BackgroundDrug transporters and metabolizing enzymes located in the epithelial lining of the intestine limit or enhance systemic drug exposure. During child development, the abundance and activity of these transporters and enzymes - determining how fast and how much of a drug is being absorbed into the circulation - changes from birth to adolescence. As most drugs given to children are taken by mouth, the aim of this project is to study the abundance and activity of transporters and metabolizing enzymes, involved in the intestinal absorption of drugs.MethodsThe ex vivo Ussing chamber with pediatric small intestinal tissue is applied to evaluate intestinal drug absorption and metabolism.1 Transport and metabolism of a selection of drug molecules is assessed across these tissues by sampling the donor and receiver compartment at different intervals and sample analysis by LC-MS/MS. Viability, functionality and integrity of the tissues are monitored using electrophysiological parameters (dP, R, I). Ussing chamber experiments are combined with a targeted proteomics approach to quantify drug transporter and metabolizing enzyme abundance in these tissues.ResultsAn Using chamber method has been successfully set up using both adult and pediatric intestinal tissue. To date samples from three children of different ages have been evaluated and show promising results. Tissue from the same patients has been stored for proteomics analysis.ConclusionThe Ussing method presents an innovative, feasible approach to study active intestinal transport in children. BackgroundTP53 is the most commonly mutated cancer driver gene but drugs that target TP53 are not yet available. A peptide derived from the most common p53 mutation R175H (HMTEVVRHC) can be presented by a common human leukocyte antigen (HLA-A*02:01) after proteasomal degradation.1 We aimed to develop T cell receptor (TCR)-mimic antibody targeting this shared neoantigen.MethodsWe constructed a single-chain variable fragment (scFv) phage display library that presents scFvs at an estimated diversity of 3.6e10. Mutant peptide-HLA (pHLA)-specific scFvs were enriched through five rounds of positive and negative selections. Mutant pHLA-specific scFv clones were converted into bispecific T cell retargeting antibodies in the single-chain diabody (scDb) format by linking with the anti-CD3 scFv UCHT1.2 These scDbs were tested in T cell co-culturs in the presence of target pulsed cells or target cells that either overexpress the p53 neoantigen or present the p53 neoantigen at endogenous levels. In vivo efficacy was assessed by administering scDb in NSG xenograft mouse model. The structural basis of the binding specificity was evaluated by X-ray crystallography.ResultsWe identified an scFv, termed clone H2, that specifically binds p53 R175H/HLA-A*02:01 pHLA and not its wild-type counterpart at a Kd of 86 nM (figure 1A). H2-scDb induced T cell cytokine release and cytotoxicity in the presence of 1) HLA-A*02:01-expressing cells pulsed with the p53R175H peptide, 2) cells transfected with HLA-A*02:01 and p53 R175H, and 3) cancer cell lines KMS26, KLE, and TYK-nu that express endogenous HLA-A*02:01/p53 R175H (figure 1B-E). T cell activation was diminished when TP53 was knocked out from these cancer cell lines using CRISPR (figure 1E). When administered to NOD scid gamma (NSG) mice systemically engrafted with the KMS-26 cell line, H2-scDb significantly suppressed tumor growth (figure 1 F, G). The structure of p53 R175H/HLA-A2 bound to the H2-Fab fragment shows that four complementarity-determining region loops of the H2 antibody formed a cage-like configuration around the C-terminus of the p53 R175H peptide, trapping the mutant histidine (His175) and the adjacent arginine (Arg174) residues in a stable interaction, which provides the structural basis for the specificity (figure 2).ConclusionsWe have developed a TCR-mimic bispecific T cell engager H2-scDb that recognized the shared neoantigen HLA-A*02:01/p53 R175H pHLA complex with exquisite specificity. It effectively activated T cells and lysed tumor cells both in vitro and in vivo. The ~2.0-1.8 Ga (billion years old) Belcher Group on the Belcher Islands in Nunavut provide a unique opportunity for studying Paleoproterozoic geobiology. The Belcher Group includes a sequence of low metamorphic grade peritidal carbonate rocks that preserve putative microbiota, as first described by Hofmann and Jackson (1969). Microbial mats, including stromatolites, are abundant in the peritidal carbonate succession. Additionally, morphologies possibly related to blue-green algae were first described in granular iron formation rocks of the Belcher Group by Moore (1918). The Belcher Group microbiota are a group of simple organisms, believed to be prokaryotic in nature. Microbiota morphologies include ellipsoids, spheroids, and filamentous chains of cells interpreted by previous workers to represent blue-green algae and acritarchs. Some microstructures are questionably biogenic and might be abiotic. The most significant field studies on the Belcher Group occurred from the late 1950s to the early 1980s, which provides the geological context for this study. This project aims to build on the previous work of H. Hofmann and others in the ‘60s and bring these microbiota into a modern context, drawing on the analytical advancements of the last 50 years. The main goal of the project is to determine if there is evidence that the microbiota are perhaps eukaryotic organisms. The emergence of eukaryotes is arguably the most significant geobiological event in Earth history, with eukaryotic cells believed to have evolved around 1.6 Ga (Knoll et al. 2006; Javaux and Lepot 2018), but some contentious fossils interpreted to represent eukaryotes have been dated to as early as 2.2 Ga (Retallack et al. 2013). In North America, the oldest discovered eukaryotic remains are around 1.5 Ga (Adam et al. 2017). If eukaryotic fossils were to be discovered in the Belcher Group, this would make them the oldest occurrence in North America. To test the hypothesis, samples from the microbiota-containing units were collected on the Belcher Islands. Both light microscopy and a collection of modern analytical techniques will be used to obtain high resolution images and chemical signatures of the microbiota and their biosignatures. Preliminary data from petrography, Raman Spectroscopy, and X-ray Photoelectron Spectroscopy (XPS) will be presented. Both Raman spectroscopy and XPS have been used as characterization tools in other studies looking at microbiota and organic matter remains (Qu et al. 2018; Arnarson and Keil 2001). Raman collects molecular and structural data from the sample, while XPS collects elemental chemical data.
