Abstract:
I uppsatsen har beneficieskyddet som finns vid utmätning av gäldenärens bostadsrätt utretts. Den skälighetsbedömning som ska göras har undersökts, där faktorerna socialt skydd och värdet på bostadsrätten varit i fokus. En kraftig ökning av bostadsrättspriserna under åren från beneficieregelns uppkomst har gjort bedömningen komplicerad. Värdet för bostadsrätter är idag jämförbart med värdet för fastigheter. Rättsläget är orättvist när en särbehandling av bostadsrättsägare sker och en borgenärs rätt till betalning kan bli beroende av vilken typ av bostad gäldenären besitter. Vidare har ett avskaffande av beneficieskyddet för bostadsrätter föreslagits när situationen ses ohållbar inför framtiden. En granskning av Kronofogdens verkställighet vid utmätning av gäldenärens fastighet har gjorts. Motsvarande skydd finns inte för fastigheter men efter målet Rousk mot Sverige i Europadomstolen har rättsläget förändrats. Kronofogden ska nu beakta de mänskliga rättigheterna och göra en proportionalitetsbedömning.

Abstract:
Utmätning är av betydelse för att vi ska kunna ha ett fungerande kreditsamhälle. Genom att tvångsvis förmå någon att utföra en prestation kan ses som ett skydd för parter i ett avtal eller kreditinstitut.   År 2013 tvångsutmättes en fastighet för en skatteskuld på 6 721 kronor. Fallet prövades i Europadomstolen där Sverige förlorade målet. Domstolen riktade kritik mot Sverige och mot systemet som sådant. Utmätningen hade utförts i enlighet med svensk rätt, men stred mot Europakonventionen för de mänskliga rättigheterna. Trots kritiken har den svenska lagstiftningen inte förändrats särskilt mycket.   Vid utmätning är det viktigt att det råder balans mellan borgenärens rätt att få sina fordringar betalda och gäldenärens rättigheter att inte betala inkorrekta skulder. Detta verkar i praktiken vara en mycket svår avvägning för Kronofogden som exekutiv myndighet. Vidare finns det andra brister med utmätningsförfarandet. Värdeförstöringen som sker vid utmätning är ett exempel.   Att svensk lagstiftning kan strida mot Europakonventionen är något som i så fall påverkar rättssäkerheten. Här uppstår emellertid frågan vad rättssäkerhet egentligen innebär. Trots att många rättsvetenskapsmän eller kvinnor och praktiskt verksamma jurister har beskriver ofta rättssäkerhet utifrån samma rättssäkerhetsfaktorer, men någon enhetlig definition av rättssäkerhet verkar således inte existera.
Execution is important in order for us to have a functioning credit market. By obligating someone to complete a performance can be seen as a protection for parties to an agreement or credit institution.  In 2013, a property was executed for a tax debt of 6 721 SEK. The case was tried in the European Court of Justice where Sweden lost the case. The court addressed criticism against Sweden and the system as such. The detention had been in accordance with Swedish law but violated the European Convention and Human Rights. Despite the criticism, Swedish legislation has not changed.  Regarding the execution procedure it is important that there is a balance between the creditor's right to receive his debts and the debtor's rights. In reality, this seems to be a very difficult balance for Kronofogden as an executive authority. Furthermore, there are other problems regarding the execution procedure. For example, the value destruction that occurs in execution procedure.   The fact that Swedish legislation violates the European Convention is something that can affect rule of law. Here, however, the question arises what rule of law really means. Although many lawyers have often described rule of law based on the same legal certainty, but a uniform definition of rule of law does not seem to exist.

A generalization of the Rouse model of the dynamics of polymers in solution is proposed. The motion of long polymer chains is considered to be due to exponentially correlated random forces driving the polymer segments, which is a more realistic model than the approximation of white thermal noise in the standard theory. Due to the fluctuationdissipation theorem such a model is consistent with the assumption that the solvent has weakly viscoelastic properties, which corresponds to the theory, originally proposed by Maxwell and later substantiated coming from first principles. A consequence of such approach is the appearance of “memory” in the polymer dynamics. To obtain a correct description at short times, we also include inertial effects into the consideration. Discrete and continuum models of the universal dynamics of polymer chains are built. Exact solutions are obtained for the center of mass motion of the polymer coil in the discrete variant of the theory. The time correlation functions describing the dynamics of internal modes are calculated in the continuum approximation. The results significantly differ from those in the standard Rouse theory and its later generalizations valid at long times.

Abstract:
This study concerns digital stories told by five young Romani students and one teacher from Roma Culture Class in Stockholm, Sweden. The theoretical framework draws from theories concerning identity, culture, Romanipe, the Romani value system and social semiotics. The material for the study is the digital stories that were analyzed qualitatively. The results show that the young Roma in the study demonstrate pride in their Romani background and culture, but also signs of changes and development in their culture. Some of the participants do not show external signs of Romani traditions, for example in the ways of dressing. However, they show their Romani descent through other signs and symbols as for example through images where the Romani tea table is especially emphasized. The study indicates that the use of multimodality seems to give the participants in the study more power over their own representations.

Abstract:
This dissertation documents the results of research evaluating policies to schedule for unanticipated disruptions in job shops. The disruptions studied in this research are of two types - machine failure and job release-time. The study was conducted using modified classical job shop problems with the minimize maximum completion time (Cmax) objective. Best random non-delay schedules (BRS) provided job sequences for each machine. Different slack policies based on frequency, duration and location of slacks in the schedule were used to strategically insert slack in the BRS schedule. The resulting robust schedules proactively managed machine or job releasetime disruptions. The change in Cmax quantified schedule robustness. Simulation was used to generate and modify the BRS schedules with and without slacks and disruptions, simulate disruption events and evaluate schedule performance. It was observed that policies that equally distribute slack to all tasks on heavily utilized machines and those that equally distribute slack to all tasks on all machines performed best. When the number of jobs to process was more than the number of machines and "big jobs" with long processing times on heavily utilized machines were present, the policy distributing slack tasks equally on heavily utilized machines exhibited superior performance. For systems with less variability both policies performed equally well. By comparing the average, minimum and maximum schedule deviations across all policies, the study concluded that strategically distributing slack to tasks on heavily utilized machines results in good robust schedules that can absorb the effects of disruptions.

The percussion ensemble is still a relatively young ensemble, with the first works by Edgard Varèse and Amadeo Roldan composed in the first third of the 20th century. Because of this youth, it is important to examine significant works for the percussion ensemble which establish themselves as staples to the repertoire. Christopher Rouse, a Pulitzer-Prize winning composer has written three such works, Ogoun Badagris (1976), Ku-Ka-Ilimoku (1978), and Bonham (1989). This study will closely examine each of these works, providing background, detailed analysis, and performance practice for each of these works.

The Avatar for trumpet and piano by Dr. Steve Rouse is one of the most challenging compositions in the trumpet repertoire. Due to The Avatar's challenges and increasing popularity, a study is necessary to aid its performance. Each movement is performed on a different instrument: Bb piccolo (with an optional A piccolo part) for Nativity, Bb Flugelhorn for Enigma-Release and Bb trumpet for Rebirth. In addition, the performer must convey one of the work's possible programmatic meanings: (1) The Hindu belief of an Avatar and its life cycle, (2) the life of Christ or (3) the human lifecycle. Chapter 1 gives historical information about the work. Chapters 2-4 discuss each movement of The Avatar programmatically and pedagogically. Facets of each movement are analyzed including differences in programmatic choices, rehearsal techniques and sound concepts. Chapter 5 provides recording suggestions, including choosing a recording engineer, preparing and planning for a recording section, choosing a venue and the benefits of hiring a tonmeister.

Idag är det ovanligt att romska ungdomar går ut grundskolan med gymnasiebehörighet. Orsakerna är många, bland annat ett flerhundraårigt utanförskap. Men vad görs egentligen för att bryta utanförskapet? Vi har tagit del av aktuell forskning i området och intervjuat sju personer som på olika sätt arbetar med romsk inkludering i skolan. Den forskning vi tagit del av rörande romers situation är på många sätt dyster. Men det finns också metoder som visat sig ge resultat. Bland annat att anställa så kallade brobyggare, romska förebilder som arbetar inom till exempel skolan. Ett återkommande problem är dock bristen på ekonomiska resurser och kontinuitet. Projekt påbörjas, men avslutas innan de hinner ge resultat för att pengarna tagit slut.

I uppsatsen analyseras representationen av romsk etnicitet i de amerikanska filmerna Dracula (1931, Browning), Thinner (1996, Holland), Drag Me To Hell (2009, Raimi), Draculas Son (1943, Siodmak) och Varulven (1941, Waggner) samt afroamerikansk etnicitet i Night of the Living Dead (1968, Romero), De Fördömdas Drottning (2002, Rymer), Draculas Son och Svart Mystik (1943, Tourneur). Syftet med uppsatsen är att blottlägga representationen av icke-amerikanska etniciteter genom analys av handling och bildspråk, samt jämföra framställningen med respresentationen av de amerikanska karaktärerna i valda filmer. Målet var att undersöka och kritiskt granska stereotypa bilder. Metoden vilar på en personlig, hermeneutisk tolkning av forskningsobjekten där den egna upplevelsen av filmerna står i centrum. Jag har även använt mig av intersektionalitet för att täcka alla identitetsskapande aspekter så som genus, etnicitet, religion, sexualitet med mera. Forskningen är av kritisk sort och influerad av Frankfurterskolans kritiska tankegångar där man anser att åskådaren blir påverkad av filmens ideologi. Den sista metoden är diskursanalysen inom vilken man anser att språket är bärare av ideologi och för att förstå samt genomskåda verket måste man blottlägga denna. Slutsatsen jag har dragit är att romska karaktärer inte porträtteras mångsidigt eller rättvist. Enligt de flesta filmerna är de en utstött grupp som inte fungerar i det amerikanska samhället. Deras påstådda kunskaper i svart magi gör dem till protagonistens ärkefiende. Representationerna utstrålar rädsla och hat. Den vita huvudpersonen står för normen och publiken nekas identifikation med någon annan grupp. De afroamerikanska karaktärerna är däremot inte lika ensidiga – två av filmerna har en negativ och stereotyp syn på gruppen medan Night of the Living Dead har en afroamerikansk protagonist. Man kan även se att gruppen inte anklagas för att vara fiender lika ofta som romska karaktärer.

The drug, N,N bis (ethylene)-P (1-adamantyl) phosphonic diamide inhibits focus formation of Rous Sarcoma Virus in tissue culture. Transformation of chick cells was inhibited when the drug was added to chick cells prior to infection. The drug did not inhibit the transformation of Normal Rat Kidney Cells infected with RSV, when the cells were grown at non-permissive temperatures and shifted to permissive temperatures upon addition of the drug. Nor did the drug revert cells transformed at permissive temperatures. These studies indicated that the inhibition of RSV is in the early stage of viral growth, possible penetration or uncoating.

The purpose of this essay is to explore the commissioning process of a wind ensemble piece by notable composer Christopher Rouse, and its significance to the wind ensemble repertoire. This essay will use the University of Miami Frost Wind Ensemble and Christopher Rouse's Wolf Rounds as the primary example. This discussion will provide an extensive background review of Christopher Rouse's early musical experience and life's accomplishments. It will focus on his creative process and document the performance preparation of Wolf Rounds as experienced by the Frost Wind Ensemble, conducted by Professor Gary D. Green. The final part of this essay will investigate the compositional insight of Wolf Rounds as well as the inspiration and motivation of Rouse to write for the wind ensemble genre. While Rouse has written for many different types of ensembles, this will be his first published addition to the wind repertoire. Wolf Rounds was chosen as the primary composition because of its significance to the composer as his first composition for winds and percussion.

Traditional bead-spring models of the polymer dynamics are based on the Einstein theory of the Brownian motion (BM), valid only at the times much larger than the particle´s relaxation time. The reason is in neglecting the inertial and memory effects in the dynamics. In the present work we use a generalized theory of the BM to build models of the dynamics of flexible polymers in dilute solution. The equations of motion for the polymer segments include the friction force that follows from the linearized Navier-Stokes hydrodynamics. It has a form of a memory integral. To get a correct description of the short-time dynamics, inertial effects are included into the consideration. For negligible hydrodynamic interactions (HI) between the beads the motion of the polymer center of mass is not influenced by internal forces within the chain and has been considered exactly. Then we include the HI int the description of motion of chains, which are assumed Gaussian in equilibrium. Analytical solutions for the observable time correlation functions describing the movement of the polymer coils significantly differ from the classical results showing, in particular, algebraic long-time tails and ballistic motion at short times.

Abstract:
The Rouse theory for the suspension of sand in the Oka estuary has been evaluated. The Rouse profile, which is often used to represent the vertical distribution of sand in suspension, is a function of the ratio of the sediment fall velocity to bed friction velocity. This ratio, ws/u*, varied with height above the bed, as it is dependent on grain diameter. The grain size of sampled suspended sand was finer near the surface and coarser near the bed. This was evident at the three anchor stations occupied at the Oka estuary, where the field work was carried out. The values of the Rouse parameter showed that the sand transport in suspension took place throughout the benthic boundary layer. The suspension threshold of Bagnold (1966) was found to discriminate accurately between bedload and suspended load. The critical Shields parameter for suspension of sand, derived from the field measurements, defines the suspension threshold better when the friction velocity is assumed constant. Laboratory work carried out to evaluate the vertical distribution of the friction velocity throughout the benthic boundary layer result in a constant distribution with height above the bed in the lower 10%. This supports the assumption made in the Rouse theory. The sand concentration profile is more complex than just Roussian, as three layers have been recognized: a Roussian layer; a buffer layer; and an inner layer. The majority (90%) of the mass flux of sand took place within the inner layer. The concentration in this layer is characterized by an exponential increase towards the bed. This profile was not asymptotic to the normally accepted bed concentration (0.65). A new simplified equation to predict sand concentration in the inner layer was generated whereby: , where z is height above bed and h is water depth. The reference concentration, Ca, of the Rouse profile is predicted using the inner layer equation and solving at the upper limit of the inner layer (z/h = 0.1). The concentration gradient and magnitude (at a given height) were not related to shear stress or near bed flow velocity. Therefore, it is recommended that a near-bed measurement be used to define concentration at the bed, a, from which the concentration profile may be constructed.

Abstract:
Both pseudorabies virus and the various strains of avian sarcoma and leukosis viruses are economically important pathogens. In Canada, pseudorabies is an exotic virus which primarily infects swine. The disease is rapidly fatal in most species although adult pigs may become carriers. Because pseudorabies virus (PrV) is exotic to Canada, imported animals are rigorously tested for the presence of antibody titers to the virus. The leukosis caused by Rous associated virus (RAV) has a major impact on the health of poultry flocks. Different strains of the avian leukosis viruses cause damage of differing severity but the general characteristics of infection include decreases in both egg and meat production. There would be significant benefits if non-infectious, antigenic fractions from both of these viruses could be produced in bacterial cells. These proteins could be used in serum antibody testing to make the detection of both viruses considerably simpler than the currently used ELISA and virus neutralization tests. As well, the production of large quantities of RAV env protein could simplify studies of the mechanisms of host specificity and provide additional information about chicken cell receptors. I have attempted to produce an antigenically recognizable protein from both pseudo-rabies virus and Rous associated virus. Attempts were made to express such a protein from pseudorabies virus by cloning either cDNA or genomic DNA fragments into plasmid vectors followed by colony immunodetection using a polyclonal antibody against PrV prepared in pig. Although several recombinants produced positive immunological results, further examination showed that none contained PrV DNA sequences. Standard screening methods were modified and improved during the course of this work. Subsequently, other laboratories identified significant difficulties in using unpurified polyclonal antibodies prepared in swine which provided an explanation for the results I observed. A cDNA library constructed from RNA prepared from RAV-1 was screened using a probe prepared from env sequences in pSR-XD2, a plasmid containing much of the RSV sequence. Transformants containing RSV homologous DNA were identified. During purification of these clones and subsequent screenings many of the plasmids identified as containing RSV homologous sequence lost their inserts. Additional studies demonstrated that plasmids which exhibited the strongest homology with the RSV probe and which contained the longest inserts, lost these inserts at a high rate. A second cDNA cloning using a different bacterial host and a different viral strain (RAV-2) produced several positive clones, with insert lengths ranging from 250 by to 950 bp. These inserts were sequenced and compared to known RSV and RAV sequences. A high degree of homology was observed. The longest RAV-2 cDNA fragments included most of the env gp37 polypeptide coding region. In order to test for expression of an antigen, inserts from two independently isolated recombinants were subcloned into lambdagt11 in each of the three reading frames. A polyclonal antibody prepared against Rous sarcoma virus was used to test for the expression of any recognizable antigenic determinants. No antigen production was detected even though nucleic acid plaque hybridizations demonstrated that the subcloning had been successful in introducing RAV-2 sequences into lambdagt11. Possible explanations for this, as well as a discussion of the composition of the RAV-2 sequences and their placement on the RSV genetic map are considered.

Abstract:
Immunochemical and immunohistochemical techniques were used to determine the role of post-translational modifications in the regulation of DNA polymerase α in Rat-1(tsLA24/RSV) cells. Immunoaffinity purification following sucrose gradient fractionation showed two immunospecific polypeptides of Mᵣ ≃ 185,000 and 220,000 only in those fractions exhibiting DNA polymerase α activity. The Mᵣ ≃ 220,000 polypeptide was shown to be phosphorylated, primarily at serine residues. Incubation of cell lysates with immobilized alkaline phosphatase reduced enzyme activity and subsequent readdition of ATP, but not ATP-γ-S, restored activity suggesting the involvement of an endogenous serine protein kinase. This kinase may be a cAMP dependent protein kinase because prior incubation of the catalytic subunit stimulated DNA polymerase α activity 3-4 fold. In the absence of serum growth factors or pp60ˢʳᶜ, DNA polymerase α activity and semi-conservative DNA replication rates in growth arrested cells were severely depressed. However, both polymerase activity and DNA synthetic rates were subsequently restored by either activation of pp60ˢʳᶜ by temperature shift or by serum addition. DNA polymerase α protein was found primarily in the nucleus of all cells in log phase, growth arrested or subsequently stimulated cultures, independent of whether the cells were replicating DNA. Stimulation by either pp60ˢʳᶜ or serum did not alter DNA polymerase α localization within the cell nor lead to a preferential synthesis of Mᵣ ≃ 220,000 peptide or proteolytic conversion of the Mᵣ ≃ 220,000 peptide to smaller peptides, but did result in phosphorylation of the Mᵣ ≃ 220,000 polypeptide. This phosphorylation was not apparent in serum deprived, growth arrested cells. It is suggested that pp60ˢʳᶜ acts to initiate DNA synthesis through the temporal activation of DNA polymerase α through a mechanism similar to that used by serum growth factors and that phosphorylation by a serine protein kinase serves an important function.

Christopher Rouse (b. 1949), winner of the Pulitzer Prize for his Trombone Concerto (1993) and a Grammy award for his Concerto de Gaudi (1999), has come to the forefront as one of America's most prominent orchestral composers. Several of Rouse's works feature quotations of and strong allusions to other composers' works that are used both rhetorically and structurally. These borrowings range from a variety of different genres and styles of works, from Claudio Monteverdi's L'incoronazione di Poppea to Jay Ferguson's "Thunder Island." Due to the more accessible filtering and funneling methods of musical borrowings (proliferation of mass media), the weighty discourses attached to them, and their variety of functions (critiquing canons, engaging in an allusive tradition, etc.), quotation has become elevated to the most prominent of musical actors that trigger narrative listening strategies, which in turn have a stronger role in the formation of narratives about music as well as narratives of music. The primary aim of this study is to adapt and apply more recent methodological narrativity frameworks to selected instrumental compositions by Rouse containing quotations, suggesting that their manner of insertion, their method of disclosure, and their referential potential can benefit from being examined through various narrative lenses as well as reveal their participation in certain roles of narrative functions. For this study, I have chosen six instrumental works by Rouse for examination - the Violoncello Concerto, Symphony No. 1, Iscariot, String Quartet No. 2, Seeing, and Thunderstuck. On a more specific level, the aim of this study is to investigate the manner, meaning, and motive of the quoted material in a select group of Rouse's compositions through various narratological lenses. To accomplish this, I intend 1) to establish a context for understanding the musical borrowing procedures of Rouse; 2) to explore how works containing quotations can be examined through various narrativity frameworks; 3) to inspect the ways in which borrowings can enhance or clarify the structural design and stylistic musical features for which he is known; 4) to investigate the various meanings that are generated from his borrowings; 5) to consider the extent to which Rouse's musical borrowings comment on various discourses, and 6) to examine the psychological needs of certain narratives triggered by quotation and the various questions they pose. This study does not attempt to systematically unify the works of Rouse that contain borrowings under a kind of "grand theory" in narrativity or borrowing studies, but rather to examine each work individually, noting the particular roles that borrowings play in regards to narratives of and about music. Fundamentally, I claim that narrativizing about music is a foundational psychological and social impulse, aiding to serve our curiosities about music's otherness qualities. Using both narratives of and about music to frame analyses, I hope to make a small contribution to the growing methodological frameworks of narrativity by featuring works containing borrowings by one individual composer, suggesting that other comprehensive approaches in borrowing studies can used for future composers.

L'objectif de la thèse a été d'étudier les propriétés dynamiques des polymères à grande densité d'interactions fortes intermoléculaires, en fonction de l'énergie et de la densité de ces interactions. Cette étude s'est effectuée par la méthode de la dynamique moléculaire. Deux types d'interactions fortes ont étés proposés et implémentés numériquement. Un premier modèle dit isotrope dans lequel des interactions fortes sont modélisées par un potentiel isotrope, représentant par exemple des interactions ioniques. Et un second modèle dans lequel nous avons choisi un potentiel directionnel, modélisant les liaisons polaires ou les liaisons hydrogènes. L'unité de l'énergie des interactions fortes intermoléculaires a été fixée par rapport aux forces de Van der Waals des monomères non polaires qui forment la partie principale des polymères considérés J'ai étudié la dynamique de la relaxation à grande échelle (mode de Rouse) en fonction des paramètres pertinents (l'énergie et la densité) dans le régime dit Williams-Landel-Ferry (WLF). Ensuite, nous avons étudié les mécanismes de relaxation à petite échelle (monomérique) en cherchant à mettre en évidence des mécanismes spécifiques à ces polymères à fortes interactions. Nous avons également étudié la relaxation du stress. Nous avons mis en évidence une relaxation à temps intermédiaire entre le temps monomérique te les temps longs pendant laquelle le stress reste à un niveau relativement élevé, sur des échelles de temps comparables à ceux des mécanismes de relaxation spécifiques mis en évidence au niveau microscopique. Ce régime spécifique de relaxation du stress qui n'est pas observé à faible fraction de liaisons fortes est susceptible de dominer les propriétés mécaniques et viscoélastiques de ces polymères à l'état fondu. Nous avons également considéré le comportement ultime de ces polymères en étudiant les mécanismes de la cavitation. L'énergie à la rupture peut être augmentée par la présence de liaisons fortes à suffisamment forte densité le long des chaînes. Les systèmes à forte densité de liaisons polaires ne font plus apparaître de cavitation mais exhibent un comportement de strain hardening

Though previous studies have shown saltmarsh adaptability to some degree of sea-level rise (SLR), sediment supply is critical to sustaining saltmarshes as SLR accelerates. Land-use activities, such as dams, often influence watershed sediment transport and delivery to the coast. Previous studies have suggested that, even in small watersheds, dams can significantly impact coastal sediment budgets. The Parker River watershed (PRW) in northeastern Massachusetts hosts 20 dams and several natural lakes, and drains into the Plum Island Sound Estuary (PIE). This research aims to evaluate the impact of dams and sediment transport in the PRW. Three approaches were used: theoretical modeling of sediment transport patterns using digital elevation models; spatial analysis of suspended sediment concentration (SSC) and remote sensing data; and empirical calculations of reservoir trap efficiency. Geomorphic modeling indicates that bankfull discharge can transport 20 μm grains (silt) as wash load throughout the PRW. Sediment deposition might happen at Crane Pond and in reservoirs, but removing dams would not change this pattern. Both SSC data and observations of satellite images during high-flow events indicate low supply and transport of sediment throughout the PRW. The estimates of sediment yield (Y) are low for the PRW. An empirical calculation indicates little-to-no trap efficiencies for all dams. Therefore, fluvial contribution to the sediment budget of the PIS estuary is limited and dam removals in the PRW are unlikely to change the rate of sediment delivery to the PIE. The proposed method of this study provides an additional scope to assess the ecological benefits of removing a dam and could be easily replicated for other locations for similar assessment. Future studies should assess sediment dynamics and management practices from a more thorough perspective incorporating the riverine, estuarine and shelf system

Afin de se répliquer, les rétrovirus doivent disposer à la fois d’ARN épissés et non épissés. Chez le virus du Sarcome de Rous (RSV), l’accumulation de l’ARN non épissé dépend de l’élément NRS (Negative Regulator of Splicing). L’élément NRS est un élément bipartite. Sa région 5’ est assimilée à une séquence ESE (Exon Splicing Enhancer) à laquelle se fixent de nombreuses protéines SR tandis que sa région 3’ contient un pseudo-site 5’ non fonctionnel qui constitue un leurre qui est responsable de l’inhibition de l’épissage à l’unique site 5’ fonctionnel du virus. Seule la structure 3D de la partie 3’ du NRS qui contient le pseudo-site a été expérimentalement établie. Dans ce travail, nous avons déterminé la structure 2D de la totalité de l’élément NRS à l’aide de sondes chimiques et enzymatiques. La comparaison de cette structure expérimentale à celles que nous avons établies pour d’autres éléments NRS mutants fonctionnels et non fonctionnel ainsi qu’à celles théoriques de la totalité des virus aviaires séquencés argumente en faveur de la forte signification biologique de notre modèle. Des expériences d’épissage in vitro réalisées sur l’élément NRS sauvage ainsi que ses formes tronquées ont permis de mettre en évidence le rôle crucial de deux structures tige-boucles dans la fonction du NRS. Les expériences de purification de complexes formés avec un extrait nucléaire de cellules HeLa sur ces différents éléments NRS par des techniques chromatographie d’affinité ont permis de démontrer l’importance de l’association de ces deux structures tige-boucles avec les protéines SR et la snRNP U1. Nous avons défini un nouvel élément NRS minimal fonctionnel capable d’inhiber l’épissage et nous avons démontré l’activation de l’inhibition de l’épissage de l’élément NRS par la protéine 9G8 in vitro et in cellulo
Retroviruses require both spliced and unspliced RNAs for productive replication. Accumulation of unspliced RNA in Rous Sarcoma Virus (RSV) depends on the NRS element, (Negative Regulator of Splicing). The NRS element is bipartite. Its 5’ terminal part is considered as an ESE that binds SR proteins and its 3’ part contains a decoy 5’-splice site (ss), which inhibits splicing at the bona fide 5’ ss. Only the 3D structure of a small NRS fragment including the decoy 5’ ss had been experimentally studied. Here, by chemical and enzymatic probing of entire RSV NRS, we determine its 2D structure. By comparative analysis of 2D structures of functional and non-functional avian NRS variants and of all sequenced avian NRSs, we bring strong arguments for a biological significance of the established structure. By in vitro splicing assays, we show a crucial role of two of the established stem-loop structures and by affinity purification of complexes formed by WT and truncated NRSs in HeLa cell nuclear extract, we demonstrate their importance for SR protein and U1 snRNP association. We define a new small NRS element retaining splicing inhibitory properties and finally demonstrate the capability of the SR protein 9G8 to increase NRS activity in vitro and in cellulo

Le but de cette thèse était tout d'abord d'étudier les théories physiques qui décrivent la dynamique des polymères à l'échelle macroscopique. Le modèle de Rouse et la théorie d'enchevêtrement de P-G. de Gennes décrivent la dynamique des polymères non enchevêtrés et enchevêtrés, respectivement. Nous avons étudié les différentes transitions entre ces deux régimes en utilisant deux techniques expérimentales : Spéctroscopie dielectrique large bande et rhéologie. Un test complet du modèle de Rouse à été effectué en comparant les prédictions de ce modèle pour la dépendance en fréquence de la permittivité diélectrique et du module de cisaillement aux données expérimentales. Les effets d'enchevêtrement sur les spectres diélectriques ont été discutés. Nous avons ensuite développé des méthodes basées sur la microscopie à force électrostatique afin d'étudier les propriétés diélectriques locales. En utilisant une simulation numérique basée sur la Méthode des Charges Equivalentes, nous avons quantifié la constante diélectrique à partir de la mesure du gradient de force crée par un potentiel statique entre une pointe et un diélectrique. Cette méthode permet d'imager la constante diélectrique avec une résolution spatial de 40 nm. Le retard de phase de la composante en 2 omega de la force ou du gradient de force crée par un voltage alternatif est relié aux pertes diélectriques. Nous avons développé un mode d'imagerie des pertes diélectriques. Cette méthode simple pourrait être appliqué en biologie ou matière molle en générale afin d'étudier des variations locales de constantes dielectriques.

Few would deny the importance of art education in the total educational development of the child. Drawing is an integral component of art education. Drawing for children appears to be a natural form of expression. Studies involving the observation of children's drawing development have had a long history in relation to child art. Recognition that children pass through identifiable stages of development in drawing and that these stages could be affected by cultural and individual differences is an important focus in art education research. This study is concerned firstly with drawing development and secondly, with differences in drawing development across cultures. Eight year old children with predominantly European background and eight year old children with Vietnamese/Kampuchean background were chosen for this study. The study was concerned with determining whether there were any significant differences in drawing development between the two ethnic groups, using a modified scale of the Rouse "Descriptive Scale for Measurement of Art Products". The results of this study indicate that there are differences in drawing abilities between the two ethnic groups.

During the replicative cycle of retroviruses, the retrotranscribed viral DNA is integrated into the host chromosome by the viral integrase protein (IN). The integration reaction is essential for the viral life cycle. Therefore, IN is a key target for antiretroviral drug design to treat HIV infection. IN consists of three domains (N-terminal, central and Cterminal) connected by flexible loops, making the enzyme difficult to crystallize. Dr C. Ronfort (Team Retrovirus and Retroviral Integration) and Pr P. Gouet (BioCrystallography Laboratory) collaborate since 2002 in Lyon to study IN from the Rous Associated Virus type 1 (RAV-1). My thesis work lies within this collaboration. Its objective was to perform crystallographic and molecular studies of the central domain of RAV-1 IN and of mutants of interest identified by the team of Dr C. Ronfort. In this aim, the IN fragment has been overexpressed and purified. Its crystal structure has been solved to a resolution of 1.8 Å. The observation of this structure reveals that the RAV-1 IN can exhibit a novel dimeric arrangement with a molecular interface stabilized by three pairs of facing α-helices. This arrangement is also characterized by the presence of a basic narrow groove at its surface. Thanks to biochemical in vitro experiments and in silico docking studies, we have shown that this median groove could allow the binding of a linear singlestranded RNA. Moreover, our experimental data can explain how the crystallization conditions as well as the mutation of a specific residue located at the surface of the enzyme favor either this novel dimeric arrangement or the classical dimeric interface. Therefore, the data obtained during this thesis suggest that IN exhibits modular structural properties, allowing it to operate in several distinct steps of the retroviral cycle in presence of dsDNA (integration) or ssRNA (reverse transcription and/or encapsidation of the retroviral RNA genome)

L'épissage est une étape majeure dans le cycle de multiplication du virus VIH-1. Un seul transcrit permet la production d'une quarantaine d'ARNm différents par l'utilisation combinée de 4 sites 5' et 8 sites 3' d'épissage. Nous avons établit la structure secondaire de la région autour d'un site 3' (A7) et mis en évidence un nouvel élément régulateur alternatif Janus qui fixe les protéines hnRNP A1 ou les protéines SR ASF/SF2. Nous avons montré que l'épissage au site A7 est régulé par les protéines SR ASF/SF2, 9G8 et hnRNP A1. Par ailleurs, nous avons développé une approche de protéomique basée sur une chromatographie d'affinité couplée à une électrophorèse mono ou bi-dimensionnel suivie d'une analyse par spectrométrie de masse qui a permis l'identification de nouvelles protéines capables d'interagir avec un ARN contenant le site A7 et l'élément Janus. Enfin, nous avons créé un plasmide avec deux gènes rapporteurs luciférases permettant de quantifier l'épissage des ARN pré-messagers dans les cellules de mammifères. Pre-mRNA splicing is a key step for HIV-1 multiplication. Four donor and eight acceptor sites are used in combination to produce 40 different mRNA. We established the RNA secondary structure around the A7 3' acceptor site and discovered a new regulatory cis acting sequence called Janus element that binds either hnRNP A1 or ASF/SF2 proteins. Moreover, the A7 splicing site is regulated by ASF/SF2, 9G8 and hnRNP A1. We used a proteomic approach based on an affinity chromatography coupled with 1 or 2D gel electrophoresis and mass spectrometry. By this approach, we identified new proteins which can interact with an RNA containing A7 3' acceptor site and the Janus element. Finally, we developed a dual luciferase reporter plasmid that can measure the pre-mRNA splicing efficiency in mammalian cells. In parallel, we established RNA secondary structure of the NRS element of the Rous sarcoma virus and identified the binding sites for SR proteins (9G8, SRp40 and ASF/SF2) and hnRNP A1

Pour étudier l'influence de la structure chimique des précurseurs sur les propriétés dynamiques des polymères à la transition sol-gel, nous avons synthétisé deux systèmes polydiméthylsiloxanes (PDMS). Nos premiers résultats tendent à montrer que la structure chimique du réticulant n'influe pas sur les exposants critiques caractérisant la transition sol-gel. Après confrontation de nos résultats à ceux de la littérature, l'interprétation que nous proposons met en évidence deux transitions définissant ainsi trois domaines de comportement. Le mécanisme de percolation des gels PDMS semble être déterminé par le rapport du nombre de segments de Kuhn entre points de branchement sur celui entre enchevêtrements. During flooding, the suspended sediment transport usually experiences a wide-range of dilute to hyper-concentrated suspended sediment transport depending on the local flow and ground con-ditions. This paper assesses the distribution of sediment for a variety of hyper-concentrated and dilute flows. Due to the differences between hyper-concentrated and dilute flows, a linear-power coupled model is proposed to integrate these considerations. A parameterised method combining the sediment size, Rouse number, mean concentration, and flow depth parameters has been used for modelling the sediment profile. The accuracy of the proposed model has been verified against the reported laboratory measurements and comparison with other published analytical methods. The proposed method has been shown to effectively compute the concentration profile for a wide range of suspended sediment conditions from hyper-concentrated to dilute flows. Detailed com-parisons reveal that the proposed model calculates the dilute profile with good correspondence to the measured data and other modelling results from literature. For the hyper-concentrated profile, a clear division of lower (bed-load) to upper layer (suspended-load) transport can be observed in the measured data. Using the proposed model, the transitional point from this lower to upper layer transport can be calculated precisely.

À l'approche de la transition vitreuse les fondus de polymères montrent une augmentation importante de la viscosité de plusieurs ordres de grandeur. Le but de cette étude est d'acquérir une compréhension plus profonde des propriétés viscoélastiques des fondus de polymères vitrifiables. Les polymères sont modélisés comme des chaînes flexibles en utilisant un modèle de bille-ressort. Les propriétés dynamiques sont analysées dans le cadre de la théorie de couplage de mode idéale. Nous constatons que la température critique de couplage de mode varie avec l'inverse de la longueur de chaîne. En étudiant la fonction de relaxation de cisaillement, nous constatons que les processus de relaxation polymériques, ne sont pas modifiés, mais décalés vers des temps plus importants en approchant la transition vitreuse
Polymer melts show a remarkable increase of their viscosity by many orders of magnitude on approaching the glass transition. The aim of this study is to gain a deeper insight into the viscoelastic properties of glass forming polymer melts. The polymers are modeled as flexible chains using a bead-spring model. The dynamic properties are analyzed in the framework of the ideal mode-coupling theory. We find that the critical temperature of the ideal mode-coupling theory scales with the reciprocal chain length. By studying the shear relaxation function we find that the polymer relaxation processes are not altered but shifted to later times in the approach of the glass transition

Abstract:
The aim of this essay is to analyze the portrayal of the Romani people in 23 history textbooks marketed towards the Swedish lower secondary school between the years 1964–2014, but also which uses of history emerge from within these descriptions. During this period, the curriculum has passed through five different stages with varying approaches towards ethnic minorities. The method used to compare different textbooks, passages and time-periods was a qualitative and comparative content analysis based on a model by Professor Lennart Hellspong. This was supported by a theoretical framework focused around the typologies of uses of history by Professor Klas-Göran Karlsson. The findings show that Romani people are portrayed in similar themes or contexts over different curriculum-stages and many of the written passages focuses on the victimization and mistreatment of Romani by either Swedish or international actors. A comprehensive account of an objective and cultural history is only found in a few of the most current textbooks. The Romani and their history are often overlooked in favor of the majority society’s historical narrative if not a specific duty is charged towards their inclusion in the schools control documents. In the year 2000 the Framework Convention for the Protection of National Minorities was ratified by the Swedish government and the subsequent revision of the curriculum made this inclusion a permanent fixture. However, some textbooks keep minority history brief and unsatisfying. The uses of history regarding Romani people vary over time and appear to be subject to the overarching sociopolitical landscape. An excluding and existential nationalistic narrative is identified to be common during the 1960’s but transforms towards a more including variety over time. Non-use and political-pedagogical use are both partial in varying degrees during all the curriculum-periods coupled with a perceived scientific presentation of historical facts, but not scientific use of history. The current curriculum introduces a widely moral and conciliatory use of history. This study confirms earlier research to an extent, but at the same time it extends to focus on the Romani people, a wider variety of books and a longer time-period compared with earlier studies on the subject. The findings open the possibility for future research on other Swedish or international minorities – or the Romani from other methodological or theoretical standpoints.
