Full textRestored peatlands are characterized by a uniform vegetation cover and a surface lacking open water bodies. Since pools are known to increase biodiversity in natural peatlands, their creation were investigated for restored sites. So far, we cannot rely on spontaneous recolonization for these created microhabitats. Mosses transfer from natural pool margins to artificial ones is a technique that failed to restore vascular plant communities associated with these microhabitats, although diaspores (rhizome, seeds, plants fragments) are transferred with mosses. Peat erosion and substrate instability around pool margins are factors affecting survival of seedlings. The main objective of this research project was to facilitate Sphagnum and vascular plants establishment around pool margins created in restored peatlands. One way to achieve this was through substrate stabilization using two species of shrubs (transplants of Andromeda glaucophylla Link and Vaccinium macrocarpon Aiton). Results show that ericaceous cover increased survival of Sphagnum along pool margins. Ericaceous shrubs -with or without mosses- also reduced frost heaving compared to the control but did not influence establishment or growth of five herbaceous species sowed around pool margin. The second objective was to evaluate different types of bryophyte seed beds to determine which one increase establishment of three herbaceous species sowed. Carex species have equally germinated on all seed beds, although Scheuchzeria palustris had a higher germination rate on Sphagnum cuspidatum seed beds. Finally, seedlings had a better growth rate on newly restored Sphagnum seed beds than on those well established. According to these results, we recommend transplanting ericaceous shrubs along pool margins as they stabilize substrate and promote Sphagnum growth. The best timing for herbaceous species sowing along pool margins would be in the first restoration process steps.

Full textThe goal of this project was to quantify the impact of sodium reduction and substitution by KCl and MgCl2 on the production of mozzarella cheese. The reduction of NaCl and its replacement by KCl or MgCl2 did not have an impact on the evolution of the growth and the enzymatic activities of thermophiles lactic bacteria in cheese slurry and mozzarella cheese. Mozzarella cheeses were salted in 12 brines solutions. The concentration of salts in the brine solutions had more impact on the cheese than the type of salts used in the brine. Low sodium mozzarella cheese may be produced without decreasing the quality and functional properties of the cheese, but the substitution of NaCl by KCl may induce a flavour defect in fresh mozzarella cheese, but this cheese could be used as food ingredient without modifying the quality of the final product.

Full textThe apple industry in Quebec is under a price management system. The Fédération des producteurs de pommes du Québec (FPPQ) manages a marketing agreement that sets a minimum for the price paid to Quebec apple producers. The floor price is determined by a committee composed of members of the FPPQ, members of the Quebec apple packers association and one representative of Quebec fresh apples buyers. This study predicts the price paid to Quebec producers for four varieties of apples (Cortland–McIntosh– Spartan– Empire) grown in Quebec and sold fresh in Quebec and in so doing will assist the FPPQ in the determination of the minimum price. Specifically, the objective of this analysis is to develop an empirical model that explains the factors that influence the price paid to Quebec producers for fresh apples. This prediction tool will provide a precise prediction of prices therefore facilitating decision by members of the Quebec apple industry. The empirical model includes variables that affect both the demand for apple and the supply of apple. The model includes variables for the stock of apple, the quantity of apples marketed, the imports of apples, the exchange rate and the prices for the four varieties of apples. The empirical model is at the cutting edge of the literature on time series that are jointly determined. The empirical model in this analysis allows for a precise prediction of the price for the four varieties of apple taking into account that the price for one variety affects and is affected by the price for all the other varieties. This approach allows for more precise prediction than under conventional models that do not account for simultaneity. The results of the estimations how that the net imports of apple from the United States and the quantity of apples marketed are the two most important factors that affect the price of apples in Quebec. The model yields a precise prediction for a time horizon below three months.

Full textIn Canada, modified starch addition in yoghurt is frequent in order to limit technological defects such as syneresis. Despite the addition of modified starch, technological defects still occur. Exopolysaccharides (EPS) produced naturally by some lactic acid bacteria can be used as stabilizers in yoghurt. Literature indicates that their properties to bind water and to modulate viscosity are not correlated to EPS concentration but to their structure and interactions with milk proteins. To our knowledge, this is the first work that has studied the effect of combined use of modified starch and EPS in yoghurt. The aim of this work was to study the structure-function relationship of EPS produced in situ by lactic acid bacteria on the rheological/physical properties (gel formation, viscosity, firmness, stiffness) and the microstructure of fermented dairy systems with modified starch. Four starters composed of one strain of Streptococcus thermophilus and one strain of Lactobacillus delbrueckii subsp. bulgaricus and producing EPS with known structure were studied: HC15/210R (control), HC15/291 (neutral, stiff, few branched EPS), HC15/702074 (neutral, flexible, highly branched EPS) and 2104/210R (anionic, stiff, linear, EPS). A final pH of 4.6 was obtained after a fermentation of 180-210 minutes à 42 C for all starters. The gel formation process was not influenced by the presence of EPS. The comparison of several EPS of known structure has shown that the anionic EPS from 2104/210R starter improved firmness and viscosity. Neutral, stiff and few branched EPS from HC15/291 starter contributed to viscosity and limited syneresis comparatively to the neutral, flexible and highly branched EPS from HC15/702074 starter. Although the smoothing process had a negative impact on the values of all rheological/physical properties of fermented dairy systems, the functionality of EPS remained. The addition of modified starch improved the rheological/physical properties of stirred fermented dairy systems but had no effect on set fermented dairy systems. To conclude, this work has shown that the rheological/physical properties of fermented dairy systems may be improved by the combination of modified starch and EPS with specific structural characteristics.

Full textThe Colorado potato beetle (CPB), Leptinotarsa decemlineata (Say), is the major insect pest of potato crops in North America, Europe, and Asia, causing important damages and financial lost. Currently, the most common mean used to control this insect consists on regularly applying chemical insecticides. However, these chemical products are often controversial because their excessive and uncontrolled use is hazardous for both the environment and the human health. Also, the CPB has developed over the years a resistance to most of the registered chemical insecticides, including those that were very effective at one time. To remedy to this lack of efficiency, producers usually use large amounts of chemicals, causing therefore health and environment problems and also raising the cost of crop protection. One of the most promising alternatives to chemical insecticides consists of taking advantage of the natural enemies of this insect pest. The use of the stink bug predator Perillus bioculatus to control the CPB has been successful at small scale in Quebec. However, this natural enemy is not abundant enough in the nature to control the CPB. Massive release of this predator on potato crops would be an adequate solution. Since labour force in agriculture is rather rare and expensive and the manual distribution of predators is laborious, it is necessary to mechanize the release process in the field, which represents a major challenge. The main objective of this research study was therefore to design and build a mechanical distributor of predator insects. A mechanical distributor of predators has been designed and built for this purpose. In this distributor, masses of predators are placed in small containers and mixed with popcorn. When the containers cross an opening system, their content drops on potato plants. Many tests carried out using this prototype revealed that the release rate of predators in the field is higher than 90% (more than 90% of the predators are released). This release rate is comparable to that obtained in the laboratory using a test bench. The prototype needs however extensive testing in the field to evaluate the survival of predators and their biological effect. Because of his revolutionary and unique device, this invention is in process to get an international patent.

Full textSoy is an important legume produced in Canada and its culture and production rate increased significantly in the recent years. Developing new competences to create by-products instead of exporting soya to other countries is of interest for the economy. The functional food and nutraceutical industry have great interests on soya due to its high bioactivity potential in regard of the protein content. Technologies using membranes offer the possibility of fractionating solutions to concentrate and isolate molecules of interest. These technologies can be divided in two main categories: Pressure-driven processes in which ultrafiltration (UF) and nanofiltration (NF) take part and electrically-driven processes in which electrodialysis with ultrafiltration (EDUF) membrane takes place. Working with protein or hydrolyzate solutions (soya in this case) can be problematic since these proteins or peptides have the tendency to foul the membranes. The aim of this study was 1) to evaluate the ion-exchange membranes potential to fouling by soy peptides 2) to compare pressure- and electrically-driven process in terms of mass flux and mass balance 3) to characterize the fractions in terms of molecular weight and amino acid profile, protein and peptide contents and 4) to evaluate their antioxidant potential and neuroprotection capacity. The study on ion-exchange membranes fouling by peptides showed that electrostatic interactions are the main cause of this phenomenon and its intensity depends on the type of membrane (cationic or anionic) used and the medium characteristics (acid or basic). Simple cleaning steps with NaCl solution could enhance performances of this process and improve its efficiency. Both processes showed a similar mass balance, however UF/NF process showed a higher mass transfer than UF/EDUF process when considering the time and membrane surface used to complete the run. UF/NF system produced a permeate with small molecular weights while UF/EDUF system produced fractions with a large range of molecular weights with a very specific molecular weight range depending on the pH value. Both processes produced similar fractions in terms of protein content. Specific fractions of EDUF were composed of a high level of polar amino acids. KCl 1 pH 3 and 6 from EDUF showed antioxidant capacities by the ORAC test. The same fractions as well as KCl 1 pH 9 and NF permeate pH 6 showed an interesting effect in the redox potential. Therefore none of these fractions showed protection against oxidative stress induced in cells.

Full textThe adoption of a centralized system of quota acquisition in the Québec egg production sector improved the availability of market information and raised the market efficiency. However the use of the T-kDA raises some questions regarding the possibilities of collusive and strategic behaviors, because of the limited number of participants to the auction’s sessions. In this study, we use the experimental economics approach to test the T-kDA in optimal collusion conditions. Our results suggest that collusion works when all the sellers and all the auction’s participants do communicate, but also when we have three sellers. Our results also show that with only one seller (monopolistic situation); his bid is smaller than the ones with three sellers communicating in contrast to the teaching of the economic theory. That is explained by the fact that in monopolistic situation, the seller does not know if he is the only seller. With three sellers in communication, the coordination between them produces a higher clearing price. The results also confirm that the T-kDA lower significantly the market price with or without collusive conditions. In other words, the truncated rule does not have a significant effect regarding the collusion when compare to the kDA.

Full textThis study was carried out to verify the biocompatibility between the mycetes of Camembert cheese and lactic cultures (probiotic and ripening strains). Most of the fungi strains used had been isolated from different milk sources over the province of Quebec (Canada) and two different kinds of milk were used to produce cheese slurries. Automated spectrophotometry (AS) was employed to screen some biocompatible pairings of mycete and bacterial strains. A milk medium was fermented by yeasts and moulds and then inoculated with bacteria. The previous growth of the mycetes was sometimes stimulatory and sometimes inhibitory, but the effects were minor and varied as a function of the strains. Subsequently, to confirm these AS results, cheese slurries were inoculated simultaneously with different strains combinations. Finally, pilot scale Camembert cheese was produced to verify its ability to support probiotic bacterial cultures viability. The absence of inhibition of the bacteria by the mycetes in SA was confirmed, but the interactions in the cheese slurries differed from those noted in AS because of the different pH patterns in the two experimental series. Camembert was shown to have potential to favour the viability of probiotic bacterial strains during ripening and storage. However, no mycete mix was systematically better than another to stimulate this viability.

Full textHigher plants have developed, over time, a variety of protection mechanisms allowing them to survive and cope with a variety of biotic stress cues in their surrounding environment. The main goal of this three-part doctoral thesis was to characterize the biochemical and the physiological responses of potato (Solanum tuberosum) to defoliation by the coleopteran insect herbivore Colorado potato beetle (Leptinotarsa decemlineata). The first objective of the project was to characterize the impact of the insect on the host plant's leaf proteome, using as a model plants treated with potato beetle larvae, mechanically wounded plants and plants infested with a sucking/piercing insect, the potato aphid (Macrosiphum euphorbiae). The second objective was to gain some insight about the proteome of potato beetle oral secretions, with the aim of assessing the relative incidence of insect and host plant proteins at wound sites generated during insect feeding. The third objective, finally, was to determine the impact of leaf proteome alterations on photosynthetic capacities of the host plant, and to determine the possible impact of the plant's own molecular constituents on the responses observed. In brief, our results showed that several proteins involved in the primary and the secondary metabolisms, including photosynthesis-related proteins, were regulated in leaves in response to potato beetle feeding. However, the negative impact of the insect on several photosynthetic proteins, notably photosystem I proteins, only had negligible effects on the light capture process by the plant. The second phase of photosynthesis, on the other hand, was significantly affected by the insect, presumably via molecular effectors from both the insect and the host plant itself. These findings suggest, overall, a specific, but somewhat limited impact of Colorado potato beetle larvae on the leaf proteome and photosynthetic capacities of the potato host. They also suggest the possible induction of compensatory mechanisms in planta and the high plasticity of primary metabolism functions in the plant upon herbivore feeding.
