Abstract:
COVID-19 is the world crisis caused by Coronavirus, which a major fall down for the tourism industry. This study is done to analyze the strategic approach in mitigating tourist destinations and to prepare the destination for the recovery after the Coronavirus pandemic. The purpose of this research is to identify and analyze strategic approaches used for mitigation in the case study of Bagak Sahwa Village Tourism in Singkawang, West Kalimantan Province. The data used qualitative research method through 8 in-depth interviews and 1 focus group discussion as the source persons. In a cause of travel restriction, the qualitative data collection method was conducted virtually. This finding uses the village tourism approaches to analyze the strategy. Based on the findings, the community of Bagak Sahwa understanding of mitigation has increased and developed several strategies in mitigating their destination. The community expects the strategy for the improvement and development of the village destination from the local tourism office. Abstract:
This research aims to ensure the active role of the Bagak Sahwa village community in making decisions on tourism by using the environmental area to obtain a fair share of income from tourism activities. Bagak Sahwa tourism village is expected to implement Community Based Tourism (CBT) effectively. However, the implementation by the local community has not been conducted as mandated in Law No. 10 of 2009. In some cases, it can even lead to community conflicts. The results showed that community participation in developing the tourism village potential was still low. The low quality of human resources also influenced this situation, and the resident-focused more on the agricultural sector. The road access is in poor shape, insufficient facilities, and infrastructure supporting tourism activities, the promotion of tourism objects is still deficient, and the quality and quantity of human resources are still low. The Social Entrepreneur model’s presence has impacted a shift in society’s perspective socially and economically. However, the Village community still upholds their ancestors’ language and behavior patterns. The administrators involving the local community transmit knowledge based on these findings. Therefore, the local community is educated to actively participate in developing the village. They can collaborate with new things by involving millennials in tourism village activities to maintain local culture and wisdom.

Abstract:
This paper tracks the relationship between religion and the state in Saudi Arabia during the past 50 years. This relationship is unique in the sense that the religious power (Wahabi) together with the ruling family (Al Sa'ud) were partners in founding the Kingdom. This cooperative relationship has been degenerating into mutual suspicion. Right now there are three main variants of religious stratification in Saudi Arabia vis-à-vis the state: the traditional Salafists who back the rulers come what may – the government is gradually reducing their prerogatives; and the opposition, which in turn is divided between Sahwa (who are discontented Salafists) and Tanweeris (advocates of enlightenment). The Sahwa current with its elderly leadership believes that the state is straying away from the teachings of religion and it recommends austere adherence to it; the dynamics of their activities are possibly pushing towards taking leadership from outside the Kingdom. The Tanweeris, on the other hand, have young leaders, the popular base is young and broad, and they are strong advocates of democracy. Despite their capacity for effective social mobilization, it seems too early to judge whether this religious stream will eventually push for reform or constrain it, especially given that it has not yet resolved its position on some key issues such as different national constituencies and women.

Violence against women and subsequent gender-based violence are issues that plague the world, harming women’s wellbeing as well as that of their families. Thirty-nine percent and twenty-one percent of Kenyan women have experienced physical and sexual violence, respectively, in their lifetimes. While there have been contested studies showing that employment can both increase and decrease the risk of suffering from violence, particularly in domestic settings, this study examines how a Kenyan woman’s experience of violence is likely to affect her level (formal or informal) of employment in the future. The results of this study indicate that emotional abuse, having a partner that drinks, educational attainment, living in a rural setting, and age are significant factors in a woman’s probability of working. Conditioned on working, experiencing controlling behaviors from a partner, educational attainment, justification of violence, ethnicity, income rank, partner’s occupation, and age at first marriage influence a woman’s probability of working informally. These results vary based on the type of employment studied, but can have wide-ranging consequences for the economic development of Kenya and empowerment of Kenyan women

Tutkimus käsittelee Martinniemen sahaa kulttuuriperinnön näkökulmasta. Suunnitelmat sahan purkamisesta herättivät paikalliset ajamaan sahan piipun suojelua. Käynnistyi kulttuuriperintöprosessi, jossa erilaisten tunteiden, muistojen ja kokemusten kautta yritettiin saada piipusta valtakunnallisesti suojeltu kulttuuriperintö kohde. Paikallisille koko sahan historia ja kulttuuriperintö kiteytyi lopulta piippuun ja piipusta tuli konkreettinen kohde, joka kiteytti ja säilytti kulttuuriperintöä. Paikallisten kautta tutkimuksessa tarkastelen yhteisöllistä kulttuuriperintönäkökulmaa. Millainen suhde paikallisilla oli sahaan ja mikä oli sahan ja sen luoman kulttuuriperinnön merkitys heille? Yksilöllisen kulttuuriperintönäkökulman tutkimukseen tuovat sahalla vierailevat ihmiset, joilla ei ole aikaisempaa suhdetta sahaan. Raunioituneissa rakennuksissa vierailevat harrastajat muodostivat erilaisia henkilökohtaisia suhteita paikkaan. Tarkoituksessa ja ihmisestä riippuen hylätty voimalaitos saattoi merkitä seikkailua, jännitystä, piilopaikkaa, leikkikenttää tai paikkaa, jossa päästä toteuttamaan itseään. Urban explorer harrastajat, valokuvaajat ja graffititaiteilijat vierailivat sahan hylätyissä, rapistuvissa rakennuksissa ja erilaisten henkilökohtaisten kokemuksiensa kautta loivat merkityksiä rakennuksille. Tutkimuksen aineistona olivat sahan voimalaitos ja piippu. Vierailin Laitakarin saarella, jossa voimalaitos ja piippu sijaitsivat, kolme kertaa tutkimusta tehdessäni 8.9.2016, 4.10.2017 ja 29.5.2018. Ensimmäisellä vierailulla tutustuin voimalaitoksen sisätiloihin, toisella vierailulla ympäristöön ja kolmannella vierailulla kävin katsomassa paikkaa, kun rakennukset oli purettu pois. Tutkimuksessa tarkastelin suojelun uutisointia ja erilaisia blogeja ja YouTube videoita sahan voimalaitoksella tapahtuneista vierailuista. Paikallisten luomaa kulttuuriperintöprosessia tarkastelin uutisissa esiin tulleiden erilaisten haastattelujen kautta, joissa perusteltiin piipun säilyttämistä. Voimalaitoksen merkitystä raunioharrastajille tarkastelin heidän tekemiensä nettijulkaisuiden kautta. Paikallisille koko sahan olemassaolo kiteytyi lopulta piippuun, jonka suojelun puolesta he taistelivat vuosia. Heille sahan kulttuuriperinnöstä muodostui miltei elävä olento, joka tarvitsi konkreettisen ilmentymän, jotta se säilyisi. Paikallisen kulttuuriperintö ja valtakunnallinen kulttuuriperintö eivät lopulta kohdanneet Martinniemen sahan piipun kohdalla ja kaikki rapistuneet rakennukset purettiin pois. Valtakunnallisella, yhteisöllinen ja yksilöllisellä kulttuuriperinnöllä on erilaiset lähtökohdat ja määritelmät. Martinniemen sahan tapauksessa valtakunnallinen ja yhteisöllinen kulttuuriperintönäkökulma ajautuivat konfliktiin, kun taas yksilöllinen kulttuuriperintönäkökulma pysyi erillään suojelusta, koska suojeltuna tai purettuna, rakennuksilla ei enää ollut heille käyttötarkoitusta.

Tässä opinnäytteessä tarkastellaan Ruukin sahayhtiötä vuosina 1900–1912 eli osakeyhtiö G. & C. Bergbom A/B:n aikana yrityksen liiketoiminnan, työtehtävien ja palkkauksen, työyhteisön arkielämän ja työläisyyden muodostumisen näkökulmista. Pruukin Saha Yhtiö oli tutkimusajanjakson aikana monialayritys, jonka toimialaan kuuluivat puutavaranjalostuksen ohella tiilenvalmistus sekä maa- ja karjatalouden harjoittaminen. Pääosan tutkielman arkistolähteistä muodostavat Ruukinsahan kassakirjat vuosilta 1901–1905 ja 1905–1907 sekä työlistakirjat vuosilta 1905–1907 ja 1909–1913. Toinen merkittävä osa tutkimuksen lähteistä koostuu tutkimusajanjakson aikana ilmestyneiden sanomalehtien Ruukin sahaa koskevasta uutisoinnista. Näistä keskeisimpiä ovat Kansan Tahto, Kaiku, Kaleva ja Raahen Sanomat. Tutkimusta täydentävinä lähteinä käytetään kolmea muistelmaluontoista kirjallista teosta. Tätä tutkimusta tehtäessä on käytetty kolmea erilaista tutkimusmenetelmää: historiantutkimukselle tyypillistä lähdekriittistä menetelmää, lähteiden lähilukua ja tilastollisia menetelmiä kassa- ja työlistakirjojen numeraalista materiaalia hyödyntäen. Maaseutumainen rauha rikkoutui Ruukissa viimeistään vuoden 1905 suurlakosta alkunsa saaneen työläisten järjestäytymisen myötä. Joukkovoima mursi paikkakunnalla siihen saakka vallinneen yksinapaisen paternalistisen järjestyksen ja tehdastyöväestö muodosti hyvin aktiivisen ja näkyvän, varteenotettavan voimatekijän kylän sosiaalisessa järjestyksessä. Tehdastyöläisiä yhdisti toisiinsa sosialismiaatteeseen nojautuva työväenyhdistys, jonka jäseninä kilpailevien yritysten työläiset pääsivät yhdessä osoittamaan joukkovoiman mahtia työnantajiaan kohtaan. Ruukin sahayhtiön tehdasyhdyskunnan elämää leimasi arkinen työ ja jatkuva muutos. Työntekijöiden oli sopeuduttava paitsi luonnonolosuhteiden vaihtelun tuomiin haasteisiin myös kahden eri tehtaan teknisten muutosten vaatimiin lukuisiin erilaisiin työtehtäviin — unohtamatta henkistä sopeutumista vuosisadan uusiin aatevirtauksiin ja nopeasti kasvavan tehdaskylään elämänrytmiin. Raahen sahayhtiön rakennuttaessa Ruukkiin suuren sahalaitoksen vuosisadan alussa, jäi ”Vanha Saha” toimialallaan kilpailijaansa huomattavasti pienemmäksi toimijaksi. Sahan työntekijät saivat opetella kokonaan uuden teollisen prosessin (tiilitehtaan) työvaiheet, heti kun olivat saaneet rakennettua tehdasrakennuksen valmiiksi. Sosiaalisia epäkohtia vuosisadan alun Ruukissa kuvastavat aikakaudelle melko tyypilliset piirteet, kuten naisten alempi palkkaustaso, tehdastyön vaarallisuus ja heikko oikeudellinen asema irtisanottaessa niin vuokra- kuin työsuhteestakin. Leimaa-antavana piirteenä koko tutkimusajanjakson aikana oli oululaisten Bergbom-sukuisten omistajien fyysinen poissaolo tehdasalueelta.

Abstract:
Il controllo epigenetico dell'espressione genica e' operato da modifiche post-translazionali della cromatina, quali acetilazione e deacetilazione. Due enzimi sono respinsabili di questo processo: le HAT (istone acetiltransferasi) e le HDAC (istone deacetilasi). Le HDAC sono un gruppo di enzimi coinvoilti nella regolazione dell'espressione genica, nella riparazione de DNA e nella risposta allo stress. L'acetilazione gioca un ruolo importante nella trascrizione poiche' responsabile del rimodellamento della cromatina aumentando l'accesso dei fattori di trascrizione al DNA. Una diminuzione dei livelli dell'acetilazione istonica sono associate con un aumento dell'attivita' di trascrizione, mentre un decremento dei livelli di acetilazione e associate con le repression della repressione genica. Infatti nelle cellule tumoralisi osserva frequentemente un'anormale espressione genica. Gli inibitori delle HDAC (HDI) sono potenti induttori dell'arresto della crescita, della differenzione e della morte dellulare per apoptosis in cellule trasformate sia in vitro che in vivo. Ad oggi vi sono sette classi distinte di inibitori delle HDAC e tutti contengono tre elementi strutturali chiave del farmacoforo: a) un gruppo legante lo zinco (R), che coordina gli ioni zinco al fondo della lunga e stretta cavita' del sito attivo; b) un cappuccio o à ¢ capà ¢ (la funzione solfonammidica), che interagisce sia con gli aminoacidi sul margine del sito di legame sia con la superficie della proteina. c) un dominio di legame, il cui ruolo e' assicurare il corretto posizionamento dei gruppi precedenti e di interagire con il canale di legame lipofilo. Seguendo il nostro recente interesse nella ricerca di HDI piu' potenti, ho progettato e sintetizzato una nuova serie di composti contenenti quattro nuove porzioni chelanti lo zinco, quali formilidrazonometilica, 2-formilidrazinocarbonilica, 2,2,2-trifluoro-N-metilencetammidica e 2,2,2-trifluoro-N-metilacetammidica, legate attraverso uno spaziatore ad una solfonammide (Schema 1), responsabile dell'interazione idrofobica con l'HDAC. Studi di docking hanno mostrato che queste molecule si inseriscono perfettamente nella tasca enzimatica dell'HDAC con un ottimo à ¯ Gbind di à ¢ 7,9 kcal/mol che e' una 1 kcal/mol piu' bassa del SAHA, un ottimo inibitore.
The epigenetic control of gene expression is operated through post-translational modifications of chromatin such as acetylation and deacetylation. Two enzymes are responsable for these processes: HAT (histone acetyl transferase) and HDAC (histone deacetylase). HDAC is a group of enzymes involved in regulating gene expression, DNA repair and stress response. Acetylation plays an important role in trascription because it remodels chromatin structure enhancing access to DNA-binding factors. In general, increased levels of histone acetylation are associated with increased transcriptional activity, whereas decreased levels of acetylation are associated with repression of gene expression. In tumor cells abnormal gene expression is frequently observed. HDAC inhibitors have been shown to be potent inducers of growth arrest, differentiation, and apoptotic cell death both in vitro and in vivo transformed cells. To date there are seven distinct classes of HDAC inhibitors and all contain three key structural elements to their pharmacophore: a) a zinc-binding group which coordinates the zinc ion at the bottom of the long narrow active site cavity; b) a capping group which interacts both with the amino acids on the rim of the binding cavity and the protein surface; c) a linker domain whose role is to ensure the correct positioning of the two former groups and to in-teract with the lipophilic binding tunnel. Following our recent interest in the research of more potent HDAC inhibitors, we have projected and synthesized a new series of compounds containing four new zinc chelant moieties, i.e. formylhydrazonomethyl, 2-formylhydrazinocarbonyl, 2,2,2-trifluoro-N-methylideneacetamidic and N-(trifluoroacetyl)amidic, linked by an aromatic spacer to an aromatic sulfonamide, that is responsible of hydrophobic interaction with HDAC. Docking studies have shown that these new molecules fit very good in the HDAC enzymatic pocket with a best ï Gbind of â 7,9 kcal/mol that is 1 kcal/mol lower of the corresponding one for SAHA, a good HDAC.

Abstract:
We are investigating silicon-based platforms for detection and analysis of breast cancer cells. Attachment and spreading of MDA-MB-231 human metastatic breast cancer cells was compared to that of non-tumorigenic human breast epithelial cells, MCF-10A, and the impact of SAHA (Vorinostat), a histone deacetylase (HDAC) inhibitor, on cell behaviors was ascertained. Our results showed the cancer cells attached to flat silicon and PECVD nitride-coated silicon more efficiently than non-cancer cells, and preferential cancer cell attachment was enhanced by SAHA. Fluorescent immunohistochemistry (IHC) revealed that SAHA stimulated actin stress fiber formation and focal adhesion to the substrates; atomic force microscopy (AFM) showed SAHA increased the cancer cell stiffness. Collectively, SAHA-induced biomechanical changes altered the cell morphology and mode of attachment to flat silicon and to three-dimensional silicon microstructures. This is the first report of the use of AFM to characterize the biomechanical effects of a HDAC inhibitor in cancer cells.

Abstract:
In this paper we report development of three dimensional silicon microenvironments in order to test the morphological changes and adhesion properties of human breast cancer cells after treatment with different anticancer drugs such as Trichostatin A (TSA), suberoylanilide hydroxamic acid (SAHA) and Scriptaid. Our results indicate that the cancer cells reorganize their cytoskeleton structure after treatment with TSA and Scriptaid. However, SAHA does not change the behavior of the cells inside the three dimensional microstructures while TSA and Scriptaid evoked striking changes in the cells morphology. TSA and Scriptaid drugs cause the cells to stretch inside the isotropic microchambers to avoid contact with curved sidewalls in contrast to their originally rounded shape. The proposed microstructures can be used to evaluate mechanical properties and the pathological grade of various cancer cell lines after different conditions i.e. drug exposure.

VUV spectra of a highly ionized argon plasma produced by a high-power subpicosecond laser are analyzed to determine the electron temperature. The calculated electron temperature is too low to support the highest observed charge state, showing the plasma charge state distribution to be out of Saha equilibrium.

The Environmental Laboratory (EL) and the Coastal and Hydraulics Laboratory (CHL) have jointly completed a number of large-scale hydrodynamic, sediment and water quality transport studies. EL and CHL have successfully executed these studies utilizing the Geophysical Scale Transport Modeling System (GSMB). The model framework of GSMB is composed of multiple process models as shown in Figure 1. Figure 1 shows that the United States Army Corps of Engineers (USACE) accepted wave, hydrodynamic, sediment and water quality transport models are directly and indirectly linked within the GSMB framework. The components of GSMB are the two-dimensional (2D) deep-water wave action model (WAM) (Komen et al. 1994, Jensen et al. 2012), data from meteorological model (MET) (e.g., Saha et al. 2010 - http://journals.ametsoc.org/doi/pdf/10.1175/2010BAMS3001.1), shallow water wave models (STWAVE) (Smith et al. 1999), Coastal Modeling System wave (CMS-WAVE) (Lin et al. 2008), the large-scale, unstructured two-dimensional Advanced Circulation (2D ADCIRC) hydrodynamic model (http://www.adcirc.org), and the regional scale models, Curvilinear Hydrodynamics in three dimensions-Multi-Block (CH3D-MB) (Luong and Chapman 2009), which is the multi-block (MB) version of Curvilinear Hydrodynamics in three-dimensions-Waterways Experiments Station (CH3D-WES) (Chapman et al. 1996, Chapman et al. 2009), MB CH3D-SEDZLJ sediment transport model (Hayter et al. 2012), and CE-QUAL Management - ICM water quality model (Bunch et al. 2003, Cerco and Cole 1994). Task 1 of the DOER project, “Modeling Transport in Wetting/Drying and Vegetated Regions,” is to implement and test three-dimensional (3D) wetting and drying (W/D) within GSMB. This technical note describes the methods and results of Task 1. The original W/D routines were restricted to a single vertical layer or depth-averaged simulations. In order to retain the required 3D or multi-layer capability of MB-CH3D, a multi-block version with variable block layers was developed (Chapman and Luong 2009). This approach requires a combination of grid decomposition, MB, and Message Passing Interface (MPI) communication (Snir et al. 1998). The MB single layer W/D has demonstrated itself as an effective tool in hyper-tide environments, such as Cook Inlet, Alaska (Hayter et al. 2012). The code modifications, implementation, and testing of a fully 3D W/D are described in the following sections of this technical note.
