Retinol (vitamin A) and its active metabolite, all-trans retinoic acid, signal through nuclear retinoic acid and retinoid X receptor (RAR/RXR) heterodimers. These complexes regulate the expression of genes involved in developmental processes such as limb development. In excess, retinoids are potent teratogens and cause marked reductive effects on the developing limb. The goal of this thesis was to elucidate the molecular mechanisms underlying retinoid-induced limb dysmorphogenesis. Retinoic Acid Metabolism Blocking Agents (RAMBAs) increase the endogenous levels of all-trans retinoic acid (RA) by inhibiting CYP26 enzymes. Retinoic acid (RA), the major biologically active form of vitamin A, is required for normal development of the mouse embryo. In particular, RA is necessary for the correct anteroposterior specification of the embryonic axis. Exposure to RA at certain stages of development leads to premature truncation of the vertebral column, accompanied by spina bifida. Vitamin A metabolites and retinoids may slow the progression of breast cancer and elicit anti-neoplastic properties similar to those of omega-3 fatty acids. Studies using animal models show a decrease in the incidence, growth and metastisis of mammary tumors in the presence of specific fatty acids. La peau des vertebres forme des annexes tegumentaires caracteristiques de chaque region du corps. L'apport ectopique d'acide retinoique tout-trans (ar-tt) provoque deux types de changements homeotiques de morphogenese (formation d'un type d'annexe en lieu et place d'un autre) : la formation de plumes sur les ecailles de la face anterieure du tarsometatarse chez le poulet et la formation de glandes en lieu et place des follicules pileux de vibrisse chez la souris.
