Full textO presente trabalho descreve o estudo quÃmico e farmacolÃgico da casca do caule da espÃcie Simarouba versicolor, pertencente à famÃlia Simaroubaceae. A espÃcie ocorre preferencialmente em Ãreas abertas de solos bem drenados, como cerrados e caatinga. Conhecida popularmente como âpau paraÃbaâ, âmata-cachorroâ ou âsimaruba do Brasilâ, a casca do seu caule possui propriedades inseticida e antihelmÃntica. Com o objetivo de estabelecer uma possÃvel relaÃÃo extrato atividade de inibiÃÃo da enzima acetilcolinesterase (AChE). Foi realizada uma triagem dos extratos das partes (folhas, talos, Galhos, Casca do caule e Cerne) da espÃcie Simarouba versicolor, atravÃs do ensaio de Ellman. O extrato etanÃlico da casca do caule apresentou elevada inibiÃÃo da AChE, desse extrato, as fraÃÃes clorofÃrmica e acetato de etila foram identificadas como sendo responsÃveis pela inibiÃÃo da enzima. A prospecÃÃo quÃmica da fraÃÃo clorofÃrmica resultou no isolamento de trÃs compostos, uma mistura de esterÃides β- sitosterol e estigmasterol, um quassinÃide, a 11-acetilamarolida e o alcalÃide 4,5- dimetÃxicantin-6-ona. Na fraÃÃo acetato de etila foi isolado o 3-β-O-β-D-glicopiranosil sistosterol. A determinaÃÃo estrutural desses compostos foi realizada atravÃs de anÃlise espectroscÃpica pelos mÃtodos de IV, EM, RMN1H e 13C-BB utilizando tÃcnicas uni e bidimensionais e por comparaÃÃo com dados descritos na literatura. Na anÃlise qualitativa de inibiÃÃo AChE os extratos e os constituintes isolados 11-acetilamarolida e 4,5-dimetÃxicantin-6-ona apresentaram bons resultados. Os extratos tambÃm apresentaram atividade citotÃxica em trÃs linhagens tumorais testados.
The present paper describes the chemical and pharmacological study of the stem bark of the species Simarouba versicolor, which belongs to the family Simaroubaceae. The species are found, mainly in open areas of well drained soils such as cerrado and caatinga. Known popularly as âpau Paraibaâ, âmata-cachorroâ or âBrazilian simarubaâ, the bark of its trunk has insecticidal and antihelminthic properties. It aims to establish a possible extract inhibition of enzyme activity of acetyl cholinesterase (AChE) Relationship. It was accomplished a triage of the parts extracts (leaves, stems, branches, stem bark and heartwood) of the species Simarouba Versicolor by the Ellman test. The ethanol extract of stem bark showed high inhibition of AChE, from this extract, the chloroform fraction and ethyl acetate were identified as being responsible for the inhibition of the enzyme. The chemical prospect of the chloroform fraction resulted in the isolation of three compounds, a mixture of steroid β-sitosterol and stigma sterol, a quassinoid, the 11-acetylamarolide and the alkaloid 4.5-dimetoxicantin-6-one. In the ethyl acetate fraction it was isolated the 3-β-O-β-D-glucopyranosyl sitosterol. The Structure determination of these compounds was performed by spectroscopic analysis through the methods of IR, MS, RMN1H and 13C-BB using uni and bidimensional techniques and by comparison with the data reported in the literature. In qualitative analysis of the AChE inhibition the extracts and isolated constituents 11- acetylamarolide and 4.5-dimetoxicantin-6-one showed good results. The extracts also showed cytotoxic activity in three tumor lines tested.

Full textObjective: To evaluate the effect of the Isobruceina B (IsoB) as antihypernociceptive and anti-inflammatory, and identify the molecular mechanism involved. Methods: Initially the effect of IsoB upon inflammatoy hypernociception on plantar tissue was evaluated by electronic von Frey. Plantar tissues were removed for indirect quantification of neutrophil infiltration by myeloperoxidase (MPO). Its effect on thermal nociception in mice was evaluated by the hot plate test at 55 ° C. Mice pretreated with IsoB (s.c.) and treated with carrageenan (i.pl.), their tissues were removed for quantification of plantar release of cytokines TNF, IL-1? and KC/CXCL1. For in vitro studies, macrophages derived from peritoneal cavity of naive mice, pretreated with 3% thioglycolate (i.p.) or line RAW 264.7 were used. Peritoneal macrophages were incubated with lipopolysaccharide (LPS) in the presence or absence of different concentrations of IsoB for the quantification of the release of cytokines TNF, IL-1?, and IL-10 KC/CXCL1 in the supernatant by ELISA. In vitro production of nitric oxide (NO) was indirectly determined by the Griess method. To verify the effect of IsoB on the pathway of NF-kB, RAW 264.7 macrophages stably transfected with a gene to expression NF-kB-dependent luciferase (RAW-Luc) were incubated with different stimuli (LPS, peptidoglycan, TNF and phorbol myristate acetate [PMA]) in the presence or absence of IsoB. The luciferase activity was detected by the Luminometer. The cytotoxicity of IsoB was assessed by the MTT, lactate dehydrogenase and cytotoxicity assays trypan blue on macrophages in vitro. The effect of IsoB on macrophages upon the IkB-? degradation and NF-kB translocation (p65 subunit for marking) to the nucleus were examined by Western blot of nuclear and cytoplasmic extracts, and Confocal Microscopy. The effect of IsoB on the TNF mRNA gene transcription induced by LPS was assessed by RT-PCR. Transfected HEK 293 cells were used to verify the effect of IsoB on the protein synthesis. Results: IsoB inhibits inflammatory hypernociception, but has no effect on thermal nociception. IsoB inhibited the production/release of cytokines IL-1? and KC/CXCL1 in vivo, and TNF, IL-1?, KC/CXCL1, IL-10 and NO in vitro in a concentration-dependent manner. The analyzed compound also inhibited luciferase activity NF-kB-dependent for all stimuli tested in a concentration-dependent manner. Cytotoxic activity was not detected in any of the concentrations tested. The IsoB not inhibit the degradation of IkB-? or translocation of NF-kB to the nucleus. Unexpectedly, the natural compound tested neither reduced transcription of TNF mRNA, but inhibited the synthesis of luciferase induced nonspecifically. Conclusion: The reduction on release of pro-inflammatory cytokines such as TNF, IL-1? and KC/CXCL1 and neutrophil migration are mechanisms involved in anti-inflammatory and anti-hypernociceptive effects of IsoB. The present results suggest a molecular mechanism based on the direct inhibition of protein synthesis.

Full textSimalikalactone E (SkE) is a potential antimalarial and anticancer quassinoid isolated from Quassia amara, a plant traditionally used against malaria in many tropical countries. To improve the extraction yield and acquire the pharmacokinetic data of this molecule, two methods of quantification of SkE were developed and validated. The fisrt method used HPLC coupled to MS using an ion trap analyzer operating in Single Ion Monitoring detection. The second used UPLC coupled to MS/MS using a hybrid triple quadrupole/linear ion trap mass analyzer operating in Multiple Reaction Monitoring. The lower limits of quantification of these two methods were 0. 224 and 0. 028 mg/L for HPLC and UPLC, respectively. The application of these two methods allowed, to indentify the critical steps of the process requiring further improvement to optimize the SkE isolation yield and also the influence of different parameters such as the extraction scale, the nature and quantity of solvents, the temperature, the agitation and the drying process. In vitro and in vivo studies were realized on the traditional antimalarial preparation obtained from Q. Amara leaves. They permitted to conclude that despite a strong antimalarial activity, SkE alone couldn't be responsible for the activity because of its very low content in the traditional decoction. Phenolic compounds (gallic acid, methyl gallate and vitexine) were therefore identified in the traditional decoction and have been found to be involved in the antimalarial activity of this preparation. The quantification methods were also used to examine the fate of SkE in biological fluids. This compound was only detectable in the blood after intravenous administration route and in a very low concentration (0. 1 mg/L 1 hour after administration) compared to the given dose. SkE was not detectable after oral or intraperitoneal administration routes. Moreover, it was found that SkE was very quickly transformed in simulated intestinal fluid and more stable in simulated gastric fluid. In conclusion, thanks to the two quantification methods, this study paves the way for improving the extraction protocol, and the research for active metabolites of this very promising drug

Full textSecond stage juveniles of Meloidogyne ore capable of moving at least 50 cm and penetrating host roots within 7 days. Vertical migrations are faster and greater than horizontal migrations. Juveniles of Meloidogyne javanica are attracted towards host plant roots. Soil and agar from tomato plant culture, germinatlng tomato seeds and root culture attract juveniles of M. Javanica. In addition, movement of juveniles is more important in soils which have supported plant growth. Soil texture influences the migration of Meloidogyne incognito. In natural soils the percentage of juveniles migrating 20 cm decreases when the percentage of clay and silt increases ; no migration is observed when the soil contains more than 30% clay plus silt. However, the clay particles appear to have a function in attracting nematodes over large distances. No migration occures in pure silica sand ; when 5 or 10% of clay are mixed to silica sand 34 and 26%, respectively of the juveniles are able to migrate 20 cm. Temperature influences migrations. The migrations of M. Incognito begin at about 18°C and reache a maximum at 22°C. Juveniles of Meloidogyne hapla have the ability to migrate at lower temperatures than M. Incognita juveniles. Mineral salts exhibit a repulsive effect on Meloidogyne juveniles. Placed in a gradient of mineral salts, these juveniles move preferentiolly towords the region having the lower salt concentration. These nematodes possess sense argons which allow them to orient their movements over gradients of salts created in agar by salt solutions at 0,26 and 10 x 10⁻²M/1. A method of control of Meloidogyne using chemicals which stop their migration is possible. Polycyclic lactones extracted from Hannoa undulata and Hannoa klaineana prevent invasion of tomato roots by stopping movement of javanica juveniles when they are in concentration of 5 ppm in the soil water

Full textSynthetic studies towards pentacyclic quassinoids, namely (-)-samaderine E and (-)-samaderine Y from (S)-(+)-carvone are described. Total synthesis of (-)samaderine Y was accomplished in 21 steps with 1.1% overall yield in which the synthesis was based on a C→CE→ABCE→ABCDE ring annulation sequence. The crucial points included: (1) a regioselective allylic oxidation to functionalize C ring; (2) a stereoselective epoxymethano bridge formation; (3) an intramolecular Diels-Alder reaction resulting in the construction of AB ring; (4) an intramolecular aldol reaction for the construction of D ring; (5) a regioselective allylic oxidation and an alpha-keto acetoxylation to functionalize the A ring.
