Full text- S'han sintetitzat les amines, 1-adamantil-9-(9,10-dihidroantril)metilamina i la 1-(1-pirenil)etilamina però en cap cas s'ha pogut avaluar la seva capacitat com a agents de solvatació quiral degut a la dificultat per resoldre els seus enantiòmers.
- S'han estudiat diverses rutes sintètiques per a l'obtenció del 2,2,2-trifluoro-1-(9-tripticil)etanol. Cap de les vies assajades ha donat lloc a l'alcohol desitjat amb rendiments acceptables. En la síntesi del 2,2,2-trifluoro-1-(9-tripticil)etanol via cicloaddició [4+2] el baix rendiment observat s'atribueix a l'electronegativitat del grup trifluorometil. Mentre que en el cas de l'obtenció de l'alcohol en estudi per reacció del 9-tripticil liti amb anhídrid trifluoroacètic i/o trifluoroacetat d'etil, aquesta no ha tingut lloc degut als impediments estèrics que apareixen en aproximar-se l'anió tripticil als diferents electròfils.
- S'ha sintetitzat, amb molt bon rendiment, l'a,a'-bis(trifluorometil)-10,10'-(9,9'-biantril)dimetanol i s'han resolt els seus dos diastereoisòmers. S'ha demostrat l'eficàcia d'aquests compost com a CSA davant de diversos soluts quirals, superant en alguns casos i igualant en d'altres la capacitat solvatant de l'alcohol de Pirkle. S'ha determinat l'estequiometria dels complexos de solvatació formats entre l'lcohol en estudi i diferents soluts observant-se una certa tendència a la complexació 1:1. S'ha realitzat també estudi sobre la seva constant d'associació (K) a temperatura ambient i a baixa temperatura, d'on s'ha pogut concloure que la K augmenta en disminuir la temperatura.
- S'ha aconseguit disposar d'un mètode de síntesi que ens permet obtenir el pirè perdeuterat en quantitats importants i a un cost relativament baix.
- S'ha sintetitzat el macrocicle ciclobis[sulfit del (R,R)-a,a'-bis(trifluorometil)-9,10-antracendimetil]. Dades experimentals i càlculs teòrics han coincidit en afirmar que la cavitat que presenta aquest compost és massa petita com per permetre la inclusió de molècules orgàniques.
- S'ha obtingut el precursor d'un nou macrocicle, el qual aportarà la mida necessària a la cavitat, per la posterior inclusió de molècules quirals en el seu interior.
The present work is based on the synthesis of new chiral solvating agents which are more efficient and versatile than the ones in the current market. In order to reach this objective a number of modifications have been introduced in the structure of the common CSA, such as Pirkle's alcohol, and 1-(1-naphtyl)ethyilamine, and then been evaluated the effect of these changes in the behaviour of these new compounds as chiral solvating agents. From this work several facts and conclusions can be stated. The amines, 1-adamanthyl-9-(9,10-dihydroanthryil)methylamine and the 1-(1-pyrenyl)ethylamine, have been synthesized but their capacity as chiral solvating agents could not be evaluated due to the difficulty of resolving their enantiomers.
Several synthetic methods to obtain 2,2,2-trifluoro-1-(9-tripticyl)ethanol have been studied. However, none of the routes has developed the expected alcohol efficiently. The synthesis of 2,2,2-trifluoro-1-(9-tripticyl)ethanol via cycloaddition [4+2] gives low yieds due to electronegativity of the group trifluoromethyl. On the other hand, the process of obtaining the studied alcohol to reaction of 9 tripticyl litium with trifluoroacteic anhydride and/or ethyl trifluoroacetate, did not work due to the steric effects found in the approach of the litium compound to the electrophil molecules
The compound a,a'-bis(trifluorometihy)-10,10'-(9,9'-biantrhyl)dimethanol has been synthesized with a very good yield and have been separeted efficiently their diastereomers. The good behaviour of this product as a chiral solvating agent behind several enantiomeric molecules has been demostrated. This alcohol has showed a large capacity to induce chirality in racemic mixtures, which is the same, or sometimes even better than the assiciation capacity of Pirkle's alcohol. The stoichiometry of the association complexes has been determined concluding that the ratio is usually 1:1. A study of the equilibrium constant (K) has been carried out and has been demonstrated that K increases when temperature decreases.
A synthetic method has been developed to obtain perdeuterio pyrene in quantity and low cost.
A macrocycle ciclobis[(R,R)-a,a'-bis(trifluoromethyl)-9,10-anthracendimethyl sulphite]has been synthesized. Experimental results and theoretical calculations coincide to confirm that the cavity created in this molecule is too little to permit the inclusion of any organic compound.
A precursor of a new macrocycle has been obtained, which will provide the appropiate size to the cavity in order to be able to include chiral molecules.

Resumo: Esta tese teve como objetivo principal estudar interações intermoleculares empregando a espectroscopia de RMN de H como ferramenta principal. O primeiro capítulo visou "construir" um "hospedeiro quiral" através de interações não covalentes que foi aplicado na discriminação quiral e síntese assimétrica, ambos discutidos, no segundo e terceiro capítulo. Este hospedeiro quiral foi obtido via complexação de calixarenos com aminas quirais ((S)-feniletilamina, (S)naftiletilamina e (R)-2-aminobutanol). A topologia dos complexos foi determinada por RMN de H através de incremento de sinal devido ao acoplamento dipolar observado nas coordenadas girantes entre o hospedeiro (calixareno) e o hóspede (amina). Os experimentos de RMN de H a baixa temperatura foram usados para determinar a conformação preferencial e as mudanças na flexibilidade do calixareno livre e nos complexados. O segundo capítulo, descreve a aplicação dos complexos supramoleculares quirais no reconhecimento quiral e na determinação de excessos enantioméricos de sulfóxidos e ácidos. A importância do solvente, temperatura e topologia foram investigados. O terceiro capítulo focalizou a aplicação dos hospedeiros quirais na redução assimétrica de iminas e sais de metil isoquinolina com boroidreto de sódio. Os excessos enantioméricos são altamente dependentes dos substratos e variam de 0-90%. Finalmente o hospedeiro quiral foi obtido e a topologia totalmente descrita sendo o mesmo aplicado em reconhecimento quiral e síntese assimétrica, consolidando a importância deste tipo de hospedeiro quiral, que pode ser sintonizado para várias aplicações. A RMN de H mostrou ser uma ferramenta bastante eficaz quando aplicada aos estudos de complexos supramoleculares.
Abstract: This thesis concems intermolecular interactions using H NMR as major tooI. The first chapter will focus on the construction of a chiral host using non covalent bondings which will be used in chiral discrimination and asymmetric synthesis both discussed in the second and third chapters. This chiral host was obtained via calixarene complexation with chiral amines ((S)-phenylethylamine, (S)-naphthylethylamine and (R)-2-aminebutanol). The complexes topologies were determined via H NMR signal enhancements due to dipolar cross relaxation in the rotatory frame between the host (calixarene) and the guests (amines). H NMR experiments in variable temperatures were used to access preferential conformation and changes in calixarene flexibility under free and complexed conditions. The second chapter describes the application of chiral supramolecular complexes to chiral recognition and to the determination of enantiomeric excess of sulfoxides and acids. The importance of the solvent choice and topologies was aIso investigated. The third chapter focuses the application of the chiral host (calixarene/amine) to asymmetric reduction of imines and methyl isoquinolonium salts with sodium borohydride. The enantiomeric excesses were highly selective depending on the substrate and ranged between 0-90%. Finally a chiral host was obtained and its topology fully described and applications in chiral recognitions and asymmetric syntheses consolidated the importance of this type of chiral host that can be tuned to the desired application. H NMR techniques have once more proved to be unreplaceable in supramolecular investigations.

Resumo: Neste trabalho foi estudado o comportamento de uma série de iminas estruturalmente análogas quando submetidas à biocatálise com fermento de pão e cenoura. Também foi testado NaBH4 em conjunto com alguns aminoálcoois de diferentes tamanhos, como 1-amino-2-propanol, prolinol e 2-a-hidroxibenzil-benzimidazol ou ácido tartárico. A utilização da biocatálise já é consolidada para a redução assimétrica de carbonilas, mas para a ligação C=N há poucos relatos na literatura, devido à facilidade com que esta ligação sofre hidrólise em meios aquosos. Os resultados foram negativos para biocatálise, apresentando hidrólise do substrato em diversas condições e solventes testados, mesmo com a utilização de 18-C-6 como aditivo e com a imobilização do substrato ou do fermento. As causas destes resultados podem ser tanto a falta de afinidade das enzimas com os substratos testados quanto a instabilidade dos mesmos nos meios utilizados. Foi aplicada uma metodologia bastante simples, com mistura manual de ambos em um almofariz. Testadas várias condições e diferentes solventes, nenhuma delas proporcionou um bom excesso enantiomérico. Uma provável causa para estes resultados pode ser a não eficiência da complexação entre o NaBH4 e os reagentes quirais. Testes de EM e RMN de B e H mostram a formação de um boroidreto quiral, mas não em quantidade suficiente para gerar maior assimetria na redução. Uma grande contribuição deste trabalho é a caracterização de um boroidreto quiral formado a partir de NaBH4 e S-(+)-2- a-hidroxibenzil-benzimidazol já reportado na literatura, mas sem um conjunto de dados conciso acerca de sua estrutura. Este composto gerou o maior excesso enantiomérico 27%, para a redução de N-benzil-(1-feniletilideno)amina
Abstract: This project studies the behavior of a series of imines structurally analogues in bioreductions with baker yeast and carrots, and also with NaBH4 using alternative chiral reagents like 1-amine-2-propanol, prolinol and 2-a- hidroxybenzyl-benzimidazole or tartaric acid. For biocatalysis we investigated different temperatures, solvents and proportions of reagents, but all conditions resulted in hydrolysis of the substrate. The instability of the imines or the lack of affinity can be the cause for these results, even using 18-C-6 as additive and immobilized yeast or substrate. By the chemical imine reduction we tried a simple method of induction, mixing manually NaBH4 and the chiral reagent in a mortar, using different solvents and temperatures, but none of them gave a good enantiomeric excess. Probably there was no coupling between NaBH4 and the chiral reagents, and consequently no preference in for one face in the reduction transition state. MS and NMR analysis have shown the formation of a chiral borohydride, but not an enough amount to give asymmetry in the reduction. A great contribution of this project is the characterization of a chiral borohydride obtained from NaBH4 and S-(+)-2-a-hydroxybenzyl-benzimidazole. It was previously reported, but with no confirmation of its structure. This compound reduced Nbenzyl-(1-phenyletilidene)amine with 27% enantiomeric excess
