'n hoë konsentrasie vrygestelde S02-gas vir 'n kort periode, gevolg deur baie lae S02 vlakke daarna, terwyl eersgenoemde 'n laer aanvanklike S02 vrystelling het, gevolg deur vergelykederwys hoër S02 konsentrasies daarna. Die ontwikkelde vervaardigingsproses en die loodsaanleg wat daaruit voortgevloei het is dus suksesvol aangewend om goeie polimeriese S02- vrystellingsvelle te vervaardig. Hierdie velle is tegnies effektief vir die beheer van Bofrifis cinerea gedurende die na-oes verpakking van tafeldruiwe en is esteties aanvaarbaar vir die Suid Afrikaanse sagtevrugtebedryf.

ENGLISH ABSTRACT: The export of Japanese plums from South Africa is challenging, since most cultivars are prone to develop chilling injury (CI) when stored at low temperatures. This injury manifests as gel breakdown or internal browning in the mesocarp tissue of the fruit on removal from low storage temperature conditions, i.e. in the consumer’s fruit basket, who subsequently does not buy plums again. Loss of cell membrane integrity and oxidative stress are, respectively, the primary and secondary physiological responses to CI. The main aim of this study was to investigate changes in cell membrane composition and levels of antioxidants in plums throughout fruit development and maturation, during forced air cooling (FAC) and storage under different temperature regimes. ‘Sapphire’ (a chilling susceptible cultivar) accumulated high levels of glutathione and polyunsaturated fatty acids (PUFAs) during fruit development. Therefore, the cultivar is protected against lipid peroxidation while developing on the tree, but the high levels of PUFAs, which are easily oxidised, may cause this cultivar to be chilling susceptible when stored at low temperatures. It is suggested that the high levels of monounsaturated fatty acids (MUFAs), which are not easily oxidised, and ascorbic acid that accumulated in ‘Angeleno’ (a chilling resistant cultivar) during fruit development, render this cultivar CI resistant during long-term cold-storage. When stored at -0.5 °C, CI development increased at a higher rate, ethylene evolution rates were higher and water soluble antioxidant activity (HAA), ascorbic acid and glutathione levels, and the MUFA:PUFA ratio were lower in H2 (more mature) ‘Sapphire’ plums than H1 fruit (less mature). Therefore, concurrent with H2 fruit having lower levels of antioxidants to quench free radicals caused by chilling stress, their cell membranes were more vulnerable to oxidation due to their phospholipid fatty acid composition. H2 fruit also had higher levels of saturated fatty acids, and hence less fluid cell membranes than H1 fruit when stored at -0.5 °C. An intermittent warming (IW) regime delayed symptom appearance and reduced CI severity in plums significantly compared to storage at -0.5 °C. Fruit stored under the IW regime had a more optimal phospholipid fatty acid composition and lower membrane sterol levels under shelf-life conditions to keep the membranes fluid. It also had higher levels of HAA and lipid soluble antioxidant activity, ascorbic acid and glutathione, which rendered fruit better protected against oxidation. Elevated storage temperatures (2.5 °C to 7.5 °C) caused higher levels of lipid peroxidation or low ascorbic acid levels and poor fruit quality compared to the IW regime in ‘Sapphire’ plums. ‘Laetitia’ plums stored at 5 °C and 7.5 °C had significantly less CI than under the IW regime, but softened quicker due to higher ethylene evolution rates. ‘Sapphire’ tolerated both long and short FAC durations, but a slower initial FAC rate prevented CI manifestation and caused a higher HAA after cold-storage in this fruit. ‘Laetitia’ cooled with a slower initial FAC rate and for a longer duration resulted in the best fruit quality and had higher HAA, total phenolic, phospholipid and saturated phospholipid fatty acid concentrations during storage.
AFRIKAANSE OPSOMMING: Die uitvoer van Japanese pruime vanaf Suid-Afrika hou talle uitdagings in, want die meeste van die kultivars ontwikkel koueskade wanneer hulle by lae temperature opgeberg word. Koueskade manifesteer as gelverval of interne verbruining in die mesokarpweefsel van die vrugte wanneer die vrugte verwyder word uit die lae opbergingstemperatuuromstandighede, m.a.w. in die verbruiker se vrugtemandjie, wat nie weer pruime koop nie. Verlies aan selmembraanintegriteit en oksidatiewe druk is, respektiewelik, die primêre and sekondêre fisiologiese reaksies op koueskade. Die hoofdoel van hierdie studie was om die veranderinge in selmembraansamestelling en antioksidantkonsentrasie in pruime te ondersoek tydens vrugontwikkeling en volwassewording, tydens geforseerde lugverkoeling (GLV) en tydens opberging onder verskillende temperatuurregimes. ‘Sapphire’ (‘n koueskade sensitiewe kultivar) het hoër konsentrasies van glutatioon en polionversadigde vetsure (POV) tydens vrugontwikkeling geakkumuleer. Dié kultivar is dus voldoende beskerm teen lipiedperoksidasie tydens vrugontwikkeling aan die boom, maar die hoë konsentrasies van POVs, wat maklik oksideer, mag veroorsaak dat hierdie kultivar meer koueskadesensitief is wanneer dit by lae temperature opgeberg word. Die hoë konsentrasies van mono-onversadigde vetsure (MOV), wat nie maklik oksideer nie, en askorbiensuur wat in ‘Angeleno’ (‘n koueskade weerstandbiedende kultivar) geakkumuleer het tydens vrugontwikkeling, verleen moontlik weerstandbiedendheid teen koueskade aan hierdie kultivar tydens langtermyn koelkopbering. Tydens opberging by -0.5 °C het koueskade ontwikkeling vinniger toegeneem, was etileenvrystellingstempos hoër en die wateroplosbare antioksidantaktiwiteit (HAA), askorbiensuuren glutatioonkonsentrasies en die MOV:POV verhouding laer in H2 (meer volwasse) ‘Sapphire’ pruime as in die H1 vrugte (minder volwasse). Dus, tesame met die laer antioksidantkonsentrasies in die H2 vrugte om die vry radikale veroorsaak deur koelopbering te verminder, was hul selmembrane ook meer vatbaar vir oksidasie a.g.v. die vetsuursamestellling van hul membraanfosfolipiede. Die H2 vrugte het ook ‘n hoër konsentrasie van versadigde vetsure, en dus minder vloeibare membrane as die H1 vrugte gehad tydens opberging by -0.5 °C. Die dubbeltemperatuurregime (DT) het simptoomontwikkeling vertraag en koueskade-intensiteit betekenisvol verminder in vergelyking met pruime wat by -0.5 °C opgeberg is. Vrugte wat met die DT regime opgeberg is, het ‘n meer optimale fosfolipiedvetsuursamestelling en laer konsentrasie van membraansterole tydens gesimuleerde raklewe gehad wat meer vloeibare membrane verseker het. Hierdie behandeling het ook hoër HAA en lipiedoplosbare antioksidantaktiwiteit (LAA), askorbiensuur- en glutatioonkonsentrasies gehad wat die vrugte beskerm het teen oksidatiewe druk. Verhoogde opbergingstemperature het hoër vlakke van lipiedperoksidasie of lae askorbiensuurkonsentrasies asook swak vrugkwaliteit in ‘Sapphire’ pruime veroorsaak in vergelyking met die DT regime. ‘Laetitia’ pruime wat by 5 °C en 7.5 °C opgeberg is, het betekenisvol minder koueskade gehad in vergelyking met die DT regime, maar het vinniger sag geword a.g.v. hoër etileenvrystellingstempos. ‘Sapphire’ kon lang en kort GLV tye weerstaan, maar ‘n stadiger inisiële GLV spoed het die manifestasie van koueskade voorkom en het ‘n hoër HAA in die vrugte tot gevolg gehad na koelopberging. ‘Laetitia’ wat met ‘n stadiger inisiële GLV spoed en oor ‘n langer tyd verkoel is, het die beste vrugkwaliteit, en hoër HAA, totale fenool-, fosfolipied- en versadigde fosfolipiedvetsuurkonsentrasies as die ander behandelings tydens koelopberging gehad

Resumo: No Brasil o uso terapêutico de plantas medicinais está cada vez mais crescente. Várias empresas nacionais e internacionais empregam matéria-prima vegetal na elaboração de seus produtos, o que torna indispensável o cultivo comercial e o beneficiamento pós-colheita destas espécies. O objetivo deste trabalho foi estudo pós-colheita de duas espécies medicinais de importância econômica, a calêndula (Calendula officinalis L.) e a carqueja (Baccharis genistelloides var. trimera (Less). Baker var. trimera (Less). Backer). Nas duas espécies foi avaliado o melhor processo de secagem, utilizando três diferentes secadores, secador convectivo vertical, secador a gás e estufa com circulação forçada de ar, em três diferentes temperaturas, 30, 45 e 60°C, bem como o armazenamento das mesmas em três embalagens diferentes, polietileno de baixa densidade, vidro e papel kraft. Os processos de secagem foram avaliados em função do teor de óleo essencial de cada espécie e sua composição química. As isotermas e as curvas de secagem foram levantadas e tratadas com os modelos matemáticos, Bet, Bet linear, Peleg. Gab, Hasley, Lagmuir, Oswin, Henderson e Chung. As diferentes temperaturas não interferiram nos parâmetros avaliados, quanto ao óleo de calêndula, em todos os secadores, mas diferiram entre os diferentes secadores utilizados, sendo que, entre os secadores o que manteve os maiores teores de óleo de calêndula foi o secador convectivo vertical. No caso da carqueja, não houve diferenças relacionadas com as temperaturas e nem com os tipos de secadores. Ao longo do armazenamento, a embalagem Kraft foi a que melhor preservou a integridade física e química do produto, assim como, manteve o teor de óleo ao longo de um ano de armazenamento nas duas espécies
Abstract: In Brazil, the therapeutical use of medicinal plants is increasing each time. Some national and international companies use vegetal raw material in the elaboration of its products, so the commercial cultivation and the improvement of harvest of these species becomes indispensable. The objective of this work was to study post-harvest of two medicinal species with economic importance: calendula (Calendula officinalis L.) and carqueja (Baccharis genistelloides var. trimera (Less). Baker). In these two species the best process of drying was evaluated, using three different dryers, convective vertical dryer, the gas dryer, and oven with forced air circulation, with three different temperatures, 30, 45 and 60°C, as well as using three different packing materials: polyethylene of low density, glass and kraft paper. The drying processes was evaluated in relation of the essential oil and its chemical composition of species. The isotherms and the drying curves were obtained and treated with mathematical model, Bet, Bet linear, Peleg. Gab, Hasley, Lagmuir, Oswin, Henderson e Chung. The differents temperatures did not show the difference in the process of drying in calendula in each different driers, but they differed among the driers, and the drier that kept more oil content of calendula was the convective vertical dryer. In the case of carqueja, no differences were found in the different temperatures or the different driers used in drying. During the storage, the kraft packing material was the one that better preserved the physical and chemical integrity of the product, as well as, it kept the oil content throughout one year of storage time for these two species

Abstract:
The purpose of this thesis was to research some innovative post-harvest techniques with potential to extend the shelf-life of berry fruit during storage and enhance their overall quality. Emphasis was given to different packaging system, in particular microperforated film, edible film, and electrostatic spraying of antimicrobial coating. The berry fruits studied were blueberry and strawberry. These fruits were selected because of their increasing worldwide consumption, among the many different types of berries. The application of microperforated polypropylene film to store freshly harvested blueberries, demonstrated the importance of selecting a proper packaging to increase the storage period and maintain the quality attributes of the fruit. The number of microperforations made to the film (1, 10, and 30) created a passive modified atmosphere within the package that led to improvement of firmness and antioxidant activity of the fruit, throughout their storage period. Equilibrium atmosphere within the packaging was achieved after two days of storage and the concentration of gases remained constant for the rest of the storage period. This stable atmospheric condition resulted in an extension of the shelf-life of blueberry for up to 16 days, at 4°C. Strawberry shelf-life extension and quality improvements were obtained by storing the fruit in clamshells that contained strawberry puree edible films infused with carvacrol and methyl cinnamate. The strawberry puree edible films served as matrix for the controlled release of natural antimicrobial’s (Carvacrol and methyl cinnamate) vapors, over time. Fresh strawberries packed in clamshells had an extended shelf life of 10 days, at 10◦C and 90% RH. The released vapors from the strawberry puree edible film extended the strawberry shelf-life by delaying spoilage of the fruit and improved main quality-related attributes, such as firmness and brightness. The natural antimicrobial vapors also increased the total soluble phenolic content and antioxidant activity of the stored strawberry. Besides microbial spoilage, that limits strawberry shelf-life, the presence of foodborne pathogens bacteria that can be carried by the fruit as result of contamination, is another important issue that can lead to serious outbreaks. Antimicrobial edible coatings can be an effective post-harvest technique to assure microbial safety and, at the same time, retain overall quality of the fruit. Antimicrobial alginate coating was developed and optimized using response surface methodology. Antimicrobial activity against Escherichia coli O157:H7 and Botrytis cinerea, as well as physical properties such as viscosity, turbidity and whitish index of the coating were also optimized based on carvacrol and methyl cinnamate concentration. After the optimization, the resulted antimicrobial coating solution was applied on the surface of freshly harvested strawberry fruit using an electrostatic spraying technology. This technology presented unique advantage with regard to transfer efficiency and evenness application of the antimicrobial coating solution. The antimicrobial coating application led to a significant increase on strawberry shelf-life. Additionally, the strawberry coated using the electrostatic spraying technology presented a significant reduction of visible decay of four days compared to not-coated fruit, and of one day compared to fruit coated with conventional spray method. Moreover, the firmness and color of the strawberries were improved by coating the fruit using the electrostatic spraying technology. Therefore, electrostatic spraying could be considered a potential technology for the commercial application of liquid coating to extend the shelf-life and improve the post-harvest quality of strawberry and other perishable fruits.
Questa tesi è stata volta alla ricerca di alcune tecniche innovative da adottare nel post-raccolta per la conservazione di piccoli frutti, al fine di allungarne la shelf-life e, allo stesso tempo, migliorarne le caratteristiche qualitative. Particolare attenzione è stata posta allo sviluppo e utilizzo di diversi tipi di confezionamento (packaging), in particolare film micro perforati, film edibili, e coating edibili applicati mediante tecnologia spray elettrostatica. Fragole (Fragaria x ananassa) e mirtilli (Vaccinium), tra i diversi tipi di piccoli frutti disponibili sul mercato, sono stati selezionati come modelli di studio a causa del loro crescente consumo globale e importanza economica. Nel primo studio, film in polipropilene micro perforati sono stati utilizzati per la conservazione di mirtilli freschi. La selezione di tali film ha evidenziato l’importanza di utilizzare un packaging opportuno (in base alle caratteristiche ambientali e del prodotto) al fine di allungare il periodo di conservazione del frutto mantenendo allo stesso tempo ottimali caratteristiche qualitative. Le tre diverse intensità di micro perforazioni (1, 10, e 30, create mediante tecnologia laser e dal diametro di 0,1 mm) hanno originato un’ atmosfera modificata passiva all’interno della confezione dovuta alla respirazione del frutto stesso. Tale atmosfera ha causato un miglioramento della consistenza dei mirtilli e un sostanziale incremento dell’attività antiossidasica durante tutto il periodo di confezionamento. All’interno delle confezioni l’equilibrio atmosferico, tra ossigeno consumato ed anidride carbonica prodotta durante il processo di respirazione, è stato ottenuto dopo due giorni di conservazione e mantenuto tale per tutto il periodo di conservazione. Questa particolare condizione di stabilità atmosferica ha determinato un allungamento della shelf-life dei mirtilli fono a 16 giorni a 4 °C. Il secondo studio ha visto l’ottenimento dell’estensione della shelf-life e il miglioramento delle caratteristiche qualitative di fragole fresche, utilizzando delle confezioni commerciali in cui erano stati applicati film edibili. Tali film ottenuti da purè di fragole erano stati arricchiti con carvacrolo e metile cinammato, due oli essenziali naturali dal potere antimicrobico. Il film edibile, fungendo da vettore, ha permesso il rilascio graduale dei vapori degli oli essenziali nel tempo. La shelf-life delle fragole conservate con questo packaging a 10 °C e con il 90% di umidità relativa è aumentata fino a 10 giorni, grazie al potere antimicrobico dei vapori degli oli essenziali che hanno ritardato lo sviluppo microbico sulla superficie del frutto, di conseguenza aumentando anche alcune caratteristiche qualitative come ad esempio la brillantezza e la compattezza. Anche la capacità antiossidasica totale e il contenuto di fenoli è aumentato a causa dell’effetto dei vapori di carvacrolo e metile cinnamato. Lo sviluppo microbico, e in particolare di muffe, è solo uno degli aspetti che limitano il consumo di fragole fresche. Infatti, la presenza di microorganismi patogeni di origine alimentare è molto diffusa soprattutto su prodotti freschi e deperibili come le fragole che non vengono lavate dopo la raccolta e possono fungere da vettore di tali batteri patogeni e causare serie epidemie. Considerando tale problematica, l’utilizzo di coating edibili con azione antimicrobica (direttamente applicati sulla superficie del frutto) possono essere considerati una valida soluzione per garantire sicurezza microbiologica e allo stesso tempo per mantenere inalterate le caratteristiche qualitative del frutto. L’attenzione del terzo studio è stata rivolta allo sviluppo e ottimizzazione di un coating antimicrobico di alginato di sodio utilizzando una collezione di metodi statistici e matematici definiti come response surface methodology (RSM). L’attività antimicrobica nei confronti di Escherichia coli O157:H7 e Botrytis cinerea, e alcune caratteristiche fisiche della soluzione, come ad esempio la viscosità, turbidità e indice del bianco (Wi) sono stati ottimizzati sulla base delle concentrazioni dei componenti antimicrobici selezionati (carvacrolo e metile cinnamato). Dopo l’ottimizzazione e lo sviluppo del coating di alginato, lo stesso coating è stato applicato su fragole fresche utilizzando la tecnologia spray elettrostatica caratterizzata dal vantaggio di creare maggiore resa e uniformità di coating rispetto ad una comune tecnologia spray. L’applicazione di tale coating elettrostatico ha reso possibile una estensione della shelf-life delle fragole trattate e una significativa e visibile riduzione di muffe presenti sui frutti pari a quattro giorni di ritardo quando paragonato alle fragole non trattate e un giorno nei confronti delle fragole trattate con una comune tecnologia spray. Inoltre, la compattezza e il colore delle fragole è migliorata durante il periodo di conservazione quando il metodo elettrostatico è stato utilizzato.

Resumo: Esta pesquisa teve por objetivo utilizar diferentes doses de irradiação gama sobre frutos de mamão 'Formosa' (Carica papaya L.) pré-cortados e refrigerados, visando facilitar as etapas do processamento mínimo e o consumo, além de encontrar a dose ideal de irradiação que proporcione total segurança alimentar com relação aos microrganismos, ampliando, desta forma, o período de vida útil dos frutos e mantendo as características de qualidade. Todo o processamento do mamão até a embalagem foi executado na Empresa Verde Limpo, situada em Botucatu/SP. Os frutos já embalados foram irradiados na Companhia Brasileira de Esterilização (CBE), em Jarinú/SP. As doses aplicadas foram 0,0; 0,2; 0,4; 0,6; 0,8 e 1,0kGy. Após a irradiação os frutos minimamente processados foram armazenados à temperatura de 9 l 1ºC e umidade relativa de 90 l 5% durante nove dias. As avaliações foram a cada dois dias, exceto as análises microbiológicas que foram nos 1º, 5º e 9º dias de conservação. As alterações na qualidade pós-colheita foram detectadas por meio das seguintes análises: perda de massa fresca (PMF), aparência, textura, acidez titulável (AT), pH, sólidos solúveis (SS), vitamina C, açúcares totais (A.Totais) e redutores (AR), sacarose (S), pectinas total (PT) e solúvel (PS), enzimas poligalacturonase (PG) e pectina metilesterase (PME), análise sensorial (cor, aroma, textura e sabor), índice de doenças e conservação pós-colheita. Abstract: The objective of this research was to evaluate the influence of gamma irradiation on 'Formosa' papaya fresh-cut fruits (Carica papaya L.), stored under refrigeration, in order to facilitate the minimal processing stage and human consumption, finding ideal dose of irradiation that provides total nutritional safety against microorganisms, enlarging fruit shelf life and sustaining quality characteristics. Processing till packing of papaya was executed in Verde Limpo Enterprise, situated on Botucatu/SP. Packing fruits were irradiated on Companhia Brasileira de Irradiação (CBE), placed at Jarinú/SP. Used dose were 0,0; 0.2; 0.4; 0.6; 0.8; and 1.0kGy. After irradiation, the fruits were stored under temperatures of 48.2ºF l 33.8ºF and relative humidity of 90 l 5%, during 9 days and appraised each 2 days. Evaluations were performed each two days, except microbiological analysis, carried up on 1st, 5th and 9th days of conservation. Changes in postharvest quality were detected in the following analysis: loss of fresh weight (LFW), appearance, firmness, titratable acidity (TA), pH, total soluble solids (TSS), vitamin C, total reducer sugars (TRS), reducer sugar (RS), sucrose (S), total pectin (TP), soluble pectin (SP), activity of polygalacturonase (aPG) and pectin methyl esterase (aPME), sensorial analysis (color, aroma, firmness and flavor), disease index and postharvest conservation. Microbiological analysis was performed to identify aerobic bacteria (mesophilic and psychrotrophic), fungi (yeasts and moulds) and coliforms (total and fecal). Data were analyzed according to a completely randomized design, with 4 repetitions per treatment, using SISVAR™ program and Scott-Knottþs test at 5% level of significance. The objective of this research was to evaluate the influence of gamma irradiation on 'Formosa' papaya fresh-cut fruits (Carica papaya L.), stored under refrigeration, in order to facilitate the minimal processing stage and human consumption, finding ideal dose of irradiation that provides total nutritional safety against microorganisms, enlarging fruit shelf life and sustaining quality characteristics. Processing till packing of papaya was executed in Verde Limpo Enterprise, situated on Botucatu/SP. Packing fruits were irradiated on Companhia Brasileira de Irradiação (CBE), placed at Jarinú/SP. Used dose were 0,0; 0.2; 0.4; 0.6; 0.8; and 1.0kGy. After irradiation, the fruits were stored under temperatures of 48.2ºF l 33.8ºF and relative humidity of 90 l 5%, during 9 days and appraised each 2 days. Evaluations were performed each two days, except microbiological analysis, carried up on 1st, 5th and 9th days of conservation. Changes in postharvest quality were detected in the following analysis: loss of fresh weight (LFW), appearance, firmness, titratable acidity (TA), pH, total soluble solids (TSS), vitamin C, total reducer sugars (TRS), reducer sugar (RS), sucrose (S), total pectin (TP), soluble pectin (SP), activity of polygalacturonase (aPG) and pectin methyl esterase (aPME), sensorial analysis (color, aroma, firmness and flavor), disease index and postharvest conservation. Microbiological analysis was performed to identify aerobic bacteria (mesophilic and psychrotrophic), fungi (yeasts and moulds) and coliforms (total and fecal). Data were analyzed according to a completely randomized design, with 4 repetitions per treatment, using SISVAR™ program and Scott-Knottþs test at 5% level of significance. The main objective of this thesis was to evaluate the influence of the pre-conditioning period prior to cold storage at -0.5 ° C and the effect of different atmosphere gas combinations during cold storage and subsequent shelf-life (20ºC) conditions, on the production of volatile compounds, physicochemical parameters and sensory evaluation in four peach varieties (Early Rich ®, Royal ® Glory, Sweet Dreamcov and Elegant Lady ®) and seven nectarine varieties (Big Top ®, Honey Blazecov, Venus ®, Honey Royalecov, August Red ®, and Nectaladycov Nectagalacov). Accordingly, four different experiments were carried out over the years 2009, 2010 and 2011 using different fruit varieties. In this research we investigated the postharvest technology of Star-of-Bethlehem cut flower (Ornithogalum saundersiae Bak.) The length of the stem of Star-of-Bethlehem cut flower ranges from 70 cm to more than 100 cm, with a corymb at the top consisting of 30 to 40 florets. When it was held in a vase at 25 , the inflorescence opened at ℃ the rate of 1~5 florets per day. Average life time of each floret is about 7 days. The inflorescence forms a perfect flower ball after 7 days, and the vase life can last about two weeks. The respiration rate of cut flower was the highest at 25℃. It is about 80mg CO2 kg-1hr-1 and decreased with the storage temperature. At 0℃, the respiration rate reached the lowest point of 20mg CO2 kg-1hr-1. The ethylene production was 1.5µl kg-1hr-1 at 25℃. Maximum production occurred at the 5th day and decreased with time. Below 15℃, ethylene production was about 0.5µl kg-1hr-1. When we treated the cut flowers with 1ppm ethylene, the total number of florets increased and the vase life was extended in comparison to that of the control group. On the other hand, 10ppm ethylene had the similar effects on the low mature florets but not on the high mature florets. In addition, ethylene treatment makes calyx of cut flower wilted and lowers the appreciation value. At low storage temperature, the cut flowers without plastic wrapping on the upper portion had greater total number of florets and longer vase life as opposed to the group with wrapping. Throughout the storing process, the inflorescence was kept dry and less likely to mold. It is considered as a promising storage method. With less than three weeks of the storage time, the flowers held at 0 had the longest vase life and the most opened florets. However, florets ℃ at the lower position had elongated pedicels and thus the flower assumed the shape of a cup rather than a ball. Flowers stored at 5 and 10 had ℃ ℃ better outcome. Flowers stored at 5 for three weeks had shorter vase ℃ life, yet they produced superior appearance. In the storage condition of 5 and during the three week period, the flowers had slightly less ℃ opened florets than that of 10 group, but looked solid and exquisite. ℃ When the flowers was held at 15 , its floret stalk elongated unevenly ℃ during the storage period, and there were nectar secreted at the base of the floret, which reduced its quality of decoration. In conclusion, 5 was the ℃ most suitable temperature for Star-of-Bethlehem cut flower to be stored. The pulsing treatment process with 5% sucrose solution+ 8HQS+STS had a better performance in increasing the total number of florets and the vase life time. But the calyx were observed to be significantly yellow and wilted. BA treatment can help increase the total number of florets and ease the problem of yellowing. During the blooming process of the cut flowers, glucose and fructose are the major components of the solvable sugar in the stem. Sucrose is hardly detectable, and the amount of glucose decreases with time.

Avocados are an important horticultural crop in South Africa, especially in the provinces of KwaZulu-Natal, Mpumalanga and Limpopo. The distance to traditional export markets, phytosanitary restrictions to lucrative markets such as China, the USA and Japan and increased competition in the European market have challenged the South African avocado industry. The industry has responded with improved logistics and shipping, a co-ordinated market access program and a global system to co-ordinate exports of avocados to the European market. To remain competitive on the global market, further improvements and innovations are required to improve the efficiency of postharvest operations. These improvements and innovations should be guided by a greater understanding of postharvest physiology. Avocados are a relatively new export crop, so there is still much to be learnt about avocado postharvest physiology and the optimisation of postharvest management. In this regard, reduced temperature storage (1°C) and modified humidity packaging (MHP) were investigated for their effect on fruit physiology and quality, the effect of a water- and ABA-infusion on ripening was examined and the effect of a cold chain break on fruit physiology and quality determined; near-infrared spectroscopy was also examined for its potential for its use in the avocado industry. As an initial study, the relationships between individual sugars, protein and oil were studied to understand the changes in avocado fruit during ripening. It was found that mannoheptulose and perseitol were the predominant sugars at harvest, but declined to very low levels during the first 10 days postharvest. The concentrations of glucose and fructose increased, while sucrose declined slightly during ripening. The concentration of protein increased sigmoidally during ripening, reflecting the increase in the ripening enzymes, particularly cellulase and polygalacturonase. The oil content fluctuated slightly during ripening. It is suggested that mannoheptulose and perseitol are important carbon and energy sources during ripening. Glucose concentration was also found to increase earlier in fast ripening fruit compared to slow ripening fruit, which is related to increased cellulase activity and may be related to the ABA functioning. Thereafter, storage and ripening trials in two consecutive seasons showed that 1°C storage and the use of MHP for 28 days reduced mass loss, water loss, ethylene production, respiration, softening and heptose consumption, without appreciably affecting fungal rots, physiological disorders or external chilling injury, compared to fruit stored at 5.5°C and regular atmosphere respectively. Also, the storage of fruit in MHP delayed the rise in the activity of cellulase during ripening, compared to fruit not stored in MHP, but there was no significant difference in the peak activity of cellulase, polygalacturonase or pectin methylesterase. In a separate experiment, fruit ripening was significantly affected by the infusion of ABA in an aqueous solution. Water slightly reduced the variation in ripening while ABA reduced the time to ripening and the variation; it is suggested that water stress and ABA are intrinsically involved in the ripening processes and may act as a ripening trigger. The water concentration in fruit was measured non-destructively using reflectance NIR; this model was used to determine the maturity of fruit and the loss of water during cold storage. In the cold chain break experiment, it was found that although fruit recovered after a cold chain break, in terms of ethylene production and respiration, there was a loss in quality because of severe shrivelling as a result of increased water loss. Fruit that were stored at 1°C were generally of a better quality at ripeness, if the cold chain was broken, compared to fruit stored at 5.5°C. In a follow-up experiment, it was found that significant changes occurred in avocado physiology over a 6h period. The respiration rate of fruit significantly increased after 4h at room temperature and mannoheptulose declined by 32% in control fruit and by 16% in ethephon-treated fruit after 6h. This demonstrates the potential for quality loss in a short amount of time. Furthermore, a model of avocado ripening is proposed, outlining the role of water, ABA, ethylene, respiration, ripening enzymes and individual sugars. This study has contributed to the understanding of avocado postharvest physiology and should aid in better management of avocados for improved fruit quality and consumer satisfaction.

This study was focused on postharvest physiology and technology of welsh onion (Allium fistulosum L.) at Sunshin area in Yilan County. The welsh onion of naked, the open polyethylene packaging and paper storage at 1, 5, 10 and 15℃. under different temperatures weight-loss all take naked as high, the paper bag and the polyethylene bag packaging is next. The different packaging on decay and the yellowing index of welsh onion storage at 1℃ and 5℃ is low. The DPPH radical scavenging activity and superoxide anion scavenging activity to be highest by fresh, however along with stores the number of days to increase reduces gradually. The ferrous ion chelating ability take fresh as low, after but stores the second day rise to present impartially. The reducing power under the different packing and the temperature, in stores the 4th day reducing power rise to drop also slowly.   The welsh onion by naked, the open polyethylene bag packaging, sealed in polyethylene bag and the sealed polyethylene bag containing ethylene absorbent packaging to storage at 5℃ for 3 weeks. The sealed polyethylene bag and the sealed polyethylene bag containing ethylene absorbent packaging on weightloss, yellowing and decay index is all low. Studies on cultivers of welsh onion to use ‘da-han’、‘er-han’、‘hei-she’ and ‘hua-shiou’ in 2007 and the welsh onion in 2008 take cultivers ‘da-han’、‘er-han’、‘hei-she’ and ‘mei-shiou’. The to use naked or open polyethylene bag packaging of welsh onion storage at 1, 10 and 20℃.Two year test results showed that, take storage at 1℃ the open polyethylene bag packaging ‘da-han’ and ‘hei-she’ weightloss as low, ‘da-han’ and ‘hei-she’ at 1℃ has the low yellowing index, the decay index take ‘da-han’ as low. The respiration rate to exhibit are low with ‘da-han’ and ‘er-han’. By 0, 0.1, 1 and the 10ppm 1- metaylcyclopropene (1-MCP) steams of dahan welsh onion, and by naked either the paper bag packaging storage at 5℃ or 15℃. The 1ppm processing with paper bag packaging at 5℃ weightloss is lowest. Various densities process the welsh onion to storage at 5℃ during 3 weeks use to naked and the paper bag packaging are not decay index. After the 0.1ppm processing, naked and the paper bag packaging storage in 5℃ under the yellowing index are few. The horizontal or vertically of intact or de-root welsh onion observation weight loss, decay and yellowing. The weightloss, yellowing index, decay index and respiration rate of welsh onion all good performance. Another defect particular to welsh onions is leaf curvature , which occurs when the product is placed horizontally. The welsh onion is leaf vertical , which occurs when the product placed vertically The weightloss, yellowing index, decay index and respiration rate of welsh onion during 5℃ for 10 days transfer to 20℃ all fast increase. The chlorophyll content, cutting force and Vitamin C content of welsh onion during 5℃ for 10 days transfer to 20℃ is decrease.

The objective of this study was to evaluate the effect of vapor heat treatment on the incidences of postharvest decay and quality of ‘Tainung No.2’ papaya fruit grown in Taiwan, and to improve the postharvest technology for export. Temperature at or lower than 12℃ delayed the ripening of papaya fruit, as evidenced by the absence of respiration and ethylene production peaks and no coloring of peel. The respiration and ethylene peaks appeared later at 15℃than 20℃. During the coloring of papaya at 15℃ and 20℃, the Fv/Fm values decreased gradually. There was no difference in the quality of fruits between harvested with and without stem, but the incidence of stem-end rot was lower in stem-remained fruit after storage and ripening. As transport from the field to packing house, the fruits held reversal had higher incidence of stem-end rot than those held vertical and horizontal. The fruits harvested in March occurred heat injury and abnormal ripening after vapor heat treatment. Hydrocooling with 1℃ cold water caused scald in peel of papaya fruit after vapor heat treatment, and with 27℃ water caused severe decay. After vapor heat treatment, the fruits were stored at 15℃, 18℃, 21℃, and 24℃, respectively. Increasing the storage temperature decreased the time of softening. The incidence of decay was higher when the fruit was held at higher temperature. Hot-water immersion could decrease the rate of decay. Complete ripening occurred after 56 hours at 30℃, 84 hours at 25℃, 7 days at 20℃, and 10 days at 15℃, respectively. Ripening temperature at or over 33℃caused abnormal ripening, as poor coloring and hard pulp. The fruits ripened at 30℃ for 24 hours were transported at 1℃, 3℃, 6℃, 9℃, 12℃, and 15℃, respectively, for 7 days and there was no significantly different in fruit quality among the transportation temperature from 3℃ to 15℃, but faster softening was occurred in higher temperature. After the fruits were stored at or below 12℃ for 30 day, to simulate long distance transportation, off flavors, decay, and skin scald developed. Delaying storage until the fruit ripened decreased chilling susceptibility.

Seasonal variations had a great influence on the ripening patterns of banana fruit. Normal ripening was enhanced or delayed by the effect of the maturing season on the fruit. Fruit maturing in summer (harvested in autumn) showed an increase in the rate of ripening, while fruit harvested in winter and spring showed a decrease in the rate of ripening. The banana fruit are sensitive to temperatures outside of their acclimated range. Due to ineffective storage and faulty handling procedures, cold and heat stress is a universal problem in bananas. Large amounts of fruit are lost due to heat and cold stress. Chilling injury occurs when bananas experience or are stored at temperatures below 13C, for a few hours to a few days. Temperatures slightly above 30C may cause extensive heat damage. The three main mechanisms suggested for the prevention of chilling and heat injury are the synthesis of small heat shock proteins, changes in the saturation of fatty acids of the membranes and suppression of the ethylene production and respiration rates. The prevention of cold and heat stress is investigated by monitoring the effect of short-term heat treatment and treatment with ethylene inhibitors on the ripening of the fruit. To determine changes in ripening the following eight ripening parameters are monitored: Yellowing, firmness of the pulp, sugar concentration, starch content, ethylene synthesis, respiration rate, rate of moisture loss and ion leakage. It was found that 1-methylcyclopropene (1-MCP) alleviates the symptoms of heat injury up to a point, but the symptoms of chilling injury were not affected. The effects of the three temperatures investigated: 35°C, 40°C and 45°C for different times. A short pre-heat treatment of 35˚C for 20 minutes proved successful to delay the ripening of the banana fruit, but did not protect the fruit against heat or chilling injury. The results of this study clearly indicate that both chilling and heat injury involve the action of ethylene and suggested that longer exposures to 1-MCP may be effective in preventing both types of injury.

The objectives of this experiment were to investigate the postharvest handling technique, to find suitable temperature for storage, and to map the symptom of chilling injury as well as physiological changes of the jicama tuberous root under low-temperature condition. Jicama is a tropical crop, and thus is prone to low-temperature injury. Decay and browning being the principal visual symptom, cause the loss of commercial values. Results indicated that the jicama tuberous root stored at a temperature lower than 9℃ would show sign of chilling injury. When the chilled jicama root was moved to a higher temperature, its respiration and ethylene production rates would increase in proportion to the severity of the damage brought about by the cold. The jicama tuberous root stored at 6℃ or below for 2 weeks, or stored at 9℃ for 3 weeks resulted in chill-induced decay and at 9℃ or below for a week led to severe browning. No obviously chilling injury was observed when it was stored at 12℃ or above for 1-4 weeks. However, a decrease in chroma and increase in lightness (L) values of root color were noticeable; the respiration and ethylene production rates remained relatively low. The critical storage temperature has found to be 9℃ for 2 weeks. After storing at 9℃ for 2 weeks, browning appeared on the tuberous roots before returning to the room temperature, and browning was deepened over time after storage. Furthermore, after storing at 1℃ for 1 week, browning was observed only after 48 hours under the room-temperature condition. Storing at 1℃ for 1 week then moved to 15℃, 20℃, 25℃, 30℃ and 35℃, the symptom of chilling injury was most significant while the root was placed above 20℃. Utilizing techniques of temperature regulation, PE packaging and hot-water treatment, it appeared that wrapping with PE bag at low temperature had limited effect on browning but reduced the severity of decaying. Temperature conditioning seemed to be ineffective either. Intermittent warmming regulation ameliorated the symptom of the chilling injury. Hot-water treatment at 50℃ before storing reduced the development of the chilling- injury symptom on the jicama tuberous root.

The objectives of this experiment were to investigate the changes in enzyme activities with regard to litchi (Litchi chinensis Sonn.) pericarp browning, and to develop an alternative method to SO2 fumigation for color retention of litchi fruits. After harvest, the water loss of litchi fruits was very rapid and the pericarp began to brown at 25℃ within 12 hours. Water loss of fruits mainly occurred through pericarp. During pericarp browning of ‘Yuh Her Bau’, ‘Hei Yeh’, and ‘Nuoh Mii Tzy’ litchi fruits, peroxidase activity of the peel was higher than the polyphenol oxidase activity. Variations in the peroxidase activity were also higher at low relative humidity, and showed a tendency to increase during storage. However, variations of polyphenol oxidase activity were not coincidental among three cultivars. It seemed that changes in the peroxidase activity were more significant than the polyphenol oxidase activity in respect of litchi pericarp browning. Experiments were also conducted to prevent pericarp browning of litchi fruits with the treatments of 1% chitosan solution, 1N hydrochloric acid (HCl) solution, and the combination of chitosan and 1 N of hydrochloric acid solution. The temperature of the solution was raised to 40℃ in the experiments. Results showed that the ‘a’ values of peel decreased in fruits dipping in 1% chitosan and 1N HCl solution at 40℃ for 5 minutes; significantly less than samples immersed in 1N HCl solution for 6 minutes. The ions leakage of the peel of fruits dipping in 1% chitosan and 1N HCl solution at 40℃ for 5 minutes was higher than that of the control. This seemed to indicate that permeability of plasma membrane of the fruit was increased after the heated 1% chitosan treatment, and made it easier for the acid to permeate into vacuole. Thus pH of the vacuole decreased, the anthocyanin molecules stabilized, the red color of the peel retained. Moreover, the hot 1% chitosan treatment obviously depressed polyphenol oxidase and peroxidase activities of the fruits, and thus reduced enzymatic browning of the peel. Therefore, the fruits could retain the red color of the peel after fruits were returned to the room temperature. Although hot 1% chitosan treatment maintained the color of the peel, there were probabilities of dehiscence, decay and sinking of peel of fruits after they were returned to room temperature. Cold storage of the fruits in the perforated polyethylene bags (0.04mm) significantly improved the sinking of peels. Further investigations are needed to improve and control of fruit decay and dehiscence.

The objectives of this study were to investigate the effects of temperatures on the storage life and fruit quality of the ‘Fuyu’ persimmon with a view to defining the best storage temperature and condition, reducing chilling injury and fruit softness, and improving the quality as well as commercial value of the fruits. The best time to harvest of ‘Fuyu’ persimmon fruit in the medium elevation mountain regions of central Taiwan should begin on 25th of November as far as maturity is concerned. At this time the average fruit weight reaches 285.2g, its total soluble solid content approaches more than 14.7∘Brix, and its peel color turns orange red. All these promises better economical values. However, in terms of storage, it is more preferable to picj them from 9 to 23, December. During this period, the fruits remain firm, with lower respiration rate and more red skin color, heavier weight, way ahead of the pre-climacteric and closer to Chinese New Year. The results indicated that persimmon fruits, which were stored at 6-15℃ for 4 weeks, developed the symptoms of translucent peel and softening flesh soon after returning to the room temperature. Softness also occurring in fruits stored at 1 or 3℃, but the damage was less extensive. It is concluded that suitable storage temperature for persimmon cv. ‘Fuyu’ should be below 1℃. The storage life of highly mature fruits is 4 weeks at 1℃. The persimmon fruits when soaked in AVG before storage could delay the occurrence of climacteric peak by inhibiting respiration and ethylene production. The same effects were obtained by spraying AVG solution on the tree 2-3 weeks before harvest. Treatment of persimmon with 1-MCP and hot water (53℃, 50min) before storage could delayed softening and turning of fruit color and prevented chilling injury from occurring. However, black scorch could appear on the peel of some fruits, which affected the appearance and marketing values. In addition, 1-MCP treatment inhibited ethylene action thus retained the firmness of ‘Fuyu’ fruits. Hot water (53℃, 50min) treatment of persimmon fruits harvested on 22nd December, the storage life was prolonged to 8 weeks under 1℃ storage condition.
