Abstract:
The drug Rauwolfia serpentina is known to Indian system of medicine since last many centuries. Because of snake like shape of drug, it has been known as sarpgandha. Although Rauwolfia serpentina contain more than 50 alkaloids but Reserpine is the principal alkaloid of Rauwolfia serpentina. Reserpine has a success application in antihypertensive even at a smaller dose. Rhizomes of Rauwolfia serpentine also have hepatoprotective activity including antihypertensive, hepatoprotective, Rauwolfia serpentina have many other medicinal uses like: Antidiahoerreal, antipsychotic, sedative, anticancer (in breast) etc. Although Rauwolfia serpentina contains major four Indole alkaloids but main object of this context is to provide knowledge about the main active alkaloid Reserpine, having more concentration in the root of plant, play a major role in antihypertensive activity of Rauwolfia serpentina. A much lesser dose of Reserpine is required to provide an antihypertensive effect otherwise it can cause some serious adverse effect like- lethargy, sedation, psychiatric depression, hypotension, nausea, bradycardia, bronchospasm and withdrawal psychosis. Because of its potent activity Reserpine is still used as antihypertensive and sedative agent.

In this study neuroprotective effects of Rauwolfia serpentina plant extract following the exposure to acute immobilization (2h) stress in rats were investigated. The extract of the plant administered orally at non-sedative dose 30mg/kg before immobilization (2h) to observe stress induced behavioral deficits. Neuroprotective efficacy of extract was assessed in terms of alteration in activities of antioxidant enzymes like superoxide dismutase (SOD) and catalase (CAT). We also monitored leptin, corticosterone and glucose levels in plasma to obtain an imminent role of Rauwolfia serpentina. Animals were orally administered with Rauwolfia serpentina (30mg/kg) while controls receive saline (1ml/kg). Each group was subdivided into stressed and unstressed groups. Behavioral deficits were monitored in the open field and light dark activity box. Animals were decapitated; plasma samples were collected for CAT, SOD, corticosterone, leptin and glucose estimation. Orally administered Rauwolfia serpentina attenuates stress induced behavioral deficits and rise antioxidant enzymes levels. Plant extract also prevents the stress-induced increase in corticosterone but glucose levels do not manifest any significant change. Immobilization stress (2h) induced decrease of plasma leptin levels were reversed by Rauwolfia serpentina. Therefore, the present study suggests that Rauwolfia serpentina has potentiality to antagonize undesirable effects of immobilization stress (2h) by reducing stress perception and inhibitory effects of stress on the activity of hypothalamic pituitary adrenal (HPA) axis and animal behaviors. Despite an apparent role of Rauwolfia serpentina the mechanism of action at molecular level causing the acute anxiolytic effects of oral administration of plant extract remains to be determined.

Abstract:
Hypertension is still a prevalent cardiovascular disorder which remains a major global health concern. Rauwolfia serpentina, renowned for its therapeutic potential in managing hypertension, harbors a diverse array of bioactive compounds. This study aimed to elucidate the molecular interactions of chemical constituents derived from Rauwolfia serpentina with key hypertensive targets through molecular docking simulations. Utilizing computational tool, a comprehensive library of phytoconstituents obtained from Rauwolfia serpentina was constructed and subjected to molecular docking analyses against human angiotensin receptor (4ZUD) as target protein. The results revealed significant binding affinities between the chemical constituents of Rauwolfia serpentina and the active sites of these molecular targets. This study bridges the knowledge gap regarding the molecular mechanisms underlying the antihypertensive effects of Rauwolfia serpentina's constituents through computational simulations. The identified compounds exhibiting strong binding affinities and favorable interactions serve as promising candidates for further in vitro and in vivo studies, offering avenues for the development of novel therapeutic agents for hypertension management.

Rauwolfia serpentina is a safe and effective treatment for hypertension. The objective of this case report is to enhance awareness for medical practitioners concerning Rauwolfia induced Parkinsonism, because recognition and appropriate management can enable proper treatment and prevent serious adverse outcomes. We report a patient, who is a known case of Systemic hypertension10 years ago and was taking 5 drops of Rauwolfia syrup, this caused the patient to suffer with rigidity and after discontinuation of Rauwolfia syrup, and full recovery was observed. The appraisal of the ADR reported, in this case, was done by applying the Naranjo probability scale of adverse drug reaction (ADR). For this patient, the Naranjo probability score of 6 was reported.

Abstract:
Objectives: About 80% of world populations are still dependent on herbal plants. Rauwolfia is also one of the wonder drugs of India, which is used since ancient time. It contains a variety of compounds with antioxidant activity and other health benefits. A wild variety of rauwolfia is became endangered due to indiscriminate use. Hence, its cultivation and collection have been started to complete the demand of rauwolfia. Therefore, the study was designed to evaluate and compare the in vitro antioxidant and antidiabetic activity of the wild and cultivated plant of rauwolfia. Methods: The methanolic extract of wild and cultivated plant was subjected to the DPPH and alpha-amylase inhibition activity for antioxidant and antidiabetic activity, respectively. Result: The study revealed that the wild and cultivated variety of Rauwolfia serpentina does not have a significant difference in their antidiabetic and antioxidant activities. Conclusion: On the basis of the in-vitro studies, it can be concluded that cultivated variety of the plant can be used as a substitute for a wild variety of R. serpentina.

Abstract:
Introduction: Hypertension (high blood pressure) is an increasingly important global medical and public health problem that is linked to a higher risk of developing cardiovascular disease. Researchers in both developed and developing nations have identified it as one of the most widespread diseases currently present. The investigators are focusing on plants for better, safer, more affordable, and more effective alternatives due to unwanted risks and negative effects of synthetic pharmaceuticals. Allium sativum possesses anticarcinogenic, antioxidant, antidiabetic, renoprotective, anti-atherosclerotic, antibacterial, antifungal, and antihypertensive properties while Rauwolfia serpentina is known to possess pharmacological actions such as hypnotic effect, sedative effect, antihypertensive activity, and anti hypercholesteremic activity. Methods: Sixty rats were divided into six groups, each containing ten rats. With the exception of the negative control group, rats from all groups were given 1% NaCI solution as water and high fat diet as food to induce hypertension. After that, among the six groups one group was treated with Allium sativum, one group with Rauwolfia serpentina and one with the mixture of Rauwolfia serpentina and Allium sativum. Results and Discussion: In case of lipid profile and blood pressure (systolic blood pressure, diastolic blood pressure, and mean blood pressure) both Rauwolfia serpentina and Allium sativum significantly (p<0.05) reversed disturbed pathological state, when compared with disease control group. However, Rauwolfia serpentina demonstrated the highest degree of activity for anti-hypertensive activity (Mean Blood Pressure 89.79±5.84 mmHg) and Allium sativum is more potent for controlling abnormal lipid profile (Total Cholesterol 125.39±5.59). Conclusion: Therefore, this study provides scientific evidence that Rauwolfia serpentina and Allium sativum have considerable anti-hypertensive and anti-lipidemic activities. More research needs to be done on these plants to isolate particular therapeutic constituents, so that it can be applied more precisely and accurately to mitigate the respective disease.

Abstract:
This study investigated the effects of ethanol and aqueous root extracts of Rauwolfia vomitoria Oliv. on tocopherol, adenine deaminase, and antioxidant parameters in albino rats with induced arthritis. A total of 135 albino rats were utilized, divided into nine groups of 15 rats each. Arthritis was induced by intradermal injection of 0.1 ml Chicken type II collagen-Complete Freund’s adjuvant into the left hind paw. Starting on day 10 postinduction, the rats were treated with the extracts at dosages of 400, 600, and 800 mg/kg body weight over a 32-day period. Analysis was performed using spectrophotometric techniques. Arthritic rats showed significantly higher (P<0.05) levels of malondialdehyde (MDA) and nitric oxide (NO), and significantly lower (P<0.05) levels of glutathione peroxidase, catalase, superoxide dismutase (SOD), reduced glutathione, and tocopherol compared to normal controls. Treatment with Rauwolfia vomitoria extracts reversed these changes in a time- and dose dependent manner. The anti-arthritic efficacy of the extracts was comparable to that of the standard drug indomethacin (P<0.05). The findings suggest that ethanol and aqueous root extracts of Rauwolfia vomitoria contain compounds that enhance antioxidant levels, reduce oxidative stress, and alleviate arthritic symptoms in adjuvant-induced arthritic rats. This study provides scientific evidence supporting the use of Rauwolfia vomitoria root extracts as a potential treatment for arthritis. Abstract:
The aim: development of the composition and technology of a medicinal drug in the form of capsules with a dry extract of Rauwolfia for the treatment of hypertension. Materials and methods. Experimental samples of masses for encapsulation contained 2.0 mg of dry Rauwolfia extract and various excipients used in the technology of solid dosage forms. The study of pharmaco-technological characteristics was carried out on the devices of the company "Pharma Test" (Germany). The time of decay was determined on the device of the company "Erweka" (Germany). A comparative analysis of the cost calculation of the production of tablets and capsules with dry rauwolfia extract was carried out by the "cost plus" method. Results. The use of PROSOLV® SMCC HD 90 improved the flowability, homogeneity and homogeneity of the mass with Rauwolfia dry extract powder, considering the low dosage of the substance of 2 mg. It has been experimentally proven that PROSOLV® SMCC HD 90 shortens the disintegration time as a disintegrant, because due to silicate moisture easily penetrates into the MCCC, hydrophilic bridges are formed, wettability increases and mass swelling occurs. The multifunctionality of PROSOLV® SMCC HD 90 three-in-one excipient, which has the properties of a filler, a disintegrant and a glidant, makes it easy to apply direct encapsulation technology, replace and reduce the number of excipients and thus increase production efficiency. Conclusions. When developing the composition of capsules with dry rauwolfia extract, the effect of various excipients on the pharmaco-technological properties of encapsulating masses and ready-made capsules was investigated. The combined excipient, namely PROSOLV® SMCC HD 90, having the properties of a filler, a disintegrant and a lubricant, in direct encapsulation technology is more effective in influencing the fluidity of the mass and disintegration of the capsules. The introduction of the direct encapsulation method will allow to expand the range of new medicines and improve existing technologies, in particular in the form of tablets, which are widely produced in industrial production. it is possible to attach specifically to Rauwolfia

Abstract:
Introduction. Rauwolfia vomitoria is a plant used as a sedative and in the treatment of psychotic tendency. This study was on the teratogenic effects of its root bark and leaf extracts on Wistar rat’s fetal femurs. Materials and Methods. Twenty-five female rats weighing between 180 and 200 g were divided into 5 groups, of 5 rats each. Group A was the control, while Groups B, C, D, and E were the experimental. The female rats were mated with mature male rats to allow for pregnancy. Groups B and C animals received orally 150 mg/kg each of the root bark and leaf extracts of Rauwolfia vomitoria, respectively, while Groups D and E animals received 250 mg/kg bodyweight each of the root bark and leaf extracts of Rauwolfia vomitoria, respectively, from day 7 to day 11 of gestation. On day 20 of gestation, the rats were sacrificed, the fetuses were examined, and their femurs were dissected out and preserved, decalcified, and routinely processed using the Haematoxylin and Eosin staining method. Results. Histological observations of the fetal femur bones showed numerous osteoblast and osteoclast, hypertrophy, and hyperplasia of bone cells compared with the control. Conclusion. Ethanolic root bark and leaf extracts of Rauwolfia vomitoria may lead to advanced skeletal development.

The wood anatomy of Aspidosperma australe Müll. Arg., Aspidosperma polyneuron Müll. Arg. and Rauwolfia sellowii Müll. Arg. are described, based on samples collected in the province of Misiones, Argentina. The three species showed the most important anatomic features, cited in the literature for Apocynaceae. Aspidosperma australe is identified by its homogeneous and predominantly uniseriate rays. Aspidosperma polyneuron has mostly biseriate rays and sclerotic tyloses in vessel members. Rauwolfia sellowii shows larger rays and no tyloses in its vessel members.

Abstract:
In this present study, a protocol was developed for the DNA profiling of medicinally important Rauwolfia species (05 species) by ISSR-PCR method. Initially genomic DNA was isolated from the fresh leaves (1 gram) of 24 months old Rauwolfia tetraphylla, R.serpentina, R.pentaphylla, R.vomitaria and R.micrantha. The content of genomic DNA isolated from 1 gram of fresh leaves of these five species ranges from 1.6 to 3.4 µg. Out of six inter simple sequence repeats (ISSR) primers used, HY3 primer has given the good amplification with distinct bands which were reproducible. The range of DNA bands produced by the HY3 primer were between 300 bp to 1500 bp among the five species. A dendrogram was constructed on the basis of similarity matrix data by unweighted pair group method with arithmetic average (UPGMA) cluster analysis using indigenous software. Average genetic similarity generated by HY3 has revealed 38% similarity between species of Sub cluster-A and species of Sub cluster-B which is consistent with their geographical distribution i.e. Sub cluster-A species R.tetraphylla and R.serpentina inhabit peninsular India, Sub cluster- B species R.micrantha and R. vomitoria inhabit the Himalayan foot hills. The developed protocol of DNA profiling in Rauwolfia species by ISSR-PCR method could be used efficiently for the differentiation and identification of the Rauwolfia species at molecular level.

Rauwolfia tetraphylla L., an endangered species, is rich in various phytochemicals and often used as a substitute of Rauwolfia serpentina and therefore aimed to produce nanopowders from different parts of this plant. These biosynthesized nanopowders were characterized by UV-Visible Spectroscopy, Scanning Electron Microscopy, X-Ray Diffraction and Fourier Transform Infrared Spectroscopy. Further bioactive components of root, stem and leaf nanopowders were analysed by Gas Chromatography-Mass Spectrometry(GC-MS), and different compounds were identified by comparison of GC-MS spectrum with library searches. Methanolic root, stem and leaf nanopowders were evaluated for antibacterial and antifungal activity. Antibacterial activity was assessed against human pathogenic bacteria Klebsiella pneumoniae, Salmonella typhi, Vibrio cholerae, Corynebacterium diphtheriae, Vibrio parahemolyticus. Human pathogenic fungi Aspergillus fumigatus, Candida albicans, Mucor hiemalis were selected for evaluating antifungal activity. The present study confirms the good antibacterial and antifungal activity of Rauwolfia tetraphylla synthesized nanopowders. As GC-MS analysis revealed different phytoconstituents, other pharmacological activities need to be evaluated.

Rauwolfia serpentina Benth. is a tropical fruticose plant that is able to produce and concentrate indole alkaloids, especially ajmaline and its derivatives, which are the most effective medications against ventricular arrhythmia with low side effects. Aim of the study. Determination of the qualitative and quantitative content of indole alkaloids in cell biomass of Rauwolfia serpentina tissue culture, obtained by the prolonged in vitro growth. Materials and methods. Object: cell biomass of Rauwolfia serpentina tissue culture (K-27 strain), obtained by methods of long-term cell selection in vitro. Alkaloids content determination: TSQ Vantage LC-MS (ThermoFischer Scientific). Results. 20 indole alkaloids are found in cell biomass of Rauwolfia serpentina tissue culture (K-27 strain). The highest content is registered for ajmaline and its derivatives (0.690 % mass. for ajmaline). The contents of reserpine and yohimbine were found to be as low as 0.009 % and 0.020 %, respectively. Conclusions. It is established that the content of indole alkaloids is higher in K-27 strain in comparison to natural plant and is stable over more than 30 years of its growth. Total alkaloids content was found to be 2.8 % of dry cell biomass, and total ajmaline-type alkaloids content (including ajmaline) was found to be 1.6 % of dry cell biomass. In contrast, the total alkaloid contents in the natural plant material is reported to be in the range of 0.8–1.3 %.

Methodology: The experiment was carried out in six groups, each having 15 specimens for 24, 48, 72, and 96 hr. Group I served as control. Groups II and III were maintained with 3 mg l-1 of Rauwolfia root extract and 0.4 mg l-1 of Cu2+, respectively. Groups IV, V, and VI were simultaneously co-exposed with 0.4 mg l-1 of Cu2+ and three different concentrations of Rauwolfia root extract 1, 2 and 3 mg l-1, respectively. For the genotoxicity assessment, blood and kidney tissues were used. Hepatic impairments were assessed after each exposure period. Results: A significant increase (p<0.05) in chromosomal aberrations, micronuclei frequency, activity of liver enzymes (SGOT, SGPT and ALP) and a decrease in protein level were recorded in Group III in comparison to the control. Groups co-exposed with Cu2+and Rauwolfia serpentine showed a significant (p<0.05) decrease in cytogenetic biomarkers, activity of liver enzymes and an increase in protein levels, as compared to Group III, with respect to control in a dose dependent manner. The enzyme is a soluble protein and catalyzes the transfer of a glucose moiety from UDPG to a wide variety of phenolic compounds with p-nitrophenol as one of the best substrates (Km = 1.21 mM, UDPG = 0.54 mM). In contrast to the membrane-bound UDPG: vomilenine-21-OH-β-D-glucosyltransferase from Rauwolfia serpentina cells, this enzyme is not able to glucosylate indole alkaloids. The enzyme activity has been detected in 14 callus cultures belonging to 10 different plant families.

Groups IV, V and VI fish were exposed to three different concentrations of ethanolic root extract of Rauwolfia serpentina (1, 2 and 3 mg l-1), simultaneously with 7.68 mg l-1 of Cr (VI). The induction of micronuclei, chromosomal aberrations, protein levels and liver enzymes-SGOT, SGPT and ALP- were assessed in fish of all six groups after designated exposure periods. Results: A significant induction (p<0.05) in chromosomal aberrations, micronuclei frequency and activities of liver enzymes (SGOT, SGPT and ALP) coupled with reduced protein level was recorded in Group III as compared to the control. Whereas, a significant (p<0.05) decrease in the frequency of chromosomal aberrations, micronuclei induction and activities of liver enzymes together with an increase in protein level was observed in Group IV, V and VI with respect to control and Group III. Abstract A method is presented for the determination of reserpine and rescinnamine in Rauwolfia serpentina powder or tablets by liquid chromatography (LC) with fluorescence detection. The sample is dispersed in CH3OH, 0.5N H2S04 is added, and the mixture is extracted with five 30 mL portions of CHC13. The extracts are separated from interfering materials on a Celite-O.IN NaOH column, and the eluates are collected in 50 mL CH3OH. After complete removal of the CHC13, reserpine and rescinnamine are determined by liquid chromatography on a normal phase column with CH3OH as the mobile phase. Because reserpine and rescinnamine produce a single peak, chromatograms are obtained at different wavelengths. Reserpine is determined at an excitation wavelength of 280 nm and an emission wavelength of 360 nm. Rescinnamine is determined at an excitation wavelength of 330 nm and an emission wavelength of 435 nm. Recovery studies were conducted at 2 different levels to simulate 100 and 50 mg Rauwolfia serpentina tablets. Samples of Rauwolfia serpentina powders and tablets were also examined, and the results were compared with those obtained by the current AOAC official method. The proposed method is applicable to the analysis of ground composites and individual tablets.

A novel enzyme was isolated, partially purified (217-fold) and characterized from cell suspen­sion cultures of Rauwolfia serpentina Benth. The enzyme catalyzes one of the late biochemical reactions in the biosynthesis of ajmaline by hydrolysis of 17-O-acetylated alkaloids of the ajmalan group forming the appropriate deacetylated compounds. This esterase exhibits an unusually high substrate selectivity and exclusively accepts acetylated ajmaline derivatives with the naturally occurring 2β(R)-configuration. The properties of the enzyme were determined showing an optimum pH at 7.5, an isoelectric point of pH 4.9 and a relative molecular weight of 33 ± 2 kDa. Inhibition studies of enzyme activity point to the necessity of SH-groups. The esterase seems not to be inhibited by ajmaline. the end product of the pathway. The highest enzyme activities were observed in leaves and cell suspension tissues of the tribe Rauwolfieae which are known to synthe­size ajmaline and its congeners. The specific function of the esterase in the biosynthesis of the later alkaloids was established.

Two different methods of in vitro multiplication of Rauwolfia serpentina from nodal explants were compared viz. multiplication via callus morphogenesis and that via shoot proliferation from axillary buds. The second method was found to be far better. The optimum shoot proliferation occurred on Murashige and Skoog (MS) medium supplemented with 1 mg/L naphthalene acetic acid (NAA) and 2 mg/L of benzyl aminopurine (BAP). The best rooting of shoots occurred on MS medium containing 4% sucrose and 1 mg/L of NAA. Solid and liquid MS media were found to be similar in supporting shoot proliferation. The plants produced were successfully hardened and established in soil. An easy, reliable and reproducible protocol was developed for in vitro micropropagation of Rauwolfia serpentina from nodal explants.

Several social, psychological and biological factors could be cause for central nervous system disorders, especially anxiety; the latter could lead to more serious neurological and psychosomatical disorders. Sedative medications of plant origin are widely used practically as an alternative and/or additive to traditional pharmacological treatments of anxiety, hyperexcitation and other neuroses. Thus, the search of new effective sedatives based on plant extracts is an actual and perspective task. Aim of the present work is to investigate the sedative effect of biomass extract from cell culture of Rauwolfia serpentina K-27 strain. Sedative action of biomass extract from cell culture of Rauwolfia serpentina high-productive strain was determined by open field method on 24 white mice males (no line). Mice weight was in 18–25 g interval. The behavior was assessed by usual behavioristic acts: motor activity, psychoemotional activity and tentative research reaction. Animals were randomized into 3 groups of 8 individuals each. Rauwolfia serpentina biomass extract at doses of 7.5 mg/kg (group 1) or 15 mg/kg (group 2) was administered orally as an aqueous solution. Animals of the control group (group 3) received a solvent (distilled water). The sedative effect of extract was discovered in doses of 7.5 and 15 mg/kg, that showed a decrease in horizontal motor activity to 63.31 and 79.76% respectively, vertical activity to 82.05 and 71.79%, grooming duration to 57.28 and 60.78% at experimental days 20 to 28. Tentative research reaction demonstrated the direct dependence of sedation intensity on duration of extract administration. As a result of the conducted researches significant differences in the number of fecal bolus of both experimental and control groups of mice were not revealed. No significant difference between the sedative effect and the dose (7.5 and 15 mg/kg) was observed. The sedative effect of biomass extract from cell culture of Rauwolfia serpentina K-27 strain was proven. This leads to perspective inclusion of the extract to traditional medicine in prophylaxis and treatment of insomnia and of physical and psychological overexcitation.

At the end of all experiment mice brain were removed and TBARS level, SOD level and infract size were determined.Results : Occlusion of both common carotid arteries significantly increased escape latency time(ELT) during acquisition trial and decrease, time spent in target quadrant on morris water maze, an increase in escape latency time was also observed on elevated plus maze when measured after 7 days of occlusion.Furthermore, TBARS levels, SOD levels and infract size were also increased when measured after 12 days of occlusion. Administration of RS specially at a dose of 20mg/kg and 40mg/kg;p.o. significantly attenuated these alteration and show neuroprotective effect against oxidative stress, infract size and learning and memory impairment. Conclusions: These finding suggest that Rauwolfia serpentina may have a promising role as a prophylactic drug in stroke induced experimental dementia due to its neuroprotective effect. The phenolic compound like phenolic acid, flavanoids and tannins present in the plant.

Abstract:
This study evaluated the effects of ethanol and aqueous root extracts of Rauwolfia vomitoria and Olax subscorpioidea on Chicken type II collagen in Complete Freund’s Adjuvant-induced arthritis in albino rats. A total of 225 albino rats, weighing between 120-200 g, were used and divided into 15 groups of 15 rats each. Rheumatoid Arthritis (RA) was induced by administering 0.1 ml of Chicken type II-Complete Freund’s Adjuvant into the left hind paw. The study duration was 32 days, during which the arthritic rats were treated with Rauwolfia vomitoria and Olax subscorpioidea root extracts at doses of 400 mg, 600 mg, and 800 mg/kg body weight. The sub-lethal toxicity study revealed significant increases (P<0.05) in WBC, platelet, and differential counts, along with significant reductions (P<0.05) in RBC count, PCV, and HB in the adjuvant induced arthritic rats compared to the negative control group. However, treatment with the plant extracts (groups 4-15) significantly reversed these trends towards normal levels compared to the untreated arthritis group (group 2) within the 32-day period. The ameliorative effects of the extracts were both time- and dose dependent (P<0.05). This study concludes that the ethanol and aqueous root extracts of Rauwolfia vomitoria and Olax subscorpioidea effectively ameliorate arthritis symptoms in albino rats, potentially due to the presence of active chemical constituents. These findings provide scientific evidence supporting the therapeutic use of these plant extracts in managing arthritis.

Abstract:
In Côte d'Ivoire, people use Rauwolfia vomitoria (Apocynaceae) as traditional medicine for the treatment of diabetes mellitus. The purpose of this study is to bring out the potentiel antidiabetic effect of an aqueous extract of roots devoid of Rauwolfia vomitoria bark (EARv), in rats made diabetic by alloxan monohydrate, a diabetogen substance, compared with the effect of glibenclamide, a reference antidiabetic substance. For this, the blood glucose level and weight evolution of healthy rats (normoglycemic), diabetic control rats and diabetic rats treated with EARv or glibenclamide are measured, as well as variations in transaminase levels during 28 days of experimentation. Administration of alloxan induces in rats made diabetic characterized by a significant and permanent increase in blood sugar, a decrease in body mass, followed by an increase in the level of serum transaminases, a sign of liver toxicity in rats. In rats made diabetic and treated daily with EARv at a dose of 1000 mg/kg body weight (B.W), there is a decrease in blood sugar and an increase in body mass, in the sense of their normalization, during the 28 days of treatment. In diabetes rats, EARv also results in normalization of serum alanine aminotransferase (ALT) concentration and a significant decrease in aspartate aminotransferase (AST). These effects of EARv at 1000 mg/kg B.W are similar to those of glibenclamide at 10 mg/kg B.W. These results show that, just like glibenclamide, EARv is an antidiabetic, hepatoprotective substance and tends to correct body mass loss in diabetic rats. These effects of EARv justify the use of Rauwolfia vomitoria in traditional medicine in the treatment of diabetes mellitus.

Design of novel herbal drugs with a wide range of activity is an important object of pharmacology. Herbal remedies having antiarrhythmic, cardiotonic and vasodilatory effects are useful for treatment of heart arrhythmias. Vasodilating drugs are prescribed for medication of hypertension and migraine. Also they are a part of comprehensive treatment of various diseases such as disturbed peripheral blood circulation and atherosclerosis of arteries of extremities as well as problems with urination and potency. Search for novel herbal preparations with vasorelaxant activity in our view is a promising matter of current interest. This work is aiming in study of vasodilatory activity of methanol extract of strain K-27 tissue culture of Rauwolfia serpentina. Evaluation of vasodilatory activity extract of high-productive strain K-27 tissue culture of Rauwolfia serpentina has been carried out by the method of auxotonic mechanography of vascular smooth muscles. Strength and frequency of spontaneous contractions of portal vein as well as relaxation of aortic smooth muscles activated with phenylephrine were evaluation indicators of extract activity. In order to estimate working concentration of extract of strain K-27 tissue culture of Rauwolfia serpentina we have studied vascular activity at sequentially increasing concentrations of extract solution on the portal vein. Overlay of sequence of growing concentrations exhibited decrease in basal level of venous tone and dose-dependent suppression of phasic contractions with complete inhibition of spontaneous activity of portal vein at 1.44 mg/ml concentration and uprise of toxicity at higher concentrations. It has been shown dose-dependent relaxation of vascular preparations followed by development of α-adrenoceptor blocking effect and loss of sensibility to phenylephrine at the end of experiment. After discontinuing exposure with a substance the activating effect of phenylephrine remained not renewed during 30-50 minutes, at the same time contractile activity of aorta on the response to the other type activation (60 mM K+) took place. It was found that at the studied range of concentrations: 0.0288 μg/ml–28.8 μg/ml the extract of strain K-27 tissue culture of Rauwolfia serpentina has distinct vasodilatory effect. Also it was discovered α-adrenoceptor blocking effect of that extract, so it can find wide application in therapy as vasorelaxant and α-blocker, for instance in treatment of prostate diseases.
