O presente trabalho tem por objetivo analisar o processo de decadência da ordem Carmelita em Pernambuco, durante a política regalista do governo imperial, e a sua posterior restauração, a partir de 1894, com a vinda dos carmelitas espanhóis. Nessa perspectiva, procuramos estudar o contexto histórico do Império no qual foram impostas medidas restritivas aos antigos institutos monásticos dos beneditinos, franciscanos e carmelitas e, nesse meio, a conseqüente situação vivenciada pelos carmelitas diante do esvaziamento de seus templos religiosos. Diante desta finalidade, a análise documental referente à decadência do Carmo pernambucano nos possibilitou um maior entendimento acerca da redução da influência que os regulares exerciam na sociedade local e, ao mesmo tempo, serviu como subsídio para que entendêssemos como ocorreu a implementação do projeto de restauração da ordem no Brasil, a partir da proclamação da República. Dessa forma, percebemos que, para restabelecer o vigor da ordem, os frades remanescentes do Recife solicitaram ajuda da Província do Doce Nome de Maria que enviou alguns grupos de religiosos para empreender tal objetivo. Salientamos que a chegada desses religiosos permitiu a reorganização do carmelo pernambucano, assim como contribuiu para consolidar a romanização da Igreja Católica. Sendo assim, na elaboração desta tese, foram utilizados correspondências, atas, ofícios, estatutos e os livros patrimoniais da ordem

Apesar das investidas da Coroa Portuguesa para restringir o poderio da Igreja ao longo da Idade Moderna, uma ação sistemática nesse sentido foi possível apenas com novas ideias do século XVIII. Durante o consulado pombalino, embasado em teorias jansenistas, febronianistas e episcopalistas, o regalismo lusitano ganhou fundamentação doutrinal que o permitiram maior força e amplitude, conhecendo então seu apogeu, tanto perante a Igreja nacional quanto a Santa Sé. Dado o poderio material, privilégios e imunidades que as tornavam arredias à desejada centralização monárquica e à submissão dos diversos corpos da Igreja, as ordens religiosas constituíram-se alvos privilegiados das políticas regalistas ora estruturadas. Apesar disso, exceto pelos jesuítas, a historiografia nacional dispensou poucas atenções à investida regalista da Coroa e de seus representantes entre as diversas províncias religiosas espalhadas pelo território colonial, limitando-se a afirmações quase sempre genéricas. Dessa forma, no presente estudo analisamos justamente as ações de enquadramento da Província de Nossa Senhora do Carmo do Rio de Janeiro de 1750 a 1808, seus sucessos e limites, passos e contrapassos.
Despite the attempts of the Portuguese Crown to restrict the power of the nacional Church throughout the Modern Age, a systematic action was only possible with the new ideas of the 18th century. During the administration of Sebastião José de Carvalho e Melo, Marquis of Pombal, based on Jansenists, Febronianists and Episcopalists theories, the Portuguese Regalism won doctrinal grounds that allowed it greater strength and breadth, reaching its heyday, both before the national Church as the Holy See. Given its opulence, privileges and immunities that made them centrifugal bodies to the desired monarchical centralization and submission of Church, religious orders constituted one of the main targets of the regalists policies then structured. Despite this, except for the Jesuits, Brazilian historiography have dismissed little attention to the Regalism of the Lusitanian Crown and its representatives among the various religious provinces throughout the colonial territory, limited to statements almost always generic. Thus, in this study we analyze precisely the actions of framework of Província de Nossa Senhora do Carmo do Rio de Janeiro, from 1750 to 1808, their successes and limits, steps and countersteps.

Abstract:
La presente investigación estudia los debates político-teológicos entre dos grupos católicos a mediados del siglo XIX en el Perú. Estos formaron parte de disputas transnacionales que cambiaron el rostro de la Iglesia católica universal. Los grupos fueron el ultramontano y el liberal-regalista. Ambos tenían posturas contrapuestas sobre la relación Estado-Iglesia y la participación de la Sede Apostólica en la Iglesia local. El ultramontanismo fomentó el proceso romanizador y buscó la firma de un concordato que legitimara la autoridad romana sobre la Iglesia peruana. Por otra parte, el liberalismo-regalista rechazó la autoridad jurídica y doctrinal del papado sobre la Iglesia local y consideró el Patronato eclesiástico como un derecho estatal. La Revolución de liberal de 1854 y la Constitución de 1856 generaron un fuerte rechazo entre los clérigos, los militares, y otros grupos de la sociedad. A pesar de ello, los ultramontanos triunfaron como los portadores del discurso auténticamente católico. Además, la política de Ramón Castilla, que evitó enfrentarse con la Iglesia y se desligó a fines de 1857 de los liberales, favoreció la victoria ideológica del ultramontanismo.

This study is dedicated to investigate the clash between the Inquisition of Goa and the Portuguese Crown by the hegemony of control over governance of the court of King José I and his Premier Sebastião de Carvalho e Mello - The Marquês of Pombal - in the second half of the eighteenth century which culminated among many others result directly contributed to the premature closure of the court in 1774, or 47 years earlier than all others in the kingdom. Finally, this work was dedicated to uncover the relationship between the government, highly inclined to dictatorship, which Carvalho e Mello, and captained the Inquisition in Goa, historically treated as the highest court of the independent and insubordinate Inquisition Portuguese. There were many actions of Pombal on the Holy Office in order to submit it to the royal authority: its transformation into a royal court, its use to punish political crimes, the replacement of his inquisitors in absentia of the will of the Church, etc.
Esse estudo dedica-se a investigar o embate entre o Santo Ofício de Goa e Coroa portuguesa pela hegemonia do comando desse tribunal durante governação de D. José I e seu Premier Sebastião de Carvalho e Mello - O Marquês de Pombal - na segunda metade do século XVIII que culminou, entre várias outras consequências, na contribuição direta para o fechamento prematuro desse tribunal, em 1774, ou seja, 47 anos mais cedo que todos os outros do reino. Por fim, esse trabalho dedicou-se a descortinar essa relação entre o governo, altamente inclinado ao absolutismo, que Sebastião de Carvalho e Mello, o Marquês de Pombal, capitaneara e a Inquisição de Goa, historicamente tratada como o tribunal mais autônome e insubordinado da Inquisição portuguesa. Foram muitas as ações de Pombal sobre o Santo Ofício com o intuito de submete-lo à autoridade real : sua transformação em tribunal régio, sua utilização para punir crimes poliíticos, a substituição de seus inquisidores à revelia da vontade da Igreja, etc.

Abstract:
Juvenile weakfish Cynoscion regalis, life history was studied in the York River estuary, Virginia. to verify daily aging methods of juvenile fish, both male and female adults were induced to spawn by injection of 200 IU Human chorionic gonadotropin/Kg wet weight. Subsequent larval and juvenile fish were reared up to 275 days with wild plankton and a daily rotating diet of squid, liver, Anchoa mitchilli, and Menidia menidia. Otoliths and scales were examined for daily microincrements patterns. Otolith ring counts were highly variable (31% varied by &>& 15% among 3 counts). Two problems were evident: (1) Microincrements frequently split to form two increments; and (2) Otoliths from a size series of fish (6.12-13.1 mm) indicated that weakfish otoliths grew by bud formation rather than concentric deposition. Scale circuli showed little variation between counts (99.5% of 2 counts from an individual scale were the same). Daily scale deposition was suggested by rearing up to 100 days, after which ring deposition was less than daily, however further research is needed because only one fish was reared past 25 days. The advantages of scale circuli counting over otolith increment counting were increased precision and ease of preparation. Field samples were collected weekly from the York River channel, at night using a 4.9 m, 1.5 mm cod end, trawl, during the weakfish nursery period (Aug-Oct 1983). The new technique of daily aging by scales, was applied to 845 of 922 weakfish collected. Counts ranged from 3 to 100 circuli/scale. Three cohorts were defined from the 1983 0-age fish. Growth rates estimated from scales (0.76-1.13 mm/d) were similar to those from length frequencies (1.0-1.2 mm/d). Analysis of covariance showed a significant difference (0.05 level) in growth rates among cohorts and among stations, but third order interactions (station, cohort, growth rate) were not detected. Mortality/migration rates estimated from decline in mean catch were significantly different between cohorts 1 and 2 (0.05 level, t-test). Weakfish were first abundant as new recruits at the river mouth, and moved upriver as they grew. In the fall a reverse migration occurred. Birthdate frequency by station and date indicated that different cohorts used different areas of the York River.

The seventeenth-century Portugal is a geographic and social space, as well as a laboratory and a bed of intellectual, philosophical, political and religious disputes, aimed at reaching the most advanced nations in that period. The order of the Society of Jesus served the interests of the Council of Trent, since the Catholic world collapsed. Inserted in Portugal they are a decisive instrument for the formation of consciences in consonance with the tridentino dogmas, through a system of education dominated by the scholastic philosophy peripatetic elaborated in Ratio Studiorum. When he realized the Portuguese cultural backwardness and the need for a global reform of Lusitanian society, in the face of a modern and cultured Europe, and in an attempt to reverse the evils caused by Jesuitism in Portugal, the Marquis de Pombal sought to make changes in all the sectors of the Portuguese crown and, mainly, in education. The Oratorians were responsible for the transformation of Portuguese society and it is they who insert Portugal into modernity.
O Portugal setecentista constitui-se no espaço geográfico e social, bem como laboratório e canteiro das disputas intelectuais, filosóficas, políticas e religiosas, que visavam alcançar as nações mais adiantadas naquele período. A Ordem da Companhia de Jesus serviram aos interesses do Concílio de Trento, uma vez que o mundo católico desmoronava-se. Inseridos em Portugal são eles um instrumento decisivo para a formação de consciências em consonância com os dogmas tridentino, através de um sistema de ensino dominado pela filosofia escolástica peripatética elaborada no Ratio Studiorum. Ao perceber o atraso cultural português e a necessidade de uma reforma global da sociedade lusitana, diante de uma Europa moderna e culta e, na tentativa de reverter os males causados pelo jesuitismo em Portugal, O Marquês de Pombal procurou, então, realizar mudanças em todos os setores da coroa portuguesa e, principalmente, a na educação. Coube aos oratorianos a responsabilidade pela transformação da sociedade portuguesa e são eles que inserem Portugal na modernidade.

Abstract:
Inherent uncertainties in the stock assessment for weakfish have precluded accurate and consistent advice concerning the management of commercial and recreational fisheries. Error within ageing techniques, used to assess relative age frequencies within commercial and recreational harvest, has been cited as a potential source for uncertainty during assessments of the weakfish fishery. The implications for age-reading error on weakfish stock assessment were explored using measurement-error growth models (i.e. Chapter 1), ageing error matrices within a statistical catch-at-age framework (i.e. Chapter 2), and Monte Carlo simulations to gauge robustness of ignoring this type of uncertainty during fisheries stock assessment (i.e. Chapter 3). Measurement-error growth models typically resulted in weakfish that grew to reach larger sizes, but at slower rates, with median length-at-age being overestimated by traditional von Bertalanffy growth curves, at least for the observed age range. Measurement-error growth models allow for incorporation of ageing uncertainty during nonlinear growth curve estimation, as well as the ability to estimate the ageing error variance. Age-reading error was further considered during statistical catch-at-age analysis of the weakfish fishery, mainly through permutations of true catch-at-age via ageing error matrices constructed from estimates of the ageing error variance, thus reflecting changes in relative age compositions as a consequence of ageing uncertainty. As a result, absolute levels of key population parameters were influenced, but general trends in those parameters tended to be similar, with strong congruency across models as to weakfish stock dynamics in most recent years. Finally, Monte Carlo simulations showed that implications for age-reading error on weakfish stock assessment are varied, depending upon the direction and magnitude of the ageing uncertainty. However, relative trends of parameter estimates over time tended to be similar, resulting in proper allocation of weakfish stock status, regardless of the type of ageing error considered. Furthermore, assuming negligible ageing uncertainty within fishery-independent surveys appears reasonable, as simulations incorporating ageing error within indices of relative abundance showed similar patterns to situations that only considered observation noise.

La ricerca di Roberta Frigeni, svolta ad ampio spettro diacronico, è condotta su di una campionatura di specula principum - editi ed inediti - elaborati tra XII e XV secolo, e ne indaga il linguaggio quale referente privilegiato, rilevandone persistenze terminologiche e nuclei sintagmatici ricorrenti, al fine di individuare concetti utili a delineare un lessico politico proprio di questa testualità, in corrispondenza al sorgere dell’entità statale europea nel XIII secolo (con particolare riguardo all’area francese, ai regni di Luigi IX e Filippo il Bello). A partire da un’analisi critica delle tesi di Quentin Skinner circa la ‘ridefinizione paradiastolica’ del sistema delle virtù classiche entro il trattato De principatibus, lo studio innesca un percorso di indagine à rebours che - sondando il linguaggio - rintraccia nella trattatistica delle institutiones regum del XV secolo (Pontano, Patrizi, Carafa, Platina) e degli specula principum medievali (Elinando di Froidmont, Gilberto di Tournai, Vincenzo di Beauvais, Guglielmo Peraldo, Egidio Romano, Guido Vernani) una consonanza di motivi nella sintassi e nell’immaginario preposti ad illustrare le potenzialità semantiche del nome di prudentia, individuata quale unica virtù sopravvissuta alla ‘ridescrizione’ del codice etico operata da Machiavelli. Indagando i progressivi ampliamenti del campo semantico sorto attorno al nome della virtù di prudenza entro la letteratura speculare, la ricerca mostra come il dialettico rapporto con i lessemi di sapientia, astutia, fides ed experientia abbia avuto un ruolo determinante per il sorgere di un’immagine del principe emancipata dalla figura biblica del “rex sapiens”, e per la formazione di un lessico ospitale delle manifestazioni concrete del vivere politico ed economico. I processi di dilatazione e rarefazione del bacino semantico di prudentia sono, infatti, funzionali ad illustrare come il linguaggio della testualità speculare registri l’acquisizione di nuove strumentazioni teoriche grazie al rinnovamento delle fonti a disposizione lungo il secolo XIII, che - sostituendo progressivamente il più recente dossier aristotelico al solo apparato veterotestamentario - permettono di integrare la concezione delle virtù in senso operativo, adattandola alle esigenze politico-economiche dei nuovi contesti istituzionali monarchici.

Abstract:
Otoliths, scales, dorsal spines, and pectoral fin rays were compared to determine the best hardpart for ageing weakfish, Cynoscion regalis. Sectioned otoliths showed the clearest marks and were validated by the marginal increment method for ages 1-5. Traditionally-used scales were found to be less-precise and to underage older fish. Most weakfish from the Chesapeake Bay region were 200-600 mm TL and ages 1-4. Weakfish were not fully-recruited to commercial foodfish grades until age 2. Maximum observed age was 17 from a Delaware Bay fish collected in 1985. Current maximum observed ages were age 12 in Chesapeake Bay and age 11 in Delaware Bay. Fish older than age 6 were rare in both areas. Weakfish growth was well-described by the von Bertalanffy growth model (R&\sp2& = 0.98, N = 857). There was no evidence Delaware Bay weakfish reached a larger asymptotic length or size-at-age than Chesapeake Bay fish. However, maximum size and age from both areas has fluctuated in the past thirty years. Maximum size greatly increased from the late 1960's until roughly 1985, as did the numbers of large fish, apparently due to a series of strong year-classes, beginning in the late 1960's. Weakfish are multiple spawners with indeterminate fecundity and a spawning season from May to August, in the Chesapeake Bay region. Sex ratios were approximately 3:1, females to males, in 1990-1992. Mean length at first maturity for males and females was 164 and 170 mm TL, respectively. Most fish were mature by age one and all fish were mature by age 2. Spawning activity was not consistent throughout the spawning season or between 1991 and 1992. Batch fecundities ranged from 75,289-517,845 eggs/female and significantly increased with both TL and somatic weight. Spawning frequency was every 2-3 days in 1991 and every 12-13 days in 1992, leading to an average annual fecundity of 7,369,750 eggs/females in 1991 and 1,808,056 eggs/female in 1992. Patterns of spawning activity within and between years appeared closely associated with feeding success. Yield-per-recruit analysis indicated that, over a likely range of natural mortality rates, growth overfishing is currently occurring in the Chesapeake Bay region. Current t&\sb{lcub}\rm c{rcub}& is &\le& age 2, whereas maximum yield consistently occurred at t&\sb{lcub}\rm c{rcub}& &\ge& age 6.

Or ces études ont souvent exclu de leur champ de recherche l’espace géographique, pourtant fondamental, que constitue le monde méditerranéen. Cette thèse va donc s’articuler autour de l’interrogation suivante : l’art du portrait, et en particulier du portrait de cour peint, a-t-il été pour les princes italiens un support de leur politique, de l’affirmation de leur pouvoir et de leur puissance ? Le portrait de cour est-il le reflet de la conception du pouvoir des princes et des principaux mouvements intellectuels de l’époque moderne ? Après avoir replacé ces tableaux dans leur contexte historique, cette étude s’attachera à appliquer aux représentations du pouvoir une véritable analyse de contenu : étude des thèmes récurrents, périodicité de leur apparition, positionnement dans les plans de l’image, superficie occupée, mais aussi connotation de chacun à des référentiels culturels bien précis, autant de sens cachés et pourtant patents. Certaines principautés méditerranéennes ont en effet utilisé les principes picturaux et politiques de la Contre-réforme pour affirmer non seulement la supériorité de l’Église catholique, mais aussi leur pouvoir. Après la période de redécouverte de l’art du portrait que constitue la Renaissance, la Contre-réforme et le XVIIe siècle apparaissent comme l’utilisation du discours « visuel » pour s’opposer au discours écrit. Au contraire, au XVIIIe siècle, c’est l’influence des Lumières et des écrits philosophiques qui vont imprégner ces portraits de cour. Si certaines permanences demeurent, les décors ou certains éléments du costume montrent une imprégnation de la société et des sphères du pouvoir par les idées nouvelles, qu’elles soient politiques ou sociales
The study of the representation of power in modern times has developed significantly in recent years under the guidance of art historians and experts in semiotics. However, these studies have often excluded from their research the fundamentally important geographical space that is the Mediterranean world. Therefore, this thesis revolves around the following questions: Was the portraiture, and in particular the state portrait, a political statement of the power and legitimacy of the person represented in the painting? Is the court portrait a reflection of the conception of the power of the princes and the main intellectual movements of the modern era? After placing these paintings in their historical context, this study conducts a content analysis of the representations of power, focusing on recurrent themes, frequency of occurrence, and the placement of the image, and also to the connotation each specific cultural references, and meanings whether hidden or evident. Some Mediterranean principalities have indeed used the pictorial and political principles of the Counter-Reformation to assert not only the superiority of the Catholic Church, but also their own power. After the period of rediscovery of portraiture as an art form during the Renaissance, the Counter-Reformation and the seventeenth century are seen as the use of visual communication to oppose the written word. On the contrary, in the eighteenth century, the influence of the Enlightenment and philosophical writings permeates the state portraits. While there was a great deal of stability in portraiture over time, gradually the background decor and elements of the clothing began to reflect the arrival of new ideas, both political and social, within the spheres of power and in society more generally

Abstract:
Reducing apple (Malus x domestica Borkh.) crop load during bloom is a reliable option for increasing fruit quality and return bloom. In this thesis, multiple approaches to improving bloom thinning practices are discussed. The first project analyzed the pollen tube growth of several crabapple cultivars. Previous research had improved the use of bloom thinning chemicals, by coordinating the application timing with the pollen tube growth between pollination and fertilization. However, pollen tube growth rates have only been measured in a few genotypes. In Chapter 2, the pollen tube growth rates of five crabapple cultivars were measured in the styles of 'Fuji', 'Golden Delicious', and 'Pink Lady' flowers, at four temperatures 12, 18, 24 and 30 C. Complex relationships were found among paternal pollen tube growth, maternal cultivar, and temperature. Chapters 3 and 4 describe projects where organically-approved chemicals, including the biofungicide, Regalia, were evaluated for their ability to simultaneously reduce crop load and decrease early season disease infection. These chemicals were applied in conventionally managed orchards (Chapter 3), and in an organically-managed 'Honeycrisp' orchard (Chapter 4). The number of chemicals approved for bloom thinning is limited, especially in the Eastern U.S. where lime sulfur and oil applications are not permitted during bloom. These studies indicate Regalia, applied during bloom, can reduce crop load and provide early season disease control. The research presented in this thesis provides new knowledge that can be incorporated into crop load management practices in both conventional and organic apple orchards.

The biological characteristics of fisheries stocks that are assessed for management considerations are rarely homogeneous over time or space. However, stock assessment scientists largely ignore this heterogeneity in their models. This thesis addresses the effects of spatial and temporal heterogeneity on stock assessment models using Atlantic Weakfish (Cynoscion regalis) as a case study. First, spatial and temporal variation was incorporated into length-, weight-, and maturity-at-age estimates using mixed-effects models (Chapter Two). The resulting heterogeneous weight and maturity parameters then were applied to per-recruit analyses to examine the sensitivity of biological reference points to spatial and temporal variation in life history attributes (Chapter Three). Mixed-effects life history models incorporating spatial and temporal variation revealed distinct regional and annual trends that were not visible from standard homogeneous models. In several instances, the homogeneous modelling approach produced life history estimates that varied significantly from the spatial and temporal means produced by the heterogeneous models. In some cases, this difference was so great that the homogeneous means were much higher or lower than the heterogeneous means for all regions or years. Minimized AIC statistics revealed that spatially and temporally integrated mixed-effects models were more robust and descriptive of Atlantic Weakfish life history than the standard homogeneous models. Per-recruit and biological reference points derived from these life history estimates in Chapter Three were found to be highly sensitive to spatial and temporal variations in weight parameters. In several cases, reference points used as management targets were so significantly different that ignoring spatial and temporal heterogeneity in Atlantic Weakfish life history would likely cause overfishing and decline of Weakfish in certain regions and years.

Quantitative population dynamics modeling is needed to evaluate the stock status and fisheries management plans to provide robust model and management strategies. Atlantic Weakfish (Cynoscion regalis), one important commercial and recreational fish species along the west coast of Atlantic Ocean that was found to be declining in recent years, was selected as an example species. My study aimed to explore the possible spatial heterogeneity of CPUE (catch per unit effort) year trend based on three fishery independent surveys and explore the influence of nonstationary natural mortality on the fisheries management through a MSE (Management Strategy Evaluation) algorithm based on the Weakfish stock assessment results. Five models for catch rate standardization were constructed based on the NEAMAP (NorthEast Area Monitoring and Assessment Program) survey data and the ‘best' two models were selected based on the ability to capture nonlinearity and spatial autocorrelation. The selected models were then used to fit the other two survey data to compare the CPUE year trend of Weakfish. Obvious differences in distribution pattern of Weakfish along latitude and longitude were detected from these three surveys as well as the CPUE year trend. To test the influence of the model selection on the MSE, five stock-recruitment models and two forms of statistical catch-at-age models were used to evaluate the fishery management strategies. The current biomass-based reference point tends to be high if the true population dynamics have nonstationary natural mortality. A flexible biomass based reference point to match the nonstationary process is recommended for future fisheries management.

Phylogenetic and historical biogeographic hypotheses were explored for three groups of teleost fishes that are hypothesized to share a common biogeographic track. Distributed across the eastern Pacific/eastern Atlantic (EP/EA) biogeographic track of Rosen (1975) are the Scomberomorus regalis (Scombridae), Strongylura marina (Belonidae) and Hyporhamphus unifasciatus (Hemiramphidae) species groups comprised of one eastern Atlantic; three, three and four western Atlantic; and two, two and four eastern Pacific species, respectively, for each group. In addition, two species in the Strongylura marina and Hyporhamphus unifasciatus groups, have invaded freshwater drainages of Central and South America. Each of the three species groups were found to be monophyletic using on average 3000 base pairs (bps) of nucleotide sequence. Genes sequenced included: the entire ATP synthase 8 (ATPase 8; 169 bp) and 6 (ATPase 6; 684 bp), NADH ubiquione oxidoreductase subunit 2 (ND2; 1047 bp), and partial cytochrome b (cyt b; 800 bp) mtDNA protein coding genes; partial 12S (12S; 429 bp) and 16S (16S; 576 bp) ribosomal mtDNA genes; and portions of the nuclear encoded aldolase exon 5/intron 5 (ALD; &\sim&400bp) and creatine kinase b introns and exons 2 and 3 (CK; &\sim&1042 bp). The analysis of multiple genes permitted an evaluation of their phylogenetic signal and usefulness in biogeographic reconstructions. Mitochondrial protein-coding genes resulted in well-supported resolution of sister species relationships, while the nuclear-encoded genes helped resolve basal relationships. However, contrary to expectations, the mtDNA ribosomal genes did not perform as well. Additionally, in Scomberomorus a morphology-based phylogeny was included in the analysis. In general phylogenetic hypotheses inferred for each species group were consistent with expectations of the EP/EA generalized biogeographic track. The results for each species group indicated a basal position for the eastern Atlantic taxa S. tritor, St. senegalensis and Hy. picarti, with closest relationships being either transisthmian (geminate) or, within the eastern Pacific or western Atlantic, for the derived New World taxa. Vicariant and dispersal hypotheses, consistent with paleogeological and paleoceanographic evidence, and life history and dispersal abilities are offered to explain the biogeographic histories of.

This thesis examines Alexander�s conception of kingship, his relationship with royal traditions in the three great kingdoms of the Near East, and the concomitant transformation of the king�s court by which Alexander created a distinctive royal insignia and introduced new court personnel and protocol. Section I ("Alexander and Near Eastern Kingship") contains Chapters I, II, and III. Section II ("The Transformation") comprises Chapters IV to VI. In Chapter I, I examine the Macedonian background of Alexander�s court and his native conception of kingship. Chapter II is a study of the kingship of Egypt. Chapter III deals with the kingship of Babylon and Persia. I then turn to an analysis of Alexander�s policies towards the Persians and the concept of the "kingship of Asia," as this was understood by Alexander. This crucial concept is to be distinguished from the kingship of Persia, a position which Alexander supplanted and replaced with his personal kingship of Asia. In Section II, three chapters are devoted to an analysis of the transformation of Alexander�s court. Chapter IV covers the origin and significance of Alexander�s royal insignia. La structuration spatiale de la diversité floristique constitue un outil de base pour la gestion durable des forêts tropicales. La complexité des forêts tropicales s’accommode mal des généralisations et des extrapolations qui aboutissent le plus souvent à des résultats imprévisibles. La présente étude a pour but (1) d’identifier les groupements végétaux des forêts matures de terre ferme de la RBD et partitionner la variance floristique ;(2) de comparer les forêts matures de terre ferme de la RBD à d’autres forêts tropicales sur la base de la diversité, de la structure et de la dominance des taxons; (3) d’évaluer la structuration spatiale de la diversité dans les forêts à Gilbertiodendron dewevrei de la RBD; (4) d’évaluer l’influence des densités de Raphia regalis sur la diversité des forêts matures de terre ferme de la RBD; (5) de déterminer le statut phytogéographique des forêts à Coula edulis du sud-est Cameroun ;(6) de Comparer les différents types forestiers de forêts matures de terre ferme du sud-est Cameroun sur la base de la diversité et de la structure. La Réserve de Biosphère du Dja (RBD) ainsi que le massif forestier Ngoyla Mintom font partie sont faiblement perturbés par les activités anthropiques et font partie de l’initiative de conservation transfrontalière TRIDOM. La variance floristique dans la RBD est principalement expliquée par les distances géographiques, l’indice de continentalité et la topographie. La part de variance floristique non expliquée est élevée et due aux facteurs stochastiques et à l’hétérogénéité environnementale non détectée. La diversité locale dans les forêts matures de terre ferme de la RBD est élevée, bien que n’atteignant pas les degrés extrêmes observés dans certaines forêts tropicales d’Asie et d’Amérique du Sud. La diversité de différenciation est faible et positivement corrélée aux distances géographiques. Quelques taxons sont dominants tandis que le nombre d’espèces rares est élevé. Dans les forêts à Gilbertiodendron dewevrei, les strates arborescentes sont moins diversifiées que celles des forêts hétérogènes aussi bien à l’échelle locale qu’à celle du site. A surface égales, les strates herbacées des forêts monodominantes sont aussi diversifiées que celles des forêts hétérogènes. La diversité de différenciation est négativement corrélée à l’amplitude de la monodominance et aux distances géographiques même en absence des effectifs de Gilbertiodendron dewevrei. Raphia regalis influence négativement les densités dans les strates arborescentes, contribuant ainsi à une diversité locale artificiellement élevée. Les forêts à Coula edulis du sud-est Cameroun correspondent à la continuité de la partie continentale du bas-guinéen. Le district congolais du Dja suit les contours de la rivière dont il porte le nom. Les types forestiers matures de terre ferme du sud-est Cameroun diffèrent aussi bien du point de vu de leur structure que de leurs diversités à différentes échelles. Toute intervention doit tenir compte des spécificités de chacune d’entre elles. Les taux élevés d’espèces rares et la faible diversité des forêts à Gilbertiodendron dewevrei, l’isolement relatif des forêts à Coula edulis du district congolais du Dja, les faibles densités d’espèces arborescentes dans les forêts à Raphia regalis et le statut conservatoire de l’espèce dominante, la diversité élevée des forêts mixtes à l’échelle du sud-est Cameroun et du bassin du Congo.

La ricerca prende avvio dagli anni giovanili, vissuti da Giancarlo Brasca nel contesto dei rami giovanili dell’Azione Cattolica ambrosiana. Il primo capitolo si propone di ricostruire quale clima e quali personalità abbiano influito sulle scelte effettuate dal giovane Brasca. Da questo punto di vista particolare attenzione è stata dedicata al rapporto con padre Agostino Gemelli e con monsignor Francesco Olgiati. L’intensità del legame con i due protagonisti del mondo cattolico milanese del Novecento costituisce un elemento significativo anche nella scelta vocazionale che legò Brasca ai Missionari della regalità di Nostro Signore Gesù Cristo, Istituto secolare fondato da Gemelli. Il secondo capitolo della tesi si prefigge dunque di ricostruire il significato dell’adesione di Brasca alla proposta di vita dei Missionari e l’apporto da lui fornito alle vicende dell’Istituto e all’evoluzione dei rapporti di quest’ultimo con l’Università Cattolica. La scelta vocazionale di Brasca rappresenta infatti la chiave di lettura imprescindibile per comprendere e ricostruire le vicende di Brasca all’interno dell’Università Cattolica dai primi anni Quaranta fino alla sua scomparsa nel 1979. Il terzo e il quarto capitolo ricostruiscono dunque le vicende di Brasca sullo sfondo degli snodi fondamentali che segnarono lo sviluppo dell’ateneo: dalla laurea in Filosofia, passando per l’esperienza di direttore della Biblioteca, per giungere agli incarichi amministrativi di primissimo piano come segretario di amministrazione e, dal 1971, direttore amministrativo.
The research begins from the years of one’s youth, lived by Giancarlo Brasca in the contest of Azione Cattolica’s young lines in Milan. First chapter proposes to reconstruct which atmosphere and which personalities influenced young Brasca’s choices. On this point of view particular attenction was dedicated to relation with Father Agostino Gemelli and Monsignor Francesco Olgiati. The intensity of relationship with two protagonists of catholic world in Milan represents a significant fact also in vocational choice that binded Giancarlo Brasca with Missionari della regalità di Nostro Signore Gesù Cristo, secular institution founded by Gemelli. So second chapter of dissertation proposes to reconstruct the meaning of Brasca’s adhesion to Missionari’s life proposal, his contribution to story of institution and to development of relation between this and Catholic University. Brasca’s vocational choice represents indeed a necessary keynote to understand and reconstruct Brasca’s story in Catholic University from ’40 to his death in 1979. So third and fourth chapters reconstruct Brasca’s events on the contest of Catholic University: from Philosophy degree moving to experience which director of Library, to arrive to very important administrative assignments like secretary of administration and, from 1971, administrative director.

Oggetto d’indagine è il complesso delle celebrazioni e dei preparativi connessi con il passaggio di Maria Teresa d’Austria ed il solenne ingresso dell’arcivescovo Stampa nel 1739, casi esemplificativi di ritualità cittadina e cerimoniali pubblici della prima metà del secolo, rappresentazioni che coinvolsero la città. A confronto è stato analizzato anche il rito di possesso di Maria Teresa, compiuto in absentia nel 1741. Attraverso l’analisi comparativa delle fonti primarie, a stampa e manoscritte, nonché iconografiche, si sono ricostruiti il ruolo delle norme di etichetta, dei cerimoniali, l’intera drammaturgia degli eventi festivi. La ricerca ha altresì riportato alla luce gli apparati allestiti dal Collegio dei Giureconsulti, ignorati dagli studi. Ha evidenziato meccanismi retorici, azioni, aspetti della rappresentazione che permangono, alcuni dei quali appartengono alla sfera degli archetipi. Benché semplificati, i cerimoniali mostrano ancora marcato il culto per la regalità. L’ingresso trionfale si rivela momento significativo per ricondurre la città verso l’immaginario delle origini dell’identità cittadina e rifondare un tempo nuovo. Si evidenzia la necessità della festa, di momenti rituali di rappresentazione e di autorappresentazione; il ruolo drammaturgico e non esornativo di elementi di decoro, quali il baldacchino, l’arco, la carrozza, il ritratto, in una cultura che attribuiva ancora grande valenza alle immagini.
The object of this research is the whole of the celebrations and preparations connected with the passage of Maria Theresa of Austria and the solemn entry of Archbishop Stampa into Milan in 1739, both exemplifications of the 18th century first half civic rituality and public ceremonials, and representations which engaged the city. A confrontation has also been made with the possesso of Maria Theresa, enacted in absentia in 1741. By means of a comparative analysis of the primary sources – printed, hamdwritten and ichnographic as well – it has been possible to reconstruct the role of etiquette norms, ceremonials and of the entire dramaturgy of festival events. The research has also brought to light the apparati set up by the Collegio dei Giureconsulti, so far ignored by studies. It has highlighted rhetorical mechanisms, actions, representational aspects which remain, some of them pertaining to the archetype sphere. Even if simplified, the ceremonials still show a strong cult of regality. The triumphal entry proves to be a meaningful moment to bring the city back to the imaginary of the origins which constitutes its identity and to renovate a new era. The necessity of the feast, of ritualistic moments of representation and auto representation is pointed out; and also the dramaturgic and not ornamental role of decorative elements like the baldachin, the arch, the carriage, the portrait, in a culture which conferred great value to images.

My thesis investigates some of the conceptual ideas related to my studio work, both in terms of theory and contemporary practice. This thesis focuses how the visual images formally operate, as well as the larger framework of discourse that surrounds my practice. In my work, the habitual, incessant process of cutting and extraction and the subsequent meticulous reconfiguring use the strategies of repetition and labor. These sustained, ongoing acts have the possibility to be generative or transformative rather than simply repetitive. This thesis also explores the found object, complicating the classification and knowledge systems of the source image with odd juxtapositions and reconfigurements. O presente trabalho dedica-se ao estudo da Mesa da Consciência e Ordens no Brasil, tribunal criado por Dom João VI, por meio do Alvará de 22 de abril de 1808, como parte da política de reprodução do aparato administrativo lisboeta no Rio de Janeiro, em virtude da transferência da corte portuguesa para o Brasil. A primeira parte deste trabalho foi dedicada à análise da Mesa da Consciência em Portugal, criada em dezembro de 1532 por Dom João III. No intuito de compreender um tribunal que incorporava funções como aconselhar a consciência do monarca, conhecer questões administrativas envolvendo a gestão de Ordens Militares Religiosas, além de diversas outras matérias, foram examinados os comandos legais expedidos com a finalidade de regulamentar a organização, funcionamento e competência do tribunal português. A segunda parte deste trabalho foi dedicada ao estudo da Mesa da Consciência e Ordens no Brasil. Com o objetivo de estudar a atuação da corte brasileira, foram analisadas as consultas proferidas pelo tribunal. A íntegra da transcrição das consultas analisadas, bem como a cópia digitalizada dos manuscritos consultados foram incluídas nos anexos a este trabalho. Por último, foi analisado o processo que levou ao encerramento das atividades da Mesa no Brasil em 1828. O objetivo deste trabalho foi o de investigar o funcionamento do Tribunal da Mesa da Consciência e Ordens no Brasil entre 1808 a 1828, com a finalidade de mellhor compreender, sob a ótica da história do direito e com lastro nas fontes jurídicas examinadas, a gênese da Mesa portuguesa, as regras que disciplinavam o seu funcionamento e competência, o modo pelo qual tal tribunal foi transferido e instalado no Rio de Janeiro, suas decisões, as causas de sua extinção e o destino de seu acervo documental.
The present work is dedicated to the study of the Mesa da Consciência e Ordens in Brazil, court created by Dom João VI through the Alvará of April 22, 1808, as part of the policy of reproduction of the administrative apparatus of Lisbon in Rio de Janeiro, due to the transfer of the Portuguese court to Brazil. The first part of this work was dedicated to the analysis of the Mesa da Consciência e Ordens in Portugal on December of 1532 by Dom João III. In order to understand a court that included functions such as advising the monarch's conscience, to know administrative matters involving the management of Military Religious Orders, besides several other matters, were analyzed the legal rules issued with the purpose of regulating the organization, functioning and competence of the Portuguese court. The second part of this work was dedicated to the study of the Mesa da Consciência e Ordens in Brazil. In order to study the Brazilian court's performance, the court's consultations were analyzed. The complete transcription of the consultations analyzed, as well as the digital copies of the manuscripts consulted were included in the annexes to this work. Finally, the process that led to the closure of Mesa's activities in Brazil in 1828 was analyzed. The objective of this work was to investigate the functioning of the Tribunal of the Mesa da Consciência e Ordens in Brazil between 1808 and 1828, in order to better understand, from the point of view of the history of law and according to the legal sources examined, the genesis of the portuguese Mesa, the legal rules issued with the purpose the organization rules issued with the purpose of regulate its organization and competence, the manner in which such court was transferred and installed in Rio de Janeiro, its decisions, the causes of its extinction and the destination of its documentary collection.

Polychlorinated biphenyls (PCBs) were first synthesized in 1881 by Schmidt and Schulz and their commercial production began in 1929. In 1966, PCBs were first identified as pollutants and subsequent investigations showed that they are ubiquitous. Many studies have even proven that PCBs are hazardous to human and other animals. Xylaria regalis is a wood-grown saprophytic fungus that is capable of PCBs degradation. After the comparison of five culture media, we found that Xylaria regalis grew in SDB medium could degrade more PCBs than in other media. Mycelium extract from 10-day growth mycelium could degrade 70% of Aroclor 1254 and culture medium from 10-day growth mycelium could degrade 67% of Aroclor 1254. Therefore, we chose SDB for the growth of Xylaria regalis and used mycelium extract for the investigation of PCBs degradation by Xylaria regalis. The results showed that the optimal reaction temperature for mycelium extract was 30 ℃. After treating mycelium extract for 4 hours at 30 ℃, 85% of Aroclor 1254 was degraded. The optimal reaction pH value for mycelium extract was pH 7. After treating mycelium extract for 2 hours at pH 7, 74% of Aroclor 1254 was degraded. Mycelium extract had the best temperature stability at 30 ℃, and it could degrade 43% of Aroclor 1254 even treated at 50 ℃ for 4 hours. At pH 7, mycelium extract had the best pH stability, and after treating at pH 3 and pH 10 for 4 hours, it could still degrade 45% and 30% of Aroclor 1254, respectively. We also tested the ability of mycelium extract for degradation of three commercial PCBs. The results showed that mycelium extract could degrade 69% of Aroclor 1242, 76% of Aroclor 1254, and 48% of Aroclor 1260, respectively. When Aroclor 1254 was added into soil to simulate the condition of soil with PCBs pollutants, we found that 78% of Aroclor 1254 in soil was degraded after the soil treated with mycelium extract for 2 hours at 30 ℃. Mycelium extract was purified by TSK gel Phenyl-Toyopearl 650 M column and there were four activity peaks in three eluents of different ammonium sulfate concentrations. After analyzing four activity peaks by SDS-PAGE, we suggested that there was more than one kind of PCBs degrading enzyme in mycelium extract.

Contextualizado no período cronológico da transição da Monarquia para a República, no Portugal de finais do século XIX e do início do século XX, o presente trabalho pretendeu indagar, por meio da decomposição do magistério de um bispo, o processo de redefinição dos modelos episcopais na contemporaneidade. Dito de outro modo, destrinçando o percurso eclesiástico de Mendes Bello até ao topo da hierarquia católica do reino, procurámos compreender as implicações decorrentes do processo de reconfiguração do catolicismo romano em Portugal, nomeadamente no que respeita à redefinição do seu enquadramento sócio-institucional, detalhando os seus dinamismos e exigências na composição dos magistérios prelatícios finisseculares, ainda herdeiros de uma eclesiologia regalista de funcionalização dos hierarcas católicos. Em simultâneo, por meio da observação da sua retórica pastoral e parlamentar, tivemos como objectivo destrinçar o protagonismo de Mendes Bello na Igreja da época, analisando a sua actuação quer no panorama da crise nacional e da transição de regime, quer na reconfiguração do movimento católico português no contexto da romanização – do laicado ao clero – tanto no Algarve como em Lisboa. Contextualized in the chronological period of the transition from the Monarchy to the Republic of the late nineteenth century and early twentieth century, the present work seeks to investigate, through the decomposition of the magisterium of an bishop, the process of redefinition of the episcopal models in contemporary times. In other words, through the unraveling of the Mendes Bello’s ecclesiastical path to the top of the Catholic hierarchy of the kingdom, we will try to understand the implications of the process of reconfiguration of Roman Catholicism in Portugal, in particular as regards the redefinition of its socio-institutional framework, detailing Its dynamisms and demands in the composition of finisecular prelattic magistrates, still heirs of a regalist ecclesiology of functionalization of the Catholic hierarchs. At the same time, through the observation of his pastoral and parliamentary rhetoric, we had as objective to unravel the prominence of Mendes Bello in the Church of the time, analyzing his action in the panorama of the national crisis, in the transition of the regime, and yet in the reconfiguration of the catholic movement portuguese in the context of Romanization - of the laity to the clergy - both in the Algarve and Lisbon. This, in order to decipher the requirements arising from ralliement politique in the exercise of his episcopal ministry.

Polycyclic aromatic hydrocarbons (PAHs) are mainly formed as products of the incomplete combustion of fossil fuels and organic compounds. They consist of benzene homologs which have two or more fused aromatic rings. As the molecular weight of PAHs increases, its toxicity and hydrophobicity increase, which indicates that it is hard to dispose PAHs in the environment. In our laboratory, we find that the enzyme of the Xylaria regalis has the ability to degrade PAHs. In this study, the fungus Xylaria regalis was incubated in the SDB medium. Mycelia of Xylaria regalis were harvested by filtration and broke by pestle. Then the crude extract was boiled in the 100℃ water and purified by ionic exchangers (CM, QAE), hydrophobic interaction (phenyl 650M) and gel filtration (HW-55, Bio-Sil SEC-400 HPLC) column. The purity of the PAHs-degrading enzyme that degraded BaP and Pyr was increased 183 and 196 fold and its specific activity was reached to 120 and 477 unit × 10-4 /mg, and the recovery was 62 and 67%, respectively. The optimal pH and pH stability for PAHs-degrading enzyme activity was 8.5. The PAHs-degrading enzyme was heat stable protein, because it could maintain the enzyme activity after treated at 100℃ for 1hr. Mg++、Sr++、Ba++、Ni++、Mn++ ions inhibited the enzyme activity. The PAHs-degrading enzyme had a molecular weight of 29 kDa as determined by HPLC gel filtration Bio-Sil SEC-400. By SDS-PAGE electropheresis, single protein band, its molecular weight about 29 kDa, was also found. Therefore, the result suggested that the PAHs-degrading enzyme was monomer.

Sendo um dos estadistas portugueses mais referenciados do século XVIII o primeiro Conde de Oeiras e Marquês de Pombal teve um papel muito relevante nas reformas realizadas no reinado de D. José. Habitualmente defende-se que foi a ideologia política de Carvalho e Melo a ditar a sua política eclesiástica, mas neste trabalho, inserindo Pombal na corrente do iluminismo católico, propomos que foi esta sensibilidade cultural e religiosa a influenciar as reformas pombalinas de um modo geral e as reformas eclesiásticas em particular.
This dissertation seeks to make an approach to the religion of Sebastião José de Carvalho e Melo. As one of the most referenced Portuguese statesmen of the eighteenth century, the first Count of Oeiras and Marquis of Pombal, he played a very important role in the reforms carried out during King José's reign. It is commonly argued that it was the political ideology of Carvalho e Melo that made his ecclesiastical policy, but in this work, inserting Pombal into the Catholic Enlightenment current, we propose that it was his cultural and religious sensibility that influenced the Pombaline reforms in general and ecclesiastical reforms in particular.

Abstract Cellulose is a linear polysaccharide of glucose residues connected by β-1,4- glucosidic linkages. Native crystalline cellulose is insoluble and occurs as fibers of densely packed, hydrogen bonded chains of 15 to 10,000 glucose units. Its density and complexity make it very resistant to hydrolysis without chemical or mechanical degradation. As a result, utilization of cellulase is the most important step during energy conversion from cellulosic biomass. A cellulosic enzyme system consists of three major components: endo-β- glucanase, exo-β-glucanase andβ-glucosidase. Endo-β-glucanase makes random scission of cellulose chains yielding glucose and cello-oligo saccharides. Exo-β- glucanase commits exo-attack on the non-reducing end of cellulose with cellobiose as the primary structure. β-glucosidase hydrolyzes cellobiose to glucose. These three enzymes act synergistically to degrade cellulose. In this study, we isolate, purify and characterize endo-β-glucanase by Ascomycete Xylaria regalis. The cellulose-containing medium is used to induce the production of the cellulose-degrading enzymes. When these enzymes are produced at maximum level, we harvest and filtrate the culture medium. The filtrate is purified by anion exchanger column (QAE) and hydrophobic interaction column (phenyl-650M). The purification fold of endo-β- glucanase increases 4.3 times, the specific activity approaches 3.7 unit/mg, and the recovery is 27.6%, respectively. SDS-PAGE shows that the molecular weight of endo-β-glucanase is 47 kDa. The optimal pH of the enzyme is 5 and the optimal temperature of the enzyme is 50℃. The pH stability of enzyme is between 4 and 5. The thermostability of enzyme is 40℃. Some metal ions, like Mg2+, Zn2+, Cu2+, Fe3+, Hg2+ and Ba2+, slightly enhance the enzyme activity; however, Co2+ ion suppresses it.

In this study, the xylanase and β- xylosidase secreted by Xylaria regalis were isolated , purified and characterized. We used the avicel and oat spelts xylan as the only carbon source to induce the production of xylan-degrading enzyme by this fungus. The fungus was grown in a rotary shaker at 25℃ for 26 days. And the culture filtrate was collected and further purified by anion exchanger column (QAE Sephadex A 25), hydrophobic interaction column (phenyl 650M) and gel filtration column (Sephadex G 75), respectively. The specific activity of the finally purified xylanase was 1.30x103 unit /mg and its purity was increased 11 fold, and the recovery was 34.6%. The molecular weight of xylanase estimated by SDS-PAGE was approximately 30.8 kDa. The optimal pH and temperature for this enzyme was pH 5.5 and 50℃. The xylanase was stable above pH 5 at low temperature. When the enzyme treated with distinct metal ions, the activity was significantly inhibited by Cu2+and Hg2+. Xylanase activity was also inhibited by EDTA or β-mercaptoethanol. The results show that the active site of xylanase may have disulfide bond which can stabilize the xylanase third structure and certain metal ions may be the cofactor of the enzyme. We collected the culture filtrate for the purification of β-xylosidase. Then culture filtrate was precipitated by 80% ammonium sulphate and further purified by hydrophobic interaction column (phenyl 650M), anion exchanger column (QAE) and gel filtration column (Sephadex G 75), respectively. The specific activity of the finally purified β-xylosidase was 4.00x10-1 unit /mg. The recovery of the enzyme was 24.2% and the purity was increased 22.97 fold. The molecular weight of β-xylosidase estimated by SDS-PAGE was approximately 45 kDa . The optimal pH and temperature for β-xylosidase was pH 5.5 at 50℃. β-xylosidase was stable above pH 5 at low temperature. Most of the tested metal ions have no remarkable effects on the activity of the β-xylosidase, whereas the Fe2+and Hg2+ significantly inhibit it. In addition, the activity of the β-xylosidase was not affected by EDTA or β-mercaptoethanol.

And the PCBs-degrading enzyme was further purified by anion exchanger column (QAE Sephadex A-25) and hydrophobic interaction column (Phenyl-650M). The purity of the PCBs-degrading enzyme was increased to 11.60 fold, the specific activity of the PCBs-degrading enzyme was reached to 8.90 x 10-2 unit/mg, and the recovery of the purified enzyme was 108.29 %. The molecular weight of the PCBs-degrading enzyme was found to be approximately 22.5 kDa by HPLC gel filtration and SDS-PAGE. The optimal pH of the enzyme was 7 and the optimal temperature of the enzyme was 30 ℃. The pH stability of the enzyme was 7 and the thermostability of the enzyme was 30 ℃. The iron ions that we studied in different concentration can inhibit the enzyme activity. When EDTA was added in the enzyme solution, the enzyme activity was inhibited.

In recently years, we have make many pollutions in our environment, such as polycyclic aromatic hydrocarbons (PAHs) are mainly formed as the products of the incomplete combustion of fossil fuels and organic compounds. These pollutants consist of benzene homologs which have two or more fused aromatic rings, and most of those are more poison and hydrophobic with the number of ring increased. When their hydrophobic property is increased, it is hard to degrad them in the environment. In this study, we isolate, purify and characterize the polycyclic aromatic hydrocarbons-degrading enzymes by Xylaria regalis. The SDB medium was used to culture the fungus for the production of the PAHs-degrading enzyme. When the enzyme was produced at maximum level, the culture medium was harvested and then filtrated. The filtrate was precipitated by 80% ammonium sulphate and furhter purified by hydrophobic interaction column (phenyl 650M), anion exchanger column (QAE), gel filtration column (HW-55). The purity of benzo(a)pyrene-degrading enzyme was increased 205 fold and its specific activity reached to 9.1636 unit/mg, and the recovery of the purified enzyme was 19.47%. The molecular weight of enzyme was found to be approximately 48 kDa by HPLC gel filtration and confirmed by SDS-PAGE. The optimal pH of the enzyme was 7 and the optimal temperature of the enzyme was 30℃. The pH stability of enzyme was 8 and the temperature stability of enzyme was 30℃. Bap-degrading enzyme catalyzed the oxidation of veratryl alcohol to veratraldehyde without H2O2 and the oxidation of Mn2+ by H2O2 to Mn3+.The metal ions that we studied in different concentration can enhance enzyme activity. When EDTA and β-mercaptoethanol was added in the enzyme solution, the enzyme activity was not inhibited.

The late seventeenth century was the golden age of the printed descriptive catalogue. Nehemiah Grew’s 1681 catalogue, Musæum Regalis Societatis, printed for London’s Royal Society, exemplifies this elaborate published genre of early museum literature during a particular moment in time when collecting and ordering were methods of understanding the world. This thesis explores the importance of ephemeral texts in historical study by analyzing the prose used in Grew’s catalogue. Musæum Regalis Societatis opens a window onto late seventeenth-century English culture, providing insight into Grew’s opinions about contemporary religious and political debates and illustrating trends within scientific thought; most notably, the influence of Francis Bacon’s new empirical methods on Grew’s object descriptions. This results in a densely descriptive catalogue with vivid object descriptions, creating a virtual guide to the Repository. However, with the eighteenth-century development of museums as sites of leisure and the rise of experts and professionals in the burgeoning scientific disciplines, there is a noticeable decline in this genre of descriptive catalogue. Thus, Grew’s catalogue exemplifies a critical moment in the late seventeenth century in which scientific catalogues were published for a broad general public.

As espécies invasoras podem interferir em processos ecológicos previamente estabelecidos num ecossistema, existindo assim uma preocupação acrescida com as espécies que possam prejudicar populações de espécies nativas com valor comercial. É o caso da corvinata-real Cynoscion regalis (Pisces: Sciaenidae), também conhecida por corvina americana ou rainha, uma espécie nativa da costa NE da América do Norte, que estabeleceu uma população invasora no estuário do Sado, tendo já sido registada em vários pontos da Península Ibérica. Os pescadores manifestaram a sua preocupação com a competição que possa existir entre a corvinata-real e os peixes nativos com valor comercial, contudo, não há evidências científicas que demonstrem que a corvinata-real está a competir com espécies nativas por alimento e espaço. Assim, o principal objetivo deste trabalho é fornecer as primeiras informações relativas à ecologia da população de corvinata-real no estuário do Sado, enquanto os seus objetivos específicos são: i) determinar a dieta e estratégia alimentar usada pela corvinata-real, através da análise de conteúdos estomacais, bem como de três espécies nativas com valor comercial – corvina-legítima Argyrosomus regius, robalo Dicentrarchus labrax e sargo Diplodus sargus; ii) inferir a sua posição trófica e a sobreposição de nicho trófico com estas três espécies nativas, através da análises de isótopos estáveis de carbono e azoto; iii) inferir o uso do habitat da corvinata-real através da análise da química de otólitos e averiguar a sobreposição de habitat com as três espécies nativas; iv) discutir os potenciais impactos desta invasão nos recursos pesqueiros do estuário do Sado, tendo em consideração a dieta e o habitat usado pela espécie ao longo do ciclo de vida; v) avaliar o potencial da corvinata-real como um novo recurso pesqueiro, através de inquéritos realizados a consumidores voluntários, e acções de sensibilização (parceria com chefs locais e publicação de dois comunicados de imprensa). Os resultados dos conteúdos estomacais apontam que no estuário do Sado, a corvinata-real é predadora de crustáceos (53%) (principalmente mysidaceos, carídeos, isópodes e caranguejos), peixes (45%) (principalmente peixe-rei Atherina sp. e anchova Engraulis encrasicolus e cefalópodes (2%). Um caso de canibalismo foi verificado. Assim, esta espécie parece alimentar-se dos mesmos grupos de presas e em proporção semelhante ao que acontece na região nativa. A estratégia alimentar é generalista e a população aparenta ser composta por indivíduos especialistas, ou seja, indivíduos que têm preferência por diferentes presas, havendo assim uma baixa sobreposição de presas entre indivíduos. Este foi certamente um fator importante que influenciou esta espécie a estabelecer-se nesta região. Os resultados da análise de isótopos estáveis revelaram que a corvinata-real, a corvina-legítima e o robalo estão a alimentar-se no mesmo nível trófico, enquanto que o sargo está um nível trófico abaixo, uma indicação de que o sargo está a consumir presas diferentes ou em diferente proporção. Não houve uma sobreposição significativa de nicho isotópico, ou índice de Pianka (calculado com base no número e tipos de presas consumidas), entre a corvinata-real e as outras três espécies, rejeitando a hipótese de que a corvinata-real está a competir por alimento com estas espécies nativas. No entanto, a probabilidade de sobreposição de nicho foi maior com a corvina-legítima, indicando uma maior probabilidade de competição por alimento, possivelmente por serem espécies taxonomicamente próximas. A análise química dos otólitos revelou que identicamente à região nativa, a corvinata-real realiza um padrão anual de migrações, usando o estuário (até à zona de água doce) na primavera e no verão, e as áreas costeiras no inverno. Confirma-se, assim, a utilização simultânea do estuário pela corvinata-real, corvina-legitima, robalo e sargo, evidenciando a competição pelo uso do habitat. Em relação aos potenciais impactos desta espécie noutros recursos pesqueiros, a corvinata-real poderá afectar a demografia das suas presas, algumas com valor comercial na região do estuário do Sado (ex: carapau Trachurus trachurus, anchova Engraulis encrasicolus, lula comum Loligo vulgaris, e choco comum Sepia officinalis. Poderá também competir com espécies nativas através da competição por alimento, uma vez que outras espécies também se alimentam das mesmas presas (ex: dourada Sparus aurata, sargo-do-senegal Diplodus bellottii, a anchova Engraulis encrasicolus, o carapau Trahurus trachurus, o choco comum Sepia officinalis, lula comum Loligo vulgaris, polvo comum Octopus vulgaris, golfinho-roaz Tursiops truncatus). Contudo é difícil responsabilizar a corvinata-real por algum impacto, uma vez que outros factores podem prejudicar espécies nativas, como a perda de habitat ou pesca excessiva. Há também a possibilidade de espécies nativas usarem a corvinata-real como alimento, uma questão que deverá ser abordada em estudos futuros com maiores amostragens de espécies nativas. Os resultados dos inquéritos revelaram que esta espécie tem um grande potencial para ser bem aceite no mercado: grande parte dos consumidores demonstrou interesse em adquirir o peixe posteriormente; a maioria prefere peixe selvagem e por isso muitos optariam por adquirir a corvinata-real em vez de peixes nativos provenientes de aquacultura; reconhecem que a espécie está subvalorizada (5€ kg-1) e estão dispostos a pagar em média mais 3€ kg-1 que o preço médio actual. Verificamos também que o principal motivo que levou os consumidores a preferirem outras espécies de peixes nativos (ex: corvina-legítima, dourada, sardinha) é a falta de informação sobre a espécie e o hábito de consumo. Assim, uma opção para contornar este conservacionismo dos consumidores será criar acções de divulgação que os informem sobre os benefícios de consumir uma espécie invasora, destacar a sua proveniência (mar), e promover o seu uso em pratos que melhorem o seu sabor e textura, nomeadamente pratos que envolvam o peixe partido, como caldeiradas ou massadas. As acções de sensibilização criadas neste trabalho (parceria com chefs locais e publicação de comunicados de imprensa) demonstraram que existe interesse na corvinata-real e na temática das espécies invasoras, permitindo assim a promoção deste novo recurso-pesqueiro e a consciencialização dos portugueses sobre o aparecimento de espécies invasoras. Assim, a comercialização da corvinata-real tem potencial para ser optimizada e poderá vir a contribuir para a redução da população no estuário do Sado e minimizar os seus possíveis impactos. Contudo, é importante considerar os riscos e benefícios desta abordagem e incluir acções de monitorização e de divulgação que clarifiquem que os benefícios da sua comercialização não superam os benefícios do controlo da população a longo prazo.
Weakfish Cynoscion regalis, a species native to the NE-coast of North America has established an invasive population in the Sado estuary, and since it is being captured in large numbers in Sado estuary and it is already being sold in fish markets, anglers expressed their concern with competition between weakfish and native prize fish; however, there are no scientific evidences showing that weakfish will outcompete native species for food and space. Therefore, we determined weakfish diet through stomach content analysis, and for three native prize fish: meagre, European bass, and white seabream. Results revealed that weakfish has a generalist feeding strategy and is preying on crustaceans (53%), fish (45%) and cephalopods (2%). We inferred trophic position through isotopic carbon and nitrogen content analysis in muscle and fin tissues, and weakfish, meagre and seabass are feeding at the same trophic level, and seabream a trophic level below. There was no significant isotopic niche overlap between weakfish and the three species, although it was higher for meagre. To infer competition for space, we determined weakfish habitat use through otolith chemistry analysis that revealed an annual pattern of migrations, using the estuary in spring/summer and coastal areas in winter, confirming the simultaneous use of the estuary among weakfish and the three natives. To evaluate the potential of weakfish as a new fishing resource, we provided inquiries to consumers and created awareness events to engage the media and assess public interest on this matter. Inquiries revealed that consumers recognize weakfish potential, since they showed interest in acquiring the fish, would pay more than the current mean price, and would opt to buy weakfish instead of farmed native fish. The feedback from the awareness events was positive, indicating that invasive species subject attracts enough interest to promote weakfish successfully and increase society’s awareness on invasive species.
