Abstract:
The aim of the study was to compare the effects of an in-season five weeks of high-intensity training (HIT) with a concurrent HIT eccentric overload training (CT) program on male amateur team-sports players’ fitness performance. Twenty-two athletes were randomly assigned to HIT (n=10) or CT (n=12). Both groups completed a HIT program. The CT group also completed two-three sets of six repetitions of eccentric overload training. Before and after training, performance was assessed for change of direction ability (COD), repeated sprint ability for best time (RSAb), mean time (RSAm) and slowest time (RSAs), jumping, and shuttle-run performance. Within-group analyses in both groups showed substantial better scores on COD, RSAb and RSAm and shuttle-run performance. Between-group analyses showed greater improvements in COD, RSAb, RSAm, RSAs, and jumping after additional CT compared to solely HIT. In conclusion, compared to HIT alone, concurrent eccentric overload and HIT training within the same session improved COD, RSA, jump, and shuttle-run performance in basketball and soccers amateur players.

Abstract This study aimed to compare the effects of a traditional warm-up with two post-activation potentiation (PAP) warm-up strategies on the repeated sprint ability (RSA) of soccer players from national (NL) and regional (RL) competitive levels. Sixteen young players (NL, n = 8, age = 20.7 ± 1.4 y, body mass = 68.5 ± 7.0 kg, body height = 177.4 ± 5.2 cm; RL, n = 8, age = 20.8 ± 1.0 y, body mass = 68.7 ± 4.0 kg, body height = 176.6 ± 5.6 cm) were recruited to complete a traditional warm-up (CONTROL), a PAP warm-up incorporating squats with a load (~60% 1RM) that allowed a high speed (1 m/s) of movement and a high number of repetitions (PAP-1), and a PAP warm-up with a load (~90% 1RM) that allowed a moderate speed (0.5 m/s) of movement and a reduced number of repetitions (PAP-0.5). A RSA test (six 20-m sprints with 20 s of recovery) was performed 5 min after the PAP warm-up to assess the effects of the different protocols on the fastest sprint (RSAb) and the mean time of all sprints (RSAm). A meaningful improvement of RSA performance was observed with PAP-0.5, attaining a large effect on NL (RSAb, ES = -1.5; RSAm, ES = -1.3) and only a small effect on RL athletes (RSAb and RSAm, ES = -0.2). Moreover, when each RSA sprint performance was compared between NL and RL players, after PAP-0.5 greater performance for all sprints was observed in the NL players. Therefore, adding a heavy strength-based conditioning exercise during the warm-up prior to a RSA test may induce significant performance improvements in NL, but only small effects in RL players.

Abstract:
Indonesia is one of the countries with the most volcanoes in the world. Observation, monitoring, recording, dissemination of information and warning of danger signs for volcanic activity is one of the focuses in the response to volcanic disasters. The instrument used to monitor volcanic activity is Real-time Siesmic Amplitude Measurement (RSAM). RSAM is a system that provides a continuous measurement of the seismic amplitude of the absolute average of the number of seismic stations. RSAM occupies a strategic role in monitoring volcanic seismic activity, especially in times of crisis before the eruption. In this study designing RSAM systems using geophone sensors to detect seismic vibrations. System testing is done by varying the test frequency. The test frequency given starts from 10 Hz to 50 Hz. The results of this study indicate that a system that has been designed to build can detect vibration signals well. This is evidenced by the Fast Fourier Transform (FFT) spectrum which shows the suitability of the test frequency with the spectrum peak produced.

Dalam KEPMENPAN No. 25/2004 dinyatakan bahwa setiap instansi pemerintah wajib menyelenggarakan survey kepuasan masyarakat. Tujuan penelitian ini adalah untuk mengetahui tingkat kepuasan pasien, kinerja pelayanan, mutu pelayanan dan prioritas peningkatan kualitas pelayanan berdasarkan KEPMENPAN No. 25/2004. Disain penelitian adalah deskriptif kuantitatif. Penelitian dilakukan pada Bulan Mei hingga Juni 2013 di RSAM. Populasi penelitian adalah semua pasien yang datang ke RSAM pada waktu penelitian dilaksanakan. Sampel penelitian 500 orang. Teknik pengambilan sampel Random Block Sampling. Hasil penelitian menunjukkan bahwa semua item pertanyaan dinyatakan valid dan reliabel. Indeks kepuasan pasien adalah 76, mutu pelayanan “B”, kinerja unit pelayanan “Baik”. Prioritas peningkatan kualitas layanan pada unsur: prosedur pelayanan, persyaratan pelayanan, kedisiplinan petugas, kecepatan pelayanan, keadilan mendapatkan pelayanan, kewajaran biaya, kepastian biaya dan kepastian jadwal pelayanan.AbstractAhmad Mochtar Hospital (RSAM) is one of the hospitals owned by West Sumatra Province. RSAM has been implementing financial management strategy of district public service agencies (BLUD) since a few years ago that required financial and service accountability. KEPMENPAN No. 25/2004 states that each public service agency should undergo a public satisfaction survey. The objective of this study is to measure patient satisfaction, service performance, service quality, and the priority of service improvement based on KEPMENPAN method. The design of this study was descriptive quantitative. The population of this study was all patients visiting RSAM from May to June 2013, 500 patients were sampled by Random Block Sampling technique. The result shows that the questionare was valid and reliable. Patient satisfaction index is 76. Most of respondents (74%) perceived that the quality of service was "B" and the service performance was "Good". Priority of service quality improvement were: service procedures, term of service, disciplinary of service officer, speed of service, service equity, the reasonableness of service charges, the certainty of services cost and schedule.The recommendation for RSAM is to improve the “quality consciousness” culture, by disseminating the results of this survey. Patient satisfaction surveys based on KEPMENPAN is recommended to be held in other hospitals.

Purpose:To examine the effects of a low-volume repeated-power-ability (RPA) training program on repeated-sprint and changeof-direction (COD) ability and functional jumping performance.Methods:Twenty-two male elite young basketball players (age 16.2 ± 1.2 y, height 190.0 ± 10.0 cm, body mass 82.9 ± 10.1 kg) were randomly assigned either to an RPA-training group (n = 11) or a control group (n = 11). RPA training consisted of leg-press exercise, twice a week for 6 wk, of 1 or 2 blocks of 5 sets × 5 repetitions with 20 s of passive recovery between sets and 3 min between blocks with the load that maximized power output. Before and after training, performance was assessed by a repeated-sprint-ability (RSA) test, a repeated-COD-ability test, a hop for distance, and a drop jump followed by tests of a double unilateral hop with the right and left legs.Results:Within-group and between-groups differences showed substantial improvements in slowest (RSAs) and mean time (RSAm) on RSA; best, slowest and mean time on repeated-COD ability; and unilateral right and left hop in the RPA group in comparison with control. While best time on RSA showed no improvement in any group, there was a large relationship (r = .68, 90% CI .43;.84) between the relative decrement in RSAm and RSAs, suggesting better sprint maintenance with RPA training. The relative improvements in best and mean repeated-COD ability were very largely correlated (r = .89, 90% CI .77;.94).Conclusions:Six weeks of low-volume (4–14 min/wk) RPA training improved several physical-fitness tests in basketball players.

Indonesia merupakan salah satu negara dengan gunungapi terbanyak di dunia. Rangkaian gunungapi membentang dari Pulau Sumatera, Jawa, Bali dan Nusa Tenggara. Dari kepulauan di Laut Bdana sampai bagian utara Pulau Sulawesi merupakan daerah gunungapi terpanjang di dunia. Indonesia memiliki 129 gunungapi aktif, 70 buah diantaranya berancaman dan 500 buah tidak aktif. Sejak tahun 1800, paling tidak telag tercatat 600 kali letusan oleh 70 gunungapi di Indonesia. Karena itu, Indonesia dapat dikatakan sebagai negara yang rawan bencana, terutama ancaman bencana gunung berapi. Pengamatan, pemantauan, pencatatan, penyebaran informasi dan peringatan tanda bahaya terhadap aktivitas gunungapi menjadi salah satu fokus dalam penanggulangan bencana gunungapi. Untuk mendukung hal tersebut diperlukan instrumentasi deteksi aktivitas vulkanik gunungapi. Salah satu instrumentasi yang digunakan untuk memantau aktivitas gunung berapi Gunung Merapi adalah Real-time Seismic Amplitude Measurement (RSAM). RSAM adalah sistem yang menyediakan pengukuran kontinu dari amplitudo seismik rata-rata absolut dari jumlah stasiun seismik. RSAM menempati peran strategis dalam memantau aktivitas seismik gunung berapi terutama pada saat krisis sebelum letusan. Dalam penelitian ini, perancangan sistem RSAM menggunakan sensor geophone untuk mendeteksi getaran seismik. Frekuensi tes yang diberikan dalam penelitian ini termasuk 10 Hz, 30 Hz, dan 50 Hz. Sistem yang telah dirancang bangun dapat mendeteksi frekuensi dengan baik sebagaimana dibuktikan bahwa semakin besar frekuensi uji maka semakin banyak output grafik yang dihasilkan. Selain itu, besaran yang dihasilkan juga semakin besar. Pada frekuensi 10 Hz, magnitudo yang dihasilkan adalah 0,997, pada 30 Hz magnitudo yang dihasilkan 1,559 sedangkan pada 50 Hz magnitudo yang dihasilkan adalah 1,962. Sistem RSAM yang dirancang bangun menghasilkan amplitudo yang memiliki hubungan linier dengan besarnya getaran sumber disediakan..

The incidence of thalassemia in the world is based on data from the World Health Organization or the World Health OrganizationPurpose To determine the relationship between chelation therapy provision and serum ferritin levels in Thalassemia B Mayor patients in Dr. Hi Abdul Moeloek, Lampung Province in 2019.Aim: To find out the relationship between the adherence to iron therapy with serum ferritin levels at RSAM in 2019.Methods: This research is quantitative with the observational analytic method with a cross-sectional approach. The total population was 115 patients. A sample of 60 patients thalassemia β mayor. This research was conducted from Feb 2020 to completion. Analysis through univariate analysis.Results: the relationship of compliance with kelasi therapy with serum feritin levels in thalasemia β mayor patients at rsam hospital 2019, 34 respondents who obeyed chelation therapy, as many as 21 respondents (61.8%) had no risk ferritin levels (≤1000), of the 36 respondents who did not comply with chelation therapy, as many as 6 respondents (23.1%) had levels ferritin No Risk (≤1000) The results of the analysis also obtained an OR value of 5.38 (95% CI 1.7-16.9), which means that respondents who are adherent to iron therapy are at risk of having ferritin levels at risk (≥ = 1000) 5.38 times greater than who do not adhere to iron therapy.Conclusion: It is known that there is a relationship between adherence to iron therapy and serum ferritin levels at rsam hospital in 2019. Hasil : hubungan kepatuhan terapi kelasi dengan kadar ferritin serum pada pasien thalsemia β mayor di RSAM bandar lampung tahun 2019 34 responden yang patuh dalam terhadap terapi kelasi, sebanyak 21 responden (61,8%) memiliki kadar ferritin Tidak Beresiko (≤1000), dari 36 responden yang tidak patuh dalam terhadap terapi kelasi, sebanyak 6 responden (23,1%) memiliki kadar ferritin Tidak Beresiko (≤1000. Hasil analisis diperoleh pula nilai OR 5,38 (95% CI 1,7-16,9) yang berarti bahwa responden yang patuh dalam terhadap terapi kelasi, berisiko memiliki kadar feritin beresiko (≥=1000) 5,38 kali lebih besar dibandingkan dengan yang tidak patuh dalam terhadap terapi kelasi.Kesimpulan : Diketahui terdapat hubungan kepatuhan terapi kelasi dengan kadar ferritin serum di RSAM Bandar Lampung tahun 2020 Kata kunci : Thalasemia, Terapi Kelasi, Feritin Serum

Fenomena anak jalanan merupakan salah satu permasalahan krusial yang menyertai proses pembangunan. Dinamika kehidupan anak jalanan yang identik dengan budaya kemiskinan, dianggap menyimpang dari fungsi sosial anak karena berbagai aktivitas yang dilakukan di jalanan. Selama 9 tahun terakhir, Kementerian Sosial telah mengimplementasikan kebijakan, strategi dan Program Kesejahteraan Sosial Anak (PKSA) yang bertujuan untuk memberdayakan dan memenuhi kebutuhan anak yang hidup di jalanan, namun demikian upaya tersebut dipandang belum berjalan secara optimal. Gaung �Gerakan Sosial Menuju Indonesia Bebas Anak Jalanan� sebagai bagian dari PKSA masih �asing� terdengar, meski data statistik menunjukkan penurunan jumlah anak jalanan realitanya masih banyak anak jalanan yang melakukan berbagai aktivitas di sudut-sudut kota seperti di traffict light, stasiun-stasiun, terminal dan di sekitar pusat-pusat perbelanjaan. Hal ini membuktikan bahwa program-program perlindungan dan pelayanan anak jalanan belum berjalan secara efektif dan belum terintegrasi dengan baik. Penelitian ini bertujuan untuk mengetahui sejauh mana manifestasi PKSA dalam mewujudkan kesejahteraan sosial anak jalanan di Rumah Singgah Anak Mandiri (RSAM) Yogyakarta. Metode yang digunakan dalam penelitian ini adalah metode penelitian kualitatif dengan menggunakan deskriptif analisis. Setelah data terkumpul maka teknik analisis data dilakukan dengan tahapan seleksi data, penyajian data dan penarikan kesimpulan. Agar hasil penelitian dapat dipercaya maka dilakukan triangulasi guna menguji keabsahan data penelitian. Berdasarkan hasil penelitian maka dapat ditarik kesimpulan bahwa pelaksanaan PKSA di RSAM Yogyakarta memiliki dampak yang positif bagi anak jalanan. Hal ini dapat dilihat dari tercapainya kebutuhan dasar anak jalanan yang menjadi binaan, tercapainya pendidikan dasar anak jalanan karena sebagian besar anak jalanan bisa kembali bersekolah, dan berkurangnya waktu anak berada di jalanan. Clients hemodialysis who were hospitalized around 43.1% experienced anxiety conditions. This is caused by the Client renal failure undergoing hemodialysis, takes 12-15 hours of dialysis every week, or at least 3-4 hours therapies. This activity will take place continuously throughout their life. This research is aimed to determine the effect of Therapy Thought Stopping and Family Psychoeducation Toward anxiety clients who undergo hemodialysis in RSAM Bukittinggi. The research design is Quasi Experimental Pre and Post test with a total sample of 64 peoples. Who received Therapy Thought Stopping and Family Psychoeducation as many as 32 peoples, and Therapy Thought Stopping just as much as 32 peoples. The using HARS scale to determine the client's level of anxiety. The results showed a significant decrease in anxiety condition in the intervention group who received Therapy Thought Stopping and family psychoeducation, compared with group who only get of therapy thought stopping (P value <0.05). Therapy Thought Stopping and Family Psychoeducation hemodialysis is recommended to clients who experience anxiety in RSAM Bukittinggi.Keywords: Anxiety, Therapy Thought Stopping, Family Psychoeducation

Latar belakang. Kematian neonatal akibat sepsis neonatorum masih sangat tinggi, salah satu kemungkinan disebabkan kegagalan terapi antibiotik. Pseudomonas aeroginosa sebagai salah satu penyebab sepsis neonatorum kemungkinan sudah resisten terhadap beberapa antibiotik.Tujuan. Mengetahui resistensi Pseudomonas aeruginosa terhadap beberapa antibiotik.Metoda. Dilakukan penelitian deskriptif dengan desain potong lintang. Populasi target adalah pasien yang dirawat di Unit Perawatan Intensif Level II Neonatal Rumah Sakit Abdul Moeloek Lampung (RSAM), selama periode Juli−Desember 2010. Kriteria inklusi, pasien menunjukkan gejala klinis sepsis neonatorum. Kriteria eksklusi, pasien yang meninggal sebelum diambil spesimennya. Pasien diambil sampel darahnya untuk pemeriksaan biakan. Biakan yang tumbuh dan dicurigai sebagai koloni kuman diidentifikasi. Uji kepekaan dilakukan dengan menggunakan metode Kirby-Bauer (disc diffusion technique). Interpretasi National Committee for Clinical Laboratory Standard.Hasil. Terdapat 14 jenis antibiotik yang diteliti >50% resisten terhadap Pseudomonas aeruginosa seperti ampisilin, eritromisin, amoksisilin, sefuroksim, seftriakson, gentamisin, tetrasiklin, sefadroksil, piperasilin, trimetroprim, tobramisin, kotrimoksazol, nalidiksid, sulfonamid kompleks. Sedangkan 11 macam antibiotik yang diteliti >50% sensitif di antaranya meropenem, klindamisin, amikasin, norfloksasin, siprofloksasin, ofloksasin, fosfomisin, seftazidim, netilmisin, kanamisin. Antibiotik lini pertama di Unit Perawatan Intensif Level II Neonatal RSAM yaitu ampisilin dan gentamisin, lini kedua seftazidim dan amikasin sedangkan lini ke tiga adalah meropenem.Kesimpulan. Pseudomonas aeroginosa telah resisten terhadap 14 jenis antibiotik, sehingga para klinisi harus berhati-hati dalam memilih antibiotik terutama golongan sefalosporin.

From the results of preliminary research in the operating room of the Central Surgical Installation (CSI) R</em><em>SUD dr. Achmad Mochtar </em><em>(RSAM) Bukittinggi in January and February 2019 showed that 81% of elective surgeries were delayed. Delay ranged from 30 minutes to several hours. </em></p><p><strong><em>Objective</em></strong><em>: To find out what causes the delay in begining the first elective surgery in the operating room of the Central Surgery Installation at RSAM in 2019 in terms of input, process and output.</em></p><p><strong><em>Method</em></strong><em>: Qualitative research using the Framework Analysis method was conducted in the operating room of the CSI RSAM. The study began in April until October 2019</em></p><p><strong><em>Conclusion</em></strong><em>: The reason for the delay in begining the surgery in terms of HR input is that the leadership in CSI is not maximized, lack of discipline and commitment of the operator doctor and anesthesiologist, the anesthesiologist's schedule is not optimized, lack of registration officers, no optimal settings for radiographer for operating rooms and the lack of skills. The problem with SOP inputs is that there are still incomplete SOPs and not yet optimal implementation of existing SOPs. In the infrastructure input the problem is in the distance from the patient's ward to the operating room, CSSD is not integrated to the operating room and picking up surgery clothes and linen to CSSD is late. From the process factor, there are obstacles in the process of delivering patients to the operating room, the arrival time of the surgeon/operator and anesthesiologist are behind schedule, the lack of optimal monitoring and evaluation, and the lack of optimal implementation of reward and punishment. Output shows that from 40 firsts surgery schedules, 35 surgeries were delayed (87.5%) and only 5 operations were carried out on time according to the schedule (12.5%).</em></p><p><strong><em>Keywords</em></strong><em>: </em><em> Efficiency, Operating Room, Monitoring, Reward, Punishment</em></p>

Thalasemia adalah penyakit anemia hemolitik yang ditandai dengan adanya kelainan sintesis rantai globin. Transfusi secara terus menerus pada pasien thalassemia bisa menyebabkan terjadi penumpukan atau penimbunan zat besi sehingga kadar ferritin meningkat dalam tubuh terutama pada hati, jantung, dan organ endokrin. Masalah di kelenjar endokrin dan kondisi anemia dapat mengganggu pertumbuhan anak sehingga menyebabkan penurunan nafsu makan dan menyebabkan kurangnya asupan gizi kedalam tubuh. Penelitian ini untuk mengetahui hubungan antara kadar ferritin dengan status gizi pada pasien Thalasemia β Mayor anak. Penelitian ini menggunakan metode observasional analitik dengan pendekatan cross sectional. Subjek dalam penelitian berjumlah 60 pasien. Instrumen yang digunakan adalah Data rekam medis. Hasil analisis statistik menggunakan uji Chi-Square menunjukkan p-value sebesar 0,02 (p-value < 0,05) yang berarti bahwa terdapat hubungan antara kadar ferritin dengan status gizi pada pasien Thalasemia β Mayor anak di Rumah Sakit Abdul Moeloek Bandar Lampung Tahun 2019 . Latar Belakang: pada saat pengambilan darah eritrosit akan mengalami kerusakan, setiap hari viabilitas eritrosit menjadi terus menurun akibat dari penurunan kadar ATP (Adenosin Trifosfat), sehingga apabila kadar ATP menurun maka terjadinya kehilangan lipid membran, membrane menjadi kaku dan bentuknya dari cakram menjadi sferis (tidak central polar dan ukuran kecil). Penelitian ini ditujukan untuk mengetahui perbedaan nilai hematokrit darah segar dengan darah simpan (30 hari) di UTD RSUD Dr. H. Abdul Moeloek Lampung tahun 2020. Metode: Jenis penelitian yang digunakan dalam penelitian ini adalah kuantitatif observasional dengan desain non probability sampling. Sampel yang digunakan pada penelitian ini responden yang bersedia mendonorkan darahnya di UTD RSUD Dr. H. Abdul Moeloek Lampung. Analisa data menggunakan rumus wilcoxon. Hasil: diperoleh p value >0,05 yang artinya tidak terdapat perbedaan yang signifikan antara darah segar dengan darah simpan (30 hari) di UTD RSUD Dr. H. Abdul Moeloek Lampung. Kesimpulan: pengelolaan darah donor dan kualitas darah donor di tempat tersebut dalam keadaan baik sehingga darah donor bias diberikan kepada resipen, tanpa memiliki perubahan komponen yang signifikan.

Pemfigus foliaseus merupakan penyakit autoimun kronik yang ditandai dengan adanya bula yang kendur dan mudah pecah sehingga menghasilkan lesi erosi, skuama serta krusta. Insidensi pemfigus foliaseus diseluruh dunia sangat sedikit 2-10 kasus per satu juta penduduk menjadikannya penyakit ini tergolong langka, umunya terjadi pada orang dewasa antara usia 40-60 tahun. Pada jurnal ini, kami melaporkan kasus perempuan berusia 50 tahun yang datang dengan keluhan timbul lepuh tipis yang mudah pecah dengan sebagian bersisik dan kemerahan pada hampir seluruh tubuh sejam 10 hari sebelum berobat. Keluhan disertai dengan panas disertai gatal ringan pada kulit yang mengalami lesi. Pasien mengatakan keluhan yang di alaminya hilang timbul sejak 16 tahun yang lalu. Dari pemeriksaan dermatologis pada hampir seluruh tubuh ditemukan makula eritem berbatas tegas, vesikel hingga bula yang multiple, bentuk bulat hingga irregular, ukuran lentikular hingga nummular, persebaran diskret hingga konfluens. Terdapat pula skuama halus hingga sedang, selapis, multiple, berwarna putih hingga kecoklatan dengan bentuk yang tidak beraturan. Pasien kemudian ditatalaksana secara umum maupun khusus.

ABSTRACT The glaciated Mount Rainier volcano in southwestern Washington State (United States) has a rich history of outburst floods and debris flows that have adversely impacted infrastructure at Mount Rainier National Park in the 20th and 21st centuries. Retreating glaciers leave behind vast amounts of unconsolidated till that is easily mobilized during high-precipitation-intensity storms in the fall months, and during outburst floods during warm summer months. Over 60 debris flows and outburst floods have been documented between 1926 and 2019 at Mount Rainier. Debris-flow activity has led to the closure of campgrounds and visitor destinations, which has limited visitor access to large swaths of the park. This paper documents efforts to characterize and seismically monitor debris flows, map hazards, and develop forecasting approaches for wet and dry weather debris flows. Using the day-of and historic antecedent weather conditions on past debris-flow days, we developed a debris-flow hazard model to help predict those days with a higher relative hazard for debris-flow activity park-wide based on prevailing and forecasted weather conditions. Debris flows are detected in near-real-time using the U.S. Geological Survey Real-time Seismic Amplitude Measurement (RSAM) tool. If an event is detected, we can then provide evacuation alerts to employees and visitors working and recreating in the areas downstream. Our goal is to accurately forecast the debris-flow hazards up to 7 days ahead of time and then use RSAM to detect debris flows within minutes of their genesis.

ABSTRACT Transport of RsaA, the crystalline S-layer subunit protein of Caulobacter crescentus, is mediated by a type I secretion mechanism. Two proteins have been identified that play the role of the outer membrane protein (OMP) component in the RsaA secretion machinery. The genes rsaF a and rsaF b were identified by similarity to the Escherichia coli hemolysin secretion OMP TolC by using the C. crescentus genome sequence. The rsaF a gene is located several kilobases downstream of the other transporter genes, while rsaF b is completely unlinked. An rsaF a knockout had ∼56% secretion compared to wild-type levels, while the rsaF b knockout reduced secretion levels to ∼79%. When expression of both proteins was eliminated, there was no RsaA secretion, but a residual level of ∼9% remained inside the cell, suggesting posttranslational autoregulation. Complementation with either of the individual rsaF genes by use of a multicopy vector, which resulted in 8- to 10-fold overexpression of the proteins, did not restore RsaA secretion to wild-type levels, indicating that both rsaF genes were required for full-level secretion. However, overexpression of rsaFa (with normal rsaF b levels) in concert with overexpression of rsaA resulted in a 28% increase in RsaA secretion, indicating a potential for significantly increasing expression levels of an already highly expressing type I secretion system. This is the only known example of type I secretion requiring two OMPs to assemble a fully functional system.

Metode dalam penelitian ini adalah metode terapan apllied research dengan desain penelitian One-Group Pretest-Postest Design. Teknik pemilihan sample pada penelitian ini menggunakan non probability sampling denganpendekatanpurposive. Dalam penelitian ini menggunakan instrument penelitian Oral Assessment Guide (OAG) untuk menilai skor mukositis. Pada peneitian ini menunjukan bahwa berkumur larutan madu dapat menurunkan skor mukositis pada kedua responden. Hasil penelitian berkumur larutan madu dapat dijadikansebagai salah satu penerapan untuk mengatasi mukositis pada pasien kanker payudara yang menjalanikemoterapi di RSAM Bandar Lampung. Rekomendasi untuk penelitian selanjutnya agar dilakukan penelitian dengan jumlah sample yang lebih banyak dana danya kelompok control dan kelompok yang diberi intervensi agar diperoleh hasil secara statistic dan lebih valid.

Insiden karsinoma sel ginjal dan karsinoma sel transisional pelvis renalis sebagai jenis histopatologis terbanyak pada keganasan ginjal menunjukkan trend peningkatan insiden di seluruh dunia. Batu saluran kemih menunjukkan trend peningkatan insiden yang serupa. Hal ini memungkinkan adanya hubungan. Penelitian sebelumnya mendapatkan hasil yang kontroversial. Tujuan penelitian ini adalah melihat hubungan antara batu saluran kemih bagian atas dengan karsinoma sel ginjal dan karsinoma sel transisional pelvis renalis di RSUP Dr. M Djamil padang dan RSAM Bukittinggi. Penelitian ini menggunakan desain case control study pada rekam medis yang dilakukan pada bulan September hingga Desember 2018. Didapatkan 34 sampel dengan hasil histopatologi karsinoma sel ginjal dengan rasio jenis kelamin 1:1,3. Sampel terbanyak adalah kelompok umur 50-59 tahun (26,47%), mean: 52,8±13,79. Didapatkan 15 sampel karsinoma sel transisional pelvis renalis dengan rasio jenis kelamin 2,75:1. Sampel terbanyak adalah kelompok umur 50-59 tahun (33,33%), mean: 57,5±11,31. Persentase yang ada batu lebih tinggi pada karsinoma sel ginjal dibandingkan dengan yang tidak karsinoma sel ginjal yaitu 62,5% : 23,1%. Secara statistik perbedaan tersebut tidak bermakna (P>0,05). Peluang untuk timbulnya karsinoma sel ginjal sebesar 5,6 kali pada yang ada batu dibandingkan dengan yang tidak ada. Persentase yang ada batu lebih tinggi pada karsinoma sel transisional pelvis renalis dibandingkan dengan yang tidak karsinoma sel transisional pelvis renalis yaitu 71,4% : 25%. Secara statistik perbedaan tersebut tidak bermakna (P>0,05). Peluang unuk timbulnya karsinoma sel transisional pelvis renalis sebesar 7,5 kali pada yang ada batu dibandingkan dengan yang tidak ada.

AbstractPolymerase chain reaction (PCR) primers specific to exon 2 of the bovine lymphocyte antigen (BoLA)-DRB3 gene were used successfully to amplify the equivalent region in 34 Murrah and 36 Surti buffaloes selected at random. The 304 bp amplified product of the DRB3 gene was separately digested with BstγI, HaeIII and Rsal enzymes. Digestion with BstγI enzyme did not reveal any polymorphism and all animals showed a single restriction pattern, which corresponded exactly to the BstγI pattern ‘b’ previously described for cattle. Digestion with HaeIII enzyme resulted in five patterns, four of which corresponded to the Haelll patterns previously reported in cattle. The new HaeIII pattern was observed in both the breeds of buffaloes studied. The fragment analysis with RsaI revealed 13 different patterns. All of these RsaI patterns corresponded to the RsaI patterns previously described for cattle. The high degree of similarity in the restriction fragment length polymorphism (RFLP) patterns of cattle and buffalo observed in the present study provide evidence for the strong conservation amongst other bovine species, of restriction sites previously reported in cattle.

Abstract:
The composition of dry mix on the basis of two-component cementing agent (aluminous cement and clay of the “Kustikha” field), mineral additives (a metakaolin, the RSAM sulfoaluminate modifier, waste of basalt fiber), Ufapore foamer and the accelerating and plasticizing “Citrate-T” additive is developed. When mixing “Citrate-T” additive with water at Water/Solid = 0.45–0.70, the subsequent mechanical binder and hardening of a foam mass heat-resistant foam concretes with a density of 300–650 kg/m3 are formed (depending on Water/Solid value). Foam concretes have strength on compression of 0.2–2.5 MPa before warming up when their initial strength depends on processes of hydration curing of aluminous cement that provides fixation of their porous structure. After annealing at 1000 °C foam concretes have final strength of 0.3–3.2 MPa due to processes of solid-phase agglomeration of clay with other components of dry mix at their heating. Foam concretes after annealing unlike foam concretes on the basis of a Portland cement and aluminous cement have big strength. Introduction of the accelerating and plasticizing “Citrate-T” additive into composition of the dry mix leads to an increase of rheological properties in expanded foam mass and time reduction of its drying and curing. It has been established that an essential role is played by the relation Water/Solid: at increase in the relation Water/Solid (with 0.45 to 0.70) occurs increase in volume of foam mass after a mechanical binder, and also heterogeneity of pores and their sizes increases that leads to reduction of density of foam concretes and strength on compression.

Data collection was carried out through in-depth interviews with six participants using purposive sampling technique. Criteria for inclusion in the sample are the head of the room or the team leader in the nursing service unit. Data were analyzed by the Colaizzi method. Penelitian ini bertujuan untuk mengeksplorasi persepsi perawat tentang EBNP di RSAM Bukittinggi. Desain penelitian kualitatif dengan pendekatan fenomenologi. Pengumpulan data dilakukan melalui wawancara mendalam kepada enam partisipan dengan teknik purposive sampling. Kriteria inklusi sampel adalah kepala ruangan atau ketua tim pada unit pelayanan keperawatan. Data dianalisis dengan metode Colaizzi. Hasil penelitian ditemukan empat tema yaitu (1) Kurangnya pemahaman perawat tentang EBNP, (2) Hambatan dalam penerapan EBNP (3) Harapan perawat dalam penerapan EBNP, (4) Keuntungan dalam menerapkan EBNP. ABSTRACT RsmA (for regulator of secondary metabolism), RsmC, and rsmB RNA, the components of a posttranscriptional regulatory system, control extracellular protein production and pathogenicity in Erwinia carotovora subsp. carotovora. RsmA, an RNA binding protein, acts as a negative regulator by promoting message decay. rsmB RNA, on the other hand, acts as a positive regulator by neutralizing the effect of RsmA. RsmC modulates the levels of RsmA and rsmB RNA by positively regulating rsmA and negatively controlling rsmB. The level of rsmB RNA is substantially higher in RsmA+ bacteria than in RsmA− mutants. We show that rsmB RNA is more stable in the presence of RsmA than in its absence. RsmA does not stimulate the expression of an rsmB-lacZ transcriptional fusion; in fact, the β-galactosidase level is somewhat higher in RsmA− bacteria than in RsmA+ bacteria. We also investigated the basis for increased levels of rsmA and rsmB RNAs in the absence of the quorum-sensing signal, N-[3-oxohexanoyl]-l-homoserine lactone (OHL). The absence of OHL activates transcription of rsmA but not of rsmB. Instead, increased stability of rsmB RNA in the presence of RsmA accounts for the elevated levels of the rsmB RNA in OHL− bacteria. Mutant studies disclosed that while RsmA, OHL, and RsmC control the levels of rsmB RNA, high levels of rsmB RNA occur in the absence of RsmC or OHL only in RsmA+ bacteria, indicating a critical role for RsmA in modulating the levels of rsmB RNA. The findings reported here firmly establish that the quorum-sensing signal is channeled in E. carotovora subsp. carotovora via the rsmA-rsmB posttranscriptional regulatory system.

Abstract Ribosome biogenesis requires at least 18 putative ATP-dependent RNA helicases in Saccharomyces cerevisiae. To explore the functional environment of one of these putative RNA helicases, Dbp6p, we have performed a synthetic lethal screen with dbp6 alleles. We have previously characterized the nonessential Rsa1p, whose null allele is synthetically lethal with dbp6 alleles. Here, we report on the characterization of the four remaining synthetic lethal mutants, which reveals that Dbp6p also functionally interacts with Rpl3p, Nop8p, and the so-far-uncharacterized Rsa3p (ribosome assembly 3). The nonessential Rsa3p is a predominantly nucleolar protein required for optimal biogenesis of 60S ribosomal subunits. Both Dbp6p and Rsa3p are associated with complexes that most likely correspond to early pre-60S ribosomal particles. Moreover, Rsa3p is co-immunoprecipitated with protA-tagged Dbp6p under low salt conditions. In addition, we have established a synthetic interaction network among factors involved in different aspects of 60S-ribosomal-subunit biogenesis. This extensive genetic analysis reveals that the rsa3 null mutant displays some specificity by being synthetically lethal with dbp6 alleles and by showing some synthetic enhancement with the nop8-101 and the rsa1 null allele.

ABSTRACT In the opportunistic human pathogen Pseudomonas aeruginosa, quorum sensing (QS) is crucial for virulence. The RsaL protein directly represses the transcription of lasI, the synthase gene of the main QS signal molecule. On the basis of sequence homology, RsaL cannot be predicted to belong to any class of characterized DNA-binding proteins. In this study, an in silico model of the RsaL structure was inferred showing that RsaL belongs to the tetrahelical superclass of helix-turn-helix proteins. The overall structure of RsaL is very similar to the N-terminal domain of the lambda cI repressor and to the POU-specific domain of the mammalian transcription factor Oct-1 (Oct-1 POUs). Moreover, residues of Oct-1 POUs important for structural stability and/or DNA binding are conserved in the same positions in RsaL and in its homologs found in GenBank. These residues were independently replaced with Ala, and the activities of the mutated variants of RsaL were compared to that of the wild-type counterpart in vivo by complementation assays and in vitro by electrophoretic mobility shift assays. The results validated the RsaL in silico model and showed that residues Arg 20, Gln 38, Ser 42, Arg 43, and Glu 45 are important for RsaL function. Our data indicate that RsaL could be the founding member of a new protein family within the tetrahelical superclass of helix-turn-helix proteins. Finally, the minimum DNA sequence required for RsaL binding on the lasI promoter was determined, and our data support the hypothesis that RsaL binds DNA as a dimer.

RsmA is a posttranscriptional regulator that controls virulence factor production and biofilm formation. In this study, we investigated the expression and activity ofrsmAand the protein that it encodes, RsmA, inP. aeruginosamucAmutant strains, which are common in chronic infections. We determined that AlgU regulates a previously unknownrsmApromoter inP. aeruginosa. Western blot analysis confirmed that AlgU controlsrsmAexpression in both a laboratory strain and a clinical isolate. RNase protection assays confirmed the presence of tworsmAtranscripts and suggest that RpoS and AlgU regulatersmAexpression. Due to the increased amounts of RsmA inmucAmutant strains, a translational leader fusion of the RsmA target,tssA1, was constructed and tested inmucA,algU,retS,gacA, andrsmAmutant backgrounds to examine posttranscriptional activity. From these studies, we determined that RsmA is active inmucA22mutants, suggesting a role for RsmA inmucAmutant strains. Taken together, we have demonstrated that AlgU controlsrsmAtranscription and is responsible for RsmA activity inmucAmutant strains. We propose that RsmA is active inP. aeruginosamucAmutant strains and that RsmA also plays a role in chronic infections.IMPORTANCEP. aeruginosacauses severe infections in immunocompromised patients. The posttranscriptional regulator RsmA is known to control virulence and biofilm formation. We identify a newrsmApromoter and determine that AlgU is important in the control ofrsmAexpression. MutantmucAstrains that are considered mucoid were used to confirm increasedrsmAexpression from the AlgU promoter. We demonstrate, for the first time, that there is RsmA activity in mucoidP. aeruginosastrains. Our work suggests that RsmA may play a role during chronic infections as well as acute infections.

The entire nucleotide sequence of the rsaA gene, encoding the paracrystalline surface (S) layer protein (RsaA) of Caulobacter crescentus CB15A, was determined. The rsaA gene encoded a protein of 1026 amino acids, with a predicted molecular weight of 98 132. Protease cleavage of mature RsaA protein and amino acid sequencing of retrievable peptides yielded two peptides: one aligned with a region approximately two-thirds the way into the predicted amino acid sequence and the second peptide corresponded to the predicted carboxy terminus. Thus, no cleavage processing of the carboxy portion of the RsaA protein occurred during export, and with the exception of the removal of the initial methionine residue, the protein was not processed by cleavage to produce the mature protein. The predicted RsaA amino acid profile was unusual, with small neutral residues predominating. Excepting aspartate, charged amino acids were in relatively low proportion, resulting in an especially acidic protein, with a predicted pI of 3.46. As with most other sequenced S-layer proteins, RsaA contained no cysteine residues. A homology scan of the Swiss Protein Bank 17 produced no close matches to the predicted RsaA sequence. However, RsaA protein shared measurable homology with some exported proteins of other bacteria, including the hemolysins. Of particular interest was a specific region of the RsaA protein that was homologous to the repeat regions of glycine and aspartate residues found in several proteases and hemolysins. These repeats are implicated in the binding of calcium for proper structure and biological activity of these proteins. Those present in the RsaA protein may perform a similar function, since S-layer assembly and surface attachment requires calcium. RsaA protein also shared some homology with 10 other S-layer proteins, with the Campylobacter fetus S-layer protein scoring highest. Background: Patients with brachial plexus avulsion (BPA) usually experience phantom sensations and phantom limb pain (PLP) in the deafferented limb. It has been suggested that evoking the sensation of touch in the deafferented limb by stimulating referred sensation areas (RSAs) on the cheek or shoulder might alleviate PLP. However, feasible rehabilitation techniques using this approach have not been reported. Objective: The present study sought to examine the analgesic effects of simple electrical stimulation of RSAs in BPA patients with PLP. Methods: Study 1: Electrical stimulation of RSAs for 60 minutes was conducted for six BPA patients suffering from PLP to examine short-term analgesic effects. Study 2: A single case design experiment was conducted with two BPA patients to investigate whether electrical stimulation of RSAs was more effective for alleviating PLP than control electrical stimulation (electrical stimulation of sites on side opposite to the RSAs), and to elucidate the long-term effects of electrical stimulation of RSAs. Results: Study 1: Electrical stimulation of RSAs evoked phantom touch sensations in the deafferented limb, and significantly alleviated PLP (p < 0.05). Study 2: PLP was alleviated more after electrical stimulation on RSAs compared with control electrical stimulation (p < 0.05). However, the analgesic effects of electrical stimulation on RSAs were observed only in the short term, not in the long term (p > 0.05). Conclusions: Electrical stimulation of RSAs not only evoked phantom touch sensation but also alleviated PLP in the short term. The results indicate that electrical stimulation of RSAs may provide a useful practical rehabilitation technique for PLP. Future studies will be required to clarify the mechanisms underlying immediate PLP alleviation via electrical stimulation of RSAs.

Salmonella contamination of pooled raw shelled egg mix (RSEM) used as an ingredient in lightly cooked or uncooked foods and high-risk kitchen hygiene practices in United Kingdom food service establishments using RSEM were investigated. Samples were collected from 934 premises. Salmonella was found in 1 (0.13%) of 764 RSEM samples, 2 (0.3%) of 726 samples from surfaces where ready-to-eat foods were prepared, and 7 (1.3%) of 550 cleaning cloths. Poor RSEM storage and handling practices were highlighted. Workers in 40% of the premises sampled failed to use designated utensils when RSEM was added to other ingredients, workers in 17% of the premises did not clean surfaces and utensils thoroughly after use with RSEM and before preparing other foods, only 42% of workers washed and dried their hands after handling eggs or RSEM, workers in 41% of the premises did not store RSEM at refrigeration temperature before use, and workers in 8% of the premises added RSEM to cooked rice at the end of cooking when preparing egg fried rice. Take-away premises, especially those serving Chinese cuisine, were least likely to have a documented food safety management system and awareness of the key food safety points concerning the use of RSEM compared with other food service premises (P &lt; 0.0001). Food service businesses using RSEM must be aware of the continuing hazard from Salmonella, must adopt appropriate control measures, and must follow advice provided by national food agencies to reduce the risk of Salmonella infection.

Using a gene fusion approach, hybrid proteins were created by linking alkaline phosphatase (PhoA) or a cellulase reporter (ΔCenA) to four large N-terminal portions of the Caulobacter crescentus surface (S)-layer protein (RsaA; 1026 amino acids). Three of the sites (amino acids 189, 220, 315) were selected on the basis of TnphoA experiments that suggested the first 250–350 amino acids of RsaA could mediate export of PhoA from the cytoplasm while the fourth lay only 21 amino acids from the C-terminus. Expression of all fusions except rsaA(315):ΔcenA and rsaA(315):phoA was toxic to C. crescentus. None of the gene fusions were toxic when expressed by Escherichia coli DH5α, where all the hybrid proteins accumulated as inclusion bodies. The toxicity of hybrid proteins encoding 189, 220, and 1005 RsaA-derived amino acids was related to the nature of the hybrid protein itself because truncated RsaA peptides lacking their reporter domains were nontoxic. Further study of RsaA(ΔC21) showed that this and presumably other truncated RsaA derivatives were neither secreted nor prone to intracellular accumulation. Although C. crescentus tolerated the expression of rsaA(3l5):ΔcenA and rsaA(315):phoA, the encoded hybrid proteins were not exported in significant quantities from the cytoplasm. These results extend and confirm earlier work that large portions of the S-layer protein N-terminus cannot mediate export of passenger proteins from the cytoplasm and that the entire native S-layer protein may be required to properly interact with the RsaA secretion machinery.Key words: surface layer, secretion, reporter protein, Caulobacter crescentus.

RsmA is a posttranscriptional regulatory protein in Pseudomonas aeruginosa that works in tandem with a small non-coding regulatory RNA molecule, RsmB (RsmZ), to regulate the expression of several virulence-related genes, including the N-acyl-homoserine lactone synthase genes lasI and rhlI, and the hydrogen cyanide and rhamnolipid biosynthetic operons. Although these targets of direct RsmA regulation have been identified, the full impact of RsmA on cellular activities is not as yet understood. To address this issue the transcriptome profiles of P. aeruginosa PAO1 and an isogenic rsmA mutant were compared. Loss of RsmA altered the expression of genes involved in a variety of pathways and systems important for virulence, including iron acquisition, biosynthesis of the Pseudomonas quinolone signal (PQS), the formation of multidrug efflux pumps, and motility. Not all of these effects can be explained through the established regulatory roles of RsmA. This study thus provides both a first step towards the identification of further genes under RsmA posttranscriptional control in P. aeruginosa and a fuller understanding of the broader impact of RsmA on cellular functions.

To better understand RsmF binding properties, we performed parallel systematic evolution of ligands by exponential enrichment (SELEX) experiments for RsmA and RsmF. The initial target library consisted of 62-nucleotide (nt) RNA transcripts with central cores randomized at 15 sequential positions. Most targets selected by RsmA and RsmF were the expected size and shared a common consensus sequence (CANGGAYG) that was positioned in a hexaloop region of the stem-loop structure. RsmA and RsmF also selected for longer targets (≥96 nt) that were likely generated by rare PCR errors. Most of the long targets contained two consensus-binding sites. Representative short (single consensus site) and long (two consensus sites) targets were tested for RsmA and RsmF binding. Whereas RsmA bound the short targets with high affinity, RsmF was unable to bind the same targets. RsmA and RsmF both bound the long targets. Mutation of either consensus GGA site in the long targets reduced or eliminated RsmF binding, suggesting a requirement for two tandem binding sites. Conversely, RsmA bound long targets containing only a single GGA site with unaltered affinity. The RsmF requirement for two binding sites was confirmed withtssA1, anin vivoregulatory target of RsmA and RsmF. Our findings suggest that RsmF binding requires two GGA-containing sites, while RsmA binding requirements are less stringent.IMPORTANCEThe CsrA family of RNA-binding proteins is widely conserved in bacteria and plays important roles in the posttranscriptional regulation of protein synthesis.P. aeruginosahas two CsrA proteins, RsmA and RsmF. Although RsmA and RsmF share a few RNA targets, RsmF is unable to bind to other targets recognized by RsmA. The goal of the present study was to better understand the basis for differential binding by RsmF. Our data indicate that RsmF binding requires target RNAs with two consensus-binding sites, while RsmA recognizes targets with just a single binding site. This information should prove useful to future efforts to define the RsmF regulon and its contribution toP. aeruginosaphysiology and virulence.

ABSTRACT In the plant-beneficial soil bacterium Pseudomonas fluorescens CHA0, the production of biocontrol factors (antifungal secondary metabolites and exoenzymes) is controlled at a posttranscriptional level by the GacS/GacA signal transduction pathway involving RNA-binding protein RsmA as a key regulatory element. This protein is assumed to bind to the ribosome-binding site of target mRNAs and to block their translation. RsmA-mediated repression is relieved at the end of exponential growth by two GacS/GacA-controlled regulatory RNAs RsmY and RsmZ, which bind and sequester the RsmA protein. A gene (rsmE) encoding a 64-amino-acid RsmA homolog was identified and characterized in strain CHA0. Overexpression of rsmE strongly reduced the expression of target genes (hcnA, for a hydrogen cyanide synthase subunit; aprA, for the main exoprotease; and phlA, for a component of 2,4-diacetylphloroglucinol biosynthesis). Single null mutations in either rsmA or rsmE resulted in a slight increase in the expression of hcnA, aprA, and phlA. By contrast, an rsmA rsmE double mutation led to strongly increased and advanced expression of these target genes and completely suppressed a gacS mutation. Both the RsmE and RsmA levels increased with increasing cell population densities in strain CHA0; however, the amount of RsmA showed less variability during growth. Expression of rsmE was controlled positively by GacA and negatively by RsmA and RsmE. Mobility shift assays demonstrated specific binding of RsmE to RsmY and RsmZ RNAs. The transcription and stability of both regulatory RNAs were strongly reduced in the rsmA rsmE double mutant. In conclusion, RsmA and RsmE together account for maximal repression in the GacS/GacA cascade of strain CHA0.

Abstract Objective The Raven’s Standard Progressive Matrices (RSPM) is a 60-item measure of non-verbal abstract reasoning. The length of the RSPM presents a potential barrier to its use. Consequently, Bilker et al. (2012) identified a 9-item short form (RSPM-SF). The purpose of this study is to (a) provide descriptive statistics for the RSPM-SF from a non-patient sample, and (b) validate the RSPM-SF by exploring correlations with congruent and divergent cognitive measures. Method Twenty men and 23 women, aged 55 to 83 (M = 67.79), completed a test battery that included the SPM-SF and global, memory, verbal, and executive measures. Results The mean score for the RSPM-SF was 4.40 (median = 4, mode = 4, SD = 1.92). RSPM-SF correlated best with the Montreal Cognitive Assessment (r = .548, CI: .297 to .729) and poorest with letter fluency (r = .065, CI: -.456 to .127). Correlations with executive measures ranged from a low of .308 (Tower Test; CI: .019 to .549) to a high of .470 (Trail Making Test Part-B; CI: .197 to .675). The average correlation with executive scores was .412. Average correlation with learning (both verbal and non-verbal) was .435, and with recall was .296. Conclusions RSPM historically was viewed as a non-verbal global estimate of cognitive ability. Present findings support using the RSPM-SF as a global measure, as it correlated well with both verbal and nonverbal, and executive and memory tests. Descriptive data suggested that the RSPM-SF items ranged from easy to difficult.
