A rede de ativismo político contemporâneo tem orientado suas ações em direção a um campo híbrido que desconsidera fronteiras entre ativismo e arte . Dentre a variedade de práticas desta rede, o prank é um exemplo da desconsideração destas fronteiras. Um prank é um golpe, uma ruptura jocosa da ordem, da hierarquia, da autoridade. Como prática, o prank é abordado ao longo deste trabalho como uma opção performativa para o ativismo político, uma vez que reúne características lúdicas, críticas, vivenciais e multiplicadoras. Os ativistas que adotam o prank como opção se relacionam com a história recente da rede de ativismo político contemporâneo, que resiste a um novo modelo de organização do capitalismo. De 1994 a 2003, esta rede estabeleceu um panorama de macroações (em especial, os Dias de Ação Global) festivas e insurgentes. O ativismo difuso, diluído no cotidiano com ações em menor escala, também tem adotado a mesma orientação irreverente. Isto é especialmente verificável na reivindicação pelo espaço, entendido de maneira ampla, tanto como espaço urbano erodido pela invasão do capital, quanto como espaço da comunicação, da ideologia e das trocas simbólicas. As modalidades de pranks adotadas neste contexto são examinadas a partir de categorias como a teatralidade e a performatividade, conforme entendidas por Josette Féral. Estas ferramentas contribuem na conclusão de que n a performance atravessada pelo prank não existem mediadores, ela é autônoma, orientada pelo princípio político do faça você mesmo.The current network of political activism has pursued a hybrid field, ignoring the borders between activism and art. Amongst the various genres of this network, the prank illustrates how such borders are blurred. A prank is a blow, a whimsical breaking of order, of hierarchy, and of authority. This thesis addresses prank as a practice, a performance option for political activism, provided that it merges experiences of play, criticism, liveness and dissemination. Activists who have chosen the prank are related with a recent history of the contemporary network of political mobilization, which resists to a new capitalist pattern of organization. From 1994 through 2003, this network established a scenario of celebrations and radical macro-actions (particularly the Days of Global Action). The spread out activism that is blended in daily life also features a similar invasive attitude. This is especially true in their claims for space, broadly understood both as urban space that has been contaminated by the invasion of capital, and as a space of communication, ideology and symbolic exchange. I have examined the various prank genres within this context on the basis of categories such as theatricality and performativity, according to Josette Férals stance. These considerations have led me to the conclusion that there are no mediators when prank merges with performance, since it is autonomous and draws on the do it yourself political principle.

Cities are expanding and the nature is being pushed into the periferi. Free flow of elements such as water, air, earth and plants are being controlled, disstorted, polluted and manipulated. Our political and economical systems are built upon the mind-set that we are separate from the nature and above the nature, but we are a part of the nature just like every other speecies on this planet. Our fast paced city-lives are creating a huge gap between the nature and ourselves and massproduction has replaced traditional craft and sustainable food production. Where is this going to end? How will it end? How can architecture help us reconnect with the nature and ourselves and inspire to a more sustainable lifestyle? Prana is not a building, it is a site. It consists of a multifunctional garden area with several buildings that together creates a whole. Prana is sanskrit meaning breath, energy or life force and has given the site its’ name because it is a platform for recreation, contemplation and knowledge exchange. We need to slow down, reflect and learn in order to feel the force and make a change, no matter if it is your lifestyle, your mind-set or your goal, no matter if you do it for the climate or for yourself. Prana is about reconnecting with the nature and yourself though arts, crafts, activities, food and food production. Prana welcomes everyone for a close nature experience of hot and cold, of fire and earth and community based learning.

El presente trabajo de investigación se desarrolla con la finalidad de implementar sesiones de mindfulness a nivel corporativo enfocado en el Top y Middle Management, una alternativa diferente que sea operativa y económicamente factible.
La motivación para desarrollar este proyecto se da por las actuales tendencias sobre el cuidado de la salud física y emocional a través de la gestión del estrés (Euromonitor, 2019) debido a los cambios en el entorno laboral y personal.
Por ello, el uso de la metodología utilizada está basado en el Modelo Canvas considerando los puntos más importantes de forma visual. Dicha información fue validada a través de entrevistas a profundidad por expertos en recursos humanos, lo cual permitió reconfirmar el público objetivo con interés en el bienestar físico y emocional.
Además, la empresa contará con una oficina administrativa en el distrito de Miraflores
Por otro lado, en la evaluación económica del plan de negocio se obtiene una rentabilidad de s/57 512.32. Los resultados muestran una inversión inicial de s/. 103 490 de los cuales 50% es capital propio y 50% será financiado.
Considerando un escenario esperado con la venta anual de 80 paquetes y la asistencia de 5 personas promedio por prácticas de 5 sesiones por programa, se alcanzaría una utilidad neta anual de s/. 74 810.65 en el primer año.
Considerando los distintos escenarios sólo en el pesimista se obtiene una TIR de 51%, en el optimista de 131% y la TIRE de 43.77%.
Los flujos económicos se reflejan positivamente a partir del primer año siendo en el año 2021 de s/.74 810.65, alcanzando los s/. 60 269.10 al 2022 y s/. 66 247.67 al 2023. Respecto al flujo financiero se registra s/. 54 515.45 en el año 2021 y llegando a s/.43 378.64 al 2023.This research work is developed with the implementation of implementing mindfulness sessions at a corporate level focused on Top and Middle Management, a different alternative that is operative and economically factible. The motivation to develop this project is given by current trends in physical and emotional health care through stress management (Euromonitor, 2019) due to changes in the work and personal environment. However, the use of the methodology used is based on the Canvas Model determined the most important points of the visual form. Such information was validated through in-depth interviews by human resources experts and meditation experts, which confirmed the target audience with an interest in physical and emotional well-being. In addition, the company has an administrative office in the Miraflores district. On the other hand, in the economic evaluation of the business plan, a return of s / 57 512.32 is obtained. The results found an initial investment of s /. 103 490 of which 50% is own capital and 50% will be financed. Considering an expected scenario with the annual sale of 80 packages annual and the attendance of 5 average people for practices of 5 sessions per program, obtain an annual net profit of s /. 74 810.65 in the first year. Considering the different aspects only in the pessimistic one obtains TIR of 51%, in the optimistic obtains131% and the TIRE obtains of 43.77%. The economic flows show positively from the first year, being in 2021 of s/.74 810.65, reaching s/. 60 269.10 to 2022 and s/. 66 247.67 to 2023. Pranks, used symbolically to subvert authority and collapse hierarchy, have become important tools for grassroots political movements. Activists utilize pranks as a form of "culture jamming" to undermine, humiliate, and educate. This thesis documents political pranks pulled by Earth First!, the Yippies, the Biotic Baking Brigade, and the Eugene Anarchists for Torrey (EAT) Campaign to show how pranks are performed and narrated within anarchist subcultures. Drawing on cultural and performance studies, as well as on anarchist theories, this thesis demonstrates how pranks can become performances of resistance and criticism that disrupt the status quo.

Les Poxviridae codent une famille originale de protéines présentant des répétitions ANK et un motif PRANC en C-terminal. Ce travail de thèse présente l'étude de 3 des 4 protéines (ANK-PRANC) codées par le virus de la myxomatose (MYXV): M148R, M149R et M150R (MNF), qui sont organisées en cluster. Les études montrent qu'elles sont exprimées séquentiellement au cours du cycle viral et qu'elles présentent des localisations cellulaires originales. In vivo, M148R, M149R et MNF sont des facteurs de pathogénicité du MYXV bien qu'elles n'aient pas de rôle dans le tropisme cellulaire ni dans son spectre d'hôte. En outre, les études d'histopathologie suggèrent que M148R et MNF inhibent de la réponse inflammatoire. De plus, nous avons montré que M148R, M149R et MNF font partie d'un complexe SCF (Skp, Culline, F-box) permettant l'ubiquitination de protéines cibles qu'il reste à identifierThe Poxviridae encode a unique class of proteins characterized by ANK repeats and PRANC motif in C-terminus. This study presents characterization of 3 of the 4 ANK-PRANC proteins encoded by myxoma virus (MYXV): M148R, M149R and M150R (MNF), organized in cluster. This work show that they are sequentially expressed during replication cycle and that they present original sub-cellular localizations. In vivo, M148R, M149R and MNF are MYXV virulence factors although they are not likely to be implicated in cell tropism or host range functions. Moreover, histological analysis suggests that M148R and MNF play a role in the subversion of host inflammatory response by MYXV. Furthermore, we show that M148R, M149R and MNF are components of SCF complexes (Skp, Cullin, F-box), allowing ubiquitination of a substrate that it remains to identify

This master´s thesis consists of two parts. The first part is a study where the possibility to use existing tools from the market is assessed. The second part describes the design, development and usability test of a web based hardware configuration tool. The usability test made in the design process, was made in order to remove common usability problems and to assess whether or not the design decisions made during the thesis were good. The usability test that was performed followed the think aloud method and gave me very useful information that helped me improve the tool.
The study that was made in the beginning of the thesis was done in order to see if there were any programs on the market which already did the required tasks or could be adjusted to do them. I could not find a satisfying alternative amongst the five alternatives investigated. This lead to the decision to design a new tool. The main functions are the ability to save the current state the hardware is in, and at a later stage load this configuration back.
The tool was designed for the PRAN unit at Ericsson AB, in Linköping, Sweden.

This diploma thesis is focused on comparison of two political approaches to a quite new e-commerce industry -- internet gambling. These approaches are prohibition on the one hand and regulation, free market environment, on the other hand. Internet gambling became the worldwide phenomenon. However, American legislators had passed the Unlawful Internet Gambling Enforcement Act 2006 in 2006 which outlawed an internet gambling. The declared purpose of this Act is protection of families against ill effects, such money laundering, underage gambling and problem gambling on society. This paper argues that regulation and free market environment established in Great Britain is much more effective way how to solve these key issues. There is demonstrated in the paper that free market stands for economically preferable option and contains better instruments for solving the issues at the same time.

The thesis deals with issues of legalization of proceeds of crime in the Czech Republic. The theoretical part is focused on defining terms and analysis of the general characteristics of organized crime, including its development since the late 20th century to the present. Further detail is aimed at analysis of the crime of money laundering focusing on stages of the process of money laundering.
The practical part describes the recover the proceeds of crime and compares previous and amended legal provisions concerning this issue. Further analyzes the statistical data of economic crime in the Czech Republic between 2008 and 2015, with a forecast of the years 2016 to 2018 and describes the analysis of actual cases of money laundering. At the end of the work deals with a summary findings and compare the characteristic features of the process of money laundering with the real practices recorded on specific cases.

Doctoral thesis deals with professional laundry care process and reduction of energy intensity and use of other operational commodities used in the process. Professional laundry care process is a specific energy intensive process; its purpose is to re-establish original properties of the now dirty laundry so that it may fulfil its functions again. This is basically a recycling and regeneration process comprising many sub-operations. Key operation is washing of the laundry in a washing bath, which removes impurities from the laundry. This thesis describes the most common technology of impurities removal, which is washing in a hot washing bath in cooperation with mechanical work and detergents. However, all of the described procedures may also be applied on alternative technologies, such as ozone washing, washing with low-temperature enzymes and ultrasound. Water is applied in all these technologies as a soaking agent. Water must have appropriate properties and must be removed from the laundry after the washing process in subsequent operations (drying, ironing, and pressing). Theoretical introduction of the thesis describes professional laundry care process in detail, and continues with mathematical modelling of the process and its main sub-operations (washing, drying) using mass and heat balance, and data acquisition and their application in research and development. Thesis further describes design of information system on the basis of relational database controlled by web application. This system enables to organize data and information from various sources – measured experimental data, data concerning measurement devices, operational records of machines, etc. Thesis further proposes how to evaluate and reduce energy intensity of a process. Descriptions are validated by examples and case studies. Key section of the thesis is a chapter describing experimental infrastructure. This infrastructure integrates several instalments of laundry equipment and is a fully functional operation of industrial laundry facility with 500kg/shift capacity, which serves as a model of energy intensive process. Sophisticated infrastructure and data acquisition system allows measuring wide spectrum of physical entities and examining factors influencing energy intensity of processes, and relationships between these factors as well as effectiveness of various savings measures in real life facilities.

In the original script of “Twelfth Night” by Shakespeare, Viola was shipwrecked and survived. Being separated from her twin brother, Sebastian, Viola decided to disguise as a boy to make a living. However, the disguise made her fall into an unusual triangular relationship with sexual confusions. This difficult situation was eventually solved perfectly in a happy ending with the appearance of Sebastian. In the performance of “12,” my character, An-an, was evolved from Viola. I think the most challenging tasks in this performance are how to “reproduce” the character in a “variation” of the original play; how to handle the “light” quality given to the character in this “variation;” and how to break the habits in my past acting and thus review the nature of acting and methods of rehearsal works in preceding attempts and adjustment. “Twelfth Night” has been praised as the “most successful comedy by Shakespeare.” The name “Twelfth Night” refers to the Epiphany in the medieval times. Therefore, the implications of “carnival atmospheres” are thus self-evident. This report will start from this point to study the western tradition of “carnival” and to trace the source of the spirit of clowns that have kept shifting between the duality of “man/god” back into the rituls of Dionysia: subversion and revelry. This context will be extended to discuss the “organic” performing state which was sought by the director in the “12,” which was born out of the “Twelfth Night.” It was an attempt to find a “free and unrestrained” acting style. After that, the report will examine the working journals to discuss the methods of works in the rehearsal progress, and will then analyze and review the problems which I faced during the performance and the ways in which I dealt with these problems.

Money laundering can be defined as a process of converting the proceeds of illegal activities to legitimate money or other assets, or as a process whose aim is to conceal the origin and ownership of proceeds that come from illegal activities. Its main phases are placement, layering and integration. The main sources of dirty money are drug trafficking, procuring, arms trafficking, extortion, robbery and property crimes. The aim of this thesis is the analysis of the fight against money laundering in terms of financial law and from institutional point of view, evaluation of its success and the formulation of my own opinions on the possible improvement of the system components. The first part of thesis deals with the term of money laundering, its phases and characteristics and history. The second part analyses the legislation at international, EU and national level. Attention is paid to the AML Directives and to the AML Act. Main obligations prescribed by Directives and AML Act to obliged persons are identification obligation, due diligence, preservation of information, reporting of suspicious transactions, postponement of client's order, creation of internal policies and staff training. Money laundering is marginalized phenomenon with high social impact which comes from necessary connection of this activity with illegal activities, especially organized crime. Since the goal of almost every illegal activity is to create profit (and since the biggest profit comes from the activities which are highly dangerous for the society, such as drug trafficking) it is vital for the society to fight money laundering effectively. It is in the best interest of the whole society to fight this phenomenon and cut the organized crime out of their resources through effective control of the money laundering. “Kuso” is part of the meaningful spirit of prank Internet games within the recent sub-culture of the younger generation. Many prankish works can be found in the networks. Originally it played a prankish role in the sub-culture and has since become a fashionable type of language. It reflects the resistance to both popular culture and traditional dogma. “Kuso” is a new belief regarding self-creation and exhibits a rebellious attitude on the part of youth. It overwhelms classical conceits. By imitating and satirizing the original creation, the copied work undermines or reconstructs the habitual significance that we have in our minds, and conveys another mode of originality. Based on the black circus, the study aims to understand the self-reflection within the creative process, by analyzing the related literatures and various works. After determining the prankish intention, four series of works are sub-divided in the study: unreasonable fairy tales, the clownery of a famous brand, funny cartoons on TV or in film and the overwhelming of ingrained responses to classical painting. However, while “Kuso” is not a regular creative behavior, it is a continuity of creation. People have been conditioned to follow the rules since they were little and their thoughts have become inflexible. The study presents diverse viewpoints for wider observation It is hoped that new thinking can be inspired and that greater originality can be generated.

The role of Prana in man was recognised and venerated since time immemorial. In the
introduction, Prana is perceived as a global, anthropological phenomenon.
Chapter one provides a theoretical framework linking Pranato Oral Tradition by focusing
on Marcel Jousse and his teachings; the Orality Perspective and the Orality - Literacy
interface. Since the earliest record of the terms; yoga, Prana and Pranayama appear in
the Vedas, the significance of the Vedas, especially as they exemplify oral expression, is
mentioned.
Chapter two focuses on: firstly, Prana within the context of yoga and secondly, Prana
and its relation to the Aramaic Rouhah. The congruency of thought on the breath
suffices to entrench Prana as a global entity.
In chapter three, the enhancement of the quality and quantity of Prana via Pranayama
(control of the breath) is discussed. Pranayama sets out with the purpose of achieving
complete harmonisation of the body-mind axis, and thereby proceeding to the
divinisatlon of energy.
A cohesive whole is established by three essential and integrated elements; the breath
(Prana), the physical gesture (asana) and the spoken gesture (mantra) in Surya
Namaskar. Oral features in these elements are analysed in chapter four.
The conclusion serves to emphasize that the wealth of oral tradition is amply evident in

Financial Aspects of Alternative Payment Systems from the Point of View of Money Laundering Abstract The aim of this thesis is to provide a general overview of the current state of alternative payment systems with regard to their inclusion in the financial market, their properties and potentials for wider use, and to evaluate their possibilities to more effectively combat money laundering, terrorist financing and the proliferation of weapons of mass destruction. In the first chapter, the thesis explains the broader context with regard to financial systems, especially within the money market systems focusing on retail, while providing a deeper explanation of the context of European law and Czech law. The second chapter is focused on closer analysis of alternative payment systems with regard to their use. Alternative payment systems are divided into two basic categories for centralized alternative payment systems and decentralized alternative payment systems. ‘Indigenous community mapping’, a new concept and practice in the fields of both indigenous community development and natural resource management, is getting more attention during recent years in Taiwan. Its emergence provides a possibility to bridge two independent and controversial issues: indigenous autonomy movement and the campaign for a new proposed Maqaw national park (i.e. new national park movement), respectively. With different social agents involved, the measure of indigenous community mapping plays a role in both of the issues as a resisting space. Adopting an action research approach to promote indigenous community mapping in aboriginal villages around the proposed national park, this thesis aims to discuss the process and function of ‘indigenous community mapping’ applied in the linkage between indigenous movement and nature conservation movement. By reconstructing the human being-land relationship that promotes sustainable local development, indigenous community mapping is used as a communicating measure, which helps organize and unite local villagers to construct the local resisting space against oppressive structures of modernization, such as the nation state. This thesis first suggests that the mechanism of local resistance comes from the ‘articulation’ of local tradition/ culture and contemporary industry development, while the former provides the basis of ecological knowledge, local identity and normative force that are needed in local autonomous development. Second, environmentalist viewpoint, which emphasizes local participation of resource management and sustainable use of natural resource, is also an effective element that helps to construct the local resisting space.

The Crime of Money Laundering under Section 216 of the Czech Criminal Code Abstract The diploma thesis aims to examine the crime of legalization of proceeds of criminal activities in its narrower sense, i.e. the crime under S. 216 (2) of the Czech Criminal Code. This crime is referred to as "the crime of money laundering" throughout the thesis, as opposed to the crime of possession of stolen goods under S. 2016 (1) of the Czech Criminal Code. The opening chapter reviews the terminology used by Czech law. Then the thesis briefly addresses the phenomenon of money laundering in a broader context; this chapter also provides with statistics of criminal prosecutions for the named crime in the Czech Republic. I will cite important references from some scholars who have had direct or indirect contact with the Moscow Art Theater, revealing little known aspects on how yoga was used by Stanislavsky on his own training and that of his actors.

This thesis deals with the issue of money laundering in terms of both criminal and criminological perspective. The aim of this thesis is to analyse the issue of money laundering and to raise questions related to de lege lata discussions and de lege ferenda proposals, to formulate my opinion and to make recommendation for legislative changes. The first part (Chapters 1, 2 and 3) contains the definition of this term and discusses methods in which money laundering occurs. In addition to well-known methods, it is dedicated to new methods focusing on the use of virtual currencies and online computer games. The question of organised crime, which is very close to money laundering, is analysed too. The dissertation thesis thoroughly examines the crime of legalization of proceeds of criminal activities (the crime of money laundering) as it is stipulated in the contemporary Czech Criminal Code. The crux of the thesis is an analysis of the most significant elements of the crime of money laundering that are legal objects (general values protected by criminal law), actus reus (physical elements of a crime), mens rea (mental elements of a crime) and offender (person who commits a crime). Within the framework of that analysis, all of the important and disputed questions relating to this crime are examined, e.g. a complex problem of legal objects of the crime of money laundering or a question of liability for so-called self-money laundering. Il y a 4 isoforme de p38 : α, β, δ, and γ. MK5, à l'origine identifié comme étant un régulateur de PRAK (Regulated/Activated Protein Kinase), est maintenant connu pour être activée par la protéine kinase p38 (qui est un mitogène activé par la protéine kinase, MAPK). Cette dernière est impliquée dans les mécanismes de fibrose et d'apoptose pendant l'hypertrophie cardiaque. De plus, MK5 est également activée par les MAPKs atypiques; ERK3 et ERK4. Bien qu’elles soient fortement exprimées dans le coeur, le rôle physiologique de MK5 et ERK3 demeure inconnu. Par conséquent, nous avons étudié l'effet de la constriction aortique transversale (TAC) – induisant un surcharge chronique de pression chez les souris hétèrozygotes knockout pour MK5 (MK5+/-) ou ERK3 (ERK3+/-) et pour leurs types sauvages (MK5+/+ et ERK3+/+).
Deux sem post-TAC; le ratio de poids du coeur/poids corporel a été augmenté chez les 2 souris MK5+/- et MK5+/+. L'échocardiographie de la trans-thoracique démontre que la surcharge de pression a altéré la fonction diastolique du ventricule gauche chez MK5+/+, mais pas chez la souris MK5+/-. De plus, nous avons observé moins de dépôt de collagène, évalué par une coloration au trichrome de Masson, 2 et 3 sem post-TAC chez les souris MK5+/-. Parallèlement, le niveau de l’ARNm de collagène type1 alpha-1 a été significativement diminué dans les coeurs des souris MK5+/-, 2 et 3 sem post-TAC. De même, ERK3, mais pas ERK5 ni p38α, co-IP avec MK5 dans les 2 modèles des coeurs TAC; aigus ou chroniques. En revanche, l’ajout exogénique de GST-MK5 a abaissé ERK4 et p38α, mais pas ERK3 dans les lysâtes de coeur de souris. Par contre, GST-ERK3 et GST-p38α ne démontrent aucune co-IP avec MK5. Ces données suggèrent que dans le coeur seul ERK3, et non ERK4 ou p38α, est capable d’interagir avec, et réguler MK5. A niveau physiologique MK5 interagit entièrement avec ERK3 et par conséquent MK5 n’est pas disponible pour lier les protéines exogéniques. Les souris hétérozygotes pour ERK3 (ERK3+/-) ont également démontré une réduction ou une absence de collagène et une faible expression d’ARNm du collagène type1 alpha1, 3 sem post-TAC. Ces résultats démontrent un important rôle pro-fibrotique de la signalisation MK5-ERK3 pendant une surcharge chronique de pression.Nous avons également démontré 5 variant d'épissage de (MK5.1-5), y compris la forme originale (MK5.1). MK5.2 et MK5.5 subissent une délétion de 6 paires de base dans l’exon 12 : MK5.3 manque l'exon 12 : MK5.4 et MK5.5 manquent les exons 2-6. L'expression des ARNm des différents variant d'épissage a été vérifiée par PCR en temps réel (qPCR). Bien que l’expression est ubiquitaire, l'abondance relative de chaque variant était tissu-spécifique (coeur, rein, pancréas, muscle squelettique, poumon, foie, et cerveau). En plus, l'abondance relative des variant d’épissage varie pendant la surcharge de pression et le développement postnatal du coeur. En outre, l'immunofluorescence a indiqué que MK5.1-5.3 se localise au noyau alors que MK5.4-5.5 est situé au niveau cytoplasmic dans les cellules HEK 293 non stimulées. Suite à une stimulation avec l'anisomycin, un activateur de p38 MAPK, MK5.1-5.3 se translocalise du noyau au cytoplasme alors qu’une petite fraction de MK5.4-5.5 translocalise vers le noyau. Ces variant d'épissage peuvent diversifier la signalisation de MK5-ERK3 dans coeur, mais leur rôle exact oblige des recherches supplémentaires.
Excepté l’isoforme δ, toutes les isoformes de p38 sont exprimées dans le coeur et la forme α est considérée comme étant l'isoforme dominante. L’analyse par qPCR et immunobuvardage de type western ont démontré que p38α et p38γ sont les deux isoformes prédominantes alors que p38β et p38δ sont exprimées aux mêmes niveaux dans le coeur de rat adulte. L'immunofluorescence a démontré que p38α et p38γ se trouvent dans le cytoplasme et le noyau. Cependant, suite à la surcharge par TAC, p38γ s'est accumulé dans noyau tandis que la distribution de p38α est demeurée inchangée. Ainsi, l'abondance de p38γ et sa translocalisation nucléaire suite à la surcharge de pression indique un rôle potentiel dans l'expression génique pendant le remodelage cardiaque.
En conclusion, nous avons mis en évidence pour la première fois un rôle pro-fibrotique pour la signalisation MK5-ERK3 pendant une surcharge chronique de pression. D'ailleurs, les niveaux comparables d'expression de p38γ avec p38α, et la localisation différentielle de p38γ pendant la surcharge aiguë ou chronique de pression suggèrent différents rôles possibles pour ces isoformes pendant le remodelage hypertrophique cardiaque.There are 4 isoforms of p38 MAP kinase: α, β, γ, and δ. p38 signaling has been implicated in fibrosis and apoptosis during cardiac hypertrophy. MK5, originally identified as a p38 Regulated/Activated Protein Kinase (PRAK), is known to be downstream of p38 mitogen activated protein kinase (MAPK). Although highly expressed in the heart, the physiological roles of MK5 remain unknown. To determine if MK5 plays a role in mediating detrimental effects downstream of p38, we studied the effect of transverse aortic constriction (TAC)-induced chronic pressure overload in mice heterozygous for a knockout of MK5 (MK5+/-). Moreover, as MK5 is also activated by the atypical MAPKs, ERK3 and ERK4, the effects of TAC were also studied in ERK3+/- mice. Wild-type (MK5+/+; ERK3+/+) littermates were used as controls. Two wks post-TAC, heart weight/body weight ratios were significantly and similarly increased in both MK5+/- and MK5+/+ hearts. Trans-thoracic echocardiography revealed that pressure overload impaired left ventricular diastolic function in MK5+/+, but not in MK5+/- hearts. In addition, less collagen deposition, assessed by Masson trichrome staining, was observed in MK5+/- hearts 2 and 3 wks post-TAC. Furthermore, TAC-induced increases in collagen alpha1 type1 mRNA levels were significantly lower in MK5+/- hearts at both 2 and 3 wks post-TAC. Immunoprecipitation of MK5 resulted in co-immunoprecipitation of ERK3 but not ERK4 or p38α in either acute or chronic sham-operated and TAC hearts. In contrast, exogenous GST-MK5 pulled down endogenous ERK4 and p38α, but not ERK3, from mouse heart lysates. Neither exogenous GST-ERK3 nor GST-p38α pulled down MK5. These results suggest that MK5 associates with, and is regulated by ERK3, but not ERK4 or p38α in heart. At physiological expressional levels, all MK5 was bound to ERK3 and hence not available to bind exogenous protein. Along similar lines, mice heterozygous for an ERK3 knockout (ERK3+/-) also showed reduced or absent collagen deposition and lower collagen alpha1 type1 mRNA levels 3 wks post-TAC. This data suggests an important pro-fibrotic role of MK5-ERK3 signaling during chronic pressure overload.We also demonstrated the existence of 5 splice variants of (MK5.1-5), including the originally published form (MK5.1). MK5.2 and MK5.5 had a 6 base pair deletion in exon 12: MK5.3 lacked exon 12: and MK5.4 and MK5.5 lacked exons 2-6. Subsequently, expression of the splice variants at the mRNA level was quantified by real time qPCR. Although ubiquitously expressed, the relative abundance of each variant was tissue-specific (heart, kidney, pancreas, skeletal muscle, lung, liver, and brain). Additionally, the relative abundance of MK5 splice variants changed in the heart during pressure overload and post-natal development. Furthermore, immunofluorescence revealed MK5.1-5.3 localized to the nucleus and MK5.4-5.5 to the cytoplasm in unstimulated HEK 293 cells. Upon stimulation with anisomycin, which activates p38 MAPK, MK5.1-5.3 translocated from the nucleus to the cytoplasm and small amounts of MK5.4-5.5 relocated to the nucleus. These splice variants may further diversify MK5-ERK3 signaling in the heart, but their exact role awaits further investigation. With the exception of p38δ, all p38 isoforms are expressed in the heart and α is considered to be the prominent isoform in this tissue. qPCR and western blot analysis revealed p38α and p38γ to be the predominant isoforms and p38β and p38δ are expressed at comparable levels in the adult heart. Confocal immunofluorescence studies revealed p38α and p38γ in both the cytoplasm and nucleus. However, in response to TAC, p38γ accumulated in the nucleus whereas the distribution of p38α remained unaffected. The high abundance of p38γ and its nuclear accumulation during chronic pressure overload suggest that this isoform may play a role in gene expression during pathological cardiac remodeling. In conclusion, we have shown for the first time a pro-fibrotic role for MK5-ERK3 signaling during chronic pressure overload. Moreover, comparable expression levels of p38γ with p38α, and differential localization of p38γ during acute or chronic pressure overload, suggest these isoforms play different roles during cardiac remodeling.

The subject of my thesis is the identification and subsequent comparison of legal instruments which are intended to fight against legalization of proceeds of crime, which is also referred to as money laundering, in the Czech Republic and in the Federal Republic of Germany. The first chapter is to familiarize the reader with the issue of money laundering, therefore it provides the definition of the phenomenon of money laundering and also describes the typical phases of this process. The second chapter is devoted to international institutions that deal with money laundering on the supranational level and whose activities are then reflected in international standards. The third chapter deals with legislation related to the fight against legalization of proceeds of crime in the Czech Republic. First I focus on the history of the development of the struggle against money laundering in the country and subsequently describe particular laws designed to regulate the rights and obligations relating to the effective fight against this phenomenon. Further I describe particular obligations that fall on subjects, which may be confronted with efforts to launder money within its activities, and characterize individual institutions that are involved in the fight against money laundering. For a better understanding of relevant issues, in the introduction of the thesis contains a brief historical excursus to that topic, and there are explained the general features of the process of legalization of proceeds from crime, including putting illustrative examples from the area of the criminal law, as well as in the area of financial law. As examples have been selected major bank cases which were related to AML/CFT, but also had much wider impact. The diploma thesis deals with measures against the legalization of proceeds from crime in area of banking and credit institutions and focuses primarily on the measures arising from major law Act no. 253/2008 Coll., on certain measures against the legalization of proceeds from crime and terrorist financing. Consequently, the thesis deals partly with law of the European Communities, which was reflected into national legislation by the Act no. 253/2008 Coll. Following this, the diploma thesis also mentions other sources that have been adopted not only at national level but also in the context of international law.
