All organisms respond to breaks in the DNA by promptly launching the DNA-damage response (DDR). This involves the recruitment of DNA repair proteins to sites of DNA damage and the activation of “checkpoint” activities that slow down or arrest cell-cycle progression, thus delaying key cell cycle transitions until the damage has been removed. In multicellular organisms, the inability to repair DNA damage and/or prolonged checkpoint activation can lead to programmed cell death or cause the cell to enter a permanent cell cycle arrest known as cellular senescence. Introduction: Therapeutic agents like doxorubicin, an anthracycline antibiotic drug, are widely used in cancer chemotherapy. The use of doxorubicin is limited however by an increased risk of cardiac damage as a side effect, and an increased cancer cell drug resistance mediated by efflux drug transporters. Strategies are needed to protect the heart and still allow the benefits of drug treatment. “Basic” fibroblast growth factor-2 (FGF-2) is a multi-functional protein. Abstract:
Nowadays, new spinal cord injury (SCI) cases are frequently due to non traumatic causes, especially vascular disorders. A prerequisite to developing mechanism-based neuroprotective strategies for acute SCI is a full understanding of the early pathophysiological changes to prevent later disability and paralysis. The immediate damage spreads from the initial site through excitotoxicity and metabolic dysfunction (ischemia, free radicals and neuroinflammation) to surrounding tissue (secondary damage). Using an in vitro neonatal rat spinal cord model, an experimental protocol (pathological medium,
