Abstract:
Three experiments with captive ruffed grouse (Bonasa umbellus) demonstrated that dietary metabolizable energy (ME) can be predicted from neutral detergent solubles, total phenols, and percent acorns of grouse diets. The weight of the fat attached to the gizzard was highly correlated with percent carcass fat in 82 grouse and was judged a useful index of body condition. Crop contents of 1005 grouse collected during fall and winter 1981-84 in Maine, New York, Wisconsin, Washington, Virginia, West Virginia, Indiana, Ohio, North Carolina, and Georgia were used to make regional comparisons of food habits and diet quality. The ME of crop contents was predicted from chemical composition. Evergreen leaves of woody plants were the most common late winter forages of grouse in southeastern states, whereas buds, twigs, and catkins were the most common late fall and winter forages in diets of grouse in northern states. Winter diets in the Southeast tended to have higher levels of predicted ME than diets in the North; however, southeastern diets tended to have higher levels of total phenols and lower levels of protein than typical northern diets. Evergreen leaf forages had higher levels of tannin phenols than buds, twigs, and catkins. Dietary ME appeared adequate in both the North and the Southeast, but low levels of protein and high levels of tannins may result in poorer quality winter diets along the southeastern edge of the range of the ruffed grouse. Acorns comprised 63% of the crop contents of 22 grouse collected in Virginia in March and April 1982, the spring following a year of high acorn production. Leaves and flowers of herbaceous forbs were the primary forages of 41 grouse collected in spring 1983 and 1984. Body fat levels were greater for females than males and declined from March to April. Fat declines appeared to be related to breeding activities. Evergreen leaves were the most abundant forages available to grouse in late winter on a study site in southwestern Virginia. Biomass of high quality herbaceous leaves-was insufficient to meet estimated energy requirements of grouse in late winter, indicating a need for a dietary shift to low quality evergreen leaves.

Abstract:
The osteoclast ruffled border is a highly convoluted, complex membrane that is necessary for bone resorption. It is thought to form following mass lysosomal fusion with the boneapposing plasma membrane and vesicular trafficking is vital for its formation and function. The aim of this PhD was to better understand the ultrastructure, formation and function of the ruffled border using TEM and advanced imaging techniques. Ruffled border reformation following calcitonin treatment was visualised and the stages of ruffled border formation were described. Ruffled borders in healthy and osteopetrotic osteoclasts were also imaged by TEM and characterised using a morphological grading system. The key findings of this thesis are as follows: (1) vacuoles, not lysosomes, are the primary contributors of membrane to the ruffled border and the membrane projections of the ruffled border form passively as a consequence of channel formation, not actively by membrane folding, (2) extracellular vesicles are located, and appear to be released, at the ruffled border. Various functional aspects of the ruffled border were also investigated. Vesicles near the ruffled border were identified and characterised by immunoelectron microscopy based on their content and morphology. We found no morphological defects in ruffled borders in mice deficient in Plekhm1. In osteoclasts derived from patients with a SNX10 mutation, we found that while the cells retained the capacity to form well-developed ruffled borders, they did so less often than healthy control osteoclasts. Importantly, we observed that even in a population of healthy osteoclasts, ruffled border morphology is highly heterogeneous because they are at different stages in the resorption cycle. In conclusion, the data in this thesis provide novel findings, previously unseen details regarding how resorbing osteoclasts interact with the bone surface, and have revealed unique insights into ruffled border morphology, formation and the vesicles with which it interacts.

Nel mercato globale la concorrenza oggi è agguerrita più che mai, perciò per essere competitivi occorre saper sfruttare ogni vantaggio a proprio beneficio. Uno degli aspetti cruciali, che secondo la filosofia kaizen, può essere sempre migliorato è l’efficienza globale di impianto (O.E.E.) la quale riassume in sé 3 diversi concetti: disponibilità, performance e qualità. L’azienda vinicola toscana Ruffino srl ha iniziato recentemente a prestare attenzione all’analisi di questo indicatore e ha voluto così intraprendere un percorso di ottimizzazione dell’efficacia e dell’efficienza produttiva della linea 1 dello stabilimento di Pontassieve (FI). In tale realtà, un peso negativo importante nel calcolo dell’O.E.E. si è confermato essere attribuibile al fattore disponibilità, impattata soprattutto dalle perdite di tempo dovute alla moltitudine di cambi formato previsti. Il presente elaborato si prefigge dunque di illustrare l’implementazione della tecnica “Single Minute Exchange of Dies” volta alla drastica ma sostenibile diminuzione dei tempi di setup. Il lavoro svolto ha previsto fasi di raccolta dati per i 3 reparti della linea in esame, importanti momenti di analisi delle operazioni di attrezzaggio tramite la stretta collaborazione con gli operatori e la formulazione di svariate proposte di miglioramento, in parte già realizzate. In riferimento al FY16, a valle del completamento dell’intero progetto esposto, si sarebbe potuto registrare un ritorno economico in termini di saving cost pari a circa 80 mila € dovuto ad un incremento della capacità produttiva di circa 28h e alla riallocazione dei costi fissi su un maggior numero di casse prodotte. Similmente l’incremento dell’O.E.E. per lo stesso periodo sarebbe potuto essere pari a 2,24%. Tali valori testimoniano la potenza dello S.M.E.D. applicato ad una linea di imbottigliamento e anche se rispecchiano una situazione passata danno idea dei benefici di cui l’azienda potrà godere per scenari futuri analoghi.

Abstract:
Feeding trials.showed that ruffed grouse (Bonasa umbellus) performed well on diets containing 20% Christmas hollyfem (Polystichum acrostichoides) or mountain laurel (Kalmia latifolia), but diets containing 40% of these forages resulted in lower protein and energy intake and the Christmas hollyfem diet caused a loss of body mass. Grouse were not able to maintain themselves solely on evergreen leaves. Glucuronide excretion was greatest for the 40% mountain laurel diet. Ornithine conjugate excretion was greatest for a diet with 40% deciduous leaves. Sulfate excretion did not vary among diets. Intake rate of leaves was an asymptotic function of bite size when the density of bites did not limit intake. Intake rate of leaves decreased at plants densities < 322 plants/m2. The maximum intake rate of leaves was 25% of the intake rate of aspen buds observed in wild grouse (Huempfuer and Tester 1988). Intake rate of raisins was an asymptotic function of bite size and was 20 times greater than the intake rate of leaves. Ruffed grouse in the Southeast must forage for> 100 min/day under ideal conditions to satisfy energy requirements.

Abstract:
Predation accounts for the majority of ruffed grouse mortality beyond the brooding period throughout grouse range. Most studies of ruffed grouse ecology have taken place in the central portion of the species range including the Great Lakes states and southern Canada where aspen (Populus tremuloides) is dominant. Populations in the central range of grouse distribution exhibit 10-year cyclic patterns of decline that have been associated with the invasions of boreal predators such as great horned owls and northern goshawks during crashes in small mammal populations. I completed an accuracy assessment of field sign to determine the role of scavenging as it related to diagnosing causes of proximate mortality. Potentially large numbers of grouse are scavenged after death, which can alter or completely falsify the reported cause of death.. My results indicated that stand-level and micro-site habitat variables did not influence scavenger detection of ruffed grouse carcasses (P > 0.05). Scavenging was limited to 24 of 64 carcasses and was entirely attributed to mammalian species. Scavenging behavior was related to the condition of the carcass following death (P = 0.003) and the ambient temperature (P = 0.01). As the temperature increased and as a carcasses entrails and muscle became more exposed, the probability of being scavenged increased. Nineteen percent of whole carcasses placed in the field were scavenged and would have been attributed to mammal predation based on the field sign; 56% of 32 mock avian kills were scavenged would have been attributed to mammal predation. I related indices of predators and weather patterns to trends in ruffed grouse predation. Using data collected at 10 study sites between February, 1997 and December, 2000, I compared predation rates, and animal and weather indices to predation rates, across sites, years, regions, seasons and month combinations pooled across years. Avian predators were the primary predators of ruffed grouse in the Appalachian region (50% of all predation). Predation rates on ruffed grouse were highest in fall (8.3%), due to high predation on juveniles, and spring (7.4%) in association with raptor migration and pre-breeding activity of grouse. Predation patterns and predator abundance did not indicate the occurrence of predator invasions during the years of this study. Predation rates on ruffed grouse were positively related to the presence of rain and negatively related to the average low temperature and number of rabbits and squirrels observed per hour. Observations of owls and Cooper's hawks per hour were correlated with predation rates on grouse while those of red-tailed, red-shouldered and broad-winged hawks were not.

Abstract:
The tense conflict over sage grouse management in the West, where livelihoods have been pitted against the possibility of an endangered species listing, has been ongoing for many years and has been described as being as tense as the spotted owl conflict in the Northwest in the 1990s. This research is designed to highlight the different frames or narratives within the sage grouse debate in Sublette County, Wyoming while exploring a resurging research methodology. Q methodology, a method intended to identify distinct viewpoints within a sample was employed to understand the different narratives among these conflict parties. The Q method suggested three distinct viewpoints or knowledge communities existed within the sample: ultra locals, classic biologists and harmonizers. Ultra locals largely consisted of ranchers (75%) and others dependent on the land for their livelihood and showed a strong preference for local county management that included local information. The narratives of the classic biologists, a group consisting solely of biologists working for agencies, consulting firms and conservation organizations, preferred that science and research point the way to a solution. Finally, agency biologists and energy industry employees made up the final group identified, the harmonizers. This group favored working with all stakeholders to work together to build a solution. A number of areas of agreement including the lack of support for an ESA listing, and disagreement such as the role of predators on sage grouse populations were highlighted. To move forward on the conflict, this research suggests that instead of pursuing issues that may only serve to increase the conflict, such as issues of predators or sources of knowledge, a path forward may be found in merging the livelihood interests of ranchers with the preservation interests of biologists. Results also show that the Q method was helpful in pinpointing distinct viewpoints on sage grouse management in Sublette County; however, without the use of an in-depth interview, the Q method results may have been difficult to clearly and meaningfully interpret.

Abstract:
Natality characteristics were calculated for 3 regions in the southern Appalachians (Ridge and Valley, Alleghany Plateau, and Ohio River Valley). I report data collected in the first 2 years of a long term (> 6 years) study conducted by the Appalachian Cooperative Grouse Research Project (ACGRP). Nesting rate, pooled over all regions, was 83.6% in 1997 and 79.7% in 1998. In the 2-year period, the Alleghany Plateau reported the highest nesting rate (97.6%) while the Ohio River Valley reported the lowest rate (54.2%). Overall hen success rates were 81.5% in 1997 and 56.9% in 1998. Yearling hen success rates were as high or higher than adults. Adult hen success was 85.7% in 1997 and 48.5% in 1998; yearling hen success was 86.7% in 1997 and 82.3% in 1998. Additionally, I found a lower renest rate (8% over 2 years) in the southern Appalachians than previous studies have reported. The mean first-nest clutch size in the southern Appalachian region was considerably lower (9.5, years and regions pooled) than that reported for other portions of ruffed grouse range. Recommendations are given on how ACGRP natality data collection may be improved in upcoming years.

Ruffed grouse chick survival estimates were calculated from data collected in the first 2 years of a long term ACGRP study as well as data collected separate from ACGRP protocol. First-week chick survival estimates ranged from 0.18 to 0.32 in 1997 and 0.45 to 0.48 in 1998. Late brood season survivorship values (0.11-0.13 at week 5, 0.07 at week 10) were considerably lower in the southern Appalachians than those reported from more northern portions of ruffed grouse range. Additionally, the mean number of chicks per brood in July was lower in the southern Appalachians than that reported in the Great Lakes region during July and August. Recommendations are given on how ACGRP chick count data collection may be improved in upcoming years.

I compared micro-habitat characteristics at known brood locations with randomly selected locations to determine which characteristics are selected by ruffed grouse hens and broods in the southern Appalachians. In the first half of the brood season (weeks 1-6) hens and broods selected sites with tall, complete, vegetative ground cover. Additionally, broods selected forested sites with a well-developed canopy, rather than areas affected by large canopy gaps or openings. Higher ground cover at brood sites may have been due to a lack of midstory structure. The abundance of arthropods, fruit, and forage at brood flush sites was higher during the first few weeks of the brood season; this was possibly due to flush sites being located in open, mid-age or mature forest. Several authors have speculated that as the chicks' diet shifts from primarily arthropods to fruit and forage at approximately 3 weeks of age, the habitat selected by hens and their broods may change to accommodate this dietary shift. In my study, a change in habitat selection did not occur between weeks 3 and 4 as expected but after week 6 and may indicate the chicks' dietary shift occurs later than some have predicted.

Abstract:
In response to extra-cellular cues, cells activate signal transduction pathways to elicit a biological response. Cell surface growth factor receptors such as the Met receptor tyrosine kinase (RTK) activate signals that result in cellular proliferation, survival, migration, as well as epithelial morphogenesis. In order for signal transduction to occur, docking proteins are recruited to the activated RTK, become phosphorylated on tyrosine residues, which then serve as docking sites for the recruitment of other signaling proteins. Docking proteins function to diversify the signal by assembling multi-protein complexes. The Gab1 docking protein is the most tyrosine phosphorylated protein upon Met receptor activation and is required for Met mediated signaling and biology.
Gab1 belongs to a family of docking proteins including the highly related Gab2 protein. Gab1 promotes signals for epithelial morphogenesis downstream of the Met receptor, however Gab2 is unable to do so. Insertion of the Gab1 Met binding Motif (MBM) which confers direct binding to the Met receptor, as well as membrane targeting of Gab2 is sufficient to switch the capacity of Gab2 to activate the morphogenic program, cell scatter and lamellipodia formation. This is achieved via activation of sustained signaling pathways, and redistribution of the Gab protein, and associated molecules to sites of lamellipodia formation at the peripheral edge of the cell.
Activation of the Met RTK, promotes the formation of dorsal ruffles on the apical surface of epithelial cells. The Met receptor, Gab1 and Gab1 associated molecules Shp2, Crk, and p8S subunit of PI3K, are localized to these structures, however only the Gab1erk complex is required to drive dorsal ruffle formation. Gab1 is required for Met induced dorsal ruffles as well as downstream the PDGF and EGF RTKs. These are a signaling micro-environment which results in enhanced receptor degradation. Inhibition or enhancement of Met mediated dorsal ruffle formation correlates with receptor stability.
Dorsal ruffle formation downstream of Met requires the enzymatic activity of PI3K and PLCgamma, both enzymes that metabolize PIP2, and form complexes with Gab1 downstream of Met. PLCgamma and the PIP3 lipid product of PI3K are co-localized with Gab1 in dorsal ruffles. Gab1 engages with elements of the cytoskeleton, actin and cortactin, providing a link between growth factor signaling and remodeling of the actin cytoskeleton. Gab1 is localized to membrane protrusions of the basal surface in organoid cultures and is required for actin protrusions of the basal surface of breast cancer cells.

I addressed the effects of landscape characteristics on ruffed grouse (Bonasa umbellus) home range size and movement, and examined grouse selection for specific landscape characteristics and cover types. Grouse home ranges and movement patterns derived from telemetry data gathered from fall 1996 through fall 1998 were overlaid onto a GIS database of Clinch Mtn. Wildlife Management Area, VA. This database was developed from GPS data and LANDSAT thematic mapping imagery (30 m pixel scale) and was composed of 22 cover types. Landscape metrics were calculated using FRAGSTATS/ARC, and multiple regression was used to relate changes in home range size and movement to these metrics. I used Wilcoxon signed-rank tests to compare the values of landscape metrics calculated for each home range to those calculated for the area encompassed by the home range plus a surrounding 300 m buffer. I used Wilcoxon rank-sum tests to compare the values of landscape metrics for the home ranges to the metrics calculated for 50 33 ha random plots. I used compositional analysis to test for preferential use of cover types. I developed 2 regression models (P < 0.01) relating changes in home range size to landscape characteristics, 1 model (P = 0.09) relating the distance between seasonal home range centers to landscape characteristics, and 1 model (P = 0.03) relating average daily movement to landscape characteristics. Grouse home range size increased as patch shape became more irregular and patch size and the number of different cover types per hectare increased, and decreased as the amount of high contrast edge in the landscape increased. The distance between seasonal home range centers increased as Shannon's diversity index and the average distance between patches of similar cover types increased, and decreased as the amount of high contrast edge increased. Average daily movement increased as the average distance between patches of the same cover type increased and as the percent cover of a full (~75%) rhododendron and/or laurel understory within a grouse's home range increased, and decreased as the amount of high contrast edge in a bird's home range increased. Ruffed grouse were selecting areas with high densities of smaller than average patches that were of uniform size and regular shape and contained higher than average amounts of high contrast edge. Areas containing a greater diversity of cover types than what was available in the study area also were preferred. Within these areas, clearcuts and mesic deciduous stands with a rhododendron/laurel understory were the most preferred cover types. Creating and maintaining a landscape with high densities of small patches that are of uniform size and regular (square) shape would provide the highest quality ruffed grouse habitat in this region. Several of these patches should be early successional cover to provide an abundance of high contrast edge. Rhododendron and/or laurel thickets also may be beneficial as supplemental winter cover, and mesic stands of mature hardwoods should be well interspersed with these cover types to provide supplemental food sources.

I investigated ruffed grouse (Bonasa umbellus) population ecology in the southern and central Appalachians as part of the Appalachian Cooperative Grouse Research Project (ACGRP). Several hypotheses have been offered to explain the low abundance of ruffed grouse in the region including inadequate quantity of early-successional forests due to changes in land use, additive harvest mortality, low productivity and recruitment, and nutritional stress. Through the cooperative nature of the ACGRP, researchers tracked >3,000 ruffed grouse between October 1996 and September 2002 and gathered data on reproduction, recruitment, survival, and mortality factors. As part of the ACGRP My objectives were (1) estimate reproductive rates, (2) estimate survival and cause-specific mortality rates, (3) determine if ruffed grouse harvest in the Appalachian region is compensatory, and (4) estimate ruffed grouse finite population growth. Ruffed grouse population dynamics in the Appalachian region differed greatly from the core of ruffed grouse range. In general, ruffed grouse in the Appalachian region had lower productivity and recruitment, but higher survival than reported for populations in the Great Lakes and southern Canada. However, within the southern and central Appalachian region, ruffed grouse population dynamics differed between oak-hickory and mixed-mesophytic forest associations. Productivity and recruitment were lower in oak-hickory forests, but adult survival was higher than in mixed-mesophytic forests. Furthermore, ruffed grouse productivity and recruitment were more strongly related to hard mast (i.e., acorn) production in oak-hickory forests than in mixed-mesophytic forests. The leading cause of ruffed grouse mortality was avian predation (44% of known mortalities). Harvest mortality accounted for only 12% of all known mortalities and appeared to be compensatory. Population models indicate ruffed grouse populations in the Appalachian region are declining, but estimates vary greatly stressing the need for improved understanding of annual productivity and recruitment. We posit ruffed grouse in the Appalachian region exhibit a clinal population structure and changes in life-history strategies due to gradual changes in the quality of food resources, changes in snow fall and accumulation patterns, and predator communities. Recommendations are presented for habitat and harvest management and future research and management needs.

Cell-substrate interactions influence various cellular processes such as morphology, motility, proliferation and differentiation. Actin dynamics within cells have been shown to be influenced by substrate stiffness, as NIH 3T3 fibroblasts grown on stiffer substrates tend to exhibit more prominent actin stress fiber formation. Circular dorsal ruffles (CDRs) are transient actin-rich ring-like structures within cells, induced by various growth factors, such as the platelet-derived growth factor (PDGF). CDRs grow and shrink in size after cells are stimulated with PDGF, eventually disappearing ten of minutes after stimulation. As substrate stiffness affect actin structures and cell motility, and CDRs are actin structures which have been previously linked to cell motility and macropinocytosis, the role of substrate stiffness on the properties of CDRs in NIH 3T3 fibroblasts and how they proceed to affect cell behavior is investigated. Cells were seeded on Poly-dimethylsiloxane (PDMS) substrates of various stiffnesses and stimulated with PDGF to induce CDR formation. It was found that an increase in substrate stiffness increases the lifetime of CDRs, but did not affect their size. A mathematical model of the signaling pathways involved in CDR formation is developed to provide insight into this lifetime and size dependence, and is linked to substrate stiffness via Rac-Rho antagonism. CDR formation did not affect the motility of cells seeded on 10 kPa stiff substrates, but is shown to increase localized lamellipodia formation in the cell via the diffusion of actin from the CDRs to the lamellipodia. To further probe the influence of cell-substrate interactions on cell behavior and actin dynamics, a two dimensional system which introduces a dynamically changing, reversible and localized substrate stiffness environment is constructed. Cells are seeded on top of thin PDMS nano-membranes, and are capable of feeling through the thin layer, experiencing the stiffness of the polyacrylamide substrates below the nano-membrane. The membranes are carefully re-transplanted on top of other polyacrylamide substrates with differing stiffnesses. This reversible dynamic stiffness system is a novel approach which would help in the investigation of the influence of reversible dynamic stiffness environments on cell morphology, motility, proliferation and differentiation in various cells types.

Au-delà des nombreuses informations archéologiques, l’étude de ces occupations a permis d’engager une réflexion sur les protocoles méthodologiques de détection et de caractérisation de ce type de campements situés en fond de vallée. Le gisement de Pont-sur-Yonne, fouillé plus sommairement, a révélé des occupations du Mésolithique ancien et moyen. La fouille intégrale de la petite occupation ancienne appartenant à la seconde moitié du Préboréal, a livré des informations de premier plan sur les questions palethnographiques grâce en particulier aux remontages très complets des éléments lithiques. La mise en application de modes de représentation originaux, a ouvert de nouvelles perspectives en particulier sur la question de la structuration spatiale. La proposition de modèles d’organisation spatiale, avec l’unité d’activités simple et l’unité d’activités à espace domestique permet de poser les premières bases formalisées d’une réflexion encore récente. La définition de modules types constitués par ces modèles autorise dans un second temps une approche de la variabilité de la forme du campement qui revêt un caractère plus ou moins extensif et un niveau d’imbrication plus ou moins fort. En définitif, les occupations ici étudiées traduisent dans leurs différentes composantes, des fréquentations de courte durée en lien avec un régime de très forte mobilité des dernières population de chasseurs-cueilleurs, proche du système économique défini sur les principes du foraging
The developpment of survey archeology explains the discovery of huge mésolithics open air sites, like Ruffey-sur-Seille and Choisey in Jura. More than numerous archeological data, the study of these occupations allowed to reflect about the methodologicals protocols of detection and characterisation from this kind of encampments located in the bottom of the valley. The Pont-sur-Yonne site, summarily excavated, revealed low and middle Mesolithic’s occupations. The entirely excavation of the small old occupation belonging to the second half of preboreal period, procured mains informations about the palethnographical question, in particular with the very completes refittings of lithics artifatcs. The application of originals way of representation, opened news perpectives, in particular, about the spatial organisation’s question. The proposal of spatial organisation’s patterns with the simple unit of activities and the unit of activities with domestic space, bring the first bases of a too young reflection. The definition of standard modulus with these patterns, allow in a second time, the consideration of the variability from the encampments form that will be more or less extensive and overlapped. Finaly, the occupations here analysed, reveal in their differents compositions, shorts frequentings related to a very high mobility of these last hunter-gathering populations, nearby of the economic system based on the foraging’s principles

By studying how captive animals use outdoor space, we can determine which areas fulfill their biological and behavioral needs to improve enclosure design for animal welfare and visitor experience. The aim of the study was to determine the differential use of outdoor space in a mixed exhibit of captive ring-tailed lemurs (Lemur catta) and red ruffed lemurs (Varecia rubra), and to assess both the effect of adding environmental enrichment and possible visitor and meteorological effects on the lemurs’ use of space and behavior. For one month, five-minute interval scan sampling was used to record lemur location and behavior, as well as the meteorological conditions and presence of zoo visitors in the lemurs’ outdoor enclosure. That baseline was followed by four alternating two-week periods of food or structural enrichment, both with and without visitors. Enrichment increased the lemurs’ use of the enriched sectors, decreased resting, and increased locomotion. Structural enrichment increased their exploratory behaviors and stimulated arboreal locomotion. Both species preferred sunny and warm conditions for sunbathing and resting, while moving more around the enclosure in cooler and cloudier weather. The visitor effect on the lemurs’ use of space was weaker than the effects of enrichment and meteorological conditions. The availability of areas to hide and possible habituation to humans may have played a role in the reduced visitor effect. The results of the study showed inter-specific differences in the lemurs’ use of space and behavior and a complex combined effect of enrichment, visitor presence and meteorological conditions.

L’étude sédimentologique de cinq sites préhistoriques, situés dans différents contextes géomorphologiques et occupés à diverses reprises entre le début du mésolithique et la fin de l’Age du fer, aboutit à une classification des faciès de dépôt en fonction des processus sédimentaires (anthropiques ou naturels) dont ils résultent. Deux grandes familles de dépôt sont différenciées : les faciès d'occupation, générés par l'homme, et les faciès d'abandon, inhérents aux processus sédimentaires ou post-sédimentaires naturels. Les premiers apportent des informations sur les aménagements réalisés, sur les activités pratiquées et sur la durée approximative des fréquentations, tandis que les seconds permettent de discuter de la fréquence et de la durée des arrêts d'occupation. A l'appui du cadre chrono-culturel, ces données sédimentologiques permettent de proposer des scenarios d'occupation pour les différents sites étudiés. Elles participent ainsi aux recherches archéologiques visant à définir les composantes socio-économiques des cultures préhistoriques. Par exemple : les implantations durables en habitat sont distinguées des fréquentations intermittentes liées à des activités saisonnières ou à l'utilisation des sites comme refuge ; des interprétations sont avancées sur l'évolution de la pratique de l'élevage, de l'agriculture, sur les rites funéraires ou sur les techniques de construction. Par ailleurs, les sites favorables à l'enregistrement sédimentaire d'événements naturels montrent qu'il existe une relation entre les variations climatiques régionales et la nature des dépôts accumulés. In last several years the amount of physical activities during our day time seriously decreased. That phenomenon was caused by reduction of physically demanding works in our jobs and sitting way of spending free time. The result of the previous fact is population fitness worsening and consequently worsening of the labour productivity and health condition. The fitness is tightly connected with the relationship between a man and a movement and sporting activities. It should become the important part of the healthy life style and influence the formation of proper manners of every single man. The fitness is one of the markers of the cardiovascular system condition. And it is considered to be the certain sum of preconditions for optimal reaction on the stress of whatever cause. The regular training, the physical activity and the daily reasonable energetic income are the safest and the least economically demanding ways how to prevent civilisation diseases. The first task of the theses was to assess the fitness of women students of The University of South Bohemia. The fitness was assessed on the base of Ruffier?s test. The second task was to compare the fitness of several groups of students which represented every single faculty of The University of South Bohemia. The both tasks were completed. Two hypotheses were settled and confirmed in purpose to reach the goal of the theses. H1: The numerological value of the Rufier?s test of women students of the Faculty of Education is lower than the result of women from the other faculties. H2: Women students whose result of Ruffier?s test is 0-5 are spending more time by performing physical activity than other students. In this theses, Ruffier?s test as a non-direct way of assessing the fitness has been chosen to confirm the both hypotheses. Ruffier?s test is a simple test to asses the fitness which uses changes of the hearth rate before and after exercising. The test is based on the general premise that the hard body exercising which last for a longer period of time is connected with the rising of heart rate. The heart rate was recorded by sport tester. Heart rate values were set into the pattern for counting the index of Ruffier?s test and consequently classified. Averages of the Ruffier?s index of each faculty were measured and compared. The questionnaire was ad to the Ruffier?s index for better assessing the fitness of students. The questionnaire was composed by thirteen questions. Some of questions were of open and some of close type. The questionnaires were anonymous. Hypothesis H1 was confirmed. The average value of the Ruffier?s index for women students of the pedagogical faculty was 10,7. And the average value of the Ruffier?s index for women students of other faculties was 13. It means the average fitness of women students of pedagogical faculty is better than the average fitness of women students of other faculties. Hypothesis H2 confirmed the correlation between the time spending by physical exercising and the better fitness. The more times weekly women students exercise for more than 30 minutes a day the lower value of the Ruffier?s index they have. The assumption of the average values of the Ruffier?s index of those women from 0 to 5 was overstated. Women student in this project had the average values from 10 to 15 which mean worse values of fitness. Despite the worse values of Ruffier?s index, the correlation between the spending time by exercising and better fitness was confirmed. To sum up, there is a recommendation to compensate the simple monotone physical activity by wide spectrum of physical activities. The sport should be introduced into the life style. The person with a good fitness has less problems with movement, can work for a longer time without break, is strong and capable to fulfil everyday tasks with reserves. The process of improving the fitness is a run for a long distance but it should be the goal of all people no matter of how old they are.
