O presente trabalho tem por objetivo analisar o processo de decadência da ordem Carmelita em Pernambuco, durante a política regalista do governo imperial, e a sua posterior restauração, a partir de 1894, com a vinda dos carmelitas espanhóis. Nessa perspectiva, procuramos estudar o contexto histórico do Império no qual foram impostas medidas restritivas aos antigos institutos monásticos dos beneditinos, franciscanos e carmelitas e, nesse meio, a conseqüente situação vivenciada pelos carmelitas diante do esvaziamento de seus templos religiosos. Diante desta finalidade, a análise documental referente à decadência do Carmo pernambucano nos possibilitou um maior entendimento acerca da redução da influência que os regulares exerciam na sociedade local e, ao mesmo tempo, serviu como subsídio para que entendêssemos como ocorreu a implementação do projeto de restauração da ordem no Brasil, a partir da proclamação da República. Dessa forma, percebemos que, para restabelecer o vigor da ordem, os frades remanescentes do Recife solicitaram ajuda da Província do Doce Nome de Maria que enviou alguns grupos de religiosos para empreender tal objetivo. Salientamos que a chegada desses religiosos permitiu a reorganização do carmelo pernambucano, assim como contribuiu para consolidar a romanização da Igreja Católica. Sendo assim, na elaboração desta tese, foram utilizados correspondências, atas, ofícios, estatutos e os livros patrimoniais da ordem

Full textDespite the attempts of the Portuguese Crown to restrict the power of the nacional Church throughout the Modern Age, a systematic action was only possible with the new ideas of the 18th century. During the administration of Sebastião José de Carvalho e Melo, Marquis of Pombal, based on Jansenists, Febronianists and Episcopalists theories, the Portuguese Regalism won doctrinal grounds that allowed it greater strength and breadth, reaching its heyday, both before the national Church as the Holy See. Given its opulence, privileges and immunities that made them centrifugal bodies to the desired monarchical centralization and submission of Church, religious orders constituted one of the main targets of the regalists policies then structured. Despite this, except for the Jesuits, Brazilian historiography have dismissed little attention to the Regalism of the Lusitanian Crown and its representatives among the various religious provinces throughout the colonial territory, limited to statements almost always generic. Thus, in this study we analyze precisely the actions of framework of Província de Nossa Senhora do Carmo do Rio de Janeiro, from 1750 to 1808, their successes and limits, steps and countersteps.

José I e seu Premier Sebastião de Carvalho e Mello - O Marquês de Pombal - na segunda metade do século XVIII que culminou, entre várias outras consequências, na contribuição direta para o fechamento prematuro desse tribunal, em 1774, ou seja, 47 anos mais cedo que todos os outros do reino. Por fim, esse trabalho dedicou-se a descortinar essa relação entre o governo, altamente inclinado ao absolutismo, que Sebastião de Carvalho e Mello, o Marquês de Pombal, capitaneara e a Inquisição de Goa, historicamente tratada como o tribunal mais autônome e insubordinado da Inquisição portuguesa. Foram muitas as ações de Pombal sobre o Santo Ofício com o intuito de submete-lo à autoridade real : sua transformação em tribunal régio, sua utilização para punir crimes poliíticos, a substituição de seus inquisidores à revelia da vontade da Igreja, etc.

The seventeenth-century Portugal is a geographic and social space, as well as a laboratory and a bed of intellectual, philosophical, political and religious disputes, aimed at reaching the most advanced nations in that period. The order of the Society of Jesus served the interests of the Council of Trent, since the Catholic world collapsed. Inserted in Portugal they are a decisive instrument for the formation of consciences in consonance with the tridentino dogmas, through a system of education dominated by the scholastic philosophy peripatetic elaborated in Ratio Studiorum. When he realized the Portuguese cultural backwardness and the need for a global reform of Lusitanian society, in the face of a modern and cultured Europe, and in an attempt to reverse the evils caused by Jesuitism in Portugal, the Marquis de Pombal sought to make changes in all the sectors of the Portuguese crown and, mainly, in education. The Oratorians were responsible for the transformation of Portuguese society and it is they who insert Portugal into modernity.
O Portugal setecentista constitui-se no espaço geográfico e social, bem como laboratório e canteiro das disputas intelectuais, filosóficas, políticas e religiosas, que visavam alcançar as nações mais adiantadas naquele período. A Ordem da Companhia de Jesus serviram aos interesses do Concílio de Trento, uma vez que o mundo católico desmoronava-se. Inseridos em Portugal são eles um instrumento decisivo para a formação de consciências em consonância com os dogmas tridentino, através de um sistema de ensino dominado pela filosofia escolástica peripatética elaborada no Ratio Studiorum. Ao perceber o atraso cultural português e a necessidade de uma reforma global da sociedade lusitana, diante de uma Europa moderna e culta e, na tentativa de reverter os males causados pelo jesuitismo em Portugal, O Marquês de Pombal procurou, então, realizar mudanças em todos os setores da coroa portuguesa e, principalmente, a na educação. Coube aos oratorianos a responsabilidade pela transformação da sociedade portuguesa e são eles que inserem Portugal na modernidade.

Full textThe study of the representation of power in modern times has developed significantly in recent years under the guidance of art historians and experts in semiotics. However, these studies have often excluded from their research the fundamentally important geographical space that is the Mediterranean world. Therefore, this thesis revolves around the following questions: Was the portraiture, and in particular the state portrait, a political statement of the power and legitimacy of the person represented in the painting? Is the court portrait a reflection of the conception of the power of the princes and the main intellectual movements of the modern era? After placing these paintings in their historical context, this study conducts a content analysis of the representations of power, focusing on recurrent themes, frequency of occurrence, and the placement of the image, and also to the connotation each specific cultural references, and meanings whether hidden or evident. Some Mediterranean principalities have indeed used the pictorial and political principles of the Counter-Reformation to assert not only the superiority of the Catholic Church, but also their own power. After the period of rediscovery of portraiture as an art form during the Renaissance, the Counter-Reformation and the seventeenth century are seen as the use of visual communication to oppose the written word. On the contrary, in the eighteenth century, the influence of the Enlightenment and philosophical writings permeates the state portraits. While there was a great deal of stability in portraiture over time, gradually the background decor and elements of the clothing began to reflect the arrival of new ideas, both political and social, within the spheres of power and in society more generally

Full textThe research begins from the years of one’s youth, lived by Giancarlo Brasca in the contest of Azione Cattolica’s young lines in Milan. First chapter proposes to reconstruct which atmosphere and which personalities influenced young Brasca’s choices. On this point of view particular attenction was dedicated to relation with Father Agostino Gemelli and Monsignor Francesco Olgiati. The intensity of relationship with two protagonists of catholic world in Milan represents a significant fact also in vocational choice that binded Giancarlo Brasca with Missionari della regalità di Nostro Signore Gesù Cristo, secular institution founded by Gemelli. So second chapter of dissertation proposes to reconstruct the meaning of Brasca’s adhesion to Missionari’s life proposal, his contribution to story of institution and to development of relation between this and Catholic University. Brasca’s vocational choice represents indeed a necessary keynote to understand and reconstruct Brasca’s story in Catholic University from ’40 to his death in 1979. So third and fourth chapters reconstruct Brasca’s events on the contest of Catholic University: from Philosophy degree moving to experience which director of Library, to arrive to very important administrative assignments like secretary of administration and, from 1971, administrative director.

Full textThe object of this research is the whole of the celebrations and preparations connected with the passage of Maria Theresa of Austria and the solemn entry of Archbishop Stampa into Milan in 1739, both exemplifications of the 18th century first half civic rituality and public ceremonials, and representations which engaged the city. A confrontation has also been made with the possesso of Maria Theresa, enacted in absentia in 1741. By means of a comparative analysis of the primary sources – printed, hamdwritten and ichnographic as well – it has been possible to reconstruct the role of etiquette norms, ceremonials and of the entire dramaturgy of festival events. The research has also brought to light the apparati set up by the Collegio dei Giureconsulti, so far ignored by studies. It has highlighted rhetorical mechanisms, actions, representational aspects which remain, some of them pertaining to the archetype sphere. Even if simplified, the ceremonials still show a strong cult of regality. The triumphal entry proves to be a meaningful moment to bring the city back to the imaginary of the origins which constitutes its identity and to renovate a new era. The necessity of the feast, of ritualistic moments of representation and auto representation is pointed out; and also the dramaturgic and not ornamental role of decorative elements like the baldachin, the arch, the carriage, the portrait, in a culture which conferred great value to images.

Full textO presente trabalho dedica-se ao estudo da Mesa da Consciência e Ordens no Brasil, tribunal criado por Dom João VI, por meio do Alvará de 22 de abril de 1808, como parte da política de reprodução do aparato administrativo lisboeta no Rio de Janeiro, em virtude da transferência da corte portuguesa para o Brasil. A primeira parte deste trabalho foi dedicada à análise da Mesa da Consciência em Portugal, criada em dezembro de 1532 por Dom João III. No intuito de compreender um tribunal que incorporava funções como aconselhar a consciência do monarca, conhecer questões administrativas envolvendo a gestão de Ordens Militares Religiosas, além de diversas outras matérias, foram examinados os comandos legais expedidos com a finalidade de regulamentar a organização, funcionamento e competência do tribunal português. A segunda parte deste trabalho foi dedicada ao estudo da Mesa da Consciência e Ordens no Brasil. Com o objetivo de estudar a atuação da corte brasileira, foram analisadas as consultas proferidas pelo tribunal. A íntegra da transcrição das consultas analisadas, bem como a cópia digitalizada dos manuscritos consultados foram incluídas nos anexos a este trabalho. Por último, foi analisado o processo que levou ao encerramento das atividades da Mesa no Brasil em 1828. O objetivo deste trabalho foi o de investigar o funcionamento do Tribunal da Mesa da Consciência e Ordens no Brasil entre 1808 a 1828, com a finalidade de mellhor compreender, sob a ótica da história do direito e com lastro nas fontes jurídicas examinadas, a gênese da Mesa portuguesa, as regras que disciplinavam o seu funcionamento e competência, o modo pelo qual tal tribunal foi transferido e instalado no Rio de Janeiro, suas decisões, as causas de sua extinção e o destino de seu acervo documental.
The present work is dedicated to the study of the Mesa da Consciência e Ordens in Brazil, court created by Dom João VI through the Alvará of April 22, 1808, as part of the policy of reproduction of the administrative apparatus of Lisbon in Rio de Janeiro, due to the transfer of the Portuguese court to Brazil. The first part of this work was dedicated to the analysis of the Mesa da Consciência e Ordens in Portugal on December of 1532 by Dom João III. In order to understand a court that included functions such as advising the monarch's conscience, to know administrative matters involving the management of Military Religious Orders, besides several other matters, were analyzed the legal rules issued with the purpose of regulating the organization, functioning and competence of the Portuguese court. The second part of this work was dedicated to the study of the Mesa da Consciência e Ordens in Brazil. In order to study the Brazilian court's performance, the court's consultations were analyzed. The complete transcription of the consultations analyzed, as well as the digital copies of the manuscripts consulted were included in the annexes to this work. Finally, the process that led to the closure of Mesa's activities in Brazil in 1828 was analyzed. The objective of this work was to investigate the functioning of the Tribunal of the Mesa da Consciência e Ordens in Brazil between 1808 and 1828, in order to better understand, from the point of view of the history of law and according to the legal sources examined, the genesis of the portuguese Mesa, the legal rules issued with the purpose the organization rules issued with the purpose of regulate its organization and competence, the manner in which such court was transferred and installed in Rio de Janeiro, its decisions, the causes of its extinction and the destination of its documentary collection.

Polychlorinated biphenyls (PCBs) were first synthesized in 1881 by Schmidt and Schulz and their commercial production began in 1929. In 1966, PCBs were first identified as pollutants and subsequent investigations showed that they are ubiquitous. Many studies have even proven that PCBs are hazardous to human and other animals. Xylaria regalis is a wood-grown saprophytic fungus that is capable of PCBs degradation. After the comparison of five culture media, we found that Xylaria regalis grew in SDB medium could degrade more PCBs than in other media. Mycelium extract from 10-day growth mycelium could degrade 70% of Aroclor 1254 and culture medium from 10-day growth mycelium could degrade 67% of Aroclor 1254. Therefore, we chose SDB for the growth of Xylaria regalis and used mycelium extract for the investigation of PCBs degradation by Xylaria regalis. The results showed that the optimal reaction temperature for mycelium extract was 30 ℃. After treating mycelium extract for 4 hours at 30 ℃, 85% of Aroclor 1254 was degraded. The optimal reaction pH value for mycelium extract was pH 7. After treating mycelium extract for 2 hours at pH 7, 74% of Aroclor 1254 was degraded. Mycelium extract had the best temperature stability at 30 ℃, and it could degrade 43% of Aroclor 1254 even treated at 50 ℃ for 4 hours. At pH 7, mycelium extract had the best pH stability, and after treating at pH 3 and pH 10 for 4 hours, it could still degrade 45% and 30% of Aroclor 1254, respectively. We also tested the ability of mycelium extract for degradation of three commercial PCBs. The results showed that mycelium extract could degrade 69% of Aroclor 1242, 76% of Aroclor 1254, and 48% of Aroclor 1260, respectively. When Aroclor 1254 was added into soil to simulate the condition of soil with PCBs pollutants, we found that 78% of Aroclor 1254 in soil was degraded after the soil treated with mycelium extract for 2 hours at 30 ℃. Mycelium extract was purified by TSK gel Phenyl-Toyopearl 650 M column and there were four activity peaks in three eluents of different ammonium sulfate concentrations. After analyzing four activity peaks by SDS-PAGE, we suggested that there was more than one kind of PCBs degrading enzyme in mycelium extract.

Polycyclic aromatic hydrocarbons (PAHs) are mainly formed as products of the incomplete combustion of fossil fuels and organic compounds. They consist of benzene homologs which have two or more fused aromatic rings. As the molecular weight of PAHs increases, its toxicity and hydrophobicity increase, which indicates that it is hard to dispose PAHs in the environment. In our laboratory, we find that the enzyme of the Xylaria regalis has the ability to degrade PAHs. In this study, the fungus Xylaria regalis was incubated in the SDB medium. Mycelia of Xylaria regalis were harvested by filtration and broke by pestle. Then the crude extract was boiled in the 100℃ water and purified by ionic exchangers (CM, QAE), hydrophobic interaction (phenyl 650M) and gel filtration (HW-55, Bio-Sil SEC-400 HPLC) column. The purity of the PAHs-degrading enzyme that degraded BaP and Pyr was increased 183 and 196 fold and its specific activity was reached to 120 and 477 unit × 10-4 /mg, and the recovery was 62 and 67%, respectively. The optimal pH and pH stability for PAHs-degrading enzyme activity was 8.5. The PAHs-degrading enzyme was heat stable protein, because it could maintain the enzyme activity after treated at 100℃ for 1hr. Mg++、Sr++、Ba++、Ni++、Mn++ ions inhibited the enzyme activity. The PAHs-degrading enzyme had a molecular weight of 29 kDa as determined by HPLC gel filtration Bio-Sil SEC-400. By SDS-PAGE electropheresis, single protein band, its molecular weight about 29 kDa, was also found. Therefore, the result suggested that the PAHs-degrading enzyme was monomer.

It is commonly argued that it was the political ideology of Carvalho e Melo that made his ecclesiastical policy, but in this work, inserting Pombal into the Catholic Enlightenment current, we propose that it was his cultural and religious sensibility that influenced the Pombaline reforms in general and ecclesiastical reforms in particular.

Abstract Cellulose is a linear polysaccharide of glucose residues connected by β-1,4- glucosidic linkages. Native crystalline cellulose is insoluble and occurs as fibers of densely packed, hydrogen bonded chains of 15 to 10,000 glucose units. Its density and complexity make it very resistant to hydrolysis without chemical or mechanical degradation. As a result, utilization of cellulase is the most important step during energy conversion from cellulosic biomass. A cellulosic enzyme system consists of three major components: endo-β- glucanase, exo-β-glucanase andβ-glucosidase. Endo-β-glucanase makes random scission of cellulose chains yielding glucose and cello-oligo saccharides. Exo-β- glucanase commits exo-attack on the non-reducing end of cellulose with cellobiose as the primary structure. β-glucosidase hydrolyzes cellobiose to glucose. These three enzymes act synergistically to degrade cellulose. In this study, we isolate, purify and characterize endo-β-glucanase by Ascomycete Xylaria regalis. The cellulose-containing medium is used to induce the production of the cellulose-degrading enzymes. When these enzymes are produced at maximum level, we harvest and filtrate the culture medium. The filtrate is purified by anion exchanger column (QAE) and hydrophobic interaction column (phenyl-650M). The purification fold of endo-β- glucanase increases 4.3 times, the specific activity approaches 3.7 unit/mg, and the recovery is 27.6%, respectively. SDS-PAGE shows that the molecular weight of endo-β-glucanase is 47 kDa. The optimal pH of the enzyme is 5 and the optimal temperature of the enzyme is 50℃. The pH stability of enzyme is between 4 and 5. The thermostability of enzyme is 40℃. Some metal ions, like Mg2+, Zn2+, Cu2+, Fe3+, Hg2+ and Ba2+, slightly enhance the enzyme activity; however, Co2+ ion suppresses it.

In this study, the xylanase and β- xylosidase secreted by Xylaria regalis were isolated , purified and characterized. We used the avicel and oat spelts xylan as the only carbon source to induce the production of xylan-degrading enzyme by this fungus. The fungus was grown in a rotary shaker at 25℃ for 26 days. And the culture filtrate was collected and further purified by anion exchanger column (QAE Sephadex A 25), hydrophobic interaction column (phenyl 650M) and gel filtration column (Sephadex G 75), respectively. The specific activity of the finally purified xylanase was 1.30x103 unit /mg and its purity was increased 11 fold, and the recovery was 34.6%. The molecular weight of xylanase estimated by SDS-PAGE was approximately 30.8 kDa. The optimal pH and temperature for this enzyme was pH 5.5 and 50℃. The xylanase was stable above pH 5 at low temperature. When the enzyme treated with distinct metal ions, the activity was significantly inhibited by Cu2+and Hg2+. Xylanase activity was also inhibited by EDTA or β-mercaptoethanol. The results show that the active site of xylanase may have disulfide bond which can stabilize the xylanase third structure and certain metal ions may be the cofactor of the enzyme. We collected the culture filtrate for the purification of β-xylosidase. Then culture filtrate was precipitated by 80% ammonium sulphate and further purified by hydrophobic interaction column (phenyl 650M), anion exchanger column (QAE) and gel filtration column (Sephadex G 75), respectively. The specific activity of the finally purified β-xylosidase was 4.00x10-1 unit /mg. The recovery of the enzyme was 24.2% and the purity was increased 22.97 fold. The molecular weight of β-xylosidase estimated by SDS-PAGE was approximately 45 kDa . The optimal pH and temperature for β-xylosidase was pH 5.5 at 50℃. β-xylosidase was stable above pH 5 at low temperature. Most of the tested metal ions have no remarkable effects on the activity of the β-xylosidase, whereas the Fe2+and Hg2+ significantly inhibit it. In addition, the activity of the β-xylosidase was not affected by EDTA or β-mercaptoethanol.

PCBs (Polychlorinated Biphenyls) are widespread pollutants. Many studies have proven that PCBs are hazardous to humans and other animals. Xylaria regalis is a wood-grown saprophytic fungus. Based on previous study, Xylaria regalis is capable of PCBs degradation, and the fungus could degrade more PCBs in SDB medium than in other media. So, We cultured Xylaria regalis in SDB medium and further purified the PCBs-degrading enzyme. We found that culture medium from 10-day growth mycelia could degrade 67 % of Aroclor 1254 (PCBs). And the PCBs-degrading enzyme was further purified by anion exchanger column (QAE Sephadex A-25) and hydrophobic interaction column (Phenyl-650M). The purity of the PCBs-degrading enzyme was increased to 11.60 fold, the specific activity of the PCBs-degrading enzyme was reached to 8.90 x 10-2 unit/mg, and the recovery of the purified enzyme was 108.29 %. The molecular weight of the PCBs-degrading enzyme was found to be approximately 22.5 kDa by HPLC gel filtration and SDS-PAGE. The optimal pH of the enzyme was 7 and the optimal temperature of the enzyme was 30 ℃. The pH stability of the enzyme was 7 and the thermostability of the enzyme was 30 ℃. The iron ions that we studied in different concentration can inhibit the enzyme activity. When EDTA was added in the enzyme solution, the enzyme activity was inhibited.

In recently years, we have make many pollutions in our environment, such as polycyclic aromatic hydrocarbons (PAHs) are mainly formed as the products of the incomplete combustion of fossil fuels and organic compounds. These pollutants consist of benzene homologs which have two or more fused aromatic rings, and most of those are more poison and hydrophobic with the number of ring increased. When their hydrophobic property is increased, it is hard to degrad them in the environment. In this study, we isolate, purify and characterize the polycyclic aromatic hydrocarbons-degrading enzymes by Xylaria regalis. The SDB medium was used to culture the fungus for the production of the PAHs-degrading enzyme. When the enzyme was produced at maximum level, the culture medium was harvested and then filtrated. The filtrate was precipitated by 80% ammonium sulphate and furhter purified by hydrophobic interaction column (phenyl 650M), anion exchanger column (QAE), gel filtration column (HW-55). The purity of benzo(a)pyrene-degrading enzyme was increased 205 fold and its specific activity reached to 9.1636 unit/mg, and the recovery of the purified enzyme was 19.47%. The molecular weight of enzyme was found to be approximately 48 kDa by HPLC gel filtration and confirmed by SDS-PAGE. The optimal pH of the enzyme was 7 and the optimal temperature of the enzyme was 30℃. The pH stability of enzyme was 8 and the temperature stability of enzyme was 30℃. Bap-degrading enzyme catalyzed the oxidation of veratryl alcohol to veratraldehyde without H2O2 and the oxidation of Mn2+ by H2O2 to Mn3+.The metal ions that we studied in different concentration can enhance enzyme activity. When EDTA and β-mercaptoethanol was added in the enzyme solution, the enzyme activity was not inhibited.

Full textWeakfish Cynoscion regalis, a species native to the NE-coast of North America has established an invasive population in the Sado estuary, and since it is being captured in large numbers in Sado estuary and it is already being sold in fish markets, anglers expressed their concern with competition between weakfish and native prize fish; however, there are no scientific evidences showing that weakfish will outcompete native species for food and space. Therefore, we determined weakfish diet through stomach content analysis, and for three native prize fish: meagre, European bass, and white seabream. Results revealed that weakfish has a generalist feeding strategy and is preying on crustaceans (53%), fish (45%) and cephalopods (2%). We inferred trophic position through isotopic carbon and nitrogen content analysis in muscle and fin tissues, and weakfish, meagre and seabass are feeding at the same trophic level, and seabream a trophic level below. There was no significant isotopic niche overlap between weakfish and the three species, although it was higher for meagre. To infer competition for space, we determined weakfish habitat use through otolith chemistry analysis that revealed an annual pattern of migrations, using the estuary in spring/summer and coastal areas in winter, confirming the simultaneous use of the estuary among weakfish and the three natives. To evaluate the potential of weakfish as a new fishing resource, we provided inquiries to consumers and created awareness events to engage the media and assess public interest on this matter. Inquiries revealed that consumers recognize weakfish potential, since they showed interest in acquiring the fish, would pay more than the current mean price, and would opt to buy weakfish instead of farmed native fish. The feedback from the awareness events was positive, indicating that invasive species subject attracts enough interest to promote weakfish successfully and increase society’s awareness on invasive species.
