Abstract:
The understanding of ageing mechanisms of antioxidants in base oils is indispensable for the development of improved lubricants. In this study, a novel artificial ageing method based on the application of peroxide as oxidant is presented for improved monitoring of thermo-oxidative degradation processes in combination with mass spectrometry. Model oils containing aminic and phenolic antioxidants were aged and chemical structures of their oxidation products were elucidated by ultrahigh performance liquid chromatography and electrospray ionization high resolution (Orbitrap) mass spectrometry. Additionally, synergistic mixtures of four antioxidants were investigated, because the formation of condensed molecules from amines and phenols would have a major influence on the antioxidant potential but could not be detected in the bulk lubricant.

Abstract:
Biogenic amines are important indicators of food quality with recognized antioxidant capacity. Diets that are rich in these compounds promote several benefits for human health, although the consumption in excess may result in food poisoning. This study aims to screen the levels of biogenic amines in four colored cauliflowers, before and after cooking (boiling, steaming, and microwaving). In addition, the levels of tryptophan and 5-hydroxytryptophan, two serotonin precursors, were analyzed. Our results reveal that thermal processing shows a tendency to increase tryptophan levels and reduce 5-hydroxytryptophan in colored cauliflowers. A reduction of the tryptophan and increase in serotonin contents in ‘Cheddar’, steamed or microwaved, was observed. A higher level of histamine was observed in the genotype ‘Forata’ after cooking, whereas melatonin levels were higher after steaming and microwaving. The lowest levels of biogenic amines and amino acids were observed in ‘Graffiti’. All the colored cauliflowers that were analyzed presented a chemical quality index (CQI) below the pre-established limits, indicating that are safe for consumption, even after cooking. We conclude that the levels of biogenic amines and amino acids in colored cauliflower are safe for human consumption and do not present health risks. Therefore, the consumption of these genotypes, raw or cooked, is a good source of bioactive compounds.

Abstract:
A new method was proposed for the determination of underivatized biogenic amines based on ion-exchange chromatography coupled with mass spectrometry detection. The method was applied to the analysis of 10 biogenic amines in fresh and processed fish products. The amines were extracted from muscle tissue with water without any additional derivative step or sample clean-up. Separation of biogenic amines was done by the IonPac (4 × 50 mm) column, applying a gradient eluent by mixing formic acid (2 mol L−1) and Milli-Q water (formic acid concentration from 400 mM to 2 M). The results demonstrated a linear response in the range of 0.01 to 10 mg L−1. The detection limits for the fish products ranged from 20 ng/g up to around 400 ng/g for histamine and putrescine, respectively. Spermidine and spermine showed significantly higher detection limits. This current method can be used for the determination of biogenic amines in both fresh and processed fish products for regulatory purposes and monitoring food-safety issues relating to these amines, particularly histamine. It is also a useful method for evaluation of other commercial analytical test kits and commonly used methods that are possibly affected by the food matrix due to processing or other drawbacks arising from the derivatization process.

Abstract:
The contents of eight biogenic amines (tryptamine, phenylethylamine, histamine, tyramine, putrescine, cadaverine, spermidine and spermine) were determined in samples of processed cheese, either pasteurised (P) or sterilised (S), and consequently stored for either 22 or 57 weeks at the temperatures of 8°C and 22°C, respectively. Tyramine was quantitatively the most important biogenic amine (1.3–29.3 mg/kg); the highest level (P < 0.05) was found in P cheese stored for 22 weeks after processing at 8°C (P1). Regardless of tyramine, and with the exception of putrescine in S cheese stored for 57 weeks (2.2 mg/kg), the content of no biogenic amine exceeded 2 mg/kgin any tested sample. Significantly higher (by five to six orders of magnitude; P < 0.05) counts of total aerobic and facultative anaerobic microorganisms in P1 sample in comparison with all other samples were indicative of the cover leakage. No lactic acid bacteria were detected in any sample, the counts of coliforms and total anaerobes, respectively, were negligible and did not differ (P> 0.05) between samples. No sample of the tested long-term stored processed cheese was considered unsafe from the toxicological viewpoint.

Abstract:
Abstract. We report measurements of ambient amines and ammonia with a fast response chemical ionization mass spectrometer (CIMS) in a southeastern US forest and a moderately polluted midwestern site during the summer. At the forest site, mostly C3-amines (from pptv to tens of pptv) and ammonia (up to 2 ppbv) were detected, and they both showed temperature dependencies. Aerosol-phase amines measured thermal-desorption chemical ionization mass spectrometer (TDCIMS) showed a higher mass fraction in the evening with cooler temperatures and lower in the afternoon with warmer temperatures, a trend opposite to the gas-phase amines. Concentrations of aerosol-phase primary amines measured with Fourier transform infrared spectroscopy (FTIR) from micron and submicron particles were 2 orders of magnitude higher than the gas-phase amines. These results indicate that gas to particle conversion is one of the major processes that control the ambient amine concentrations at this forest site. Temperature dependencies of C3-amines and ammonia also imply reversible processes of evaporation of these nitrogen-containing compounds from soil surfaces in daytime and deposition to soil surfaces at nighttime. During the transported biomass burning plume events, various amines (C1–C6) appeared at the pptv level, indicating that biomass burning is a substantial source of amines in the southeastern US. At the moderately polluted Kent site, there were higher concentrations of C1- to C6-amines (pptv to tens of pptv) and ammonia (up to 6 ppbv). C1- to C3-amines and ammonia were well correlated with the ambient temperature. C4- to C6-amines showed frequent spikes during the nighttime, suggesting that they were emitted from local sources. These abundant amines and ammonia may in part explain the frequent new particle formation events reported from Kent. Higher amine concentrations measured at the polluted site than at the rural forested site highlight the importance of constraining anthropogenic emission sources of amines.

Abstract:
The history of young French people born to parents who have settled in France is completely different from that of their parents. The acculturation processes involved in their parents’ history have occurred in a complete social and political silence, raising neither polemic nor intensive research. By contrast, children are more visible, they protested and up to now they continue to request the attention of researchers in social sciences. As a political and electoral stake, they have unwillingly become an object of ethnicisation of the social, cultural and economic relationships. Such a position became susceptible to simplifications and drifts. This thesis explores, with a cross-cultural and psychosocial approach, the factors determining the processes involved in the identity construction of people who were born and grew up in conditions of cultural contact. We have chosen to work on the feelings of exclusion, social representations in the French society, identity-making orientations and the choices of identification. The participants in this research are young French people, 334 of whom issued from Maghrebian immigration and 158 from Turkish immigration, girls and boys. Their ages ranged between 15 and 25 years old. In order to evaluate the studied processes, we have built up a multidimensional research tool (questionnaire, scales, “Who am I ?” test). The results highlight the dynamic and strategic characters of identity, which result from social interactions in a cross-cultural situation. The comparative approach that was adopted and the intersection of the variables (i. E. Origin, sex and age) made it possible to clarify how the macro-social dimensions and the way social relationships are perceived may constitute a framework to understand the individual

Abstract:
This thesis concerns the development of novel catalytic approaches for the construction of stereocentres bearing a nitrogen atom. In 2011, the Dixon group reported a Ag(I)/cinchona-derived amino phosphine catalytic system for the activation of isocyanoacetates in asymmetric aldol and Mannich reactions. During this thesis work it was sought to extend the scope of this catalytic system to Mannich additions of other isocyanide pronucleophiles, then the focus was broadened to include Reformatsky and α-alkylation reactions of ketimine substrates. Chapter 1 gives an overview of the state of the art with particular emphasis on catalytic enantioselective additions to ketimines and the use of activated isocyanides as pronucleophiles. Chapter 2 describes the application of the Ag-catalysed enantio- and diastereoselective aldol reaction of isocyanoacetates to the concise asymmetric synthesis of the antibiotic chloramphenicol, which possesses a chiral stereodefined α-amino β-hydroxy motif. Chapter 3 details our efforts to expand the scope of the Ag(I)/amino phosphine catalytic system to the activation of more challenging isocyanides lacking an electron-withdrawing group in the α-position by investigating aldol and Mannich reactions of benzyl isocyanide. Chapter 4 describes how the scope of the Ag(I)/amino phosphine catalytic system was successfully extended to another pronucleophile, the versatile p-toluenesulfonylmethyl isocyanide (TosMIC). The first catalytic enantio- and diastereoselective addition of TosMIC to N-diphenylphosphinoyl (N-DPP) ketimines was developed, affording 2-imidazolines possessing two contiguous stereocentres with high yields and excellent levels of stereocontrol. Chapter 5 describes the development of a Ni(II)-catalysed Reformatsky reaction of N-DPP ketimines with ethyl bromoacetate and diethylzinc, providing racemic amines bearing a quaternary stereocentre in the α-position in good yields. Chapter 6 reports the serendipitous discovery of the α-alkylation of N-DPP ketimines with ethyl bromoacetate using visible light photoredox catalysis. The transformation, catalysed by ruthenium(II) and nickel(II) complexes under mild conditions, was optimised, its scope assessed and the mechanism investigated.

Abstract:

Abstract:

Full text
Abstract:
In this work, a sequential organocatalytic process for the stereodivergent synthesis of β-trifluoromethyl-α-amino acids using Erlenmeyer azlactones as starting material is presented. The strategy developed consists of a sequential catalytic approach, employing two catalysts that act independently to control the absolute configuration of two different stereocenters. The first step is a catalytic asymmetric hydrogen transfer of the activated double bond of the azlactone promoted by a Jacobsen type thiourea and Hantzsch ester as hydride donor. The second step involves a nucleophilic addition of an alcohol to the carbonyl moiety controlled by a chiral bifunctional catalyst typically used in the dynamic kinetic resolution of azlactones. The catalyst structure for the second synthetic step was thoroughly investigated in order to maximize the selectivity. Both products were achieved with a good diastereoselectivity and high enantioselectivity. Taking into account the obtained result it was possible to set up an initial study for the feasibility of straightforward one-pot procedure. In conclusion, with this work it was possible to set up a synthetic strategy for the synthesis of all four diastereoisomers starting from the set of starting material.

Abstract:
Amino acid transport is controlled by a system of transporter proteins whose function remains poorly defined in CHO cells. In this thesis, the relationship between amino acid transporter activity and culture performance parameters was investigated and used as a basis to explore new opportunities for improving cell line productivity. Transcriptomic analysis of transporter expression was first performed in two non-producing CHO cell lines and one antibody-producing CHO cell line during fed-batch cultures. Seven transporters were highly expressed across the three cell lines (ASCT1, CAT-1, GLAST, LAT1, SNAT2, xCT and y+LAT2). For all cell lines, the cystine-glutamate xCT transporter was significantly upregulated at stationary phase and formed part of a larger adaptive response to support the cellular availability of glutathione. The antibodyproducing cell line also upregulated ASCT1 and LAT1 transporter expression during stationary phase, which allowed the cells to maintain a high consumption rate of amino acids abundant in the antibody. Cells were next treated with inhibitors that block amino acid transport through individual or groups of transporters. Inhibition results confirmed that xCT transport activity is a key determinant of culture viability in all cell lines, and along with LAT1, also supports specific productivity during late-stage culture in the antibody-producing cell line. A directed evolution strategy was subsequently developed to increase xCT transport capability in the host. Evolved host cells demonstrated an increased capacity for GSH synthesis, increased resistance to a ROS insult, and a heritable improvement in cell growth but were not able to outperform the unevolved host in the transient production of an IgG. Finally, a mechanistic model to describe essential amino acid transport processes in CHO cells was constructed from transcriptomic and inhibitor data. This model can be used to direct future optimisation of amino acid concentrations in culture media to maximise cell growth and antibody production.

Abstract:
The thesis addresses several issues in prebiotic chemistry in the context of the origins of life through a systems chemistry approach. The first part is devoted to the study of chemical activation processes that are not only important in the formation of polymers, but also to feed the system with energy in order that a far from equilibrium state is maintained, a prerequisite for self-organization. It has been suggested that 5(4H)-oxazolones intermediates formed by C-terminus peptide activation could be involved in self-organization of life. To this aim, we have checked the reactivity of relevant prebiotic reagents previously proposed to activate α-amino acids. None of them led to a satisfactory C-terminus activation of peptides, showing that no general process for feeding a protometabolism of peptides with energy is identified yet, with the notable exception of N-carboxyanhydrides (NCAs) that can be formed through prebiotically relevant pathways. Additionally, we demonstrated that carbodiimides reagents are as efficient in the activation of N-carbamoyl amino acids as in that of the C-terminus of peptides in diluted aqueous media. The second part of the dissertation discloses new results in support of a process of coevolution of peptides and nucleotides. Firstly, a study of non-enzymatic aminoacylation reagents of the 3’-terminus of RNA is presented. Secondly, we assessed co-polymers of α-amino acids and nucleotides bound by phosphoramidate and ester linkages as potential players in chemical evolution. The kinetic relevance of these structures was demonstrated as well as potential chemical processes that allow their formation

Abstract:
The landmark study discussed in this chapter is ‘The contribution of excitatory amino acids to central sensitization and persistent nociception after formalin-induced tissue injury’, published by Coderre and Melzack in 1992. Previous studies in this field implicate a contribution of excitatory amino acids (EAAs), specifically l-glutamate and l-aspartate, to injury-induced sensitization of nociceptive responses in the dorsal horn of the spinal cord. Repetitive stimulation of primary afferent fibres demonstrated that l-glutamate and NMDA can produce ‘wind-up’ of neuronal dorsal horn activity, and this is blocked by application of NMDA antagonists. This study uses the formalin test as a behavioural model to investigate the mechanisms underlying central sensitization and the role of EAAs, NMDA, their receptors, and their antagonists in this process.

Abstract:
As Chapter 17 makes clear, the asexualist/sexualists controversy continued even as Johann Hedwig and Karl von Nägel demonstrated the existence of sex in cryptogams by discovering the Alternation of Generations (1782, 1784), hybridizers A. F. Wiegman and Carl Friedrich von Gaertner recieved prestigious prizes for their work, and Giovanni Battista Amici and Adolphe-Theodore Brongniart discovered—and confirmed—the pollen tube. Unconvinced, Matthias Jacob Schleiden, co-founder of the cell theory, insisted that ferns grow asexually from spores, and that spores, not seeds, are the primary units of propagation in seed plants also. He argued (1853) that the entire life-cycle of seed plants is based on duplicative cell divisions that produce seeds entirely by vegetative processes. Following the Aristotelian doctrine that the female parent provides the material substance of the embryo, he concluded pollen must be a female structure that reproduces vegetatively—thus making the case for a unisexual, plants-as-female model.

Crawford, Margo Natalie. The Counter-Literacy of Black Mixed Media. University of Illinois Press, 2018. http://dx.doi.org/10.5406/illinois/9780252041006.003.0004.

Abstract:
The third chapter brings the mixed media of the BAM and the 21st century together as Crawford shows that black art, after the Black Arts Movement, continues to create an alternative way of approaching art as process, not as object. The first part of this chapter shapes this process-oriented counter-literacy around the Black Arts Movement textual productions of the black book as the open book. She explores the openness of word and image texts and argues that they produce the lack of closure of black post-blackness. Through the text paintings of Glenn Ligon and the word and image books of Amiri Baraka, June Jordan, John Keene, Christopher Stackhouse, and others, this chapter unveils the unbound nature of mixed media as one of the most innovative legacies of the Black Arts Movement.

Full text
Abstract:
In the next two decades epigenetics could revolutionize understanding and treatment of diseases of the central nervous system. New research already demonstrates that manipulation of epigenetic mechanisms in vivo and in vitro can ameliorate a host of pathogenic processes associated with neurodegenerative disorders such as Alzheimer’s disease (AD), Parkinson’s (PD), amyotrophic lateral sclerosis (ALS), Huntington’s (HD), and multiple sclerosis (MS), among others. These advances have come relatively rapidly for a field that is still in its infancy compared to the much longer history of epigenetics in developmental biology. Epigenetic modifications are all-encompassing, from nucleotides to amino acids. They are capable of altering transcriptional to biochemical activity in a consistent manner across thousands of genes and hundreds of biologic pathways, yet they can do so differentially even in individuals or cells with identical gene codes. As such, epigenetic modifications are likely to touch on virtually all the mechanisms described in this book.

Burton, Derek, and Margaret Burton. Excretion. Oxford University Press, 2017. http://dx.doi.org/10.1093/oso/9780198785552.003.0008.

Full text
Abstract:
Excretion is the removal of metabolic wastes such as ammonia, carbon dioxide, ions and water as well as toxic xenobiotics and metals. The process involves the gills, kidney, liver and rectal gland (elasmobranchs and coelacanth). In the liver, amino acids, haemoglobin, steroids and molecules resulting from human activities are transformed to excretable products. The rectal gland excretes ions, notably Na+ and Cl−. The kidney in teleosts has a distinction between an anterior head-kidney containing haematopoietic tissue and endocrine tissue and the posterior region with nephrons (kidney tubules). Fish nephrons generally have a Malphigian corpuscle with a glomerulus but the structure varies between fish taxa and some marine teleosts lack a glomerulus. Control systems for fish excretion are unclear but it is expected that various hormones influence excretory homeostasis.

Abstract:
AbstractMetabolism is a fundamental process for all cellular functions. For decades, there has been growing evidence of a relationship between metabolism and malignant cell proliferation. Unlike normal differentiated cells, cancer cells have reprogrammed metabolism in order to fulfill their energy requirements. These cells display crucial modifications in many metabolic pathways, such as glycolysis and glutaminolysis, which include the tricarboxylic acid (TCA) cycle, the electron transport chain (ETC), and the pentose phosphate pathway (PPP) [1]. Since the discovery of the Warburg effect, it has been shown that the metabolism of cancer cells plays a critical role in cancer survival and growth. More recent research suggests that the involvement of glutamine in cancer metabolism is more significant than previously thought. Glutamine, a nonessential amino acid with both amine and amide functional groups, is the most abundant amino acid circulating in the bloodstream [2]. This chapter discusses the characteristic features of glutamine metabolism in cancers and the therapeutic options to target glutamine metabolism for cancer treatment.

Abstract:
Abstract Airborne molecular contamination poses a serious problem for advanced wafer fabrication as the devices are continually scaled down. The amount of this contamination may be only a few monolayers, which are extremely difficult to detect by the commonly used analytical techniques, such as FTIR. ToF-SIMS has extremely high surface sensitivity for the analysis of trace contaminants on wafer surfaces. The high mass resolution of ToF-SIMS is also a powerful tool for the identification of the contaminants. In the current study, ToF-SIMS is used to monitor the build-up of airborne amine contamination on Black Diamond1 surfaces. It has been found that cleaning of the Black Diamond surfaces using wet chemicals can lead to photoresist poisoning. Thermal desorption-GC-MS analysis revealed that wet cleaning would result in the accumulation of hydrocarbons on the Black Diamond surfaces. ToF-SIMS shows that amines can build up gradually on the Black Diamond surfaces after wet cleaning, probably via airborne molecular contamination. For the Black Diamond wafers which did not go through the wet cleaning process, there was no significant increase of amines on the wafer surfaces. The amount of amines on the Black Diamond surfaces depends on the chemicals used in the cleaning processes and the wafer storage conditions. The level of amine contamination can be significantly reduced after the samples are heated up to 300°C for a few minutes in inert atmosphere.

In SPE Nigeria Annual International Conference and Exhibition. SPE, 2021. http://dx.doi.org/10.2118/207183-ms.

Full text
Abstract:
Abstract This work has been carried out to model and simulate a typical acid gas absorption process using Aspen HYSYS process simulator. The chemical components involved in the process development were water, methane, ethane, propane, higher alkanes, carbon dioxide, hydrogen sulphide, nitrogen and amines: monoethanolamine (MEA), diethanolamine (DEA), triethanolamine (TEA) and methyldiethanolamine (MDEA). The fluid package selected for the simulation before entering the simulation environment was Acid Gas – Chemical Solvents. In the simulation environment, the model was developed by picking an absorber from the Model Palette, placing it and assigning the input and the output streams involved before inputting the parameters required for model convergence. The carbon dioxide-rich feed gas was made to enter the absorber at the bottom inlet stream while the lean amine stream entered at the top inlet and showered down on the uprising gas thereby trapping the carbon dioxide molecules within the gas. The top product from the absorber was the treated gas while the amine solution and the trapped carbon dioxide left the absorber as the bottom product. Different simulations were run to investigate the performance of the amines under the same operating conditions. It was discovered that, of all the four amine solvents considered in this work for the removal of carbon dioxide by chemical absorption, MEA had the highest efficiency but would require more dehydration because it had the highest water content. DEA was also found to scrub the carbon dioxide down to acceptable levels. However, TEA and MDEA barely scrubbed any carbon dioxide under these conditions, as their carbon dioxide compositions were found to be unacceptable. The analyses of the results obtained from the simulations indicated that Aspen HYSYS can be used to study the process of acid gas absorption successfully.

Abstract:
Research goal of this paper represents a novel method of synthesis of an amino-modified, highly effective natural polymer adsorbent kraft lignin based, functionalized with magnetite nanoparticles, for effective removal of chromate anions from water solution. Amino-modified lignin microspheres (LMS-NM) were synthesized via inverse suspension copolymerization of kraft lignin with poly(ethylene imine) grafting-agent and epichlorohydrin cross-linker. The porous structure of the synthesized adsorbent was formed using a 5.0 wt.% sodium-alginate solution. Additional efficiency in the removal of chromium anions from water was achieved by functionalization of lignin microspheres with magnetite (NM) nanoparticles obtained by co-precipitation and additional introduction of amino groups. Structure of formed LMS-NM was confirmed via Furrier Transform Infrared (FTIR) Spectroscopy. Optical microscopy analyses showed the creation of sphere-shaped materials and showed the diameter of prepared LMS-NM in the range of 500 to 800 µm. The removal of chromate anions from water solution was performed in a batch system. The result confirmed high adsorption efficiency and potential application of this nature-based adsorbent for heavy metal anions removal. Abstract:
Acid gas removal is a critical process step in natural gas processing and syngas production for ammonia and other uses. Application of a liquid phase turbocharger to the acid gas removal unit (AGRU) results in significant energy savings and improvement to reliability, availability and maintainability (RAM) of the plant. This paper describes conventional configurations with high pressure pumps and new configurations utilizing liquid phase turbochargers. Design of the equipment, process operations and controls and reliability analysis are included. The results of a RAM study comparing conventional configurations to those incorporating liquid phase turbochargers in multiple cases are also presented. From the RAM study, it can be concluded that flow sheet configurations that include a liquid phase turbocharger consistently provide lower plant downtime and maintenance costs as compared with conventional flow sheet configurations. This is in addition to the energy savings that result from energy recovery with the application of the liquid phase turbocharger to the AGRU. For the reference plant used in the study, the maintenance cost savings are as great as $2.5M over the 20 year lifetime of the plant and average annual downtime reduction is as much as 19.8 hours.
