Full textCommon heat treatments applied to milk cause undesirable modifications. The goal of this project was to gain knowledge on the effect of an electroreductive treatment on heat-induced modifications in milk. The evaluation of rennet coagulation kinetics showed that the lagtime of electroreduced milk is lower than that of degassed or reference milk gel, whereas firmness is higher. Physico-chemical analyses show that electroreduced milk also has a higher thiol concentration. The study of heat coagulation time shows that electroreduced milk thermal stability, with pH higher than 6,90, is up to 5 times higher than that of the control. Moreover, electroreduced and degassed milks, heated at 140 ºC, have a higher thiol concentration than reference milk.

Full textThe goal of this research was to evaluate the behavioural variability of three genetic lines of growing pigs: Large White (LW), synthetic Meishan line (GM) and synthetic paternal line (SG) and its relationship with residual energy intake. Daily behaviour, behaviour at weighing and at slaughter showed that GM pigs were calmer and tamer. Large White pigs were more aggressive and more nervous at weighing. The synthetic paternal line was more nervous and more difficult to handle. Residual energy intake, measured as the difference between observed and predicted energy intake, was lower in GM pigs, likely because of differences in maintenance energy requirements. Both physical activity, estimated by the analysis of postures, and stress, represented by vocalisations, induced variation in residual energy intake.. Individual and genotypic behavioural variability may have an impact on adaptability, welfare and energy utilisation in growing pigs.

Full textProgress in genetic engineering has led to the development of a new system to produce therapeutic molecules in plants: molecular pharming. The interest to transform plants into molecular factories for human purposes is explained by its lower cost, its biosecurity and the capacity of plants to synthesize recombinant proteins with appropriate folding, glycosylation and activity. Floral ablation represents one of the best ways to establish both male and female genetic confinement in open field conditions. With the final objective to develop this strategy in alfalfa (Medicago sativa), the promoter of a MADS box gene, MsMADS1, was isolated from the alfalfa genome. The genomic organisation of MsMADS1 appears to be unusual with a first intron longer than 1,9 kp without homology to any known sequence. This promoter was fused to the GUS reporter gene and a cytotoxic protein gene (saporin). Following Agrobacterium-mediated transformation of Arabidopsis thaliana (Arabidopsis) with various constructs, transgenic plants were regenerated and analysed. Transient expression was also assayed to evaluate GUS expression in alfalfa cells. Additional MsMADS1 genomic sequence was obtained by genome walking. In transgenic Arabidopsis, the MsMADS1 promoter allowed GUS expression preferentially in flowers. Surprisingly, saporin constructs resulted in the production of viable transgenic plants with a phenotype identical to wild Arabidopsis.

Full textThe eukaryotic DNA mismatch repair system (MMR) is responsible for the repair of replication errors, and plays a role in genetic recombination. AtPMS1, a MMR gene from Arabidopsis thaliana, has already been shown to play a role in DNA repair but its involvement in meiosis has yet to be examined. We characterised two lines of insertional mutants at AtPMS1. A first mutant line has abnormal flowers, which is probably a phenotype unrelated to the loss of AtPMS1 activity but rather caused by cosuppression. The second mutant line shows many phenotypes, likely linked to AtPMS1 inactivation: a 65 to 80% reduction in seed production, an abnormal segregation of the mutant allele and cytogenetic abnormalities in 16% of the microsporocytes (chromosome fragmentation and missegregation). The observed bias in segregation as not been observed in other eukaryotes, and seems to be a particularity of MutL homologues in Arabidopsis or plants as a whole.

Full textDuring the growth and the maturation of the oocyte, organelles, proteins and mRNA known as maternal are stockpiled in order to prepare and supply material to the developing embryo in the main goal to activate the embryonic genome. An oocyte that is able to supply to this step is called developmental competent. The objective of the first experiment was to determine an embryos population with different level of developmental competence. The triple selection lies in two characters related to the oocyte, which are the follicular size and the morphology of the COC and one is related to the embryo, which is the time of first cleavage post insemination. The different population generated by this selection has provided a population highly competent to development and a population lacking developmental competence that will be useful for the study of developmental competence. The objective of the second experiment was the study of the proteomic of the translated maternal mRNA during the early embryo development. We wanted to know which proteins play the role of maternal housekeeping proteins during oocyte maturation and early embryo development. This experiment has clearly demonstrated that when the oocyte resume meiosis (Coenen et al., 2004) and after fertilization, the translated protein pattern follows a continuum events which conducts embryo to his activation. Although they are supply by the same reservoir of mRNA, the needs of the oocyte and the embryo are different. Only about fifty proteins are commonly translated during oocyte maturation and early development. Eleven of them have been identified: HSC71; HSP70; CypA (2 times); UCHL1; GSTM5; Cct5; E-FABP; 2,3-BPGM, ubiquitin-conjugating enzyme E2D3; and β- actin/γ-actin. The third experiment consists of the discovery of factors related to developmental competence. Three populations from the triple selection with high, low and developmental competence was compared by a proteomic study. This study has shown that developmental incompetent embryos translate a minimum of maternal mRNA, which the majority is the embryo housekeeping found in the previous study. Whereas high and low developmental iv competent embryos present shared and exclusive translated proteins, these proteins represent potential factors related to developmental competence. By trying to identify these proteins, we have found that most translated proteins are transient and are not accumulated in the cytoplasm. In spite of this observation, we have identified six proteins: cytosolic thiolase, isocitrate dehydrogenase 1, peroxiredoxin 6, proteasome-α6, hypothetical protein My027 and thiamine triphosphatase. Two maternal housekeeping proteins were also missing for the incompetent 2-cell embryos: UCH-L1 and CypA. From these experiments, a new method of protein identification confirmation, called in silico confirmation, was developed. As the genome and the proteome of Bos taurus is limited, the protein identification was made in foreign species or from Bos taurus EST databases. This method as made possible to rebuilt the bos taurus messager, to translate the bovine protein, find his theoretical peptide mass fingerprint and compare it to the experimental spectrum and thus increase the number of matched peptides. In conclusion, the proteomic study on the developmental competence of the bovine oocyte has identified of a reduce number of candidates compared to genomic studies. A new method has been developed during this work. The in silico confirmation allowed the confirmation of protein identification without additional material.

Full textIn 2001 and 2002, 89 strawberry fields from nine regions in Québec were sampled to determine the diversity and abundance of root weevil species. Soil characteristics were also assessed. We further studied the seasonal ecology of the strawberry root weevil, Otiorhynchus ovatus (L.) in the Québec City area. The strawberry root weevil was the most abundant species with >82% of all captured specimens. Its abundance was positively correlated with sand in soil sample. Two cohorts of insects have been identified with a peak of abundance at the end of the fruit harvesting period. This period is most suitable for O. ovatus control as females have not yet initiated oviposition.

Full textProductivities, decomposition rates and nutrient turnovers are low in polygon fens of Bylot Island. The Greater snow geese population could modify these parameters since goose faeces contain nutrients that are easily uptaken. However, it is important to consider the moss carpet. This study showed that mosses play an important role on the polygon fen dynamic by sequestrating the nutrients added by goose faeces. Grazed plants were able to uptake nutrients only when the mosses are saturated and consequently, when nutrients reach their roots. The decomposition rates and the turnover of nutrients were not affected by nutrient additions. In addition, in the short-term, it seems that fertilizing effect of geese was visible only on moss growth. As a secondary result, this study showed that innate marker method is the most appropriate to estimate moss primary production in arctic wetlands.
