Resumo: Nosso estudo debruça-se por sobre as canções de Torquato Neto, anteriores a sua participação no Tropicalismo, que tem um perfil semelhante à canção engajada, de protesto, de meados da década de 1960, e durante seu envolvimento com o movimento, tomando-as como uma entrada para o questionamento dos discursos críticos estabelecidos sobre a sua poética. Procuramos estabelecer um diálogo entre a nossa escuta das canções e as leituras críticas que as tomam isoladamente, como texto poético apenas. De modo a questionar os dois pólos extremos em que é confinada sua poética: Ou o poeta é visto enquanto um artista de vanguarda continuador de uma certa tradição do novo, de inventividade e rigor construtivo; ou como romântico libertário radical, que por coerência com seu projeto poético-existencial, renuncia a vida
Abstract: Our study bends over Torquato Neto¿s songs, prior to his participation at the Tropicalism, which has a similar profile of the activist song, of protest, in the middle of the 1960s, and during his involvement with the movement, turning them into a beginning of questioning the established critical discourses about his poetry. We try to establish a dialogue between our listening to the songs and the critical readings which consider them separately, as a poetic text only. So that we can question the two opposite extremes in which his poetry is confined to: either the poet is seen as an avant-garde artist who continues a certain tradition again, of ingenuity and constructive rigidity, or as a romantic radical libertarian who, in accordance with his political-existential project, abandons his life

Full textThis work is a study of contemporary protest song in the USA through the songs and social and political activism of Bruce Springsteen. It contends that Springsteen has been able to reexamine American protest songs and frame a new genre under the umbrella of rock music by adapting his music to the social and political context of contemporary America. The thesis that I defend shows that the reason behind the commercial success of Springsteen can be explained by the fact that he has adopted several artistic strategies which have brought him an audience composed of dedicated fans who admire him. I argue that Springsteen adopts four main strategies: firstly, singing about ordinary people and reporting their daily hardships; secondly, composing somber songs that deal with the despair of blue collars and marginalized Americans; thirdly, adopting mimesis (imitation) which has a cathartic effect on Springsteen’s audience; and finally, getting involved in social and political activism. The thesis contends that Springsteen is among the top selling rock singers in the USA and several other countries thanks to these four strategies. Springsteen has reinvented protest song as a genre and has become a reference in a world where everything is co-opted including the very act of rebellion. I, therefore, suggest a new term, reflective songs, to describe the songs of Bruce Springsteen when they reflect on the condition of the socially excluded instead of protesting pointlessly. Springsteen’s reflective song is undoubtedly the most relevant artistic medium to shed light on the most deprived people of contemporary America. Perhaps, the reflective song of Bruce Springsteen will not change the world, but at least it gives people a reason to hope for a better future

During this study we focused our research on dialogues between the Protest Song of 1960s and 1970s, especially those occurred in certain works created by Gilberto Gil, Caetano Veloso and Chico Buarque de Holanda and a few songs produced by politicized and protest rock launched by bands from Brasilia such as Capital Inicial, Plebe Rude and Legião Urbana in the 1980s, contextualizing this cultural production within an important period in the recent history of Brazil. Through the theory developed by Bakhtin, we were confronted with dialogues between speakers and speeches, which allowed us to produce analysis of the characteristics and content of the dialogues between the "Old" Protest and the "New" Protest. The investigation allowed us to point out some similarities between the songs analyzed, and also enabled us to interpret them as articulating elements of the period in which they arose, which is evident in the performance of songs from both periods as a political tool of contestation.
No decorrer deste estudo, procuramos focar nossa investigação sobre os diálogos estabelecidos entre a Canção de Protesto dos anos 1960 e 1970, em especial, os ocorridos em determinadas obras criadas por Gilberto Gil, Caetano Veloso e Chico Buarque de Holanda e algumas canções produzidas pelo rock politizado e de protesto lançado pelas bandas de Brasília: Capital Inicial, Plebe Rude e Legião Urbana, nos anos 1980. Assim, pudemos contextualizar a produção cultural, dentro desse período importante da história recente do Brasil. Através da teoria desenvolvida por Bakhtin, foram confrontados os diálogos entre interlocutores e discursos, o que nos permitiu produzir análises acerca das características e conteúdos dos diálogos travados entre o Velho Protesto e o Novo Protesto. A pesquisa admitiu apontar algumas convergências entre as canções analisadas, bem como, também, possibilitou-nos interpretá-las como elementos articuladores do período em que surgiram que, por conseguinte, evidencia-nos a atuação das canções de ambos os períodos como instrumento político de contestação.

Full textThe objective of this research is to analyze the Latin America integration project conceived by the artists involved with the protest song in the 1960s and 1970s. In this period, the impact of Cold War resulted in a polarized debate that affected the cultural field, and especially the artistic field. The common political experiences favored the exchange of ideas and the intense movement of artists across the continent. Using the most prominent artists discography as source, the purpose is to analyze three moments of the history of \"transnational connections\" in the protest song in Latin American: a) the first, between 1963 and 1966, time of development and consolidation of nueva canción movement in the Southern Cone, when the engagement was manifested fundamentally by social criticism; b) the second, between 1967 and 1969, marked by the impact of the I Encuentro de la Canción Protesta held in Cuba, that highlighted two important themes: revolution and anti-imperialism; c) the third, which comprises the 1970s, a period marked by the radicalization of political discourse and by the intensification of connections. Especially in this third phase, I analyze the Brazilian artists efforts to integrate the circuits of Latin American protest song.

Full textIt has become imperative in the current context of Brazilian public school the reading practices as an agenda of constant reflection and research for alternatives that can contribute to the development of the reading comprehension in various levels of middle school students, given the Saeb data and Prova Brasil (2014), whose large-scale assessments showed difficulties of the students to select implicit information, inferring the meaning of a word or expression, among others; there by, indicating, serious deviations of understanding and the appropriation of the writing production, in the act of reading. Based on this fact, the present report has as a guiding objective to promote the formation of a proficient and critical reader through the analysis of Brazilian social protest songs in activities of reading and understanding the implicit textual, enabling recognize them as situated discursive practices in daily life that can contribute to the formation of active citizens in society. Was taken as theoretical support the studies of textual linguistics, the sociocultural perspective; using the sociointeractional language design, subject and text for which the textual and discursive understanding can only be understood when it exceeds the limits of decoding linguistic elements and reaches the horizons of mobilization of the socio-historical and cultural knowledge and reconstruction of these within a communication situation in a given context. Therefore, we used a qualitative research, with the methodological approach by the application of didactics sequence in a 9th grade of elementary school, that used the song of social protest genre as mediator. Taking into account the proposal prepared for the research and what was executed, it is possible to say that the understanding of the text work from the songs analysis is configured as a positive and effective alternatives, as the students found new forms of meanings construction for the studied texts. The results obtained from this work fulfilled the ultimate goal of this trajectory that is becoming theoretical and practical support for the development of a concrete tool for socialization of experienced knowledge: the Pedagogical Section, whose main purpose is to share successful experiences with other Portuguese-speaking teachers in teaching and in the reading learning process.
Tornou-se imperativo, no atual contexto da escola pública brasileira, que as práticas de leitura sejam pauta de constante reflexão e da busca por alternativas que colaborem para o desenvolvimento da compreensão leitora dos alunos do ensino fundamental (EF), em vários níveis, haja vista os dados do Saeb e da Prova Brasil (2014), cujas avaliações em larga escala apontaram dificuldades dos discentes para selecionar informações implícitas, inferir o sentido de uma palavra ou expressão, entre outros; indicando, desse modo, os graves desvios de compreensão e apropriação da produção escrita, no ato de ler. Partindo dessa realidade, o presente relatório tem como objetivo norteador promover a formação de um leitor proficiente e crítico por meio da análise de canções brasileiras de protesto social em atividades de leitura e compreensão dos implícitos textuais, possibilitando reconhecê-las como práticas discursivas situadas no cotidiano que contribuem com a formação de cidadãos ativos na sociedade. Tomaram-se como aporte teórico os estudos da Linguística Textual, na perspectiva sociocultural; adotando-se a concepção sociointeracional de língua, de sujeito e de texto para a qual a compreensão textual e discursiva só pode ser entendida quando ultrapassa os limites da decodificação de elementos linguísticos e alcança os horizontes da mobilização dos saberes sociohistórico e cultural e da reconstrução destes no interior de uma situação de comunicação, num dado contexto. Para tanto, adotou-se a pesquisa de cunho qualitativo, tendo como caminho metodológico a aplicação de Sequência Didática em uma turma de 9º ano (EF), que utilizou a canção de protesto social como gênero textual mediador. Levando-se em conta a proposta elaborada para a pesquisa e o que foi executado, é possível dizer que o trabalho de compreensão do texto a partir da análise de canções configurou-se como uma alternativa positiva e eficiente, na medida em que os alunos descobriram novas formas de construção de sentidos para os textos estudados. Os resultados obtidos a partir desse trabalho atendem ao objetivo final dessa trajetória, que é a elaboração de um instrumento concreto de socialização dos conhecimentos teórico-práticos vivenciados: o Caderno Pedagógico, cujo principal propósito é compartilhar experiências exitosas, testadas no ensino e na aprendizagem da leitura, com outros professores de língua portuguesa.

Full texte)Objectives: Delineating a profile of the modal melodic procedures present in the ethnic and popular manifestations of the Northeast region of Brazil in order to verify them in authors connected to the Brazilian popular music from the 1960s, comparing the different harmonic procedures produced. f)Methods: The main method used was the analysis of transcripts of audio samples. From these analyses a series of comparisons with similar materials found in the bibliography was made. g)Results: As a result of this research, the main modes and cadential traces used in the ethnic and popular repertoire chosen from the Northeast of Brazil were indicated. These traces were important in the construction of the songs here selected, characteristics of the so-called MPB of the 1960s presenting, from those analyses, the main harmonic procedures of those authors related to the cadential modes and traces found.

Full textIn 1958, a new musical genre appears in Brazil: the Bossa nova, which looks like a revolution in the traditional musical landscape. Twenty years later, 1978 is the year of political openness, Chico Buarque, poet musician who dominated the decade through his songs as well as his resistance against the millitary dictatorship, launches an album on which he records some songs that were censored until then. This event puts an end to a period of great artistic creativity, punctuated with social and cultural changes, statements of intents, ruptures, standpoints on the political or cultural life of the country, which often are one. These twenty years witnessed the appearance of the most striking figures of Brazil’s 20th century music scene, and of the acronym whereby the urban song gained respectability: MPB, initials of Música Popular Brasileira. Rooted in tradition though defending itself from it, exchanging with outside influences which it influences as well, distancing itself till the rupture of the movements of which it was the closest, today this generation seems to us much more united and limited in time than it thought it was. That’s why the time has come, with historical perspective, to look at the musical production of that period with fresh eyes, in order to define the characteristics that unite and distinguish it at the same time
1958, nasce no Brasil un gênero musical novo, a Bossa Nova, que se anuncia comme uma revolução na paisagem musical. Vinte anos depois, 1978, é o ano da "Abertura" política, Chico Buarque, poeta músico que dominou a década tanto pelas suas canções como pela sua atitude de resistência contra a ditadura, lança um disco no qual ele grava músicas até então proibidas pela censura. Esse acontecimento conclui um período de intensa criatividade artística, acompanhada por grandes mudanças sociais e culturais, declarações de intenções e rupturas, tomadas de posição em relação com a vida política ou cultural do país, que muitas vezes faz uma só. Esses vinte anos viram emergir as figuras mais marcantes da canção brasileira do século XX, concomitantemente à sigla pela qual a canção urbana moderna foi consagrada : MPB, iniciais de Música Popular Brasileira. Ancorando-se na tradição sem reivindicá-lo, dialogando com as influências externas que ela influencia por sua vez, demarcando-se até à ruptura dos movimentos que lhe eram próximos, essa geração nos aparece hoje muito mais unida e circunscrita no tempo do que ela o percebia. Por essa razão, o tempo veio, com o recuo histórico, de questionar com um olhar novo a produção musical daquela época, para definir as características que a unificam e distinguem ao mesmo tempo

Full textThe aim of the present work is to analyze the movie Im Not There (USA, 2007), by the American director Todd Haynes. The analysis will have as its main goal pinpointing the central theme of the movie, which portrays the life and career of Bob Dylan through six very distinct characters. Therefore, three characters will be thoroughly observed, the ones more closely connected to the musical universe, namely the boy who presents himself as Woody Guthrie, Jack Rollins and Jude Quinn. The other three characters (Robbie Clark, Arthur Rimbaud and Billy the Kid) will be discussed insofar as they raise relevant issues for understanding the film. This analysis leads to the conclusion that the main topic in Im Not There is the culture industry. It is thus possible to see in which way the movie deals with this issue and how the theme works as a starting point for a broader discussion concerning the social and political panorama of the 1960s in the United States. Finally, it is necessary to debate the importance of a contemporary movie having as its topic the 1960s and how Im Not There itself, as a cultural object, relates to the discussion it proposes regarding the culture industry

Full textThis research joins Costaâs (2001) conception that the literary and musical speech as a discursive practice serves as a reference to the subjects, legitimizing truths and spreading ways how those subjects interact with reality, with other subjects and with themselves. Our association with Costasâs mind increased further when the objective of our project was defined as the engaged songs and the songs of protest and neoprotest of the speech community of rock80, called Brock, a musical moment with great integration of youth, when the amused discourse of the rockers served as interaction and was confused with the copyright word of those young people, changing their ideas and the way they thought, felt and acted. The theory that guided our paper was the Speech Analysis by French line, the theoretic material provided by Maingueneau (2001). Our research set out to investigate the following categories ethos, set design and linguistics code in the speech of those songs, taking as reference the seminal engagement of the protest line which was in the songs of the sixties, choosing original songs of Geraldo VandrÃ, ZÃ KÃti e SÃrgio Ricardo as emblem. In this paper, we proved the involvement of rock songs of eighties observing the linguistics materiality inside the code, at the capture of its ethos by those speakers, which was reflected in the construction of the scenery. The research clarified and confirmed our initial position about the existence of marks of protest in both the gestures: the literary and musical community of the sixties and the rock tribe of eighties, as well. The first one left its trademark in an analog, black and white speech, whereas the last one left its speech in a colorful, digital format. Using a visual media language, only the formats were subservient to the protest of those songs.

Full textThe recycling to the plasma membrane of receptors and lipids is tightly regulated and is essential for PM homeostasis, adhesion and cell migration. It requires small GTPase Rab proteins and their effectors. The MICAL-L1 protein, an effector of several Rabs including Rab 8, 11, 13 and 35, has been shown to play an important role in the recycling. Here, we report a novel interaction between MICAL-L1 and the BAR domain containing protein PACSIN3/Syndapin3 that contributes to generate tubular recycling endosomes. MICAL-L1 is required for the localization of PACSIN3 to endosomal membranes. Importantly, disruption of MICAL-L1/PACSIN3 interaction promotes the transferrin receptor (TfR) delivery back to the plasma membrane. The MICAL-L1/PACSIN3 complex accumulates in elongated tubules that contain transferrin carriers. The dynamic of transferrin positive endosomes segregation from MICAL-L1/PACSIN3 tubules suggests that MICAL-L1/PACSIN3 complex controls TfR recycling endosomes delivery to the plasma membrane. Our data provide novel mechanistic insights on the dynamical regulation of the plasma membrane recycling pathway

This qualitative study utilized a focus group setting to gather information. Within two weeks of the focus group 6 participants withdrew, the focus group ultimately included 6 participants. Participants voiced their need to educate their sons about police, Blackness being a risk factor, the emotional burdens of ensuring safety, strategies used to promote safety in potential police interactions, and possible solutions to strained community police relations. Participants developed strategies to increase safety by asking their sons to be compliant, stay under the radar, utilize family support, recreational and community resources. While there was a lack of confidence and mistrust of the police, participants maintained hope that with training and positive community interactions there can be improvement in safety during police interactions.

Full textThe current growth in the number of known 3-dimensional proteins structures is producing a database of structures that is very useful as starting point for the development of new medically relevant molecules such as drugs and vaccines. In this work part of the project of developing new drugs against the sleeping sickness has been done based on crystallographic, ligand-binding and molecular modeling studies of two glycolytic glycosomal enzymes from T. brucei. As the target for new drugs is the glycolysis pathway, which is present also in the human being, the new ligand must be selective. Fructose-1,6-bisphosphate aldolase (ALDO) is one of the target protein for the rational drug design. The structural analysis and crystallographic studies of ALDO from T. brucei have been done, from overexpressed protein. Several possible sites for selective inhibitor were found in T. brucei ALDO when compared with the three human isoenzyme sequences, based on the human ALDO A structure. The flexible C-terminal arm is a promising target for selective compounds, due to the non-conserved sequence. Crystals were grown, and X-ray diffraction was obtained up to 3.0Å, but still the crystals are small and show radiation damage. Triosephosphate isomerase (TIM) is the other target protein analyzed in this work. The structure of TIM from T. brucei is already known at high resolution. Crystals from overexpressed protein were obtained after extensive experiments. It was identified that the use of fresh protein is essential to grow good diffracting crystals. After obtaining crystals from overexpressed protein, it has been done soaking of new inhibitors, diffraction data collection and analysis of the structures of the possible complexes. The well known flexible loop in TIM closes upon the binding of either the substrate or an inhibitor analogue to the substrate in the active site. Rational drug design has been followed based on the \"closed\" conformation of the flexible loop. Nevertheless, with a new complex structure, in which the flexible loop adopts the \"open\" conformation, it has been done a search for new compounds in the database, non-derived from phosphate and phosphonate, through the program DOCK. Based on this procedure it was proposed new lead compounds which will be used on the rational drug design cycle. The structure of the thermostable triosephosphate isomerase from Bacillus stearothermophilus in complex with the competitive inhibitor 2PG was determined by X-ray crystallography to a resolution of 2.8Å. The structure was solved by molecular replacement using XPLOR. Two fold averaging and solvent flattening were applied to improve the quality of the map. Both of the active sites were occupied by the inhibitor and the flexible loop adopts the \"close\" conformation in both subunits. The crystallographic R-factor is 17.6% with good geometry. Bacillus stearothermophilus is considered a moderate thermophile organism. There are already five triosephosphate isomerase structures available from mesophilic organisms. The structure reported here is the first thermostable TIM reported. It has been conducted in this work a comparison between the thermophilic and the mesophilic protein structures available. Several reports had listed thermostable factors, which have been analyzed in all TIM structures available, from which it has been concluded that the hydrophobic interaction upon the dimmer formation and the higher number of proline residues are the most important factors that contribute to the thermostability of B. stearothermophilus TIM. Also it has been concluded that the ratio Arg/(Arg+Lys) is increased in B. stearothermophilus TIM more due to lower number of lysine instead of a higher number of arginine residues. Arginines have been analyze in B. s,tearothermophillis TIM as well as the interactions of arginines and lysines in all other TIM structures and it has not been possible to relate the increased ratio Arg/(Arg+Lys) with thermo stability, which was previously reported as important in thermostability

A hipoxia e caracterizada pela reducao de oxigenio no sangue e esta relacionada a diversas patologias, entre elas a Sindrome da Apneia Obstrutiva do Sono (SAOS). Suas consequencias ainda nao estao completamente esclarecidas, mas evidencias da literatura demonstram que a hipoxia induz alteracoes bioquimicas com reflexos em aspectos comportamentais e importantes consequencias em diversos sistemas, como o sistema nervoso central, respiratorio e cardiovascular. A hipoxia e a idade apresentam uma intima correlacao com o desenvolvimento da Doenca de Alzheimer. A neprilisina (EC 3.4.24.11) e uma metaloprotease presente em diversos tecidos como rins, intestino, pulmao, cerebro, entre outros. Ja esta demonstrado que uma das principais atividades da enzima no cerebro e a degradacao do peptideo β amiloide, um dos principais responsaveis pelo surgimento e agravamento da Doenca de Alzheimer. Assim, nosso objetivo foi avaliar os efeitos da hipoxia intermitente, um modelo de SAOS, na expressao e atividade da neprilisina e na memoria e aprendizado dos animais. Metodos: Ratos machos wistar foram separados em 3 grupos: Controle, Hipoxia e Rebote. Os grupos hipoxia intermitente e rebote foram submetidos ao protocolo de hipoxia intermitente (HI-ciclos de 3 min de 21% a 5% de O2) das 8 as 16h, por seis semanas. Apos a HI, o grupo rebote foi mantido por 2 semanas em condicoes de normoxia. Realizou-se os ensaios do Labirinto aquatico de Morris (LAM), de RT-PCR, Western blot e de atividade enzimatica para neprilisina e para proteina precursora amiloide (APP) nas regioes cerebrais do hipocampo, cortex frontal e temporal. Resultados: A HI nao promoveu alteracoes na memoria e aprendizado dos animais, avaliados por LAM. Os ensaios de RT-PCR, Western blot e atividade enzimatica demonstraram um aumento da enzima seletivamente na regiao do cortex temporal. A expressao proteica da proteina precursora amiloide se manteve inalterada. Conclusoes: Apesar da HI nao afetar o desempenho dos animais no LAM, houve o aumento da expressao e atividade da enzima seletivamente no cortex temporal, uma area relacionada a cognicao, podendo levar a alteracoes ainda nao esclarecidas

Full textCaveolin-1, a 21 kDa membrane protein, is the principal membrane protein ofcytoplasmic membrane domains named caveolae. Specifically enriched incholesterol and sphingolipids those domains are important in many aspect of the cell's life and make up a proteins and lipids interaction platform. In the past years, despite a large number of publications stating the implication of caveolin-1 or caveolae in many cell processes and pathologies, very few is known about theway this protein is organized at the cell membrane. Hence, the main purpose of thiswork is to contribute to the acquisition of structural data on this protein. At the base of this work is the heterologous expression within a bacterial host, and as a fusion protein, of the canin beta isoform of cavéolin-1 or one of its fragment. These expressions lead to the formation of cytoplasmic vesicles composed mainly ofthe expressed protein. Thence, the first part of this work focus on developping a method to purify those vesicles that does not rely on using any kind of detergent which could enable structural studies in a native environnment. The second part present a potentiel application of those vesicles and inparticular the use of those vesicles to characterize a membrane enzyme, namelythe murin microsomal glutathion-Stransferase. The last part will be a contribution to data analysis within the context of molecular dynamics simulationof membraneous systems

Full textIDPs are proteins devoided of a unique and stable 3D structure in solution and in the absence of their partners. Those proteins possess unique properties, as well as mechanisms of interaction with their partners.The C-terminal region of the nucleoprotein of measles virus, NTAIL, is an IDP and interacts with XD, the globular C-terminal domain of the viral phosphoprotein, via the box2 region that is an (alpha-MoRE). This interaction allows to recruit the L protein in order to form the viral polymerase.The aim of my work was to characterize the molecular basis of NTAIL-XD interaction. I was able to show that the contribution of the amino acids among box2 depends on the orientation of their lateral chain, and that the substitution of one single amino acid has drastic effect upon the viral replication.IDPs keep a non-negligible amount of residual disorder among the complex. This fuzziness can have multiple forms, like the presence of fuzzy regions from either side of the MoRE. The impact fuzzy regions have within the complex is not well known. We demonstrated that the fuzzy region upstream box2 inhibits the settlement of the interaction between NTAIL and its partners.When interacting with their partners, IDPs generally undergo a folding associated with binding that can take place either before or after the interaction. The interaction between NTAIL and XD was investigated and monitored by fluorescence kinetic measurements, using variants bearing a tryptophan substitution. We concluded without any doubt that NTAIL folds under an induced folding mechanism.Those studies together contribute to enlighten the molecular mechanisms that govern partner recognition by the IDPs

Full textThe spermatozoon is a highly specialized cell that must be able to perform specific functions tofertilize the oocyte. It must be able to perform the acrosome reaction, an exocytosis of a giant vesicleof secretion, attached to the nucleus. This vesicle contains enzymes that allow the sperm to penetratethe zona pellucida surrounding the oocyte. The aim of my work was first to study the effect of anenzyme present in the acrosome, the sPLA2 of group X in mouse (mGX). This is the onlymammalian phospholipase among the five tested that has an inhibitory effect on sperm specificpopulation with low mobility. My work has confirmed the specificity of this phospolipase on theregulation of sperm physiology. Second, I participated in the discovery of the gene DPY19L2involved in male infertility, globozoospermia. The globozoospermia is characterized by round headspermatozoa without acrosome. DPY19L2 gene is specifically expressed in the testis and is absent in80% of globozoospermic patients. I then identified the role of this protein, which is involved in theattachment of the acrosome to the nucleus. I showed that this protein belongs to the inner nuclearmembrane where it likely interacts with the protein sun5 which also belongs to the inner nuclearmembrane and whose expression is specific to spermiogenesis. Sun5 is involved in the complexformation, called LINC that connects the cytoskeleton to the nucleoskeleton lamina. The role ofDpy19l2 could help stabilizing the anchoring of protein sun5 in order to transmit the forces exertedby the cytoskeleton to the nucleus of the spermatid during acrosome spreading. Dpy19l2 belongs to aprotein family containing 4 members, Dpy19l1 to l4, which has not been poorly studied so far. Arecent study shows that the knock-down of Dpy19l1 resulted in defective glutamatergics neuronsmigration on the radial glia. The results obtained during my work would improve the knowledge ofCytoskeleton-nucleoskeleton interaction, and give new insight on this new family of proteins

This research joins Costa’s (2001) conception that the literary and musical speech as a discursive practice serves as a reference to the subjects, legitimizing truths and spreading ways how those subjects interact with reality, with other subjects and with themselves. Our association with Costas’s mind increased further when the objective of our project was defined as the engaged songs and the songs of protest and neoprotest of the speech community of rock80, called Brock, a musical moment with great integration of youth, when the amused discourse of the rockers served as interaction and was confused with the copyright word of those young people, changing their ideas and the way they thought, felt and acted. The theory that guided our paper was the Speech Analysis by French line, the theoretic material provided by Maingueneau (2001). Our research set out to investigate the following categories ethos, set design and linguistics code in the speech of those songs, taking as reference the seminal engagement of the protest line which was in the songs of the sixties, choosing original songs of Geraldo Vandré, Zé Kéti e Sérgio Ricardo as emblem. In this paper, we proved the involvement of rock songs of eighties observing the linguistics materiality inside the code, at the capture of its ethos by those speakers, which was reflected in the construction of the scenery. The research clarified and confirmed our initial position about the existence of marks of protest in both the gestures: the literary and musical community of the sixties and the rock tribe of eighties, as well. The first one left its trademark in an analog, black and white speech, whereas the last one left its speech in a colorful, digital format. Using a visual media language, only the formats were subservient to the protest of those songs
Essa pesquisa se filia à concepção de COSTA (2001) de que o discurso literomusical enquanto prática discursiva serve de referência para os sujeitos, como legitimador de verdades e disseminador de maneiras de como esses sujeitos interagem com a realidade, com os outros sujeitos e com eles próprios. Essa nossa associação ao pensamento de Costa(2001) recrudesce ainda mais posto que o escopo de nosso projeto foram as canções engajadas, de protesto, ou neoprotesto da comunidade discursiva do rock80, o chamado Brock, momento musical de grande inserção no meio da juventude, onde o discurso alheio dos rockeiros serviu de alteridade e em muito se confundiu com a palavra autoral dessa juventude lhe modificando idéias, maneira de pensar, sentir e agir. A teoria de base que norteou o nosso trabalho foi a Análise do Discurso de linha francesa, material teórico fornecido por Dominique Maingueneau(2001). Nossa pesquisa se propôs a investigar as categorias ethos, cenografia e código lingüístico no discurso dessas canções, tendo como referencial o engajamento seminal da linha de protesto das canções da década de sessenta, escolhendo canções matrizes emblemáticas do cancioneiro de Geraldo Vandré, Zé Kéti e Sérgio Ricardo. Comprovamos o engajamento das canções rockeiras de 80 via materialidade lingüística expressa no código, na captação do ethos de seus enunciadores quando da construção da cenografia. A pesquisa esclareceu e comprovou nossa posição inicial acerca da existência das marcas de protesto nos dois gestos enunciativos: tanto na comunidade literomusical dos anos sessenta quanto na tribo rockeira de 80, aquela deixando sua marca registrada em preto e branco, de forma analógica, e esta pontuando no seu discurso o colorido e o formato digital. Usando uma linguagem midiática visual, apenas os formatos é que se emprestaram subservientes ao protesto das canções

Full textArf1 is a small GTPases, essentially involved in the vesicular traffic. Arf1 switch between two conformations, an active form bound to GTP and an inactive form bound to GDP. Arno is one of the exchange factors (GEF) that can activate Arf1, through its catalytic Sec7 domain, promoting the exchange of GDP by GTP. Activated in breast cancer cells, Arf1 plays an important role in the migration and proliferation of cancer cells.The aim of my thesis was the study and the modulation of the interaction between small G proteins and their GEFs, more precisely the Arf1-Arno interaction. My work has been planned around two axes: (1) the study of the interaction between Arf1 and Arno, and its modulation with a known inhibitor Brefeldin A (BFA). (2) The development of a rational strategy for designing inhibitors of protein-protein interaction for the Arf1-Arno complex.In the first part of my PhD work, we set up a Surface Plasmon Resonance (SPR) method allowing to determine the kinetic parameters of the interaction between Arf1 and Arno. We also studied the effects of allosteric partners such as GDP, GTP and Mg2+ as well as the known uncompetitive inhibitor (Brefeldin A). This SPR approach allowed a very informative analysis at qualitative and quantitative levels of the various complexes taking place during the exchange reaction that should help to solve the inhibitory mechanism for the known inhibitors reported in the literature. In the second part of my thesis, we propose a strategy for targeting the interaction between Arf1and Arno. This approach is based on virtual screening of fragments at hotspot regions. Using biophysical techniques such fluorescence techniques, SPR, NMR and X-Ray crystallography, we identified and validated Hits, showing by crystallographic structural data their modes of interaction with the target protein Arno. A fluorescence polarization test was also developed to identify false positive fragments to eliminate promiscuous aggregators. Taken together, our work proposes a method based on SPR allowing the study of known inhibitors of GEFs, understanding at molecular level their mode of action. We also propose a general strategy for finding Hit fragments that designing competitive inhibitor of the interaction small G protein with its GEFs, that can be the scaffold for designing more powerful inhibitors

Full textABCs (ATP binding cassettes) transporters constitute a large family of transmembrane proteins present in all organisms. ABC transporters hydrolyze ATP to translocate an immense quantity of amphiphilic substrates, such as lipids, steroids, peptides... Some ABCs confer a multiresistance cellular phenotype to drugs from bacteria against antibiotics to humans against anticancer agents, antivirals...A fundamental question for understanding drug transport at the molecular level is how the properties of membranes modulate the function and spatial temporal organization of ABCs. We studied in detail these coupling with BmrA, a bacterial ABC of B. subtillis using different in vitro membrane systems and different biochemical and membrane biophysical approaches. Firstly, after expression and purification of proteins in detergent, we characterized the hydrolysis of ATP of BmrA according to its membrane environment, solubilized in detergent micelles and in mixed lipid/detergent micelles. Proteoliposomes were characterized according to protein orientation, incorporation rate, size and lamellarity. This allowed us to modulate in a controlled manner lipid composition, protein density and conformation and membrane curvature to quantitatively determine the respective contributions of these membrane parameters. Thus, we show that ATP hydrolysis is sensitive to lipid specificity when the protein is embedded in a bilayer. This lipid specificity is provided by negative lipids and phosphatidylethanolamine type lipids that synergistically stimulate hydrolytic activity. However, ATP hydrolysis was decreased in high positive membrane curvature. Secondly, we determined the conditions of reconstitution of BmrA in Giant Unilamellar Vesicles, which then allowed our collaborator to study the respective roles of membrane curvature and tension in the spatial organization of BmrA. Nanotube pulling experiments performed in collaboration show that BmrA has a strong preference for highly curved membrane regions leading to protein cluster formation and that this preference varies according to the catalytic state of the protein. Finally, we developed a method to study the dynamics of NBDs by Förster resonance energy transfer at the single molecule level in reconstituted system via fluorescence cross-correlation spectroscopy.The data set suggest that spatial organizations of ABC transporters in bacterial and eukaryotic cells are different with the possibility of sorting during membrane remodeling of eukaryotic membranes in areas of strong membrane curvatures but without significant change in function

Full textHIV-1, the causative agent of AIDS, is a retrovirus that infects immune cells and hijacks their cell machinery to achieve rapid replication. In the course of infection, the RNA genome is reverse transcribed into DNA by the viral Reverse Transcriptase (RT) before the obligatory insertion of the proviral genome into the host cell DNA catalyzed by the viral enzyme Integrase (IN). The interaction of IN with its essential cofactor, the nuclear protein LEDGF/p75, targets integration within gene introns in highly transcribed chromatine regions, which allows efficient production of new virions. IN-LEDGF Allosteric Inhibitors (INLAIs) are a novel class of antiretroviral molecules binding IN at the LEDGF/p75-binding site. Designed to competitively inhibit IN-LEDGF/p75 protein-protein interaction, they are also capable of inhibiting IN enzymatic activities and raising the IN multimerization level.We studied several new INLAI series from the company Mutabilis. We could demonstrate that these molecules inhibit integration, but also have a more potent, LEDGF-independent, antiretroviral effect during virion maturation, resulting in the production of non-infectious virions. Virions produced upon INLAI treatment have an eccentric morphology characterized by an encapsidation defect of the viral genome, and lead to an infection block at reverse transcription. Yet, we showed that these virions package a stable and functional viral genome, an active RT and the tRNALys3 primer for reverse transcription, and also keep their immunoreactivity towards B- and T-cell lymphocytes. When evaluating the influence of polymorphism at the edge of the binding site, we identified the IN Ala125 polymorphic variant which causes the concomitant loss of MUT-A effect on virion maturation and IN multimerization, whereas inhibition of integration and IN-LEDGF interaction are maintained. This proves that INLAIs exert their late stage effect through induction of IN multimerization. We could associate IN multimerization to INLAI-induced dimer destabilization by analyzing MUT-A co-structures with polymorphic integrases. Full textHemophilia is a disease caused by the lack of Factor VIII (FVIII), leading to insufficient thrombin production, and therefore bleeding. Recently, new therapeutic strategies for hemophilia treatment, that do not rely on clotting factor replacement, have emerged. We propose an innovative approach consisting in targeting a natural and potent thrombin inhibitor, expressed by platelets and called protease nexin-1 (PN-1). Our results demonstrated that blocking PN-1 increased, in vitro, thrombin generation in plasma from patients with hemophilia, and reduced blood loss in hemophiliac mice. Our study provides proof-of-concept that PN-1 neutralizing can be a a novel approach for future clinical care in hemophilia. The potential role of PN-1 in regulating inflammatory processes is not known. Vascular cells, platelets and inflammatory cells express PN-1, but no data are available concerning its expression and potential function in neutrophils. For the first time, we demonstrated the presence of PN-1 in neutrophils. Our data have shown that neutrophil recruitment was much less important in peritoneal cavity of PN-1-/- mice than in those of WT mice.
