Aging is characterized by a stochastic accumulation of molecular damage, progressive failure of maintenance and repair, and consequent onset of age-related diseases. Applying hormesis in aging research and therapy is based on the principle of stimulation of maintenance and repair pathways by repeated exposure to mild stress. In a series of experimental studies we have shown that repetitive mild heat stress has anti-aging hormetic effects on growth and various other cellular and biochemical characteristics of human skin fibroblasts undergoing aging in vitro. These effects include the maintenanc

DNA Helicase B (HELB) is a conserved helicase in higher eukaryotes with roles in the initiation of DNA replication and in the DNA damage and replication stress responses. HELB also inhibits homologous recombination by reducing long-range end resection. After phosphorylation by cyclin-dependent kinase 2 (CDK2) at the G1 to S transition, HELB is pr

Abstract:
Telomeres are regions of repetitive DNA bound with a set of specialized proteins required to protect chromosomes from fusing with each other and from eliciting DNA damage response. Dysfunctional telomere maintenance can lead to premature cellular senescence, premature organismal aging and cancer predisposition. In the last few years the evidence has emerged indicating a link between dysfunctional maintenance of telomeres and defective DNA damage response. The objective of this project was to explore further this link by examining effects of some DNA damage response proteins on telomeres that h

New environmental regulations as well as the increasing industrial competitiveness have set new more demanding rules on the manufacturing industry. In order to abide by those rules not only from the legal point of view but also to be able survive, manufacturing has to be more sustainable from many aspects, especially the economical one. One way to achieve the previous target is an unfortunately often oversighted aspect of the industry sector, the maintenance strategy. Telomeres are physical ends of chromosomes consisting of (TTAGGG)n DNA sequence and a specialized set of proteins that protect chromosomal ends from degradation and from eliciting DNA damage response. These specialized set of proteins, known as shelterin, directly bind to telomeric DNA. In addition, some DNA double-strand break (DSB) repair proteins such as, DNA-PKcs and KU70/80, play active roles in telomere maintenance. Mouse knock-out experiments have revealed that deletion of either DNA-PKcs or Ku70/80 resulted in elevated levels of telomeric fusion, indicative of dysfunctional telomeres.

In the late 1980s and early 1990s few transportation agencies around the world considered performance-based specifications as an alternative to improve the efficiency of the services provided to the public. These initiatives are better known as Performance-Based Road Maintenance (PBRM). PBRM calls for performance-based work, in which a desired outcome is specified rather than a material or method. This type of specification promises to be an excellent tool to improve government efficiency in maintaining transportation networks; however, without proper monitoring, it could likely yield adver
