Abstract:

A series of 2-alkyl-3-arylpyrido[2,3-d]pyrimidones were synthesized for testing as potential antiepileptic drugs. The goal was to achieve better neurological activity and/or lower toxicity than displayed by a series of 2-alkyl-3-aryl-4(3H)-quinazolinones prepared previously in our research group. From the pharmacological testing data of these target compounds, we have found that the additional nitrogen at the C-8 position of the quinazolinone framework increased the anticonvulsant activity. New methodology for the synthesis of several 4H-pyrido[1,2-a]pyrimidin-4-ones has been developed from commercially available 2-aminopyridines and β-oxo esters catalysed by Montmorillonite under solvent-free conditions in good yields. Eight mononuclear complexes were prepared from the N-(2-pyridyl)-4-amino-4-oxobutanoic acid (H2L1) ligand. H2L1 chelated to cobalt(II), nickel(II) and copper(II) through the pyridine and amido nitrogen atoms while the carboxylic acids did not coordinate. 1D and 3D hydrogen bonding networks were formed through interactions between non coordinated solvent molecules and anions within the crystal lattices along with carboxylic acid interaction to the metal centres. La protéine du TGF-[bêta]1 est une protéine multifonctionnelle qui agit dans plusieurs types cellulaires. Son action varie selon le type de cellulaire. Bien qu'elle ait un rôle inhibiteur chez les astrocytes normaux, elle posséderait un rôle principalement activateur de nombreuses voies carcinogéniques chez les tumeurs astrocytaires primaires malignes. L'isoforme du TGF-[bêta]1 est celle qui est la plus impliquée dans ces processus.
