Full textThe prostaglandin D2 (PGD2) pathway is involved in numerous biological processes and while it has been identified as a partner of the embryonic sex determining male cascade, the roles it plays in ovarian function remain largely unknown. PGD2 is secreted by two prostaglandin D synthases (Pgds); the male-specific lipocalin (L)-Pgds and the hematopoietic (H)-Pgds. Here, we report the localization of H-Pgds mRNA in the granulosa cells from the primary to pre-ovulatory follicles. We used adult female mice treated with HQL-79, a specific inhibitor of H-Pgds enzymatic activity, to provide evidence of an interaction between H-Pgds-produced PGD2 signaling and FSH signaling. This leads to the activation of steroidogenic Scc and StAR gene expression through increased FshR and LhR receptor expression leading to progesterone secretion. We also identify a role whereby H-Pgds-produced PGD2 is involved in the regulation of follicular growth through inhibition of granulosa cell proliferation in the growing follicles. Indeed, we report an altered H-Pgds expression in human ovarian tumors alongside a partial or complete absence of H-Pgds protein in granulosa cell tumors, suggesting a potential association between decreased levels of H-Pgds expression and a tumoral phenotype. Together, these results show PGD2 signaling to be essential for FSH action within granulosa cells, thus identifying an important and unappreciated role for PGD2 signaling in controlling the balance of proliferation, differentiation and steroidogenic activity of these cells

Full textLipid metabolism affects the maturation and the differentiation of bone cells. The aim of my PhD thesis is to explore two unknown sides of lipid metabolism which are the actions of phospholipase D (PLD) and those of prostaglandin PGE2 receptors during cell differentiation. Human lineage, Saos-2 cells and primary osteoblasts from calvaria of mice were used as cellular models of physiological mineralization. The ex vivo aorta culture under hyperphosphatemia conditions has been used to reproduce the calcification of the aorta, which is an ex vivo model of cardiovascular calcification (CVC). We showed that the expression and the activity of PLD increased in Saos-2 and primary osteoblasts after the fifth day of differentiation while in the aorta under hyperphosphatemia condition, PLD activity increased at the end of the sixth day. PLD inhibitors decreased the activity of alkaline phosphatase (TNAP) in osteoblasts and in calcified aorta while the overexpression of PLD1 in the Saos-2 increased it. In the second part of this work, we monitored the variation of the expression of PGE2 receptors during the maturation of Saos-2 cells. The EP3 gene expression increased in the late stage of the mineralization while that of EP4 decreased. In conclusion, these results indicated that the PLD activity by affecting the activity of TNAP could modulate the physiological mineralization and CVC. We showed that the mineralization is dependent of the change of the expression of PGE2 receptors in Saos-2 cells

This work aimed to study the effect of PGF2α administration on anestrous post-partum period in 80 goats, with continuos or controlled suckled, rised in semiextensive system with water and mineral salt ad libitum. The females, with age between two and six years, were randomly distributed in three groups (GI, GII e GIII). The females of GI (n = 30) received 250 μg of PGF2 α in vulvar muscle on days 6th and 12th after delivery. The females of GII (n = 30) received the treatment of Group I, but every day from days 6th to 10th after delivery. The GIII (n = 20) was the control group. The estrous detection was made by using teasers and the mating with bucks with confirmed fertility. The obtained data were analyzed by ANOVA and by analysis of standard errors of difference among proportions. Difference in the occurrence of estrous among the three experimental groups was not observed (P > 0,05), however the average time of post partum anestrous was not observed (P < 0,05) in animals treated with of post PGF2 α. It was also detected a relevant reduction (P > 0,05) of post-partum anestrous in females with controlled suckled. Difference was not registered (P > 0,05) on pregnant percentages among the different groups. It may be concluded that the administration of PGF2α in the beginning of puerperium in caprine is efficient to reduce the anestrous pos-partum period, specially in females controlled suckled; however do not influence the fertility in meat goats. In the second study the objective was to verify the viability of the PGF2α administration association or not with dexamethasone in the induction of birth in milk goats and its implication in the delivering of the afterbirth placenta,puerperal disturb and viability you create of them. This work was lead in the Pendencia Experimental Station, EMEPA, located at Soledade – PB. A total of 80 milk goats from different races had been used such as Anglo Nubian, Brown Alpine and British Alpine, presenting age between two and six years and gestational period between 143 and 146 days. Two experiments had been made, being experiment1 (n = 40) carried through in the dry period and experiment 2 (n = 40) carried through in the rainy period. The females had been randomly distributed in eight experimental groups (GI, GII, GII, GIII e GIV) for each experimental type. The GI ( n= 10) received 5,0 mg from PGF2α byintramuscular to vulvae way; the GII (n = 10) received 5,0 mg from associated PGF2 α the 8,0 mg of corticoids saw to intramuscular; the GII ( n= 10) was submitted the administration of 0,125 mg of PGF2 α by intramuscular to vulvae way and the GIV (n = 10) received 0,125 mg from the PGF2α by intramuscular to vulvae way, associated the 8,0 mg of dexamethasone by intramuscular way. On the other hand group GII presented difference statistics in the dry period, however it did not present difference statistic between groups in the different periods. It was concluded that the use of the synthetic prostaglandins associated or not to dexamethasone do not promote the artificial induction of the births without compromise the viability of the kids and the deliver of the afterbirth placenta.
Foram conduzidos dois experimentos para avaliar a eficácia do d-cloprostenol na redução do anestro pós-parto e a indução do parto com d-cloprostenol, Dinoprost trometamina e dexametasona em cabras. No primeiro estudo objetivou-se avaliar os efeitos da administração da PGF2α sobre o período de anestro pós-parto de 80 cabras, em aleitamento contínuo ou controlado, criadas em sistema semi-intensivo com acesso a água e sal mineral ad libitum. As fêmeas, com idade entre dois e sis anos, foram aleatoriamente distribuídas em três grupos experimentais (GI, GII e GIII). As do GI (n = 30) receberam, na musculatura vulvar, 250 μg de PGF2α no 6º e 12º dias após o parto. As do GII (n =30) foram tratadas, pela mesma via e com a mesma dose de PGF2α, no 6º, 7º, 8º, 9º e 10º dias do pós-parto e as do GIII (n=20) serviram de controle. A detecção do estro foi realizada por rufião com auxílio de pessoal habilitado e as coberturas efetuadas com reprodutores de fertilidade comprovada. Os dados foram analisados através da ANOVA e do cálculo do erro padrão da diferença entre proporções. Não foi verificada diferença (P>0,05) das porcentagens de estro entre os três grupos experimentais, todavia, a duração média do anestro pós-parto foi significativamente reduzida (P<0,05) nos animais tratados com PGF2α. Houve redução significativa (P<0,05) do anestro pós-parto das fêmeas em aleitamento controlado. Não foi registrada diferença (P<0,05) nas porcentagens de prenhez entre os diferentesgrupos. Os dados obtidos permitem concluir que a administração da PGF2α no início do puerpério é eficiente para reduzir o período de anestro pós-parto, especialmente nas fêmeas em aleitamento controlado, contudo, não exerce influência sobre a fertilidade de cabras SRD com aptidão para produção de carne. No segundo experimento objetivou-se verificar a viabilidade da administração da PGF2α associada ou não a dexametasona na indução de parto em cabras leiteiras e sua implicação no delivramento das secundinas,distúrbios puerperais e viabilidade das crias. Este trabalho foi conduzido na Estação Experimental de Pendência, pertencente à EMEPA, no município de Soledade-PB. Foram utilizadas 80 cabras de aptidão leiteira das raças Anglo-Nubiana, Parda Alpina e Alpina Britânica, apresentando idade entre dois e seis anos e período gestacional entre 143 e 146 dias. Foram feitos dois experimentos, sendo o experimento 1 (n = 40) realizado no período chuvoso. As fêmeas foram aleatoriamente distribuídas em oito grupos experimentais, sendo quatro grupos (GI, GII, GIII e GIV) para cada tipo experimental. O GI (n = 10) recebeu 5,0 mg de PGF2α, via intramuscular; o GII (n = 10) recebeu 5,0 mg de PGF2α associada a 8,0 mg de corticóides, via intramuscular; o GIII (n = 10) foi submetido à administração de 0,125 mg de PGF2α via intramuscular vulvar e o GIV (n = 10) recebeu 0,125 mg de PGF2α via intramuscular vulvar, associado a 8,0 mg de dexametasona via intramuscular. Apenas o GII apresentou diferença estatística no período seco, porém não apresentou diferença estatística nos diferentes períodos. O intervalo entre o parto e nos diferentes períodos. Conclui-se que os protocolos avaliados mostraram-se eficientes na indução artificial dos partos, as prostaglandinas sintéticas foram eficazes na promoção da luteólise e o tempo do delivramento das secundinas não foi afetado bem como a viabilidade dos cabritos.

Full textOsteoarthritis is a complex multifactorial disease; many factors are involved in its pathogenesis, among those factors prostaglandins. Some prostaglandins have inflammatory role and some have anti-inflammatory role. Prostaglandin D2 (PGD2) is a well-established anti-inflammatory PG which is synthesized by the Lipocalin prostaglandin-D synthase (L-PGDS) enzyme. Upon the initiation of the inflammatory process in osteoarthritis, chondrocytes try to save themselves by activating salvage pathways, among these pathways is the induction of L-PGDS gene. In this study we are addressing the signaling pathways involved in the regulation of (L-PGDS) gene expression, in chondrocytes treated with different inflammatory mediator. Rationale: understanding the regulation of L-PGDS will allow us to design therapeutic targets that can switch on the intrinsic anti-inflammatory pathway. Method and findings: In vivo, osteoarthritis was induced in mice knees surgically or naturally following aging, the development of osteoarthritis was then confirmed histologically. The expression levels of L-PGDS were detected by Immunohistochemistry. In vitro, human chondrocytes were treated with different inflammatory mediators (interleukin-1 beta, interleukin-17, hydrogen peroxide and tert-Butyl hydroperoxide). Interestingly, in most cases chondrocytes increased expression of L-PGDS in a dose and time dependent manner. We discovered that chondrocytes release L-PGDS into the extracellular space in response to inflammation, in vivo and in vitro. Lastly we observed different isoforms of L-PGDS generated in response to inflammation.
