Abstract:
Background/Aims: Photodynamic therapy (PDT) is a promising noninvasive technique, which has been successfully applied to the treatment of human cancers. Studies have shown that the Bcl-2 family proteins play important roles in PDT-induced apoptosis. However, whether Bcl-2-interacting mediator of cell death (Bim) is involved in photodynamic treatment remains unknown. In this study, we attempt to determine the effect of Bim on Photofrin photodynamic treatment (PPT)-induced apoptosis in human lung adenocarcinoma ASTC-a-1 cells. Methods: The translocation of Bim/Bax of the cells were monitored by laser confocal scanning microscope. The levels of Bim protein and activated caspase-3 in cells were detected by western blot assay. Caspase-3 activities were measured by Caspase-3 Fluorogenic Substrate (Ac-DEVD-AFC) analysis. The induction of apoptosis was detected by Hoechst 33258 and PI staining as well as flow cytometry analysis. The effect of Bim on PPT-induced apoptosis was determined by RNAi. Results: BimL translocated to mitochondria in response to PPT, similar to the downstream pro-apoptotic protein Bax activation. PPT increased the level of Bim and activated caspase-3 in cells and that knockdown of Bim by RNAi significantly protected against caspase-3 activity. PPT-induced apoptosis were suppressed in cells transfected with shRNA-Bim. Conclusion: We demonstrated the involvement of Bim in PPT-induced apoptosis in human ASTC-a-1 lung adenocarcinoma cells and suggested that enhancing Bim activity might be a potential strategy for treating human cancers.

Abstract:
This study aims to study the graduation rate of living and post growth of vaname shrimp larvae using multilevel salinity acclimatization methods maintained at low salinity. This study consisted of five treatments, and each treatment had 3 repeats. Thus in this study there were 15 experimental units. As a treatment is the adedation period of acclimatization decreases salinity as follows: Treatment 1: No decrease in salinity (30 ppt). Treatment 2: 6 day decreased salinity (30 ppt, 24 ppt, 18 ppt, 12 ppt, 6 ppt, 0 ppt). Treatment 3: 7 days decrease in salinity (30 ppt, 25 ppt, 20 ppt, 15 ppt, 10 ppt, 5 ppt, 0 ppt). Treatment 4: 8 days decrease in salinity (30 ppt, 26 ppt, 21 ppt, 17 ppt, 13 ppt, 9 ppt, 4 ppt, 0 ppt). Treatment 5: 9 days decrease in salinity (30 ppt, 26 ppt, 23 ppt, 19 ppt, 15 ppt, 11 ppt, 8 ppt, 4 ppt, 0 ppt). The results showed that the larvae of vaname shrimp that were kept at low salinity using acclimatization methods had no noticeable effect on survival rates. The survival rate is between 72% - 94.67%, has an absolute weight between 1.75 grams – 2.36 grams and an absolute length between 1.47 cm – 1.87 cm with a fcr value of 3.17 – 3.83. The oxygen consumption rate in vaname shrimp is 1.76 – 2.2.

Abstract:
Testing processes and workflows in information and Internet of Things systems is a major part of the typical software testing effort. Consistent and efficient path-based test cases are desired to support these tests. Because certain parts of software system workflows have a higher business priority than others, this fact has to be involved in the generation of test cases. In this paper, we propose a Prioritized Process Test (PPT), which is a model-based test case generation algorithm that represents an alternative to currently established algorithms that use directed graphs and test requirements to model the system under test. The PPT accepts a directed multigraph as a model to express priorities, and edge weights are used instead of test requirements. To determine the test-coverage level of test cases, a test-depth-level concept is used. We compared the presented PPT with five alternatives (i.e. the Process Cycle Test (PCT), a naive reduction of test set created by the PCT, Brute Force algorithm, Set-covering-Based Solution and Matching-based Prefix Graph Solution) for edge coverage and edge-pair coverage. To assess the optimality of the path-based test cases produced by these strategies, we used 14 metrics based on the properties of these test cases and 59 models that were created for three real-world systems. For all edge coverage, the PPT produced more optimal test cases than the alternatives in terms of the majority of the metrics. For edge-pair coverage, the PPT strategy yielded similar results to those of the alternatives. Thus, the PPT strategy is an applicable alternative as it reflects both the required test coverage level and the business priority in parallel.

Abstract:
Pain perception threshold (PFT) in the head was assessed with a pressure algometer in 58 cluster headache (CH) patients (52M, 6F; 41 episodic and 17 chronic). Fourteen patients in cluster period were retested in remission. Thresholds were assessed at 10 symmetrical points on each side of the head and at the deltoid. Compared with controls ( n = 80), CH patients had lower PPT in the head and in the deltoid. PPT was lower on the symptomatic side than on the non-symptomatic side in patients with episodic CH during a cluster period ( p<0.001) and in patients with chronic CH ( p<0.05). This pattern was more evident during a cluster period than during remission ( p<0.05). A reduced PPT did not correlate with illness duration and pain side. The lowest PPT mean values were found at the anterior and intermediate levels of the temporal muscle on the symptomatic side. These results imply a central mechanism underlying the pathogenesis of CH.

Abstract:
Tilapia fish is a freshwater fish commodity that has the potential to be developed for fisheries business. Tilapia fish have many advantages such as easy breeding, fast-growing, and can survive at high salinity. To increase durability at high salinity it is necessary to prepare saline tilapia seeds produced from parentage that is tolerant to high salinity or through the salination process since the size of the seed. The purpose of this study was to determine the survival rate, absolute growth, and water quality during the adaptation period of increasing salinity of tilapia juvenile. The experimental design used was a completely randomized design with three replications. The salinity increase treatments used were A (1 ppt salinity), B (2 ppt salinity), C (3 ppt salinity), D (4 ppt salinity), and E (5 ppt salinity). The results showed the survival rate of tilapia seeds each treatment is treatment A (1 ppt) by 100%, treatment B (2 ppt) by 100%, treatment C (3 ppt) by 100%, treatment D (4 ppt) by 99.2%, and treatment E (5 ppt) by 98.6%. The results of each absolute growth of each treatment are treatment A (1 ppt) of 0.002 grams / day, treatment B (2 ppt) of 0.002 grams / day, treatment C (3 ppt) of 0.001 grams / day, treatment D (4 ppt) of 0.0007 grams / day, and treatment E (5 ppt) of 0.001 grams / day. It can be concluded that the adaptation of fish seeds at different salinities (1 ppt, 2 ppt, 3 ppt, 4, ppt, and 5 ppt) still shows a good survival rate, but the absolute growth of tilapia seeds on the adaptation of salinity increases of 4 ppt and 5 ppt has not been optimal.

Abstract:

Abstract:
ABSTRACT Evidence from knockout studies in male mice and from experiments in male rats,in which expression of the estrogen receptor alpha (ERα) gene was inhibited in the medial preoptic area (MPO), suggests that ERα is important in the control of male rat mating behavior. Therefore, in this experiment, we tested the hypothesis that activation of ERα in the MPO is sufficient to maintain mating behavior in castrated male rats receiving subcutaneously (s.c.) dihydrotestosterone (DHT), a non-aromatizable androgen. Accordingly, castrated rats treated with DHT s.c. received MPO implants of either: (i) propyl-pyrazole-triol (PPT) (Stauffer, et al 2000; Katzenellenbogen, et al 2000), a selective ERα agonist, (ii) E2 (positive controls) or (iii) cholesterol (negative controls)and sexual behavior was monitored. PPT was as effective as E2 at maintaining mating behavior suggesting that, in the MPO, ERα is sufficient to mediate responses to E2 that underlie male rat mating behavior.

Persson, Anna, and Jessica Tedenhag.

Abstract:
In 2012 the BEPS project started with the purpose of preventing tax subjects from using loopholes in the tax treaties and national tax laws in order to receive tax benefits. In 2013, the OECD published an action plan that identifies 15 actions that is meant to be taken regarding this issue. Action 6 regulates treaty abuse through treaty shopping, which means that a tax subject is searching for the tax jurisdiction with the tax treaty that leads to the most beneficial taxation. To prevent this, Action 6 suggests that a specific LOB-rule and a general anti-avoidance rule, PPT-rule, should be included in the OECD model convention. The LOB-rule regulates in which specific situations a treaty benefit can be granted. The PPT-rule is more general and contains general wordings to cover situations that is difficult to foresee. The purpose of this thesis is to examine whether the proposed rules can fulfill the purpose of Action 6, to prevent treaty abuse. If not possible, it will be determined if Sweden can prevent treaty abuse by applying the general anti-avoidance rule against tax evasion stated in the Swedish skatteflyktslagen. The authors of the thesis are of the opinion that the LOB-rule is too complex in its current wording which makes it difficult to apply. Since the PPT-rule is vague, there is a wide scope for arbitrary assessments leads to an unpredictable outcome when the guidance is unclear. Therefore, the authors find that the rules in its current wordings cannot satisfy the purpose of Action 6.  According to the Peru-judgement the skatteflyktslagen can be applicable to situations that are covered by a tax treaty. The transaction should be tested against the general antiavoidance. The fourth prerequisite, in contravention of the purpose of the legislation, is difficult of apply and the judgements of the court varies. The authors believe that if the treaty preamble includes a statement which clarifies that the intention of the treaty is not to allow treaty abuse, the Swedish general anti-avoidance rule can prevent treaty abuse and thereby protect the Swedish tax base against BEPS regarding treaty abuse.

McAlpin, Jennifer, and Cassandra Ross. Houston Ship Channel Expansion Channel Improvement Project (ECIP) numerical modeling report : BABUS cell and Bird Island analysis. Engineer Research and Development Center (U.S.), August 2021. http://dx.doi.org/10.21079/11681/41581.

Full text
Abstract:
The Houston Ship Channel (HSC) is one of the busiest deep-draft navigation channels in the United States and must be able to accommodate increasing vessel sizes. The US Army Engineer District, Galveston (SWG), requested the Engineer Research and Development Center, Coastal and Hydraulics Laboratory, perform hydrodynamic and sediment modeling of proposed modifications in Galveston and Trinity Bays and along the HSC. The modeling results are necessary to provide data for hydrodynamic, salinity, and sediment transport analysis. SWG provided three project alternatives that include closing Rollover Pass, Bay Aquatic Beneficial Use System cells, Bird Islands, and HSC modifications. These alternatives and a Base (existing condition) will be simulated for present (2029) and future (2079) conditions. The results of these alternatives/conditions as compared to the Base are presented in this report. The model shows that the mean salinity varies by 2–3 ppt due to the HSC channel modifications and by approximately 5 ppt in the area of East Bay due to the closure of Rollover Pass. The tidal prism increases by 2.5% to 5% in the alternatives. The tidal amplitudes change by less than 0.01 m. The residual velocity vectors vary in and around areas where project modifications are made.

Trace explosives detection by cavity ring-down spectroscopy (CRDS). Engineer Research and Development Center (U.S.), August 2021. http://dx.doi.org/10.21079/11681/41520.

Full text
Abstract:
We built three successive versions of a thermal decomposition cavity ring-down spectrometer and tested their response to explosives. These explosive compound analyzers successfully detected nitroglycerine, 2,4,6-trinitrotoluene (TNT), pentaerythryl tetranitrate, hexahydro-1,3,5-trinitro-s-triazine and triacetone triperoxide (TATP). We determined the pathlength and limits of detection for each, with the best limit of detection being 13 parts per trillion (ppt) of TNT. For most of the explosive tests, the peak height was higher than the expected value, meaning that peroxy radical chain propagation was occurring with each of the explosives and not just the peroxide TATP.

Abstract:
Review question / Objective: P:Adult patients (age ≥ 18 years) diagnosed with stroke based on relevant clinical examination; I:Intervention group with rTMS alone or in combination with other treatments with rTMS; C:Control group received sham treatment or no rTMS; O: Upper extremity function:the Fugl-Meyer Assessment Upper Extremity (FMA-UE); Hand function:box and block test(BBT), nine-hole peg test(NHPT), and Purdue pegboard test(PPT); S:Randomized controlled trials (rather than crossover designs). Condition being studied: In Europe, more than 1 million new cases of stroke are reported each year. The absolute number of stroke patients is expected to increase in the near future due to the progressive aging of the population. Approximately 50-80% of stroke survivors present with upper extremity dysfunction. Recovery of upper extremity function is associated with improvements in activities of daily living and mental health. However, few stroke survivors show full recovery of upper extremity function 6 months after stroke. In addition, rehabilitation has a limited impact on the recovery of hand motor function.

Gadd, M. G., J. M. Peter, and D. Layton-Matthews. Genesis of hyper-enriched black shale Ni-Mo-Zn-Pt-Pd-Re mineralization in the northern Canadian Cordillera. Natural Resources Canada/CMSS/Information Management, 2022. http://dx.doi.org/10.4095/328013.

Full text
Abstract:
Polymetallic (Ni-Mo-Zn-Pt-Pd-Au-Re) hyper-enriched black shales in the northern Canadian Cordillera consist of thin, semi-massive sulfides interbedded with black shale. We studied HEBS deposits at Nick, Peel River, Monster River, and Moss in northern Yukon, and at a single locality underlying the Cardiac Creek Pb-Zn-Ag deposit in northeastern British Columbia. High-grade mineralization contains up to 7.4 weight per cent Ni, 2.7 weight per cent Zn, 0.38 weight per cent Mo, 400 ppb Pt, 250 ppb Pd, 160 ppb Au, and 58.5 ppm Re. Sulfide mineralization formed during syngenesis to later diagenesis. Analyses by LA-ICP-MS indicate that pyrite is the principal host of platinum-group elements, Au, and Re. Mineralization and sedimentation were coeval based on the overlap between Re-Os geochronology of HEBS at Nick and Peel River (390.7 ± 5.1 and 387.3 ± 4.4 Ma, respectively) and conodont biostratigraphic ages of sedimentary host rocks. Bulk S isotope composition of HEBS is uniformly negative, indicating that bacterial reduction of seawater sulfate generated sulfur to precipitate sulfide minerals. The initial Os ratios at Peel River (0.25 ± 0.07) and Nick (0.32 ± 0.20) overlap with Middle Devonian seawater, suggesting that elemental enrichment was derived from seawater.

Dickman, Martin B., and Oded Yarden. Role of Phosphorylation in Fungal Spore Germination. United States Department of Agriculture, August 1993. http://dx.doi.org/10.32747/1993.7568761.bard.

Full text
Abstract:
Spore germination is a common and fundamental event in fungal development and in many instances an essential phase of fungal infection and dissemination. Spore germination is also critical for hyperparasites to function as biocontrol agents as well as in fermentation proceses. Our common objective is to understand the mechanisms which regulated spore germination and identify factors involved in pathogenicity related prepenetration development. Our approach is to exploit the overall similarity among filamentous fungi using both a plant pathogen (Colletotricum trifolii) and a model system that is genetically sophisticated (Neurospora crassa). The simulataneous use of two organisms has the advantage of the available tools in Neurospora to rapidly advance the functional analysis of genes involved in spore germination and development of an economically important fungal phytopathogen. Towards this we have isolated a protein kinase gene from C. trifolii (TB3) that is maximally expressed during the first hour of conidial germination and prior to any visible gene tube formation. Based on sequence similarities with other organisms, this gene is likely to be involved in the proliferative response in the fungus. In addition, TB3 was able to functionally complement a N. crassa mutant (COT-1). Pharmacological studies indicated the importance of calmodulin in both germination and appressorium differentiation. Using an antisense vector from N. crassa, direct inhibition of calmodulin results in prevention of differentiation as well as pathogenicity. Both cAMP dependent protein kinase (PKA) and protein kinase C (PKC) like genes have been cloned from C. trifolii. Biochemical inhibition of PKA prevents germination; biochemical inhibitors of PKC prevents appressorium differentiation. In order to analyze reversible phosphorylation as a regulatory mechanism, some ser.thr dephosphorylative events have also been analyzed. Type 2A and Type 2B (calcineurin) phosphatases have been identified and structurally and functionally analyzed in N. crassa during this project. Both phosphatases are essential for hyphal growth and maintenance of proper hyphal architecture. In addition, a first novel-type (PPT/PP5-like) ser/thr phosphatase has been identified in a filamentous fungus. The highly collaborative project has improved our understanding of a fundamental process in fungi, and has identified targets which can be used to develop new approaches for control of fungal plant pathogens as well as improve the performance of beneficial fungi in the field and in industry. In addition, the feasibility of molecular technology transfer in comparative mycology has been demonstrated.

Zevotek, Robin, and Steve Kerber. Fire Service Summary Report: Study of the Effectiveness of Fire Service Positive Pressure Ventilation During Fire Attack in Single Family Homes Incorporating Modern Construction Practices. UL Firefighter Safety Research Institute, May 2016. http://dx.doi.org/10.54206/102376/ncck4947.

Full text
Abstract:
There is a continued tragic loss of firefighter and civilian lives, as shown by fire statistics. One significant contributing factor is the lack of understanding of fire behavior in residential structures resulting from the use of ventilation as a firefighter practice on the fire ground. The changing dynamics of residential fires as a result of the changes in home construction materials, contents, size and geometry over the past 30 years compounds our lack of understanding of the effects of ventilation on fire behavior. Positive Pressure Ventilation (PPV) fans were introduced as a technology to increase firefighter safety by controlling the ventilation. However, adequate scientific data is not available for PPV to be used without increasing the risk to firefighters. This fire research report details the experimental data from cold flow experiments, fuel load characterization experiments and full scale fire experiments. During the project it was identified that the positive pressure attack (PPA) and positive pressure ventilation (PPV) were often used interchangeably. For the purpose of this report they have been defined as PPA for when the fan is utilized prior to fire control and PPV for when the fan is used post fire control. The information from the full scale tests was reviewed with assistance from our technical panel of fire service experts to develop tactical considerations for the use of PPV fans in residential single family structures.
