En los últimos años, diferentes técnicas y frameworks han surgido con el fin de proveer una solución para crear aplicaciones cross-platform. El objetivo es desarrollar un solo código que puede ser desplegado en diferentes sistemas operativos y proveer una experiencia completamente nativa. Desde 2015, React Native ha sido promocionado como una opción para desarrollar aplicaciones móviles con el objetivo de escribir el código una sola vez y desplegarlo en cualquier plataforma perfectamente. Al momento no hay otra librería JavaScript que permita el desarrollo de aplicaciones móviles nativas. Este trabajo realiza un análisis experimental con una visión imparcial, basada en la experiencia del desarrollo de aplicaciones móviles utilizando React Native y código nativo. Analiza varios escenarios para elegir analíticamente código nativo, React Native o una combinación de ambos para desarrollar una aplicación móvil. Algunas conclusiones encontradas sostienen que la elección de la tecnología depende de la naturaleza del proyecto, tiempo, presupuesto, experiencia del equipo de desarrollo y los requerimientos de la aplicación.

Abstract:
Cross-platform framework is becoming more and more popular. Many giant tech companies have their own offering with different programming languages. With so many choices, sometimes it is difficult for a start-up developer to choose which framework they will use for their project. Therefore, this research aims to give insight for start-up developers on which cross-platform framework is better between Flutter and React Native by comparing their performance. This research achieved its goal by creating two identical mobile applications using Flutter and React Native and then comparing their performance using a theoretical framework that measures load time, average frame rate, and memory usage. The result of this testing is that Flutter has a shorter load time at 1.69 second compared to React Native at 4.26 second and lower memory usage at 6.52MB compared to React Native at 27.6MB. Their average frame rate is very comparable hovering around 60 frames per second. With faster load time and lower memory usage Flutter has better performance wise compared to React Native.

Mobile application development was growing very rapidly along the increase of mobile devices and technology. An application must be built at least 2 (two) types according to the current popular operating system (Android and iOS) in order to be able to get large application users.Hybrid Application is an application development model that combines native application approach and web application to be a mobile application. It can be converted into more than one mobile operating system platform later. The advantages of hybrid applications is the rapid development process because one source code can run on more than one mobile operating system, while the disadvantage is it use more of resources than the application built with the original language of the operating system platform used.Adobe Phonegap (Phonegap) and React Native are two frameworks that can be used to build mobile hybrid applications. Since Covid-19 was declared a global pandemic because it has spread throughout the world, every effort has been made to help prevent and tackle the transmission of Covid-19, including information technology. Information technology developed to determine the shortest distance for Covid-19 cases around us needs to be developed. This research implements Dijkstra's Algorithm written in the React Native programming language to build a Covid-19 tracking application. The system can display the closest distance with a radius of at least one meter, and the test results can map the nearest radius of 41 meters and the most immediate radius of 147 meters. This system is built for the compatibility of Android OS and iOS applications with React Native programming.

This paper outlines the findings of a two-year evaluation of learner use of software tools in a multimedia software package for teaching Indonesian listening comprehension and culture. This software package was introduced into the Introductory language program to help address the need of total beginners for exposure to native speakers using daily language in ordinary ways, on common, everyday topics. The package is designed to introduce, develop, and reinforce appropriate learning strategies at the same time. Data on learner perceptions of the ease of use, and appropriateness of the software interface were investigated, in order to improve our understandings of the perceptions and reactions of novices compared to experienced users of multimedia software packages. This investigation includes an analysis of the differences in strategy use between high, medium and low proficiency learners, and their use of the software Help tools such as grammar reference notes, language laboratory simulation, replay facility, and mid-task answer checking.

Di Indonesia terdapat banyak platform donasi online, namun kebanyakan donasi yang diberikan berbentuk uang. Padahal kebutuhan pertama para korban bencana alam bukanlah uang melainkan barang primer. Selain itu, berdasarkan Laporan Tren Preloved Indonesia 2017 oleh Carousell, di Indonesia terdapat 82% dari 1.000 responden menumpuk barang yang tak terpakai dirumahnya. Maka dari itu, dibuatlah aplikasi donasi barang berbasis mobile, dimana barang yang didonasikan merupakan barang-barang bekas yang sudah tidak terpakai. Tujuannya yaitu memudahkan pengguna untuk berdonasi barang, dimana pun dan kapan pun. Aplikasi dibangun menggunakan framework React Native dengan RESTFul API dan MySQL sebagai bahasa pemrograman database. Pengujian yang dilakukan yaitu pengujian fungsional sistem dengan menggunakan teknik black-box testing dan User Acceptance Test (UAT) serta pengujian dengan menggunakan kuesioner. Berdasarkan hasil pengujian, fungsional sistem berjalan sesuai dengan requirement yang telah didefinisikan. Hasil pengujian tersebut berdasarkan dari hasil black-box testing 100% dan UAT 100% serta kuesioner dengan rata rata nilai 87,4%.

However, if the note must contain details of the transactions carried out, it can be considered less efficient. The use of Optical Character Recognition will be able to answer this problem, by taking a picture of the transaction, all transaction details will be recorded on the smartphone, and the user can validate the results obtained and save the record on the smartphone user. Users can also immediately see the total transactions made according to the selected time range without having to calculate each transaction made manually. The application will be designed using the react native framework which allows it to run on various platforms.

Dengan demikian, hal tersebutmasih menjadi salah satu parameter dalam penilaian kinerja individu maupun perusahaan. Olehkarena itu dibutuhkan sistem yang dapat memonitoring kehadiran pegawai berbasis mobile agarlebih efektif dan efisien. Penelitian ini menggunakan metode analisa PIECES, permodelansistem menggunakan UML (Unified Modelling Language) untuk menggambarkan secaravisualisasi, lalu diimplementasikan dengan bahasa pemrograman PHP dan JavaScript,Framework React Native dan basis data MySQL sebagai database. Synthetic peptides representing different areas of the CEA molecule were used as immunogens for the development of anti-CEA antibodies. Both polyclonal and monoclonal antibodies were generated using peptides composed of CEA amino acid positions 99–128 and 585–613, respectively. One MAb, designated CP4, generated using the CEA peptide 99–128, was chosen for a more detailed analysis of reactivity. MAb CP4 reacts in solid phase RIAs with CEA peptide 99–128 immunogen and purified native CEA. CP4 did not react with purified non- specific cross reacting antigen (NCA), even though there were two single amino acid differences between NCA and CEA in the 29 amino acid peptide. The affinity constants of CP4 for the CEA peptide 99–128 and native CEA are 4.07 × 109 M−1 and 5.75 × 108 M−1, respectively. When CP4 was reacted with purified CEA in Western blotting experiments, the Mr 180,000 glycoprotein characteristic of CEA was detected, but CP4 reacted to various size entities in tumor cell extracts. The results of liquid competition RIAs showed that the epitope that MAb CP4 recognized on native CEA is not available for binding when CEA is in solution. Physical (adsorption to a solid matrix) or chemical (deglycosylation or formalin-fixation) alteration of CEA is required for binding of CP4 to CEA. MAb CP4 reacted approximately 1,000-fold greater to deglycosylated CEA than native CEA. Immunohistochemical studies using formalin-fixed paraffin-embedded tissue sections demonstrated that, among carcinomas, CP4 reacts selectively with colorectal carcinomas, while normal colon is negative. Although stomach carcinoma is negative, dysplastic lesions and areas of intestinal metaplasia are reactive. Two of 7 normal stomach tissues showed focal cytoplasmic reactivity of the surface epithelium. CP4, therefore, appears to react with an epitope with highly restricted expression in colorectal carcinoma. These studies demonstrate the complexities in dealing with an anti-peptide MAb with reactivity to an epitope which is accessible only under certain conditions.

The use of fluorescence microscopy to investigate fungal growth in wood often causes interference due to the strong autofluorescence of wood lignin, unless fluorescent probes which specifically react to fungal hyphae, are used. Techniques to enable differentiation of hyphae from wood have been recently reported (Singh et al., 1997; Xiao et al., 1997). The authors demonstrated that while glutaraldehyde can be used to detect fungal native proteins, wheat germ agglutinin (WGA) which reacts with cell wall chitin of hyphae, considerably improved detection of fungi growing in wood.

Pada proses bisnis distro ini, ternyata para pelaku bisnis distro ini menjalankan bisnisnya secara manual tanpa memanfaatkan perkembangan teknologi saat ini, sehingga mereka masih kesusahan dalam melakukan pencatatan transaksi, manajemen produk, dll. Berdasarkan permasalahan tersebut, maka perlu untuk membuat aplikasi Point of Sales untuk mempermudah pegawai distro dalam pekerjaannya. Metode penelitian yang digunakan pada penelitian ini adalah classic life cycle dan untuk pengumpulan data yaitu dengan melakukan wawancara, observasi dan dokumentasi pada distro yang dijadikan tempat penelitian. Berdasarkan hasil dari pengumpulan data, maka dibuatlah rancangan sistem, rancangan database dan rancangan antarmuka. Rancangan tersebut kemudian dituangkan kedalam coding untuk membuat aplikasi dengan menggunakan framework react native dan setelah aplikasi selesai dibuat, maka dilakukan pengujian terhadap aplikasi tersebut dengan metode black box testing. Hasil dari pengujian yang telah dilakukan sudah sesuai dengan yang diharapkan sehingga aplikasi Point of Sales ini berhasil dibuat dan sudah disesuaikan dengan kebutuhan pihak distro.

Surface modification is achieved by coating the aluminum surface with a powder mix of {Si + flux + a metal (Me)} and heating at ∼600 °C for a time interval of the order of 1 min. During heating, the flux melts and dissolves the native surface oxide film on aluminum, thus allowing Si to react with the substrate to form a film of liquid Al-Si alloy of near-eutectic composition. The component Me of the powder coating then reacts with the molten metal layer to modify the metallurgical properties of the surface after solidification.

Grammaticality judgments reflect a compound product of both grammatical and processing factors. But because they interact in a symbiotic way, very often grammatical and processing constraints are difficult to separate. According to generally accepted grammatical theory, (a) Who do you think John told Mary he fell in love with? and (b) Who do you think John told Mary fell in love with Sue? are equally grammatical. We have observed, however, that native speakers strongly accept sentences like (a) as grammatical but react quite variably to sentences like (b). A possible explanation is that native English speakers exhibit a processing preference, in searching for the extraction site for the wh- word, for object position over subject position. Proficient nonnative judgmental data offer additional support for a processing account. Nonnatives whose L1 grammars do not bias them toward objects also show preferences similar to those of natives. We provide a processing account based on Frazier's Minimal Attachment principle.

Abstract:
The properties of two carnitine acyltransferases (CPT) purified from bovine liver are compared to confirm that they are different proteins. The soluble CPT and the inner CPT from mitochondria differ in subunit Mr, native Mr, pI and reactivity with thiol reagents. All eight free thiol groups in soluble CPT react with 5,5′-dithiobis-(2-nitrobenzoate) in the absence of any unfolding reagent, and activity is gradually lost. The inner CPT activity is completely stable in the presence of 5,5′-dithiobis-(2-nitrobenzoate), and only one thiol group per molecule of subunit is modified in the native enzyme. Antisera to each enzyme inhibit that enzyme, but do not cross-react. CPT activity in subcellular fractions can now be identified by titration with these antibodies. The soluble CPT from bovine liver is probably peroxisomal in origin, but, although antigenically similar, it differs from the peroxisomal carnitine octanoyltransferase found in rat and mouse liver in its specificity for the longer-chain acyl-CoA substrates.

Pada penelitian ini menggunakan metode Research and develpoment (R&D) dengan model penelitian ADDIE yakni, Analysis, Design, Development, Implementation, and Evaluation. Hasi dari penelitian ini adalah aplikasi yang diimplementasikan dengan bahasa pemrograman Javascript dengan framework react native dan database yang digunakam yaitu firebase. Hasil yang diperoleh dari pengujian black box dan pengujian whitebox, yaitu semua proses dapat bekerja dengan baik dan sesuai dengan fungsinya. This contribution discusses the relationship between sport and citizenship by exploring the citizenship paths described by young cricketers of Bangladeshi origin living in Venice (Italy). In particular, it focuses on the processes of misrecognition, enacted both by natives and their older countrymen, that these youths are suffering in their everyday life and that are rooted and reflected in their playing cricket in the neighbourhood. Starting from these premises, their aspired citizenship paths are described, revealing how the European passport, often an aspiration in itself, may become a passe-partout to react to misrecognition, allowing them to describe aspirations, reflected also in their sports practices, that are nationally, transnationally and globally deployed and that may aim, although through an individual claim, to restore the disruptions lived by the whole Bangladeshi diaspora. In this sense, within their distinctive aspired citizenship paths, the borders between distinction/integration with their older countrymen and native people are blurred, thus revealing their willingness to enjoy the same rights as their native peers as well as to overcome the differential inclusion suffered by their parents.

Bilinguals react to cultural information in a language-dependent fashion, but it is unknown whether this is influenced by the individual’s emotional state. Here, we show that induced mood states increase cultural bias—measured using the Implicit Association Test (IAT)—but this effect occurs asymmetrically across languages. In the native language, bilinguals show a strong cultural bias, which is not influenced by mood. But in the non-native language, a relatively low cultural bias significantly increases as a function of a positive or negative mood. Our findings suggest that the native language promotes an inherent cultural bias, which is impervious to fluctuations in the bilingual’s mood state. In the second language, however, bilinguals are culturally impartial, unless they are in a heightened mood state.

We purified a protein from Drosophila S3 tissue culture cells that has many of the diagnostic features of spectrin from vertebrate organisms: (a) The protein consists of two equimolar subunits (Mr = 234 and 226 kD) that can be reversibly cross-linked into a complex composed of equal amounts of the two subunits. (b) Electron microscopy of the native molecule reveals two intertwined, elongated strands with a contour length of 180 nm. (c) Antibodies directed against vertebrate spectrin react with the Drosophila protein and, similarly, antibodies to the Drosophila protein react with vertebrate spectrins. One monoclonal antibody has been found to react with both of the Drosophila subunits and with both subunits of vertebrate brain spectrin. (d) The Drosophila protein exhibits both actin-binding and calcium-dependent calmodulin-binding activities. Based on the above criteria, this protein appears to be a bona fide member of the spectrin family of proteins.

(1) Background: Forestry will have to react to climate change because many tree species suffer. Mitigation can be realized either by planting non-native trees from regions with high climatic stress or by utilizing native tree provenances already adapted to stressful environments. Non-native trees have often generated problems in the past due to uncontrolled invasiveness. The use of native trees pre-adapted to the prospective climatic conditions is far less risky for the respective ecosystems. We offer a tool for selecting ecotypes of native trees as provenances for future forestry. (2) Methods: We propose the selection of tree species native to Middle Europe from a database of vegetation relevés of ± natural forest stands. By calculating the mean ecological indicator values of stands from their vegetation, cover sites can be elected that can provide seeds of provenances well adapted to future climatic conditions. (3) Results: By selecting the 10% partition of the most extreme stands of European tree species, seeds can be sampled and propagated for re-cultivating forests fit for future climate. (4) Conclusions: One can expect ecotypes of tree species that grow well on dry sites, since generations have faced evolutionary selection, for survival under stressful environments. This approach helps to avoid ecological risks of non-native trees.

Three stable hybridoma cell lines (AF8, BC11, CE2) have been produced that secrete antibodies specific for cathepsin B. These have been characterized by ELISA, SDS-PAGE immunostaining, immunoprecipitation and immunofluorescent staining. CE2 immunoprecipitated native cathepsin B with retention of enzymic activity, but failed to cross-react with the alkali-denatured enzyme. BC11 bound only to the denatured form of cathepsin B and AF8 cross-reacted with both native and denatured cathepsin B. However, unlike CE2-immunoprecipitated enzyme, activity could be detected only after dissociation of the antigen-AF8 antibody complex. No cross reaction was found with any lysosomal protein includihg the cysteine proteinases, catbepsins H and L.

Murine monoclonal antibodies specific for titin have been elicited using a chicken heart muscle residue as antigen. The three antibodies T1, T3, and T4 recognize both bands of the titin doublet in immunoblot analysis on polypeptides from chicken breast muscle. In contrast, on chicken cardiac myofibrils two of the antibodies (T1, T4) react only with the upper band of the doublet indicating immunological differences between heart and skeletal muscle titin. This difference is even more pronounced for rat and mouse. Although all three antibodies react with skeletal muscle titin, T1 and T4 did not detect heart titin, whereas T3 reacts with this titin both in immunofluorescence microscopy and in immunoblots. Immunofluorescence microscopy of myofibrils and frozen tissues from a variety of vertebrates extends these results and shows that the three antibodies recognize different epitopes. All three titin antibodies decorate at the A-I junction of the myofibrils freshly prepared from chicken skeletal muscle and immunoelectron microscopy using native myosin filaments demonstrates that titin is present at the ends of the thick filaments. In chicken heart, however, antibodies T1 and T4 stain within the I-band rather than at the A-I junction. The three antibodies did not react with any of the nonmuscle tissues or permanent cell lines tested and do not decorate smooth muscle. In primary cultures of embryonic chicken skeletal muscle cells titin first appears as longitudinal striations in mononucleated myoblasts and later at the myofibrillar A-I junction of the myotubes.

We studied the distribution of fibronectin (FN) in rat uterus, rat tail tendon, and rooster comb, using LM and EM immunocytochemistry. Special attention was paid to the interaction of FN with collagen (COL). Various labeling protocols and dot-blot experiments were performed to confirm the results. Under conditions of labeling specificity, FN distribution over native COL fibrils was usually sparse, especially when these were organized into thick bundles. No labeling was observed over section surfaces of COL fibrils when postembedding methods were used, which indicates that no FN is present within these fibrils. Under conditions in which exogenous FN could react with tissues, e.g., when preincubation with normal serum for background blocking was performed, artifactual staining appeared over COL. Such a reaction also occurred when anti-FN antiserum completely blocked by liquid-phase adsorption was used. Therefore, the FN present in soluble FN-anti-FN immune complexes must have still been able to react with COL. The artifactual labeling was, in all cases, almost exclusively localized on the section surfaces of COL fibrils. These results suggest that FN has a very low affinity for the surface of native COL fibrils.

In-situ and ex-situ spectroscopic ellipsometry (SE), atomic force microscopy (AFM), transmission electron microscopy (TEM), and time of flight medium energy backscattering (ToF MEBS), are used to investigate the properties of 30 and 60 Å ZrO2 films deposited at different temperatures on hydrogen terminated silicon (H-Si) and native silicon oxide surfaces. Results show that the initial-stage deposition of ZrO2 on H-Si and native silicon oxide surfaces are different. A 3-dimesional (3D) type nucleation process of ZrO2 on H-Si leads to high surface roughness films, while layer-by-layer deposition on native silicon oxide surfaces leads to smooth, uniform ZrO2 films. An interfacial layer, between the substrate and the metal oxide, is formed through two independent mechanisms: reaction between the starting surfaces and ZTB or its decomposition intermediates, and diffusion of reactive oxidants through the forming ZrO2 interfacial stack layer to react with the substrate.

1. Bovine inositol monophosphatase reacts with thiol reagents such as 5,5′-dithiobis-(2-nitrobenzoic acid) (DTNB), N-ethylmaleimide (NEM) and iodoacetic acid (IAA). 2. Modification by NEM results in nearly total loss of enzyme activity, whereas modification by IAA causes a slight increase in activity. 3. The loss of activity caused by NEM can be prevented by the inclusion of Ins1P, or better Ins1P and LiCl in the reaction mixture. 4. Two equivalents of p-nitrothiobenzoate (NTB2-) are released from the native enzyme on reaction with DTNB, and six equivalents of NTB2- are released from the SDS-denatured enzyme, suggesting that none of the six cysteine residues per molecule of enzyme is involved in intra- or inter-molecular disulphide bridges. 5. Both NEM and IAA react with two cysteine residues (residues 141 and 184 in the sequence) in a mutually exclusive manner. 6. NEM also reacts stoichiometrically with residue 218. 7. The NEM-induced loss of enzyme activity is accompanied by a 15% decrease in protein fluorescence. 8. A mutant of the enzyme which has an Ala-218 replacement for Cys-218 has full activity and is not sensitive to NEM, showing that the modification of this cysteine by NEM causes inhibition of the native protein by steric effects and that Cys-218 is not essential for activity.

An arabinogalactan-protein (AGP) purified from the filtrate of liquid-suspension-cultured Italian-ryegrass (Lolium multiflorum) endosperm cells by affinity chromatography on myeloma protein J539-Sepharose was deglycosylated with trifluoromethanesulphonic acid to remove polysaccharide chains that are covalently associated with hydroxyproline residues in the peptide component of the proteoglycan. The protein core, which accounts for less than 10% (w/w) of the intact proteoglycan, was purified by h.p.l.c. It has an apparent Mr of 35,000, but reacts very poorly with both Coomassie Brilliant Blue R and silver stains. Amino-acid-sequence analysis of the N-terminus of the h.p.l.c.-purified protein core and of tryptic peptides generated from the unpurified protein reveals a high content of hydroxyproline and alanine. These are sometimes arranged in short (Ala-Hyp) repeat sequences of up to six residues. Polyclonal antibodies raised against the protein core do not cross-react with native AGP, the synthetic peptide (Ala-Hyp)4, poly-L-hydroxyproline or poly-L-proline. The results suggest that the polysaccharide chains in the native AGP render the protein core of the proteoglycan inaccessible to the antibodies and that the immunodominant epitopes include domains of the protein other than those rich in Ala-Hyp repeating units.

The present short report reveals how teachers of English in Hong Kong (HKTEs) react to Hong Kong English (HKE). By employing a mixed method approach consisting of 100 survey responses and 28 interviews, types of feedback and activities teachers use when they encounter HKE in classroom were recorded and reported. The results showed that the two types of teachers of English – Native and Non-native English speakers – provided different kinds of responses because of the differences in attitude they held toward new varieties of English. The current study potentially sheds light on how different varieties of English could fit in traditional ESL curricula. Further research is warranted on how the feedback may affect English acquisition among Hong Kong students and whether the feedback brings positive or negative effects to the students.

A monoclonal antibody (CC-212), obtained in a fusion experiment in which basal bodies from quail oviduct were used as immunogen, has been shown to label the apical pole of ciliated cells and to react with a 200-kD protein. This monoclonal antibody was demonstrated to be an anti-myosin from smooth muscle or from nonmuscular cells using the following criteria: On Western blots it reacted with the myosin heavy chains from gizzard and platelet extracts and from cultured cell line extracts, but did not react with striated muscle myosin heavy chains. By immunofluorescence it decorated the stress fibers of well-spread cells with a characteristic striated pattern, while it did not react with myotubes containing organized myofibrils. On native ciliated cells as well as on Triton-extracted ciliated cortices from quail oviduct, this monoclonal antibody decorated the apical pole with a stronger labeling of the periphery of the apical area. Ultrastructural localization was attempted using the immunogold technique on the same preparation. Myosin was associated with a filamentous material present between striated rootlets and the proximal extremities of the basal bodies. No labeling of the basal body itself or of axoneme was observed.

The effect of organotin compounds (OTs) on the accumulation of the lipid peroxidation (LPO) carbonyl by-products, which react with thiobarbituric acid (TBARS) in fresh and cryopreserved sperm of Russian sturgeon, was studied. It was found that incubation (1 hour) of Russian sturgeon sperm with OTs (CH3SnCl3, (CH3)2SnCl2, (CH3)3SnCl, (n-C4H9)2SnCl2, (n-C4H9)3SnCl, (C6H5)2SnCl2, (C6H5)3SnCl) in concentration 0.1 mM led to the promotion of the accumulation of TBARS in native semen. Dimethyl- (DMT) and diphenyltin dichlorides (DPT) exhibited the greatest promoting activity, and the LPO level of both native and cryopreserved sperm of Russian sturgeon, including those in modified Stein's cryomedium, increased in the presence of these compounds. It was found that Russian sturgeon's cryopreserved sperm had lower sensitivity to the promotion of sperm LPO by DMT and DPT compared with the native sperm. The protective effect of Stein’s cryomedium decreased in the presence of the studied OTs. The results suggest that accumulation of OTs by gonad of fish is another stress factor affecting the cell productivity in the cryopreservation process.

Postcolonial discourses describe colonization as a process of invention to impose the will of a conquering West on “backward” societies. The will to power conjugated with the need for raw materials served as the main catalysts. They put side by side a hegemonic intruder bent on duplicating itself, and a powerless and compliant native unable to react to the blitz of transformations. Hence, the master/slave or father/child relationships that describe the colonial framework. The task is to interrogate these generally accepted assumptions and binary oppositions. Although marginalized, the Congolese native was unwilling to become an object for the colonizer’s gaze. In fact, the inability to expel the “invader” did not prevent the creation of legitimacies out of what was precipitously brought in. This mechanism of transformation is perceptible in Paul Lomami Tchibamba’s novel Ngando (1948), the object of this study. Ngando’s imagined colonial city stands out as a site of contrasts and contradictions. However, the duplicated model shows the “transformability” of the new space into “normalcy” by a subversive native.

Brucellosis is a global disease and the world’s most prevalent zoonosis. All cases in livestock and most cases in humans are caused by members of the genus Brucella that possess a surface O-polysaccharide (OPS) comprised of a rare monosaccharide 4-deoxy-4-formamido-D-mannopyranose assembled with α1,2 and α1,3 linkages. The OPS of the bacterium is the basis for serodiagnostic tests for brucellosis. Bacteria that also contain the same rare monosaccharide can induce antibodies that cross-react in serological tests. In previous work we established that synthetic oligosaccharides, representing elements of the Brucella A and M polysaccharide structures, were excellent antigens to explore the antibody response in the context of infection, immunisation and cross reaction. These studies suggested the existence of antibodies that are specific to the tip of the Brucella OPS. Sera from naturally and experimentally Brucella abortus-infected cattle as well as from cattle experimentally infected with the cross-reactive bacterium Yersinia enterocolitica O:9 and field sera that cross react in conventional serological assays were studied here with an expanded panel of synthetic antigens. The addition of chemical features to synthetic antigens that block antibody binding to the tip of the OPS dramatically reduced their polyclonal antibody binding capability providing conclusive evidence that the OPS tip (non-reducing end) is a potent epitope. Selected short oligosaccharides, including those that were exclusively α1,2 linked, also demonstrated superior specificity when evaluated with cross reactive sera compared to native smooth lipopolysaccharide (sLPS) antigen and capped native OPS. This surprising discovery suggests that the OPS tip epitope, even though common to both Brucella and Y. enterocolitica O:9, has more specific diagnostic properties than the linear portion of the native antigens. This finding opens the way to the development of improved serological tests for brucellosis.

The facile rearrangement of “S-acyl isopeptides” to native peptide bonds via S,N-acyl shift is central to the success of native chemical ligation, the widely used approach for protein total synthesis. Proximity-driven amide bond formation via acyl transfer reactions in other contexts has proven generally less effective. Here, we show that under neutral aqueous conditions, “O-acyl isopeptides” derived from hydroxy-asparagine [aspartic acid-β-hydroxamic acid; Asp(β-HA)] rearrange to form native peptide bonds via an O,N-acyl shift. This process constitutes a rare example of an O,N-acyl shift that proceeds rapidly across a medium-size ring (t1/2 ∼ 15 min), and takes place in water with minimal interference from hydrolysis. In contrast to serine/threonine or tyrosine, which form O-acyl isopeptides only by the use of highly activated acyl donors and appropriate protecting groups in organic solvent, Asp(β-HA) is sufficiently reactive to form O-acyl isopeptides by treatment with an unprotected peptide-αthioester, at low mM concentration, in water. These findings were applied to an acyl transfer-based chemical ligation strategy, in which an unprotected N-terminal Asp(β-HA)-peptide and peptide-αthioester react under aqueous conditions to give a ligation product ultimately linked by a native peptide bond.

Abstract:
In this work, a series of Bi2Te3/X mol% MoS2 (X = 0, 25, 50, 75) bulk nanocomposites were prepared by hydrothermal reaction followed by reactive spark plasma sintering (SPS). X-ray diffraction analysis (XRD) indicates that the native nanopowders, comprising of Bi2Te3/MoS2 heterostructure, are highly reactive during the electric field-assisted sintering by SPS. The nano-sized MoS2 particles react with the Bi2Te3 plates matrix forming a mixed-anion compound, Bi2Te2S, at the interface between the nanoplates. The transport properties characterizations revealed a significant influence of the nanocomposite structure formation on the native electrical conductivity, Seebeck coefficient, and thermal conductivity of the initial Bi2Te3 matrix. As a result, enhanced ZT values have been obtained in Bi2Te3/25 mol% MoS2 over the temperature range of 300–475 K induced mainly by a significant increase in the electrical conductivity.

Invasive mammalian predators are linked to terrestrial vertebrate extinctions worldwide. Prey naïveté may explain the large impact invasive predators have on native prey; prey may fail to detect and react appropriately to the cues of novel predators, which results in high levels of depredation. In Australia, the feral cat ( Felis catus ) and the red fox ( Vulpes vulpes ) are implicated in more than 30 animal extinctions and the naïveté of native prey is often used to explain this high extinction rate. Reptiles are one group of animals that are heavily preyed upon by F. catus and V. vulpes . However, very few studies have examined whether reptiles are naive to their cues. In this study, we examine the ability of two native reptile species ( Morethia boulengeri and Christinus marmoratus ) to detect and distinguish between the chemical cues of two invasive predators ( V. vulpes and F. catus ) and three native predators (spotted-tailed quoll, Dasyurus maculatus; dingo, Canis lupus dingo ; eastern brown snake, Pseudonaja textilis ), as well as two non-predator controls (eastern grey kangaroo, Macropus giganteus and water). We conducted experiments to quantify the effects of predator scents on lizard foraging (the amount of food eaten) during 1 h trials within Y-maze arenas. We found both study species reduced the amount they consumed when exposed to predator scents—both native and invasive—indicating that these species are not naive to invasive predators. An evolved generalized predator-recognition system, rapid evolution or learned behaviour could each explain the lack of naïveté in some native Australian reptiles towards invasive predators.

Abstract A group of 19 anti-class II monoclonal antibodies produced in different laboratories were tested in ELISA for their ability to bind to a panel of synthetic peptides selected from HLA-DQ alpha and beta chains. No one of the antibodies tested was found to react with the synthetic fragments, thus confirming the common finding that MoAbs generally fail to recognize fragments of the native antigen. The possibility that this result might be partly due to the procedure used for screening hybridoma supernatants is discussed.

Polyclonal antibodies to native RNA polymerase II of Achlya ambisexualis and Agaricus bisporus were produced in rabbits and in mice. Monoclonal antibodies were produced against the α-amanitin resistant RNA polymerase II of the mushroom A. bisporus. These antibodies were used in comparative cross-reactivity studies with five purified RNA polymerases II (A. bisporus, A. ambisexualis, Saccharomyces cerevisiae, wheat germ, and calf thymus). A method for quantitatively comparing cross-reactivity was developed utilizing an enzyme-linked immunosorbant assay (ELISA). ELIS A comparisons indicated that the two filamentous fungi cross-reacted effectively with one another and depending upon the preparation reacted less effectively with yeast and wheat germ RNA polymerases II. Cross-reactivity measurements were also made by immunoblotting sodium dodecyl sulfate – polyacrylamide separated RNA polymerases II. The mouse anti-A. bisporus RNA polymerase II immunoglobulin G (IgG) and the monoclonal antibody preparations did not react with high molecular subunits of A. bisporus RNA polymerase II. The sera did, however, cross-react with high molecular weight subunits of A. ambisexualis. Similarily, rabbit anti-A. ambisexualis RNA polymerase II IgG reacted only with low molecular weight subunits of A. bisporus RNA polymerase II, but reacted with high molecular weight subunits of A. ambisexualis and wheat germ. Our results indicate differences in the cross-reactivity of native and denatured RNA polymerases II and suggest differences in the tertiary and quaternary organization of the enzymes examined.

ABSTRACT Phage Q38, a representative member of the c2 species, was purified by CsCl gradient and used to immunize BALB/c mice. Monoclonal antibodies (MAbs) were raised and then characterized by enzyme-linked immunosorbent assay. Two MAbs of isotype immunoglobulin G2a, designated 2A5 and 6G7, reacted only with phages belonging to the c2 species and not with phages of the 936 and P335 species, with a Lactococcus lactis cell extract, or with phage DNA. Immunoelectron microscopy showed that both MAbs recognized only phage head proteins. They did not react with any denatured phage proteins in Western blot assays. However, when the nitrocellulose membranes were treated with a Triton-based buffer to assist in protein renaturation, MAbs 2A5 and 6G7 recognized the two major capsid proteins with molecular masses of 80 and 170 kDa. Competitive inhibition tests showed that the two MAbs bind to overlapping epitopes. These MAbs may be a useful tool for monitoring c2 bacteriophages during dairy fermentation and in genetic studies.
