Full textSalmonella is a bacterial enteropathogen representing the main cause of food poisoning worldwide. Persistence of Salmonella Enteritidis in poultry flocks in the form of intestinal carriage represents an important source of contamination of poultry derived food products. Improving immunity of the mucosal barrier of the bird intestine to fight bacterial colonisation is a strategy alternative to the controversial use of antibiotics. During the study of chicken response to Salmonella infection, it had been shown that avian ß-defensin genes (AvBD1 and 2) were highly expressed in intestinal tissue of birds resistant to Salmonella colonisation. ß-defensins are cationic peptides able to kill bacteria and also, in mammalian species, to attract cells of the animal immune defence. In this context, understanding the mechanism of action of these two avian ß-defensins against intestinal infection of birds by Salmonella represented the main objective of the thesis work. While AvBD1 and 2 were initially identified from heterophils, we were interested in determining whether the intestinal epithelial cells, which constitute the first barrier encountered by the bacteria, were able to express them. A primary culture of intestinal epithelial cells has been developed from chicken embryos of divergent inbred lines either resistant or susceptible to Salmonella intestinal carriage. The cells exhibited characteristics of epithelial type according to morphology and expression of specific cytoskeletal proteins. They were shown to express the two ß-defensin genes AvBD1 and AvBD2 according to the chicken line. Furthermore, S. Enteritidis interfered with AvBD2 expression only in the cells from the susceptible line. In parallel, we were interested in preparing these ß-defensins in order to assess their antibacterial activity. Chicken bone marrow was used to isolate ß-defensins by acetic acid extraction followed by chromatographic separation. Three purified ß-defensins, AvBD1, AvBD2 and AvBD7 were characterized by mass spectrometry, revealing post-translational modifications in AvBD1 C-terminal end and AvBD7 N-terminal end. Antimicrobial activity measured by radial diffusion assay has shown a high efficiency against Gram + and Gram – except for AvBD2 with a lower potency against Gram – bacteria. Antibacterial spectrum of AvBD2 showed specificity towards Gram + bacteria as well as the king penguin AvBD103b whose three-dimensional structure has been described. This prompted us to resolve the structure of AvBD2 by nuclear magnetic resonance in order to analyze structure-activity relationships by comparing these two avian ß-defensins. This approach has led us to propose a mechanism of action for these two ß-defensins and particularly for AvBD2 in the intestinal mucosa. Finally, taken together the results allow to further define the role of avian ß-defensins in the phenotype of resistance/susceptibility to intestinal invasion by Salmonella. In addition to their antibacterial activity, which can be overcome to some extend by Salmonella, they could display other functions such as chemotaxis allowing recruitment of immune competent cells. The characterization of the activity spectrum of these avian ß-defensins opens perspectives of therapeutic application in a domain where antibiotics use should be avoided

Full textIn chickens, the muscle glycogen stores available at death are determinant for meat quality via their effect on the ultimate pH. This thesis aimed at identifying through functional genomic approaches genes causing changes in glycogen and therefore meat quality in chicken. A focused approach on the AMP-activated protein kinase (AMPK) and several enzymes that control glycogen metabolism suggested that transcriptional regulation of several genes (PRKAG2, GYS, PYG, …) could be involved in the control of muscle glycogen stores. This study, which compared lean and fat chickens also showed activation levels 3 times higher in lean chickens whose glycogen stores are the lowest. Transcriptomic analyses performed on oligonucleotide microarrays have highlighted 439 genes differentially expressed between individuals with different levels of muscle glycogen store. Among them, some as CEBPB, PDK4 or UGDH could play a direct role in the regulation of glycogen metabolism in muscle. Analysis of positional data (Quantitative Trait Loci or QTL) allowed identifying other genes which were both regulated at the functional level and located within a QTL of meat quality. Those genes enriched the list of candidates revealed by expressional studies. Altogether these studies are a first step in identifying genes involved in meat quality changes in chicken. After validation within commercial genotypes to appreciate the real interest in term of selection, our results should ultimately help to develop molecular tools useful for selection but also for optimizing rearing practices to improve the quality of poultry meat

Full textCephalic neural crest (CNC) is a pluripotent structure giving rise to entire skeleton of the face and skull. Absence of CNC is associated with forebrain and eye defects that mimic human congénital malformations: microcephaly, holoprosencephaly, aniridia, missing anterior chamber, coloboma, congenital dislocation of the lens, agenesis of eyelid and early strabismus. (Alward 2000 Creuzet 2009, Williams and Bohnsack 2015). In our study, heterochronic increased Bmp7 signaling in the prechordal plate (Pcp), caused disruption of CNC migration associated with fusion of two optical fields into one cyclopic eye. This cyclopia, unlike previous studies (Chiang et al. 1996, Golden et al. 1999), is preceded by a disturbance of the apical constriction in optic vesicles at Hamburger-Hamilton (HH)12 stage witch is responsible for dorsal invagination of OVs and their separation from the rest of the neural tube. At the molecular level, we recorded an increase in Wnt1 expression very early after treatment (2h) in CNC and this increase extends rostrally suggesting that Wnt1 could be involved in the disturbance of eye development. To test this hypothesis, we attempted rescue experiments by inactivating Wnt1 signaling by RNA interference and we observed an improvement of the phenotype but the eyes remained morphologically in hypotelorism, too close,. At E8 (32-33HH) stage the eyes are remained fused to the forebrain and are synophtalmic, two eyes in a single orbit,. We also observed a decrease in Wnt2b expression in OV in the cyclopic embryo and that this decline persists despite Wnt1 rescue. To understand Wnt2b signaling, we attempted a second rescue experience with RNA interference against Wnt1 and a plasmid vector expressing Wnt2b in the CNC and OV respectively. Coronal sections at E8 stage resemble the control but with persistence of dorsal fusion between OVs and telencephalon. In situ hybridization showed a decrease in Foxg1 expression at 12HH in the dorsal part of OVs in the RD/T boundry in the two rescue attempts suggesting that this gene may be involved in the separation of OVs at 12HH stage rostrally from the telencephalon

Full textMsx genes are highly conserved homeobox genes which are expressed prominently at sites of induction during development. In vertebrates, they are expressed at the dorsal midline of the neural tube. Using Msx1 and Msx2 mutant mice, we investigated their functions at this site. Msx1nlacZ mutant mice display a hydrocephalus which is characterised by the enlargement of the third and lateral ventricules. Histological analysis shows that the sub-commissural organ (SCO), a neurosecretory organ that derives from the dorsal part of prosomere 1, is abnormal in the mutant. Furthermore BMP6, Wnt3a, Wnt1, and Msx1nlacz expression profile is interrupted in the dorsal midline of prosomere1. Therefore, morphological and gene expression data indicate that a functional midline is not maintained along the whole prosomere 1 in these mutant mice. This results in the downregulation of genes expressed laterally to the midline in prosomere 1, confirming the importance of the midline as a signaling centre. Wnt1 is essential for dorso-ventral patterning. In the Msx1 mutant, Wnt1 is downregulated before the midline disappears, suggesting that its expression depends on Msx1. Electroporation in the chick embryo further demonstrates that Msx1 can induce Wnt1 expression in the diencephalon neuroepithelium and in the lateral ectoderm. In double Msx1/Msx2 mutants, Wnt1 expression is completely abolished at the dorsal midline of the diencephalon and rostral mesencephalon. This indicates that Msx genes regulate Wnt1 expression at the dorsal midline of the neural tube. Based on these results, we propose a model in which Msx genes are intermediary between BMP and WNT at this site
