Abstract:

The Internet of Things is changing all sectors such as manufacturing, agriculture, city infrastructure, and the automotive industry. All these applications ask for secure processors that can be embedded in the IoT devices. Furthermore, these devices are restricted in terms of computing capabilities, memory, and power consumption. A major challenge is how to meet the need for security in such resource-constrained devices. This paper presents a customized version of LEON3, the ReonV RISCV (Reduced Instruction Set Computer-five) processor, dedicated for IoT applications that has strong effective security mechanisms built in at the design stage. Firstly, efficient lightweight cipher designs are elaborated and validated. Then, the proposed cryptographic instructions (PRESENT and PRINCE) are integrated into the default instruction set architecture of the ReonV processor core. The instruction set extensions (ISE) of lightweight cipher modules can be instantiated in software routines exactly as the instructions of the base architecture. A single instruction is needed to implement a full lightweight cryptographic instruction. The customized ReonV RISCV processor is implemented on a Xilinx FPGA platform and is evaluated for Slice LUTs plus FF-pairs, frequency, and throughput. Obtained results show that our proposed concepts not only can achieve good encryption results with high performance and reduced cost but also are secure enough to resist against the most common attacks.

Abstract:

INTRODUÇÃO: Métodos clínicos e laboratoriais estão sendo cada vez mais desenvolvidos para avaliar os diferentes aspectos do equilíbrio postural e estabelecer parâmetros para identificação de idosos com maior risco de quedas. OBJETIVOS: Analisar a correlação entre três testes utilizados para avaliar o risco de quedas em idosos. MATERIAIS E MÉTODOS: Tratou-se de um estudo analítico transversal realizado com 49 idosos comunitários, hígidos, com diferentes níveis de condicionamento físico. Foram utilizados dois testes clínicos, o Timed Up and Go (TUG) e o QuickScreen Clinical Fall Risck Assessment (QuickScreen), e um laboratorial, o Biodex Balance System - Modo Fall Risck (BBS-FR). A correlação dos dados foi realizada por meio da aplicação do coeficiente de correlação de Spearman, com nível de significância de 5% (p < 0,05). RESULTADOS: Verificou-se correlação positiva e moderada entre o QuickScreen e o BBS-FR (r = 0,35; p = 0,01) e entre o QuickScreen e o TUG (r = 0,32; p = 0,02). Não houve correlação significativa entre o BBS-FR e o TUG (r = 0,10; p = 0,47). CONCLUSÃO: Os testes analisados são complementares, tendo em vista que não se correlacionaram fortemente e mostraram-se com particularidades e limitações distintas. Entretanto, sugerimos a utilização do instrumento QuickScreen no ambiente clínico, já que este foi o que melhor se correlacionou com o dispositivo laboratorial, o BBS-FR, e o que mais identificou idosos ativos com risco de quedas. Dessa forma, o QuickScreen avaliou o risco de quedas de forma mais ampla, uma vez que engloba questões não abordadas pelos dois outros testes estudados.

Abstract:

ABSTRACTAdenoviruses encode a set of highly abundant microRNAs (mivaRNAs), which are generated by Dicer-mediated cleavage of the larger noncoding virus-associated RNAs (VA RNAs) I and II. We performed deep RNA sequencing to thoroughly investigate the relative abundance of individual single strands of mivaRNA isoforms in human A549 cells lytically infected with human adenovirus 5 (Ad5) at physiologically relevant multiplicities of infection (MOIs). In addition, we investigated their relative abundance in the endogenous RNA-induced silencing complexes (RISCs). The occupation of endogenous RISCs by mivaRNAs turned out to be pronounced but not as dominant as previously inferred from experiments with AGO2-overexpressing cells infected at high MOIs. In parallel, levels of RISC-incorporated mRNAs were investigated as well. Analysis of mRNAs enriched in RISCs in Ad5-infected cells revealed that only mRNAs with complementarity to the seed sequences of mivaRNAs derived from VA RNAI but not VA RNAII were overrepresented among them, indicating that only mivaRNAs derived from VA RNAI are likely to contribute substantially to the posttranscriptional downregulation of host gene expression. Furthermore, to generate a comprehensive picture of the entire transcriptome/targetome in lytically infected cells, we determined changes in cellular miRNA levels in both total RNA and RISC RNA as well, and bioinformatical analysis of mRNAs of total RNA/RISC fractions revealed a general, genome-wide trend toward detargeting of cellular mRNAs upon infection. Lastly, we identified the direct targets of both single strands of a VA RNAI-derived mivaRNA that constituted one of the two most abundant isoforms in RISCs of lytically infected A549 cells.IMPORTANCEViral and cellular miRNAs have been recognized as important players in virus-host interactions. This work provides the currently most comprehensive picture of the entire mRNA/miRNA transcriptome and of the complete RISC targetome during lytic adenovirus infection and thus represents the basis for a deeper understanding of the interplay between the virus and the cellular RNA interference machinery. Our data suggest that, at least in the model system that was employed, lytic infection by Ad5 is accompanied by a measurable global net detargeting effect on cellular mRNAs, and analysis of RISC-associated viral small RNAs revealed that the VA RNAs are the only source of virus-encoded miRNAs. Moreover, this work allows to assess the power of individual viral miRNAs to regulate cellular gene expression and provides a list of proven and putative direct targets of these miRNAs, which is of importance, given the fact that information about validated targets of adenovirus-encoded miRNAs is scarce.

Abstract:

Abstract Diverse classes of silencing small (s)RNAs operate via ARGONAUTE-family proteins within RNA-induced-silencing-complexes (RISCs). Here, we have streamlined various embodiments of a Q-sepharose-based RISC-purification method that relies on conserved biochemical properties of all ARGONAUTEs. We show, in multiple benchmarking assays, that the resulting 15-min benchtop extraction procedure allows simultaneous purification of all known classes of RISC-associated sRNAs without prior knowledge of the samples-intrinsic ARGONAUTE repertoires. Optimized under a user-friendly format, the method – coined ‘TraPR’ for Trans-kingdom, rapid, affordable Purification of RISCs – operates irrespectively of the organism, tissue, cell type or bio-fluid of interest, and scales to minute amounts of input material. The method is highly suited for direct profiling of silencing sRNAs, with TraPR-generated sequencing libraries outperforming those obtained via gold-standard procedures that require immunoprecipitations and/or lengthy polyacrylamide gel-selection. TraPR considerably improves the quality and consistency of silencing sRNA sample preparation including from notoriously difficult-to-handle tissues/bio-fluids such as starchy storage roots or mammalian plasma, and regardless of RNA contaminants or RNA degradation status of samples.

A discussão sobre a concepção de risco é ampla e permeia desde uma perspectiva mais geral, que busca contextualizá-lo na dinâmica da mudança da sociedade, até a abordagem mais específica na área da saúde, particularmente nos estudos associativos da epidemiologia. A palavra risco tem sido cada vez mais freqüente nas revistas médicas nas últimas três décadas. Esse fenômeno, no entanto, não é exclusivo da área da saúde e é permeado pela diversidade de uma noção que esconde uma lacuna conceitual. Diante dessa diversidade, o presente texto, a partir de revisão bibliográfica, procura sistematizar a discussão sobre risco. O resultado está organizado em três seções. Na primeira, é apresentada uma perspectiva mais geral da discussão sobre risco no âmbito do debate sobre a mudança da sociedade, na transição da modernidade para uma nova fase de organização social; na segunda parte, estão sintetizados os usos da noção de risco na área de conhecimento da saúde; na última, a constituição do conceito epidemiológico de risco e sua articulação com a clínica.

Abstract:

Abstract RNA interference (RNAi) is mediated by small, 20-24-nt-long, non-coding regulatory (s)RNAs such as micro (mi) and small interfering (si) RNAs via the action of ARGONAUTE (AGO) proteins. High-throughput sequencing of size-separated sRNA pools of plant crude extracts revealed that the majority of the canonical miRNAs were associated with high molecular weight RNA-induced silencing complexes co-migrating with AGO1 (HMW RISC). In contrast, the majority of 24-nt-long siRNAs were found in association with low molecular weight complexes co-migrating with AGO4 (LMW RISC). Intriguingly, we identified a large set of cytoplasmic sRNAs, including mature miRNA sequences, in the low molecular size range corresponding to protein-unbound sRNAs. By comparing the RISC-loaded and protein-unbound pools of miRNAs, we identified miRNAs with highly different loading efficiencies. Expression of selected miRNAs in transient and transgenic systems validated their altered loading abilities implying that this process is controlled by information associated with the diverse miRNA precursors. We also showed that the availability of AGO proteins is a limiting factor determining the loading efficiency of miRNAs. Our data reveal the existence of a regulatory checkpoint determining the RISC-loading efficiencies of various miRNAs by sorting only a subset of the produced miRNAs into the biologically active RISCs.

Abstract:

Aging can be defined as the progressive loss of physiological homeostasis that leads to a decline in cellular and organismal function. In recent years, it has become clear that small RNA pathways play a role in aging and aging related phenotypes. Small RNA pathways regulate many important processes including development, cellular physiology, and innate immunity. The pathways illicit a form of posttranscriptional gene regulation that relies on small RNAs bound by the protein components of the RNA-induced silencing complexes (RISCs), which inhibit the expression of complementary RNAs. In Drosophila melanogaster, Argonaute 1 (Ago1) is the core RISC component in microRNA (miRNA) silencing, while Argonaute 2 (Ago2) is the core RISC component in small interfering RNA (siRNA) silencing. The expression of Ago1 and Ago2 is regulated by stress response transcription factor Forkhead box O (dFOXO) increasing siRNA silencing efficiency. dFOXO plays a role in multiple stress responses and regulates pathways important for longevity. Here we use a next-generation sequencing approach to determine the effects of aging on small RNA abundance and RISC loading in male and female Drosophila. In addition, we examine the impact of the loss of dFOXO on these processes. We find that the relative abundance of the majority of small RNAs does not change with age. Additionally, under normal growth conditions, the loss of dFOXO has little effect on the small RNA landscape. However, we observed that age affects loading into RISC for a small number of miRNAs.

Abstract:

A presença de estafilococos coagulase positiva e coliformes a 45ºC em alimento que será consumido cru representa potencial risco à saúde do consumidor. Buscou-se averiguar a presença de bactérias pertencentes aos grupos dos estafilococos e coliformes em sushis confeccionados à base de salmão comercializados em Parnaíba/PI. Foram realizadas análises para determinar as contagens de Bactérias Heterotróficas Cultiváveis (BHC), estafilococos coagulase positiva e NMP de coliformes a 45ºC. Os resultados foram comparados aos limites aceitáveis pela RDC nº 12 de 2001 (ANVISA). As contagens de BHC variaram de 1,3 x 10 5 a 4,0 x 10 7 UFC/g e 4,6 x 10 4 a 2,2 x 10 5 UFC/g para os estabelecimentos A e B. As contagens de estafilococos coagulase positiva para as amostras coletadas no estabelecimento A indicaram níveis acima do permitido e ausência para o estabelecimento B. A presença de coliformes a 45ºC foi confirmada para os dois estabelecimentos, no entanto, 25% das amostras do estabelecimento B expressaram níveis acima do permitido. O presente estudo demonstra o risco microbiológico em sushi preparado à base de salmão cru ao confirmar a presença de bactérias patogênicas em concentrações acima do permitido pela legislação nacional vigente.

As manobras de ressuscitação cardiopulmonar (RCP) durante a pandemia de COVID-19 em pacientes internados graves é requerida mais habitualmente. De uma maneira geral, o paciente sofre a parada cardiorrespiratória em decorrência da hipóxia que a Síndrome Respiratóra Aguda Grave (do inglês severe acute respiratory sundrome – SARS) leva. Entretanto, pacientes com arritmias ventriculares e choque já estão predispostos à parada cardiorrespiratória. Entretanto, é importante a equipe médica saber quando ressuscitar ou não o paciente, principalmente em decorrência da possível falta de Equipamentos de Proteção Individual (EPIs) da equipe, o que pode levar a um risco alto de contaminação dos envolvidos. Desta forma, as etapas, equipes e protocolos devem ser seguidos à risca na salvaguarda do pacente e da equipe médica.

Apesar dos avanços observados no tratamento do HIV/AIDS nas últimas décadas, denota-se o aumento das condutas sexuais de risco, que contribuem no aumento de novos casos de HIV+. Este artigo teve como objetivo principal analisar se os constructos psicossociais, resiliência e conhecimento sobre HIV/AIDS, protetores contra a vulnerabilidade à infecção pelo vírus HIV, estão associados às características sociodemográficas. Contou-se com 600 pessoas adultas da população em geral (M= 27,31 anos; DP=9,37), sendo 59,2% mulheres e 40,8% homens. Utilizaram-se os seguintes instrumentos: questionário de dados sociodemográficos e comportamentos sexuais, escala de conhecimento sobre HIV/AIDS e a escala de resiliência CD-RISC-BR. Empregou-se o software SPSS versão 22 para a análise dos dados. Observou-se que os níveis de resiliência variaram significativamente em função das variáveis sociodemográficas: sexo, escolaridade, situação laboral, iniciação sexual e realização do teste de HIV. No que se refere ao conhecimento sobre HIV, encontrou-se diferenças estatisticamente significativas em função das características: orientação sexual, iniciação sexual e realização do teste de HIV. Por fim, espera-se que este estudo possa contribuir na formulação de intervenções preventivas em saúde aos comportamentos sexuais de risco ao HIV.

Este artigo tem o propósito de relatar a implementação física de um processador de 32-bits de arquitetura do tipo RISC, de conjunto de instruções reduzidas, denominado de RISCO. Este processador é projetado para uma arquitetura simples com a capacidade reconfigurável, proporcionando uma fácil adaptação para os limites especificados dentro de um projeto de SoC. O processador foi implementado na tecnologia XFAB 600nm usando o conjunto de ferramentas EDA Cadence. O principal objetivo do projeto é obter e compartilhar experiências em projetos ASIC e no desenvolvimento de síntese física de circuitos integrados. A implementação física do processador em questão resultou em um circuito de 15,18 mm² com 9.247 células e uma potência estimada de 254 mW.

Abstract:

Intestinal ischemia is an abdominal emergency with a mortality rate >50%, leading to epithelial barrier loss and subsequent sepsis. Epithelial renewal and repair after injury depend on intestinal epithelial stem cells (ISC) that reside within the crypts of Lieberkühn. Two ISC populations critical to epithelial repair have been described: 1) active ISC (aISC; highly proliferative; leucine-rich-repeat-containing G protein-coupled receptor 5 positive, sex determining region Y-box 9 positive) and 2) reserve ISC [rISC; less proliferative; homeodomain only protein X (Hopx)+]. Yorkshire crossbred pigs (8–10 wk old) were subjected to 1–4 h of ischemia and 1 h of reperfusion or recovery by reversible mesenteric vascular occlusion. This study was designed to evaluate whether ISC-expressing biomarkers of aISCs or rISCs show differential resistance to ischemic injury and different contributions to the subsequent repair and regenerative responses. Our data demonstrate that, following 3–4 h ischemic injury, aISC undergo apoptosis, whereas rISC are preserved. Furthermore, these rISC are retained ex vivo in spheroids in which cell populations are enriched in the rISC biomarker Hopx. These cells appear to go on to provide a proliferative pool of cells during the recovery period. Taken together, these data indicate that Hopx+ cells are resistant to injury and are the likely source of epithelial renewal following prolonged ischemic injury. It is therefore possible that targeting reserve stem cells will lead to new therapies for patients with severe intestinal injury. NEW & NOTEWORTHY The population of reserve less-proliferative intestinal epithelial stem cells appears resistant to injury despite severe epithelial cell loss, including that of the active stem cell population, which results from prolonged mesenteric ischemia. These cells can change to an activated state and are likely indispensable to regenerative processes. Reserve stem cell targeted therapies may improve treatment and outcome of patients with ischemic disease.

No campo da educação pública, os acintes têm sido agudizados de forma reiterada, sobretudo no que concerne à redução de recursos ou disputa pelos fundos públicos. Enquanto elaboramos esse editorial, em 24 de fevereiro de 2021, diferentes associações acadêmicas e sindicais realizam manifestações contrárias à Proposta de Emenda Constitucional (PEC) n. 186 (BRASIL, 2019), de autoria do senador Marcio Bittar, a qual compromete a manutenção dos dispositivos constitucionais de vinculação de recursos da receita de impostos para a educação no âmbito da União, dos estados, do Distrito Federal e dos municípios. De fato, isso impacta negativamente as ações do Estado brasileiro na ampliação e priorização dos investimentos para garantir o direito inalienável à educação.

Por meio da análise de alguns discursos políticos, busca-se verificar o uso de “pequenas verdades” para operar revisionismos históricos e trazer ao debate público certos argumentos, legitimando-os e operando no campo ideológico com fins de normalizar determinados pensamentos e, ao mesmo tempo, atacar ideias e ideologias opostas. Vários fatores contribuem para essa ineficiência. Na busca por alternativas que contribuam para uma efetiva conservação dos bens de valor histórico e cultural, destaca-se a necessidade de uma participação mais efetiva da sociedade civil. Incentivos precisam ser criados para que investimentos privados sejam destinados ao patrimônio. A Lei Rouanet poderia ter esse objetivo, mas somente uma pequena parcela de seus recursos é destinada a essa finalidade. Recentemente aprovou-se a Lei que permite os endowments no Brasil e pode ser um caminho interessante a ser trilhado.
