Abstract:
Abstract Retinoic acid (RA) is a key morphogen involved in the establishment of various cell-fates in the developing embryo. The establishment of certain tissues, including mesodermal derivates such as the somites and heart depends on restricted exposure to RA during embryonic development, however, in regard to the possible involvement of RA for the emergence of blood in the human embryo very little is known. By using an inhibitor of cellular RA synthesis (DEAB) together with our human iPS-to-blood differentiation system we report enhanced generation of human hematopoietic progenitors cells (CD43/45+,CD34+), and early progenitor cells with an adult hematopoietic stem cell phenotype (CD43/45+,CD34+,CD38,CD90+,CD45RA-) possessing myeloid and lymphoid differentiation capacity. RA inhibition increased the output of these early progenitor cells by 2.7-fold (p-value: 0,006, n=6) compared to DMSO control, and increased the number of clonogenic progenitors (CFUs) by 2-fold (p-value: 0,022, n=6). This improvement is consistent when using both a human ES line (Hues3) and a human iPS-line (RB9-CB1, generated from cord blood). RA inhibition also increased the number of iPS-derived blood progenitors capable of differentiating into the lymphoid lineage, generating phenotypic T-lineage and natural killer cells after sub-culture on OP9-DL1 stroma. Conversely, and in support of our findings, directly adding RA was found to severely decrease the blood generation efficiency, with the early progenitor fraction significantly decreased even at low levels of RA exposure. In order to assess how the RA inhibition led to the increase in early progenitors, whether by directing pluripotent stem cell differentiation of mesodermal precursors towards blood, or by inferring an early progenitor cell maintenance and/or expansion, we performed the following analyses. Firstly, the loss of clonogenic progenitors was reduced 1.6 fold with extended culture in the presence of the inhibitor. Secondly, within the iPS derived blood (CD43/45+) fraction of the cultures, RA inhibition increased the frequency of the CD34+,CD38-,CD90+,CD45RA- compartment by 2.1-fold (p-value: 0,021, n=6). Similarly when comparing the CFU-forming capacity from equal numbers of sorted hematopoietic progenitors (CD43/45+,CD34+), cells generated with the inhibitor demonstrated 2-fold higher CFU frequency suggesting increased preservation of CFU-forming cells within the CD34+ progenitor fraction. Together this suggests that progenitor maintenance is improved with the inhibitor, however, the contribution to the total fold expansion seen with RA inhibition cannot be attributed solely to the increased maintenance of early progenitor cells in the culture. Using multi-timepoint RT-qPCR of samples obtained during the differentiation protocol, we analysed expression of key developmental genes and saw improved commitment of the differentiating culture towards mesoderm (BRACHYURY), decreased amounts of anterior lateral plate/cardiogenic mesoderm (NKX2.5), and enhanced expression of genes known to be relevant for definitive hematopoiesis and hemogenic endothelium (RUNX1, FLK1, APLNR, PDGFRa). In addition, preliminary data show that RA inhibition increases the frequency of the CD34hi,CD90hi,CD43neg population observed at day 8, recently described as the precursor population of definitive hematopoiesis, indicating that RA inhibition confers an early positive effect on the developmental commitment towards the hematopoietic mesoderm. Together, our data indicate that modulation of retinoic acid signaling improves blood generation from human pluripotent stem cell lines both by improving the development towards the hematopoietic mesoderm by favoring mesodermal blood precursor commitment, and by preserving the immature progenitor population from differentiation. Therefore, we propose that RA is a negative regulator of mesodermal formation and the specification towards hematopoietic precursors, and that it promotes the exhaustion of definitive hematopoietic progenitors. This suggests the possibility that blood evolutionarily formed in a region of the mesoderm where RA exposure from neighboring tissues, such as RA producing ectoderm, was restricted. Therefore, using small molecules to inhibit RA signaling is a novel approach to improve the generation of human blood in vitro. Die Verfügbarkeit neuer, gezielter Therapieformen unterstreicht den Bedarf an sensitiven bildgebenden Verfahren, die in der Lage sind, auch frühe Stadien der Erkrankung zuverlässig zu erfassen, eine Prognose zu erheben und die Wirksamkeit der eingesetzten Therapien zu dokumentieren. Die konventionellen Röntgenaufnahme als etablierter Goldstandard kann Gelenkdestruktionen bei Patienten mit bereits bekannter RA nachweisen, häufig entgehen jedoch frühe Krankheitsmanifestationen wie Weichteilentzündungen oder begin-nende knöcherne Erosionen der Diagnose. Magnetresonanztomographie (MRT) und Sonographie erfassen durch unmittelbare Darstellung von Weichteilstrukturen hochsensitiv frühe entzündliche und destruktive Veränderungen. Durch einen Überblick über veröffentliche Daten zu Rönt-gen, Computertomographie, MRT, Sonographie und Szintigraphie erörtert der folgende Artikel den aktuellen Stand sowie die zukünftige Rolle einzelner bildgebender Verfahren in der Diagnostik entzündlicher Gelenkerkrankungen.

Abstract:
The Swedish Water Management Research Programme (VASTRA) focuses on the development and demonstration of tools for more efficient eutrophication control when implementing the EU water framework directive in Sweden. During the first half of the programme, models for nitrogen flow were developed, and at present, similar models for phosphorus are under construction (e.g. HBV-P). The programme is interdisciplinary, and scientists are collaborating in actor-games and focus group evaluations including scenario analysis. The scenarios modelled in VASTRA phase I, show that (i) changed agricultural practices can be the most effective and least expensive way to reduce nitrogen transport from land to the sea; (ii) constructed agricultural wetlands may only have small impact on riverine nitrogen transport in some regions, due to natural hydrometeorological dynamics; (iii) removing planktivorous fish may be an efficient way of reducing the algal concentrations in lakes without the undesired side-effect of increased nutrient load to the down-stream river system. In VASTRA phase II, one of the highlights will be interdisciplinary scenario-modelling of different measure strategies in a pilot catchment of southern Sweden (Rönne å).

The rating curve enables the translation of water depth into stream discharge through a reference cross-section. This study investigates coupling national scale airborne laser scanning (ALS) and acoustic Doppler current profiler (ADCP) bathymetric survey data for generating stream rating curves. A digital terrain model was defined from these data and applied in a physically based 1-D hydraulic model to generate rating curves for a regularly monitored location in northern Sweden. Analysis of the ALS data showed that overestimation of the streambank elevation could be adjusted with a root mean square error (RMSE) block adjustment using a higher accuracy manual topographic survey. The results of our study demonstrate that the rating curve generated from the vertically corrected ALS data combined with ADCP data had lower errors (RMSE = 0.79 m3/s) than the empirical rating curve (RMSE = 1.13 m3/s) when compared to streamflow measurements. We consider these findings encouraging as hydrometric agencies can potentially leverage national-scale ALS and ADCP instrumentation to reduce the cost and effort required for maintaining and establishing rating curves at gauging station sites similar to the Röån River.

Älgförvaltningsområden har sedan 2012 varit en av de rumsliga skalor där älgförvaltningen i Sverige sker. I denna studie har Riksskogstaxeringens data över förekomst, täckningsgrad och betningsgrad av viltfoder per älgförvaltningsområde (2011–2015) analyserats tillsammans med Skogsstyrelsens prognos över foderproducerande ungskog (fodpro) och avskjutningsstatistik (avskjutningstäthet) från den nationella jaktdatabasen (Viltdata.se). Resultaten visar på både likheter och skillnader mellan undersökta älgförvaltningsområden (ÄFO). Björk var den viltfoderart med högst täckningsgrad i alla ÄFO samtidigt som den hade lägst betestryck. Rönn var den enda enskilda viltfoderart där täckningsgraden skiljde sig signifikant åt mellan några ÄFO, vilket tyder på att älgförvaltningsområdena är lika varandra i fråga om kvalitet av viltfoder. Trots signifikanta skillnader i den totala täckningsgraden av viltfoder mellan några av älgförvaltningsområdena tyder resultatet på att områdena även är lika varandra i fråga om kvantitet av viltfoder. Både avskjutningstätheten och några av betningsgraderna för tall skiljde sig signifikant åt mellan alla ÄFO. Det fanns svaga, positiva signifikanta samband mellan avskjutningstätheten och betningsgraden ”hård” för både tall och salix. Dessa samband skulle kunna bero på att en ökad älgtäthet genererat ett högre betestryck på tall och salix vilket i sin tur sannolikt medfört en högre avskjutning av älg. Det fanns inga signifikanta skillnader mellan Riksskogstaxeringens och Skogsstyrelsens skattningar av viltfoder. Metoderna för skattningarna skiljer sig dock åt i flera avseenden vilket gör att de får olika tillämpningsområden.

I Nacka kommun pågår sedan 2008 ett projekt med att skapa två museer i Fisksätra. De två museerna består dels av ett historiecenter som berättar om Slaget vid Stäket 1719 då svenska och ryska styrkor drabbade samman, dels består det av Fisksätra museum 2.0 där Fisksätraborna får dela med sig av sina egna berättelser från platsen. Syftet med museerna är bland annat att skapa identitet och inkludering i samhället och att locka fler besökare till området kring Stäket och Skogsö. Med hjälp av bland annat modeller, arkeologiska fynd och ett skylt- och guideprogram vill man förmedla Baggenstäkets historia och en basutställning kommer att finnas i Fisksätra Marina. Många utställningar använder sig av texter för att berätta om föremål och företeelser i utställningen. Kring dessa utställningstexter cirkulerar problem och många kritiska röster har höjts mot deras kvalitet. De anses bland annat vara för långa och svåra; styrande och opersonliga och skapade utan tanke på målgruppen. Vissa menar till och med att utställningar mår bättre utan text där föremålen får tala för sig själva. Den här uppsatsen handlar om denna kritik och alla de råd om hur man skriver bättre utställningstexter. Syftet är att ansvariga för projektet med de nya museerna ska utveckla ett kritiskt förhållningssätt till utställningstexter för att på så viss kringgå de fallgropar som man annars riskerar att hamna i.

Syftet med detta arbete har varit att undersöka möjligheten att konkretisera hjärnforskningens rön i förhållande till historiedidaktiken för att finna sätt att öka elevers motivation inför historieämnet. En genomgång av relevant forskning om motivation, historiedidaktik samt neuro- och kognitionsvetenskap kopplad till pedagogik, det senare en del av hjärnforskningen, visar på betydande överlappning vad gäller metodiska rekommendationer. Den empiriska undersökningen hade som syfte att undersöka två elevgruppers uppfattningar om historieundervisning, speciellt med avseende på begrepp som är centrala inom motivationsforskningen: erfarenheter, förväntningar, meningsfullhet och nytta. Undersökningens resultat visar att elevgrupperna, som studerar vid ettnaturbruksgymnasium, hade ganska höga förväntningar på ämnet historia. Trots detta varmåluppfyllelsen i kursen bristfällig. Detta arbete argumenterar för att hjärnforskningens rön kan användas för att belägga vissa didaktiska metoders effektivitet. Slutsatsen är dock att flera pedagogiska perspektiv måste kombineras för att skapa inre motivation, vilket kan öka elevens måluppfyllelse i ämnen som eleven initialt inte är intresserad av.

Rapport från senaste European Social Science History Conference i Glasgow, där kritiska rön om ADHD-diagnostisering presenterades under rubriken: "Changing conceptions on normal childhood behaviour: International perspectives on ADHD and learning disability". Rapporten behandlar den forskning som presenterades vid konferensen om kraftigt stigande diagnostisering av ADHD, orsakerna till detta, konkurrerande perspektiv samt centrala problem som relaterades till den ökande diagnostiseringen.<br>A report from the latest European Social Science History Conference in Glasgow 2012 presented critical findings about ADHD diagnoses under the title: ”Changing conceptions on normal childhood behaviour: International perspectives on ADHD and learning disability". The report covers research on rapidly rising diagnosis rates of ADHD, including possible reasons, competing perspectives, and key issues related to the increasing diagnosis. The evidence suggests that the precautionary principle is increasingly rare among the occupations that are central for the diagnosis of the condition. Historical and social scientific aspects of the disorder may give a different picture than the dominant and widespread definition, which is understood solely in a medical context with a focus on pharmacology.

Abstract:
This paper deals with village history around millstone quarry Eastern Utsjö in Malung, Sweden, in the hope of finding deposits that could date the quarry, in that the quarry in the current situation is dated only by the millstone fragments outside the resort. My purpose was to conduct a discussion on the quarry alone can be dated by deposits outside the resort. I have used FMIS RAÄ:s fornsök and a lot of literature to identify village history. The results showed that it is not possible to date the millstone quarry from village history, but through deposits outside the resort of Malung, which have been found around Mälardalen, Sweden through archaeological excavations, and derived from the millstone quarry Eastern Utsjö, can put an age on the quarry.
