Toxigenic Clostridium sordellii causes uncommon but highly lethal infections in humans and animals. Recently, an increased incidence of C. sordellii infections has been reported in women undergoing obstetric interventions. Pathogenic strains of C. sordellii produce numerous virulence factors, including sordellilysin, phospholipase, neuraminidase, and two large clostridial glucosylating toxins, TcsL and TcsH. Recent studies have demonstrated that TcsL toxin is an essential virulence factor for the pathogenicity of C. sordellii. In this study, we identified and characterized TcsR as the toxin gene (tcsL) regulator in C. sordellii. High-throughput sequencing of two C. sordellii strains revealed that tcsR lies within a genomic region that encodes TcsL, TcsH, and TcsE, a putative holin. By using ClosTron technology, we inactivated the tcsR gene in strain ATCC 9714. Toxin production and tcsL transcription were decreased in the tcsR mutant strain. However, the complemented tcsR mutant produced large amounts of toxins, similar to the parental strain. Expression of the Clostridium difficile toxin gene regulator tcdR also restored toxin production to the C. sordellii tcsR mutant, showing that these sigma factors are functionally interchangeable.

The intensifying nature and extent of disasters together with the associated devastation and astronomical costs required to manage the rippling effects of disasters, enunciates the national and international focus on disaster risk reduction. Further the ever evolving and complex dynamics of risk as the decisive contributor to disasters has heightened the urgency to pursue effective disaster risk assessment as a prerequisite to inform the disaster risk management planning and disaster risk reduction intervention processes. A structured and systematic approach to disaster risk assessment assists in maintaining rigour thus promoting the quality and validity of the process and its outcomes. Appropriate models serve as valuable tools in enabling this methodological perspective to undertake disaster risk assessment. The nonexistence of an appropriate disaster risk assessment model in South Africa has initiated the emphasis and purpose of this study thereby underscoring the critical need for the development of an effective, holistic and integrated disaster risk assessment model for local government in South Africa. In spear heading the process towards the development of an appropriate disaster risk assessment model, the research commenced with establishing and asserting the fundamental link between disaster risk assessment and disaster risk reduction as an avenue to contextualise and ground the key issues in effective disaster risk reduction. The exploratory analysis engaged in presenting a theoretical construct of disaster risk assessment examined the core components informing the disaster risk assessment process. This discussion led to the comparative review of three disaster risk assessment models viz the Community-Wide Vulnerability and Capacity Assessment (CVCA) Model, the Community-Based Risk Reduction Model and the South African Disaster Risk Assessment Model interrogating the significant characteristics, structure and application of the models. The results of the comparison of the above three models provided the necessary insight for the development of the disaster risk assessment model for local government in South Africa. Further influenced by the outcomes of the applied research on the critical analysis of the current disaster risk assessment practice within the four selected municipalities representing local government in South Africa; viz eThekwini Metropolitan, Ekurhuleni Metropolitan, Bojanala District and Stellenbosch local Municipalities. Through the data coding, classification and interpretive process, constructive and correlated research findings were immanent guiding the final development of the integrated disaster risk assessment model for local government in South Africa.

Abstract

G-protein coupled receptors (GPCRs) are having the high medical importance since almost half of the medicinal drugs are designed as modulators of receptor molecules. Crystal structure or NMR structures of GPCRs are very difficult to determine because all GPCRs are typically bound to the cell membrane and thus their molecular activation mechanism is still unclear. The recent publication of the crystal structure of the 2-adrenoreceptor will provide new insights in the field of GPCR research.

Ghrelin is a peptide growth hormone which binds to the growth hormone secretagogue receptor (GHS-R) and stimulates the release of growth hormone. Based on the known ghrelin receptor binding core sequences wFwLL (upper letter and lower letter representative for L-form and D-form of the amino acids respectively), we prepared two novel peptide analogs with terminal S-(2-aminoethylsulfenyl) cysteine residues. These peptides were tested for their ability to suppress the binding of ghrelin to transfected COS7 cell-line (Kidney fibroblast line from the green African monkey) cells expressing the ghrelin wild-type receptor or certain mutants thereof. As a result we observed a significant reduction of the total number of binding sites accessible for ghrelin, which increased with the time the cells were incubated with our test compounds. This observations support our hypothesis that the peptides we tested form a covalent bond with free thiols located closely to the ligand binding-site of the receptor protein by disulfide thiol exchange which is an interesting target for development of anti-obesity drugs.

In this work, new solutions based on the Boundary Element Method (BEM) are established for the linear analysis of bending and stability problems of Reddy-Bickford beams. All mathematical steps to write the BEM representation are properly presented: transformation of governing differential equations into equivalent integral equations, deduction of fundamental solutions, formation and solution of algebraic representation.In addition, elastic foundations (winkler and pasternak’s types) attached to Reddy-Bickford beams are solved by BEM as well. It is also addressed a convenient strategy for discontinuities in the area such as abrupt change in geometry of the cross section (stepped beam), intermediate axial load, intermediate supports (continuous beam). Numerical examples incorporating various types of discontinuities and boundary conditions in the field are presented to validate the solutions proposed BEM.
Neste trabalho, novas soluções, baseadas no Método dos Elementos de Contorno (MEC), são estabelecidas para a análise linear de problemas de flexão e estabilidade de barras de Bickford-Reddy. Todos os passos matemáticos para estabelecer a representação do MEC são apresentados: transformações das equações diferenciais governantes em equações integrais equivalentes, dedução das soluções fundamentais, obtenção e solução do sistema alébrico. Além disso, fundações elásticas (Winkler e Pasternak) em barras de Bickford-Reddy também são analisados pelo MEC. É também abordada uma conveniente estratégia para de discontinuidades no domínio tais como: mudança abrupta de geometria da seção transversal (viga escalonada), carga axial intermediária, apoios rígidos no domínio (viga contínua). Exemplos numéricos incorporando vários tipos de condições de contorno e discontinuidades no domínio são apresentadas para validar as soluções do MEC propostas.

Abstract:
This study focuses on the analyses of the stylistic elements and challenges of technique and improvisation in three late piano works by the South African composer Surendran Reddy. Reddy’s personal identity as a crossover artist is apparent in his stylistic approach. The fusion of Western art music, jazz, contemporary and indigenous music from various parts of the world ensued in a distinctive compositional style, which he termed clazz. Three of his shorter late piano compositions, namely African Funk for Felix (2005), Ballad for Thomas (2005) and Toccata for John Roos (2007), contain key characteristics of Reddy’s clazz style. It also represents the composer’s own pianistic virtuosity that renders them challenging to performers. The technical, stylistic and improvisational difficulties in these three works are investigated through a practice-led approach. The methodologies used to study and perform these works were researched by means of two interviews conducted with two professional South African pianists who have performed these works. The technical difficulties in these compositions are delineated and potential solutions to overcome virtuoso passages through fingering suggestions or recommendations for alternative practising methods are examined. The diverse stylistic influences on these works, such as gospel, rock, funk, soul, pop, jazz and various elements of Western art music, are identified, discussed and explored, with emphasis on the influence of South African jazz on Reddy’s conceptualisation of clazz. Prescribed improvisation sections are examined in-depth, and potential performance strategies are devised in the form of notated exercises that would hopefully serve as an accessible methodological approach.

This research examined the relationship of female employment with three major contributing factors: ecological, motivational, and personal ability factors, in the state of Andhra Pradesh and its two highly urbanized districts (Ranga Reddy and Hyderabad). The 1991 census data were utilized as a major source. Specifically, the study tested the following six hypotheses: 1. Female work force participation rates vary by rural-urban residence, industrial categories, and National Industrial Classification (NIC). 2. Differential female work force participation rates can be observed by class of worker and occupational type. 3. Female workers vary by their educational attainment, age, and marital status. 4. The growth rate of female work force participation is significantly effected by change in industrial structure, time-period, and rural-urban residence over a period of time. 5. Female employment, especially in non-agricultural and non-household sectors, is significantly effected by education and urbanization, independently as well as combined. 6. The structure of female employment is the same between the state of Andhra Pradesh and its districts, Ranga Reddy and Hyderabad, but the size of participation may vary. The results of the data analysis confirmed all hypotheses.

The present research is carried out from the viewpoint of primarily space applications where human lives may be in danger if they are to work under these conditions. This work proposes to develop a one-degree-of-freedom (1-DOF) force-reflecting manual controller (FRMC) prototype for teleoperation, and address the effects of time delays commonly found in space applications where the control is accomplished via the earth-based control stations. To test the FRMC, a mobile robot (PPRK) and a slider-bar were developed and integrated to the 1-DOF FRMC. The software developed in Visual Basic is able to telecontrol any platform that uses an SV203 controller through the internet and it allows the remote system to send feedback information which may be in the form of visual or force signals. Time delay experiments were conducted on the platform and the effects of time delay on the FRMC system operation have been studied and delineated.

This research work has been prepared with the purpose of implementing a new innovative business idea in the city of Metropolitan Lima. Basically, our research work contains some preliminary steps before making our product known in the market. L’obésité s’accompagne d’une inflammation chronique du tissu adipeux qui est impliqué dans le développement de la résistance à l’insuline et du diabète de type 2. la sécrétion de cytokines pro-inflammatoires, l’augmentation du LPS circulant et l’hypoxie du tissu adipeux contrôlent la mise en place de cet état inflammatoire. Parmi les gènes induits par l’hypoxie, nous avons étudié l’implication de REDD1, un inhibiteur de mTORC1, dans le développement de l’inflammation in vitro et in vivo. 1. REDD1 est impliqué dans l’activation de l’inflammasome NLRP3 dans des macrophages. L’expression de REDD1 est induite par le LPS dans des macrophages dérivés de moelle osseuse de souris. L’absence de REDD1 inhibe l’activation des MAPKs, de NFκB et de l’inflammasome NLRP3 en réponse au LPS. Bien que REDD1 soit connu pour inhiber l’activité de mTORC1, REDD1 contrôlerait l’activation des voies pro-inflammatoires indépendamment de mTORC1. L’invalidation de REDD1 s’accompagne d’une diminution de la quantité de ROS (reactive oxygen species), associée à une diminution de l’expression de Nox1 et une augmentation de l’expression de GPx3. Dans un modèle de coculture entre des adipocytes et des macrophages, l’absence de REDD1 dans les macrophages inhibe l’établissement de la résistance à l’insuline adipocytaire. 2. Caractérisation métabolique des souris REDD1-/- soumises à un régime riche en graisses. Les souris REDD1-/- ont une légère amélioration de leur tolérance au glucose par rapport aux souris sauvages associée à une diminution de l’expression de marqueurs de l’inflammation du tissu adipeux
Obesity leads to the development of inflammation within the adipose tissue leading to the establisheent of insulin résistance and type 2 diabetes. Secretion of pro-inflammatory cytokines, increase circulating LPS and adipose tissue hypoxia regulate this inflammation. Among all the genes induced by hypoxia, we have studied the implication of REDD1, a mTORC1 inhibitor, in the development of inflammation in vitro and in vivo. 1. REDD1 is involved in the activation of NLRP3 inflammasome in macrophages. REDD1 expression is induced by LPS in bone marrow derived macrophages. Absence of REDD1 inhibits MAPKs, NFκB and NLRP3 inflammasome activation in response to LPS. Although REDD1 is known to inhibit mTORC1 activity, REDD1 regulates inflammation pathways independently of mTORC1. Absence of REDD1 is followed by a decrease in level of ROS (reactive oxygen species), associated to a decrease Nox1 expression and an upregulation of GPX3 expression. In a coculture cellular model between adipocytes and macrophages, invalidation of REDD1 in macrophages prevents adipocyte insulin résistance. 2. Metabolic characterization of REDD1-/- mice under high fat diet. REDD1-/- mice have better glucose tolerance than wildtype mice associated to a decrease of the expression of pro-inflammatory markers. After high fat diet, REDD1 -/- mice gain less weight than wild-type mice, without improvement of their glucose tolerance or adipose tissue inflammation pattern. In conclusion, REDD1 seems to be implicated in the regulation of inflammation

concept conciergeries is a relatively new concept on the European market, which is well

known on the US one. However, entering a new market with a new concept is not always easy

as there are some factors to be considered by the company.

The objective of this thesis is to provide the reader with a theoretical part, which is led by our

defined model in the introduction. In the empirical part the gathered information is presented

in order to compare the theory and the data gathered in the analysis. Furthermore the aim of

this thesis is to prove if a particular market – namely Sweden – is ready for introducing such a

concept. During the survey, the Swedish market and the Swedes has been regarded in more

detail to compare it with the pioneer (US) and the given market (France). To confirm/not

confirm the first statement, the authors made an interview with one potential customer in

Sweden. In the end a conclusion and some recommendations are given to the reader.

The Chinese tourists market has become the 4th largest inbound market for New Zealand in 2007; it also ranks 6th in terms of expenditure. Chinese visitors spent a total of NZ $352 million to the year December 2006, an average of NZ $3,340 per person (Ministry of Tourism, 2006). The latest statistics released from New Zealand Tourism Research Council show that 122,045 Chinese visited New Zealand by the year ended January 2008. These crucial statistics – market size, length of stay and average spend – indicate the necessity for the New Zealand tourism industry to understand and provide for Chinese expectations and requirements. Auckland is New Zealand's key gateway, and it is vital that Auckland provides a first good impression for the rest of country. Chinese travellers visit New Zealand with great desire and expectations of experiencing an exotic land. They expect Auckland, as the biggest city in New Zealand, to provide them with a memorable city experience in a developed country. The purpose of this research is to study Chinese tourists' expectations and travel experiences, with a particular focus on investigating whether Auckland can provide suitable services and experiences which match their requirements and expectations. The research will build on the study by Bull (1991) and Ryan (1995), which looked at supply demand relationships and at the tourism industry attempts to balance the supply with the actual demands. It will also build on work by Zhao (2006) and Qu (2006) which explored the dimensions of Chinese demand and distribution channel factors at both origin and destination. The goal is to establish which factors are adequate or inadequate. A synthesis of the demand supply situation will enable the development of recommendations. It is envisage that these recommendations could be adopted by local government and the tourism industry, either as actions or as improvements to policy. With regards to the purpose of this research, a qualitative method was decided as the most relevant approach. Qualitative methods are useful for revealing and understanding what lies are behind any phenomenon about which little is known. Grounded theory is a qualitative research method that was developed for the purpose of studying a social phenomenon, and to generate a theory relating to a particular situation (Strauss & Corbin, 1994). The historical bases of the grounded theory approach matches the aim of this research, namely to reveal the relationships between tourists and the tourism industry and to gain a better understanding of Chinese tourists' expectation and satisfaction, and the level of the service which the Auckland tourism industry provides to Chinese travellers needs to match the Chinese market and requirements of travellers. The interviews were conducted with both Chinese travellers and the Auckland tourism industry, including hotel managers, restaurant managers, gift/souvenirs shop assistants, tour guides, and so on. The research shows Auckland is not ready for Chinese market yet. This result comes from three aspects: firstly, most Chinese tourists do not have clear requirements or expectations of Auckland before they come to New Zealand. This is mainly because of the limited promotional materials available in China about Auckland. Secondly, Chinese tourists do not have many opportunities to get to know Auckland's attractions and activities after they arrive in Auckland. Auckland city is New Zealand's most popular urban tourism destination in terms of population and gateway function provides tourism facilities and services to Chinese tourists. However, the findings highlighted most Chinese tourists found Auckland's tourism attractions to be unattractive to them due to the language barrier and itinerary issues. Thirdly, the research also found Chinese tourists have been driven away due to lack of hotel accommodations in Auckland, especially in the shoulder or high seasons. Auckland's accommodation facilities are facing a challenge. The increasing domestic and international visitors' numbers and insufficient hotel accommodations will be the major problem for Auckland for the next decade. Most of the Chinese tourists who were interviewed expressed their hope of knowing more about Auckland, however, these hopes will go unrealised until the city provides better attractions and more services for them.

Introduction Trauma and emergencies contribute to the quadruple burden of disease in South Africa and being prepared for an emergency requires rapid access to emergency equipment, drugs and emergency trolleys to optimally manage an emergency. This is the first descriptive study looking specifically at essential emergency equipment, drugs and the emergency trolley required for the provision of optimal emergency care at Community Health Centres (CHCs) in the Western Cape Metropole. Aims and Objectives The aim of the study was to evaluate whether eight 24 hour emergency units at CHCs in the Western Cape Metropole had the appropriate and essential emergency equipment, drugs and emergency trolleys necessary for the delivery of optimal emergency care, using the Emergency Medicine Society of South Africa (EMSSA) guidelines as the audit tool. Objectives included: 1. To assess availability of essential emergency equipment 2. To assess availability of essential emergency drugs 3. To assess the functionality of existing emergency trolleys Methodology EMSSA guidelines were used as the evaluation audit tool to perform a survey of emergency equipment, drugs and emergency trolleys at eight 24 hour CHCs in the Western Cape Metro pole. Data collection for the study was conducted at the eight 24 hour CHCs over a 3 month period during the months of June 2012 to August 2012. The data was analyzed using the Statistical Package for Health Sciences (Statistica, version 10 of 2012) and Microsoft Excel. Results A total of 81 emergency equipment items, 43 emergency drug items (37 emergency drugs, 6 intravenous fluids) and 78 emergency trolley items were required to be in each emergency unit. An average of 62% of all recommended emergency equipment items, 80% of all emergency drugs and 52.4% of all emergency trolley items were found to be present in this survey. Essential emergency paediatric equipment including bag ventilation devices, Magill’s forceps, masks, intraosseous needles and appropriate blood pressure cuffs were found to be absent at 2 CHCs. All CHCs had access to a defibrillator and ECG machine but these were found to be dysfunctional at 2 CHCs due to expired batteries and no tracing paper being available. Expired first line emergency drugs (adrenaline and atropine) were found at certain CHCs. The recording of emergency trolley checklists and stocking of essential emergency items were found to be incongruent, inconsistent and not up to the recommended standard. Conclusion Essential emergency equipment and drugs and the functionality of emergency trolleys were found to be generally inadequate. Considerable deficiencies of essential emergency items were found, particularly paediatric equipment and drugs and this may negatively impact on resuscitative efforts and outcome in both paediatric and adult emergency care at CHCs in the Western Cape Metropole.
