Abstract:
Ptaquiloside (PTA) is a carcinogenic compound naturally occurring in bracken ferns (Pteridium aquilinum). It is highly water soluble and prone to leaching from topsoil to surface and groundwaters. Due to possible human exposure via drinking water, PTA is considered as an emerging contaminant. We present a sensitive and robust method for analysis of PTA and its degradation product pterosin B (PtB) in groundwater. The method comprises two steps: sample preservation at the field site followed by sample pre-concentration in the laboratory. The preservation step was developed by applying a Plackett–Burman experimental design testing the following variables: water type, pH, filtering, bottle type, storage temperature, transportation conditions and test time. The best sample preservation was obtained by using amber glass bottles, unfiltered solutions buffered at pH 6, transported without ice, stored at 4 °C and analysed within 48 h. The recovery was 94% to 100%. The sample purification step had a pre-concentration factor of 250, and the recovery percentages of the entire method were 85 ± 2 (PTA) and 91 ± 3 (PtB). The limits of detection (LOD) of the full method were 0.001 µg L−1 and 0.0001 µg L−1 for PTA and PtB, respectively. The method enables sensitive monitoring of PTA and PtB in groundwater. Carcinogenic PTA was detected in one groundwater well (0.35 µg L−1).

Abstract:
Povezanost površinske temperature mora (PTM) i površinske temperature zraka (PTZ) izmjerene na malome otoku Hvaru (Jadransko more) ispituju se na godišnjim i mjesečnim vremenskim ljestvicama, koristeći se podacima za razdoblje od pedeset pet godina (1964. – 2018.). Analizirana su sljedeća tri indeksa PTM-a i PTZ-a: (1) apsolutni minimum, (2) srednja vrijednost, (3) apsolutni maksimum. Rezultati su istaknuli statistički značajan trend porasta za sva tri godišnja indeksa PTM-a. U slučaju PTZ-a trend niza apsolutnih minimalnih godišnjih vrijednosti nije statistički značajan. Za nizove srednjih i apsolutno maksimalnih godišnjih vrijednosti PTZ-a trend je statistički značajan na razini p < 0,01. Mjesečne analize indeksa PTM-a gotovo u svim mjesecima (osim u listopadu) imaju statistički značajne trendove povećanja. U slučaju PTZ-a, statistički značajan trend porasta svih analiziranih indeksa dogodio se preko ljeta (srpanj i kolovoz). Rezultati pokazuju da je analizirana regija, posebice mali jadranski otoci, ugrožena klimatskim promjenama, tj. globalnim zatopljenjem tijekom ljeta. Primjenom RAPS metode na minimalnom godišnjem PTM-u zabilježen je statistički značajni pomak 1988. godine. Deset godina poslije, 1998. godine, otkriven je statistički značajan pomak prema gore na srednjem i maksimalnom indeksu PTM-a i PTZ-a. U slučaju minimalnih godišnjih vrijednosti PTZ-a otkriven je statistički značajan pomak prema dolje, počevši od 1979. godine. U analiziranom slučaju otoka Hvara, zagrijavanje indeksa PTM-a veće je od zagrijavanje indeksa PTZ-a i događa se tijekom cijele godine (osim u listopadu).

Platinum nanoparticles were synthesized using the gamma-ray irradiation as a reducing factor and chitosan as a stabilizer. The prepared platinum nanoparticles were characterized using ultraviolet-visible spectroscopy (UV-Vis), transmission electron microscopy (TEM), and Fourier-transform infrared spectroscopy (FT-IR). The saturated conversion dose to reduce the Pt4+ to Pt0 was found to be about 14 kGy for initial Pt4+ concentration of 1 mM. The size of the platinum nanoparticles (1.4–1.6 nm) did almost not change with the increase of chitosan concentration from 0.5 to 2.0%. Thus, gamma Co-60 ray irradiation method is favorable for synthesizing PtNPs with the small sizes.

Abstract:
Abstract A flow-injection biamperometric method for determination of pantoprazole (PTZ) in pharmaceutical tablets is reported for the first time. The reversible redox couples Fe3+/Fe2+, Fe(CN)63−/Fe(CN)64−, Ce4+/Ce3+, VO3−/VO2+, and I2/I− were tested as indicating redox systems for biamperometric determination of PTZ in a flow-injection assembly with optimized flow parameters. The best results were obtained using VO3−/VO2+, which showed to be a selective and sensitive biamperometric indicating system for PTZ even in the presence of excipients and antioxidants that typically are found in drugs. The analytical graph was linear (r = 0.99945) in the range from 10 to 100 mg/L using 25 mmol/L VO3− as the reagent and water as the carrier stream and applying 100 mV between the 2 platinum wire electrodes. The limits of detection and quantitation were 200 and 667 μg/L, respectively, with a sensibility of calibration of 22.6 mV/mg/L. The proposed method was successfully applied to determine PTZ in commercial pharmaceutical tablets with a mean relative error of 1.60% (n = 5) and mean relative standard deviation of 3.10%. Recoveries close to 100% showed good agreement between the expected amount of PTZ in tablets (40 mg) and the results found by the application of the proposed method and demonstrated that the formulations used in the tablet compositions do not interfere in the PTZ analysis. The system had good stability, with a relative standard deviation of 3.80% for 9 sequential injections of a 60 mg/L PTZ solution. A sampling rate of about 100 samples/h was obtained.

A simple, sensitive, and accurate HPLC-DAD method has been developed and validated for the simultaneous determination of pantoprazole and etodolac in rat plasma as a tool for therapeutic drug monitoring. Optimal chromatographic separation of the analytes was achieved on a Waters Symmetry C18 column using a mobile phase that consisted of phosphate buffer pH~4.0 as eluent A and acetonitrile as eluent B in a ratio of A : B, 55 : 45 v/v for 6 min, pumped isocratically at a flow rate of 0.8 mL min−1. The eluted analytes were monitored using photodiode array detector set to quantify samples at 254 nm. The method was linear withr2=0.9999for PTZ andr2=0.9995for ETD at a concentration range of 0.1–15 and 5–50 μgmL−1for PTZ and ETD, respectively. The limits of detection were found to be 0.033 and 0.918 μgmL−1for PTZ and ETD, respectively. The method was statistically validated for linearity, accuracy, precision, and selectivity following the International Conference for Harmonization (ICH) guidelines. The reproducibility of the method was reliable with the intra- and interday precision (% RSD) <7.76% for PTZ and <7.58 % for ETD.

Abstract:
A reversed-phased high-performance liquid chromatography–diode-array detection (HPLC–DAD) method has been developed for investigating the stress-dependent degradation of pantoprazole (PTZ) by a photolytic and oxidative mechanism. The developed method separated PTZ from its degradation products on a C18 column with a mobile phase consisted of methanol and water (60:40, v/v; pH 3.0) at a flow rate of 1 mL/min. The linear regression coefficient of 0.9995 was obtained for a concentration range from 5 to 25 μg/mL. The % relative standard deviation for repeatability and intermediate precision were below 0.5% and 1.5%, respectively, while the sensitivity of the method was demonstrated by a limit of detection value of 0.25 μg/mL. The stress sample analyses for PTZ results revealed the formation of a total of 18 degradation products, and out of them, 9 degradation products were common for both photolytic and oxidative degradations. Further, the oxidation by azobisisobutyronitrile produced the highest number of degradation products (11 impurities), 3 of which are more hydrophobic than PTZ. In photolytic degradation, 8 and 7 degradation products were observed with UV radiation and sunlight exposure, respectively. Furthermore, the degradation of pantoprazole sodium injection formulation was carried out under the same stress conditions, and it revealed the formation of 3 common impurities under both stress conditions, but other impurities were not detected in the formulations. Finally, 3 common impurities formed in formulations of PTZ injections, viz., sulfone, N-oxide, and N-oxide sulfone impurities, were identified by spike analyses.

Pretrombin merupakan prekursor dari trombin yang memiliki aktivitas proteolitik. Trombin merubah fibrinogen menjadi benang fibrin yang salah satu aplikasinya adalah dapat digunakan sebagai lem untuk menggantikan teknik jahitan pasca bedah. Aplikasi trombin untuk pembuatan lem fibrin menuntut diproduksinya trombin rekombinan. Tujuan dari penelitian ini adalah ekspresi gen pretrombin-2 (PT2) manusia rekombinan menggunakan sistem ekspresi Pichia pastoris. Gen pengode PT2 dirancang sesuaidengan kodon preferensi P. pastoris. Fragmen PT2 diamplifikasi dengan metoda PCR dengan penambahan sisi restriksi EcoR1 pada ujung 5’ dan sisi restriksi SacII pada ujung 3’. Produk PCR yang berukuran 924 pb diligasi dengan vektor ekspresi pPICZaB untuk P. pastoris dan disubkloning dalam inang Escherichia coli. Urutan nukleotida dikonfirmasi dengan metoda dideoxy Sanger. Plasmid rekombinan pPICZaB-PT2 kemudian digunakan untuk mentransformasi P. pastoris SMD1168 defisien protease dengan metoda elektroporasi. Hasil penelitian menunjukkan bahwa gen PT2 berhasil diamplifikasi dan dikloning dalam E. coli. Analisis restriksi dan penentuan urutan DNA menunjukkan bahwa PT2 rekombinan 100% homologi dengan hasil rancangan. Hasil ekspresi PT2 oleh P. pastoris menggunakan metanol sebagai inducer memperlihatkan bahwa PT2 dengan berat molekul 35 kDa berhasil diekspresikan. Optimasi kondisi ekspresi melalui variasi konsentrasi metanol sebagai inducer dan sorbitol sebagai sumber karbon tambahan menunjukkan bahwa metanol 2% dan sorbitol 2% merupakan kondisi optimum ekspresi PT2.
