Abstract:
The thesis deals with water electrolysis focusing in particular on pulse electrolysis. The theoretical part characterizes hydrogen and its properties as well as the ways of its use and storage. It also analysis various methods of hydrogen generation. It examines in details water electrolysis and its energetic and chemical balance. There is also an evaluation of water electrolysis efficiency and its improvement followed by pulse electrolysis description. Laboratory equipment for direct current and pulsed direct current hydrogen production are suggested. Theoretical expectations are tested experimentally. Finally the thesis presents the results of measurements, compares and summarizes the data. It points out the contribution of this type of electrolysis with its effectiveness. It proposes uses of electrolysis and the subsequent research.

Abstract:
The aim of the Master's thesis was to study contemporary methods for human pulse detection from standard video and suggest a method, which can be used to detect the pulse. Approaches of detecting miniature changes between frames of a video are presented. Position changes of the feature points or changes in colour of some part of an image are detected. It capitalize on the fact that those changes are caused by the pulse of blood. The method for color changes magnification is selected as a base for pulse detector. Face regions of interest are analyzed to detect frequency of changes of intensity between frames. 1D signal is gained and its analysis leads to heart rate. Approach to create heat map of frequency changes is also presented.

In the semestral thesis is given basic methods of non-contact measurement heart rate. There is explained Eulerian video magnification method deals with the visualization of the pulse in the videosequence of face. The semestral thesis describes algorithm Viola-Jones face detection in images and algorithm Kanade-Lucas-Tomasi for tracking faces in the videosequence. Part of the work includes design and realization of measurement. There is explained realization of the program and documented execution results, which are discussed. From the results it is designed to guide for optimal recording.

Cílem této práce je seznámení se s problematikou detekce ionizujícího záření a jeho detekcí zejména pomocí proporcionálních detektorů a navrhnutí algoritmu pro jejich klasifikaci a jeho následná implementace do FPGA. V první části práce je obecně popsáno, jak jednotlivé typy plynem plněných detektorů fungují. V druhé části je věnován prostor algoritmům pro klasifikaci pulzů, které se objevují v literatuře. Následuje návrh vlastního algoritmu, jeho rozbor a rozebrání výsledků. Ve třetí části následuje popis samotné realizace navrhnutého řešení na platformě RedPitaya. Je zde rozebrána celková architektura navrhnutého systému, detailně popsány jak jednotlivé bloky v logice FPGA, tak i ostatní skripty zajišťující zpracování naměřených výsledků a jejich vizualizaci.

Abstract:
The main goal of this thesis is to design control board for the experimental electroporation device and to develop control firmware. The first chapter of this work focuses on the electroporation phenomenon itself. Behaviour of the cell model in external electrical field is described there as well as simulation and overview of how electroporation affects living tissue. It also explains the main requirements for parameters of the electroporation pulses as well as need for ECG synchronization. Furthermore, some remarks are given about novel high frequency electroporation method, which involves use of nanosecond bipolar high voltage pulse bursts. The second chapter briefly introduces commercial electroporation device called Nanoknife, including control part, power part, and it's limits. The third chapter consists of introduction of the novel experimental electroporation device developed at BUT. Power part of this device is discussed as well. Next chapter focuses on design of the control board for this device and also on description of the particular schematic parts. There is a control algorithm explanation in the fifth chapter of this thesis followed by the brief manual to machine operation.

Abstract:
The phagocytic cells circulating in the bloodstream play a key role in both the defense of the body and the pathology of inflammatory diseases. Thus, targeting their functions has potential to modulate an immune response, especially during the inflammatory phase. This master's thesis was focused on the influence of electric pulses on the most abundant phagocyte population in human peripheral blood, namely neutrophils. The theoretical part describes the role of neutrophils in the development of the immune response and the effects of the electric field on various cells. Consequent part of the thesis was the optimization of the electrical stimulation of neutrophils using a unique platform with a network of gold electrodes. In stimulated cells by electrical pulses, activation of selected signaling pathways, degranulation, ROS production, citrullination of histone H3 and expression of surface markers were monitored. Overall, electrical stimulation was observed to induce neutrophil activation but only electrical pulses of size 1 V were found to be statistically significant in the case of ROS production and 10 mV and 100 mV electrical pulses in the case of metalloproteinase MMP8 degranulation. The absence of significant effects in the most observed parameters was probably due to unwanted activation of neutrophils in control samples.

The lead-acid batteries are the most commonly used electrochemical power sources and are the oldest type of secondary battery cells. This master thesis provides a short introduction to the lead-acid batteries and shows possible ways of sorting from different points of view. Thesis describes construction of lead-acid batteries, parameters of lead-acid batteries, electrochemical and physical reactions inside the battery. Futher the thesis deals with pulse charging of lead-acid batteries. In the next part of the thesis is described the production of a set of experimental electrodes for electrochemical workplace resources. The final part of this thesis are experiments focusing on the pulse charging using negative current pulses and evaluate the effect of different pulse modes on cells.

Subsequently, the work includes strength analysis of the designed turbine housing. The aim of this diploma thesis is to develop a linear positioning stage for Ultra High Vacuum (UHV) environment. Simple prototypes of the linear positioning stage were designed and incorporated as part of a multiaxis sample manipulator for a UHV Scanning Tunneling Microscopy / Atomic Force Microscopy (STM/AFM). Different types of position encoders and linear guideways are discussed. Implementation of the homodyne interferometer as an optimization tool for a slip-stick based linear stage is described. Scalar diffraction theory is used to model the diffraction grating optical position encoder behavior.

Abstract:
Epsilon toxin (Etx) produced by Clostridium perfringens type D is responsible for a fatal Enterotoxemia (Pulpy Kidney Disease) in economically significant livestock such as sheep and goats. The only practical means of controlling this disease is by immunisation and avoiding the circumstances that are conducive to its occurrence. All currently available Pulpy Kidney Disease vaccines are based on a formalinised toxoid of Clostridium perfringens (Welchii) type D. This is either as an alum-precipitate, oil-emulsion formulation of the whole cell culture or a bacterial culture filtrate (Deepika, 2010). The current vaccine has several drawbacks. First, it is generally accepted that its chemical inactivation with formalin is difficult to standardise. Second, the classical methods of detoxification usually alter the overall protein structure in a random manner. Consequently, the immunogenicity of this type of a vaccine is decreased substantially. Third, there is a narrow range in balancing the detoxification (the strength of formalin commonly used is 1%) and the immunogenicity of the vaccine (Robertson et al. 2011). Alternatives to formalin inactivation have been proposed because of these challenges. These include ectopically expressing the mutated toxoids in other gram-positive microorganisms. Apparently, plants provide a genuine alternative for the expression of an immunogenic and non-toxic vaccine for the Pulpy Kidney Disease. Thus, in this study, the hypothesis was that the EtxD protein could be transiently expressed efficiently in Nicotiana benthamiana plants via deconstructed viral vectors, targeting the EtxD protein expression into the apoplast or the cytosol. This resulted in the investigation of the following six research topics: 1. The feasibility of producing the Epsilon toxin recombinantly in N. benthamiana. 2. The role of subcellular targeting in the observed level of expression and accumulation of the target protein. 3. The methods of purification that could be explored to recover and purify the EtxD protein that was transiently expressed recombinantly in the N. benthamiana leaves. 4. The suitability of the plant-derived purified EtxD protein as an antigen for Pulpy Kidney Disease vaccine production. 5. An evaluation of the toxicity of the plant-derived EtxD protein. 6. Testing the efficacy of the recombinant plant-derived EtxD antigen. In this context, the EtxD gene, accession number AY858558, and the genetic region from the NCBI and EMBL databases was targeted for this study since the EtxD gene is well-reported in published data and is similar to the EtxD-gene strain produced in South Africa. The targeted gene was then codon-optimised to be expressed in N. benthamiana plants and chemically synthesised by GeneArt. The codon-optimised EtxD gene was directly cloned into an Icon-deconstructed vector and vacuum infiltrated via an Agrobacterium- mediated transfer. Leaves of N. benthamiana were transfected by vacuum infiltration to deliver the EtxD gene transiently into the apoplast and the cytosol respectively. The plant-derived EtxD protein was then isolated from the plant matrix and biochemically analysed by SDS-PAGE, N-terminal peptide sequencing and Western Blot analysis. The EtxD protein was visible as a 34 kDa protein band on an SDS-PAGE gel. The protein band was isolated, and the sequence was confirmed by N-terminal sequencing, as well as by Western Blot analysis using a secondary polyclonal guinea pig anti-EtxD antibody. The plant-derived EtxD protein was then quantified by ELISA and thereafter the expression levels were established at 380 mg/kg fresh weight when targeted to the apoplast, and 300 mg/kg fresh weight when targeted to the cytosol. The apoplast plant-made EtxD protein was purified using a two-step chromatography method, namely ion exchange and size exclusion, with a 50% recovery of the EtxD protein on the final step of purification. To investigate the suitability of the plant-derived purified EtxD protein as an antigen for the Pulpy Kidney Disease vaccine, the toxicity of the EtxD protein was evaluated and the efficacy of the derived EtxD protein was tested. For these, both in vitro and in vivo studies were conducted. The LD50 studies on mice revealed that the plant-derived EtxD protein was slightly toxic, which correlated with the IC50 results on MDCK cells. For the animal-challenge results, two formulations of vaccines were prepared from the recombinant antigen EtxD protein and administered intravenously to mice. The formulations that contained the plant-derived EtxD protein that were not activated by trypsin were unable to protect mice against the Epsilon toxin challenge. This indicates that the Epsilon toxin in the purified plant extract was not immunogenic. When the plant-derived EtxD protein was treated with trypsin, inactivated with formalin and formulated with the adjuvant, alum, it was also non-protective. However, the formulation containing the plant-derived EtxD protein and Disease Control Africa (DCA) immune stimulant was protective. These findings indicated that the plant-derived Epsilon toxin is a viable recombinant antigenic vaccine when formulated with the immune stimulant DCA. In conclusion, this study has demonstrated that tobacco is a suitable host for the production of the EtxD protein. The ELISA results of the infiltrated tobacco leaf samples have demonstrated the successful expression of the 34 kDa EtxD protein together with glucan-endo-1, 3-beta-glucanase of about 25 kDa. The cytosol targeted strategy generated the lowest EtxD protein production at 300, 200 and 10 ?g/kg fw of the protein. For large scale-production of the EtxD protein, transient expression targeting to the apoplast is preferable because of the high yield of protein per fresh leaf weight achieved in this study.

This diploma thesis deals with the feasibility of using PVDF nanofibers as an active sensor layer generating electrical signal. PVDF and related electromechanical effects are described. A research study is conducted regarding existing PVDF nanofiber applications and based on it, an event sensor design utilizing triboelectric effect and electrostatic induction is proposed. The electrical response of the layers is experimentally investigated and a pulse detection algorithm is conceived and implemented. Finally, a way of integrating the sensor into a rail track is proposed.

Abstract:
This master thesis concerns with emotional states and gender recognition on the basis of speech signal analysis. We used various prosodic and cepstral features for the description of the speech signal. In the text we describe non-invasive methods for glottal pulses estimation. The described features of speech were implemented in MATLAB. For their classification we used the GMM classifier, which uses the Gaussian probability distribution for modeling a feature space. Furthermore, we constructed a system for recognition of emotional states of the speaker and a system for gender recognition from speech. We tested the success of created systems with several features on speech signal segments of various lengths and compared the results. In the last part we tested the influence of speaker and gender on the success of emotional states recognition.

Abstract:
This thesis deals with the cartilage imaging using magnetic resonance. At first, there is mentioned physical principle of the magnetic resonance phenomenon and the most commonly used excitation sequences, followed by the description of the 9.4 T MR imaging system Bruker BioSpec 94/30 USR, which was used for measurement in the practical part. The next part is dedicated to the composition of cartilages and describes the temporomandibular joint, due to its suitability as an object for cartilage imaging. The series of MR scans of temporomandibular joint were taken with different acquisition parameters and evaluated by program designed through the MATLAB software. The program can be used for viewing scanned images, evaluating their contrast and determining the T1 relaxation time of the tissues by creating T1 maps.

Abstract:
The presented thesis is focused on knowledge extension in the area of partial discharge measurements and evaluation at frequencies higher than 1 kHz. The thesis includes the design and set up of the measuring workplace equipped for measuring partial discharges, including the methodology of the measuring and calibration procedures necessary for the achievement of reproducible results. Another part of the thesis deals with the technology of acoustical and electromagnetic sensors convenient for the diagnostics and localization of partial discharges in devices, that work at voltages above 500 V. Electrical circuits that enable data evaluation by using the signal from a sensor working on amplitude analysis principle were developed. For PD detection a PC controlled measuring device working on amplitude analysis principle had to be designed and realized. The possibilities of the workplace have been demonstrated in the process of development of the insulating systems for pulse transformers. Thesis results can significantly contribute to a higher quality of newly designed electronic devices and systems working at voltages above 500 V.

This master’s thesis describes possibilities of usage of ZigBee communication protocol and integrated circuit ELM 327 in problem of realization universal diagnostic device, running by OBDII communication standards, which can be used to monitor function of all car´s electronic systems. Thesis also deals with design and realization of necessary electro technical devices and modules followed by compilation of control software and experimental measurements of these devices in real traffic flow. In the first part of project, there are reminded some basic features and parameters of OBDII, followed by description of ways how to use OBDII and adumbrating how to realize universal diagnostic system. The analysis of ZigBee system focused on XBee circuits follows. Construction, ways of data transfer and possibilities of controlling are described. Integrated circuits produced by ELM Electronics Company are introduced and possibilities of usage ELM 327 are described as requested by main task. Second part talks about designing of electronic circuits of this system, assembling schematic diagram and designing sources for production of PCB. Physical realization of these circuits and its first start is also included. Thesis finishes by dealing with practical experiences and measurements using real vehicles. This part talks also about means to improve system by several software solutions to reach more effective functioning of created device.

In this thesis we present the realization of a source of 1.5-cycle carrier-envelope phase stable laser pulses in the mid-infrared spectral region. We used ytterbium laser system generating 1 µm pulses as a pump of setup, where the beam is split into several parts and interact in nonlinear optical media. 2 µJ pulses with duration 18 fs at 50 kHz repe- tition rate are produced. By spectral broadening in crystal GGG, 9,9 fs pulses can be achieved. The mid-IR pulses was characterized by third harmonics generation frequency resolved optical gating in the interferometric configuration. Fourier filtering of the mea- sured interferogram allows for the complete reconstruction of amplitude and phase of the ultrashort pulses generated by our setup. The pulses will in future serve for experimental investigation of ultrafast strong-field phenomena in solids. 1

Abstract:
The aim of this dissertation study was to optimize the isolation and long term cultivation protocols for human dental pulp stem cells. The protocols which showed best results were used for cultivation of dental pulp stem cell isolated from exfoliated teeth (SHED). Additional aims were to characterize DPSC and SHED and prove their ability to proliferate over Hayflick's limit and differentiate into mature cell lines (osteoblasts, chondroblasts and adipocytes). In order to find optimal protocols for isolation of dental pulp from tooth, we tried three different approaches. During optimization of cultivation protocol we focused on decreasing amount of fetal calf serum (FCS) from 10 % FCS in cultivation media (most often used in literature) into 2 % and thus get closer to cultivation conditions suitable for clinical usage. We compared DPSC cultivated in three different media (medium with 10 % FCS, 2 % FCS supplemented with growth factors and media with 2 % FCS supplemented with ITS and growth factors). For characterization of DPSC and SHED we used basic biological properties (proliferation activity, viability, morphology), their phenotype and karyotype. The main focus of this thesis is the characterization of the charge transport in CdZnTe radiation detectors and the study of the effect of the detector illumination on charge transport. The transport properties are evaluated using Laser-induced Transient Current Technique and the Monte Carlo simulation is used for fitting the measured current waveforms. The properties of the detector prepared from semi-insulating CdZnTe single crystal with a platinum Schottky contacts were measured in the dark in the unpolarized and polarized state and under the anode and cathode continuous LED above-bandgap illumination.

Ondřej Novák, Ph.D., Hilase centre, Institute of Physics of CAS Abstract: High average power wavelength tunable picosecond mid-IR source based on optical parametric generation (OPG) and optical parametric amplification (OPA) is being developed. The conversion system is pumped by an Yb:YAG thin-disk laser delivering 100 W of average power at 100 kHz repetition rate, 1030 nm wavelength, and 2-3 ps pulse width. Part of this fundamental beam pumps an OPG process in a PPLN crystal. The generated wavelength is determined by PPLN's poling period and temperature. Tunability of the signal wavelength between 1.46 µm and 1.95 µm was achieved, the signal beam of 20 mW was generated at 2 W of pump power, when double pass of the beams through PPLN crystal was used. The corresponding idler wavelengths were in range 2.18-3.50 μm. The signal beam was further amplified by OPA process in two KTP crystals, which was pumped by the fundamental beam. The signal beam was amplified up to 2 W at pumping of 38 W. Tuning of the output wavelength was realized by change of the phase-matching angle in KTP crystals. The research design was to investigate the “Application of Flying Puly on Elderly Outdoor Resistance Fitness Equipment”, targeting the senior population, where such fitness equipment is placed in public places, such as parks. To effectively exercise the muscles and bones in human body to prevent deterioration in skeletal muscles and bone density due to aging, a continuous variable transmission system was used, since there was no speed limit ratio and it could allow user to manually adjust accordingly to need. The resistance came from non-contact magnetic force, which was mainly considering the cost of frictional wear. In the experiment, the combination of parameters of the continuous variable transmission system for elderly exercise was: Ply-disc with long bullet-shaped pulley balls of pure lead weight and the variable spring with the minimum spring coefficient of 0.12 kg/cm at a wheelbase of 25 cm, measuring the minimum speed of output shaft of the CVT system at 413 rpm. The speed of the bicycle when stepped upon by human is generally between 33 rpm and 80 rpm. Considering the low speed upon stepping that the CVT system may even lose its momentum, a combination of increasing speed gear at ratio 1:12 was added in the system. The experimental result of system with and without magnetic force as resistance showed that the output shaft speed of CVT without resistance was 460 rpm, while it was 706 rpm for CVT with magnetic force as resistance, yielding a difference of 246 rpm. In the fixed-speed test, the operational balance point speed without additional magnetic resistance was 1133 rpm and 1223 rpm for system with magnetic force as resistance. The Ply-disc exercise system in this study was matched with various parameters and applied with magnetic force as the source of resistance to test and present the trend graphs of its action at each operational point, at each operational point in balance and at the highest rotational speed. By adding an incremental mechanism for treading at low speed by the user, the magnetic force would increase to produce more resistance as speed increased to allow a progressive increase in exercise.

Abstract:
In the presented thesis the topic of femtosecond pulse dispersion and methods of characterizing pulse profile are briefly introduced. Then, a functionality of a spatial light modulator is described. The spatial light modulator was used in an experimental scheme called the pulse shaper, which allowed independent amplitude and phase modulation of pulses. Duration and dispersion of pulses was measured by two methods called MIIPS and PICASO. MIIPS was also used for a reconstruction of a spectral phase of pulses. The autocorrelator was constructed on a design of the Mach-Zehnder interferometer. The duration of the shortest measured pulse (13.3 ± 0.5) fs was retrieved from measured interferometric autocorrelations by PICASO. Furthermore, theoretical dependence of pulse duration on the group delay dispersion was confirmed for pulses shorter than 120 fs. The group velocity dispersion was measured for fused silica windows and for a pair of diffraction gratings in the pulse shaper. Both values confirmed theoretical expectations.

Spintronics is a dynamically developing branch of electronics which for transfer, processing and storing of information use not only electron charge but also its spin. Materials appropriate for a construction of spintronic devices should sustain the spin order for a sufficiently long time enabling a manipulation of spins. Simultaneously, in these materials the spin of electrons should be possible to transport fast and long enough across the device. In this work, heterostructure of GaAs/AlGaAs fulfilling these requirements is investigated by a pump-probe technique. In particular, long spin lifetimes of electrons in the heterostructure are studied using the resonant spin amplification method and with a use of the pulse picker. The nuclear spin polarization and the effect of an applied electric current on the studied heterostructure are also investigated in this work. Spectral filters are used to improve the existing experimental setup for the pump-probe technique and to measure a magnetooptical spectrum of the investigated heterostructure.

Intense few-cycle laser pulses can significantly affect the properties of transparent solids during the interaction. These processes take place on femtosecond time scales and they can be studied using ultrafast spectroscopic methods. This dissertation deals with highly nonlinear processes in diamond. In the interaction with a crystalline diamond, we observed a strong nonlinear broadening of the spectrum due to self-phase modulation effect, which allowed us to create a simple technique for compression of pulses from laser oscillator. At the same time, strongly nonlinear five-photon absorption was observed, in which we found a significant anisotropy and dependence on the polarization state. With two-beam pump and probe experiments we present a study of anharmonic phenomena in the dynamics of coherent phonons in diamond, additionally we created a new detection technique of lattice vibrations using multiphoton absorption. Finally, the high time resolution of the experiments revealed that the sub-picosecond electron dynamics strongly depends on the composition and morphology of the polycrystalline diamond thin films. The experimental results of this work provide comprehensive research into the interaction of diamond with few-cycle laser pulses and the development of new spectroscopic methods.

The circadian system controls the timing of most of the physiological functions and behaviour within approximately 24 hours period. The light/dark cycle aligns this circadian period with the exact length of the solar day. Light information is conveyed via ipRGC cells in the retina, with maximum sensitivity in the blue part light (460-480nm wavelength). The information proceeds to the main circadian clocks located in the suprachiasmatic nuclei. This clock regulates the synthesis of neurohormone melatonin in the pineal gland whose high night level can be rapidly downregulated by the light at night. The major aim of this study was to test the effectiveness of the blue or green light-emitting device, potentially utilized in clinical practice, to suppress the night melatonin level and provide thus the first evidence that this device may affect the circadian system. The results show that the light emitted by this device has the potential to lower melatonin levels in most subjects with the higher efficiency in the blue range. Abstract:
The aim of the study was to evaluate the effect of specific rehabilitation concept on the motor deficit in patients after stroke. This concept is used at the Department of inpatient rehabilitation FN Motol, the main component is exercise on the principles of dynamic neuromuscular stabilization (DNS) and is based on the principles of developmental kinesiology. The study included 12 patients after stroke. Probands participated in the 3 week therapy, exercise was carried out twice a day. Significant (p <0.05) improvement in motor function of the arm, hand, leg and foot was demonstrated by using Student`s paired t-test. There was a statistically significant improvement in postural control and reducing shoulder pain (all assessed using a specific scale: Chedoke McMaster Stroke Assessment). Changes in the degree of spasticity were evaluated by Modified Ashworth scale. A statistically significant improvement in the degree of spasticity was observed in these muscle groups: the elbow flexors , adductors of lower limb, knee extensors and hamstrings. It has been shown that the concept of dynamic neuromuscular stabilization can be advantageously used in selected patients after stroke. It is appropriate to supplement the DNS therapy with training of ADL.

The diploma thesis presents the development of selected monitoring techniques of basic life functions in historical continuity. From the measured variables, this work includes measurements of cardiac activity, blood pressure, pulse, respiration and body temperature of the adults patients with intensice care in a historical context. This is a theoretical diploma thesis, which is divided into several chapters describing the history of nursing, a view of the monitoring in intesive care in the historical context to the presents. The first part of this historical work describes the beginnings of nursing. Its own origin and development, until now. The prestige of the nurse´s profession is formed by the society. In the leading positions in medicine previously dominated the men, in nursing care mainly women. A balanced relationship between a nurse and a doctor has been and is very important. The next part is devoted to the development of intensive care and the development of the role of the nurse in these departmenst. The history of intensive care has led to new partnership relationships between physician and nurse. There was an unfamiliar environment in which critical patients had to be cared for. This new role for the sister led to the increase of copetencies, there was a need to increase the education activity that focused on intensive care area. The view of urgent condition has changed over time. Already in the early sources, the urgent condition was described as a condition in which the health and life of the affected person is urgently threatened. These urgent conditions gave rise to the first beds for intensive care. Futher, the development of all monitoring techniques to date, servise of the monitoring technology. The negatives associated with monitoring technology are described too. The thesis will be used as a preview of changing nursing care in the context of changes in monitoring techniques of the intensive care.
