Full textA new polyphenolic compound was found in 2011 in maple syrup. It is formed during maple syrup processing of Acer saccharum sap. This compound, named quebecol, is structurally similar to common bioactive agents and showed activity against breast and colon cancer cell lines in some preliminary in vitro biological assays. However, the study of this compound has as yet been limited by its low abundance in its vegetal source. To facilitate further research on this compound by solving its availability problem, we decided to work on the total synthesis of quebecol. In addition to getting access to this natural product, we were interested in developing a general synthetic strategy to produce a variety of 2,3,3-triarylacrylic acid esters, a class of tetrasubstituted olefins whose preparation has not been much explored, and involves many challenges. The following thesis will present our group’s progress over the last years on the synthesis of 2,3,3-triarylacrylic acid esters, the preparation of quebecol and its derivatives, and the evaluation of the anti-inflammatory activity of quebecol and several related structures. First, we will present our synthetic strategy to form quebecol. It involves two key steps. The first is a C-1 homologation reaction on an -ketoester to produce the dibromoalkene analog. This product is used in the second key step: a double Suzuki-Miyaura cross-coupling with the appropriate arylboronic acid. We will also discuss the extension of this strategy to the development of a methodology for the preparation of 2,3,3-triarylacrylic acid esters with one, two, or three different aryl groups. Secondly, we will present our evaluation of the anti-inflammatory activity of quebecol and its derivatives. We will discuss the synthesis of some of these derivatives, including isoquebecol (an unprecedented isomer). Finally, we will establish a series of structure-activity relationships regarding the anti-inflammatory activity of quebecol and its general structure by comparing the tested compounds.
