Abstract:
Adamo's article on Ebed-Melech's protest brings fresh insight into my earlier article on Song of Songs 1:5-7, prompting me to reread the text as a protest song (essay) against the racial stigmata that continue to bedevil black people in the world. The current article, using hermeneutics of appropriation, maintains the meaning of שְׁחוֹרָה as a black person, who in the Song of Songs protests against the racism, which transformed her status to that of a socioeconomic other. The study is informed by the contemporary and historical contexts of racial injustices and stigma suffered by Blacks for 'being' while Black. The essay investigates this question: In which ways does Adamo's reading of Jer 38:1-17 influence an African reading of Song 1:5-7 as a protest against racism? The article employs African Biblical Hermeneutics, as part of a creative and literary art in the protests against racism, to read the biblical text as our story-a divine story, which in the language of Adamo, has inherent divine power that can empower oppressed black people.

Abstract:
On Cape Breton Island, where coal mining and steel making were once an essential part of the region’s culture and economy, protest song and verse are found in abundance. This article explores some previously unexamined protest songs and verses of Cape Breton Island. The body of songs is culled largely from the Maritime Labour Herald, a newspaper of the 1920s that included both locally and internationally composed works. Some earlier folklorists ignored protest songs because their paradigms did not permit them to view these forms as authentic cultural expressions. Their approach raises complex issues of how authenticity is constructed and by whom. My intent is to show that a well-developed protest song tradition was alive and well, and played an important role in the labour struggles of the 1920s. Indeed, these vernacular materials were used for solidarity during times of upheaval and change in Cape Breton Island.

Abstract:
In an increasingly interconnected and globalised world, the need for cross cultural understanding is greater than ever before. Exploring and analysing songs from different cultures can be an effective ‘entry point’ into learning about the nature of other peoples and societies lives and for developing a sense of ethnocultural empathy. Protest songs can provide a lens for intercultural analysis, especially for understanding minority or subcultural perspectives. Translating songs into different languages makes these works more accessible to a broader cross-section of people. We present translations of protest songs, two from Indonesian to English, and one from English to Indonesian. We discuss the respective importance of meaning and poetics in making song translations. Strict adherence to song rhymes is a challenge for translators and one which may impede meaning. The optimal approach depends on the format in which the translation is likely to be presented. Fluency in the target language, rather than the source language, is more helpful for successful translation. Finally, we make some recommendations about the usefulness of intercultural song (text) translation analysis and intercultural awareness.

Abstract:
The article focuses upon some aspects of the impact of English song names resulting from secondary nomination from the names of narcotic substances on young subjects. A large number and variety of song names of different genres encouraged us to introduce a special term «songonyms». As the research material, the name of the song LSD (performed by the rock band Hawkwind) has been chosen. It is difficult to clearly assume what effect song names have on the listener: whether they call for any actions, or, on the contrary, cause a protest and rejection. Therefore, in this study, it seemed important to verbalize the image of the LSD song name in the linguistic consciousness of young people and to reveal what implications this name carries - calling for actions or protest. By means of free associative experiment, according to the authors, individual meanings inserted by an individual into the song name are "revealed". The experiment has been conducted among 2-4-year students in one of the universities of Chelyabinsk. Through the making up of associative fields, it has been revealed that the song name LSD is perceived by young people not as calling for any actions, but as a protest element of rejection.

The goal of this article is to discuss the relationship between popular music (song writing, in particular) and issues of social protest, as portrayed in the various repertoires. The interface of the analysis is of a semiotic type, and the steps will follow a path that goes from problematizing the issue as such (with an emphasis on the current difficulties of identifying protest songs in terms of "genre"), to the definition of those stylistic elements pertaining to the context (in/for which these songs are written and/or played), the themes (as appearing from the lyrics) and the music itself (with the implication that the latter aspect is normally underrated in its significance, within such a process).

Abstract:
AbstractThe rediscovery of democratic traditions of folk song in Germany after the Second World War was not just the counter-reaction of singers and academics to the misuse of German folk song by the Nazis. Such a shift to a more ‘progressive’ interpretation and promotion of folk tradition at that time was not distinct to Germany and had already taken place in other parts of the Western world. After firstly examining the relationship between folk song and national ideologies in the nineteenth century, this article will focus on the democratic ideological basis on which the 1848 revolutionary song tradition was reconstructed after the Third Reich. It will look at how the New Social Movements of West Germany and the folk scene of the GDR functioned in providing channels of transmission for this, and how in this process a collective cultural memory was created whereby lost songs – such as those of the 1848 Revolution – could be awakened from extinction. These processes will be illustrated by textual and musical adaptations of key 1848 songs such as ‘Badisches Wiegenlied’ (Baden Lullaby), ‘Das Blutgericht’ (The Blood Court) and ‘Trotz alledem’ (For all that) within the context of the West German folk movement and its counterpart in the GDR.

Abstract:
Protest song has been an important component of grassroots political struggles, and the Palestinian resistance to Zionist settler-colonization is no exception. This article draws on original research with activists in the Palestinian popular resistance on the impact of song during the first intifada (1987 to 1993) and more recently in the opposition to the segregation wall and accelerated colonization of the West Bank. The significance of international solidarity to the Palestinian struggle is noted, and the role of protest song in international solidarity is explored. The activities of Edinburgh-based community choir San Ghanny in using song as an expression of solidarity with the Palestinian popular anti-colonial struggle is analysed. Protest song is a globally recognizable form, which can help to build connections with social movements in different parts of the world and in different periods of history, which is both rooted in individual places and struggles, and also transcends these at the level of global solidarity.

In the late 1950s, Pete Seeger received a letter from his manager, Howie Richmond, begging him to write a new hit song. &hellip; [Richmond] believed that &ldquo;protest songs&rdquo; were not marketable. Seeger was angry&mdash;he had a new song in mind, with words from a poem that he had set to music, and he believed it was, in a deep and significant sense, a song of protest.&hellip;. The song, of course, was &ldquo;Turn! Turn! Turn! (To Everything There Is a Season),&rdquo; which continues to be performed and recorded by many artists, and most famously became a huge folk-rock hit for The Byrds. It was as though, despite himself, Seeger produced a hit song, even when commercial popularity was the furthest thing from his mind&mdash;an example of how inseparably his songwriting talents and political principles were bound together.<p class="mrlink">This article can also be found at the <a href="http://monthlyreview.org/index/volume-66-number-7" title="Vol. 66, No. 7: January 2015" target="_blank"><em>Monthly Review</em> website</a>, where most recent articles are published in full.</p><p class="mrpurchaselink"><a href="http://monthlyreview.org/index/volume-66-number-7" title="Vol. 66, No. 7: January 2015" target="_self">Click here to purchase a PDF version of this article at the <em>Monthly Review</em> website.</a></p>

Abstract: Our study bends over Torquato Neto¿s songs, prior to his participation at the Tropicalism, which has a similar profile of the activist song, of protest, in the middle of the 1960s, and during his involvement with the movement, turning them into a beginning of questioning the established critical discourses about his poetry. We try to establish a dialogue between our listening to the songs and the critical readings which consider them separately, as a poetic text only. So that we can question the two opposite extremes in which his poetry is confined to: either the poet is seen as an avant-garde artist who continues a certain tradition again, of ingenuity and constructive rigidity, or as a romantic radical libertarian who, in accordance with his political-existential project, abandons his life

Rose, Mark. "Striking a discordant note : protest song and working-class political culture in Germany, 1844-1933." Thesis, University of Southampton, 2010. https://eprints.soton.ac.uk/367356/.

Abstract:
This thesis examines the role played by protest song in the development of the political culture of Germany’s industrial working class between 1844 and 1933. Protest song was an integral component in the struggle of the German working class to achieve some measure of political and social equality in the nineteenth and early twentieth centuries. Throughout this period, the working class found itself subjected to varying levels of political repression by the German authorities, and in order to promote their political views, industrial workers used the medium of song to protest against injustice. The thesis seeks to determine the significance of protest song for the political development of the German industrial working class through an analysis of song lyrics. The study of working-class protest song lyrics has largely been the preserve of historians from the former German Democratic Republic, where scholarship was shaped by the unique political imperatives of history writing under the Communist regime. This thesis seeks to redress the historiographical imbalance that this approach engendered, arguing that protest song produced under the auspices of the Social Democrats was both a culturally valid and politically significant feature of German working-class political life, albeit one that offered a different ideological approach to that of the overtly revolutionary output of the Communist movement. Additionally this thesis will acknowledge that working-class song was not merely used as an instrument of protest, but also as a medium to communicate political ideology. Protest song was an integral part of the cultural capital of the working class milieu, creating a distinct canon upon which German industrial workers drew in a variety of political, social and cultural situations. This study will engage with this canon in order to establish how the cultural practice of singing endowed working-class protest songs with an intrinsic political significance.

Abstract:
This work is a study of contemporary protest song in the USA through the songs and social and political activism of Bruce Springsteen. It contends that Springsteen has been able to reexamine American protest songs and frame a new genre under the umbrella of rock music by adapting his music to the social and political context of contemporary America. The thesis that I defend shows that the reason behind the commercial success of Springsteen can be explained by the fact that he has adopted several artistic strategies which have brought him an audience composed of dedicated fans who admire him. I argue that Springsteen adopts four main strategies: firstly, singing about ordinary people and reporting their daily hardships; secondly, composing somber songs that deal with the despair of blue collars and marginalized Americans; thirdly, adopting mimesis (imitation) which has a cathartic effect on Springsteen’s audience; and finally, getting involved in social and political activism. The thesis contends that Springsteen is among the top selling rock singers in the USA and several other countries thanks to these four strategies. Springsteen has reinvented protest song as a genre and has become a reference in a world where everything is co-opted including the very act of rebellion. I, therefore, suggest a new term, reflective songs, to describe the songs of Bruce Springsteen when they reflect on the condition of the socially excluded instead of protesting pointlessly. Springsteen’s reflective song is undoubtedly the most relevant artistic medium to shed light on the most deprived people of contemporary America. Perhaps, the reflective song of Bruce Springsteen will not change the world, but at least it gives people a reason to hope for a better future

During this study we focused our research on dialogues between the Protest Song of 1960s and 1970s, especially those occurred in certain works created by Gilberto Gil, Caetano Veloso and Chico Buarque de Holanda and a few songs produced by politicized and protest rock launched by bands from Brasilia such as Capital Inicial, Plebe Rude and Legião Urbana in the 1980s, contextualizing this cultural production within an important period in the recent history of Brazil. Through the theory developed by Bakhtin, we were confronted with dialogues between speakers and speeches, which allowed us to produce analysis of the characteristics and content of the dialogues between the "Old" Protest and the "New" Protest. The investigation allowed us to point out some similarities between the songs analyzed, and also enabled us to interpret them as articulating elements of the period in which they arose, which is evident in the performance of songs from both periods as a political tool of contestation.

Abstract:
This dissertation presents a historical study of intersections between music and activism in Poland from the election of Cardinal Karol Wojtyła as Pope John Paul II in 1978 to Poland’s first democratic elections in 1989. Musical action in three cultural spheres shapes the project: (1) the political activism of musicians, (2) activists who turn to music as a political instrument, and (3) the musical ambitions of the communist authorities, the Polish United Workers’ Party. I critique the repercussions of politics in music as well as music’s significance for policy makers and dissidents, and I assume that neither course of influence is intrinsic or inevitable under state socialism. In doing so, I highlight the complex relationship between activist culture and music at the end of the Cold War. Throughout the decade, religious hymns, patriotic anthems, experimental music, and popular songs shared spaces in Polish society, projected analogous ambitions, reflected communal responses, and partook in debates about culture’s capacity to effect political action. The plurality of musical genres and music histories during the Cold War reflects the political tensions in the Polish opposition to state socialism. The diverse materials I investigate in this dissertation respond both to the tumultuous politics of the 1980s and to the ethnographic, historical, and analytical methods I employ to write music history. My thesis—that political activism offered politicians, activists, and musicians the opportunity for constructive creative action—provides a model for rethinking Cold War music history. I begin with an explanation of the Communist Party’s program for music and the practical means by which it carried out this vision through the decade. Two chapters examine specific historical moments: I critique the ways in which music has come to be associated with the August 1980 strikes that brought about the formation of Solidarity, the first independent trade union in the Eastern bloc, and map the sites of music making in the weeks after martial law’s imposition in December 1981. I explore the resonance of popular sacred hymns and plainchant for musicologists, composers, and members of the opposition through the final decade of the Cold War. The dissertation concludes by analyzing the unofficial musical discourse on independence, drawing out the concept’s resonance for artists invested in their own musical autonomy.

Abstract:
This study investigates the role music played in the Civil Rights Movement as a form of political protest. The first part of the studies analyzed how political protest music was used in the early part of the twentieth-century leading up to the Civil Rights Movement of the 1960s. An analysis of the role of music in African-American culture also provides a historical background to the music-making of the Civil Rights Movement. Specific musical forms such as topical ballads, freedom songs, and spirituals are examined. In addition, musical influences of African culture as well as religious influences on music-making during the Civil Rights Movement are also examined. The second section of the paper investigates the life and murder of NAACP organizer Harry T. Moore of Mims, Florida. Moore's life and death became the subject of a topical ballad, “The Ballad of Harry T. Moore”, composed in 2001 by musical group Sweet Honey In The Rock. An analysis of the song's, literary, political, and musical connections to the ideology and music of the Civil Rights Movement, as well as subject matter, gives evidence that places the song within the tradition of the musical protest activities of the Civil Rights Movement.

Abstract:
The objective of this research is to analyze the Latin America integration project conceived by the artists involved with the protest song in the 1960s and 1970s. In this period, the impact of Cold War resulted in a polarized debate that affected the cultural field, and especially the artistic field. The common political experiences favored the exchange of ideas and the intense movement of artists across the continent. Using the most prominent artists discography as source, the purpose is to analyze three moments of the history of \"transnational connections\" in the protest song in Latin American: a) the first, between 1963 and 1966, time of development and consolidation of nueva canción movement in the Southern Cone, when the engagement was manifested fundamentally by social criticism; b) the second, between 1967 and 1969, marked by the impact of the I Encuentro de la Canción Protesta held in Cuba, that highlighted two important themes: revolution and anti-imperialism; c) the third, which comprises the 1970s, a period marked by the radicalization of political discourse and by the intensification of connections. Especially in this third phase, I analyze the Brazilian artists efforts to integrate the circuits of Latin American protest song.

It has become imperative in the current context of Brazilian public school the reading practices as an agenda of constant reflection and research for alternatives that can contribute to the development of the reading comprehension in various levels of middle school students, given the Saeb data and Prova Brasil (2014), whose large-scale assessments showed difficulties of the students to select implicit information, inferring the meaning of a word or expression, among others; there by, indicating, serious deviations of understanding and the appropriation of the writing production, in the act of reading. Based on this fact, the present report has as a guiding objective to promote the formation of a proficient and critical reader through the analysis of Brazilian social protest songs in activities of reading and understanding the implicit textual, enabling recognize them as situated discursive practices in daily life that can contribute to the formation of active citizens in society. Was taken as theoretical support the studies of textual linguistics, the sociocultural perspective; using the sociointeractional language design, subject and text for which the textual and discursive understanding can only be understood when it exceeds the limits of decoding linguistic elements and reaches the horizons of mobilization of the socio-historical and cultural knowledge and reconstruction of these within a communication situation in a given context. Therefore, we used a qualitative research, with the methodological approach by the application of didactics sequence in a 9th grade of elementary school, that used the song of social protest genre as mediator. Taking into account the proposal prepared for the research and what was executed, it is possible to say that the understanding of the text work from the songs analysis is configured as a positive and effective alternatives, as the students found new forms of meanings construction for the studied texts. The results obtained from this work fulfilled the ultimate goal of this trajectory that is becoming theoretical and practical support for the development of a concrete tool for socialization of experienced knowledge: the Pedagogical Section, whose main purpose is to share successful experiences with other Portuguese-speaking teachers in teaching and in the reading learning process.

Abstract:
The object of those researches is to study in a country where the cultural substratum is massively invested by the oral character, a way of expression, of communication, of resistance, of mobilization, of contestation, of claiming and affirmation of their identity, that was constituted by a great part of the Algerian population, an indispensable outlet to the maintenance of an individual and collective equilibrium strongly compromised, but also and mostly a non negligible possibility of self affirmation : the song. In deed, carrying feelings and resentments of a population taken between terrorism and repression and whose speech is excluded of the official circuits, denouncing the violence of the religious extremism like the abuses and the deficiencies of the power in place, transgressing the interdicts and the taboos, claiming a plural culture and identity, the Algerian songs of the 1990 are a vector of contestation and diversity, a real way of communication that we have to apprehend by linking it to the whole system of which it participates

Abstract:
Protein S (PS) is a plasma glycoprotein that serves as a cofactor for activated protein C (PC) anticoagulant activity. Inherited PS deficiency has been found to be associated to thrombotic disease in several families. In the present study, we report on a Spanish family with type II PS deficiency. The propositus is a 40 year-old male that was referred to our center for study after having suffered from multiples thrombotic events since he was 20 year-old. After his first episode of deep vein thrombosis (DVT) he had 4 recurrences, three of them complicated with pulmonary embolism. It should be remarked that one of the episodes occured while the patient was under oral anticoagulant treatment. The basic screening of haemostasis and hepatic function were normal for a patient that was being treated with oral anticoagulants. Functional and antigenic levels of antithrombin III, protein C and plasminogen were also normal. When total and free protein S levels (method of Comp et al.) were measured using both an electroimmunoassay and an ELISA assay ,almost indetectable levels of free protein S (between 0 and 10%) and very low levels (20%) of total plasma PS, were found. These results were also confirmed by crossed-electriimnunophoretic studies. When the family of this patient was studied it was found that his two sons, aged 15 and 8 years, as well as one of his sisters, aged 35 years, and her daughter of 4 years, were also affected (free PS levels between 38-60% and total PS between 35 and 39%). All these members had been assymptomatic up to now and are not under oral anticoagulants.

Abstract:

Abstract:
Kapahaka is not simply the song and dance of Aotearoa’s Indigenous people. Deeply steeped in mātauranga Māori, kapahaka is a way of simultaneously exemplifying Māori histories, the present, and the future; meanwhile it is a community-focused cultural practice, methodology, and pedagogy. Contemporary kapahaka – both competitive and for entertainment – fosters, develops, validates, and celebrates the Māori world, the language, and our ‘ways’: arguably the fundamental building blocks of Māori ‘popular culture’. The research project Kia Rite! Kapahaka for Screens, from which this presentation is a tiny proportion, will focus on the influence and impact of screen production on the art’s ebbs and flows, and the conflicts between maintaining ‘traditions’ and exploring innovation in and towards the future. Over the last century, the kapahaka art-form has evolved exponentially, and as the wider project will explore, in large part as a response to the advancement of screen technologies. An important strand in Kia Rite! will investigate the kapahaka audience. It employs a refined iteration of Rōpū Whānau, a focus group methodology where closely linked relations will be asked to respond to archival through to contemporary kapahaka footage as a generational screen audience study. Exploring responses to screened kapahaka in this way revisits a whakawhiti kōrero-based audience study method designed to reflect and embody the fundamental whakataukī ‘he aha te kai o ngā rangatira? He kōrero’ (what is the food of chiefs? It is talk.) Rōpū Whānau was developed to move beyond the ‘safety in numbers’ focus group methodology to more of a ‘safety within the whānau’ format. By inviting participants from the same family, a duty to protect the under 18s and inherently control researcher behaviours provides an extra layer of a kind of ‘Māori ethics’. This critical presentation brings forward the fundamental elements of Rōpū Whānau and unpacks how it has been used in various research projects in the past. This is to plot the way forward for Indigenous community-led research methodologies, and encourages the consideration of Indigenous research approaches.

Abstract:

Abstract:
Context: Creutzfeldt-Jakob disease (CJD) is a rapidly progressive neurodegenerative disease. Heidenhain’s variant has isolated visual symptoms that persist even without any cognitive decline for a few weeks. Objective: To report a case of the Heidenhain variant of the CJD with evolution documented. Methods: Case report:A 67-year-old woman was admitted to the emergency room with a report of “seizures and dementia for 1 month”. It all started in August 2020 with a decrease in visual acuity. The son started to record the evolution of the disease in vídeos on his mobile phone. It got worse, with an inability to reach objects and optical apraxia, psychomotor slowing, abnormal repetition of acts and impairment of judgment. MRI of the brain (normal) and EEG was performed, with diffuse slowing of the base activity. It evolved with intermittent myoclonus and abulia. Extensive investigation for encephalitis: normal. Treatment with acyclovir and pulsetherapy with methylprednisolone were performed. Results: new brain MRI: restricted area following the diffusion and T2 / FLAIR hypersignal exclusively cortical involving both posterior hemispheres. Also new EEG: generalized periodic discharges. The 14-3-3 protein was detected in her CSF.The patient died 4 months after the onset of the condition due to infectious complications. Conclusions: The Heidenhain variant of CJD should be considered a differential diagnosis in all patients who have isolated visual complaints, especially if associated with cognitive complaints. The video recording of the patient allows a detailed analysis of the clinical picture and becomes an important diagnostic complementation tool.

Abstract:

Full text
Abstract:
In plasma exchange donation (PED), several liters of fresh plasma are removed fran a donor with a pheresis instrument as a source of cryoprecipitate, and replaced with autologous cryoprecipitate-supernatant from the previous donation. Repetitive PED can produce large quantities of factor VIII from individual donors over time, with a favorable impact on donor exposure for factor VIII recipients. To clarify the implications for donor safety, we report our experience with several donors who have undergone multiple PEDs. Detailed observations are presented for one donor who has undergone PED 101 times between 5/83 and 1/87, and has provided all the factor VIII needed by his son (now age 14) with severe hemophilia A during this period. Exchange volume was gradually increased while donation frequency was gradually decreased. There were 23 exchanges of 2 L, 52 of 2.5 L, and 26 of 3 L for a total of 254 L plasma exchanged. Desmopressin (20 meg tV) was given before 45 more recent donations to augment factor VIII yield. A total of 343,274 IU factor VIII have been collected; the mean (±SD) yield from a 3 L, desmopressin- stimulated PED is 5598 ± 899 IU. The donor has remained in good health; he has noted no adverse effects fran any PED, and none have been found in laboratory monitoring. Prior to the 100th donation the following were within normal limits: CBC,platelet count, urinalysis, SMA-18, protein electrophoresis, IgG, IgA, IgM, hemolytic complement, C3, C4, fibronectin, prothrombin time, partial thromboplastin time, thranbin time, factor VIII:C (140%), factor VIII:Ag (134%), von Willebrand factor (86%) and fibrinogen (215 mg/dL). In another family, the father has donated 40 times since 1981 and the paternal grandmother has donated 31 times since 1984 with no untoward effects detected in clinical or laboratory monitoring. They have supported two moderately affected patients now ages 7 and 9. Extensive experience with these donors suggests that repeated PED is safe, and that a highly motivated donor can sometimes provide single donor support, even for a severe hemophiliac.

Abstract:
This study examines Mozambican popular music to investigate three questions: Are notions of empowerment and accountability present in popular music in Mozambique? If so, what can these existing notions of empowerment and accountability reveal about relations between citizens and state institutions in general and about citizen-led social and political action in particular? In what ways is popular music used to support citizen mobilisation in Mozambique? The discussion is based on an analysis of 46 protest songs, interviews with musicians, music producers and event promoters as well as field interviews and observations among audiences at selected popular music concerts and public workshops in Maputo city. Secondary data were drawn from radio broadcasts, digital media, and social networks. The songs analysed were widely played in the past two decades (1998–2018), a period in which three different presidents led the country. Our focus is on the protest song, conceived as those musical products that are concerned with public affairs, particularly public policy and how it affects citizens’ social, political and economic life, and the relationship between citizens and the state.

Nine North American al organizations contributed semen to the Dairy Bull DNA Repository (DBDR), which currently has 65,743 units from 3366 bulls. Semen was obtained for 31 out of 35 grandsires. Semen of 28 and 23 sons of two Israeli bulls was also collected. Eighteen grandsires were genotyped for 75 microsatellites. One thousand, three hundred and sixty-two sons with evaluation from 17 families were genotyped for 24 markers. Eleven thousand, six hundred and twenty sons genotypes were determined, of which 8,802 were informative. The genotype data was matched to the bulls' daughter yield deviations (DYD) for seven traits; milk, fat, and protein production; fat and protein percent; somatic cell concentration (SCS); and productive herd life. Seven loci had significant effects at p<0.05, but only two loci, TGLA263 and MGTG7, had significant effects at p<0.01, and the effect of TGLA263 on fat percentage was significant at p<0.0001. There was at least one significant effect for each of the seven traits analyzed.

Abstract:
The objectives were to continue collection of semen for the US dairy bull DNA repository, to conduct a systematic search of the Holstein genome for economically significant economic trait loci (ETL), to develop and refine statistical techniques for the analysis of the data generated, and to confirm significant effects by genotyping daughters i Israel and additional US sons. One-thousand-seventy-six sons of eight US grandsires were genotyped for 174 microsatellites located on all 29 autosomes. ETL were detected for milk production traits on seven chromosomes. ETL for milk and fat yield and fat and protein percentage on BTA3 was mapped to between the markers BL41 and TGLA263. The 95% confidence interval for the ETL affecting fat percentage on BTA14 localized this ETL between the contromere and chromosome position 11 cM. This ETL was verified in the Israeli cattle population by genotyping an independent sample of cows from seven families. The radiation hybrid data for the centromeric region of BTA14 is defined by a single linkage group. Order of Type I genes within this region, CYC-FADK-TG-SQLE, is conserved between human and cattle. Thus, HSA8, the human homologue of BTA14, can be used to identify candidate genes for the ETL.

Abstract:
The project’s general objectives were to determine specific polymorphisms at the DNA level responsible for observed quantitative trait loci (QTLs) and to estimate their effects, frequencies, and selection potential in the Holstein dairy cattle breed. The specific objectives were to (1) localize the causative polymorphisms to small chromosomal segments based on analysis of 52 U.S. Holstein bulls each with at least 100 sons with high-reliability genetic evaluations using the a posteriori granddaughter design; (2) sequence the complete genomes of at least 40 of those bulls to 20 coverage; (3) determine causative polymorphisms based on concordance between the bulls’ genotypes for specific polymorphisms and their status for a QTL; (4) validate putative quantitative trait variants by genotyping a sample of Israeli Holstein cows; and (5) perform gene expression analysis using statistical methodologies, including determination of signatures of selection, based on somatic cells of cows that are homozygous for contrasting quantitative trait variants; and (6) analyze genes with putative quantitative trait variants using data mining techniques. Current methods for genomic evaluation are based on population-wide linkage disequilibrium between markers and actual alleles that affect traits of interest. Those methods have approximately doubled the rate of genetic gain for most traits in the U.S. Holstein population. With determination of causative polymorphisms, increasing the accuracy of genomic evaluations should be possible by including those genotypes as fixed effects in the analysis models. Determination of causative polymorphisms should also yield useful information on gene function and genetic architecture of complex traits. Concordance between QTL genotype as determined by the a posteriori granddaughter design and marker genotype was determined for 30 trait-by-chromosomal segment effects that are segregating in the U.S. Holstein population; a probability of <10²⁰ was used to accept the null hypothesis that no segregating gene within the chromosomal segment was affecting the trait. Genotypes for 83 grandsires and 17,217 sons were determined by either complete sequence or imputation for 3,148,506 polymorphisms across the entire genome. Variant sites were identified from previous studies (such as the 1000 Bull Genomes Project) and from DNA sequencing of bulls unique to this project, which is one of the largest marker variant surveys conducted for the Holstein breed of cattle. Effects for stature on chromosome 11, daughter pregnancy rate on chromosome 18, and protein percentage on chromosome 20 met 3 criteria: (1) complete or nearly complete concordance, (2) nominal significance of the polymorphism effect after correction for all other polymorphisms, and (3) marker coefficient of determination >40% of total multiple-regression coefficient of determination for the 30 polymorphisms with highest concordance. The missense polymorphism Phe279Tyr in GHR at 31,909,478 base pairs on chromosome 20 was confirmed as the causative mutation for fat and protein concentration. For effect on fat percentage, 12 additional missensepolymorphisms on chromosome 14 were found that had nearly complete concordance with the suggested causative polymorphism (missense mutation Ala232Glu in DGAT1). The markers used in routine U.S. genomic evaluations were increased from 60,000 to 80,000 by adding markers for known QTLs and markers detected in BARD and other research projects. Objectives 1 and 2 were completely accomplished, and objective 3 was partially accomplished. Because no new clear-cut causative polymorphisms were discovered, objectives 4 through 6 were not completed.

Abstract:

Rights and Resources Initiative, October 2017. http://dx.doi.org/10.53892/vnib5422.

Abstract:
