Abstract:
A new species of Theraphosidae, genus Psalmopoeus Pocock, 1895 is described from the Chocó region of Ecuador, Psalmopoeus pristirana n. sp. (male and female). The identity of the female of the recently described Psalmopoeus satanas Peñaherrera-R. & León-E., 2023 is discussed. Images of Psalmopoeus ecclesiasticus Pocock, 1903 are given for comparison and a distribution map of all Ecuadorian Psalmopoeus species is presented.

Abstract:
The genera Psalmopoeus Pocock, 1895, Tapinauchenius Ausserer, 1871 and Pseudoclamoris Hüsser, 2018 are revised and cladistics analyses carried out including most species of these genera. In order to test the monophyly of Aviculariinae and Psalmopoeinae, species of all genera in these two subfamilies were included, as well as of Harpactirinae, Selenocosmiinae, Theraphosinae, Stromatopelmatinae, Ischnocolinae, Schismatothelinae, Poecilotherinae, and a barychelid was used to root the cladogram. The matrix with 50 terminal taxa, 1 continuous and 85 discrete characters was analysed with TNT 1.5. The results show a monophyletic Psalmopoeinae as sister group of Aviculariinae. Psalmopoeus comprises 9 species: Psalmopoeus cambridgei Pocock, 1895 (type species), Psalmopoeus ecclesiasticus Pocock, 1903, Psalmopoeus emeraldus Pocock, 1903, Psalmopoeus irminia Saager, 1994, Psalmopoeus langenbucheri Schmidt, Bullmer & Thierer-Lutz, 2006, Psalmopoeus plantaris Pocock, 1903, Psalmopoeus pulcher Petrunkevitch, 1925, Psalmopoeus reduncus (Karsch, 1880), and Psalmopoeus victori Mendoza, 2014. Psalmopoeus intermedius Chamberlin, 1940 is considered a junior synonym of P. reduncus. Psalmopoeus copanensis Gabriel & Sherwood, 2020, P. sandersoni Gabriel & Sherwood, 2020 and P. petenensis Gabriel & Sherwood, 2020 are considered junior synonyms of P. victori. Psalmopoeus maya Witt, 1996 is considered nomen nudum. Tapinauchenius comprises 8 species: Tapinauchenius plumipes (C. L. Koch, 1842) (type species), Tapinauchenius sanctivincenti (Walckenaer, 1837), Tapinauchenius latipes L. Koch, 1875, Tapinauchenius brunneus Schmidt, 1995, Tapinauchenius cupreus Schmidt & Bauer, 1996, Tapinauchenius polybotes Hüsser, 2018, Tapinauchenius rasti Hüsser, 2018, and T. gretae n. sp. The female of T. brunneus is described for first time. Tapinauchenius violaceus (Mello-Leitão, 1930), T. purpureus Schmidt, 1995, T. concolor (Caporiaccco, 1947), and T. gigas Caporiacco, 1954 are considered junior synonyms of T. plumipes. With the synonymy of T. gigas (type species of Pseudoclamoris Hüsser, 2018), it was necessary to describe a new genus for the two species formerly included in it. Thus, the new genus Amazonius n. gen. is erected to include A. elenae (Schmidt, 1994) n. comb., A. burgessi (Hüsser, 2018) n. comb. as well as two new species A. giovaninii n. gen. n. sp. and A. germani n. gen. n. sp. A discussion on the relationship of Psalmopoeinae and Aviculariinae and maps with the distributions of all species are provided.

Abstract:
Two novel species of Psalmopoeus Pocock, 1895 are described from the north-western and central-western slopes of the Cordillera Occidental of the Andes mountain range in Ecuador. The new species are easily differentiated from other congeners of Psalmopoeus by spermathecae and male palpal bulb morphology and a comparatively distant distribution to the type localities of the geographically nearest known congeners. The diagnosis of P. ecclesiasticus Pocock, 1093 is revised and updated, considering the novel species and observations on spermatheca of this species. Likewise, an evaluation is provided for the new species in terms of conservation due to the various threats impacting ecosystems and ecosystem services of their type localities. Finally, the importance of theraphosid spiders in Ecuador and South America and their possible conservation requirements are discussed and assessed.

The tarantula genus Ephebopus Simon 1892 is reviewed and includes the type species, E. murinus (Walckenaer 1837), and E. uatuman Lucas, Silva &amp; Bertani 1992, E. cyanognathus West &amp; Marshall 2000, E. rufescens West &amp; Marshall 2000 and Ephebopus foliatus, sp. nov., from Guyana. Ephebopus violaceus Mello-Leitão 1930 is transferred to Tapinauchenius Ausserer, where it is a senior synonym of Tapinauchenius purpureus Schmidt 1995 new synonymy. Ephebopus fossor Pocock 1903 is considered a nomen dubium. Ephebopus occurs in northeastern South America where it is known only from Brazil, Guyana, Suriname, and French Guiana. Spiders of the genus are generally fossorial; however, Ephebopus murinus has a developmental stage that is arboreal. A cladistic analysis of the Theraphosidae retrieves the Aviculariinae as monophyletic, including Avicularia Lamarck, Iridopelma Pocock 1901, Pachistopelma Pocock 1901, Tapinauchenius, Psalmopoeus Pocock, Ephebopus, Stromatopelma Karsch and Heteroscodra Pocock, having as a synapomorphy the well-developed scopulae on tarsi and metatarsi I–II that is very laterally extended.

Abstract:
Psalmopeotoxin I (PcFK1) is a 33-residue peptide isolated from the venom of the tarantula Psalmopoeus cambridgei. This peptide specifically inhibits the intra-erythrocyte stage of Plasmodium falciparum in vitro. It contains six cysteine residues forming three disulfide bridges and belongs to the superfamily of natural peptides containing the inhibitor cystine knot (ICK) fold. We produced the wild-type and mutated forms of the recombinant peptide to examine the mechanism of action of PcFK1. The purified toxins were consistently produced as two isobaric peptides (r-PcFK1-1 and r-PcFK1-2) with different retention properties but identical anti-plasmodial biological activity. Comparison of 15N-NMR heteronuclear single quantum correlation spectra revealed that although rPcFK1-1 was highly structured, rPcFK1-2 does not have a stable three-dimensional structure. We used high-energy collision-induced fragmentation of the peptides with a matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometer to further investigate the structure of the native peptides in its natural form and produced in E. coli. The fragmentation spectra of the native peptides were very complex due to the occurrence in the spectrum of ions resulting from (1) cross-linking of fragments through a disulfide bridge and (2) asymmetric fragmentations of the disulfide bridges and (3) multiple neutral losses. The tandem mass spectrometry fragmentation pattern of r-PcFK1-1 was similar to that of the natural peptide isolated from crude venom, but r-PcFK1-2 had a clearly distinct fragmentation pattern, more closely resembling the fragmentation spectra of reduced and alkylated peptides. Observed ions could be attributed to specific fragments by comparing spectra between the wild-type and selected variants with point mutations (Y11W, R20T, Y26W, K28V). The disulfide connections in r-PcFK1-2 differed from those of the native peptide and showed a rare disulfide bridge between vicinal cysteine residues. The r-PcFK1_(R20T) variant showed a very limited fragmentation pattern when analyzed in positive mode but displayed much more fragmentation in negative mode pointing out the importance of the R20 residue in the fragmentation of PcFK1. Using the reductive matrix 1,5-diaminonaphtalene promoted strongly in source decay fragmentation of the peptides in MS mode. Our findings illustrated the critical role of the electronic environment around the central Cys18–Cys19 doublet in PcFK1 in internal fragmentation of the peptide.

The genus Avicularia Lamarck, 1818 is revised and all species are rediagnosed. The type species, described as Aranea avicularia Linnaeus, 1758, is the oldest mygalomorph species described and its taxonomic history is extensive and confusing. Cladistic analyses using both equal and implied weights were carried out with a matrix of 46 taxa from seven theraphosid subfamilies, and 71 morphological and ecological characters. The optimal cladogram found with Piwe and concavity = 6 suggests Avicularia and Aviculariinae are monophyletic. Subfamily Aviculariinae includes Avicularia Lamarck, 1818, Typhochlaena C. L. Koch, 1850, Tapinauchenius Ausserer, 1871, Stromatopelma Karsch, 1881, Ephebopus Simon, 1892, Psalmopoeus Pocock, 1895, Heteroscodra Pocock, 1899, Iridopelma Pocock, 1901, Pachistopelma Pocock, 1901, Ybyrapora gen. n., Caribena gen. n., and Antillena gen. n. The clade is supported by well-developed scopulae on tarsi and metatarsi, greatly extended laterally. Avicularia synapomorphies are juveniles bearing black tarsi contrasting with other lighter articles; spermathecae with an accentuated outwards curvature medially, and male palpal bulb with embolus medial portion and tegulum's margin form an acute angle in retrolateral view. Avicularia is composed of twelve species, including three new species: Avicularia avicularia (Linnaeus, 1818), Avicularia glauca Simon, 1891, Avicularia variegata (F. O. Pickard-Cambridge, 1896) stat. n., A. minatrix Pocock, 1903, Avicularia taunayi (Mello-Leitão, 1920), Avicularia juruensis Mello-Leitão, 1923, Avicularia rufa Schiapelli &amp; Gerschman, 1945, Avicularia purpurea Kirk, 1990, A. hirschii Bullmer et al. 2006, Avicularia merianae sp. n., A. lynnae sp. n., and A. caei sp. n.. Avicularia species are distributed throughout Mexico, Costa Rica, Panama, Trinidad and Tobago, Venezuela, Guyana, Suriname, French Guiana, Colombia, Ecuador, Peru, Bolivia, and Brazil. Three new genera are erected to accommodate former Avicularia species: Caribena gen. n., composed of Caribena laeta (C. L. Koch, 1842), comb. n. and Caribena versicolor (Walckenaer, 1837), comb. n.; Antillena gen. n., with a single species, Antillena rickwesti (Bertani &amp; Huff, 2013), comb. n., both from the Caribbean; and Ybyrapora gen. n., composed of Ybyrapora sooretama (Bertani &amp; Fukushima, 2009), comb. n., Ybyrapora gamba (Bertani &amp; Fukushima, 2009), comb. n. and Ybyrapora diversipes (C. L. Koch, 1842), comb. n. from Brazilian rainforest. The subspecies Avicularia avicularia variegata F. O. Pickard-Cambridge, 1896 is elevated to species status, resulting in the combination Avicularia variegata (F. O. Pickard-Cambridge, 1896) stat. n.. The following new synonymies are established: Avicularia velutina Simon 1889, Avicularia exilis Strand, 1907, Avicularia ancylochyra Mello-Leitão, 1923, Avicularia cuminami Mello-Leitão, 1930, and Avicularia nigrotaeniata Mello-Leitão, 1940 are junior synonyms of A. avicularia; Avicularia bicegoi Mello-Leitão, 1923 is a junior synonym of A. variegata stat. n., and Avicularia urticans Schmidt, 1994 is a junior synonym of Avicularia juruensis Mello-Leitão, 1923.
