With recent advances and cost reductions in next generation sequencing (NGS), the amount of genetic sequence data is increasing rapidly. For example, although large numbers of genetic variations are discovered during clinical use of NGS, annotating and understanding the impact of such coding variations on protein phenotype remains a bottleneck (i.e. what is the molecular mech

The need to high quality therapeutic proteins has grown significantly in the past 30 years. Recombinant proteins are often produced from vectors inserted into E. coli cell lines for large scale production. However, over-expression of the protein within the cell can lead to the formation of large, insoluble aggregates known as inclusion bodies. Native monomer protein can be isolated from inclusion bodies through a refolding process. This entails disruption of the aggregate structure with high concentrations of denaturant and renaturation in native-promoting solution. Our work characterises prot

<p>Forty-two high producing Holstein cows were paired by body condition and mature equivalent milk production and fed either a high or low energy complete ration ad libitum during the last 16-20 weeks of lactation. Cows fed the high energy ration ate more feed, produced more milk and gained more body condition than cows fed the low energy ration. Cows were fed to maintain condition during the dry period. During weeks 3-15 of the next lactation, half the cows in each condition group (fat or thin) were fed a mixed ration with soybean meal (SBM) as the major protein source. The remaining

Abstract:
Obesity and particularly visceral adiposity are important factors in insulin resistance, and contribute to blunted protein anabolism that is observed with diabetes. This insulin sensitivity of protein metabolism in obese adults with type 2 diabetes, does not improve when normalizing glycemia with medication, although rates of protein turnover decrease. On the other hand, insufficient protein intake in weight-loss diets that keep protein at a fixed percentage of energy could exacerbate insulin sensitivity of protein metabolism. Thus the objective of this thesis was to perform a pilot study in 6
