The Pasteuria genus comprises gram-positive bacteria that are obligate parasites of arthropods and nematodes. Species of this genus are ubiquitous, being present in both aquatic and terrestrial environments all around the world. Pasteuria was first described as a genus at the end of the 19th century and has undergone considerable reclassification regarding its member species. Starting in the 1980s, a more meticulous classification effort regarding the identification of Pasteuria spp., and its parasitic habits began. These studies were strongly motivated by the ability of individuals of this genus to parasitize phytopathogenic nematodes of several plant species. Each species of the genus Pasteuria establishes a strict parasitic relationship with a specific genus of phytonematode. As an example of this interaction, Pasteuria thornei is a parasite restricted to the genus Pratylenchus, which comprises the nematodes popularly known as root-lesion-nematodes, a pest of several agronomically important crops. Considering the current relevance of studies involving the biological control of phytonematodes, in the present work three experiments were carried out, each one containing a replicate, totaling, therefore, six experiments. Two experiments were intended to verify the efficacy of P. thornei as a biological control agent (BCA) of Pratylenchus brachyurus in soybean. The remaining four experiments had a similar objective in the scope of the Pratylenchus zeae - maize pathosystem. Two experiments were carried out to verify the efficacy of P. thornei as a biological control agent for P. zeae in maize, and afterwards, two additional experiments were performed in order to verify the capacity of the BCA to reduce productivity losses in corn plants due to the parasitism of this nematode. For the soybean experiments, the following treatments were added to the seeds of the cultivar SYN1080: three different concentrations of P. thornei endospores per seed (5x106, 107 e 1,5x107), a commercial control group for comparison containing abamectin (0.58mg/seed) and a mixed treatment containing abamectin (0.58 mg / seed) and 107 P. thornei endospores. Untreated seeds were used as a control group. The treatments were sown in 500 cm3 plastic cups containing soil inoculated with 1000 nematodes (experiment 1) and 600 nematodes (experiment 2). Fresh root mass and nematodes extracted from the roots of each plant were used as parameters of evaluation, taking place 60 and 90 days after inoculation (DAI). Only the treatment with the highest concentration of P. thornei (1.5x107) reduced the final population of nematodes significantly, reaching 30-50% of reduction compared to the untreated seeds. However, treatments containing the commercial control abamectin were superior in reducing the final population of nematodes in all experiments evaluated. Regarding the maize efficacy experiments, CELERON hybrid seeds were treated as described: four concentrations of P. thornei endospores per seed (5x106, 107, 1,5x107, 2x107), a commercial control group for comparison containing abamectin (0.58 mg / seed) and a mixed treatment containing abamectin (0.58 mg / seed) and 107 P. thornei endospores. Untreated seeds were used as a control group. The treated maize seeds were planted in 500 cm3 plastic cups containing soil inoculated with 4000 and 1000 individuals for the efficacy experiments 1 and 2, respectively. Evaluations occurred at 60 and 90 DAI. For the productivity assays, the experiments 3 and 4 were carried out under a screened greenhouse, with experimental plots consisting of 9L pots filled with artificially infested soil. Seeds of the CELERON hybrid received the following treatments: abamectin (0.58mg / seed), P. thornei (107 endospores / seed) and mixed treatment containing both abamectin (0.58mg / seed) and P. thornei (107 endospores / seed). Two additional treatments containing untreated seeds served as controls, with and without the presence of Pratylenchus zeae. The evaluation measured several agronomic traits, such as dry weight of the aerial parts, fresh mass of roots at harvest and total weight of grains. In addition, the nematode population was measured in fresh roots at 45, 90 days and at the time of harvest. Efficacy trials showed that the highest concentrations of P. thornei (1.5x107 and 2x107) have a considerable potential of P. zeae control. The nematode population reduction was 54 and 47% in experiments 1 and 2, respectively, for the highest P. thornei concentration treatment. The commercial formulation containing abamectin showed a reduction of P. zeae population above 90% in both experiments. Regarding the maize productivity experiments, control potential of nematodes by P. thornei was similar to that observed in the efficacy study. The treatments containing abamectin had an effect on the mitigation of yield losses caused by P. zeae in both experiments. The mixed treatment (abamectin and P. thornei) and the one containing exclusively P. thornei presented a positive performance in both replicates. In none of the experiments synergistic or additive effects were observed between P. thornei and abamectin. With the data obtained in these experiments, the control potential of P. thornei on P. brachyurus and P. zeae in soybean and corn, respectively, is evident. Additionally, P. thornei and abamectin in the form of seed treatment, show potential in mitigating yield losses caused by P. zeae in maize. This highlights the importance of P. thornei as an additional tool for the management of root lesion nematodes in soybean and maize, and should encourage subsequent work.<br>O gênero Pasteuria compreende bactérias gram-positivas parasitas obrigatórias de artrópodes e nematoides. A distribuição das espécies deste gênero pelo mundo é ubíqua, podendo ser encontradas em ambientes aquáticos e terrestres. Este gênero foi descrito no final do século XIX e sofreu consideráveis reclassificações em relação às espécies nele compreendidos. A partir da década de 80, deu-se início a um esforço de classificação mais minucioso com relação à identificação de Pasteuria spp. e seus hábitos parasitários. Estes estudos foram motivados, principalmente, pela capacidade dos indivíduos deste gênero em parasitar nematoides fitoparasitas de diversas culturas. Cada espécie do gênero Pasteuria estabelece relações parasitárias com um gênero específico de fitonematoide. A exemplo desta interação, Pasteuria thornei é um parasita restrito ao gênero Pratylenchus, que compreende os nematoides causadores das lesões radiculares, daninhos a diversas culturas de importância agronômica. Considerando a relevância atual de estudos envolvendo o controle biológico de fitonematoides, no presente trabalho foram realizados três experimentos, cada um contendo uma réplica em época distinta, totalizando, portanto, seis experimentos. Dois experimentos tiveram por objetivo verificar a eficácia de P. thornei como agente de controle biológico (ACB) de Pratylenchus brachyurus na cultura da soja. E os demais quatro experimentos abordaram o patossistema Pratylenchus zeae-milho. Para esse objetivo, foram realizados dois experimentos com o intuito de verificar a eficácia de P. thornei como agente de controle biológico de P. zeae em milho, e outros dois experimentos para testar a capacidade do ACB em reduzir a perda de produtividade em plantas de milho decorrente do parasitismo do nematoide. Para os experimentos de soja, às sementes da cultivar SYN1080 foram adicionados os tratamentos como se segue: três concentrações de endósporos de P. thornei por semente (5x106, 107 e 1,5x107), um grupo de controle químico comercial para comparação contendo abamectina (0,58 mg / semente) e um tratamento misto contendo abamectina (0,58 mg / semente) e 107 endósporos de P. thornei. Sementes não tratadas foram utilizadas como testemunha. As sementes tratadas foram semeadas em copos de plástico de 500 cm3 contendo solo inoculado com 1000 nematoides (experimento 1) e 600 nematoides (experimento 2). A massa de raiz fresca e os nematoides extraídos das raízes de cada planta foram utilizados como critério de avaliação dos experimentos, a qual foi realizada aos 60 e 90 dias após a inoculação (DAI). Apenas o tratamento com a maior concentração de P. thornei (1,5x107) reduziu a população final de nematoides de maneira significativa atingindo 30-50% de redução, comparado àquele contendo sementes não tratadas. No entanto, os tratamentos que contém abamectina foram superiores na redução da população final de nematoides em todos os experimentos avaliados. Em relação aos experimentos de eficácia em milho, sementes do híbrido CELERON foram tratadas como explicitado: quatro concentrações de endósporos de P. thornei por semente (5x106, 107, 1,5x107 e 2x107), um grupo de controle comercial para comparação contendo abamectina (0,58 mg / semente) e um tratamento misto contendo abamectina (0,58 mg / semente) e 107 endósporos de P. thornei. As sementes tratadas de milho foram semeadas em copos de plástico de 500cm3 contendo solo inoculado com 4000 e 1000 indivíduos para os experimentos de eficácia 1 e 2, respectivamente. As avaliações ocorreram aos 60 e 90 DAI. Para os estudos de produtividade, foram realizados os experimentos 3 e 4 sob um telado com parcelas experimentais constituídas por vasos de 9L preenchidos de solo infestado artificialmente. Sementes do híbrido CELERON foram utilizadas contendo os seguintes tratamentos: abamectina (0,58mg / semente), P. thornei (107 endósporos/semente) e um tratamento misto contendo abamectina (0,58mg / semente) e P. thornei (107 endósporos/semente). Dois tratamentos adicionais contendo sementes não tratadas serviram de testemunhas, com e sem Pratylenchus zeae. A avaliação consistiu na medição de várias características agronômicas, como peso seco da parte aérea, massa fresca de raízes no momento da colheita e peso total dos grãos. Adicionalmente, foi mensurada a população de nematoides em raízes frescas aos 45, 90 dias e no momento da colheita. Os ensaios de eficácia mostraram que as concentrações mais elevadas de P. thornei (1,5x107 e 2x107) possuem um potencial mensurável de controle de P. zeae. A redução da população de nematoides foi de 54 e 47% nos experimentos 1 e 2, respectivamente. A formulação comercial de abamectina mostrou uma redução da população de nematoides superior a 90% em ambos os experimentos. No que diz respeito aos experimentos de produtividade de milho, o potencial de controle de nematoides por P. thornei foi semelhante ao observado no estudo de eficácia. O tratamento com abamectina teve efeito na redução das perdas de rendimento causadas por P. zeae em ambos os experimentos; assim como os tratamentos misto (abamectina e P. thornei) e aquele contendo apenas P. thornei que apresentaram desempenho positivo em ambas as repetições. Em nenhum dos experimentos foi observado efeito sinérgico ou aditivo entre P. thornei e abamectina. Com os dados obtidos nestes experimentos, fica evidente o potencial de controle de P. thornei sobre P. brachyurus e P. zeae em soja e milho, respectivamente. Ainda, tanto P. thornei quanto abamectina apresentam o potencial de mitigar as perdas de rendimento causadas por P. zeae em milho através do tratamento de sementes. Isso evidencia a importância de P. thornei como uma ferramenta adicional para o manejo desses nematoides, e deve encorajar trabalhos subsequentes.

Os danos causados por Pratylenchus brachyurus e P. zeae às variedades de cana-de-açúcar SP70-1143 e SP71-1406 foram avaliados em dois ensaios em vasos de 150 litros. Cada ensaio constou da inoculação de 0, 200, 400 ou 800 nematóides por litro de solo de uma ou outra espécie de Pratylenchus. As populações de nematóides nas raízes e parâmetros de crescimento e produção foram avaliados ao longo do ciclo e por ocasião do corte, aos 17 meses. P. braghyurus se multiplicou nas raízes das duas variedades sem causar prejuízos a nenhuma delas, que foram consideradas tolerantes ao nematóide. Na variedade SP70-1143, P. zeae só se multiplicou nas plantas inoculadas com 200 nematóides por litro de solo, porém não causou reduções nos pesos frescos dos colmos e total da parte aérea, em relação às demais. SP71-1143 foi tida como resistente a P. zeae. Na variedade SP71-1406, as maiores populações de P. zeae foram registradas nas plantas que receberam 400 ou 800 nematóides por litro de solo. Estas plantas também mostraram reduções significativas nos pesos frescos dos colmos e total da parte aérea, em relação as demais. SP71-1406 foi considerada susceptível a P. zeae<br>Damage to two sugarcane varieties resulting from infestation by Pratylenchus brachyurus and P. zeae were evalueted in two experiments. Each experiment consisted of inoculation with 0, 200, 400 or 800 nematodes/l soil from one or another Pratylenchus species on the varieties SP70-1143 and SP71-1406, the most cultivated in São Paulo State, nowadays. Single buds from both varieties were grown in 150 l pots, containing sterilized sandy soil and were inoculated 30 days after planting. Nematode population in roots and parameters were evalueted along the plant growth cicle and at the harvest, 17 months after planting. P. brachyurus reproduction was observed at the roots of two varieties, but no growth reduction was caused by the parasities. Both varieties were considered tolerant to P. brachyurus. On SP70-1143, P. zeae reproduction ocurred only at initial population of 200 nematodes/l soil. At the harvest, these plants only showed significant decreases in fresh weight of top cane plus leaves and dry weight of roots when compared with the plants without nematodes. SP-1143 was classified as resistant to P. zeae. As far as SP71-1406 is concerned, the higher P. zeae populations were observed in the pots inoculated with 400 or 800 nematodes/l soil. In these cases, significant reduction on fresh weight of shoots and fresh weight of aerial parts observed. SP71-1406 was considered susceptible to P. zeae

There are several phytosanitary problems that affect the culture and cause losses in production, including plant nematodes. Pratylenchus brachyurus is one of the most widespread nematodes in soybean producing areas of Brazil. The search for genotypes that have resistance to this plant nematode has been widespread, but there is still disagreement regarding the behavior of several brazilian cultivars. Evaluation of soybean cultivars in infested areas has not indicated the existence of materials resistant or tolerant. Thus, this study was to evaluate the reaction to P. brachyurus of six soybean cultivars and eight F1 populations originated from crosses in partial diallel arrangement. We also evaluate the behavior of cultivars exposed to different inoculum concentrations in three planting dates. The first study aimed to evaluate the reaction of cultivars and F1 populations of the plant nematode P. brachyurus. Experiment were conducted in naturally infested field conditions to evaluate the cultivars MT/BR51 (Xingu), BRSGO Caiapônia, MSOY 8360RR, BRS 8560RR, Emgopa 313RR and MSOY 7639RR and another in greenhouse conditions, which were evaluated in addition to these cultivars, the F1 progenies obtained by crossing between the parents of group I (Xingu and BRSGO Caiapônia) and group II (MSOY 8360RR, BRS 8560RR, Emgopa 313RR and MSOY 7639RR). In the field experiment all cultivars were susceptible to plant nematode, except the cultivar Xingu that showed behavior of resistance in the evaluation at 60 days after planting (DAP) with a RF of 0,80. However, this cultivar enabled the multiplication of P. brachyurus in the evaluation at 30 DAP. In the greenhouse condition, all parents behaved as resistant. The diallel analysis revealed the presence of non-additive and additive effects in controlling resistance. The Xingu cultivar showed the best GCA and cross between the cultivars Xingu and MSOY 8360 RR was the most promising in generating progeny with resistance reaction. The second study aimed to evaluate the reaction of soybean cultivars BRSGO Caiapônia, MT/BR51 (Xingu), MSOY 8360RR, MSOY 7639RR, BRS 8560RR and Emgopa 313RR, subjected to different inoculums concentrations in three planting dates. For this, three experiments were conducted under greenhouse conditions to evaluate the reaction of soybean cultivars under initial inoculum concentrations of 200, 400 or 600 juveniles and/or adults of P. brachyurus per plant in three planting dates. The planting date interferes with the infection of P. brachyurus and in the development of plants. Temperatures recorded in the second planting date, probably favored the multiplication of the nematode resulting in the lowest root development of cultivars. In this planting date was observed the highest population density of the nematode. In the third planting date the average temperature of the environment presented milder. At this planting date it was noted that all cultivars had lower population densities of P. brachyurus and resistant behavior (RF<1). There was a trend towards growing the plant nematode population density increased as the inoculums concentration in the first and second planting dates. In their results, we can observe the vulnerability of Brazilian soybean genotypes to P. brachyurus and the existence of variability of the pathogen. More research is needed to identify genotypes that have resistance to this nematode, as well as population studies related to the nematode. These studies should take into account the annual variation of nematode population due to low infectivity observed in dates of warmer temperatures.<br>A soja é a cultura agrícola mais importante do Brasil, o qual se destaca por ser o segundo maior produtor mundial do grão. Diversos são os problemas fitossanitários que afetam a cultura e ocasionam perdas na produção, dentre eles os fitonematoides. Pratylenchus brachyurus é um dos nematoides mais disseminados na cultura da soja nas áreas produtoras do Brasil. A busca por genótipos de soja que apresentem resistência a este fitonematoide tem sido muito difundida, porém, há ainda divergência em relação ao comportamento de diversas cultivares brasileiras. A avaliação de cultivares de soja em áreas infestadas não tem indicado a existência de materiais resistentes ou tolerantes. Dessa forma, neste estudo avaliou-se a reação ao P. brachyurus de seis cultivares de soja e de oito populações F1 originadas por cruzamentos em esquema de dialelos parciais. Avaliou-se também o comportamento dessas cultivares, submetidas a diferentes concentrações de inóculo, em três épocas de plantio. O primeiro estudo teve como objetivo avaliar a reação das cultivares e das populações F1 ao fitonematoide P. brachyurus. Para isso, foram conduzidos dois experimentos, um em condições de campo naturalmente infestado, avaliando-se as cultivares MT/BR51 (Xingu), BRSGO Caiapônia, MSOY 8360RR, BRS 8560RR, Emgopa 313RR e MSOY 7639RR, e outro em condições de casa de vegetação, em que foram avaliadas, em esquema de dialelo parcial, além destas cultivares, as progênies F1 obtidas pelo cruzamento entre os genitores do grupo I (Xingu e BRSGO Caiapônia) e do grupo II (MSOY 8360RR, BRS 8560RR, Emgopa 313RR e MSOY 7639RR). No experimento de campo todas as cultivares comportaram-se como suscetíveis, com exceção da cultivar Xingu que apresentou comportamento de resistência na avaliação aos 60 dias após a semeadura (DAS) com um FR de 0,80. Porém, na avaliação aos 30 DAS esta cultivar possibilitou a multiplicação de P. brachyurus. Já em casa de vegetação, todos os genitores comportaram-se como resistentes. A análise dialélica evidenciou a presença de efeitos aditivos e não aditivos no controle da resistência. A cultivar Xingu apresentou a melhor capacidade geral de combinação e o cruzamento entre as cultivares Xingu e MSOY 8360RR foi o mais promissor em gerar progênies com reação de resistência. O segundo estudo teve como objetivo avaliar o comportamento das cultivares BRSGO Caiapônia, MT/BR 51 (Xingu), MSOY 8360RR, MSOY 7639RR, BRSG 8560RR e Emgopa 313RR submetidas a diferentes concentrações de inóculo de P. brachyurus, em três épocas de plantio. Para isso, foram conduzidos três experimentos sob condições de casa de vegetação, para avaliar a reação das cultivares com relação às concentrações iniciais de inóculo de 200, 400 ou 600 juvenis e/ou adultos de P. brachyurus por parcela, semeadas em três épocas diferentes. A época de semeadura interferiu na infecção de P. brachyurus e, consequentemente, no desenvolvimento das plantas. As temperaturas registradas na segunda época de semeadura, provavelmente, favoreceram a multiplicação de P. brachyurus ocasionando o menor desenvolvimento das raízes das cultivares. Nesta época observou-se a maior densidade populacional de P. brachyurus. Na terceira época, a temperatura média do ambiente apresentou-se mais amena, quando se notou que todas as cultivares apresentaram baixas densidades populacionais de P. brachyurus e comportamento resistente. Observou-se tendência de aumento na densidade populacional do fitonematoide à medida que se aumentou a concentração de inóculo, nas primeira e 5 segunda épocas de plantio. Com base nesses resultados observa-se a vulnerabilidade dos genótipos brasileiros de soja ao P. brachyurus e a existência de variabilidade do patógeno. Mais pesquisas são necessárias visando à identificação de genótipos que possuam resistência a este nematoide, assim como estudos populacionais relacionados ao P. brachyurus. Nestes estudos deve-se levar em conta a variação anual da população do nematoide em razão da baixa infectividade observada em épocas de temperaturas mais amenas.

Para um manejo bem sucedido, devem-se integrar diversas estratégias de controle. A utilização de cultivares resistentes é a maneira mais eficiente, porém as informações quanto às formas de avaliar a reação de cultivares são escassas. Devido à importância de Pratylenchus brachyurus e a falta de padronização de metodologias para avaliação de ensaios em casa de vegetação, objetivou-se estudar metodologias para seleção de genótipos de soja quanto à reação a P. brachyurus baseadas em diferentes níveis de inóculo e épocas de avaliação, bem como a contribuição da utilização de uma escala de notas em função da percentagem de escurecimento das raízes nas avaliações. Foram conduzidos dois ensaios em casa de vegetação na estação da GDM GENÉTICA DO BRASIL LTDA, em Cambé – PR, em 2015. No primeiro ensaio verificou-se a reação de 45 genótipos de soja a P. brachyurus aos 45, 60 e 90 dias após a inoculação de 600 espécimes/planta. Para o segundo foram selecionados nove dos genótipos estudados anteriormente e acrescentado o genótipo 46, não utilizado no primeiro ensaio e avaliados aos 45 e 60 dias após a inoculação de 800 e 1000 espécimes/planta. O delineamento experimental de ambos foi o inteiramente casualizado, em esquema fatorial, com cinco repetições. As variáveis avaliadas foram: fator de reprodução (FR), número de nematoides por grama de raiz (nem/g raiz) e escala de notas para escurecimento de raízes. Os dados foram submetidos à análise de variância e as médias comparadas pelo teste de Scott-Knott a 5% de probabilidade. Os dois ensaios apresentaram interação significativa entre os fatores, apontando dependência entre genótipo, época de avaliação e nível de inóculo para as variáveis respostas. No primeiro ensaio, houve diferença significativa entre os genótipos para FR e nema/g raiz quando se avaliou aos 60 e 90 dias após a inoculação, mas não aos 45 dias. De modo geral, maiores valores de FR e nem/g raiz foram obtidos na avaliação mais tardia, o que não se repetiu para nota, que apresentou maiores médias aos 60 dias. No segundo ensaio, aos 45 dias de avaliação não houve diferença significativa entre os genótipos para as três variáveis avaliadas, nos dois níveis de inóculo utilizados. Por outro lado, aos 60 dias, para FR e nem/g raiz os genótipos diferiram entre si, apenas no nível de 800 espécimes/planta. No entanto, de acordo com as notas de escurecimento, nos dois níveis de inóculo estudados, os genótipos apresentaram diferença estatística entre si. No entanto, não houve diferença de escurecimento entre os níveis de inóculo utilizado. Portanto, nas condições deste trabalho, não recomenda-se a avaliação aos 45 dias após a inoculação dos nematoides; recomenda-se a inoculação de 800 indivíduos por planta com avaliação aos 60 dias; e a escala visual de notas pode ser uma ferramenta importante para auxiliar na seleção de genótipos para programas de melhoramento; porém são necessários mais estudos em nível de campo.<br>For a successful management should be integrated various control strategies. The use of resistant cultivars is the most efficient way, but the information about the reaction of cultivars are scarce. Due to the importance of Pratylenchus brachyurus and the lack of standardization of methodologies for assessment of testing in the greenhouse, aimed to study methods for selection of soybean genotypes for resistance to P. brachyurus based on different levels of inoculum and evaluation periods, as well as the contribution of the use of a visual rating scale according to the percentage of dark roots in the ratings. Two experiments were conducted in a greenhouse in the GDM GENETICS OF BRAZIL LTDA station in Cambé - PR in 2015. In the first study was checking the reaction of 45 soybean genotypes to P. brachyurus at 45, 60 and 90 days after inoculation of 600 specimens / plant. For the second nine genotypes were selected previously studied, added the genotype 46, not used in the first test and evaluated at 45 and 60 days after inoculation of 800 and 1000 specimens / plant. The experimental design of both was completely randomized in a factorial design with five replications. The variables evaluated were: reproduction rate (FR), number of nematodes per root gram (nema / g root) and degree of root darkening. Data were submitted to analysis of variance and means were compared by the Scott-Knott test at 5% probability. The two trials showed a significant interaction between the factors pointing dependence between genotype evaluation time and inoculum level for the response variables. In the first trial, there were significant differences among genotypes for FR and nema / g root when evaluated at 60 and 90 days after inoculation, but not 45 days. In general, higher FR values and or / g root were obtained in later evaluation, which was not repeated to note, that showed higher averages 60 days. In the second trial, the 45-day trial there was no significant difference between genotypes for the three variables evaluated in two inoculum levels used. Furthermore, at 60 days, and even for FR / g root genotypes differed only in the level of 800 specimens / plant. However, according to the blackout notes, the two inoculum levels studied, the genotypes showed statistical difference between them. However, no difference between the dimming levels of inoculum used. Therefore, in the conditions of this work, not recommended to the evaluation at 45 days after inoculation of nematodes; it is recommended to inoculate 800 specimens per plant and evaluation at 60 days; and the visual rating scale can be an important tool to assist in the selection of genotypes for breeding programs; but more studies are needed in the field level.

Due to inadequate soil management and the fact that there is not efficient nematicide in the market. The, P. brachyurus known as the nematode root lesions, has been spreading rapidly and increased its importance in the Brazil fields. The aim of this study was to evaluate the effect of organic fertilizers in soil and liquid form in control of the nematode in soybean roots, on Fepagro 36RR.The design was a randomized block design with six treatments and nine replicates. There treatments were evaluated with fertilizers in solid and liquid form: soil+ vermicompost manure (VERM), soil+ compost (manure and straw, COMP), soil +organic compound Ecocitrus (ECO) and two control without fertilization, with the presence or absence of the nematode. Samples were collected at 25, 50 and 75 days after inoculation of phytonematode, evaluating height, fresh shoot and root mass the juvenils number of P. brachyurus in the roots. The results show that after 50 days, the treatments applied in solid form were suppressive to control P. brachyurus and development of soybean plants. 75 days, it is observed that plants grown with VERM were less affected, with the number of nematodes penetrated 13% and 26% lower than that observed in the ECO and COMP fertilizer. New tests should be conducted using compost of cattle manure, vermicompost and commercial waste to confirm the potencial of organic fertilizer in control P. brachyurus in soybean field.<br>Os fitonematoides podem limitar significativamente a produtividade da soja. Devido ao manejo inadequado do solo e ao fato de não existir nematicidas químicos eficientes no mercado. O Pratylenchus brachyurus conhecido como nematoide das lesões radiculares, tem se disseminado rapidamente e aumentado sua importância nas lavouras do Brasil. O objetivo deste trabalho foi avaliar a ação de fertilizantes orgânicos na forma sólida e líquida no controle de P. brachyurus, em raízes de soja, cultivar Fepagro 36 RR. O delineamento utilizado foi de blocos ao acaso com seis tratamentos e três repetições. Foram três tratamentos avaliados com os fertilizantes na forma sólida e líquida: solo+vermicomposto de esterco bovino (VERM), solo+composto (esterco bovino e palha, COMP), solo+fertilizante orgânico Ecocitrus (Eco) e dois controles sem adubação, com a presença e ausência do P. brachyurus. As coletas foram realizadas aos 25, 50 e 75 dias após a inoculação do fitonematoide. Avaliou-se a altura, massa seca da parte aérea e raiz e o número de juvenis de P. brachyurus no interior das raízes. Os resultados demonstram que partir dos 50 dias os tratamentos aplicados na forma sólida foram eficientes no controle de P. brachyurus e ao desenvolvimento das plantas de soja. Aos 75 dias, observa-se que as plantas cultivadas com VERM foram menos prejudicadas, sendo o número de nematoides penetrados 13% e 26% menor que o observado nos adubos ECO e CEBP. Novos ensaios deverão ser realizados utilizando compostos de esterco bovino, resíduos comerciais e o vermicomposto para confirmar o potencial dos fertilizantes orgânicos no controle de P. brachyurus em soja a campo.

Plant-parasitic nematodes devastate a wide range of plant species across the world including many economically important crops such as coffee. Determining the mechanisms that underpin nematode entry and parasitism of the root is of great interest for the design of control strategies. The first aspect of crop protection is to accurately and reliably diagnose the nematode species of concern. A molecular pipeline is described in this thesis that utilises PCR to rapidly detect and quantify the major Meloidogyne and Pratylenchus nematode species that are capable of parasitising coffee. This methodology assessed the prevalence of these species across the major coffee growing regions in Brazil, Vietnam and Indonesia. Distinct profiles of Meloidogyne species were detected across the regions whereas congruent Pratylenchus species were associated with coffee in all three countries. Furthermore, the pathogens were more numerous around intercropped plants such as banana and black pepper compared to coffee, suggesting that these crops may aggravate the issue. There was a high abundance of Meloidogyne in soil in which Pratylenchus was low, suggesting that the success of one genus may deter another. There is a clear widespread, yet differential nematode problem within coffee plantations that is likely to be effecting production and the issue is compounded by local practices and choices of intercrops. The global scale of the problem and the cost to coffee production could be elucidated with wider application of the approach. Plant-nematode interactions must be defined in order to act upon diagnoses from the field. As obligate plant parasites, Meloidogyne and Pratylenchus nematodes must be able to locate and feed from their host in order to survive. The plant cell wall provides protection and support to plant cells and is a major barrier that the nematode must overcome to initiate feeding. This projects shows that Pratylenchus coffeae, a key species detected in the field, regulates the expression of selected cell-wall degrading enzyme genes relative to the abundance of substrate in root exudates, thereby tailoring gene expression to maximise the chances of successful parasitism. Treatment with the substrates directly or with root exudates deficient in the substrates conferred a specific gene expression response with no effect on expression of another cell wall degrading enzyme gene. This indicates that host-specific gene expression in this plant-parasitic nematode is influenced by cell wall components that derive from plant secretion or degradation of root tissue. The illustrated transcriptional plasticity may have evolved as an adaptation for recognition and increased root invasion of a wider range of host species.

The experiment was conducted in the municipality of Caiabu in an area with nematode infestation. Two genotypes of sugarcane which the action of nematodes were used in the experiment SP81-3250 and RB867515. The employee was a randomized complete block design with five replications. Treatments were two genotypes of sugarcane combined with four ways to control nematodes (carbofuran, B. subtilis applied at planting, B. subtilis applied in post -emergence control and without the application of chemical or biological nematicide) total of 40 plots. During the conduct of the culture three assessments for quantification of nematodes on the roots was carried out. Was also carried out an assessment of chlorophyll content, tillering and biomass energy potential by the calorimeter method at 180 days after planting. At harvest yield was determined and made technological analysis of sugarcane. Chemical and biological control with carbofuran with Bacillus subtilis were effective for controlling nematodes reviews. There were no differences in the efficiency of the control in the evaluation of two forms of application of bacteria in soil. There increments Brix and Pol in variety SP81-3250 when we applied the treatment with B. subtlis.<br>O objetivo do presente trabalho foi o de avaliar duas formas de aplicação de Bacillus subtilis no controle biológico de nematoides em duas variedades de cana-de-açúcar comparando-se com o controle químico convencional. O experimento foi desenvolvido no município de Caiabu, em uma área com histórico de infestação de nematoides. Foram utilizados no experimento dois genótipos de cana-de-açúcar susceptíveis a ação de nematoides, SP81-3250 e RB867515. O delineamento empregado foi de blocos casualizados, com cinco repetições. Os tratamentos foram dois genótipos de cana-de-açúcar combinados com quatro formas de controle de nematoides (carbofurano, B. subtilis aplicado no sulco de plantio, B. subtilis aplicado em pós-emergência e controle sem aplicação de nematicida químico ou biológico), totalizando 40 parcelas. Durante a condução da cultura foram realizadas três avaliações para quantificação de nematoides nas raízes. Foi também realizado uma avaliação de teor de clorofila, perfilhamento e potencial energético da biomassa pelo método do calorímetro aos 180 dias após o plantio. No momento da colheita foi determinada a produtividade e realizadas análises tecnológicas da cana-de-açúcar. O controle químico com carbofurano e biológico com Bacillus subtilis foram eficientes para controlar os nematoides avaliados. Não houve diferenças quanto à eficiência de controle na avaliação das duas formas de aplicação da bactéria no solo. Ocorreram incrementos do Brix e do Pol na variedade SP81-3250 quando aplicou-se o tratamento com B. subtlis.

Until now, there is no clean and low cost alternative to successfully reduce its population in contaminated fields. However, recent studies has shown the possibility of inducing the defense mechanisms in plants as a strategy for the management. The objective of this study was to evaluate the biochemical response and the development of soybean cultivars when subjected to parasitism by the nematode Pratylenchus brachyurus and treat with abiotic resistance inducer acibenzolar-S- methyl (ASM). Soybean was sown in polyethylene bags, and seven days after, the treatments were inoculated with the nematode. The experiment was conducted in a completely randomized design in a 5x4 factorial scheme, with eight replications. The treatments consisted of combinations of five soybean cultivars with: Control (plants not inoculated with the nematode and not sprayed with ASM), Pb (plants inoculated with the nematode), ASM (plants sprayed with the plant activator) and Pb + ASM (plants inoculated with the nematode and sprayed with ASM). The treatments with ASM were sprayed at 14, 21 and 28 days after the nematode inoculation (DAI) and, 24 hours later, the plant height and the enzymatic activity were evaluated. At 60 DAI it was evaluated the plant height, the shoots fresh weight (SFW), the roots fresh weigh (RFW), the reproduction factor (Rf) and the nematode density in the roots. There was higher total protein concentration after the third ASM application, with higher activity of chitinase (CHI) and β-1,3- glucanase (GLU) in the first two applications, higher peroxidase (POX) in the second and lipoxygenase (LOX) and phenylalanine ammonia-lyase (PAL) after the third. The results showed a possible synergistic effect between the activities of CHI and GLU and antagonistic effect between the POX and the FAL during the evaluation timings. ASM increased the activity of POX, LOX and FAL in the absence or presence of the nematode. Activities of CHI and GLU were higher in plants parasitized by the P. brachyurus. During the three evaluation periods the greatest heights were recorded at 28 DAI with lower height in plants with nematode. There was no statistical difference in height between treatments. However, there were differences between cultivars. The cultivars BRSGO 8560 RR, Emgopa 313 RR e M-Soy 8360 RR showed greater heights. The plants had higher SFW and RFW in treatments containing the nematode. In plants treated with ASM the nematode density was higher, while the Rf was lower. Thus, the results showed that the plant activator ASM and the nematode P. brachyurus affect the plant development and that the enzymatic activity is influenced by the number of ASM sprays and by the presence of nematode. The resistance inducer, ASM, shows potential for reducing the nematode`s reproduction.<br>O nematoide das lesões radiculares (Pratylenchus brachyurus) é um dos principais fitoparasitas da sojicultura brasileira. Até o momento, não há uma alternativa limpa e de baixo custo de controle que reduza satisfatoriamente a população desse nematoide nas áreas infestadas ou que proporcione pleno desenvolvimento das plantas parasitadas. Porém, estudos recentes mostram que há a possibilidade de induzir as defesas das plantas como estratégia a ser inserida nos programas de manejo das culturas a fitopatógenos, como os nematoides. Diante do exposto, o objetivo deste trabalho foi avaliar a resposta bioquímica e o desenvolvimento de cultivares de soja quando submetidas ao parasitismo pelo nematoide Pratylenchus brachyurus e tratadas com o indutor de resistência abiótico acibenzolar-S- metil. A soja foi semeada em saquinhos de polietileno e, após sete dias, os tratamentos foram inoculados com o nematoide. O experimento foi conduzido em delineamento inteiramente casualizado em esquema fatorial 5x4, com oito repetições. Os tratamentos foram compostos pela combinação das cinco cultivares de soja com: Testemunha (plantas não inoculadas com o P. brachyurus e não pulverizadas com o ASM), Pb (plantas inoculadas com P. brachyurus), ASM (plantas pulverizadas com o indutor de resistência) e Pb+ASM (plantas inoculadas com o nematoide e pulverizadas com o ASM). Os tratamentos que receberam ASM tiveram três aplicações (14, 21 e 28 dias após a inoculação do nematoide), sendo a atividade enzimática avaliada 24 horas após a aplicação do produto. Aos 60 DAI foi avaliado a altura, a massa fresca da parte aérea (MFPA) e das raízes (MFR), fator de reprodução (FR) e densidade do P. brachyurus nas raízes. Observou-se maior concentração de proteínas totais após a terceira aplicação do ASM, com maior atividade da quitinase (QUI) e β-1,3-glucanase (GLU) nas duas primeiras aplicações, da peroxidase (POX) na segunda aplicação e da lipoxigenase (LOX) e fenilalanina amônia-liase (FAL) após a terceira aplicação. Os resultados apresentam um possível efeito sinérgico entre as atividades da QUI e GLU e antagonista entre a POX e a FAL durante essas épocas. O ASM contribuiu para o incremento da atividade da POX, LOX e da FAL na ausência ou presença do nematoide e as atividades da QUI e da GLU foram maiores nas plantas parasitadas pelo P. brachyurus. Não houve diferença estatística para altura entre os tratamentos. No entanto, houve diferença entre as cultivares. A BRSGO 8560 RR, Emgopa 313 RR e M-Soy 8360 RR apresentaram as maiores alturas. As plantas apresentaram maior MFPA e MFR nos tratamentos contendo o nematoide. Nas plantas tratadas com o ASM a densidade de nematoides foi maior, enquanto que, o FR foi menor. Sendo assim, os resultados mostram que o indutor e o nematoide interferem no desenvolvimento da planta e a atividade enzimática é influenciada pelo número de aplicações do ASM e pela presença do P. brachyurus. O indutor de resistência mostra potencial para a redução da reprodução deste fitopatógeno.

Variações morfológicas e biológicas são observadas entre os isolados de Pratylenchus coffeae, o nematóide das lesões radiculares do cafeeiro, indicando a existência de raças dentro da espécie. Este estudo foi realizado com o objetivo de caracterizar dois isolados de P. coffeae coletados do Brasil. Um dos isolados foi coletado de raízes de cafeeiro em Marília, SP, e o outro de raízes de Aglaonema sp. no Rio de Janeiro, RJ. Ambos isolados foram multiplicados em calos de alfafa no laboratório. Sete experimentos foram conduzidos com o objetivo de caracterizar os isolados através de seus hospedeiros. Os resultados mostraram que esses isolados são muito diferentes do isolado típico de Pratylenchus coffeae, uma vez que a taxa reprodutiva foi baixa banana e citros. Além disso, a população do isolado do Rio de Janeiro diminuiu no cafeeiro, o qual é um hospedeiro típico de Pratylenchus coffeae, porém aumentou em gergelim, um hospedeiro desfavorável desse nematóide. Baseado nos resultados da caracterização biológica, a qual mostrou que o cafeeiro é um hospedeiro desfavorável para ambos isolados, um experimento foi conduzido como objetivo de definir a importância desses nematóides como patógeno do cafeeiro. Plantas de cafeeiro cv. Mundo Novo receberam 8.000 nematóides de cada isolado, o crescimento das plantas, peso fresco das raízes, e peso fresco da parte aérea, foi avaliado 160 e 261 após a inoculação. A reprodução dos nematóides também foi avaliada. Ambos isolados reduziram o crescimento das plantas de cafeeiro, porém houve uma redução na população do nematóide. O efeito do isolado de Marília foi mais danoso do que o do isolado do Rio de Janeiro, resultando em necroses nas raízes e morte de algumas plantas. As características morfológicas também podem ser utilizadas para diferenciar os isolados. The root lesion nematode (RLN), Pratylenchus thornei, is a biotrophic migratory pest of plant roots and its infestation causes losses in many economically important crops. RNA interference (RNAi) is a naturally occurring eukaryotic phenomenon and can be used to silence parasitism effector genes of P. thornei using host-mediated RNAi. This may be developed as an environmentally friendly and a cost-effective control strategy. The overall aims of this research were to investigate the effects of in vitro and in planta RNAi silencing of putative P. thornei parasitism effector genes, and their nematicidal effects in two host plants. Five putative target parasitism genes vital for nematode entry into roots (Pt-Eng-1, Pt-PL), feeding (Pt-CLP) and suppressing host defence responses (Pt-UEP, Pt-GST) were identified, validated in silico using comparative bioinformatics, cloned into suitable in vitro transcription and binary vectors, and advanced to RNAi studies. Partial sequences for four of these target effector genes (Pt-Eng-1, Pt-PL, Pt-CLP, Pt-GST) were identified using Rapid Amplification of cDNA (RACE) PCRs and annotated in silico. Protein families, conserved domains, taxonomic and phylogenetic relationships for all four effectors were studied. This sequence information will help inform future investigations involving gene expression and proteomics of the selected putative effectors. In vitro RNAi was used for functional characterisation of the five effector sequences. Effects on nematode phenotype, behaviour, gene expression, and longer-term effects on reproduction were assessed after soaking nematodes in dsRNA through infection of healthy wild type soybean and alfalfa roots. Soaking of mixed stage P. thornei in 1mg/mL dsRNA of target genes for 16 h did not cause phenotypic changes except for Pt-PL, which exhibited straight or slightly curved phenotypes after soaking compared to the normal sigmoid body movement, also evident for green fluorescent protein (gfp) and no dsRNA treated controls. Semi-quantitative PCRs and densitometry analysis revealed a significant reduction of transcript accumulation for all five putative parasitism effector genes. Longer-term effects assessed at 21 dpi reduced nematode reproduction by 40 to 70% for all target genes compared to respective control treatments suggesting that the effectors studied were required for nematode infectivity, survival or reproduction. In planta RNAi involved Agrobacterium-mediated plant transformations to develop axenic transgenic hairy root events of soybean (Glycine max var. Williams 82) and alfalfa (Medicago sativa), and non-axenic hairy roots (composite plants) of soybean. Both hosts were amenable to Agrobacterium-mediated transformation, but hairy root induction was faster in alfalfa than soybean. However, more events were generated for soybean than alfalfa. Transgenic hairy roots confirmed by molecular analyses were challenged with P. thornei and their presence confirmed after 14 dpi. After 21 dpi, nematode numbers and transcript abundance was assessed using semi-quantitative PCRs and densitometry analysis. Host-mediated silencing of the five putative parasitism effector genes using transgenic soybean and alfalfa hairy roots showed a significant reduction in target transcript accumulation and approximately 38 to 75% reduction in P. thornei numbers compared to untransformed wild-type controls. For some events, there was a positive correlation between reduced transcripts and nematode numbers. Based on percent reduction in transcript accumulation of the target genes relative to 18S rRNA as assessed by densitometry, the extent of gene knockdown measured (from most to least) was: Pt-Eng-1, Pt-PL, Pt-CLP, Pt-UEP, and Pt-GST. Similarly, Pt-Eng-1, Pt-PL and Pt-CLP were ranked in the same order, from the lowest to highest reproduction on soybean and alfalfa, indicating a positive correlation between the level of knockdown and reduced reproduction. In soybean, these genes were followed by Pt-GST and Pt-UEP for the percentage of reproduction recorded, whereas, in alfalfa, reduction in reproduction for these two target genes did not differ significantly. Composite soybean with wild-type shoots and transgenic hairy roots expressing Pt-Eng-1 and Pt-PL genes were developed and provided an opportunity to test the effectiveness of silencing target genes in planta and on nematode numbers in conditions that mimicked natural host infections. For both Pt-Eng-1 and Pt-PL genes, there was a significant reduction in percentage of transcript accumulation relative to 18S rRNA, which correlated with a reduction in nematode numbers by 53.4% and 48.5% for Pt-Eng-1 and Pt-PL, respectively. The amenability of P. thornei to host-mediated RNAi using effector gene sequences, and the overall results of this study, point towards the potential use of this technology to control P. thornei and related RLN species effectively in different host crops.

Australia is the driest inhabited continent with about 70% of the land arid or semi-arid, and soils which are geologically old, weathered, and many are infertile. This is a challenging environment for agricultural production, which is further impacted by biotic constraints such as root lesion nematodes (RLNs), Pratylenchus spp. These soil-borne nematodes cause significant economic losses in yields of winter cereals, and in other crops, particularly under conditions of moisture and nutrient stress. RLNs are widely distributed in Australian broadacre cropping soils, and losses in cereal production are greater when more than one RLN species is present, a situation which often occurs in Western Australia (WA). Hence, to develop appropriate management regimes, accurate identification of RLN species is needed, combined with understanding the biology of host-nematode interactions. The initial aim of this research was to extend the molecular and biological characterisation of P. quasitereoides, a recently described species of root lesion nematode from WA. Morphological measurements of two important characters, tail shape and the per cent distance of the vulva from the anterior end of the nematode body, were made from nematodes collected from the four locations of WA. These measurements did not match with the published data on P. quasitereoides. As a result, nematode samples from Pingelly were sent to Dr. M. R. Siddiqi in the UK for detailed morphometric measurements: he concluded that the nematodes from Pingelly differed from other recognised species of Pratylenchus in Australia. The longer conus (more than half of the spear length), three annules in the lip region, large rounded basal knobs, phasmids in the anterior region of the tail, usually about one-fourth the tail length behind the anus, and the absence of males were good characters for species comparisons. The morphometric measurements suggested that the species under study was in fact P. curvicauda, a species which had been reported previously and described from WA (Siddiqi et al., 1991). Analyses of SEM images also supported the morphological description of the nematode samples from Pingelly as being P. curvicauda. Study of the genetic variation of this species with other known species of Pratylenchus was undertaken based on analysis of sequences of both the ribosomal DNA (rDNA) ITS and D2-D3 regions. Sequence analyses showed that there was considerable variation in P. curvicauda rDNA in samples from four wheatbelt regions, and this was also the case for P. neglectus, P. penetrans and P. thornei. The ITS sequences of P. curvicauda differed by 39% from those of P. neglectus and P. thornei, but were more similar to those of P. penetrans. Nucleotide variations between sequences of P. curvicauda and those of P. neglectus, P. penetrans and P. thornei were mainly in the ITS1 and ITS2 regions, which differed by 44% and 49% in nucleotide sequences of these regions with P. neglectus and P. thornei, whereas with only 10% in both regions compared to P. penetrans sequences. For the D2-D3 region, sequences from P. curvicauda, P. neglectus, P. penetrans and P. thornei differed by up to 30% at the nucleotide level. The D2-D3 sequences of P. curvicauda were very different from the published sequences for P. quasitereoides, with up to a 39% variation in nucleotide sequence. These results support the morphological data that Pratylenchus sp. collected from the four WA locations did not appear to be any of the species of Pratylenchus currently described as being in WA (i.e. P. neglectus, P. penetrans, P. thornei or P. quasitereoides). This result prompted a more detailed study of the biology of the nematode. The ability of different plant taxa/genotypes to support the reproduction of P. curvicauda was assessed using soil collected from Pingelly under glasshouse conditions. Crop and pasture species challenged included wheat, barley, pasture species, brassicas, and model plants, both by manual inoculation and grown in naturally infested field soil. Wheat, barley and sorghum were good hosts; canola, tomato and other brassicas were poor hosts; most pastures species and tobacco were non-hosts. A part of this research focused on how P. curvicauda was attracted to and penetrated roots of host plants identified as resistant or susceptible. Since attraction to and entry into roots of hosts classified as resistant or susceptible was essentially the same, host resistance is probably expressed later in the interaction. The life cycle of this species was also studied. It took 35-42 d to complete one generation in susceptible wheat roots under glasshouse conditions. Histological studies of roots infested by P. curvicauda were also undertaken. Cellular changes in infested roots included deformation of cell wall, loss of cytoplasmic contents and death of invaded cells, and there was some evidence of changes in cells further from the invading nematodes. There were no clear differences in the damage caused in susceptible and resistance hosts, at least up to 7 d after infection. This study is the first to investigate the biology and molecular characteristics of P. curvicauda.

Na literatura, encontram-se trabalhos acerca dos nematoides na cultura da goiabeira principalmente quando se trata de M. enterolobii, entretanto, até o momento não se registrou estudos solucionando os problemas com os nematoides e as variedades de goiabeira. Diante disto, os objetivos deste trabalho foram: a) Avaliar o desenvolvimento das variedades de goiabeiras Paluma, Pedro Sato, Século XXI e Tailandesa quando parasitadas por M. enterolobii, b) avaliar a reação de variedades de goiabeiras Paluma, Pedro Sato, Século XXI e Tailandesa à Pratylenchus brachyurus, c) Estudar a flutuação populacional de M.enterolobii em goiabeira Paluma, d) Avaliar o desenvolvimento vegetativo das goiabeiras Tailandesa e Paluma em formação sob quatro níveis crescentes de inóculo de M. enterolobii. Todos os experimentos foram realizados em casa de vegetação. This specie leads to significant yield losses on field and prevents orchards. Because of nematode damage the growers use to cultivate the area with other crops. In literature, there are some studies about rootknot nematode in guava tree mainly when they refer to M. enterolobii. However, there is not any result discussing how to solve the problems caused by root-knot nematodes considering the use of guava varieties. The objectives of this study were: a) evaluate the development of guava varieties Paluma, Pedro Sato, Século XX1 and Tailandesa when infected by M. enterolobii b) evaluate the reaction of guava varieties Paluma, Pedro Sato, Século XX1 and Tailandesa to lesion nematode Pratylenchus brachyurus c) study the dynamics population of M.enterolobii to Paluma guava variety d) evaluate the vegetative growth of Tailandesa and Paluma guava varieties under four increasing levels of M. enterolobii inoculum. To investigate nematicidal activity against the root-lesion nematode, Pratylenchus coffeae, the methanolic extracts were obtained from 5 medicinal plants in Vietnam. Methanol extracts of the 5 samples were screened for nematicidal activity against Pratylenchus coffeae in vitro. Of the plant extracts tested, a 5 mg/ml concentration of Terminalia nigrovenulosa extract showed the highest level (95.0%) of nematicidal activity against Pratylenchus coffeae at 9 h after treatment. The remainder of the plant extracts was followed by Cinnamomum camphora (66.7%), Jasminum suptriplinerve (30.8%), Premna integrifolia (6.7%), and Treptocaulon juventas (5.8%). A 2.0 mg/ml concentration of T. nigrovenulosa resulted in 13.3, 42.5, 62.5 and 86.7% at 1, 3, 5 and 7 h after treatment, respectively against Pratylenchus coffeae. Concentration of 0.1, 0.5, 1.0 and 2.0 mg/ml of T. nigrovenulosa resulted in 26.7, 71.7, 78.3 and 86.7% effectiveness respectively, at 7 h after treatment. Also, at 3 days after exposure to T. nigrovenulosa, hatch was inhibited by 60.0, 45.8, 17.5 and 5.8% at 0.1, 0.5, 1.0 and 2.0 mg/ml, respectively. Hatch was inhibited by 65.0, 50.8, 21.7 and 6.7% at 0.1, 0.5, 1.0 and 2.0 mg/ml respectvely, at 6 days after exposure to T. nigrovenulosa. The mortality rate and hatch inhibition increased with increasing incubation time for crude extract of T. nigrovenulosa<br>Trong nghiên cứu này, 5 cây thuốc (Cinnamomum camphora, Jasminum suptriplinerve, Premna integrifolia, Terminalia nigrovenulosa, Treptocaulon juventas) đã được sử dụng làm nguyên liệu chiết xuất bằng methanol để thử hoạt tính kháng tuyến trùng Pratylenchus coffeae. Kết quả cho thấy chiết xuất từ cây Terminalia nigrovenulosa biểu hiện hoạt tính kháng tuyến trùng cao nhất (95,0%) sau 9 giờ xử lý với nồng độ 5 mg/ml. Ở cùng thời điểm sau xử lý 7 giờ, các nồng độ 0,1; 0,5; 1,0 và 2,0 mg/ml chiết xuất của T. nigrovenulossa có hoạt tính gây chết tuyến trùng Pratylenchus coffeae lần lượt là 26,7; 71,7; 78,3 và 86,7%. Bên cạnh đó, chất chiết xuất của T. nigrovenulossa cũng cho thấy tỷ lệ ức chế trứng nở cao. A crescente preocupação com nematoides está associada a sua facilidade em encontrar alimento durante a safra e entressafra. A sucessão de culturas popularizada pelo Sistema de Plantio Direto (SPD) permite a multiplicação constante destes patógenos em campo. Pratylenchus brachyurus, o nematoide das lesões radiculares, foi extremamente favorecido por este sistema devido a seu alto grau de polífagia, parasitando inclusive as principais culturas agrícolas nacionais. A soja é o a cultura que mais sofre perdas por este patógeno e o sistema onde é empregada, em rotação com gramíneas, é uma condição agravante para a cultura. Devido a isto a busca por alternativas de sucessão em que o nematoide não seja capaz de aumentar a população é uma ferramenta importante para o controle do nematoide. O objetivo deste trabalho foi buscar alternativas de coberturas vegetais para o controle de P. brachyurus e testar o efeito em sucessão com soja. Foram realizados dois experimentos, sendo o primeiro para avaliar a reação de hospedabilidade de plantas de cobertura a P. brachyurus, e o segundo com sucessão de sorgo com soja. No primeiro experimento foram avaliados 18 tratamentos na primeira etapa e 10 na segunda, totalizando 19 coberturas vegetais diferentes. Os ensaios foram conduzidos em casa de vegetação em vasos de plástico com 1L de solo autoclavado. Três plantas de cada tratamento foram mantidas por parcela. Foram inoculados 1000 espécimes do nematoide por parcela e a avaliação foi realizada aos 90 dias após a inoculação (DAI). Neste período os nematoides, foram extraídos das raízes por método de liquidificação, peneiramento e flutuação em centrifuga, e contados em Câmara de Peters sobre microscópio ótico. Com a população final encontrada foi avaliado o Fator de Reprodução (FR) do nematoide na planta. Nos dois ensaios foram encontrados resistência para seis tratamentos. Leucena, estilosantes, tefrosia, C. grantiana, C. spectabilis e C. breviflora apresentaram resistência nos dois experimentos. Guandu-mandarim apresentou resistência no primeiro ensaio e suscetibilidade no segundo enquanto que, C. ochroleuca ocorreu o inverso. Os resultados demostraram o potencial de redução da população do fitonematoide com o uso de espécies vegetais de cobertura. No segundo experimento foi conduzido ensaio de sucessão com cinco cultivares de sorgo e um de milheto em sistema com gramínea-soja. Os experimentos foram conduzidos em casa de vegetação com oito parcelas por tratamento. Cada parcela foi inoculada com 1000 espécimes de P. brachyurus em vasos de 1L com solo autoclavado. Foram mantidas três plantas por vaso. Quatro parcelas foram colhidas aos 90 DAI e realizada a extração para avaliação de FR. Das quatro parcelas restantes foi removida a parte aérea das gramíneas e semeado soja. As avaliações em soja foram realizadas na senescência da cultura. Não foram encontrados tratamentos resistentes. O cultivar de milheto BRS 1501 apresentou os menores valores de FR, multiplicando menos o nematoide, enquanto o cultivar de sorgo BRS 716 apresentou os valores mais elevados. As avaliações em soja demonstraram a menor produtividade e massa fresca de raiz de soja conforme o aumento da população do nematoide na parcela devido à sucessão. Nenhum dos tratamentos foi eficiente para diminuir a população do nematoide e o aumento da população inicial na soja agravou os danos causados pelo nematoide. Por tanto nenhum dos tratamentos com gramíneas é recomendado para rotação com soja ou cultivo em solos infestados. Neste trabalho foi possível visualizar os danos causados pelo nematoide na cultura e também foi possível encontrar potenciais de controle cultural com plantas resistentes e a supressão da população no solo.<br>The growing concern with nematodes is associated with their ease in finding food during the harvest and in the off season. The crop rotation, popularized by the No-till farming allows the constant multiplication of these pathogens in the field. Pratylenchus brachyurus the root-lesion nematode was greatly favored by this system due to its polyphagous feeding behavior, parasitizing even the main national agricultural crops. Soybean is the host of this pathogen, and the system where it is employed, in rotation with grasses, is an aggravating condition for the crop. Due to this the search for rotation alternatives where the nematode is not able to increase the population is an important tool for the control in the off season and to avoid losses in the main crop. The objective of this work was to search for plant cover alternatives for the control of P. brachyurus and to test the effect in succession with soybean. Two experiments were carried out, the first one to evaluate the host planting of P. brachyurus, and a second with succession of grasses with soybean in soil infested with nematodes. In the first experiment, eighteen treatments were evaluated in the first stage and ten in the second, totaling nineteen different cover crops. The assays were conducted in a greenhouse in plastic vats with 1L of steam sterilized soil. Three plants of each treatment were maintained per plot. 1000 specimens of the nematode were inoculated per plot and the evaluation was performed at 90 days after inoculation (DAI). During this period the nematodes were extracted from the roots by means of liquidification, sieving and centrifugal flotation, and counted in Peters\' Chamber on an optical microscope. With the final population found, the reproductive factor (RF) of the nematode in the plant was evaluated. In both tests resistance was found for six treatments. Leucaena, stylers, tephrosia, C. grantiana, C. spectabilis and C. breviflora showed resistance in both experiments. Pigeonpea cv \'mandarim\' presented resistance in the first trial and susceptibility in the second while C. ochroleuca occurred the inverse. The results demonstrated the potential of reducing the phytonematoid population with the use of plant cover species. In the second experiment, a succession test was carried out with five cultivars of sorghum and one of millet in a grass-soybean system. The experiments were conducted in greenhouse with eight plots per treatment. Each plot was inoculated with 1000 specimens of P. brachyurus in 1L vessels with autoclaved soil. Three plants were maintained per pot. Four plots were harvested at 90 DAI and extraction was performed for FR evaluation. Of the four remaining plots was removed the aerial part of the grasses and sown soybean. The soybean evaluations were carried out in the senescence of the crop. No resistant treatments were found. The millet cultivar BRS 1501 presented the lowest values of RF, multiplying the nematode less, while the sorghum cultivar BRS 716 had the highest values. Soybean yields showed lower productivity and fresh soybean root mass as the nematode population increased in the plot due to succession. None of the treatments with grasses were efficient to decrease the nematode population and the initial population increases in soybean aggravated nematode damage. None of the treatments are recommended for soybean rotation or cultivation on infested soil. In this work it was possible to visualize the damage caused by the nematode in the crop and it was also possible to find potentials of cultural control with resistant plants and the suppression of the population in the soil.

Para isso, foram montados quatro diferentes experimentos. No primeiro experimento plantas de cafeeiro Catuaí Vermelho e de citros Limão Cravo foram inoculadas 100 nematóides (P. coffeae ou P. jaehni) por planta, que foram mantidas em casa de vegetação. Após 148 dias as plantas foram avaliadas utilizando como parâmetros: altura e peso seco da parte aérea, peso fresco do sistema radicular e população final do nematóide inoculado no solo e na raiz, seguindo a metodologia proposta por Jenkins (1964) e Coolen e D Herde (1972), respectivamente. No segundo experimento plantas de cafeeiro Catuaí Vermelho e de citros Limão Cravo foram inoculadas 220 nematóides (P. jaehni) por planta, as quais foram mantidas em casa de vegetação. Após 136 dias as plantas foram avaliadas utilizando os mesmos parâmetros acima relacionados. No terceiro experimento sete genótipos de cafeeiro (Icatu Vermelho, Apoatã, Catuaí Vermelho, Mundo Novo, Laurentii, EP 355 e IAC 457) foram inoculadas com 500 nematóides (P. coffeae ou P. jaehni) por planta. Após 120 dias as plantas foram analisadas utilizando como parâmetros massa fresca do sistema radicular e população final do nematóide inoculado no solo e na raiz, seguindo a metodologia proposta por Jenkins (1964) e Coolen e D Herde (1972), respectivamente. No quarto experimento, também conduzido em casa de vegetação, três genótipos de cafeeiro (Catuaí Vermelho, Icatu Vermelho e Mundo Novo) foram 2 inoculadas com 1.000 nematóides (P. coffeae ou P. jaehni) por planta. Bibliography: leaves 307-329. This study concludes that in soils in South Australia where root-rotting fungi and P. neglectus exist together, root disease of wheat is caused by their combined effect. Evidence suggests that P. neglectus not only contributes to this interaction through mechanical wounding of roots, but also causes biochemical and physiological changes in plants, making them more prone to fungal infection.

O gênero Eucalyptus possui diversas espécies que apresentam grande valor comercial, sendo utilizado principalmente para produção de energia (carvão e lenha) e celulose-papel. Na literatura, entretanto, existem poucas informações acerca de nematoses nessa cultura. Além dessa carência e de sua importância econômica, o eucalipto é comumente cultivado em locais de alta incidência de Pratylenchus brachyurus e Meloidogyne incognita, além de ser consorciado com diversas outras culturas intolerantes, como por exemplo a soja. Dessa forma há uma necessidade de conhecer a reação das espécies de eucalipto a estes nematoides. Dito isso, o presente trabalho teve por objetivo testar a reação de algumas espécies de eucalipto a P. brachyurus e M.incognita. Foram realizados 6 experimentos, 5 envolvendo o nematoide das lesões e 1 com o nematoide de galhas. Os tratamentos foram inoculados com uma população inicial de nematoides e, após 90 dias, estimava-se o fator de reprodução (FR) e nematoides por grama de raiz (Nem/g). Adotou-se o critério baseado em Oostenbrink (1966), onde a espécie foi considerada suscetível quando apresenta FR >= 1 e resistente quando FR < 1. Dos eucaliptos testados, 6 apresentaram reação suscetível e 4 reação resistente para P. brachyurus. As espécies com maiores FR estão entre as mais utilizadas no Brasil, exceto E. saligna. Ressalta-se que estas não necessariamente são resistentes, necessitando apenas de um período maior para o estabelecimento do patógeno. Observou-se indícios de que E. saligna e E. camaldulensis sejam intolerantes a P.brachyurus. Para M. incognita raça 3 todas as espécies testadas foram resistentes. Aconselha-se a realização de experimentos com períodos superiores a 100 dias para uma correta classificação da reação de hospedeiro. O eucalipto é uma opção para aproveitamento de áreas infestadas com M. incognita raça 3, porém seu uso mostra-se um risco em áreas infestadas com P. brachyurus.<br>The Eucalyptus has several species with high commercial value and is used mainly for energy purpose (Charcoal and firewood) and paper. In the literature, however, there is little information about plant parasitic nematodes in this woody specie. In addition to this lack and its economic importance, eucalypts is also commonly cultivated in areas with high incidence of Pratylenchus brachyurus and Meloidogyne incognita, and consorted with several intolerant crops, such as soybean. Thus, there is a need to know the host status of the eucalypts species to these nematodes. With that, this work aimed to test the reaction of some Eucalyptus species to P.brachyurus and M. incognita. For this purpose six experiments were performed, five to test the lesion nematode and one to test the root-knot nematode. The treatments were inoculated and, after 90 days, it was estimated the reproduction factor (FR) and nematodes per gram of root (Nem/g). The present work adopted the criteria based on Oostenbrink (1966), where the species was considered susceptible when presenting FR >= 1 and resistant when FR < 1. Of species tested, six were susceptible hosts and four resistant host to P. brachyurus. The species with highest FR are among the most used eucalypts in Brazil, except E. saligna. It is emphasized that those species with FR < 1 are not necessarily resistant, requiring only a longer period for the pathogen establishment. There was some evidence that E. saligna and E. camaldulensis are intolerant hosts to P. brachyurus. To M. incognita Race 3 all tested species were resistant. It is advisable to perform experiments for periods longer than 100 days to a correct classification of host status. Eucalyptus is an option for utilization of areas infested with M. incognita race 3, but its use appears to be a risk in Pratylenchus brachyurus infested areas.

Em ensaios de casa-de-vegetação, primeiramente foi avaliada a reprodução de Pratylenchus brachyurus em oito cultivares de algodoeiro, que foram: IAPAR 71-Paraná 3; IAC 20; CS 50; CNPA 717; IAC 87; CNPA-Precoce 2; IAC 21 e IAC 22.Posteriormente foram selecionadas duas cultivares para estudar a patogenicidade do nematóide, inoculando-se diferentes níveis de população inicial; 0 (testemunha); 333; 1000; 3000; 9000 e 27000 nematóides/planta. As avaliações foram com base nos números de nematóides extraídos do sistema radicular e do solo e nos fatores de reprodução. Nos estudos de patogenicidade foram coletados também os seguintes dados de crescimento das plantas; altura de planta, massa fresca de sistema radicular e massa seca de parte aérea. Entre as oito cultivares em que se avaliou a reprodução, foi possível identificar IAPAR 71-Paraná 3 e IAC 20 como as melhores hospedeiras do nematóide, nas quais a população final do nematóide foi significativamente maior que em IAC 22. Para os estudos de patogenicidade foram selecionadas as cultivares IAC 20 e IAC 22, que, entre as mais utilizadas comercialmente, permitiram a maior e a menor reprodução do nematóide, respectivamente. Ambas as cultivares tiveram o crescimento prejudicado pelo nematóide e as reduções de crescimento das plantas inoculadas em relação à testemunha foram mais acentuadas nos níveis mais altos de inóculo. Pratylenchus brachyurus é um dos nematóides mais disseminados na cultura do algodão nas áreas produtoras do Brasil. Sua patogenicidade ao algodoeiro, entretanto, é pouco estudada. Os objetivos deste trabalho foram: i) correlacionar níveis populacionais iniciais crescentes de P. brachyurus (0, 12.000, 30.000 e 75.000 exemplares/ planta) com os danos causados ao algodoeiro \'Delta Opal\'; ii) avaliar a patogenicidade de populações de P. brachyurus em algodoeiros \'Delta Opal\' e \'Fibermax 966\'; iii) testar cultivares de algodão em relação à reprodução de três populações de P. brachyurus ; iv) caracterizar a relação parasito-hospedeiro (em termos de suscetibilidade/resistência) de alguns adubos verdes, coberturas vegetais e pastagens a Pratylenchus brachyurus; v) caracterizar molecularmente populações de P. brachyurus, através de PCR-RFLP e seqüenciamento da região ITS-1 do rDNA. Os resultados sugerem que P. brachyurus é patógeno pouco agressivo da cultura do algodão, já que não se verificaram danos significativos às plantas em densidades populacionais do nematóide inferiores a 12.000 exemplares/ planta. Em relação às cultivares, todas foram suscetíveis a P. brachyurus . Entre as espécies vegetais testadas, as que se mostraram resistentes a P. brachyurus foram Crotalaria spectabilis, C. breviflora, amaranto \'BRS Alegria\', nabo forrageiro \'Comum\' e as cultivares de aveia preta Campeira Mor, IPFA 99006, Comum, CPAO 0010 e Garoa. As análises de PCRRFLP revelaram variabilidade genética entre as diferentes populações de P. brachyurus estudadas, em função dos diferentes padrões de bandas encontrados para as populações estudadas. O seqüenciamento da região ITS-1 do rDNA confirmou a variabilidade observada pela digestão enzimática, além de evidenciar heterogeneidade das regiões 18S e ITS-1 do rDNA de P. brachyurus<br>Although Pratylenchus brachyurus is widespread in Brazilian cotton fields, information about its importance as a cotton pathogen is scarce. The objectives of this work were: i) correlate crescent initial population densities (0; 12,000; 30,000; and 75,000 nematodes/ plant) with damage on cotton \'Delta Opal\'; ii) measure the pathogenic effect of P. brachyurus on cotton \'Delta Opal\' and \'Fibermax 966\'; iii) characterize the reaction of cotton cultivars to three populations of P. brachyurus ; iv) characterize the host reaction (in terms of susceptibility/ resistance) of some green manures, cover crops and pastures to two populations of P. brachyurus; v) characterize different populations of P. brachyurus by PCR-RFLP and sequencing of ITS-1 rDNA region. Results suggest that P. brachyurus is an eventual pathogen of cotton, since high population levels were necessary to reduce plant growth (< 12,000 nematodes/ plant). All cotton cultivars tested were rated as susceptible to P. brachyurus In relation to crop species tested, Crotalaria spectabilis, C. breviflora, amaranth \'BRS Alegria\', oil radish \'Comum\', and the black oat cultivars Campeira Mor, IPFA 99006, Comum, CPAO 0010, and Garoa were resistant to P. brachyurus PCR-RFLP showed intraspecific variability for different population of P. brachyurus studied. Sequencing of the ITS-1 rDNA region confirmed the results of the enzymatic digestion and demonstrated heterogeneity of 18S and ITS-1 rDNA regions of P. brachyurus.

Orientador: Silvia Renata Siciliano Wilcken<br>Banca: Mário Massayuki Inomoto<br>Banca: Roberto Kazuhiro Kubo<br>A presente pesquisa visou obter a taxa de reprodução de Pratylenchus coffeae (Zimmermann, 1898) e Pratylenchus jaehni Inserra,Duncan, Santos, Kaplan, Vovlas 2001 em cafeeiro e em citros a fim de encontrar nesses hospedeiros semelhanças ou diferenças entre as duas espécies de nematóides estudadas. Para isso, foram montados quatro diferentes experimentos. No primeiro experimento plantas de cafeeiro ‘Catuaí Vermelho’ e de citros ‘Limão Cravo’ foram inoculadas 100 nematóides (P. coffeae ou P. jaehni) por planta, que foram mantidas em casa de vegetação. Após 148 dias as plantas foram avaliadas utilizando como parâmetros: altura e peso seco da parte aérea, peso fresco do sistema radicular e população final do nematóide inoculado no solo e na raiz, seguindo a metodologia proposta por Jenkins (1964) e Coolen e D’Herde (1972), respectivamente. No segundo experimento plantas de cafeeiro ‘Catuaí Vermelho’ e de citros ‘Limão Cravo’ foram inoculadas 220 nematóides (P. jaehni) por planta, as quais foram mantidas em casa de vegetação. Após 136 dias as plantas foram avaliadas utilizando os mesmos parâmetros acima relacionados. No terceiro experimento sete genótipos de cafeeiro (Icatu Vermelho, Apoatã, Catuaí Vermelho, Mundo Novo, Laurentii, EP 355 e IAC 457) foram inoculadas com 500 nematóides (P. coffeae ou P. jaehni) por planta. Após 120 dias as plantas foram analisadas utilizando como parâmetros massa fresca do sistema radicular e população final do nematóide inoculado no solo e na raiz, seguindo a metodologia proposta por Jenkins (1964) e Coolen e D’Herde (1972), respectivamente. No quarto experimento, também conduzido em casa de vegetação, três genótipos de cafeeiro (Catuaí Vermelho, Icatu Vermelho e Mundo Novo) foram 2 inoculadas com 1.000 nematóides (P. coffeae ou P. jaehni) por planta. A ocorrência de estresses abióticos e bióticos em plantas imaturas de soja, o que inclui a presença de nematoides, podem resultar em morte prematura da planta ou maturação forçada e produzir sementes esverdeadas e de baixo potencial fisiológico. Assim, o objetivo deste trabalho foi avaliar a presença de Meloidogyne javanica e Pratylenchus brachyurus em plantas de soja e a relação com o desenvolvimento da planta, com a ocorrência de sementes esverdeadas e com o potencial fisiológico das sementes. Para isso, foram conduzidos três experimentos. O experimento I foi realizado utilizando plantas da cultivar Pintado, as quais foram submetidas à diferentes tratamentos de inóculo inicial e avaliadas quanto à fenologia e à produtividade e as sementes quanto ao potencial fisiológico. No experimento II foi analisada a resistência de dez cultivares de soja à M. javanica e P. brachyurus. No experimento III foram utilizadas plantas de duas cultivares de soja (TMG-115RR e M 7908 RR, previamente selecionados no experimento II), dois tratamentos de população de nematoides (0 e 7000 nematoides) e dois tratamentos de estresse hídrico (com e sem), cujas plantas foram avaliadas quanto a fenologia e produção de vagens e sementes, as raízes quanto à multiplicação de nematoides e as sementes quanto as determinações do grau de umidade, do potencial fisiológico (testes de germinação, tetrazólio, envelhecimento acelerado, condutividade elétrica, emergência de plântulas em areia e análise computadorizada de imagens de plântulas), ocorrência de sementes esverdeadas (método visual e fluorescência de clorofila) e citometria de fluxo. Com os resultados do experimento I, observou-se que a presença de M. javanica e de P. brachyurus em plantas de soja, independentemente da população inicial, provoca a redução do ciclo da planta devido exclusivamente à diminuição do estádio de senescência destas, porém a qualidade de sementes não foi afetada com população inicial de nematoides mais elevada (2400 espécimes). Todas as cultivares testadas no experimento II foram classificadas como suscetíveis, porém a cultivar TMG-115RR e M 7908 RR foram escolhidas para a condução do experimento III por permitirem boa reprodução dos nematoides em estudo e por serem consideradas por algumas empresas produtoras de sementes com sendo sensíveis a ocorrência de sementes esverdeadas. Finalmente, com os resultados do experimento III verificou-se que a população inicial de 7000 nematoides afeta, negativamente, a produção, a ocorrência de sementes esverdeadas e a qualidade de sementes. Entretanto, a intensidade do efeito de M. javanica e de P. brachyurus no desenvolvimento de plantas de soja, na ocorrência de sementes esverdeadas e no seu potencial fisiológico depende da população inicial e da espécie do nematoide, da cultivar e das condições ambientais as quais as plantas são submetidas.<br>Abiotic and biotic stresses in immature soybean plants, including nematodes presence, can result in premature plant death and can produce green seeds with low physiological potential. The aim of this study was to evaluate the Meloidogyne javanica and Pratylenchus brachyurus presence on soybean plants and the relationship with plant development, the green seeds occurrence and the seed physiological potential. For this, three experiments were done. The experiment I was carried out using the cultivar Pintado, which was inoculated with different nematode population and the phenology, productivity, and the seed physiological potential were evaluated. In experiment II it was analyzed the resistance of ten soybean cultivars to M. javanica and P. Brachyurus. In experiment III plants of two soybean cultivars (TMG-115RR and M 7908 RR, chosen by experiment II) were submitted to two nematode population treatments (0 nematodes and 7000) and two levels of water stress (with and without) and they were evaluated as phenology and production of pods and seeds, the multiplication of nematodes on roots and the determinations of moisture content, physiological potential (germination, tetrazolium, accelerated aging, electrical conductivity, sand seedling emergence and seedling computerized image analysis), the occurrence of green seeds (visual method and chlorophyll fluorescence) and flow cytometry on seeds. It was observed in the results of experiment I that the M. javanica and P. brachyurus on soybean plants, regardless of the initial population, decreases of cycle plant due the decrease on senescence phase, however, the seed quality was unaffected with higher initial nematode population (2400 nematodes). All the cultivars in experiment II were classified as susceptible, but the cultivar TMG-115RR and M 7908 RR were chosen for the experiment III because they allowed a good nematode reproduction and because some seed companies considered them as sensible to soybean green seed occurrence. Finally, the results of experiment III showed that the initial population of 7000 nematodes affected negatively the production, the soybean green seeds occurrence and the seed quality. However, the effect of M. javanica and P. brachyurus in the soybean plantas development, in the soybean green seeds occurrence and their physiological potential depends on the initial nematode population and species, cultivar and environmental conditions which the plants are exposed.

A series of crop rotations were evaluated for their impacts on soil-borne populations of root lesion nematodes, Pratylenchus neglectus. Population changes, measured as the ratio of P. neglectus adults counted at harvest to those counted at planting time, was recorded under a series of two-year rotations alternating winter wheat with either fallow, barley, pea, lentil, canola, and camelina. Fallow, barley, pea, and camelina were found to have a neutral effect on nematode populations (p &lt; 0.001). Winter wheat and canola caused significant increases in populations, while lentils caused significant decreases (p &lt; 0.001). Populations were sustained through winter following winter wheat and barley, but not canola, camelina, pea, lentil, or fallow treatments (p &lt; 0.001). In addition to the rotation study, cultivars of barley and canola were evaluated for their resistance to P. neglectus. Greenhouse trials for barley showed significant differences among 19 cultivars tested (p &lt; 0.001), with a 5-fold difference in P. neglectus multiplication separating the least- from the most resistant. Separation among cultivars was not found with canola (p = 0.20). The information gathered in this study will help Montana wheat growers better understand the impacts of their crop selections on this important pest.

Merr.) is present in practically every Brazilian state occupying an area of 23 million ha. The Central West Region is considered the greatest grain producer. The annual losses due to diseases are estimated around 15% and 20%. Among the main crop diseases in the Cerrados region, the ones caused by nematodes, especially H. glycines and P. brachyurus, are very important. Occurrence of P. brachyurus has been increasing, especially in areas of no till and pastures. The corn crop usually planted in crop rotation with soybean is also susceptible to P. brachyurus, which makes more difficult the nematode management. The use of genetics resistance is one of the most important control strategies due to its compatibility with other practices and for not being environmentally harmful. The purpose of this study was to evaluate the soybean and corn genotypes reaction to the nematode P. brachyurus, aiming the identification of resistance that could be of immediate use for growers in crop rotation system or to identify sources of resistance that could be used in plant breeding programs. The reaction of 50 soybean genotypes and 38 corn genotypes to P. brachyurus was studied under naturally infested field conditions. The experiments were conducted in 2008/09 in a randomized block design with split plot scheme, with evaluations at 30 and 60 days after emergence (DAE). Soybean experiment had eight replications and corn experiment, ten replications. The root samples were collected and taken to the laboratory for nematode extraction. The results were expressed as number of nematodes/ 10 g of roots. The reproduction factor (RF) was calculated considering the evaluation at 30 DAE as the initial population and the evaluation at 60 DAE as the final population. The soybean cultivars were separated into two groups with high and low susceptibility to P. brachyurus. Twelve genotypes presented interaction with the evaluation timing. Ten out of these twelve genotypes had the P. brachyurus population reduced from 45% to 70% at 60 DAE. The cultivars BRSGO Luziania RR, BRS Juliana, M-Soy 7211 RR, M-Soy 8411, TMG 103 RR and Emgopa 316 had 70%, 62%, 61%, 58%, 58% and 57% reduction in the population densities at 60 DAE. The cultivars were also separated into to groups according to the RF. The corn genotypes were separated into three groups. Seventeen corn genotypes had significant interaction with the evaluation timing and only the genotype Agromen 30A06 had the population density reduced (57%) at 60 DAE. All the other 16 genotypes had the nematode population increased between 30 and 60 DAE with increasing variation from 42% to 85%. The genotypes with lowest population density at the final evaluation (60 DAE) were: P 30F80, P30S31, Agromem 30A06, AS1575, DKB 499, GNZ 2500, DKB 350, DG 501 e NK Impacto. The soybean and corn genotypes that presented lower population density and RF along the evaluation period must be preferred for planting in infested areas and can be considered for future investigation for genetic resistance.<br>A soja (Glycine max L. Merr.) é cultivada em praticamente todos os estados do Brasil em um total de 21 milhões de ha, e a região Centro Oeste se destaca como a maior produtora do grão no Brasil. As perdas anuais de produção por doenças são estimadas em cerca de 15% a 20%. Dentre as principais doenças da cultura atualmente na região dos Cerrados, destacam-se aquelas causadas por nematóides, principalmente Heterodera glycines e Pratylenchus brachyurus. Tem-se observado o aumento da ocorrência de P. brachyurus, principalmente em áreas sob sistema de plantio direto e áreas com cultivos anteriores de pastagens. O milho, comumente utilizado em rotação ou sucessão com a cultura da soja, é também suscetível a P. brachyurus dificultando o manejo das áreas infestadas com esse patógeno. A utilização de resistência é uma das estratégias de controle mais importantes devido à sua compatibilidade com outras práticas de manejo e por não ser prejudicial ao meio ambiente. O objetivo do presente trabalho foi avaliar a reação de genótipos de soja e de milho ao nematóide das lesões radiculares P. brachyurus visando identificar resistência, ou possíveis fontes de resistência, que possam ser usadas em programas de melhoramento genético. Foi estudada a reação de cinquenta genótipos de soja e trinta e oito genótipos de milho ao nematóide das lesões radiculares em campo naturalmente infestado. Os experimentos foram conduzidos na safra 2008/09 em delineamento em blocos ao acaso em esquema de parcelas subdivididas no tempo, com avaliações aos 30 e 60 dias após emergência (DAE) com oito repetições para a soja e dez repetições para o milho. As raízes coletadas foram processadas em laboratório, sendo os resultados para densidade populacional expressos em número de indivíduos por dez gramas de raízes e o fator de reprodução (FR) foi calculado considerando a população final (pf) como sendo a segunda avaliação (60 DAE) e a população inicial (pi) a densidade populacional aos 30 (DAE). Para as cultivares de soja, o teste de médias para a densidade populacional separou os genótipos em dois grupos. Desses genótipos analisados, 12 apresentaram interação significativa com as épocas de avaliação, sendo que 10 delas apresentaram redução na densidade populacional aos 60 DAE variando de 45% a 70% de redução. As cultivares BRSGO Luziânia RR, BRS Juliana RR, M-Soy 7211 RR, M-Soy 8411, TMG 103 RR e Emgopa 316 RR destacaram-se com as maiores reduções na densidade populacional de P. brachyurus sendo de 70%, 62%, 61%, 58%, 58% e 57%, respectivamente. As cultivares BRSGO Jataí, BRSGO Silvânia RR, BRSGO Gisele RR, Emgopa 313 RR, M-SOY 7908 RR e Emgopa 313 destacaram-se por apresentarem baixas densidade aos 60 DAE. Da mesma forma, o teste de média para FR separou as cultivares em dois grupos, com maior e menor FR. Para os genótipos de milho o teste de médias para densidade populacional separou os genótipos em três grupos, sendo oito genótipos com as maiores densidades populacionais, quinze intermediários e quinze genótipos com as menores densidades populacionais aos 60 DAE. Dos genótipos avaliados, dezessete apresentaram interação significativa com as épocas de avaliação, sendo que apenas o genótipo Agromem 30A06 diminuiu a densidade populacional na segunda avaliação (57%). As demais apresentaram aumento na densidade populacional dos 30 até 60 DAE, variando de 42% a 85% de aumento. Os genótipos que apresentaram as menores médias populacionais na avaliação final foram: P 30F80, P30S31, Agromem 30A06, AS1575, DKB 499, GNZ 2500, DKB 350, DG 501 e NK Impacto. Com relação ao FR dos genótipos de milho o teste de média separou-os em três grupos. Os genótipos de soja e milho que apresentaram as menores densidades populacionais e menores FR devem ser preferidas para semeadura em áreas infestadas e para investigações futuras visando identificação de possível resistência genética, embora mais pesquisas sejam necessárias visando identificação de genótipos que possuam resistência a P. brachyurus.

In the Central West region of Brazil, the main soybean-producing region, the lesion nematode, Pratylenchus brachyurus, has become a major problem for culture can lead to significant losses. Weeds are also a limiting factor for obtaining high yields in soybean, and herbicide application to control it can affect the population of nematodes. This study aimed to evaluate the effect of herbicides, including glyphosate, on the population density of P. brachyurus on soybean conventional and transgenic cultivar. Two experiments were conducted in field conditions and another in a greenhouse in 2010/2011 harvest. The experimental design of the first was completely randomized in factorial arrangement 2x4+2, with four replicates. In greenhouse the design was completely randomized, with the same factorial design and six replications. The factors of the treatments consisted of cultivars, with two levels (BRSGO Iara and M-SOY 8001), and methods of weed control, with four levels (pull-off manual and chemical control with lactofen, chlorimuron-ethyl, and haloxifop-r). Two additional treatments consisted of transgenic cultivar BRS Valiosa RR with manual control of weeds and glyphosate. The assessments for the population of P. brachyurus, in the field were made at 45 and 90 days after emergence (DAE). We also evaluated the biomass, leaf area index (LAI), fresh and dry mass of pods. In the greenhouse were measured wet weight of roots and aboveground biomass at 30 and 60 DAI. The reproduction factor (RF) was determined in both trials. In the field, to calculate the RF was considered the population of nematodes 45 DAE as initial population. The use of transgenic cultivar and glyphosate did not affect the population density of P. brachyurus. There was no difference in the behavior of cultivars in relation to nematode P. brachyurus. Herbicides clorimuron, lactofen and haloxifop also did not affect the population of P. brachyurus. However clorimuron and lactofen reduced the growing of soybean cultivars BRS Iara and M-SOY 8001.<br>A soja (Glycine max L.) é hoje um dos principais produtos de exportação do Brasil e uma das principais commodities do mundo. Na região Centro Oeste do Brasil, a principal região produtora de soja, o nematoide das lesões radiculares, Pratylenchus brachyurus, tornou-se um grande problema para a cultura, podendo ocasionar importantes perdas. As plantas daninhas também constituem fator limitante para a obtenção de altos rendimentos na cultura da soja, e a aplicação de herbicidas para o seu controle pode afetar a população de nematoides. O presente trabalho teve como objetivo avaliar o efeito de herbicidas, incluindo glyphosate, sobre a densidade populacional de P. brachyurus, em cultivares de soja convencionais e uma cultivar transgênica. Foram conduzidos dois ensaios, um em campo e outro em casa de vegetação, na safra 2010/2011. O delineamento experimental utilizado no campo foi blocos casualizados, em arranjo fatorial 2x4+2, com quatro repetições. Em casa de vegetação foi utilizado inteiramente casualizado, com o mesmo esquema fatorial, e seis repetições. Os fatores dos tratamentos consistiram em: cultivares, com dois níveis (BRSGO Iara e M-Soy 8001) e métodos de controle das plantas daninhas, com quatro níveis: arranquio manual e controle químico com lactofen, chlorimuron-ethyl e haloxifop-r. Os dois tratamentos adicionais consistiram da cultivar transgênica BRS Valiosa RR, com controle manual de plantas daninhas e glyphosate. As avaliações para a população de P. brachyurus, no campo, foram efetuadas aos 45 e 90 dias após emergência (DAE). Também avaliaram-se biomassa, índice de área foliar (IAF), massa fresca e massa seca das vagens. Em casa de vegetação foram avaliados o peso fresco de raízes e a biomassa da parte aérea, aos 30 e 60 dias após a inoculação (DAI). O fator de reprodução (FR) foi determinado em ambos os ensaios. No campo, para cálculo do FR considerou-se a população do nematoide aos 45 DAE como população inicial. O uso da cultivar transgênica e do herbicida glyphosate não afetou a densidade populacional de P. brachyurus. Não houve diferença no comportamento das cultivares em relação ao nematoide P. brachyurus. Os herbicidas clorimuron, lactofen e haloxifop também não influenciaram a população de P. brachyurus. Entretanto, clorimuron e lactofen reduziram o crescimento vegetativo das cultivares de soja BRSGO Iara e M-Soy 8001.

The area harvested in 2014, was 55 million hectares, which represents in increase of 4.2 % compared to 2013 (52.8 million hectares). Rice, corn and soybean crops are the three main products that together accounted for 92.5 % of the grain production and accounted for 85.5 % of the harvested. Together these three crops produced around 177 million tonnes in the 2013/2014 season . Among the major diseases of soybean currently in the Cerrado region, we highlight those caused by nematodes, especially Heterodera glycines and Pratylenchus brachyurus. The maize and rice are used in rotation succession withis soybean crops and are host to the nematode making the management of areas infested with this pathogen more complex. The use of resistance is one of the most important strategies for plant disease control due to its compatibility with other management practices and for not being harmful to the environment. The objective of this study was to evaluate the reaction of of soybean, corn and rice genotypes to the root lesion nematode in order to obtain sources of resistance that could be included in plant breedind or be readily used by the farmers. Nine experiments were performed in the present study. Three experiments were conducted with soybeans in a completely randomized design. The first (experiment I) was conducted in field conditions with seventy cultivars (treatments) and six replications. The second experiment (experiment II) was conducted under controlled with 26 cultivars and seven replications. The third experiment (experiment III) was conducted in naturally infested field with 12 cultivars and six replications with assessments at 30, 60 and 90 DAS and harvest at 131 DAS. For corn, two experiments were conducted, the first with 30 genotypes and seven replications. The second (experiment II) was conducted in field conditions with ten genotypes and six replications evaluated at 30, 60 and 90 DAS and harvest at 119 DAS. For the rice crop, four experiments were conducted, three under controlled conditions (experiments I, II and III) and one in the field (experiment IV). All four experiments were set as a completely randomized design with six replications. In the present study no genotypes of soybean, corn or rice were found as resistant to P. brachyurus but some had promising results. The soybean cultivars TMG 132 RR, Emgopa 313 RR, M-Soy 8360 RR and CD 237 RR, rice cultivars BRS 01600, BRS Primavera and BRS Monarca and the corn hybrids, P30K75 and P30S31 presented lower nematode population densities or reproduction factors (RF). The soybean cultivars P 98Y11, BRSGO 8560 RR and P 98Y51 and the corn hybrids AG 1051, P3862H and SHS 3031 showed tolerance to the nematode. Given the results presented here, it is necessary for the studies to be continued in order to obtain resistant genotypes and to identify other promising crops that can be used in rotation with soybeans in areas with high infestation by the nematode. Also its important to study the behavior of this nematode in ecosystems where these crops are cultivated. Genotypes that provided lower nematode population densities should be preferred for future research in order to identify resistance.<br>A estimativa nacional de grãos para a safra 2013/2014 é de 193,9 milhões de toneladas. A área a ser colhida em 2014, de 55 milhões de hectares, apresenta acréscimo de 4,2% frente à de 2013 que foi de 52,8 milhões de hectares. O arroz, o milho e a soja são os três principais produtos em volume deste grupo, que somados representam 92,5% da produção e respondem por 85,5% da área colhida na safra 2013/2014. Juntas essas três culturas produzirão em torno de 177 milhões de toneladas na safra 2013/2014. Dentre as principais doenças da cultura da soja atualmente na região dos Cerrados, destacam-se aquelas causadas por nematoides, principalmente Heterodera glycines e Pratylenchus brachyurus. O milho e o arroz são utilizados em rotação ou sucessão com a cultura da soja e são culturas hospedeiras de P. brachyurus dificultando o manejo das áreas infestadas com esse patógeno. A utilização de resistência é uma das estratégias de controle mais importantes devido à sua compatibilidade com outras práticas de manejo e por não ser prejudicial ao meio ambiente. O objetivo do presente trabalho foi avaliar a reação de genótipos de soja, milho e arroz ao nematoide das lesões radiculares P. brachyurus a fim de obter fontes de resistência, que possam ser usadas em programas de melhoramento genético ou imediatamente por agricultores em áreas infestadas pelo nematoide. Foram realizados nove experimentos no presente estudo. Para a cultura da soja foram realizados três experimentos, o primeiro (Experimento I) em condições de campo com setenta cultivares (tratamentos) em delineamento inteiramente casualizado e seis repetições. O segundo experimento (experimento II) foi realizado em condições semi-controladas, com delineamento inteiramente casualizado, com 26 cultivares e sete repetições, e avaliação aos 60 dias após inoculação (DAI). O terceiro experimento (Experimento III) foi realizado em campo naturalmente infestado, em delineamento inteiramente casualizado com 12 tratamentos (cultivares) e seis repetições, com avaliações aos 30, 60 e 90 dias após semeadura (DAS), e colheita dos grãos aos 131 DAS. Para o milho foram conduzidos dois experimentos, sendo o primeiro em condições semi-controladas, com 30 tratamentos (genótipos), sete repetições e avaliação aos 60 DAI. O segundo (experimento II) foi conduzido em condições de campo com dez tratamentos (genótipos), seis repetições e avaliação aos 30, 60 e 90 DAS e a colheita dos grãos foi realizada aos 119 DAS. Para a cultura do arroz foram conduzidos quatro experimentos, sendo três em condições controladas (experimentos I, II e III) e um em condição de campo (experimento IV) todos em delineamento inteiramente casualizado e seis repetições. Nos experimentos I, II e III as avaliações foram realizados aos 45, 55 e 76 DAI. No experimento IV avaliou-se aos 86 DAS. No presente estudo, não foram encontrados genótipos de soja, milho e arroz resistentes a P. brachyurus, mas, encontrou-se resultados promissores. As cultivares de soja TMG 132 RR, Emgopa 313 RR, M-Soy 8360 RR e CD 237 RR, as cultivares de arroz BRS 01600, BRS Primavera e BRS Monarca e os híbridos de milho, P 30K75 e P 30S31 foram promissores por apresentarem menores densidades populacionais ou FR para o nematoide. As cultivares de soja P 98Y11, BRSGO 8560 RR e P 98Y51e os híbridos de milho AG 1051, P 3862H e SHS 3031 apresentaram tolerância a P. brachyurus. Diante dos resultados aqui apresentados, é necessário que os estudos sejam continuados com o intuito de se obter genótipos resistentes e também identificar outras culturas promissoras que possam ser usadas em rotação ou sucessão com a soja, em áreas com alta infestação por P. brachyurus, além de ser importante o estudo do comportamento desse nematoide nos ecossistemas onde essas culturas são produzidas. Os genótipos que proporcionaram menores densidades populacionais devem ser preferidos para investigações futuras visando à identificação de genótipos que possuam resistência a P. brachyurus.

Os nematoides que atualmente tem sido classificados como P. coffeae, demonstram ampla variabilidade morfológica, molecular e quanto à reação à diferentes hospedeiros. Por conseguinte, é essencial que se identifique corretamente as espécies e que se conheça a capacidade de parasitismo em alguns hospedeiros, para que se possa estabelecer medidas de controle e estimar eventuais riscos da entrada de um patógeno em uma área com hospedeiro suscetível. Neste sentido, a presente pesquisa propõe, numa primeira parte, uma medida de controle de P. jaehni (K5), que é muito agressivo à cafeeiro arábico, baseada no uso de plantas má hospedeiras. Na segunda parte deste trabalho é realizada uma caracterização da reação de diferentes espécies vegetais à quatro populações de Pratylenchus spp. Em virtude do exposto, este estudo objetivou avaliar a reação de diferentes cultivares de feijoeiro comum frente à P. jaehni (K5), visando seu possível uso no manejo de áreas cafeeiras infestadas e caracterizar a reação de diferentes espécies vegetais de importância econômica (café, porta-enxertos cítricos, banana e sorgo) frente à quatro populações de Pratylenchus spp. As populações inciais utilizadas nos experimentos variaram entre 180 e 200 nematóides. Em todos os experimentos, os nematóides foram extraídos das raízes pelo método de Coolen e D´Herde (1972) e, eventualmente, do substrato pelo método de Jenkins (1964). Foram realizados três ensaios: i) o primeiro com feijoeiro comum, onde todas as cultivares utilizadas foram resistentes à P. jaehni (K5), inclusive na réplica; ii) o segundo com porta-enxertos cítricos, no qual somente o limão-cravo foi hospedeiro de P. jaehni (K5); iii) o terceiro, no qual foi utilizado café, limão-cravo, banana e sorgo para o conhecimento das respectivas reações frente à 4 populações de Pratylenchus spp. Neste último experimento ocorreu uma reação hospedeira diferenciada para cada população. De acordo com os resultados, concluise que as cultivares de feijoeiro comum utilizadas apresentam potencial de uso em áreas cafeeiras infestadas por P. jaehni (K5), em consórcio ou em áreas de renovação de cafezal, e que as populações de Pratylenchus spp. são capazes de se reproduzir de forma diferenciada frente às espécies vegetais testadas.<br>Nematode populations that have been classified as P. coffeae show wide morphological, molecular and host range variability. Therefore, is essential the correct identification of the species and the knowledge about its parasitism capacity on some hosts, to ensure appropriate control measures and to estimate the entry risks of a pathogen in an area with susceptible host. In this sense, the present research report, firstly, propose a control measure of P. jaehni (K5), which is very aggressive to the arabic coffee, based on the use of poor host plants. In the second part, was carried out a host status evaluation of different plant species to four populations of Pratylenchus spp. In this context, were evaluated the reaction of different common bean cultivars to P. jaehni (K5), for its possible use in management of coffee areas infested with this nematode, and characterized the response of different economic important plant species (coffee, citrus rootstocks, banana and sorghum) to four populations of Pratylenchus spp (IB01P, IB02P, K5 e C1). The initial population used in experiments ranged between 180 and 200 nematodes. In all experiments, nematodes were extracted from roots by Coolen e D\'Herde´s method (1972) and eventually from the substrate by Jenkins´ method (1964). Three assays were conducted and results was as following: i) in the first one, with common bean, all tested cultivars were resistant to P. jaehni (K5), including the replica; ii) in the second one, with rootstocks, only rangpur lime was a good host of P. jaehni (K5); iii) in the third one, in which was used coffee, rangpur lime, banana and grain sorghum, was observed a differential host reaction for each nematode population evaluated. According to the present results, we suggest that common bean cultivars tested have great potential for use, or in intercropping or in crop rotation, in coffee areas infested by P. jaehni (K5) and populations of Pratylenchus spp. have different reproductive fitness in the plants species tested.

O nematóide das lesões Pratylenchus coffeae é um dos principais parasitos do cafeeiro e de outras culturas e sua variabilidade biológica, que dificulta a adoção de métodos de controle, contribui para aumentar a sua importância no Brasil. Pela importância da cafeicultura e a falta de estudos com esse nematóide no Brasil, foram realizados experimentos com dois de seus isolados (K5 e M2), com os objetivos de correlacionar densidades populacionais do nematóide aos danos causados e estabelecer possíveis fontes de resistência de cafeeiros ao isolado K5. Foram testadas diferentes densidades populacionais iniciais do isolado M2 em plantas (seis pares de folhas) e plântulas (dois pares de folhas) do cafeeiro arábico ‘Catuaí Vermelho’. As densidades populacionais utilizadas foram de 0, 333, 1.000, 3.000 e 9.000 nematóides por plântula ou planta. A avaliação ocorreu aproximadamente cinco (plântulas) e sete (plantas) meses após a inoculação. Os resultados mostraram que houve uma acentuada redução do crescimento das plântulas, bem como massa fresca das raízes e massa seca da parte aérea, já a partir das densidades mais baixas. A variação populacional (Pf/Pi) foi menor que um (1,0) para todas as densidades de inóculo, indicando que esta cultivar, no estágio de plântulas com dois pares de folhas, mostrou-se intolerante ao parasitismo. Em relação à inoculação das plantas, já com seis pares de folhas, não houve diferenças significativas nas variáveis analisadas e ocorreram decréscimos populacionais do nematóide, indicando que, nessas condições, ‘Catuaí Vermelho’ mostrou-se resistente ao isolado M2. Em relação ao isolado K5, foram realizados cinco experimentos, visando caracterizar as reações de genótipos de Coffea canephora ('Robusta' e 'Conilon'), além de C. arabica ‘Mundo Novo’, comparado às reações frente ao nematóide de galhas Meloidogyne incognita raça 2. No Experimento 1, foram utilizadas plantas de C. arabica ‘Mundo Novo’, inoculadas com 1.480 nematóides por planta (isolado K5 e M. incognita). Após sete meses da inoculação foi feita a avaliação, mostrando que o crescimento populacional dos nematóides foi alto e a reação de suscetibilidade. Mesmo em mudas desenvolvidas de cafeeiro ‘Mundo Novo’, o isolado K5 destacou-se como tão agressivo quanto M. incognita. Os outros genótipos testados, de C. canephora, foram inoculados com 3.000 nematóides por planta. Nos Experimentos 2 e 3, as linhagens IAC 4804 e IAC 4810 de ‘Robusta’ foram suscetíveis ao isolado K5, mas em um deles (IAC 4804) ocorreu grande variação entre as repetições em relação à M. incognita. Apenas o isolado K5 promoveu redução do crescimento do cafeeiro, evidenciado na variável massa fresca das raízes, em ambas as linhagens, sendo que IAC 4810 comportou-se como resistente a M. incognita. No caso de C. canephora ‘Conilon’, ambas as linhagens testadas (IAC 4764 e IAC 4765) foram resistentes ao isolado K5 e suscetíveis a M. incognita.<br>The lesion-nematode Pratylenchus coffeae is a major pest of coffee and other economic crops and its biological variability, which often makes difficult the adoption of control methods, contributes to increase the importance of this parasite in Brazil. Due to the importance of coffee production and the lack of studies involving this nematode species in Brazil, experiments were set with two of its available isolates (K5 and M2) to correlate initial population densities with the damage caused on coffee plants and to establish possible resistance sources in relation to the isolate K5. Different population densities of isolate M2 were tested in plants (six pairs of leaves) and seedlings (two pairs of leaves) of Coffea arabica ‘Catuaí Vermelho’. The population densities (Pi) were: 0, 333, 1.000, 3.000 and 9.000 nematodes per seedling or plant. The evaluation was done at approximately five (seedlings) and seven (plants) months after inoculation. The results showed that there was a marked reduction of the height, as well as root fresh weight and shoot dry weight of the seedlings, starting from the lower Pi values. The nematode population decreased (Pf/Pi < 1), indicating that this cultivar, at the seedling stage, was intolerant to parasitism. In relation to the inoculation of older plants, there were no significant differences in the growth parameters and the nematode population also decreased allowing ‘Catuaí Vermelho’ to be rated as resistant to the isolate M2. In relation to isolate K5, five experiments (referred to as 1, 2, 3, 4, and 5) were set to characterize the reaction of different genotypes of Coffea canephora ('Robusta' and 'Conilon') and C. arabica ‘Mundo Novo', as compared with their reaction to the root-knot nematode Meloidogyne incognita race 2. In Experiment 1, plants of C. arabica ‘Mundo Novo’ were inoculated with 1,480 nematodes per plant (K5 and M. incognita). The final evaluation after seven months of the inoculation showed a high populational increase of the nematodes and that both were pathogenic at a same extent. The other genotypes tested, belonging to C. canephora, were inoculated with 3,000 nematodes per plant. The genotypes (IAC 4804 and IAC 4810) of ‘Robusta’ were susceptible to isolate K5, but in one of them (IAC 4804) there was great variation among the repetitions in relation to M. incognita. The isolate K5 caused marked reduction in the growth of coffee Robusta plants as evidenced particularly through the root fresh weight values in both tested genotypes; in addition, IAC 4810 was rated as resistant to M. incognita. With regard to C. canephora 'Conilon', both tested genotypes (IAC 4764 and IAC 4765) were resistant to isolate K5 and susceptible to M. incognita.

To investigate nematicidal activity against the root-lesion nematode, Pratylenchus coffeae, the methanolic extracts were obtained from 5 medicinal plants in Vietnam. Methanol extracts of the 5 samples were screened for nematicidal activity against Pratylenchus coffeae in vitro. Of the plant extracts tested, a 5 mg/ml concentration of Terminalia nigrovenulosa extract showed the highest level (95.0%) of nematicidal activity against Pratylenchus coffeae at 9 h after treatment. The remainder of the plant extracts was followed by Cinnamomum camphora (66.7%), Jasminum suptriplinerve (30.8%), Premna integrifolia (6.7%), and Treptocaulon juventas (5.8%). A 2.0 mg/ml concentration of T. nigrovenulosa resulted in 13.3, 42.5, 62.5 and 86.7% at 1, 3, 5 and 7 h after treatment, respectively against Pratylenchus coffeae. Concentration of 0.1, 0.5, 1.0 and 2.0 mg/ml of T. nigrovenulosa resulted in 26.7, 71.7, 78.3 and 86.7% effectiveness respectively, at 7 h after treatment. Also, at 3 days after exposure to T. nigrovenulosa, hatch was inhibited by 60.0, 45.8, 17.5 and 5.8% at 0.1, 0.5, 1.0 and 2.0 mg/ml, respectively. Hatch was inhibited by 65.0, 50.8, 21.7 and 6.7% at 0.1, 0.5, 1.0 and 2.0 mg/ml respectvely, at 6 days after exposure to T. nigrovenulosa. The mortality rate and hatch inhibition increased with increasing incubation time for crude extract of T. nigrovenulosa.<br>Trong nghiên cứu này, 5 cây thuốc (Cinnamomum camphora, Jasminum suptriplinerve, Premna integrifolia, Terminalia nigrovenulosa, Treptocaulon juventas) đã được sử dụng làm nguyên liệu chiết xuất bằng methanol để thử hoạt tính kháng tuyến trùng Pratylenchus coffeae. Kết quả cho thấy chiết xuất từ cây Terminalia nigrovenulosa biểu hiện hoạt tính kháng tuyến trùng cao nhất (95,0%) sau 9 giờ xử lý với nồng độ 5 mg/ml. Hoạt tính kháng tuyến trùng của các chất chiết xuất được biểu hiện theo thứ tự giảm dần lần lượt là C. camphora (66,7%), J. suptriplinerve (30,8%), P. integrifolia (6,7%) và T. juventas (5,8%). Với nồng độ 2 mg/ml của chất chiết xuất từ T. nigrovenulossa biểu hiện hoạt tính gây chết tuyến trùng Pratylenchuss coffeae là 13,3; 42,5; 62,5 và 86,7% sau 1, 3, 5 và 7 giờ xử lý theo thứ tự. Ở cùng thời điểm sau xử lý 7 giờ, các nồng độ 0,1; 0,5; 1,0 và 2,0 mg/ml chiết xuất của T. nigrovenulossa có hoạt tính gây chết tuyến trùng Pratylenchus coffeae lần lượt là 26,7; 71,7; 78,3 và 86,7%. Bên cạnh đó, chất chiết xuất của T. nigrovenulossa cũng cho thấy tỷ lệ ức chế trứng nở cao. Cụ thể là sau 3 ngày xử lý với chất chiết xuất của T. nigrovenulossa, tỷ lệ ức chế trứng không nở là 60,0; 45,8; 17,5 và 5,8% at 0,1; 0,5; 1,0 và 2,0 mg/ml theo thứ tự. Sau 6 ngày xử lý với chất chiết xuất của T. nigrovenulossa, tỷ lệ ức chế trứng nở là 65,0; 50,8; 21,7 và 6,7% tại nồng độ 0,1; 0,5; 1,0 và 2,0 mg/ml theo thứ tự. Tỷ lệ gây chết và tỷ lệ ức chế của chất chiết xuất phụ thuộc vào cả thời gian và nồng độ xử lý.

Raspberry fields in the Lower Mainland of British Columbia are often prepared for replanting by fumigating and/or applying broiler manure in order to control soil borne pathogens such as those causing raspberry root rot. Broiler manure applications pose a threat of nitrate leaching and contamination of groundwater, while fumigation poses environmental risks because it is a broad-spectrum biocide. The aim of my research was to evaluate the effectiveness of readily available composts to reduce the impacts of plant parasitic nematodes and Phytophthora rubi and to improve plant growth. In 2013, two outdoor pot experiments using ‘Malahat’ cultivar red raspberry plants were conducted with raspberry field soils naturally infested with Pratylenchus penetrans. The first experiment held during the spring compared two concentrations of two composts, two concentrations of manure, and fumigation, in the presence or absence of P. rubi and arranged in a randomized complete block design. A 72-h flooding period after P. rubi inoculation was used to facilitate P. rubi infection. The second experiment during the summer compared the same amendment treatments without P. rubi inoculation to optimize conditions for P. penetrans. Soil samples taken from each pot were analyzed for nematode populations on the day of planting and at harvest (13 weeks later). The number of shoots and the biomass of shoots and roots were assessed at the end of both experiments, while root rot ratings were performed at the end of the first experiment. P. rubi was not detected in experiment 1 due to suboptimal environmental conditions after inoculation. Broiler manure suppressed root lesion nematode populations and improved plant growth relative to the control nearly as well as fumigation in experiment 1. There was no significant treatment effect on plant growth in experiment 2. However, broiler manure and fumigation did suppress P. penetrans populations relative to the control as in the first experiment. In contrast with earlier field studies, compost treatments did not suppress nematode populations or improve plant growth relative to the control. Overall, plant vigor and nematode suppression in compost treatments were limited. Based on previous compost studies, I speculate that longer experiment periods may be needed in order to detect benefits from composting.<br>Irving K. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior<br>The production expansion areas of soybean monoculture system result in increased fertilizers and pesticides application, and the intensification of some phytosanitary problems such as the damage caused by plant-parasitic nematodes. The arbuscular mycorrhizal fungi (AMF) increase plant growth and reduce the problems caused by nematodes in soybean. The objective of this work was to study the effect of arbuscular mycorrhizal fungi Rhizophagus clarus in the growth of soybean plants and the nematode Pratylenchus brachyurus suppression. Two trials were conducted in a greenhouse. In the first experiment evaluated the growth of soybeans in the presence and absence of the mycorrhizal fungus R. clarus. In the second trial was carry out the damage caused by the P. brachyurus in soybean grown in the presence and absence of R. clarus. In soybean flowering period evaluated the height, stem diameter, the dry matter of root and shoot, nodulation, the relative chlorophyll index, the nutrient content of the plant tissue, the number of the nematode in soil and penetrated to the root system, the reproduction factor of the nematode, the number of fungal spores in the soil and mycorrhizal colonization of roots. The results showed that mycorrhiza increased the growth, nodulation and nutrient uptake of soybean plants grown in soil with or without nematode. The mycorrhizal colonization reduced the penetration of the nematode in soybean roots, despite the greater number of nematodes in the soil. The presence of the nematode influenced mycorrhizal colonization of soybean plants. The use of sustainable management practices that favor the populations of AMF in the soil, can be a strategy for increasing and/or maintaining crop yields and reducing the affect caused by the P. brachyurus.<br>A expansão das áreas de produção e o sistema de monocultivo da soja resultam no aumento da aplicação de fertilizantes e agrotóxicos, e na intensificação de alguns problemas fitossanitários, como os danos causados pelos fitonematoides. Os fungos micorrízicos arbusculares podem contribuir para aumentar o crescimento das plantas e reduzir os problemas causados pelos nematoides na cultura da soja. O objetivo do presente trabalho foi estudar o efeito do fungo micorrízico arbuscular Rhizophagus clarus no crescimento das plantas de soja e na supressão do nematoide Pratylenchus brachyurus. Dois ensaios foram conduzidos em casa de vegetação. No primeiro ensaio foi avaliado o crescimento da soja na presença e na ausência do fungo micorrízico R. clarus. No segundo ensaio foi avaliado os danos causados pelo nematoide P. brachyurus na cultura da soja cultivada na presença e na ausência do R. clarus. No período de florescimento da soja avaliou-se a altura, o diâmetro de colmo, a fitomassa da parte aérea e radicular, a nodulação, o índice relativo de clorofila, o teor de nutrientes do tecido vegetal, o número de P. brachyurus no solo e penetrados no sistema radicular, o fator de reprodução dos nematoides, o número de esporos fúngicos no solo e a colonização micorrízica das raízes. Os resultados demonstraram que a micorrização aumentou o crescimento, a nodulação e a absorção de nutrientes das plantas de soja, cultivadas em solo com ou sem o nematoide. A colonização micorrízica reduziu a penetração do P. brachyurus nas raízes da soja, apesar do maior número de nematoides no solo. A presença do nematoide influenciou a colonização micorrízica das plantas de soja. A adoção de práticas de manejo sustentáveis que favoreçam as populações de FMAs no solo, pode ser uma estratégia para o aumento e/ou manutenção da produtividade das lavouras e redução dos danos causado pelo P. brachyurus.

This study analyzed the reaction of soybean lines to the nematode Pratylenchus brachyurus. The reaction of 70 genotypes, including commercial cultivars and under development by Pioneer Seeds were evaluated in a field experiment conducted in the city of Tupirama-TO. The major objective was to select tolerant or resistant cultivars to Pratylenchus brachyurus. Resistance, or tolerance, was determined by observing reproductive parameters in the plant root system, and geospatial aspects of the test and of the root system with a rating scale, developed during the work. The experimental design was completely randomized, with six replicates. The evaluation of the test and processing of soil and root samples to estimate the nematode population was done 80 days after planting. The results showed that all soybean cultivars evaluated hosted Pratylenchus brachyurus, but to different degrees. A smaller reproduction degree of P. brachyurus was observed in the lines of the Breeding Program of Pioneer Soybean Seeds, PHI022, PHI007, PHI027 and PHI031 and other lines us P99R01, M8352RR, Doko RC, P98R31 and PI200490 with final population = 0.71 nematodes g-1 of roots. It not possible to establish any correlations between the parameters evaluated: development of root and shoot and final nematode population.<br>O objetivo deste trabalho foi identificar a reação de linhagens de soja ao fitonematóide Pratylenchus brachyurus. Para isso, avaliou-se a reação de 70 genótipos, entre eles, cultivares comerciais e em desenvolvimento, pela Pioneer Sementes, em um experimento conduzido em área com lavoura de soja no município de Tupirama/TO. O delineamento experimental utilizado foi inteiramente casualizado, com seis repetições. Para tanto, foram avaliadas características como reprodução do nematóide no sistema radicular das plantas, aspecto geoespacial do ensaio e sintomas no sistema radicular, através de uma escala de notas, desenvolvida durante o trabalho. A avaliação do ensaio e processamento de amostras de solo e raízes para estimativa da população do nematóide realizou-se 80 dias após a semeadura da soja. Os resultados mostram que todas as cultivares de soja avaliadas hospedam Pratylenchus brachyurus, porém com populações finais variando de 0,71 a 27,95 nematóides. g-1. Uma menor taxa de reprodução de P. brachyurus foi observada em linhagens do Programa de Melhoramento de Soja da Pioneer Sementes, PHI022, PHI007, PHI027 e PHI031, como também em P99R01, M8352RR, Doko RC, P98R31 e PI200490 com população final = 0,71 nematóides. g-1 de raízes. Memoria para optar al título profesional de Ingeniero Agrónomo<br>Se realizó un estudio para determinar el estado sanitario de un huerto de manzanos (Malus domestica Borkh.) cultivar Brookfield sobre patrón M26, con alta infestación de nemátodos fitoparásitos, ubicado en la comuna de Las Cabras, Región del Libertador Bernardo O'Higgins, durante la temporada 2010-2011. Se tomaron muestras de suelo y raíces. Los individuos del suelo se extrajeron mediante el método de tamizado y en raíces mediante la incubación de tejidos. La determinación de géneros y especies, indicó la presencia dominante de Pratylenchus, y otros géneros en bajas poblaciones, tales como Mesocriconema, Helicotylenchus, Hemicycliophora, Tricodorus y Xiphinema. La identificación de especies de Pratylenchus spp. se realizó con claves taxonómicas, en base a caracteres morfométricos y morfológicos de la hembra, como la región labial, estilete y estructuras asociadas, intestino medio y recto, sistema excretor y órganos reproductivos, y presencia o ausencia de machos. Las mediciones correspondieron a los índices propuestos por de Man. Se estudiaron además los efectos de dos nematicidas granulares, oxamil (Vydate 10G) y cadusafos (Rugby 10G), en dosis de 30 kg/ha, sobre las poblaciones de nemátodos fitoparásitos, siendo aplicados en dos oportunidades, diciembre de 2010 y abril de 2011.

However the risks and damages caused by nematodes have been prominent among the diseases of soybean. Recently in soybean areas in Brazil has increased the occurrence of root-lesion nematode, Pratylenchus brachyurus, causing high damage to culture and rising due to the sequential planting of susceptible crops. The objectives of this study was to evaluate the density population of P. brachyurus on soybean growing areas under no-tillage in the northern state of Mato Grosso, and from that sampling to perform experiments under controlled conditions on the hostel of soybean and millet genotypes grown over most abundant in weed found in the region . Were sampled four properties of commercial soybean crops under no-tillage system, which occurred in the soybean harvest 2009/2010 and 2010/2010, 2010/2010 off-season and pre-planting of the 2010/2011 crop soybeans. Inoculum was obtained from populations of P. brachyurus from samples of soil and root crops in the areas of commercial soybean harvest of 2009/2010 and was made multiplying the population of P. brachyurus on susceptible crops. Upon obtaining the inoculum, three experiments were conducted under controlled conditions to check the reaction of nematodes in plants. The first experiment was conducted to evaluations the host of P. brachyurus in nineteen soybean genotypes and four pearl millet genotypes, compared with two standard species susceptibility to P. brachyurus. The second experiment was to evaluate the host of P. brachyurus in four nematode species of weeds. In the third experiment evaluated the effectiveness of seed treatment with Avicta Completo to control P. brachyurus in different soybean genotypes. All areas and seasons sampled showed P. brachyurus, at different levels of population density. Pearl millet cultivars ADR 300 and ADR 7010 showed low values of both reproduction factor (RF) of 0.2, being considered bad hosts on nematode. Over all soybean cultivars evaluated were susceptible to P. brachyurus. All weed species evaluated were susceptible to P. brachyurus and allowed the increase of the nematode population. The seed treatment with Avicta Completo contributed to reducing the reproduction rate of P. brachyurus on soybean plants.<br>A soja é uma das mais importantes leguminosas cultivadas no Brasil e no mundo. No entanto a cultura sofre riscos fitossanitários, e os danos causados por fitonematoides têm se destacado entre as doenças. Recentemente, nas áreas de cultivo de soja do Brasil, vem aumentando a ocorrência do nematoide das lesões radiculares, Pratylenchus brachyurus, ocasionando danos elevados e crescentes. Os objetivos do trabalho foram avaliar a densidade populacional de P. brachyurus em áreas de cultivo de soja sob o sistema de plantio direto na região Médio-Norte do estado de Mato Grosso, e a partir dessa amostragem efetuar experimentos em condições controladas sobre a hospedabilidade de cultivares de soja e milheto mais cultivados e plantas daninhas mais abundantes na região. Foram amostradas quatro propriedades de lavouras comerciais sob sistema de plantio direto, nas safras agrícola de soja 2009/2010 e 2010/2010, safrinha 2010/2010 e no pré-plantio de soja safra 2010/2011. Obteve-se inóculo de populações de P. brachyurus extraída das amostras de solo e raízes das áreas de lavouras comerciais da safra agrícola 2009/2010 e foi efetuada a multiplicação da população de P. brachyurus em culturas suscetíveis. No primeiro experimento avaliou a hospedabilidade do fitonematoide em 19 cultivares de soja e quatro cultivares de milheto, comparados com duas espécies padrão de suscetibilidade ao P. brachyurus. O segundo experimento consistiu em avaliar a hospedabilidade de P. brachyurus em quatro espécies de plantas daninhas. No terceiro experimento avaliou-se a eficiência do tratamento de sementes com Avicta Completo no controle de P. brachyurus em diferentes cultivares de soja. Todas as áreas e épocas amostradas apresentaram P. brachyurus, em diferentes níveis de densidade populacional. As cultivares de milheto ADR 300 e ADR7010 apresentaram baixos valores de fator de reprodução ambos com FR de 0,2, sendo considerados maus hospedeiros do nematoide. De modo geral as cultivares de soja avaliadas apresentaram suscetibilidade a P. brachyurus. Todas as espécies de plantas daninhas avaliadas foram suscetíveis a P. brachyurus e permitiram o aumento populacional do nematoide. O tratamento de sementes com Avicta Completo contribuiu para a redução do fator de reprodução de Pratylenchus brachyurus em plantas de soja.
