Abstract:
Proliferative vitreoretinopathy (PVR) is the most common cause of late anatomical failure of retinal detachment surgery. Efforts to modify this vitreoretinal scarring response have so far proved clinically unsuccessful, with surgical and visual outcomes remaining poor. This work is aimed at identifying strategies to improve outcomes in eyes at high risk of PVR development following open globe trauma (OGT), and those with established PVR disease. Two prospective clinical trials investigating the benefit of adjunctive corticosteroids in these two populations were conducted in a total of one hund

Abstract:
Cellular migration and membrane contraction are two vital steps in pathogenesis of proliferative vitreoretinopathy (PVR). Several factors, related to cell activity, are involved in induction of cell migration and cell-induced membrane contraction. However, the roles of some of these factors in PVR pathophysiology are still to be clarified. The studies described in this thesis were undertaken to investigate the factors affecting cellular migration, mainly HRPE cells, which may modify cellular migration during proliferative vitreoretinopathy. Bioengineering
Ph.D.
Central to ocular health is the vitreous body, a complex, gelatinous tissue filling the space between the lens and retina. It is a natural polymeric hydrogel whose delicate architecture of collagen and hyaluronic acid loses its mechanical structure under the influence of degeneration or destruction, leaving the retina vulnerable to injury and disease. Since World War II, combat ocular trauma has increased six-fold while the population of aging veterans continues to grow in tandem. With the advancement of vitreoretinal surgery, the prognosis of PVR has been improved. Long term success, however, is often prevented by the recurrence of proliferative contractile membranes. Pharmacological management using various agents to inhibit cellular proliferation has been tried as an adjuvant therapy of PVR. Alternatively, using anti-adhesion therapy to prevent cells attac

Abstract:
Proliferative vitreoretinopathy (PVR) is characterized by the formation of fibrocellular membranes in the vitreous and on the surfaces of the retina. These membranes exert tractional forces on the retina, causing retinal tears and/or detachments. The purpose of this study is to develop a reproducible animal model that closely mimics PVR in the clinical setting. The retinal pigment epithelial (RPE) cell is the major cell type found in PVR membranes. We cultured RPE cells from a human epiretinal membrane (ERM) obtained at surgery from a patient with PVR, and injected them into the right e
