Abstract:
The presence of a motto is not just a word sweetener. But it must have meaning that can be implemented in a concrete form. The values of Islamic education contained in the prostration motto can be implemented in the form of policies by the government as the formulator of the motto. So, it becomes a necessity if the motto "prostrate" is applied by the community, it will be able to bring to the regional development goals. This study aims to describe the policy of Islamic education based on the prostration motto in Tanah Bumbu Regency, as well as the obstacles and carrying capacity of the government in implementing Islamic education policy based on the prostration area motto. The method used by researchers is descriptive qualitative research with a policy study approach. Collecting data with direct interview techniques to respondents, namely Dr. Zairullah Azhar Regent of Tanah Bumbu Regency for the first period (2003-2010), as well as people involved in the policy as participants and the local community as a crosscheck who felt directly the impact of the policy. The results of this study indicate that the policy of Islamic education based on the prostration area motto begins with the formulation of the concept of the prostration area motto by Dr. Zairullah Azhar, prostration is an abbreviation of the values of Islamic education that is clean, thankful, honest, and peaceful. The implementation of policies based on this prostrate values includes Divine management, the obligation to pray in congregation for the ASN (State Civil Apparatus), the midday prayer in congregation, the Al Qur'an program (before work), the Duha prayer in the morning. , grave pilgrimage as a strategy for not corruption, Islamic education in the non-formal form of recitation of the majelis ta ‟lim. However, this policy does not touch on formal institutions, and society at large. Some people's acceptance of these policies follows and some does not. Because the policy can only be felt directly by the ASN / PNS. The policy of Islamic education based on prostration motto has not been able to be implemented in general in all fields. So that finally led to a conclusion that the policy of Islamic education based on the prostrate motto has not been fully implemented properly.

Abstract Prostratin is a unique phorbol ester that stimulates protein kinase C activity but is nontumor promoting. Remarkably, prostratin is also able to inhibit de novo human immunodeficiency virus type 1 (HIV-1) infection yet up-regulate viral expression from latent proviruses. Prostratin's lack of tumor promotion, coupled with its ability to block viral spread yet induce latent proviral expression, prompted studies to determine whether this compound could serve as an inductive adjuvant therapy for patients treated with highly active antiretroviral therapy (HAART). The current experiments indicate that prostratin is a potent mitogen for mononuclear phagocytes possessing many of the activities of phorbol myristate acetate (PMA) with notable functional differences. Prostratin, like PMA, accelerates differentiation of the myeloid cell-lines, HL-60 and THP-1, as well as mononuclear phagocytes from bone marrow and peripheral blood. Enzyme-linked immunosorbent assay and gene array analyses indicate significant changes in the expression of proteins and messenger RNA after treatment of cells with prostratin, consistent with phagocyte activation and differentiation. Prostratin blocks HIV-1 infection relating to down-regulation of CD4 receptor expression. The array analysis indicates a similar down-regulation of the HIV-1 coreceptors, CXCR4 and CCR5, and this may also reduce viral infectivity of treated host cells. Finally, prostratin is capable of up-regulating HIV-1 expression from CD8+ T lymphocyte–depleted peripheral blood mononuclear cells of patients undergoing HAART. This novel observation suggests the agent may be an excellent candidate to augment HAART by inducing expression of latent HIV-1 with the ultimate goal of eliminating persistent viral reservoirs in certain individuals infected with HIV-1.

Prostratin, a non-tumor promoting 12-deoxyphorbol ester, has been reported as a protein kinase C (PKC) activator and is shown to have anti-proliferative activity in certain cancer cell types. Here we show that GRC-2, a prostratin analogue isolated from Euphorbia grandicornis, is ten-fold more potent than prostratin for inhibiting the growth of human non-small cell lung cancer (NSCLC) A549 cells. Flow cytometry assay revealed that GRC-2 and prostratin inhibited cell cycle progression at the G2/M phase and induced apoptosis. The cytotoxic effect of GRC-2 and prostratin was accompanied by activation and nuclear translocation of PKC-δ and PKD as well as hyperactivation of extracellular signal-related kinase (ERK). Knockdown of either PKC-δ, PKD or ERK significantly protected A549 cancer cells from GRC-2- and prostratin-induced growth arrest as well as apoptosis. Taken together, our results have shown that prostratin and a more potent analogue GRC-2 reduce cell viability in NSCLC A549 cells, at least in part, through activation of the PKC-δ/PKD/ERK pathway, suggesting the potential of prostratin and GRC-2 as anticancer agents.

ABSTRACT To design strategies to purge latent reservoirs of human immunodeficiency virus type 1 (HIV-1), we investigated mechanisms by which a non-tumor-promoting phorbol ester, prostratin, inhibits infection of CD4+ T lymphocytes and at the same time reactivates virus from latency. CD4+ T lymphocytes from primary blood mononuclear cells (PBMC) and in blocks of human lymphoid tissue were stimulated with prostratin and infected with HIV-1 to investigate the effects of prostratin on cellular susceptibility to the virus. The capacity of prostratin to reactivate HIV from latency was tested in CD4+ T cells harboring preintegrated and integrated latent provirus. Prostratin stimulated CD4+ T cells in an aberrant way. It induced expression of the activation markers CD25 and CD69 but inhibited cell cycling. HIV-1 uptake was reduced in prostratin-stimulated CD4+ T PBMC and tissues in a manner consistent with a downregulation of CD4 and CXCR4 receptors in these systems. At the postentry level, prostratin inhibited completion of reverse transcription of the viral genome in lymphoid tissue. However, prostratin facilitated integration of the reverse-transcribed HIV-1 genome in nondividing CD4+ T cells and facilitated expression of already integrated HIV-1, including latent forms. Thus, while stimulation with prostratin restricts susceptibility of primary resting CD4+ T cells to HIV infection at the virus cell-entry level and at the reverse transcription level, it efficiently reactivates HIV-1 from pre- and postintegration latency in resting CD4+ T cells.

Abstract:
In this article, I explore the prostration accumulation portion of the Preliminary Practices of a specific group of Tibetan Buddhist women in Bongwa Mayma, a rural area of Yushu Tibetan Autonomous Prefecture in Qinghai Province. I focus specifically on the nuns and lay women who utilize this set of teachings and practices. The Preliminary Practices not only initiate practitioners into a specific tradition (that of the Drikung Kagyu and more specifically the Amitabha practices of this lineage), but also more fundamentally into Vajrayāna Buddhism as it is practiced in contemporary Tibet. Although monks and male lay practitioners in this region also tend to perform the same Preliminary Practices, I focus specifically on women because of their unique relationship with bodily labor. I begin this article with a discussion of the domestic and economic labor practices of contemporary Tibetan women in rural Yushu, followed by an analysis of Preliminary Practices as understood through the Preliminary Practice text and oral commentaries utilized by all interviewees and interviews (collected from 2016–2020) with female practitioners about their motivations, experiences, and realizations during the Refuge and prostration accumulation portion of their Preliminary Practices. Women themselves view bodily labor as a productive and inevitable aspect of life. On the one hand, women state openly that their domestic duties impede upon their ability to achieve religious realization. On the other, they frequently extol the virtues of hard work, perseverance, patience, and fortitude that their lives of labor helped them to cultivate. Prostration is meant to embody the act of going for Refuge, of submitting oneself to the teachings of the Buddha, to the path of the dharma, and to the community of religious practitioners with whom they will study and grow. Prostrations are meant to embody the extreme difficulty of Refuge, to remove obscurations, to crush the ego, and to confirm a dedication to endure the hardships on the path to realization. Buddhist women, despite their ambiguous relationship with physical labor, see the physical pain of this process as a transformative experience that allows them a glimpse of the spaciousness of mind and freedom from attachment-filled desire promised in the teachings they receive.

In the past decade, much emphasis has been put on the transcriptional activation of HIV-1, which is proposed as a promised strategy for eradicating latent HIV-1 provirus. Two drugs, prostratin and hexamethylene bisacetamide (HMBA), have shown potent effects as inducers for releasing HIV-1 latency when used alone or in combination, although their cellular target(s) are currently not well understood, especially under drug combination. Here, we have shown that HMBA and prostratin synergistically release HIV-1 latency via different mechanisms. While prostratin strongly stimulates HMBA-induced HIV-1 transcription via improved P-TEFb activation, HMBA is capable of boosting NF-κB-dependent transcription initiation by suppressing prostratin-induced expression of the deubiquitinase A20, a negative feedback regulator in the NF-κB signaling pathway. In addition, HMBA was able to increase prostratin-induced phosphorylation and degradation of NF-κB inhibitor IκBα, thereby enhancing and prolonging prostratin-induced nuclear translocation of NF-κB, a prerequisite for stimulation of transcription initiation. Thus, by blocking the negative feedback circuit, HMBA functions as a signaling enhancer of the NF-κB signaling pathway.

Abstract:
L’infection par HIV-1 représente un des problèmes de santé publique majeurs de notre société actuelle. Le traitement HAART (Highly Active AntiRetroviral Therapy) inhibe le cycle réplicatif viral mais ne permet pas l’éradication du HIV-1. La principale cause de cet échec thérapeutique est la persistance de réservoirs cellulaires infectés de manière latente par HIV-1, qui, lors de l’arrêt du traitement HAART, sont à l’origine d’un rebond de la charge plasmatique virale. Le défi actuel est donc de découvrir de nouvelles méthodes d’élimination des cellules réservoirs. Une des stratégies envisagées est de forcer l’expression virale dans les cellules infectées de manière latente afin d’entraîner leur destruction suite à leur détection par le système immunitaire ou suite aux effets cytopathiques viraux. Parallèlement, le traitement HAART serait maintenu afin de limiter la propagation des virions néo-synthétisés. Plusieurs éléments sont impliqués dans la répression transcriptionnelle associée à la latence post-intégrationnelle du virus HIV-1 :la nature du site d’intégration ;l’absence de facteurs cellulaires inductibles tels que NF-κB ;la structure chromatinienne du provirus et les modifications post-traductionnelles des histones ;l’absence de niveaux suffisants de la protéine trans-activatrice Tat. De plus, notre laboratoire a précédemment mis en évidence un lien entre deux de ces éléments, en démontrant, dans une lignée modèle de latence post-intégrationnelle, que la cytokine pro-inflammatoire TNFα, un activateur de la voie de signalisation NF-κB, permet une réactivation synergique de l’expression virale combinée à l’inhibiteur d’histone-désacétylases (HDACI) TSA. Cependant, l’utilisation thérapeutique du TNFα et de la TSA est inenvisageable en raison de leurs toxicités.

Abstract:
Treatment options for patients with advanced prostate cancer (PC) still remain limited and rarely curative. The prostatic acid phosphatase (PAP) is prostate specific protein over-expressed in more than 90% of prostate tumours. Although an FDA-approved vaccine for the treatment of advanced prostate disease, PROVENGE® (sipuleucel-T), has been shown to prolong survival, the precise sequence of the PAP protein responsible for the outcome remains unknown. As the PAP antigen is one of the very few prostate-specific antigens for which there is a rodent equivalent with high homology, pre-clinical studies using PAP have the potential to be directly relevant to the clinical setting. The current study identified HLA-A2 and HLA-DR1 PAP-derived peptides using the transgenic HHDII/DR1 and C57Bl/6 mice. The PAP-114-128 (15-mer) peptide was shown to elicit CD4+ and CD8+ T-cell-specific responses in C57Bl/6 mice. Furthermore, when immunised in a DNA vector format (ImmunoBody), PAP-114-128 was able to prevent and reduce the growth of TRAMP C1 prostate cancer cell-derived tumours in both prophylactic and therapeutic settings. This anti-tumour effect was associated with an enhanced infiltration of CD8+ tumour-infiltrating lymphocytes (TILs) and the generation of high avidity T cells secreting elevated levels of IFNγ. Importantly, PAP-114-128 specific IFNγ response was also seen in PBMC isolated from PC patients. Also, immunisation of C57Bl/6 and HHDII/DRI mice with the analogue peptide epitope (obtained by altering the second amino acid of PAP-114-128) showed significantly enhanced IFNγ response compared to PAP-114-128 epitope. Collectively, PAP-114-128 appears to be a highly relevant peptide on which to base vaccines for the treatment of advanced PC.

(Uncorrected OCR) Abstract of thesis entitled Telomerase activation and telomeric repeats alteration in sex hormone-induced prostatic carcinogenesis in the Noble rat submitted by Chan Ching for the Degree of Master of Philosophy at the University of Hong Kong In November, 1998 Despite its distinction as the most frequently diagnosed cancer and the�second leading cause of cancer deaths in men, little is known about the causes and mechanisms of prostatic carcinogenesis. The animal model, based on the existing sex hormone-induced prostate carcinogenesis in the Noble rat, by substantially increasing the dosage of testosterone while keeping the level of estrogen unchanged has been reported. With this modified protocol, we have successfully induced high incidence of prostate carcinoma in Noble rats. The earliest time of development of dysplasia was two months, carcinoma in situ at 4 months and fully developed carcinoma at six months. The tumor incidence was 92% after 12 months hormonal implantation. This animal model is very useful in the investigation of prostate cancer as its multi -step nature mimics the human situation and its high incidence in relative short time. Telomerase activation is a characteristic of immortalized tumor cells and is thought to contribute to the mechanism by which these cells abort the normal process of senescence. Telomerase activity has been detected in various human cancers and there are reports showing that telomeric repeat fragment (TRF) length may be correlated to the staging of tumors. These findings suggested that telomerase activation and TRF length alteration might be useful in prognosis of different cancers. Study of prostate cancer on animal model can compensate for the difficulties in human cancer research, as stage-by-stage investigations are available. In this study, we proposed that telomerase would be activated in sex hormone-induced prostatic carcinogenesis and TRF length alterations might be correlated to the different stages of prostatic carcinogenesis. We have examined the telomerase activation as well as telomeric repeat fragment content alteration in the hormonal induced prostatic carcinogenesis in Noble rat. Telomerase activation is common in prostate carcinoma and also can be detected in 12% of non-malignant tissues. For the non-prostatic tissues tested, all the testis tissues (n=5) were strongly positive for telomerase activity and only one liver tissue (n=5) showed weak. telomerase activity. The high frequency of telomerase activation in prostatic carcinoma specimens suggested that it might be a useful malignancy marker for prognosis evaluation in prostatic carcinogenesis. There were alterations of TRF content in dorsal lateral prostate. The ventral prostate tissues have the similar results as the dorsal lateral prostate. It seems that the TRF content in normal tissues is less than that of hyperplasia, dysplasia and carcinoma tissues. However, no obvious relationships between the TRF content and clinicopathological properties of prostatic carcinogenesis were observed. Also, there was no significant relation between the telomerase activation and TRF content in this study. Hence, the TRF content appear to have no significant correlation with prostatic carcinogenesis.

In Sweden, close to 10 000 men are annually diagnosed with prostate cancer (PCa) and approximately 2400 men die of their disease each year. Today there is no reliable marker that can separate patients who will have an aggressive type of disease that requires treatment, from patients who will have a more indolent clinical course and can be left untreated. This further leads to the current problem of over treatment of men with PCa. Hence, there is an urgent need for reliable prognostic markers that can be used at time of diagnosis. With the discovery of recurrent gene rearrangements in PCa, most commonly ERG rearrangements, hope came that this aberration could play a role in diagnosis and/or prognosis of the disease. The aim of this thesis was to investigate the clinical implication of ERG rearrangements in the management of PCa. The work in this thesis supports the findings from previous studies, suggesting that the ERG rearrangement is a sign of a more aggressive type of cancer. The major findings are that in multifocal PCa, the ERG rearranged cancer foci are more prone to metastatic dissemination compared to foci without the ERG rearrangement and that patients harboring the ERG rearrangement have a faster disease progression leading up to earlier start of hormonal treatment. Furthermore, the results add an additional level of complexity in a subset of PCa tumors that harbor multiple gene rearrangements on the cellular level. The result also show that the newly available ERG antibody is highly predictive of ERG rearrangement and is appropriate to use when faced with limitations in tissue amounts. The findings in this thesis indicate that the ERG rearrangement has a potential role in the clinical management of PCa but further studies arerequired.

Prostrate juniper heath (H15 Calluna vulgaris-Juniperus communis subspecies nana heath) is of considerable conservation importance. Its infrequent distribution in northwest Scotland is often attributed to historical burning. The intraspecific taxonomy of Juniperus communis L (Cupressaceae) is unclear. This thesis seeks to understand the history and current status of the community, and to clarify the taxonomy of juniper in Scotland. The impact of current levels of land use on extant HI5 was found to be low. A study of the environmental conditions associated with HI5 revealed that the community was absent from large areas of north-west Scotland with suitable conditions. Other communities with only infrequent juniper exist in many such areas. Sites with environmental conditions suitable for HI5 were investigated using palaeoecological methods. Fire had occurred in both HI5and non-H15 sites, but there was a prolonged recent absence in HI5. A study of annual growth rings showed juniper in HI5 to be twice as old as in other communities, emphasising the lack of recent disturbance. Evidence of greater past juniper cover was not found on non-H15 sites. It is argued that the history of prostrate juniper heath is more dynamic and complex than previously believed. Comparative morphological, genetic marker, and reciprocal transplant studies of juniper populations showed that Juniperus communis ssp. nana is characterised by genotypic prostrateness, imbricate leaf arrangement, and abrupt leaf tips. It is argued that the conservation importance of HI5 and juniper are increased by the findings of this study. Implications for conservation practice, and future research, are considered.

Abstract:
March 2021, a 12-month-old infant was brought to the HPS East Zone Little Jonh, in the capital of the State of Amazonas, Brazil, where she was admitted with fever, vomiting, intense prostration, little acceptance of breast milk, difficulty in walking, associated with left-facing strabismus and neck stiffness. At admission, the main author performed a lumbar puncture, and serological tests on the CSF to identify the causative agent were referred to a reference laboratory, which suggested that they be referred to the Central Laboratory - LACEN for suspecting the presence of agent Angionsytrongylis Canyonensis. Although, the identification of the causative agent is difficult to be detected and confirmed, this case report shows us the importance of the clinical laboratory in the identification of Eosinophilic Meningitis. The sub-detection of eosinophils in the CSF helps to underestimate the prevalence of eosinophilic meningitis. Thus, CSF analysis, slide preparation, sediment staining, as well as knowledge about the importance of adequate leukocyte differentiation in CSF are extremely important, to properly conduct the final diagnosis of the disease and prevent possible endemicity. The knowledge of this condition and early detection allows an effective therapy and improvement of public health actions. ? ?,
