Pyrazolo[3,4-d]pyrimidines were known as adenosine antagonists at the rat A1 and A2A adenosine receptors based on our previous studies. In this study, 245 pyrazolo[3,4-d]pyrimidines derivatives with various benzyl substitutents at N-1 and various hydrophobic side chains at C-4 and C-6 were synthesized and screened at the human A1, A2A and A3 adenosine receptors. 14 out of 245 compounds were resynthesized and purified to determine the Ki values of these compounds at the human A1 adenosine receptor. In this study, the synthesis of 5-ferrocenyl/aryl-4-(phenylselenyl)-1H-pyrazole derivatives was investigated since the integration of ferrocenyl and selenium moieties into pyrazole derivatives may increase their current biological activities. Initially, the starting propargyl aldehydes were synthesized from corresponding acetylenes. Subsequently, propargyl aldehydes were reacted with hydrazines to yield corresponding hydrazones. Initially the perfluoroacylation of a range of commercially available vinyl ethers and conversion of the resulting perfluoroacylated enol ethers to 1-H-pyrazoles via reaction with hydrazine is reported. The selective synthesis of a range of alpha-aryl vinyl ethers using Heck chemistry is then reported. 3-Chloro-4,4,5-trimethyl-3,5-diphenyl-4,5-dihydro-3H-pyrazole (3b) and 3-bromo-4,4,5-trimethyl-3,5-diphenyl-4,5-dihydro-3H-pyrazole (3c) were prepared for the thermolysis project. The thermal decompositions of 3b and 3c were monitored using 1H NMR spectroscopy. Plots of ln (% starting material) vs. time (sec) were linear for at least two half lives and the first order rate constants were determined over at least a 30o temperature range. The relative reactivity was found to be 3c > 3b. The activation parameters determined for the thermal decomposition of the pyrazoline at 150oC were found to be

We found that treatment of (Z)-1-phenyltrideca-4-en-2,6-diyn-1-one (78b) with 5 eq hydrazine and 4 eq copper(I) chloride in refluxing CH3CN afforded pyrazolo[1,5-a]pyridine (81) in 85 % yield.

Using phenyl pyrazole calix[4]arenes as basic synthesis frameworks, we synthesized phenylanthryl pyrazole calix[4]arenes 76 and 77 and control compounds 78 and 79 as fluorogenic ions sensors. Based on the results from UV-Vis, fluorescence and 1H-NMR spectrometry, we found that pyrazole calix[4]arenes 77 and 79 recognized Ag+ and Hg2+ with excellent selectivity in MeOH/CHCl3 (v/v = 4:1), respectively.
