Abstract:
Chlorinated aromatic compounds are an important group of compounds. Many of them have been produced in large quantities and they are indispensable to technological and societal benefits. But regulatory agencies have tightened regulations on the use and release of chlorinated aromatic compounds because of the scientific understanding of their toxicity, persistence, behavior in the environment and their potential to cause adverse effects on the ecosystem and human health.
Pentachlorophenol (PCP), octachloronaphthalene and decachlorobiphenyl are all highly chlorinated aromatic compounds, of which, PCP has been used mainly as a biocide. Octachloronaphthalene and decachlorobiphenyl don't have practical use, but their congeners have been used widely as chemicals in industry. These compounds are toxic, recalcitrant and bio-accumulated within organisms. As the conventional treatment, incineration of these compounds can cause more serious problems, so that suitable alternatives need to be developed for their detoxification.
When compared with biodegradation or the thermal treatment of these compounds, chemical degradations have several merits. (Abstract shortened by UMI.)

Abstract:
Recalcitrant organic compounds remain a key challenge in landfill leachate management as they are resistant to microbial degradation and have potential to damage the water environment. Conventional leachate characterisation methods are time consuming and limited by their inability to provide compositional analysis. This research therefore investigates the characteristics of recalcitrant organic compounds in leachates and undertakes a feasibility study of the possible use of UV absorption and fluorescence spectroscopy for a rapid and economical compositional analysis of recalcitrant organic compounds. Two laboratory experiments are carried out in this regard. In one experiment, a laboratory scale aerobic biodegradation is carried out on four untreated and two treated leachate samples collected from two UK MSW landfills (Pitsea and Rainham), and leachates are characterised using conventional methods (COD and DOC) as well as UV absorption and fluorescence spectroscopy. It is found that the leachates which have low organic content are easily biodegradable whereas the leachates which have high organic content are not easily biodegradable. UV and fluorescence spectroscopy allow compositional analysis of recalcitrant organic compounds and show that humic and fulvic compounds are the key components of the recalcitrant organic compounds. A rapid biodegradability assessment of different leachates using these novel techniques is in agreement with conventional method showing that a rapid characterisation of leachates using spectroscopic methods is feasible. It is also found that organic compounds in these leachates are aromatic in nature and the leachates containing large amounts of aromatic compounds, condensed aromatic structures are difficult to degrade. In another experiment, the influence of the solid waste component on the development of recalcitrant organic compounds in leachates is investigated by carrying out an anaerobic biodegradation experiment for fresh waste, composted waste, newspaper waste and synthetic waste. Composted waste contributes significantly to the development of the recalcitrant compounds due to the removal of readily biodegradable organic compounds during composting and hence the increased proportion of biologically resistant compounds during the subsequent anaerobic biodegradation. Newspaper waste also contributes significantly to the presence of recalcitrant organic compounds due to the relatively less resistant cellulose and high lignin present in this waste. This research validates the application of UV and fluorescence spectroscopy for rapid on-site monitoring of landfill leachates that would help scientist and engineers to assess leachate quality, identify various organic compounds and to optimise leachate treatment processes. The analysis performed on Pitsea and Rainham leachates is a promising step towards developing a database of representative information of characteristics of recalcitrant organic compounds in leachates for different UK landfills. This research also provides an understanding of the composition of leachates from different wastes and it can be conclude that disposals enriched with composted and newspaper waste would favour the development of leachates with high concentrations and condensed aromatic structures of recalcitrant organic compounds in a landfill system

Abstract:
This study reviews catalytic ozonation AOPs and traditional ozone-based AOPs to compare their efficiency for degradation of ozone-recalcitrant compounds. With the world’s population increasing and water becoming a scarce resource, it is important to improve current water recycling methods. Recycling water will play a large role in accommodating the increasing demand, but it will also be necessary to be improve the level of treatment in order to account for emerging contaminants (ECs) such as pharmaceuticals and personal care products. Advanced oxidation processes (AOPs) have been developed to degrade ECs that are not effectively removed by conventional wastewater treatment. The goal of implementing AOPs is to promote the formation of hydroxyl radicals (•OH), which are stronger oxidants than ozone, to degrade recalcitrant compounds. Current AOPs under investigation include ozonation of metal and carbon-based catalysts, known as catalytic ozonation. Traditional ozone-based AOPs currently in use include UV combined with ozone (UV/O3) and hydrogen peroxide combined with ozone (H2O2/O3). Seventeen studies were reviewed to analyze the effectiveness of multiple carbon- and metal oxide-catalytic ozonation AOPs, compared to traditional AOPs. These studies varied in reactor type, water source, pH, catalyst pretreatment, inclusion of competitor species, and flow regime. The variety of testing conditions made comparison difficult, so all studies were compared based on contaminant removal efficiency and degradation rate, as well as general EC degradation and removal of TOC. The addition of metal oxides during ozonation consistently increased rate of removal and in some cases, even doubled the reaction rate. Catalytic ozonation consistently decreased total organic carbon (TOC) levels amongst multiple studies, even in the presence of competitor species. Future work should study the formation and subsequent breakdown of reaction intermediates, role of competitor species, and impact of sorption to the ozonation catalyst.

Abstract:
Tannins are polyphenolic compounds produced by plants that are used in the vegetable tanning of leather at an industrial scale. Tannins differ from most other natural phenols since they precipitate proteins and are used in the tanning process to bind to the collagen proteins of the animal skin to make leather more durable and not putrescible. Tannins represent one of the low-biodegradability substances in tannery wastewaters with a highly recalcitrant soluble chemical oxygen demand; moreover, at high concentrations they can inhibit biological treatment. This soluble recalcitrant fraction of tannery effluents is usually removed by means of chemical processes. Therefore, a biological treatment that could remove effectively this fraction would have both environmental and economic advantages. Despite the antimicrobial properties of tannins, there are organisms that are able to grow on them; in fact their biodegradation in the environment is mainly associated with fungi rather than bacteria. Fungi could thus be exploited for the bioremediation of wastewater streams of the tanning industry. However, in environmental biotechnology applications, fungi tend to be outcompeted by bacteria. The application of a fungal-based bioreactor, that has a similar performance under sterile and non-sterile conditions in long-term operations, is still a challenging task. The present thesis was aimed at developing and testing technologies to remove tannins with an engineered ecosystem based on fungi and bacteria for application in typical tannery wastewater treatment trains (non-sterile conditions). Aspergillus tubingensis MUT 990 was selected as a suitable fungal strain. Literature research and previous experiences were used to design and build lab-scale (4 litres volume) and pilot-scale (1.5 m3 volume) reactors. The selected fungal strain was immobilised in polyurethane foam cubes carriers and inoculated in a novel rotating, submerged, packed bed reactor. The first test in bioreactors was carried out to evaluate the effect of rotation and the co-substrate addition on tannin removal. The second test in bioreactors was carried out in two submerged packed bed reactors run in parallel, the first one was fed with a condensed tannin (Quebracho tannin), and the second with hydrolysable tannin (Tara tannin), both were inoculated with Aspergillus tubingensis in attached form. The hydraulic retention time and the concentration of tannins in the medium were varied to maximise the removal capacity and to analyse the substrate inhibition. A stable biofilm was maintained in the first reactor during the 180 days of operation. On the other hand, in the second reactor there was a biomass detachment during the start-up and then grown as a suspended culture throughout the operational period (226 days). Soluble chemical oxygen demand removal up to 53% and 90% were achieved in the first and second bioreactor, respectively, without the addition of co-substrates. The microbial communities of the reactors, made up of fungi and bacteria, were characterised with a metagenomic analysis. In addition, an innovative technique, the heterogeneous respirometry, was applied to assess the biological activity of immobilised cells (biofilm). The data obtained were used to develop a mathematical model and to perform a kinetic and stoichiometric characterisation of the biomass. Since fungal biomass is poorly characterised with modelling and respirometry, dedicated experiments were set-up along with a respirometric procedure. The lab-scale results were used to design a pilot-scale reactor. Finally, after a start-up phase the pilot-scale reactor was fed with real tannin-rich wastewater collected from tanneries. At the end of the experimentation the biofilm was stable and an encouraging performance had been achieved. We believe the results obtained represent the first step for a future real-scale application to reach an efficient biological removal of tannins from tannery wastewater.

Abstract:
This thesis presents the design and analysis of photo-electro catalytic oxidation technology for its potential use in water treatment applications. Doping titanium dioxide can effectively improve oxidative reactions. Using minute reactor design adjustments, such as electrolysis, the bases of degrading soluble organic compounds can be improved within the reactor. Applications of doping and secondary catalysis are effective in increasing the process of photoactivity within the catalytic reactor, theoretically increasing the production of sacrificial electron acceptors. Higher degradation's were accomplished using doped chromium titanium dioxide photocatalyst, which degraded significantly larger amounts of organics compared with pure titanium dioxide. Techniques used to accomplish the increased photo-reactivity were doping chromium (III) ion into titanium dioxide crystal lattices and silver ions into the photocatalytic coat. Degradation curves were determined by total organic carbon and the chemical oxygen demand. Calculations of the final rate constants show that lighter molar concentrations have higher rates of degradation.

Abstract:
This thesis reports on the degradation of two different models of organic pollutants in two contrasting Scottish river sediments. The first type of sediment (sediment 1) was collected close to an urban area with high percentage of organic matter content (1.3%). The second type of sediment (sediment 2) was collected from a pristine area and was less organic (0.2%). As organic pollutant model, radiolabelled (14C) 2,4-Dichlorophenoxyacetic acid (Acid-ring-UL-14C) and Naphthalene (Naphthalene-1-14C) were amended to individual microcosms (50g of sediment and 50ml of river water) at different concentrations (0.5, 5.0 and 50 mg per microcosm). Degradation was assessed by emission of 14CO2 during a period of 40 and 124 days for 2,4-D and Naphthalene, respectively. 2,4-D was mostly degraded between 20-35 days, and degradation rates (5 to 750 mgd-1) were proportional to initial concentrations. Almost 25% of applied Naphthalene was degraded by 40 days, at different degradation rates (2.7 to 405 mgd-1) according to sediment type. Assessment of the impact of 2,4-D and Naphthalene on the sediment microbial community was made using Biolog, DGGE and PLFA analysis to provide for a comparative physiological, molecular and chemotaxonomic assessment, respectively. These analyses linked differences in the rates and extents of 2,4-D degradation to differences in composition of the microbial communities. To assess the impact of 2,4-D, DGGE-profiling was used and show genetic changes were more significant in sediment 2 microcosms than sediment 1. Biolog suggested differences in the microbial structure of both sediments, but PLFA demonstrates that sediment 1 treatments were more affected by 2,4-D amendments than sediment 2. The extent of Naphthalene biodegradation was restricted by bioavailability, sediment type and volatilisation.

Leachates are wastewaters generated at sites of solid waste disposal through organic matter decomposition, added to infiltration waters and suspended materials. It contains biodegradable and recalcitrant compounds - difficult to biological degradation (biodegradation). This wastewater requires treatment before being disposed in the environment. The aim of this study was to evaluate the use of ozone (O3) and hydrogen peroxide (H2O2) as advanced oxidation process (AOP), in order to removal recalcitrant compounds in landfill leachate. It was used two leachates from São Leopoldo/RS landfill: raw and treated in pounds, which were treated only with O3 at a concentration of 29 mg/L (generated from the corona process) or with addition of 1000 mg/L H2O2 to O3. The tests occurred on a pilot scale, treating 460 L of leachate in each test, which lasted 96 hours. OAP (operating with the oxidants O3 and O3 + H2O2) was more efficient in the treatment of leachate that had the lowest initial concentrations of BOD, COD and TOC (both raw and treated leachate).. High removals of apparent color, COD and TOC were observed, reaching 99%, 88% and 66%, respectively. The greater removal of inorganic carbon was 23%, but the group that started with the highest concentration of these parameter, achieve the greatest removals for all others parameters studied. The addition of H2O2 increased the efficiency of removal of all parameters, but not significantly. It was observed need of additional treatment to the POA of this study.

Abstract:
This work assessed the feasibility of using biological and advanced oxidation processes (AOP) as post-treatments for old leachate from stabilization ponds in series. Additionally, an analytical methodology for detection and quantification of aromatic compounds, organochlorides, phthalate esters and hormones, was developed. These compounds are considered as recalcitrant and rarely investigated in treated leachates, mainly in wastewater treatment plants located in the state of CearÃ. Initially, a physico-chemical characterization and a metal speciation was carried out, from which it was verified that COD and ammonia were above the limits permitted by environmental legislation, and also some metals. In the development of the analytical methodology for detection and quantification by GC/FID, the solid phase extraction using C-18 was the best technique for extraction and pre-concentration of the recalcitrant compounds and the best results in terms of recuperation were achieved for the aromatic compounds and phthalate esters. In the post-treatment experiments using biological reactors, aerated submerged reactor (ASR) and sequential batch reactor (SBR), both were efficient in the leachate nitrification reaching ammonia removal efficiencies above 90%. However, they were not efficient on phosphorus removal. In terms of recalcitrant compounds removal, SBR presented better results when compared with ASR for the aromatic compounds and phthalate esters. The advanced oxidation process UV/H2O2 reached turbidity and COD removal and decolourisation efficiencies above 90% from a reaction time of 120 minutes by using recirculation batch mode. The UV/H2O2 system was efficient in the removal of aromatic compounds and the phthalate esters benzyl butyl phthalate (BBP) and diethyl hexyl phthalate (DEHP), achieving 100% removal. The photo-Fenton process with solar radiation assessed with the leachate from Portugal showed to be an excellent option for the leachate post-treatment when compared to the other systems studied (UV/H2O2 and TiO2/UV). Mineralization of organic compounds has reached approximately 80% reduction of dissolved organic carbon (DOC), beside aromaticity and decolourisation reductions and increasing leachate biodegradability. The optimal iron concentration for the leachate studied was 60 mg L-1 Fe+2, with H2O2 consumption of 310.6 mM and approximately 110 kJ L-1 of accumulated energy. However, it was not efficient in the removal the ester DEHP, one of most important phthalates, which is considered an endocrine disruptor. The UV/H2O2 and TiO2/UV processes using solar energy were slower than the photo-Fenton process and low COD removals were achieved, which indicated low mineralization of the organic compounds present in the leachate.

Phenol and phenolic compounds have been listed as priority-pollutants by the Environmental Protection Agency (EPA USA). They are often found in industrial effluents such as from pulp and paper, timber, plastics and synthetic polymer, pharmaceutical, pesticide, oil and petrochemical industries. Most of the traditional wastewater treatments have shown inefficiency against high phenol concentrations. This work aims to treat phenol solution via Advanced Oxidation Processes (AOP´s) to enhance medium biodegradability and using it as an effluent in a continuous activated sludge process. The purpose of the pre treatment is to modify the phenol structure leading it to its byproducts such as aromatics compounds and carboxylic acids. The experiments have shown the effectiveness of the pre-treatment by the biodegradability parameter BOD5/COD which was achieved 0,19 at 30 min of photo- Fenton reaction with a initial phenol concentration of 1000 mg.L-1 of TOC, 1 mmol.L-1 of FeSO4 and 100 mmol.L-1 of H2O2, added with a peristaltic pump along 2 hours of reaction. After the photochemical pre treatment, the same solution was used as a carbon source in a continuous activated sludge system for removing residual organic matter, with a reduction of 92% of the TOC initial. Those results have shown the efficiency when integrating photo-Fenton and activated sludge processes on the treatment of effluents containing recalcitrant organic compounds.

Abstract:
With the environmental problems caused by man-induced pollution by persistent toxic compounds, the importance of finding remediation solutions is immense. As an emerging field, microbial environmental biotechnology may provide the tools to achieve novel solutions. Microbial communities in the environment have biodegradation capacities which could be, and historically have been, exploited for bioremediation. The novelty lies in being able to access the capacity of the uncultured majority of the microbial community. Every day, more and more knowledge is gained in the field and thanks to new approaches such as metagenomics, along with the access to databases and archives where scientists share information and data, the quest becomes considerably facilitated. Microorganisms are highly diverse in metabolic pathways and some have become highly developed during evolution; detoxification and biotransformation of naturally occurring toxic compounds are therefore not novel concepts. The environmental problem occurs when synthetically manufactured compounds are less efficiently biodegraded. However, improved knowledge about the degradation potential in nature and the involved enzymes may help in developing bioremediation procedures. For this reason, an enzyme involved in catabolic pathways of chlorinated aromatic compounds, dienelactone hydrolase, which has been less well studied, was selected as a target. This study investigated the biogeographical distribution of the dienelactone hydrolase gene identified in metagenomes sampled from different environments globally in order to detect potential environmental patterns. Results may cast light on its significance for degradation of chlorinated aromatic compounds in nature. The results indicate a broad biogeographical distribution of dienelactone hydrolase in varying microbial habitats in the environment. The enzyme was found in environments ranging from water and soil habitats to hypersaline-, dechlorinating-, hot-spring- and other extremophillic habitats, in which the gene sequences shared high similarity within each group. A broad environmental distribution suggests that dienlactone hydrolase could be useful in bioremediation.

Abstract:
Creosote, containing many high molecular weight hydrophobic polycyclic aromatic hydrocarbons (PAH's), is present in the subsurface environment at the Oneida Tie-Yard in Oneida, Tennessee. Phytoremediation using hybrid poplar trees was chosen as the remedial technology on-site. Since monitoring began, the contaminant plume has been shrinking consistently and evidence has shown that remediation is taking place. However, remediation may be rate-limited by the desorption and dissolution kinetics of the PAH's on-site. The objectives of this research are to: (1) estimate the desorption and dissolution rates of 10 PAH's found in the subsurface and (2) estimate the amount of each PAH and total mass of contaminant that is irreversibly sorbed to the soil. Three laboratory desorption and dissolution experiments were performed using contaminated soil samples from the Oneida Tie-Yard site. The first experiment was a batch desorption equilibrium experiment, the second was a batch desorption kinetics experiment, and the third was a soil column dissolution kinetics experiment. The target compounds in this study were: naphthalene, acenaphthylene, acenaphthene, fluorene, phenanthrene, anthracene, fluoranthene, pyrene, chrysene, and benzo(b)fluoranthene. The resulting data for the desorption equilibrium experiment revealed that rates of equilibrium were truly not instantaneous in the systems studied. However, because approximately 76% of PAH's desorbed by the first sampling event (3 days), an equilibrium isotherm was considered appropriate. Results showed that there is a sorbed reversible concentration that readily desorbs to the aqueous phase for each PAH. Additionally, it was determined that the percent removal of sorbed PAH's decreases with increasing molecular weight. Desorption curves based on experimental data were found to exhibit linear behavior over large variations in aqueous concentration, but showed exponential behavior as concentrations approached zero. Freundlich sorption equilibrium isotherms for the 10 monitored PAH's on-site were generally found to have N coefficient values over 1, especially over large variations in solution phase concentration, indicating a non-uniform sorbent. Dissolution of resistant PAH's under field-like conditions was determined to occur over long periods of time. Dissolution rates calculated from experimental data were shown to generally decrease with increasing molecular weight. Overall, desorption and dissolution kinetics of PAH's were shown to be rate-limiting factors to remediation at the Oneida Tie-Yard.

Abstract:
Effluents with high concentration of organic matter and xenobiotic compounds, especially in the case of the agrochemicals, cause environmental concern. In the present work, the adaptative ability of two filamentous fungi, which cause white rot (Geotrichum candidum Link & Pers. and Phanerochaete chrysosporium Burds.), to tolerate high concentration of xenobiotic substances was analyzed. In solid medium, P. chrysosporium and G. candidum presented excellent development under aerated and not aerated conditions (30ºC and dark), in potato-dextrose-agar and malt extract-agar, respectively. In liquid medium, the same was observed in broth-potato-dextrose and malt extract. The fungi also developed well in liquid media deficient in proteins and carbohydrates, but with high concentrations (10 and 25 mg/L) of the agrochemicals Boral, Decanote, Gliphosate, Paraquat and Plenum - all used in fields of sugar-cane, as the ones of the "S.A Usina Coruripe Açúcar e Álcool" (Coruripe - AL). These chemicals are classified as persistent, belong to the toxicological classes II and III, and have different solubilities. The fungi presented tanninolitic activity when inoculated in samples of the crude effluent with supplement of nutrients, under aeration or not. The activity of the laccase of G. candidum in the same medium was higher in the 3rd day of the growth without aeration, occurring a reduction of that in the 5th day. On the other hand, the fungus P. chrysosporium presented a higher activity of laccase in the 7th day of the growth, also without aeration. Both fungi did not interfere in the color, turbidity, concentration of nitrates and nitrites of this medium. G.candidum revealed equal efficiency in the reduction of the concentration of phosphates of the crude effluent, under aeration (78.18%) and not aeration (79.33%) conditions. However, it did not reduce the COD. P. chrysosporium was efficient in the reduction of this COD, and the difference between the aerated growth (26.77%) and the not aerated one (29.55%) was very small. It did not reduce the total tanning bark concentration, but it diminished the concentration of phosphates under aerated (55.55%) and under not aerated (61.65%) condition. The addition of nutrients in the medium of diluted effluent (2 X, 4 X and 8 X) showed that there is a direct effect of the concentration of the COD on its percentage of removal by the inoculated fungi. There is also a direct effect on the remotion of total phenolic compounds and reduced glycids. The conversion of COD in the diluted effluent (8X) was higher in the growth of P. chrysosporium (65.66%) than in the one of G.candidum (57%). The same it can be said regarding to the best rates of phenol degradation (59.66% and 50.33%, respectively). The pH measured in the media supplemented with nutrients was always higher (7,56-7,8) than the initial one (5,5). In the higher dilution of the effluent (8X) plus the supplement of nutrients, the maximum removal of COD and total phenols after the treatment occurred in 7th day of incubation of both fungi. In all the cases of diluted samples plus supplement of nutrients, the discoloration drastically increased after the inoculation of the fungi. Therefore, the use of these microorganisms in the bioremediation of the effluent of sugar-alcohol industry is recommended only after a correction of the ratio C: N: P to about 100:1,6:1.
