Full textAdipocytes are a cell population largely located in the human bone marrow cavity. In this specific microenvironment where adipocytes can interact with a variety of different cells, the role of fat is mainly unknown. To our knowledge, this report is the first to characterize mature adipocytes isolated from human bone marrow (BM-A) molecularly and functionally to better understand their roles into the hematopoietic microenvironment. Healthy BM-A were isolated after collagenase digestion and filtration. We studied the morphology of BMA, their gene expression and immunophenotypic profile and their functional ability in the hematopoietic microenvironment, comparing them with adipocytes derived from adipose tissue (AT-A). BM-A showed a unilocular lipid morphology similar to AT-A and did not lose their morphology in culture; they showed a comparable pattern of stem cell-surface antigens to AT-A. In line with these observations, molecular data showed that BM-A expressed some embryonic stem cells genes, such as Oct4, KLf4, c-myc, Gata4, Tbx1, and Sox17, whereas they did not express the stem cell markers Sox2 and Nanog. Moreover, BM-A had long telomeres that were similar to bone marrow mesenchymal stem cells. Notably, BM-A supported the survival and differentiation of hematopoietic stem cells in long-term cultures. These results showed that BM-A are stromal cells with a gene expression pattern that distinguished them from AT-A. BM-A showed stem cell properties through their hematopoietic supporting function, which was certainly linked to their role in the maintenance of the bone marrow microenvironment. Depending on specific demands, BM-A may acquire different functions based on their local environment.

Full textThe work analyses the outsourcing processes on the point of view of industrial relations, with the purpose to verify the effectiveness of the collective bargaining provisions to “manage” these processes. The dissertation moves from the assumption that the second-level agreements should be implemented, considering the applicability to all employees and the greater proximity to the interests concerned. The major part of the analysis revolves around the service contracts and the heterogeneous related provisions of the national collective labour agreements (such as the ban on outsourcing, the information to trade unions, the entitlement to certain treatments or to hold down the employment relationship), which should be developed by company or territorial level agreements. Then the dissertation focuses on the transfer of business, highlighting the role of the second-level bargaining, which could be able to find a compromise between competitiveness and social protection, as well as to control the compliance of the transactions. The work also considers the staff leasing contract, hoping for a system of collective representation and negotiation which includes the temporary workers. Finally, with reference to the so called “working project consulting agreements”, the author observes that the trade unions should give to the consultants a major collective protection.

Full textThe thesis reflects on the need to rethinking the anthropological sciences in the direction of a public use and of a public recognition for this disciplines by the world outside academy. The debate is introduced from the different positions shared in the international context about the use of applied anthropology. In the United States the reflection is developed by the proposal of public anthropology, that has some differences respect the traditional figure of the European public intellectual. The dissertation highlight the various positions about ethical, methodological and epistemological standpoint application of anthropology. First of all, different methodologies developed from the tradition of Applied Anthropology are taken into account, from the first proposals dating back to the second postwar period. Secondly the analysis focus on the relationship between anthropology, colonial power and the armed forces until the recent case of anthropologists embedded in the Human Terrain System. Different forms of anthropological engagement shows researchers involved in different roles up to the extreme case in which they becoming activists in the same causes of their partners. The issue about the role played by the researcher, and the role that the fieldwork attribute to him, is deepened through the category of implication drawn up in the French context. Through some fieldwork experiences, concrete forms of intervention in the Italian overviews are presented to shift the focus on the anthropologists’ action possibility in the society. Finally, the dissertation present the Italian ongoing debate about the crisis that the discipline is passing through, and the resulting work in progress for the constitution of a national association for professional anthropology.

Full textMy research project focused on the role of ultrasound in dogs and cats with oncologic diseases and it included two studies. The aim of the first was to evaluate the relationship between the sonographic honeycomb pattern (HCP) of the spleen and cyto-histological diagnosis in cats, and to assess the influence of the type of transducer on HCP visualization. Abdominal ultrasounds of cats with HCP were reviewed to record spleen size, shape and margin appearance, other parenchymal alterations, lymphadenopathy. Images acquired with high-frequency and curvilinear transducers were compared to see if HCP was more consistent on high resolution images. A retrospective review of the corresponding splenic cyto-histopatologic samples was performed. Based on our findings, HCP in cats is not predictive of lymphoma and it can be associated with both benign and malignant disorders. The use of high-frequency transducers is recommended to properly recognize HCP or subtle changes in splenic parenchyma. The second study aimed to compare performances of contrast enhanced ultrasound (CEUS) and bidimensional ultrasound (US) guided fine needle aspiration (FNA) of abdominal and thoracic masses and to determine the effect of the following independent predictors: location, size, presence of avascular areas within the mass, operator’s experience, on the adequacy of the cytological sample. Dogs and cats with masses were retrospectively selected and included into 2 groups: 43 patients were included in the CEUS-guided group and 39 in the US-guided group. The cytological samples were reviewed by two pathologists and a final interpretation of the cytological adequacy was provided. According to a univariate and multivariate logistic regression analysis both CEUS and US guidance are suitable for FNA. Mass location and presence of avascular areas can also affect the adequacy. Thus, in case of large masses CEUS should be performed before FNA as presence of avascular areas may lead to inadequate sampling.

Full textIn Neoclassical finance theory, there is no role for investor sentiment in valuation, markets are efficient and all movements in stock prices rationally reflect changes in cash flows or discount rates. In sharp contrast, the behavioral finance literature posits that investor sentiment and limits to arbitrage play a role in the determination of asset prices which is independent of market fundamentals. Real Estate Investment Trusts (REITs) are unique in that the pricing of the asset class parallels two markets. Specifically, a dual asset market situation exist for trading real estate assets in the private real estate market, trading properties directly, and the public real estate market for trading REIT shares that provides ownership of underlying properties indirectly. The contribution of this analysis to the existing literature of REIT pricing dynamic is twofold. First, this analysis address the question of whether REITs suffer from stock market sentiment following the presence of noise traders or the real market sentiment is an important force in REIT pricing. To this extent this work aims to disentangle the driving forces leading to sentiment as “rational” sentiment, related to fundamental changes and “irrational” stock market sentiment. Secondly, this analysis is one of the first attempts to investigate how REIT-specific characteristics are related to their sensitivity to investor sentiment (“sentiment beta”), focusing the attention on the role played by the institutional investors that are usually view as rational traders by the literature. Several econometric issues arise in the measurement of the investor sentiment index and the time series analysis; therefore, Principal Component Analysis (PCA) and Vector Error Correction Model (VECM) are used to address those issues. The remainder of this work is structured as follow: after an introduction of the role of investor sentiment in the real estate market, chapter 2 analyzes more in detail the investor sentiment literature in the finance and real estate field; subsequently, chapter 3 discusses the construction of the sentiment indices; the empirical analysis of the influence of sentiment on the dynamic pricing of REITs in the US Market is presented in chapter 4; chapters 5 focus on the development of the sentiment beta measure to test the “Hard-to-Arbitrage, Difficult-to-Value Hypothesis”. Lastly, Chapter 6 ends up with the main conclusions and future research developments.

Dysfunction of many protein kinases, particularly the tyrosine kinases, has been identified in a variety of human cancers, and has just begun to be investigated in spontaneous tumours of dogs and cats. We present the results of four studies aimed at characterizing the dysregulation (aberrant immunohistochemical expression and gene mutations) of Kit receptor tyrosine kinase in feline cutaneous mast cell tumors and the overexpression of the epidermal growth factor receptor (EGFR) in feline squamous cell carcinoma and in canine primary lung cancer. The potential prognostic implications of the observed dysregulation are discussed.

Full textBackground/Aims: High-fat dietary intake and low physical activity lead to insulin resistance (IR) and nonalcoholic fatty liver disease (NAFLD)/steatohepatitis (NASH). Recent studies have shown an effect of Glucagon-like-peptide-1 (Glp1) on hepatic glucose metabolism, although the presence of Glp-1 receptor (Glp-1R) in human livers is still unclear. The aim of this study was to investigate the presence of hepatic Glp-1R and the effect of exendin-4, a Glp-1 analogue, on hepatic signaling. Methods: Expression of Glp-1R was evaluated in human liver biopsies and in the livers of high-fat diet-treated rats. The effect of exendin-4 (100nM) was evaluated in hepatic cells of rats fed 3 months with the high-fat diet. Results: Glp-1R is expressed in human hepatocytes, although reduced in patients with NASH. Similarly, in rats with NASH resulted from 3 months of the high-fat diet, we found decreased expression of Glp-1R and PPARγ, and reduced PPARα activity. Incubation of hepatocytes with exendin-4 increased PPARγ expression that exerted also an insulin-sensitizing action by reducing JNK phosphorylation. Moreover, exendin-4 increased PKA activity, Akt and AMPK phosphorylation and determined a PKA-dependent increase of PPARα activity. Conclusions: Glp1 has a direct effect on hepatocytes, by activating genes involved in fatty acid β-oxidation and insulin-sensitivity. Glp1 analogues could be a promising treatment approach to improve hepatic insulin resistance in patients with NAFLD/NASH.

Full textBackground and aims: Non-Alcoholic Fatty Liver Disease (NAFLD) is the most common form of chronic liver disease. The NLRP3-inflammasome can elicit a pro-inflammatory response when activated but it also regulates intestinal homeostasis and gut microbiota composition. The NLRP3-inflammasome has been implicated in the pathogenesis of obesity, diabetes and in the progression of chronic liver injury, but its role in NAFLD is controversial. Therefore, the aim of this study was to investigate the role of NLRP3-inflammasome in NAFLD. Methods: Nlrp3-/- and wild-type (WT) C57BL/6 mice were fed with a high-fat diet with fructose in drinking water (HFHC), or a chow diet, for 12 weeks. Results: Nlrp3-/- HFHC showed reduced energy expenditure and this led to increased body weight, higher fat mass and adipose tissue TNF-α expression. Nlrp3-/- HFHC developed more hepatic steatosis, measured by triglyceride content, compared to WT HFHC because of augmented SCD-1 activity (a regulator of hepatic lipogenesis) and PPARγ2 expression (that regulates lipid uptake and storage). Increased mitochondrial fatty acid oxidation and reduced expression of NRF2, the “master regulator” of antioxidant response, led to increased superoxide production in Nlrp3-/- HFHC. After HFHC, lack of NLRP3-inflammasome was associated with significant alteration in intestinal microbiota (higher Firmicutes/Bacteroidetes ratio, and increased levels of mucus-degrading bacteria Akkermansia and Desulfovibrio) which was associated to a “leaky” intestinal barrier. Concomitantly, hepatic TLR4 and TLR9 expression, inflammatory macrophages in the liver, and NAS score were increased in Nlrp3-/- HFHC mice, indicating increased inflammatory response and liver injury. Gut decontamination by antibiotics reduced body weight, bacterial translocation and liver injury in Nlrp3-/- HFHC mice. Conclusions: Lack of NLRP3-inflammasome is linked to severe metabolic alterations and development of NASH possibly due to translocation of bacterial products.

Full textCorporate reporting has undergone many changes. One main reason is that stakeholders see their needs change and evolve, thus making it necessary for them to express their various requests for information which, in turn, force companies to rethink their external reporting practices. Recent studies support the position that the traditional annual report does not provide stakeholders with an adequate picture of the assets and the value created by the company. In response to this, Integrated Reporting (IR) has been adopted as a solution capable of providing both a holistic view of a company’s ability to create value in a sustainable way and an integrated vision of the company from a financial and a non- financial point of view. Thus, this work aims to examine the real reasons that have pushed major Italian companies to opt for IR and, especially, to explore the benefits that can now be identified a few years beyond the first publication of an Integrated Report. To investigate this topic, an exploratory field study involving eleven Italian companies was conducted. The methodology relied on a qualitative approach, as it allows researchers to deal with complex “how” questions and, at the same time, offers a wide and complete picture of the phenomenon. The work gives senior managers an overview of Italian companies that have already developed this new form of information reporting, while providing valuable insights and an understanding of the perceived organizational benefits of implementing IR requirements. The study also contributes to the debate on managerial motives for producing an Integrated Report and how that may affect future decisions. The findings reveal a common thread among all the study participants, summarized by the completeness and transparency of the results. They also highlight an exhaustive list of benefits reported by these companies, which may serve as an incentive for those that have not yet adopted IR.
