Full textThe current network of political activism has pursued a hybrid field, ignoring the borders between activism and art. Amongst the various genres of this network, the prank illustrates how such borders are blurred. A prank is a blow, a whimsical breaking of order, of hierarchy, and of authority. This thesis addresses prank as a practice, a performance option for political activism, provided that it merges experiences of play, criticism, liveness and dissemination. Activists who have chosen the prank are related with a recent history of the contemporary network of political mobilization, which resists to a new capitalist pattern of organization. From 1994 through 2003, this network established a scenario of celebrations and radical macro-actions (particularly the Days of Global Action). The spread out activism that is blended in daily life also features a similar invasive attitude. This is especially true in their claims for space, broadly understood both as urban space that has been contaminated by the invasion of capital, and as a space of communication, ideology and symbolic exchange. I have examined the various prank genres within this context on the basis of categories such as theatricality and performativity, according to Josette Férals stance. These considerations have led me to the conclusion that there are no mediators when prank merges with performance, since it is autonomous and draws on the do it yourself political principle.

Full textThis research work is developed with the implementation of implementing mindfulness sessions at a corporate level focused on Top and Middle Management, a different alternative that is operative and economically factible. The motivation to develop this project is given by current trends in physical and emotional health care through stress management (Euromonitor, 2019) due to changes in the work and personal environment. However, the use of the methodology used is based on the Canvas Model determined the most important points of the visual form. Such information was validated through in-depth interviews by human resources experts and meditation experts, which confirmed the target audience with an interest in physical and emotional well-being. In addition, the company has an administrative office in the Miraflores district. On the other hand, in the economic evaluation of the business plan, a return of s / 57 512.32 is obtained. The results found an initial investment of s /. 103 490 of which 50% is own capital and 50% will be financed. Considering an expected scenario with the annual sale of 80 packages annual and the attendance of 5 average people for practices of 5 sessions per program, obtain an annual net profit of s /. 74 810.65 in the first year. Considering the different aspects only in the pessimistic one obtains TIR of 51%, in the optimistic obtains131% and the TIRE obtains of 43.77%. The economic flows show positively from the first year, being in 2021 of s/.74 810.65, reaching s/. 60 269.10 to 2022 and s/. 66 247.67 to 2023. Regarding financial flow, s/. 54 515.45 in the year 2021 and reaching s/ .43 378.64 by 2023.

Full textThis thesis examines the performance of political pranks by contemporary radical activists and anarchists. Pranks, used symbolically to subvert authority and collapse hierarchy, have become important tools for grassroots political movements. Activists utilize pranks as a form of "culture jamming" to undermine, humiliate, and educate. This thesis documents political pranks pulled by Earth First!, the Yippies, the Biotic Baking Brigade, and the Eugene Anarchists for Torrey (EAT) Campaign to show how pranks are performed and narrated within anarchist subcultures. Drawing on cultural and performance studies, as well as on anarchist theories, this thesis demonstrates how pranks can become performances of resistance and criticism that disrupt the status quo.

Full textThe Poxviridae encode a unique class of proteins characterized by ANK repeats and PRANC motif in C-terminus. This study presents characterization of 3 of the 4 ANK-PRANC proteins encoded by myxoma virus (MYXV): M148R, M149R and M150R (MNF), organized in cluster. This work show that they are sequentially expressed during replication cycle and that they present original sub-cellular localizations. In vivo, M148R, M149R and MNF are MYXV virulence factors although they are not likely to be implicated in cell tropism or host range functions. Moreover, histological analysis suggests that M148R and MNF play a role in the subversion of host inflammatory response by MYXV. Furthermore, we show that M148R, M149R and MNF are components of SCF complexes (Skp, Cullin, F-box), allowing ubiquitination of a substrate that it remains to identify

Full textThis master´s thesis consists of two parts. The first part is a study where the possibility to use existing tools from the market is assessed. The second part describes the design, development and usability test of a web based hardware configuration tool. The usability test made in the design process, was made in order to remove common usability problems and to assess whether or not the design decisions made during the thesis were good. The usability test that was performed followed the think aloud method and gave me very useful information that helped me improve the tool.
The study that was made in the beginning of the thesis was done in order to see if there were any programs on the market which already did the required tasks or could be adjusted to do them. I could not find a satisfying alternative amongst the five alternatives investigated. This lead to the decision to design a new tool. The main functions are the ability to save the current state the hardware is in, and at a later stage load this configuration back.
The tool was designed for the PRAN unit at Ericsson AB, in Linköping, Sweden.

In the original script of “Twelfth Night” by Shakespeare, Viola was shipwrecked and survived. Being separated from her twin brother, Sebastian, Viola decided to disguise as a boy to make a living. However, the disguise made her fall into an unusual triangular relationship with sexual confusions. This difficult situation was eventually solved perfectly in a happy ending with the appearance of Sebastian. In the performance of “12,” my character, An-an, was evolved from Viola. I think the most challenging tasks in this performance are how to “reproduce” the character in a “variation” of the original play; how to handle the “light” quality given to the character in this “variation;” and how to break the habits in my past acting and thus review the nature of acting and methods of rehearsal works in preceding attempts and adjustment. “Twelfth Night” has been praised as the “most successful comedy by Shakespeare.” The name “Twelfth Night” refers to the Epiphany in the medieval times. Therefore, the implications of “carnival atmospheres” are thus self-evident. This report will start from this point to study the western tradition of “carnival” and to trace the source of the spirit of clowns that have kept shifting between the duality of “man/god” back into the rituls of Dionysia: subversion and revelry. This context will be extended to discuss the “organic” performing state which was sought by the director in the “12,” which was born out of the “Twelfth Night.” It was an attempt to find a “free and unrestrained” acting style. After that, the report will examine the working journals to discuss the methods of works in the rehearsal progress, and will then analyze and review the problems which I faced during the performance and the ways in which I dealt with these problems.

“Kuso” is part of the meaningful spirit of prank Internet games within the recent sub-culture of the younger generation. Many prankish works can be found in the networks. Originally it played a prankish role in the sub-culture and has since become a fashionable type of language. It reflects the resistance to both popular culture and traditional dogma. “Kuso” is a new belief regarding self-creation and exhibits a rebellious attitude on the part of youth. It overwhelms classical conceits. By imitating and satirizing the original creation, the copied work undermines or reconstructs the habitual significance that we have in our minds, and conveys another mode of originality. Based on the black circus, the study aims to understand the self-reflection within the creative process, by analyzing the related literatures and various works. After determining the prankish intention, four series of works are sub-divided in the study: unreasonable fairy tales, the clownery of a famous brand, funny cartoons on TV or in film and the overwhelming of ingrained responses to classical painting. However, while “Kuso” is not a regular creative behavior, it is a continuity of creation. People have been conditioned to follow the rules since they were little and their thoughts have become inflexible. The study presents diverse viewpoints for wider observation It is hoped that new thinking can be inspired and that greater originality can be generated.

‘Indigenous community mapping’, a new concept and practice in the fields of both indigenous community development and natural resource management, is getting more attention during recent years in Taiwan. Its emergence provides a possibility to bridge two independent and controversial issues: indigenous autonomy movement and the campaign for a new proposed Maqaw national park (i.e. new national park movement), respectively. With different social agents involved, the measure of indigenous community mapping plays a role in both of the issues as a resisting space. Adopting an action research approach to promote indigenous community mapping in aboriginal villages around the proposed national park, this thesis aims to discuss the process and function of ‘indigenous community mapping’ applied in the linkage between indigenous movement and nature conservation movement. By reconstructing the human being-land relationship that promotes sustainable local development, indigenous community mapping is used as a communicating measure, which helps organize and unite local villagers to construct the local resisting space against oppressive structures of modernization, such as the nation state. This thesis first suggests that the mechanism of local resistance comes from the ‘articulation’ of local tradition/ culture and contemporary industry development, while the former provides the basis of ecological knowledge, local identity and normative force that are needed in local autonomous development. Second, environmentalist viewpoint, which emphasizes local participation of resource management and sustainable use of natural resource, is also an effective element that helps to construct the local resisting space.

Full textIn this work I intend to show the influence of yoga on the artistic life of Stanislavsky. I will parallel Stanislavsky's way of thinking with the thinking involved in yoga with regard to life, the actor, the work ethic, and the physical, mental, and spiritual development of the human being. I will discuss the energy (prana) concepts of yoga origin used by Stanislavsky in the preparation of the actor and in the construction of the character. This concept of prana and its use through correct breathing must have been one of the main components that Stanislavsky used to achieve his main goal which was to reveal a human spirit and to show it in artistic and beautiful form. I will raise the question of whether it is actually useful to use yoga techniques in the preparation of the actor and in the construction of the character, considering that yoga provides physical, mental and spiritual work. I will cite important references from some scholars who have had direct or indirect contact with the Moscow Art Theater, revealing little known aspects on how yoga was used by Stanislavsky on his own training and that of his actors.

Full textThere are 4 isoforms of p38 MAP kinase: α, β, γ, and δ. p38 signaling has been implicated in fibrosis and apoptosis during cardiac hypertrophy. MK5, originally identified as a p38 Regulated/Activated Protein Kinase (PRAK), is known to be downstream of p38 mitogen activated protein kinase (MAPK). Although highly expressed in the heart, the physiological roles of MK5 remain unknown. To determine if MK5 plays a role in mediating detrimental effects downstream of p38, we studied the effect of transverse aortic constriction (TAC)-induced chronic pressure overload in mice heterozygous for a knockout of MK5 (MK5+/-). Moreover, as MK5 is also activated by the atypical MAPKs, ERK3 and ERK4, the effects of TAC were also studied in ERK3+/- mice. Wild-type (MK5+/+; ERK3+/+) littermates were used as controls. Two wks post-TAC, heart weight/body weight ratios were significantly and similarly increased in both MK5+/- and MK5+/+ hearts. Trans-thoracic echocardiography revealed that pressure overload impaired left ventricular diastolic function in MK5+/+, but not in MK5+/- hearts. In addition, less collagen deposition, assessed by Masson trichrome staining, was observed in MK5+/- hearts 2 and 3 wks post-TAC. Furthermore, TAC-induced increases in collagen alpha1 type1 mRNA levels were significantly lower in MK5+/- hearts at both 2 and 3 wks post-TAC. Immunoprecipitation of MK5 resulted in co-immunoprecipitation of ERK3 but not ERK4 or p38α in either acute or chronic sham-operated and TAC hearts. In contrast, exogenous GST-MK5 pulled down endogenous ERK4 and p38α, but not ERK3, from mouse heart lysates. Neither exogenous GST-ERK3 nor GST-p38α pulled down MK5. These results suggest that MK5 associates with, and is regulated by ERK3, but not ERK4 or p38α in heart. At physiological expressional levels, all MK5 was bound to ERK3 and hence not available to bind exogenous protein. Along similar lines, mice heterozygous for an ERK3 knockout (ERK3+/-) also showed reduced or absent collagen deposition and lower collagen alpha1 type1 mRNA levels 3 wks post-TAC. This data suggests an important pro-fibrotic role of MK5-ERK3 signaling during chronic pressure overload.We also demonstrated the existence of 5 splice variants of (MK5.1-5), including the originally published form (MK5.1). MK5.2 and MK5.5 had a 6 base pair deletion in exon 12: MK5.3 lacked exon 12: and MK5.4 and MK5.5 lacked exons 2-6. Subsequently, expression of the splice variants at the mRNA level was quantified by real time qPCR. Although ubiquitously expressed, the relative abundance of each variant was tissue-specific (heart, kidney, pancreas, skeletal muscle, lung, liver, and brain). Additionally, the relative abundance of MK5 splice variants changed in the heart during pressure overload and post-natal development. Furthermore, immunofluorescence revealed MK5.1-5.3 localized to the nucleus and MK5.4-5.5 to the cytoplasm in unstimulated HEK 293 cells. Upon stimulation with anisomycin, which activates p38 MAPK, MK5.1-5.3 translocated from the nucleus to the cytoplasm and small amounts of MK5.4-5.5 relocated to the nucleus. These splice variants may further diversify MK5-ERK3 signaling in the heart, but their exact role awaits further investigation. With the exception of p38δ, all p38 isoforms are expressed in the heart and α is considered to be the prominent isoform in this tissue. qPCR and western blot analysis revealed p38α and p38γ to be the predominant isoforms and p38β and p38δ are expressed at comparable levels in the adult heart. Confocal immunofluorescence studies revealed p38α and p38γ in both the cytoplasm and nucleus. However, in response to TAC, p38γ accumulated in the nucleus whereas the distribution of p38α remained unaffected. The high abundance of p38γ and its nuclear accumulation during chronic pressure overload suggest that this isoform may play a role in gene expression during pathological cardiac remodeling. In conclusion, we have shown for the first time a pro-fibrotic role for MK5-ERK3 signaling during chronic pressure overload. Moreover, comparable expression levels of p38γ with p38α, and differential localization of p38γ during acute or chronic pressure overload, suggest these isoforms play different roles during cardiac remodeling.
