Abstract:
The work presented in this thesis evaluated the potential of integrated management of root-knot nematodes. Emphasis was given to the use of two complementary biological control agents, Paecilomyces lilacinus and Bacillus firmus, together with appropriate cultural practices including soil solarization and organic soil amendments, in comparison with the judicious use of chemical control. Use of soil amendments for nematode population suppression, demonstrated that application of fresh-chopped grass and brassicacaeous amendments suppressed root-knot nematode populations. Root-knot nematodes (Meloidogyne spp.) are serious root pathogens that affect commercial crops world-wide. The overall aim of this study was to develop techniques to identify plant genes expressed in specialised cells induced to feed these nematodes in order to develop a novel approach to host resistance. Little data exists on which root-knot nematode species are present in Western Australia. An initial study was carried out to identify species present in the major horticultural areas of WA. Root-knot nematodes (Meloidogyne sp.) are sedentary endoparasites that infect roots of a wide range of plant species and cause considerable economic loss to many crops. Root-knot nematodes (RKN) transform selected root vascular cells into enlarged, multinucleate feeding sites called giant-cells that arise from repeated karyokinesis without cytokinesis. Giant-cells undergo extensive modifications of the cell wall architecture including cell wall thickening and the formation of ingrowths that act to increase the surface area of the plasma membrane to facilitate solute uptake by the nematode. Plant parasitic nematodes are economically important crop pests: of these root-knot nematodes (Meloidogyne spp.) have the widest host range and are recognised as the most significant nematode pests of crop plants worldwide. An alternative control strategy chosen here was to target and silence a conserved mechanism (RNA interference – RNAi) in the nematodes via transgenic plants. The RNAi pathway itself was studied
