The goal of this master's thesis is to perform strategic analysis and assessment of corporate strategy by using the system of Balanced Scorecard. The thesis itself consists of two parts. The first (theoretical) part describes mission, vision, strategy and the most common tools and methods for creating strategic analysis. This analysis contains external and internal analysis. The results are summarized in SWOT analysis, which is the baseline for creation of corporate strategy. The second (practical) part combines theoretical part and real data of analyzed company.

Swahili ist keineswegs eine besonders „exotische“ Sprache, aber dennoch fällt es schwer, eine gu-te moderne Grammatik des Standard-Swahili (Kiswahili sanifu) zu finden. Insbesondere die Inter-pretation der vorkommenden Tempora, Aspekte oder Modi des Verbs sind in den erhältlichen Grammatiken diskrepant bis widersprüchlich. Der Artikel versucht, einen systematischen Ansatz für eine strukturelle Matrix des TAM-Systems im Swahili herauszuarbeiten. Dabei beziehe ich mich auf die von Shankara Bhat in The Prominence of Tense, Aspect and Mood (1999) vorge-schlagene typologische Methode. Bhat legt dar, dass nicht jede Sprache jede Dimension von TAM gleich gewichtet, sondern dass jeweils eine davon vorherrscht. Eine Analyse von TAM in einer be-stimmten Sprache muss sich daher an der prominenten Dimension orientieren. Im Artikel wird herausgearbeitet, dass das Tempus die strukturierende Dimension im Swahili darstellt und dass im Gegensatz dazu der Aspekt nicht systematisch grammatikalisiert ist. Dabei war es nötig, die im Swahili vorkommenden TAM-Formen funktional zu interpretieren und zu benennen, da diese in der aktuellen Literatur teilweise sehr unterschiedlich analysiert werden.

ENGLISH ABSTRACT: This study is divided into five chapters. The first chapter which marks the introductory chapter deals with the general introduction to the entire study, the scope of the study, statement of the aim and objectives of this study, also looks at the research methods and gives St J Page Mbalana Yako's brief biographical sketch. The second chapter provides a brief theoretical background to the study of poetry. This forms the basis upon which the entire study will rest as it provides different poetic devices and stylistics, which reveal what makes poetry. The third and fourth chapters concentrate on a critical evaluation of Yako's poems. The poetry, which is presented in these chapters, is selected from Yako's anthology entitled Ikhwezi. It represents a particular genre from a wide range of poetic forms that Yako has written. Chapter three concentrates on educators and the achievements of certain individuals. Chapter four concentrates on traditional leaders. It is in this chapter that Yako displays his expertise in the use of excellent and appropriate poetic devices and stylistics in his poetry. The fifth chapter contains general conclusions drawn from the entire study. Translations are provided for each poem and are contained in the appendix that is found at the end of this study, which is immediately followed by the bibliography. Yako's endeavour to bring light to the nation through poetry is admirable. Further research will unearth more art and craft in this author's poetry.
AFRIKAANSE OPSOMMING: Hierdie studie is verdeel in vyf hoofstukke. Die eerste hoofstuk bied In algemene inleiding tot die hele studie. Dit omskryf die omvang van die studie, die doelstelling en oogmerke, die navorsingsmetodologie, en gee In kort biografiese skets van St. J. Page Mbalana Yako. Die tweede hoofstuk gee 'n kort teoretiese agtergrond oor die studie van poësie. Dié hoofstuk dien as die basis vir die hele studie, omdat dit verskillende poëtiese middele en stylvorme uitlig wat die verskynsel van poësie verklaar. In die derde en vierde hoofstukke word gefokus op In kritiese evaluering van Yako se gedigte. Die gedigte wat in hierdie hoofstukke aangebied word, is geselekteer uit Yako se bloemlesing, getiteld Ikhwezi. Dit verteenwoordig 'n sekere genre uit In wye reeks poëtiese vorme wat deur Yako gebruik is. Abstract:

Prominent Feature Analysis (PFA) is a reliable and valid writing assessment tool, derived from the writing it is used to assess. PFA, used to assess on-demand expository essays in Grades 3–12, uncovers positive and negative characteristics of a sample. To extend PFA to a new academic level and genre, I assessed scientific writing of 208 undergraduates, identifying 35 linguistic and 20 scientific prominent features. An essay could earn up to 28 positive (24 linguistic and four scientific), and up to 27 negative marks (11 linguistic and 16 scientific). The minimum prominent features number in a paper was 3, the maximum was 25 (M = 12.45, SD = 3.88). The highest positive and negative prominent features numbers noted were 17 (M = 4.11, SD = 3.96), and 16 (M = 8.34, SD = 3.25) respectively.

Rasch analysis revealed a good data-model fit, with item separation of 5.81 (.97 reliability). The estimated feature difficulty of items spanned over 10 logits; common errors were easier to avoid than “good writing” characteristics to exhibit. Significant correlations among linguistic, but not between linguistic and scientific features, suggest writing proficiency does not assure excellence in scientific writing in novices. Ten linguistic features significantly strongly and moderately inter-correlated with each other, appearing to represent writing proficiency. Student GPA correlated significantly with the raw prominent features scores (r = .37; p < .01), and negatively with the sum of negative linguistic features (r = –.40, p < .01), providing support for scale’s validity, and suggesting that good students are better at avoiding common writing errors than less able learners. Additionally, PFA scores positively significantly correlated with composite ACT scores.

To investigate PFA’s ability to track change in writing over time, I compared 2 sets of prominent features scores of 25 students. In comparison with earlier essays, later (longer) essays exhibited significantly more positive, and more negative features. Prominent features scores did not correlate significantly between the sets. This suggests, that while PFA is a valid and appropriate tool for analysis of undergraduate scientific writing, it was not suitable for tracking change in writing ability in this small sample.

Abstract:
Water is the most common solvent for most biological reactions, therefore it is vital that we fully understand water and all its properties. The complex hydrogen bonding network that water forms can influence protein-protein and protein-substrate interactions and can slow protein conformational shifts. Here, I examine an important property of water known as energetic roughness. The network of interactions between individual water molecules affect the energy landscape of proteins by altering the underlying energetic roughness. I have attributed this roughness to the making and breaking of hydrogen bonds as the network of hydrogen bonds constantly adopts new conformations. Through a novel computational approach I have analyzed five prominent water models and have determined their inherent roughness to be between 0.43 and 0.62 kcal/mol.

Abstract:
Agriculture receipt data were obtained from USDA Agriculture Statistics on four prominent tobacco producing states-North Carolina, Kentucky, Tennessee, and Virginia--for the period of 1946 through 1995. The data were adjusted for inflation according to the Consumer Price Index and averaged for five-year periods beginning with 1946-1950 and continuing through 1991-1995. There were four objectives of the study. The first objective was to compare these states for annual agriculture receipts, crop receipts, and livestock receipts. State total agriculture receipts ranged from 1.8 to 3.2 billion dollars in 1946-50 and from 1.4 to 4.1 billion dollars in 1991-95. For total agriculture receipt, rankings were as follows: North Carolina highest, Kentucky intermediate, and Tennessee and Virginia lowest and about equal. State rankings for crop and livestock receipts were the same as those for total agriculture receipts except the North Carolina ranking for livestock receipts went from lowest to highest among the states during the period of study. In Kentucky and Tennessee, crop and livestock contributions were approximately equally throughout the study. Crop receipts exceeded livestock receipts in NC until the 1980s, while in Virginia livestock receipts consistently exceeded crop receipts. The second objective was to determine the contribution of tobacco receipts and their relationships to the receipts of other agriculture commodities. Average state tobacco receipts in dollars ranged from 254 million to 1.8 billion dollars in 1946-50 and from 129 to 723 million dollars in 1991-95. State rankings for tobacco receipts were as follows: North Carolina highest, Kentucky intermediate, and Tennessee and Virginia lowest and approximately equal. Tobacco receipts (dollars) for Kentucky, Tennessee, and Virginia were relatively consistent over the 50-year period; however, the tobacco receipts for North Carolina decreased from 1.8 billion to 723 million dollars during the period. When tobacco receipts were considered as a component of total agriculture receipts, the tobacco contribution decreased from 54% to 18% in North Carolina and from 35% to 26% in Kentucky, whereas the proportion remained rather consistent in the 10% to 15% range for Tennessee and Virginia. When tobacco receipts were expressed as a percentage of crop receipts, the percentages were highest in Kentucky, intermediate in North Carolina, and lowest in Virginia and Tennessee. Correlations between tobacco and total agriculture receipts were positive and significant for each state except North Carolina. The third objective of the study was to examine the contribution of specific crop and livestock enterprises to agriculture receipts during the 1981-1995 period. Total agriculture receipts increased in North Carolina but decreased in the other states. The increase in North Carolina resulted from increases in livestock receipts (collectively), swine, poultry, vegetables, and other crops. The decrease in Kentucky, Tennessee, and Virginia resulted from decreases in livestock (collectively), dairy, swine, crops (collectively), fruit and tobacco receipts. North Carolina agriculture income has increased and reflects more contribution from livestock (collectively), swine, poultry and less from tobacco. Kentucky agriculture income has decreased and reflects decreases from livestock and crops. Kentucky agriculture receipts are more dependent upon tobacco than are the receipts in the other states. The fourth objective was to relate diversification patterns from the other states to Kentucky's present status and future opportunities. In 1995, tobacco contributions to agriculture receipts were North Carolina (15%), Kentucky (21%), Tennessee (11%), and Virginia (8%); poultry contributions were North Carolina (29%), Kentucky (4%), Tennessee (10%), and Virginia (31%). Although considered minor contributors at present, hay and other crops (greenhouse and nursery plants) show promise for further diversity of Kentucky agriculture.

Abstract:
In this work a high resolution data set of ionic composition from a firn core collected at GV7 site (East Antarctica) is presented. An efficient decontamination procedure in cold room and chemical analysis by Ion Chromatography were performed. The most relevant sources delivering aerosol to the Antarctic continent were studied and these results can be pointed out: - as concerning primary marine source (sea salt), a good correlation was found between the main chemical markers related to this input (Na+, Cl-, Mg2+). The highest peaks of sea salt components were found to occur in winter time; - crustal source can be studied through Calcium content and especially its non sea salt fraction (nssCa2+). No clear seasonal pattern was observed in Calcium record at this site; - the markers of secondary marine source (biogenic activity), i.e. Sulfur oxidized compounds (MSA and nssSO42-), show a clear seasonal pattern with concentration maxima in summer, when a large phytoplanktonic bloom occurs in the surrounding ocean causing larger emissions of dimethylsulfide; - NO3- was found not to be affected here by post-depositional effects, that are able to modify its concentration in low accumulation sites. Moreover, thanks to the relatively high accumulation rate at GV7, it was possible to achieve an accurate dating of the core, by counting successive annual snow layers, marked by those parameters presenting a clear seasonal trend along the stratigraphy, or by the presence of specific dated events such as documented volcanic eruptions. nssSO42- was chosen for dating purpose, by using its maxima occurring in full summer period and, starting from the surface, 26 summer peaks were identified along the whole GV7 core. These preliminary results appear to be promising for the obtainment of a record at high temporal resolution on the climatic variability in the last millennium by the chemical and isotopic analysis of the 250 m deep ice core drilled in this site.

Abstract:
The aim of this study was to investigate the composition of an acoustic scattering layer in the North Sea that is particularly strong at 38 kHz. A full definition of the biological composition of the layer, along with its acoustic properties, would allow for it to be confidently removed from data collected during acoustic fish surveys, where it presents a potential source of bias. The layer, traditionally and informally referred to as consisting of zooplankton, appears similar to others observed internationally. The methodology utilised in this study consisted of biological and acoustic sampling, followed by application of forward and inverse acoustic modelling techniques. Acoustic data was collected at 38, 120 and 200 kHz in July 2003, with the addition of 18 kHz in July 2004. Net samples were collected in layers of relatively strong 38 kHz acoustic scattering using a U-tow vehicle (2003) and a MIKT net (2004). Acoustic data were scrutinised to determine actual backscattering, expressed as mean volume backscattering strength (MVBS) (dB). This observed MVBS (MVBSobs) was compared with backscattering predicted by applying the forward problem solution (MVBSpred) to sampled animal densities in order to determine whether those animals were responsible for the enhanced 38 kHz scattering. In most instances, MVBSobs > MVBSpred, more pronounced at 38 kHz. It was found that MVBSpred approached MVBSobs more closely with MIKT than with U-tow samples, but that the 38 kHz mismatch was present in both. Inversion of candidate acoustic models predicted gas-bearing scatterers, which are strong at 38 kHz, as most likely to be responsible for this. Potential sources of inconsistencies between MVBSpred and MVBSobs were identified. The presented forward and inverse solutions infer that although the layer often contains large numbers of common zooplankton types, such as copepods and euphausiids, these are not the dominant acoustic scatterer at 38 kHz. Rather, there remains an unidentified, probably gas-bearing scatterer that contributes significantly to observed scattering levels at this frequency. This study identifies and considerably narrows the list of candidates that are most likely to be responsible for enhanced 38 kHz scattering in the North Sea layer, and recommendations are made for potential future studies.

Five well-known Zr-based alloys of the systems Zr–Cu–Al–(Ni–Nb, Ni–Ti, Ag) (Cu 5 15.4–36 at.%) with the highest glass-forming ability were comparatively analyzed regarding their pitting corrosion resistance and repassivation ability in a chloride-containing solution. Potentiodynamic polarization measurements were conducted in the neutral 0.01 M Na₂SO₄ 1 0.1 M NaCl electrolyte and local corrosion damages were subsequently investigated with high resolution scanning electron microscopy (HR-SEM) coupled with energy dispersive x-ray spectroscopy (EDX). Both pitting and repassivation potential correlate with the Cu concentration, i.e., those potentials decrease with increasing Cu content. Pit morphology is not composition dependent: while initially hemispherical pits then develop an irregular shape and a porous rim. Corrosion products are rich in Cu, O, and often Cl species. A combination of low Cu and high Nb or Ti contents is most beneficial for a high pitting resistance of Zr-based bulk metallic glasses. The bulk glassy Zr₅₇Cu₁₅.₄Al₁₀Ni₁₂.₆Nb₅ (Vit 106) and Zr₅₂.₅Cu₁₇.₉Al₁₀Ni₁₄.₆Ti₅ (Vit 105) alloys exhibit the highest pitting resistance.

Abstract:
The gut microbiota play an important role in human health and nutrition. Despite this, the molecular mechanisms that underpin the ability of specific microorganisms to survive and flourish in the highly competitive environment of the large intestine are not well understood. The genome of Bacteroides thetaiotaomicron (Bt), a prominent member of our normal gut microbiota, contains a large number of genes that are predicted to encode products dedicated to polysaccharide sensing, acquisition and utilization. These genes are organised into co-regulated clusters, termed polysaccharide utilization loci (PULs). Microarray data from our collaborators revealed Bt activates a single PUL of 5 genes (Bt3086 to Bt3090) and an orphan gene (Bt458I) when grown on the a-I, 6-linked glucose-polymer, dextran. The dextran locus encodes two proteins predicted to be involved in polysaccharide binding and translocation (homologues of SusD and SusC, respectively), a hypothetical protein downstream of the susD homologue and glycoside hydrolases belonging to families GH66, GH3I and GH97. Here we have characterised all components of the dextran utilisation system to further our understanding of polysaccharide utilization by this prominent gut bacterium. Bt309I resides upstream of the locus and its putative topology is suggestive of a membrane bound one-component system (OCS), comprising of an N-terminal periplasmic domain, a transmembrane region and a C-terminal cytoplasmic domain. Here we reveal Bt309I to be the regulator of the dextran locus and demonstrate the first example of direct carbohydrate binding. to the periplasmic sensor domain (Bt309I pd) of this class of protein. In attempt to identify the activating ligand we characterised the putative glycoside hydrolases of the system which display endo-dextranase/transglycosidase (Bt3086), endo-dextranase (Bt3087) and a- glucosidase activity (Bt4581). We show that Bt3087 is likely surface located, whereas Bt3086 and Bt458I are likely periplasmic. Bt309Ipd binds to dextran degradation products of Bt3087 and Bt3086. Based on the respective activities of the GHs, we propose that the activating ligand is an a-I,6 linked oligosaccharide of 4 residues or more. The cytoplasmic domain of Bt309I binds directly to sequence specific regions of DNA upstream of the dextran PUL and the orphan gene; this binding interaction is mediated by residues residing on the third helix of the predicted helix-turn-helix domain. This is the first demonstration of direct DNA binding in a Bacteroides OCS. We also show that following signal perception the cytoplasmic domain remains tethered to the membrane and therefore postulate that the DNA is constitutively bound to Bt3091. Initial acquisition of extracellular polysaccharides occurs via proteins located on the cell surface, yet information on how these components contribute to polysaccharide utilization is sparse. Here we reveal Bt3089 - a SusD homolog - to be essential for growth on dextran oligosaccharides >2 glucoses and demonstrate direct glycan binding with a preference for dextran oligosaccharides with a degree of polymerization >6 glucoses. The susE- positioned gene product from the dextran locus, Bt3088, is essential for growth on dextran polysaccharide, but not oligosaccharides of <30 sugar residues; this targeted function is reflected in its high affinity for polymeric dextran. A model for dextran utilization in this prominent gut bacterium is proposed.

Abstract:
The human gut microbiota (HGM) represents a diverse community of bacteria with cell numbers in the trillions. The HGM is presented with dietary glycans, primarily from plant material consumed in the human diet, and host glycans which allow the bacterial community to proliferate in the gut. As such carbohydrate active enzymes (CAZymes) and multi-protein glycan utilisation systems are commonly expressed by members of the HGM. The human genome encodes few CAZymes rendering the vast majority of carbohydrates consumed accessible to the HGM. During bacterial fermentation of glycans short chain fatty acids (SCFAs), acetate, butyrate and propionate, are released into the gut lumen and are utilised for energy by colonocytes. SCFAs have also been shown to repress genes relating to proliferation in cancerous gut cells and activation of Treg cells. Bacteroidetes have been shown to possess a unique glycan utilisation strategy involving degradation of the target glycan at the cell surface before transport into the periplasm where degradation is completed before fermentable substrates are transported into the cytoplasm. Bacteroides ovatus deploys a xylan utilisation system expressed from two loci which is capable of targeting the relatively simple glucuronoxylans (GX) and arabinoxylans (AXs), but also the highly complex glucuronoarabinoxylans (GAXs) from corn. Work presented in this thesis has contributed to our understanding of xylan degradation. The data showed that xylan degradation begins at the cell surface with endo-acting xylanases from family GH10, targeting the backbone of simple xylans, while a GH98 xylanase targets specific structures in complex GAXs generating oligosaccharides that are transported into the periplasm. Co-culturing B. ovatus alongside Bifidobactertium adolescentis on GX and AX demonstrated cross-feeding of xylooligosaccharides, allowing Bi. adolescentis access to previously inaccessible substrate. This interaction provides further evidence for use of xylans and xylooligosaccharides as prebiotic supplements to the human diet with the potential to enrich for both butyrate-producing Bifidobacterium and propionate producing Bacteroides. Bacteroides thetaiotaomicron is regarded as a glycan generalist with upwards of 88 polysaccharide utilisation loci (PULs), the majority of which target different glycans in the gut. Among these target glycans are pectic polysaccharides, homogalacturonan, galactan, arabinan, rhamnogalacturonan I (RGI) and rhamnogalacturonan II (RGII). Galactan, which represents a relatively simple polysaccharide, required only two glycoside hydrolases for complete degradation, a surface galactanase (GH53) and a periplasmic galactosidase (GH2). Arabinan required two surface endo-arabinanases (GH43), a periplasmic α-1,2-arabinofuranosidase (GH43) and α-1,3-arabinosidase (GH51) for the removal of sidechains and a final periplasmic α-1,5-arabinosidase (GH51) to complete degradation of the arabinan backbone. Remaining galactooligosaccharide sidechains appended to rhamnose of RGI were degraded by three galactosidases (two GH2s and a GH35), each displaying different substrate preferences. During B. thetaiotaomicron pectin utilisation a degree of surface degradation was shown to be required prior to import; each of the resulting oligosaccharides can be scavenged by a B. thetaiotaomicron mutant lacking the specific surface enzymes required for polysaccharide degradation. These data demonstrate cross-feeding is prevalent during B. thetaiotaomicron utilisation of pectic polysaccharides in the HGM. Fructans are particularly well described prebiotics found to be a bifidogenic growth substrate. B. ovatus and B. thetaiotaomicron are capable of utilising inulin- and levan-type fructans, respectively. Co-culture of B. ovatus and Bi. adolescentis on inulin demonstrates that the Bifidobacterium sp out competes the Bacteroides sp, despite B. ovatus being capable of utilising long and short inulin chains. Cross-feeding also occurs when Bi. longum or Bacteroides vulgatus is the oligosaccharide recipient. B. thetaiotaomicron growth on levan is able to support growth of Bi. adolescentis indicating levan oligosaccharides have probiotic potential. Cross-feeding has the potential to have a dramatic influence on how probiotics are processed, utilised and distributed in the gut and their contribution to the HGM food web must be considered when designing dietary supplements.

Abstract:
Plant mortality following fire has been a concern of many people and agencies. There is little information, relating to the direct effect fire has on grass mortality, biomass production, and tillers at the community and individual levels. The objectives of this research were to determine the survival, tiller numbers, and biomass response to fuel load and direct measures of heat of four prominent graminoid species at the individual and community levels. The community level consisted of 24 plots measuring 20 X 20 meters. The plots were burned in autumn 2008 and 2009. Regression analyses were used to assess relationships of fuel load, degree-seconds, duration of heat, and maximum temperature, with relative biomass and frequency of species and species groups. The results illustrated fuel load, degree seconds, and duration were good predictors of total biomass. Threadleaf sedge increased in biomass to pre-treatment measures. Threadleaf sedge and needle-and-thread biomass was negatively related to fuel load, degree-seconds, and duration of heat. Neither blue grama nor western wheatgrass changed in biomass. A burn cage was used to apply the burn treatments for the individual plant study using range of fuel loads 500-9000 kg•ha-1. Relationships of fuel load with degree-seconds, heat duration, and maximum temperature were tested independently. Probabilities of plant mortality were estimated based on fuel load, duration of heat, maximum temperature, and degree-seconds. Of 120 plants of each species, only one western wheatgrass and one threadleaf sedge plant died following fire. Mortality occurred for 20 blue grama plants and 17 needle-and-thread plants. Degree-seconds, duration, maximum temperature, and fuel load were good predictors of mortality for blue grama and needle-and-thread. Neither duration of heat or maximum temperature explained changes in biomass or tillers for any species. Plant response was less in the field than the burn cage even at heavier fuel loads. This study found that as fuel load increased, degree-seconds, duration, and maximum temperature increased. Fuel load was the primary factor increasing degree-second, duration, and maximum temperature and provided an alternative way to predict plant mortality following fire. Understanding direct fire effects on plants will provide better management decisions following a fire.

ENGLISH ABSTRACT: Chromosome arm 7DL of common wheat carries genes for agronomically important traits such as leaf rust, stem rust, Russian wheat aphid and eye spot resistance. Some of these genes occur on introgressed foreign chromatin, which restricts their utility in breeding. The 7DL genetic maps are poorly resolved, which seriously hampers attempts to manipulate the genes and introgressed regions in breeding. This dissertation represents an attempt to improve our knowledge of the relative map positions of three resistance genes that have significant potential for use in local breeding programmes. The leaf rust resistance gene, Lr19, is located on a Thinopyrum ponticum-derived translocation which occupies a large part of the terminal end of 7DL. The translocation also carries genes for less favourable traits such as yellow flour colour. Attempts have been made to reduce the size of the translocation through allosyndetic pairing induction; the primary aims being to remove deleterious genes and to minimise the amount of foreign chromatin associated with Lr19 so it can be recombined with other useful 7DL genes. Twenty-nine 'Indis'-derived Lr 19 deletion mutants were previously produced by gamma irradiation and a physical map was constructed. In this study, the set of mutant lines were further analysed using 144 Sse8387I/Msei and 32 EcoRI/Msel amplified fragment length polymorphism (AFLP) primer combinations. The previous physical map, which was based on five restriction fragment length polymorphism (RFLP) markers and five structural gene loci, was extended and now includes 95 novel AFLP markers (86 Sse8387I/Msei and 9EcoRI!Msel markers), of which seven map close to Lr 19. Most of the deletions could be ordered according to size and the improved map has already been used to characterise shortened recombinant forms of the Lr 19 translocation. An unsuccessful attempt was made to convert one of the seven markers closest to Lr 19 into a sequence-specific marker. However, an AFLP marker located distally from Lr 19 was successfully converted into a sequence-specific marker in collaboration with other researchers. An attempt was also made to map and tag the Russian wheat aphid (RWA) resistance gene, Dn5. A doubled haploid mapping population consisting of 94 lines was created and typed for Dn5, four microsatellite loci and the endopeptidase locus, Ep-Dl. The Dn5 locus mapped 25.4 cM and 28.6 cM distally from Xg.vm111 and Xg.vm437, respectively, but was not linked to Xgwm428, Xgwm3 7 or Ep-Dl. Tagging of Dn5 was attempted by screening twelve homozygous resistant and seven homozygous susceptible F2 lines from a cross between 'Chinese Spring' and 'PI 294994' with 70 Sse8387IIi\1sei AFLP primer combinations. Only two potentially useful polymorphisms (one in coupling and one in repulsion phase) were identified. Conversion of the coupling phase marker to a sequence-specific marker was not successful. The eyespot resistance gene, Pchl , was derived from Triticum ventricosum and is present in the wheat VPM-1. Close association between Pchl and the endopeptidase Ep-Dlb allele has been reported previously. Pchl/Ep-Dl was tagged by screening ten wheat genotypes (each homozygous for the confirmed presence or absence of Pchl and/or Ep-Dl b) with 36 Sse83 87I/ Msei AFLP primer combinations. Three AFLP markers were closely associated with Pchl I Ep-D 1, one of which was targeted for conversion into a sequence-specific marker. The sequence-specific marker contained a microsatellite core motif and was found to be useful for tagging Pchl!Ep-Dl. A genetic distance of 2 cM was calculated between the novel microsatellite marker and Ep-Dl. The microsatellite marker was also polymorphic for the Lr 19 translocation and it was possible to map it between the Wsp-Dl and Sr25 loci. In this dissertation, mapping and/or tagging of three important resistance genes were achieved. Due to the fact that all markers used in these studies were not polymorphic between all of the targeted regions, it was not possible to fully integrate the data obtained for the three regions.

Hierdie proefskrif verteenwoordig 'n paging om kennis van die relatiewe kaart liggings van drie weerstandsgene, met betekenisvolle potensiaal in plaaslike tee! programme, te verbreed. The November, 2007, street vendor riots in Dakar, Senegal, were one of the country's most powerful expressions of political and economic disintegration in recent times. Almost equally striking was the absence from these events of Enda Tiers Monde, Senegal's oldest, largest, and most influential indigenous NGO and the champion of the "popular economy." This thesis uses the vendor riots as a window into the response of an organization, Enda, to changing institutional contexts (i.e. different political regimes) and emerging development challenges (i.e. informal street vendors and urban spatial access). In its thirty years of operation, Enda has made significant contributions to urban development in Dakar and in cities throughout the global south. These contributions include extending access to basic services and improving housing in slums, organizing and advocating for the urban poor, and training local leaders in participatory governance. However, as demonstrated by its lack of involvement and influence in events surrounding the vendor riots, Enda's role and relationship with both grassroots actors and national policy-makers has changed. This thesis demonstrates that different types of relationships between Enda and the state affect the organization's relationship with its base, and therefore Enda's legitimacy with and connection to both actors. I also argue that these changing institutional contexts have affected Enda internally.

Since an important political turnover in the year 2000 and the death of Enda's founder in 2002, Enda has been attempting to align its internal structure and functioning with its external institutional context. The challenge for Enda is to develop a system that will grant its teams the flexibility to experiment while supporting them with evaluation for learning and adaptation. This is essential for Enda's renewed visibility and influence in urban development in Dakar.

Abstract:
This study explored the experiences of prominent jazz vocalists regarding the collaboration between the vocalist and pianist during duo performances. The work relationship between a jazz vocalist and accompanist is a unique phenomenon due to the improvisatory and collaborative nature of jazz. Effective co-performer interaction and communication are two essential components within an improvisational music setting. A spirit of spontaneity ir required in order for improvisation and interpretation to take place in real-time and on stage; aspects which cannot be duplicated or replicated during rehearsals. A qualitative research approach was employed by means of a collective case study, exploring the views and experiences of three prominent South African and three international jazz vocalists. Data was gathered through semi-structured interviews. Furthermore, artefacts including audio and audio-visual recordings of each of these performers were studied to ascertain aspects of the vocal-piano collaboration. Thematic data analysis presented four main themes: (1) individuality; (2) interactive relationships; (3) musicianship; and (4) the present moment. The findings revealed that the roles of both partners within the vocal-piano duo collaboration are evenly distributed, yet have distinctive functions. Each new performance between the same vocal-piano duo is an innovative execution, a skilful and artistic equilibrium. It embodies a companionship playing out on stage in view of an audience, yet with the subtle sharing of musical messages.

The  European  health  care  is  facing  challenges  with  an  increasing  ageing  population, with a higher frequency of chronic diseases, which have resulted in rising health care costs. Meanwhile, the trend shows how patients and citizens are becoming more active in their personal health care, with the number of existing doctors and nurses subsiding furthermore entailing problems. The area of eHealth, which involves information and communication technologies with health care, is hence seen as a partial long-term solution and is considered being a rapidly growing market both in Sweden, but also in Europe. eHealth services further consider to promote increased access, mobility and interoperability in the health care, but the lack of wholehearted commitment, financial support and complex EHR-systems in Sweden's municipalities and county councils might partially impeding down the development. The purpose of this report is therefore targeting to explore, identify and analyze prominent aspects for the continued development of the Swedish health care and eHealth services. The study also examines what subsequent benefits an implementation of an eHealth service entails, which also has been related to the identified prominent aspects. This master thesis is based on  a thorough literature review extracted from a theoretical framework   including   an   interoperability-,  security-,   mobility-   and   business-modeling perspective, which are used as a foundation for the building of a set of hypotheses, which are subsequently verified with the aid of gathered empirics. The empirics are obtained from 10 semi-structured qualitative interviews, as well as two case studies, which together resulted in key-findings and conclusions. Firstly: in relation to the interoperability perspective, it became clear how there should exist both   a   technical   and   social   interoperability   that   communicate   with   each   other. The  EHR-systems  of  today  are  often  considered  difficult  to  learn,  non-intuitive  and lacking interfaces that are user-friendly designed for the end-user. Increased interoperability was also seen as enabling and simplifying the access to the patient’s medical history, which the EHR-system TakeCare evidently demonstrated. Furthermore, it was acknowledged how there is no correlation between the increased time spent by health care professionals with administrative tasks and documentation with an increased interoperability. It also emerged that patients and the dominant part of the population had either no or very limited knowledge regarding the underlying security and overall management of personal health information in health care. Patients instead often blindly trust the Swedish health care system being secure, and prioritizing other things during medical appointments. The knowledge of security issues in the health care is predicted to increase among patients if they in the future would obtain full access to their own medical records. There is also a general opinion among health care professionals and related instances how new security risks will arise alongside the eHealth wave,  with  a  particular  concern  for  the  increasing  involvement  of  mobile  devices. Relationships between an increased interoperability also seem to favor increased mobility in health care, but security aspects often prevent the mobility development. Finally, it was unanimously espoused how non-financial values must not be ignored, where the on-going debate argues whether what real impact these non-financial values have, where inter alia strict budgets and large gaps between the decision-makers and end-users appeared as issues. Similar arguments were encountered regarding the actual impact of the opinions of patients in relation to business modeling, where a tripartite-problem and the patients’ limited access to their medical records was partly seen as a primary issue. Secondly:  the  case  studies  demonstrated  how  a  transition  to  the  EHR-system  TakeCare generally did result in cost- and resource savings in terms of local servers, IT-maintenance and inventory management. The TakeCare implementation also led to an increased visibility among health care centers by enabling and simplifying the access to patient medical history. Increased communication, awareness, and more effective internal processes due to integrated modules and direct connections to ePrescriptions could also be accessed from the TakeCare transition. Finally, it emerged that relations existed between simplified access to the patient’s medical   history   and   how   it   subsequently   resulted   in   an   increased   interoperability. A correlation was also seen as the health care become generally more mobile due to increased interoperability.
Hälso- och sjukvården i Europa står inför utmaningar i och med en stigande åldersgrupp med en större andel kroniska sjukdomar, vilket resulterat i stegrande sjukvårdskostnader. Samtidigt ses en trend i hur patienter och medborgare börjar bli mer aktiva i sin egen vård och efterfrågan på sjukvårdspersonal ökar, med ökande utmaningar som följd. eHälsa-området, vilket involverar informations- och kommunikationsteknik inom sjukvården, ses därför som en potentiell långsiktig del-lösning och anses samtidigt vara ett starkt växande område i Sverige, men också på den övriga europeiska marknaden. eHälsa betraktas vidare främja en ökad åtkomst, mobilitet och interoperabilitet inom sjukvården, men bristen på helhjärtat engagemang, finansiellt stöd och det stora antal komplexa journalsystem i Sveriges olika kommuner och landsting ses delvis ligga till grund för en bromsad utveckling. Syftet med denna rapport är därför att undersöka, kartlägga och analysera de mest centrala aspekterna för den fortsatta utvecklingen av svensk sjukvård och eHälso-tjänster. Studien undersöker även vad implement av en eHälso-tjänst praktiskt har medfört, med ett fokus på journalsystem där de enskilda förändringarna även har relaterats till de identifierade centrala aspekterna. Examensarbetet är baserat på en gedigen litteraturstudie som utifrån ett teoretiskt ramverk inkluderande ett interoperabilitets-, säkerhets-, mobilitets- och affärsmoduleringsperspektiv ligger till grund för framtagandet av hypoteser som sedan verifierats med hjälp av empiriskt insamlad information. Empirin är erhållen från tio semi-strukturerande kvalitativa intervjuer, samt två fallstudier, vilka tillsammans har resulterat i ett flertal slutsatser. Utifrån ett interoperabilitetsperspektiv framgick det hur det bör finnas både en teknisk och social interoperabilitet som kommunicerar med varandra, då journalsystem idag anses vara svåra att lära sig, icke intuitiva och ej användarvänligt utformade för slutanvändaren. Ökad interoperabilitet ses även möjliggöra och förenkla åtkomsten av patienthistorik, vilket journalsystemet TakeCare tydligt påvisat. Vidare kunde det konstateras att det inte finns en korrelation mellan den progressivt ökande avsatta tiden som sjukvårdspersonal idag  illbringar med administrativa uppgifter och dokumentation med en förhöjd interoperabilitet. Det framkom även att patienter har väldigt liten eller obefintlig kunskap rörande den underliggande säkerheten och hanteringen av personlig information i sjukvården, då de ofta blint litar på att svensk sjukvård anses vara säker samt att patienter prioriterar annat vid läkarbesök. Kunskaper om säkerheten i sjukvården bland patienterna anses dock öka ifall de  framtiden får tillgång till sin journal. Det finns även en allmän oro bland sjukvårdspersonal och närbesläktade instanser för att nya säkerhetsrisker kommer att uppstå i och med eHälsa-vågen, med ett särskilt orosmoln för den ökande användningen av mobila enheter. Relationer  mellan  hur  ökad  interoperabilitet  även  gynnar  förhöjd  mobilitet  sågs  även  förekomma,  men  att  det  ofta  samtidigt  är  säkerhetsaspekter  som  hindrar  den  mobila utvecklingen.  Avslutningsvis  förespråkades  det  hur  icke-finansiella  värden  inte  får  bli ignorerade, men problemet kring hur verkningsfull dess faktiska påverkan är, relateras bland annat  till  strikta  budgetar  samt  stora  avstånd  mellan  beslutstagare  och  slutanvändare. Liknande argument påträffades angående den faktiska inverkan av åsikter från patienter vid affärsmodulering,  där  ett  trepartsproblem  och  patienternas  begränsade  åtkomst  till  sina journaler delvis sågs ligga till grund. Fallstudierna påvisade hur övergången till journalsystemet TakeCare generellt har lett till resursbesparingar i form av lokala servrars underhåll och lageranvändning, samt en ökad synlighet  i   vården   med   förbättrad  tillgång   till   patienthistorik  jämfört   med   tidigare journalsystem. En ökad kommunikation och medvetenhet samt effektivare interna processer på  grund  av  integrerade  moduler  och  direktkoppling  till  eRecept  kunde  även  påvisas. Slutligen framgick det att relationer förekom mellan den ökande åtkomsten av patienthistorik och andra journaler, och hur förhöjd interoperabilitet medfört detta. Abstract:
Numerous studies have attempted to measure scientists' influence by measuring the quantity of citations to their works. The problem with "citation counting," as it is called, is that it assumes that each listing of an author in a citation index is equal to another without bothering to explore the substantive uses of citations in the source article. The present study attempts to alleviate this problem by content analysis of citations in a limited sphere: reference to major philosophers of science by sociologists. In just over 100 sociology journals, citations to Thomas Kuhn, Karl Popper, Ernst Nagel, and Carl Hempel (overall, the most frequently cited philosophers of science) from 1971-1982 were randomly sampled. Each citation was classified according to the following criteria: 1) philosopher cited; 2) work cited, 3) exclusivity (whether cited with others); 4) multiplicity (number of citations by the philosopher in the same article); 5) type of article; and 6) purpose of citation. Purposes of citation included seven categories: 1) listing as relevant literature; 2) definition of a concept; 3) modification or extension of a philosopher's theory; 4) formulation of a research problem; 5) interpretation of results; 6) critical of philosopher's work; and 7) other. Analysis of these data revealed the following conclusions: 1) the major use of philosophy was the furnishing of concepts and their definitions; 2) philosophy of science played little or no role in directing research or interpreting results; 3) the use of citations differed greatly among the philosophers; 4) simple citation counting would have severely distorted the relative influences of each philosopher; and 5) the dialogue between sociology and the philosophy of science has, in the last decade, been dominated by Kuhn's theory of scientific revolutions.

Millions of people in South Africa eat out every day, utilising the food service sector. Although the lack of an effective reporting system makes it difficult to know how many of these people suffer from food-borne illness, statistics from the developed countries show that this number may be significant. There is, therefore, the need to ensure that the food service sector, which encompasses fast food outlets, hotels and similar accommodation outlets offering food and beverage services, restaurants, caterers, etc., implement effective food safety management systems. Internationally, the trend has been that food safety management systems should be based on the internationally accepted Hazard Analysis Critical Control Point (HACCP) principles. In South Africa, the implementation of HACCP as a food safety management system has been driven by international trade requirements where foods are exported to countries such as the European Union or the United States of America. A national regulation requiring HACCP implementation was promulgated in 2003, but compliance is not yet required for the food service sector. Currently, neither of the above mentioned factors put adequate pressure on the food service sector to implement formal food safety management systems. However, increasing international tourism and the hosting of international sporting events has brought this sector under scrutiny. Food handlers have been implicated in many outbreaks of food-borne illness and much research has been done to investigate causal factors in this regard. Food handler training has been proposed as a strategy to improve food safety practices. However, research has shown that the traditional provision of food safety and food hygiene knowledge does not equate to improved food safety behaviours. Some authors postulate that the organisational context, created largely by the management of an organisation, is of greater significance than training. Less research is available on these management factors – defined as the situational factors when discussing organisational culture, or defined as enabling and reinforcing factors when discussing food handler behaviour. This study commenced with the hypothesis that food handlers are not able to implement the correct food safety behaviours in the absence of sufficient management support. This support would require appropriate policies regarding food safety, the provision of training and infrastructure and enforcing the correct behaviours by line management, as a minimum. The aim of this study was to investigate and assess the role of line management in relation to food safety at a prominent South African entertainment facility. In order to achieve this, the following objectives were defined for the study: to conduct a qualitative assessment of the role of management in food safety, to assess the role of management in the provision of food safety training and to assess the role of management in the provision of a basic hygiene infrastructure at the study site in order to allow food handlers to carry out the correct behaviours. The objective of conducting a qualitative study of management practices, policies and resource provision with respect to food safety revealed that there was no formal evidence of management commitment to food safety other than the recent provision of food handler training. The findings also indicated a lack of a formal management system for food safety at the study site. In the exploratory survey of food safety training and knowledge, results showed that only 60 % of staff in the survey had received training. This indicates that at the time of the survey, the study site did not fully comply with the minimum legal requirements for food handler training. The results of the employee survey further indicated that employees were aware of the importance of hand washing although it was not possible to determine whether this knowledge was as a result of the training intervention or prior knowledge. Many of the supervisors were not yet trained in food safety and the impact of the food safety training intervention on related behaviours at the site will require further in-depth assessment. Upon investigating the food hygiene infrastructure provided at the study site to allow food handlers to carry out the correct behaviours, findings indicated that although the personnel hygiene programme addressed most of best practice requirements in design, the implementation of the hand washing requirements was not aligned with accepted norms due to the lack of sufficient hand wash basins. The provision of facilities such as sufficient and conveniently located hand wash basins is a management function and findings suggest that, as a priority, management should ensure that they are not contributing to the lack of implementation of the correct food safety behaviours of food handlers as a result of failing to provide the necessary resources. The results of this study should be of value in the food service sector, specifically hotel kitchens, as a guideline to ensure that management plays an effective role in facilitating food safety management systems. A robust food safety and food hygiene training programme for all levels of the organisation is essential in ensuring adequate knowledge of food safety hazards and correct practices. Training should be supported by daily supervision of food safety controls, management commitment and a work environment that supports the implementation of the correct behaviours. Literature has shown that undesirable practices are often deeply rooted in kitchen culture. It has further been commented that culture changes require a top-down approach which usually involves working with the leadership of the organisation. Important policies and procedures generally originate from the management tiers and will always require the concurrence of management in providing resources, altering priorities or otherwise changing how things are done in the organisation. The results of this study are invaluable in highlighting areas in an organisation that could be targeted to change the kitchen culture. Such changes are primarily the responsibility of management. Ultimately, this study endeavours to contribute to the body of knowledge pointing to the role of social-behavioural aspects in ensuring food safety and thereby consumer well-being.

The European health care is facing challenges with an increasing ageing population, with a higher frequency of chronic diseases, which have resulted in rising health care costs. Meanwhile, the trend shows how patients and citizens are becoming more active in their personal health care, with the number of existing doctors and nurses subsiding furthermore entailing problems. The area of eHealth, which involves information and communication technologies with health care, is hence seen as a partial long-term solution and is considered being a rapidly growing market both in Sweden, but also in Europe. eHealth services further consider to promote increased access, mobility and interoperability in the health care, but the lack of wholehearted commitment, financial support and complex EHR-systems in Sweden's municipalities and county councils might partially impeding down the development. The purpose of this report is therefore targeting to explore, identify and analyze prominent aspects for the continued development of the Swedish health care and eHealth services. The study also examines what subsequent benefits an implementation of an eHealth service entails, which also has been related to the identified prominent aspects. This master thesis is based on a thorough literature review extracted from a theoretical framework including an interoperability-, security-, mobility- and business-modeling perspective, which are used as a foundation for the building of a set of hypotheses, which are subsequently verified with the aid of gathered empirics. The empirics are obtained from 10 semi-structured qualitative interviews, as well as two case studies, which together resulted in key-findings and conclusions. Firstly, in relation to the interoperability perspective, it became clear how there should exist both a technical and social interoperability that communicate with each other. The EHR-systems of today are often considered difficult to learn, non-intuitive and lacking interfaces that are user-friendly designed for the end-user. Increased interoperability was also seen as enabling and simplifying the access to the patient’s medical history, which the EHR-system TakeCare evidently demonstrated. Furthermore, it was acknowledged how there is no correlation between the increased time spent by health care professionals with administrative tasks and documentation with an increased interoperability. It also emerged that patients and the dominant part of the population had either no or very limited knowledge regarding the underlying security and overall management of personal health information in health care. Patients instead often blindly trust the Swedish health care system being secure, and prioritizing other things during medical appointments. The knowledge of security issues in the health care is predicted to increase among patients if they in the future would obtain full access to their own medical records. There is also a general opinion among health care professionals and related instances how new security risks will arise alongside the eHealth wave, with a particular concern for the increasing involvement of mobile devices. Relationships between an increased interoperability also seem to favor increased mobility in health care, but security aspects often prevent the mobility development. Finally, it was unanimously espoused how non-financial values must not be ignored, where the on-going debate argues whether what real impact these non-financial values have, where inter alia strict budgets and large gaps between the decision-makers and end-users appeared as issues. Similar arguments were encountered regarding the actual impact of the opinions of patients in relation to business modeling, where a tripartite-problem and the patients’ limited access to their medical records was partly seen as a primary issue. Secondly, the case studies demonstrated how a transition to the EHR-system TakeCare generally did result in cost- and resource savings in terms of local servers, IT-maintenance and inventory management. The TakeCare implementation also led to an increased visibility among health care centers by enabling and simplifying the access to patient medical history. Increased communication, awareness, and more effective internal processes due to integrated modules and direct connections to ePrescriptions could also be accessed from the TakeCare transition. Finally, it emerged that relations existed between simplified access to the patient’s medical history and how it subsequently resulted in an increased interoperability. A correlation was also seen as the health care become generally more mobile due to increased interoperability.
Hälso- och sjukvården i Europa står inför utmaningar i och med en stigande åldersgrupp med en större andel kroniska sjukdomar, vilket resulterat i stegrande sjukvårdskostnader. Samtidigt ses en trend i hur patienter och medborgare börjar bli mer aktiva i sin egen vård och efterfrågan på sjukvårdspersonal ökar, med ökande utmaningar som följd. eHälsa-området, vilket involverar informations- och kommunikationsteknik inom sjukvården, ses därför som en potentiell långsiktig del-lösning och anses samtidigt vara ett starkt växande område i Sverige, men också på den övriga europeiska marknaden. eHälsa betraktas vidare främja en ökad åtkomst, mobilitet och interoperabilitet inom sjukvården, men bristen på helhjärtat engagemang, finansiellt stöd och det stora antal komplexa journalsystem i Sveriges olika kommuner och landsting ses delvis ligga till grund för en bromsad utveckling. Syftet med denna rapport är därför att undersöka, kartlägga och analysera de mest centrala aspekterna för den fortsatta utvecklingen av svensk sjukvård och eHälso-tjänster. Studien undersöker även vad implement av en eHälso-tjänst praktiskt har medfört, med ett fokus på journalsystem där de enskilda förändringarna även har relaterats till de identifierade centrala aspekterna. Examensarbetet är baserat på en gedigen litteraturstudie som utifrån ett teoretiskt ramverk inkluderande ett interoperabilitets-, säkerhets-, mobilitets- och affärsmoduleringsperspektiv ligger till grund för framtagandet av hypoteser som sedan verifierats med hjälp av empiriskt insamlad information. Empirin är erhållen från tio semi-strukturerande kvalitativa intervjuer, samt två fallstudier, vilka tillsammans har resulterat i ett flertal slutsatser. Utifrån ett interoperabilitetsperspektiv framgick det hur det bör finnas både en teknisk och social interoperabilitet som kommunicerar med varandra, då journalsystem idag anses vara svåra att lära sig, icke intuitiva och ej användarvänligt utformade för slutanvändaren. Ökad interoperabilitet ses även möjliggöra och förenkla åtkomsten av patienthistorik, vilket journalsystemet TakeCare tydligt påvisat. Vidare kunde det konstateras att det inte finns en korrelation mellan den progressivt ökande avsatta tiden som sjukvårdspersonal idag tillbringar med administrativa uppgifter och dokumentation med en förhöjd interoperabilitet. Det framkom även att patienter har väldigt liten eller obefintlig kunskap rörande den underliggande säkerheten och hanteringen av personlig information i sjukvården, då de ofta blint litar på att svensk sjukvård anses vara säker samt att patienter prioriterar annat vid läkarbesök. Kunskaper om säkerheten i sjukvården bland patienterna anses dock öka ifall de i framtiden får tillgång till sin journal. Det finns även en allmän oro bland sjukvårdspersonal och närbesläktade instanser för att nya säkerhetsrisker kommer att uppstå i och med eHälsa-vågen, med ett särskilt orosmoln för den ökande användningen av mobila enheter. Relationer mellan hur ökad interoperabilitet även gynnar förhöjd mobilitet sågs även förekomma, men att det ofta samtidigt är säkerhetsaspekter som hindrar den mobila utvecklingen. Avslutningsvis förespråkades det hur icke-finansiella värden inte får bli ignorerade, men problemet kring hur verkningsfull dess faktiska påverkan är, relateras bland annat till strikta budgetar samt stora avstånd mellan beslutstagare och slutanvändare. Liknande argument påträffades angående den faktiska inverkan av åsikter från patienter vid affärsmodulering, där ett trepartsproblem och patienternas begränsade åtkomst till sina journaler delvis sågs ligga till grund. Fallstudierna påvisade hur övergången till journalsystemet TakeCare generellt har lett till resursbesparingar i form av lokala servrars underhåll och lageranvändning, samt en ökad synlighet i vården med förbättrad tillgång till patienthistorik jämfört med tidigare journalsystem. A low power plasma thruster, initially dubbed the Diverging Cusped-Field Hall Thruster, was built at MIT in 2007 with the aim of providing an alternative to comparably sized conventional Hall thrusters able to provide enhanced lifetimes and thus deliver higher total impulses. The initial design was constrained in some respects by availability and inexpensiveness of materials rather than on experience based optimization due in part to the dearth of literature available for similar devices. While experiments suggest that the prototype Diverging Cusped-Field Thruster performs as well as mature Hall thruster designs, they have also revealed several operational quirks which could potentially pose problems for spacecraft integration. Dominant among these peculiarities are the existence of at least two distinct operational modes, marked by a steep jump in anode current and current oscillation amplitude, and, somewhat separately, the coalescence of the ion plume into a hollow cone-like structure with peak fluxes around 30 degrees from the thruster axis. This thesis describes the experimental and numerical work undertaken in an attempt to uncover the physical underpinnings of these two prominent cusped-field thruster phenomena. Stratification of the plasma plume into a wide-angle conical shell represents both a loss in thrust efficiency and a potential hazard for beam impingement on spacecraft surfaces. A method was envisaged to decrease the divergence of the beam using an external solenoid to re-contour the magnetic field near the thruster exit, where measurements suggest the critical ion acceleration occurs. Implementation of this method demonstrated clear vectoring of a large portion of the ion beam towards smaller angles in line with hypothesized effects, however a distinct, low-density, outer shell remained at the initial angle of divergence. Separately, the observed bifurcation in operational mode at normal conditions represents both a physically interesting display of plasma dynamics in the thruster discharge chamber and a potentially highly problematic feature, as operational envelopes in a particular mode have been observed to change unpredictably with experimental conditions. Attempts are made to experimentally isolate the factors which lead to bifurcation. The steady-state characteristics of the two modes are investigated using infrared thermography to monitor chamber temperatures while the unsteady behavior is monitored with an AC current probe on the discharge power supply. A thermal model of the thruster, using commercial finite-element-analysis software, is developed to validate plasma power deposition estimates from the thermography experiments. A prominent feature of the bifurcation is observed in the anode current spectrum, which changes from a quiescent mode to one exhibiting a strong relaxation oscillation at a fundamental frequency of several kilohertz. The nature of the oscillatory mode, and its transition to a highly damped state, is investigated through parametric scans of the thruster operating conditions. The main oscillation appears, from experiment, to be a breathing mode similar to those observed in Hall thrusters. An unsteady 1-D fluid code is adapted from the Hall thruster literature to model the interaction of neutral gas flow with the magnetically-limited electron current believed to be the main source of the instability. Trends in stability of the breathing mode with both thruster and model parameters, such as propellant flow rate, anode voltage, magnetic field, electron mobility and discharge chamber divergence are examined. A turbulent shear model is introduced to capture the effects of a time-evolving cross-field electron mobility on mode transitions and is shown to reproduce the experimentally observed oscillation damping at high voltages if the eddy turnover times are allowed to increase by modest intervals. The shear model is also able to naturally reproduce a feature of the anode current waveform, where a series of possible ionization avalanche events are muted before eventually leading to a large burst in current, which has not been captured by prior models.

Durant les situations de crise, telles que les catastrophes, le besoin de recherche d’informations (RI) pertinentes partagées dans les microblogs en temps réel est inévitable. Cependant, le grand volume et la variété des flux d’informations partagées en temps réel dans de telles situations compliquent cette tâche. Contrairement aux approches existantes de RI basées sur l’analyse du contenu, nous proposons de nous attaquer à ce problème en nous basant sur les approches centrées utilisateurs tout en levant un certain nombre de verrous méthodologiques et technologiques inhérents : 1) à la collection des données partagées par les utilisateurs à évaluer, 2) à la modélisation de leurs comportements, 3) à l’analyse des comportements, et 4) à la prédiction et le suivi des utilisateurs primordiaux en temps réel. Dans ce contexte, nous détaillons les approches proposées dans cette thèse afin de prédire les utilisateurs primordiaux qui sont susceptibles de partager les informations pertinentes et exclusives ciblées et de permettre aux intervenants d’urgence d’accéder aux informations requises quel que soit le format (i.e. texte, image, vidéo, lien hypertexte) et en temps réel. Ces approches sont centrées sur trois principaux aspects. Nous avons tout d’abord étudié l’efficacité de différentes catégories de mesures issues de la littérature et proposées dans cette thèse pour représenter le comportement des utilisateurs. En nous basant sur les mesures pertinentes résultant de cette étude, nous concevons des nouvelles caractéristiques permettant de mettre en évidence la qualité des informations partagées par les utilisateurs selon leurs comportements. Le deuxième aspect consiste à proposer une approche de modélisation du comportement de chaque utilisateur en nous basant sur les critères suivants : 1) la modélisation des utilisateurs selon l’évolution de l’évènement, 2) la modélisation de l’évolution des activités des utilisateurs au fil du temps à travers une représentation sensible au temps, 3) la sélection des caractéristiques les plus discriminantes pour chaque phase de l’évènement. En se basant sur cette approche de modélisation, nous entraînons différents modèles de prédiction qui apprennent à différencier les comportements des utilisateurs primordiaux de ceux qui ne le sont pas durant les situations de crise. Les algorithmes SVM et MOG-HMMs ont été utilisés durant la phase d’apprentissage. La pertinence et l’efficacité des modèles de prédiction appris ont été validées à l’aide des données collectées par notre système multi-agents MASIR durant deux inondations qui ont eu lieu en France et des vérités terrain appropriées à ces collections
During crisis events such as disasters, the need of real-time information retrieval (IR) from microblogs remains inevitable. However, the huge amount and the variety of the shared information in real time during such events over-complicate this task. Unlike existing IR approaches based on content analysis, we propose to tackle this problem by using user-centricIR approaches with solving the wide spectrum of methodological and technological barriers inherent to : 1) the collection of the evaluated users data, 2) the modeling of user behavior, 3) the analysis of user behavior, and 4) the prediction and tracking of prominent users in real time. In this context, we detail the different proposed approaches in this dissertation leading to the prediction of prominent users who are susceptible to share the targeted relevant and exclusive information on one hand and enabling emergency responders to have a real-time access to the required information in all formats (i.e. text, image, video, links) on the other hand. These approaches focus on three key aspects of prominent users identification. Firstly, we have studied the efficiency of state-of-the-art and new proposed raw features for characterizing user behavior during crisis events. Based on the selected features, we have designed several engineered features qualifying user activities by considering both their on-topic and off-topic shared information. Secondly, we have proposed a phase-aware user modeling approach taking into account the user behavior change according to the event evolution over time. This user modeling approach comprises the following new novel aspects (1) Modeling microblog users behavior evolution by considering the different event phases (2) Characterizing users activity over time through a temporal sequence representation (3) Time-series-based selection of the most discriminative features characterizing users at each event phase. Thirdly, based on this proposed user modeling approach, we train various prediction models to learn to differentiate between prominent and non-prominent users behavior during crisis event. The learning task has been performed using SVM and MoG-HMMs supervised machine learning algorithms. The efficiency and efficacy of these prediction models have been validated thanks to the data collections extracted by our multi-agents system MASIR during two flooding events who have occured in France and the different ground-truths related to these collections

Images as important sources of information, powered by the newest robotic microscopy technologies, make the volume of data available larger than ever before. From the images, hundreds of variables can be calculated. The medical research group within the HASTE project conducts a LNP (Lipid Nano-particle) experiment that is designed to transport a drug to target cells. In the LNP experiment, microscopy images are taken over time to record if the drug is uptaken. We propose that the non-parametric change point analysis can be used to identify the variable which shows the earliest state change (potentially signifying the drug uptake) among all variables calculated from the images. Two algorithms for non-parametric change point analysis, an agglomerative and a divisive, are studied through simulation leading us to implement the agglomerative algorithm on the LNP experiment data. Objectives: To assess the effects of orthodontic treatment for Class III incisors in children and adolescents. Design: A Cochrane systematic review. Method: The following databases were searched up to 7th January 2013: Cochrane Oral Health Group Trials Register, Cochrane Central Register of Controlled Trials (CENTRAL), MEDLINE via OVID, EMBASE via OVID. Selection criteria: All randomised controlled trials of orthodontic treatments to correct Class III incisors. Trials were eligible for inclusion in the review if they recruited children and/or adolescents (aged 16 or less) receiving orthodontic treatment to correct Class III incisors. Trials including patients with a cleft lip and/or palate or other cranio-facial deformity/syndrome were excluded as were trials that had recruited less than 80% children or adolescents or patients who had previously received surgical orthognathic treatment. Active interventions included: orthodontic braces, chin cups, facemasks, reverse headgear, bone-anchored appliances or any other intra or extra-oral appliance aiming to correct Class III incisors. Controls included: No treatment, delayed treatment, other active intervention. Types of Outcome Measures - Primary: Prominence of the lower front teeth (measured in mm or by any index of malocclusion). Secondary: Relationship between upper and lower jaw; psychosocial measures; patient satisfaction; jaw joint problems. Adverse effects: Health of the gums; damage to the teeth e.g. tooth decay. Outcomes were recorded at all ages reported. The results were reported according to the most common endpoints. Adverse effects were recorded and the results reported in descriptive terms. Data collection and analysis: The titles and abstracts of the search results were examined to exclude obviously irrelevant reports. Full text reports of potentially eligible studies were examined for compliance with the eligibility criteria. Screening of references, data extraction and assessment of the risk of bias of included studies, was performed independently and in duplicate by two review authors. The mean differences, with 95% confidence intervals, were calculated for continuous data. Meta-analysis was only used when studies of similar comparisons were reporting comparable outcome measures. A fixed-effect model was used. I2 statistics were used as measures of statistical heterogeneity. Results: Seven randomised controlled trials were included in this review. Of these, four reported on the use of a facemask, two on the chin cup, one on the tandem traction bow appliance and one on mandibular headgear. One study reported on both the chin cup and mandibular headgear appliances. Three trials (n=155) reported ANB differences immediately after treatment with a facemask when compared to an untreated control. The pooled data, for ANB difference, showed a statistically significant mean difference of 3.93 degrees (95%CI 3.46 to 4.39; P<0.0001) in favour of the facemask. There was significant heterogeneity between these studies (I2=82%). One well designed trial, with a low risk of bias, reported outcomes of the use of the facemask compared to an untreated control at 3 years’ follow-up. This showed that improvements in overjet and ANB were still present at 3 years. However, there was no evidence of improved self-concept. The remaining trials each evaluated a different comparison and reported different outcomes so no meta-analysis was possible. Conclusions: There is some evidence that the use of a facemask, to correct prominent lower front teeth in children, is effective when compared to no treatment on a short term basis. However, in view of the general poor quality of the included trials, these results should be interpreted with caution. Further randomised controlled trials, with long follow-up, are required.

The purpose of this study was to identify Core components perceived by faculty to be effective in their prominent graduate entrepreneurship education programs. The study sought to identify the best practices in graduate entrepreneurship education programs from the perceptions of faculty in the field. Research questions guiding the study were: (1) What Core components related to the following Broad question areas are perceived by faculty to be effective in their prominent graduate entrepreneurship education programs: Activities and initiatives; Adult education principles and practices; Alumni and mentoring; Course offerings; Curriculum and degrees; Faculty data; Institutional characteristics; Instructional methods; Student companies; and Student data. (2) What other Core components and/or general observations are identified by faculty in the survey comments? (3) What are faculty perceptions of the popular marketplace publication rankings of graduate entrepreneurship education programs? A survey was distributed via the internet to faculty at 54 prominent graduate entrepreneurship education programs identified by The Princeton Review, US News & World Report, or the AACSB Entrepreneurship Spotlight Challenge. The survey was developed through several phases using panels of individuals with expertise related to this study. The resulting 106 Core components were divided into 10 Broad question areas and were evaluated individually and averaged for each Broad question area. Questions regarding the accuracy of graduate program rankings and student intent were also included, along with demographics, open-ended questions regarding additional Core components, and additional survey comments. The results of the study indicated the most important Core components in the Broad question areas were Alumni and mentoring and Institutional characteristics, while the Curriculum and degrees area was perceived to be much less important to graduate entrepreneurship education effectiveness. The results also indicated that student intent and popular marketplace publications were only moderately accurate in evaluating entrepreneurship education effectiveness. Four top programs dominated the perceptions of faculty as effective programs: Stanford, Babson, MIT, and Harvard. Findings indicated that faculty perceptions differed from other measures of effectiveness of graduate entrepreneurship education programs.

Abstract:
This thesis explores the manner in which the conflict in the oil-rich Niger Delta in Nigeria is represented in western Anglophone media. Large oil reserves in the Niger Delta have contributed millions of dollars towards the growth of Nigeria's export economy. Despite this, the Niger Delta is the least developed region in the country and is characterised by high rates of inequality. Residents of the Niger Delta have been outraged by the lack of action on the part of the Nigerian government and multinational oil corporations. Their discontent over the inequalities in the region has resulted in the proliferation of armed groups and militants who often use violent and criminal tactics to communicate their disgruntlement. This thesis closely examines the representations of the violent insurgency in the Niger Delta by conducting a Critical Discourse Analysis of 145 news texts selected from four western Anglophone newspapers from 2007 to 2011. The depiction of the conflict as it appears in the four newspapers is discussed in relation to an overview of scholarly literature which explores the portrayal of Africa not only in western media, but also in other forms of western scholarship and writing. The research undertaken in this study reveals that to a significant extent representations of the Niger Delta conflict echo and reflect some of the stereotypical and age-old negative imagery that informs meanings constructed about the African continent. However, the analysis of the news texts also shows that there are certainly efforts amongst some newspapers to move beyond simplistic representations of the conflict. The disadvantage however, is that these notable attempts tend to be marred by the use of pejorative language which typically invokes negative images associated with Africa. This study argues that the implications of these representations are highly significant as these representations not only affect the way in which the conflict is understood, but also the manner in which the international community responds to it.

Efforts to improve educational achievements in Israel are at the forefront of national concern. In acknowledging the crucial role of the teacher in this endeavour, this enquiry explores the prominent instructional practices of effective teachers of English as a Foreign Language in Israeli elementary schools. The qualitative study is placed within a social constructivist perspective of learning that, in turn, serves as a foundation to the authentic pedagogical approach as determined by Newmann et al (1996). A multiple case study was conducted, whereby observations were carried out in the classrooms of five teachers recognised to be effective in their practice, and followed up with post- observation and post-data analysis interviews. The data collected from each case individually was subsequently used to engage in a cross-case analysis. The analysis offers clear indication that all teachers provide significant opportunity for pupils to construct knowledge using a variety of techniques and strategies. There is also evidence to show –albeit to a lesser extent, that relevance in instruction, promoted primarily through the topic of the lesson and opportunities to develop intrapersonal skills, plays an important role in the practice of the effective teachers in this study. Both aspects identified are central to the notion of authenticity in instruction. The enquiry concludes with the suggestion that the findings from this study could be used as a basis to explore the extent to which the aspects of instruction identified feature in the practice of effective teachers in a wider context, with a view to developing a national model of pedagogy that would promote much-needed and sustainable educational improvements in the Israeli context.

Swahili ist keineswegs eine besonders „exotische“ Sprache, aber dennoch fällt es schwer, eine gu-te moderne Grammatik des Standard-Swahili (Kiswahili sanifu) zu finden. Insbesondere die Inter-pretation der vorkommenden Tempora, Aspekte oder Modi des Verbs sind in den erhältlichen Grammatiken diskrepant bis widersprüchlich. Der Artikel versucht, einen systematischen Ansatz für eine strukturelle Matrix des TAM-Systems im Swahili herauszuarbeiten. Dabei beziehe ich mich auf die von Shankara Bhat in The Prominence of Tense, Aspect and Mood (1999) vorge-schlagene typologische Methode. Bhat legt dar, dass nicht jede Sprache jede Dimension von TAM gleich gewichtet, sondern dass jeweils eine davon vorherrscht. Eine Analyse von TAM in einer be-stimmten Sprache muss sich daher an der prominenten Dimension orientieren. Im Artikel wird herausgearbeitet, dass das Tempus die strukturierende Dimension im Swahili darstellt und dass im Gegensatz dazu der Aspekt nicht systematisch grammatikalisiert ist. Dabei war es nötig, die im Swahili vorkommenden TAM-Formen funktional zu interpretieren und zu benennen, da diese in der aktuellen Literatur teilweise sehr unterschiedlich analysiert werden.

Background Growing evidence emphasizes the relevance of sphingolipids for metabolism and immunity of antigen-presenting cells (APC). APCs are key players in balancing tolerogenic and encephalitogenic responses in immunology. In contrast to the well-known prominent effects of sphingosine-1-phosphate (S1P) on lymphocyte trafficking, modulatory effects on APCs have not been fully characterized. Methods Frequencies and activation profiles of dendritic cell (DC) subtypes, monocytes, and T cell subsets in 35 multiple sclerosis (MS) patients were evaluated prior and after undergoing fingolimod treatment for up to 24 months. Impact of fingolimod and S1P on maturation and activation profile, pro-inflammatory cytokine release, and phagocytotic capacity was assessed in vitro and ex vivo. Modulation of DC-dependent programming of naïve CD4+ T cells, as well as CD4+ and CD8+ T cell proliferation, was also investigated in vitro and ex vivo. Results Fingolimod increased peripheral slanDC count—CD1+ DC, and monocyte frequencies remained stable. While CD4+ T cell count decreased, ratio of Treg/Th17 significantly increased in fingolimod-treated patients over time. CD83, CD150, and HLADR were all inhibited, but CD86 was upregulated in DCs after incubation in the presence of fingolimod. Fingolimod but not S1P was associated with reduced release of pro-inflammatory cytokines from DCs and monocytes in vitro and ex vivo. Fingolimod also inhibited phagocytic capacity of slanDCs and monocytes. After fingolimod, slanDCs demonstrated reduced potential to induce interferon–gamma-expressing Th1 or IL-17-expressing Th17 cells and DC-dependent T cell proliferation in vitro and in fingolimod-treated patients. Conclusions We present the first evidence that S1P-directed therapies can act additionally as immunomodulators that decrease the pro-inflammatory capabilities of APCs, which is a crucial element in DC-dependent T cell activation and programming.

Embryonic stem (ES) cell pluripotency is sustained by a network of transcription factors centred on Oct4, Sox2 and Nanog. Whilst Oct4 and Sox2 expression is relatively uniform, ES cells fluctuate between states of high Nanog expression possessing high self-renewal efficiency, and low Nanog expression exhibiting increased differentiation propensity. Moreover, modulation in the level of Nanog expression determines the efficiency of ES cell self-renewal. To identify genes regulated by Nanog, genome-wide transcriptional profiling was performed on ES cells expressing different Nanog levels and Nanog-null ES cells expressing a Nanog-ERT2 fusion protein in which nuclear Nanog activity can be regulated by tamoxifen. Surprisingly, only a minor fraction of the genes to which Nanog binds showed significant changes in response to Nanog induction. Prominent amongst Nanog-responsive genes is Estrogen-related receptor b (Esrrb). Nanog binds directly to Esrrb, enhances binding and pause-release of RNAPolII from the Esrrb promoter and stimulates Esrrb transcription. Consistent with these findings, elevation of Nanog produces a cell population that expresses uniformly high Esrrb levels. Moreover, double fluorescent reporter lines show that Esrrb and Nanog levels are strongly correlated in individual cells. Loss of Nanog is required for downregulation of Esrrb, which coincides with commitment to differentiate. Esrrb overexpression results in LIF independent self-renewal, and blocks neural differentiation, even in the absence of Nanog. Cell fusion experiments between ES and neural stem (NS) cells show that elevated Esrrb levels allow the reprogramming of the NS cell genome in the absence of Nanog. Esrrb can rescue stalled reprogramming during the derivation of Nanog-/- induced pluripotent stem (iPS) cells. Moreover, targeted knock-in of Esrrb at the Nanog locus rescues the ability of Nanog null ES cells to maintain germ cell development beyond E12. Finally, Esrrb deletion abolishes the defining ability of Nanog to confer LIF-independent selfrenewal to ES cells. Together these data identify Esrrb as a critical downstream mediator of Nanog function.
