Abstract:
The integrity of the genome relies on the maintenance of chromosomes, the structural embodiment of the genetic material. Disruption of chromosome replication can lead to extensive genomic rearrangements, spanning kilobase (Kb) to megabase (Mb) regions. Some chromosome rearrangements are inherently dynamic, beginning as a single unstable rearrangement from which multiple rearrangements emerge. The rare formation and transient behavior of unstable chromosomes renders their study challenging. Here I characterize the genetic ontogeny of unstable chromosomes in a budding yeast model, from initial r

Abstract:
Eukaryotic DNA replication is composed of a complex array of molecular biological activities compounded by the pressure for faithful replication in order to maintain genetic and genomic integrity. The constraints governing DNA replication biology is of fundamental importance to understand the degree of replication error and strategies employed by organisms to tackle the threats to replication fidelity from such errors. We apply a simple conceptual model, formalized by the use of probability theory and statistics, to discern fundamental pressures and constraints that optimise complete DNA repli

Abstract:
Experimental psychology is said to be having a reproducibility crisis, marked by a low rate of successful replication. Researchers attempting to respond to the problem lack a framework for consistently interpreting the results of statistical tests, as well as standards for judging the outcomes of replication studies. In this paper I introduce an error-statistical framework for addressing these issues. I demonstrate how the severity requirement (and the associated severity construal of test results) can be used to avoid fallacious inferences that are complicit in the perpetuation of unreliable

Abstract:
The success of screening experiments hinges on the effect sparsity assumption, which states that only a few of the factorial effects of interest actually have an impact on the system being investigated. The development of a screening methodology to harness this assumption requires careful consideration of the strengths and weaknesses of a proposed experimental design in addition to the ability of an analysis procedure to properly detect the major influences on the response. However, for the most part, screening designs and their complementing analysis procedures have been proposed separately i

The error statistical account provides a basic account of evidence and inference. Formally, the approach is a re-interpretation of standard frequentist (Fisherian, Neyman-Pearson) statistics. Informally, it gives an account of inductive inference based on arguing from error, an analog of frequentist statistics, which keeps the concept of error probabilities central to the evaluation of inferences and evidence. Error statistical work at present tends to remain distinct from other approaches of naturalism and social epistemology in philosophy of science and, more generally, Science and Technolog
