Full textThe study relates to adjustments of expenditures, by budget components, when households are facing an important fall of income. It begins with a presentation, in a methodological perspective, of the most important empirical studies on the subject. It goes on with a synthesis of the principal results arising from studies which relate to unemployed and presents the results obtained by the author from two surveys : "conditions de vie" from INSEE (1986) and "enquête socio-économique auprès des ménages lorrains" (1986). Eight components of household’s budget are considered: clothing, leisure, transport, home equipment, food, housing, health, expenditures for children. For each of this components are examined: its place in the hierarchy of outlays adjustments, the ways by which expenditures are reduced, the dynamic of adjustments, the relation between diminishing expenditures and different characteristics of households. For this last point, two complementary approaches are used: a descriptive one and an econometric one. The study points out the necessity of studying thoroughly the dynamic of the ways by which expenditures are reduced

Recent developments in analytical philosophy and normative economics are giving some ways to better understand the problem of inequality and the policies to solve it. In this thesis, the post-welfarist approaches (answering to the question: "equality of what?") and their scientifical and ethical interests are particularly studied. Taking the question of minimum guaranteed income and studying the relevance of social justice as impartiality theories, we are trying to analyses the way the national minimum guaranteed income schemes (particularly in France but also in northern Europe) are meeting social justice or local justice principles. The public goods features are also analyzed. At last, the impact of such schemes, in terms of standard of living and well-being, is analyzed

Full textWoodroll® är en förgasningateknik som utvecklats av Cortus som producerar syntesgas (syngas) från biomassa. Det finns en rad olika användningsområden för gasen. Ett intressant sådant är att omvandla den till biometan, vilket kan användas som drivmedel eller ersätta naturgas i gasnät. Dock så är intäktern och lönsamheten starkt beroende av stödsystem. Dessa subventioner kan antingen vara i form av en inmatningstariff, där biometanproducenten får en fast peng för biometanproduktion, eller i form av indirekt stöd där konsumtion snarare än produktion stimuleras. I detta arbete har det utretts vilka intäkter som kan förväntas för biometan-produktion genom Woodroll® i Sverige, Nederländerna, Tyskland, Storbritanninen, Frankrike och Italien, både i termer av belopp och risk. En lönsamhetsanalys har också gjorts för att preliminärt jämföra avkastningen för de olika länderna. Två olika scenarier för olika intäkter har analyserats för två olika råvarupriser. Resultatet visade att samtliga länder kan erbjuda attraktiv avkastning. Dock är samtliga fall förknippade med osäkerhetsfaktorer. Sverige och Tyskland erbjuder indirekt stöd och negativa marknadstrender. Nederländerna och Storbritannien är de enda alternativen som ger en feed-in-tariff för biometanproduktion. I Nederländerna kan tariffen säkras innan investeringsbeslut fattats men betalas endast i 12 år. Storbritannien ger en fast tariff i 20 år men kan justeras kvarstalsvis och nivån säkras först efter uppstart av anläggningen. Tariffen har reducerats med 40 % de senaste 24 månaderna, men det pågår diskussioner om att introducera en särskild tariff för förgasning. Italien erbjuder stödsystemet som ger högst potentiella intäkter men biometan från förgasning är inte berättigat för stödet. Det senare gäller också för Frankrike som kan bli ett intressant fall i framtiden. Om det önskas att minimera risken så bör Cortus fokusera antingen på Nederländerna eller invänta diskussionerna om förgasningstariffen i Storbritannien och Frankrike. Arbetet kring standardisering av biometan-användning bör också följas eftersom Italien erbjuder de potentiellt högsta intäkterna. Det rekommenderas också att Cortus tittar vidare på andra alternativ. Det bästa fallet för den risk-aversiva är fallet som ger en fast tariff i 20 år och där stödet kan säkras innan investeringsbeslut fattas.

Full textThis thesis is devoted to the analysis of subjective well-being, with a special focus on the geographical aspects. Two questions are asked : to what extent do the measures of subjective well-being provide a good proxy for utility ? And more specifically, what can they tell us on the determinants of well-being ? The first chapter focuses on peer effects and income comparisons. The analysis reveals that neighbors' median income has a very significant impact on respondents' well-being in the United States - meaning that well-being is relative. The second chapter further analyses the impact of a local reform on residents' well-being in Switzerland. The results show that political, administrative and fiscal centralization at the regional level reduces significantly people's lift satisfaction. These two first chapters also deal with the impact of local public goods on well-being. The third chapter of this thesis takes a broader perspective and shows that the increase in public spending observed since the 60s in most developed countries, has had significant distributional effects in terms of well-being. Finally, the fourth chapter focuses on one dimension of well-being : health. It shows that health predicts life satisfaction belter than income or unemployment do

Full textAs competitiveness increases and environmental legislation toughens, companies can no longer solely rely on traditional supply chain OEM to remain competitive. Indeed for some companies such as copy machine or computer manufacturers, a unique value creation chain is no longer sufficient. These companies have created, or are trying to create, a second valuecreation chain, the reverse supply chain. This organizational innovation is mainly based on creating value through remanufacturing of end of the life products. The benefits of a reverse supply chain clearly emerge in academia and industrial test studies. Yet the number of companies that have been able to create and sustain a viable reverse supply chain is rather limited.This dissertation has been conducted under a multidisciplinary research project. The goal of the project was to create a methodology to facilitate the development of remanufacturing systems based on an intervention research conducted with a company seeking to promote the remanufacturing of its end of life products. This intervention also allowed us, beside to ourcontribution to the development of the remanufacturing methodology, to look into the strategic factors which influence the emergence of a reverse supply chain

Full textAIDS is an infection transmitted mainly in the genital mucosa. It is at this level that an effective barrier should be developed to block infection from the initial entry of HIV-1 in the genitals, before establishment of mucosal reservoirs. IgA is the main protective antibody at the genital mucosa and has been suggested to be protective in vivo against HIV-1. Hence, mucosal gp41-specific IgA with strong cross-clade antiviral activity are a major protective correlate in Highly HIV- Exposed individuals that remain SeroNegative (ESN) despite unprotected sexual intercourse with infected partners. However, genital mucosa are also protected by IgG whose origin and role are currently reevaluated. In this work, we have analyzed the influence of the antibody isotype, IgA or IgG, on the epitope specificity, antiviral functions of HIV-1 protective antibodies and evaluated new antiviral functions mediated by the constant IgA regions. For this purpose we have used a library of protective Fab IgA derived from ESN women that are specific for conserved regions of the HIV-1 envelope gp41 subunit, already characterized in the laboratory (Tudor et al., Mucosal Immunol, 2009). By genetic engineering, we have first transformed two of these Fab IgA (FabA) into their corresponding Fab IgG (FabG), having the same paratopes but differing from each other by their CH1 domain (alpha or gamma). These two FabA/FabG couples were then analyzed comparatively for their epitope specificity and functional activities. We have shown that the isotype and particularly the CH1 domain influences (i) the affinity of Fabs for their antigens (gp41 and P1 of clades A, B and C) using surface plasmon resonance and ELISA : FabA have a 50- to 100-fold higher affinity than corresponding FabG ; (ii) antiviral functions: FabA more effectively neutralize CD4+ T-cells infection and transfer of HIV-1 from Langerhans cells to CD4+ T-cells than their corresponding FabG. In a second step, taking as experimental model the neutralizing HIV-1 antibody 2F5 as IgA, we demonstrated that IgA specific for gp41 were able to lyse HIV-1 infected cells in a process mediated by their Fc alpha domain, namely antibody mediated cell cytoxicity (ADCC), a function usually attributed to IgG. Furthermore, anti-gp41 IgA and IgG work in synergy to increase HIV-1-infected cell lysis. Altogether, the results show the isotype (the CH1 domain) has an influence on the recognition and affinity for the antigen, but also on the quality of the secondary immune response (antiviral functions). Finally, we characterized epitopes specific from the two FabA/FabG couples using biopanning followed by in silico analysis. Cross clade 3-dimensional epitopes were mapped on gp41 from Clade A, B and C, each in pre- or post-fusion conformation. These analyses allowed to define cross clade 3-dimensional epitopes targeted by these protective antibodies derived from ESN subjects. These epitopes could serve as immunogen in a vaccine strategy aiming at recapitulating the protective ESN IgA response. Altogether, we have used the strategy of Reverse Vaccinology 2.0 to characterize novel immunogens based on 3D-epitopes targeted by mucosal antibodies from individuals resisting HIV infection. These results should be taken into account in the design of an effective vaccine against the active clades of HIV-1 in the world (A, B and C). Such mucosal vaccine approach is necessary to decrease the number of new HIV infections among women (the most affected) but also in men, which remains a huge public health challenge

This thesis entitled ``Standard of living of the older people in Morocco: A construction of an equivalence scale with elderly'' aims to analyze deeply the question of standards of living of older people in Morocco, by estimating an appropriate equivalence scale to this country. Full textThe human immunodeficiency virus type 1 (HIV-1) is a retrovirus responsible of a disease currently affecting 34 million people and caused the death of 25 million people from its discovery in the 1980s. Despite the great progresses made in the development of new drugs to block the replication of the virus, the rapid emergence of resistant mutant strains requires the development of new therapeutic strategies to combat this agent.The Reverse transcriptase (RT) plays an essential role in the replication of human immunodeficiency virus type 1 (HIV-1) and remains a primary target of anti-HIV-1 drugs. The biologically active form of HIV-1 RT is a heterodimer of two subunits, p51 and p66, each consisting of distinct sub-domains: the fingers, the palm, the connection and the thumb are present in both of them. p66 contain also an RNAse H sub-domains. The formation of RT is a two-step mechanism, involving first the rapid association of the two subunits (Dimerization), followed by conformational changes (Maturation) yielding the biologically active form of the enzyme.In order to inhibit the function of RT, we have proposed and elaborated a new strategy based on short interfacial peptides that target protein-protein interfaces involved in RT- dimerization and activation. The dimerization of two subunits involves interaction between their connection domains. In the first part of this thesis, we screened and isolated peptides capable of interfering with the interaction, involving a "cluster" of aromatic residues. We identified a short peptide PID4, and demonstrated that induces a conformational change in the RT, resulting in dissociation of the heterodimer complex. Maturation involves the interaction of residues located at "hot spot" in the RNase H domain of p66 and the thumb domain of p51. The second part of this thesis is based on a screening of peptides, derived from the thumb domain of the enzyme. We have identified a short peptide (P27) which inhibits the maturation step and abolish replication of HIV-1 LAI. These two classes of peptides block viral replication and are effective in several viral strains and mutants resistant to NNRTIs and NRTIs.Taken together, the results of our two classes of peptides inhibitors, demonstrate that the dimerization and maturation of RT constitutes an attractive target for AIDS chemotherapeutics and for the design of more specific new antiviral drugs that can bypass resistance limitation

Full textThe rapid emergence of drug-resistant viruses against all approved HIV clinical drugs together with inaccessible latent virus reservoirs and side effects of currently used compounds have limited the potency of existing anti-HIV-1 therapeutics. Therefore, there is a critical need for new safer drugs, active against resistant viral strains. Reverse transcriptase (RT) plays an essential role in the replication of human immunodeficiency virus type 1 (HIV-1) and remains a primary target of anti-HIV-1 drugs. To develop specific HIV inhibitors, we have elaborated a new strategy based on short antibody fragments derived from the unique Heavy-chain antibodies present in Camelidae called Nanobodies that targets RT-activation. The immunization of dromedaries with RT has lead to the isolation of a panel of Nanobodies that tightly bind the two subunits of RT and inhibit its DNA-dependent DNA polymerase activity at nanomolar range. From that screen we have elaborated an intrabody (cell penetrating anti-RT Nanobody) NbRT20 that constitutes a potential interesting anti-HIV compound.We demonstrated that NbRT20 inhibits RT polymerase activity and exhibiting a potent antiviral activity with a subnanomolar IC50. NbRT20 binds the thumb subdomain and restricts its flexibility and mobility resulting in an inactive/non processive dimeric conformation of the enzyme. From a mechanistic point of view, we have showed that NbRT20 is a conformational inhibitor. it prevents proper binding of primer/template and of dNTP and destabilizes the enzyme in an inactive/non processive dimeric conformation.Taken together, these results demonstrated that, the Nanobody platform may be highly effective at generating extremely potent and selective intrabody to neutralize RT and HIV proliferation.Key words: HIV-1, RT, Nanobodies, cell penetrating peptide
