Full textViolence against women and subsequent gender-based violence are issues that plague the world, harming women’s wellbeing as well as that of their families. Thirty-nine percent and twenty-one percent of Kenyan women have experienced physical and sexual violence, respectively, in their lifetimes. While there have been contested studies showing that employment can both increase and decrease the risk of suffering from violence, particularly in domestic settings, this study examines how a Kenyan woman’s experience of violence is likely to affect her level (formal or informal) of employment in the future. The results of this study indicate that emotional abuse, having a partner that drinks, educational attainment, living in a rural setting, and age are significant factors in a woman’s probability of working. Conditioned on working, experiencing controlling behaviors from a partner, educational attainment, justification of violence, ethnicity, income rank, partner’s occupation, and age at first marriage influence a woman’s probability of working informally. These results vary based on the type of employment studied, but can have wide-ranging consequences for the economic development of Kenya and empowerment of Kenyan women

Full textThe epigenetic control of gene expression is operated through post-translational modifications of chromatin such as acetylation and deacetylation. Two enzymes are responsable for these processes: HAT (histone acetyl transferase) and HDAC (histone deacetylase). HDAC is a group of enzymes involved in regulating gene expression, DNA repair and stress response. Acetylation plays an important role in trascription because it remodels chromatin structure enhancing access to DNA-binding factors. In general, increased levels of histone acetylation are associated with increased transcriptional activity, whereas decreased levels of acetylation are associated with repression of gene expression. In tumor cells abnormal gene expression is frequently observed. HDAC inhibitors have been shown to be potent inducers of growth arrest, differentiation, and apoptotic cell death both in vitro and in vivo transformed cells. To date there are seven distinct classes of HDAC inhibitors and all contain three key structural elements to their pharmacophore: a) a zinc-binding group which coordinates the zinc ion at the bottom of the long narrow active site cavity; b) a capping group which interacts both with the amino acids on the rim of the binding cavity and the protein surface; c) a linker domain whose role is to ensure the correct positioning of the two former groups and to in-teract with the lipophilic binding tunnel. Following our recent interest in the research of more potent HDAC inhibitors, we have projected and synthesized a new series of compounds containing four new zinc chelant moieties, i.e. formylhydrazonomethyl, 2-formylhydrazinocarbonyl, 2,2,2-trifluoro-N-methylideneacetamidic and N-(trifluoroacetyl)amidic, linked by an aromatic spacer to an aromatic sulfonamide, that is responsible of hydrophobic interaction with HDAC. Docking studies have shown that these new molecules fit very good in the HDAC enzymatic pocket with a best ï Gbind of â 7,9 kcal/mol that is 1 kcal/mol lower of the corresponding one for SAHA, a good HDAC.

Full textIn this work two approaches to obtain new secondary metabolites from fungi were studied. Suberoylanilide hydroxamic acid (SAHA) and five analogues were synthesized and assessed as epigenetic modifiers in eight marine-derived fungi. The fungus Penicillium sp. and Acremonium sp. presented changes in their metabolomic profile when they were grown on a small scale in presence of 4F-SAHA derivative and Microphaeropsis sp. presented a modification in the presence of the 3 AM-SAHA. In the second approach a new extraction strategy using XAD 2-4-7 resin mixture allowed the extraction and subsequent isolation of six new peptide alkaloids (72-77) and two peptides (78-79) from the aqueous fraction of the culture medium of the Antarctic fungus Penicillium solitum IS1-A. Using the same methodology, the secondary metabolism of the Antarctic fungus Thelebolus globosus E2 was studied. From the methanolic extract of the PDB liquid medium cyclopenine (86) and cyclopenol (87) were isolated and the viridicatine (88) and viridicatol (89) alkaloids were detected. The fungus Thelebolus globosus was shown to be a producer of statins, mainly of mevastatin (90).

In Iraq, since ever, tribalism constituted an organizational structure and a pattern of social relations, as well as dynamic of power’s exercise. Under the British Mandate and the Monarchy, politic manipulation of tribal’s culture and structures strengthened government’s legitimation; during the Republican period, as the Army emerged as main political driver and the influence of Modernity, tribalism transformed itself even if its presence into the society was still very strong. In the initial phase of Ba’thist’s regime, tribal culture was opposed, since it was considered as an obsolete heritage. But, facing the difficulties stemming from the war with Iran, the invasion of Kuwait and the embargo, the regime of Saddam Hussein exploited tribalism as a dynamic of power’s control, as well as propaganda and social bond. In the political void ensuing to the collapse of the regime, the tribes surfaced as “battlefield” between insurgency and Coalition’s Forces. American Surge and the tribes’ intervention in the Sahwa, as a reaction to jihadist’s bloodbath, avoided a sectarian civil war. Also al Maliki was able to exploit the tribal system. Nowadays, tribes are still an invaluable constituency for every political party, as well as diversified and a cross-parties political force.

Full textHemangiosarcoma is a very common canine neoplasm of vascular endothelial origin. In the dog, the most frequent primary site for hemangiosarcoma is the spleen. Typically, canine hemangiosarcoma has a very aggressive biologic behavior with metastases, especially to lung and liver, occurring early in the course of the disease. In the majority of cases, death is related to acute hemoabdomen secondary to tumor rupture and/or metastases. Surgery remains the main method of treatment for most dogs with this type o cancer, but adjuvant chemotherapy is highly recommended due to the high risk of metastasis and the poor outcome associated with surgery alone. A retrospective study was performed to determine survival times and potential risk factors in dogs diagnosed with visceral hemangiosarcoma. Medical records of 42 dogs were reviewed. Age and baseline weight at the time of diagnosis, breed, sex, tumor location, clinical stage of the disease, treatment type and median survival time were evaluated. Twenty-three dogs were treated with surgery alone, while 19 dogs were treated with surgery and adjuvant chemotherapy. There was significant difference in survival between dogs treated with surgery alone (66 days) and with surgery followed by Doxorubicin (274 days). Dogs with splenic hemangiosarcoma had a longer median survival time than dogs with hemangiosarcoma of other sites, and with metastasis (274 versus 117 versus 38 days, respectively, p = 0,013). The overall median survival time for these 42 dogs was 237 days, and the one-year survival rate was estimated to be 26.32%. In conclusion, primary tumor location was associated with prognosis and the addition of doxorubicin after surgery did improve survival. In this study, clinical stage had no association with prognosis

Full textThis study on death, mourning and funerals conducts, is a tentative reflexion on man in three dimensions : ontological, ethical, and metaphysical. For example with the Sawa of Cameroon, there exist two aspects of funerals conducts; one liturgical and the other therapeutic. These two aspects manage the equilibrium of the individual as well as that of the community involved. There is significance in the analysis of these conducts. Then, the scene of the funerals is entirely representational structure. The space of death, modified on the arrival of death is sacred. This dramatic production allows the emergence of significants majors in order to expel the "evil" of death. The principal significant persons being the dying and the widow. The body of the dying person concerned is magnified and subinised by the way of aesthetic and harmony, until it becomes a symbolical "figure". The widow is taken care on three stages: the "prosecuted", the condemnation and the rebirth. The purpose of which is significant on two levels: the change of the name and the attribution of a colour symbolical of widowhood. Thus the widow as metaphor of the community as well as the metonymy of "evil". The parallel setting of these two persons engenders two paradigms: the precariosity the disorder, the unbalance, the order of death; the permanence, the order, the harmony, and order of life. From a therapeutic point of view, the treatment of dying person and the charge of the widow on the scene of funerals, permits the emergence of a funeral "us" as an affirmation of the community "us". This brings us to the reflexion of death as "evil" and the search for meaning of death as a search for the meaning of life

Full textThe spatial dependence is a fundamental concept in spatial analysis. The spatial dependence results from that everything is related to everything else, but near things are more related than distant things. Spatial data analysis may use heuristic algorithms as Genetic Algorithms; however, these algorithms ignore the spatial dependence. In this work we present the SAHGA Spatially Aware Hybrid Genetic Algorithm a Hybrid Heuristic Algorithm with explicit representation of spatial relationships. The proposed algorithm includes a Generalized Proximity Matrix (GPM) to represent the spatial association between objects in space, in other words the spatial relationships. SAHGA embody two heuristics: Genetic Algorithms and Simulated Annealing. We developed two software with SAHGA: SAHGA Model Breeder (SAHGA MB) and SAHGA Species Distribution Models (SAHGA SDM). The SAHGA MB was used in social data analysis and the SAHGA SDM was used to model the potential distribution of Strix varia Barton (1799) and Thalurania furcata boliviana Boucard (1894) species. Both software may be used to create models with and without spatial relationships. We compared the adjusted models, with and without spatial relationships, to observe the effects of spatial relationships. We also compared the created SDM with GARP models. The results obtained show that the developed software may adjust useful models. Model breeder and species distribution models were affected by spatial relationships; the effects of relationships are function of the knowledge represented by the GPM.

Full textPursuing our exploration on anticancer activity of tamoxifen ferrocene derivatives, we have synthesized a series of hybrid complexes resulting from the substitution of ferrocifen (FcTAM) side chain with SAHA, a histone-deacetylase inhibitor (HDACi) used in the treatment of certain lymphomas. From FcTAM-SAHA hybrid, five types of modifications have been performed. They are [1] modification of the hydroxamic acid function (CONHOH) with a primary amide (CONH2) and a carboxylic acid (COOH), [2] variation of the side chain length, [3] with or without an aromatic substituent, [4] substitution of the metallocene and [5] substitution of the ferrocenyl unit with a phenyl group. The antiproliferative effects of the new complexes have been studied on two breast cancer cell lines, MDA-MB-231 (hormone-independent and triple-negative cell line) and MCF-7 (hormone-dependent cell line). A synergistic effect of the toxicity was observed in series FcTAM-SAHA on both cell lines. Surprisingly, no increase in toxicity was observed in the phenolic (FcOHTAM-SAHA-type) and in ferrocenophane (FnTAM-SAHA-type) hybrids. This observation suggests that the redox properties of the molecules are not expressed in the same way for precursors and hybrid compounds. In general, the hydroxamic acids and primary amides are more cytotoxic than carboxylic acids and these effects are not influenced by the length of the side chain. Finally, we found that the ferrocenic compounds are always more cytotoxic than their organic analogs. This difference highlights the importance of the presence of the ferrocenic moiety associated with its unique redox properties

Full textMucoepidermoid carcinoma (MEC) is the most common malignant salivary tumor compromising about 30% of all salivary malignances. Managing MEC patients remain challenging especially due to the heterogeneous response of tumor cells to available therapy. For this reason clinical outcome remains unpredictable. Current treatment of MEC encompasses surgical resection with eventual adjuvant radiotherapy, which frequently leads to functional and aesthetic complications. The use of chemotherapy is often reserved for recurrent and metastatic tumors. Administration of single-agent or combination therapy has showed activity, however overall response rates are unsatisfactory and of short duration. Emerging evidences suggest that the modest response of tumor cells to therapy resulting in high recurrence rates and poor survival, are associated with the presence of cancer stem cells (CSC). Quiescence of CSC is achieved by the reduced levels of transcription in a process that requires tight folding of DNA driven by core histone proteins. Changes in DNA folding are responsible for different cellular phenotypes mediated by a cell type-specific chromatin organization. Of interest, we also found that acetylation of HNSCC tumor histones driven by histone deacetylase (HDAC) inhibitors abrogate tumor resistance to chemotherapy. We investigate the effects of HDACi and cisplatin in the population of CSCs of MEC. Initially, we found that MEC tumors are composed by a heterogeneous population of squamous-like and mucous-like cells presenting distinct acetylation levels of histone 3 (Lys9). Tumor cells where treated with cisplatin and SAHA, a Food and Drug Administration (FDA) approved histone deacetylase inhibitor. Surprisingly, we found that administration of SAHA resulted in complete depletion of MEC CSCs. In facts, SAHA alone surpassed the inhibitory therapeutic effects of cisplatin and the combined therapy using SAHA and cisplatin over the population of CSCs. We also found that administration of cisplatin to MEC tumor cells result in unexpected accumulation of a sub population of CSC (paraclones), suggesting a correlation between the administration of intercalating agents such as cisplatin to the development of resistance of MEC cells to chemotherapy.

El tratamiento que se administra en etapas avanzadas incluye radioterapia, quimioterapia y cirugía. El fármaco más utilizado es cisplatino, a pesar de que se ha evidenciado una alta tasa de fracaso y recidiva, además de los efectos secundarios producido por el tratamiento. Es por esto que se han evaluado diversos compuestos farmacológicamente activos, con actividad citotóxica, como quimiosensibilizadores o moduladores de la resistencia a fármacos. Es en este ámbito que el desarrollo de inhibidores de histona desacetilasas (iHDAC) pueden ser utilizados en el tratamiento para COCE, ya que el aumento de las histonas desacetilasas contribuye a la expresión aberrante de genes, existiendo por ende una relación entre la desacetilación de histonas y la iniciación y progresión tumoral. El propósito de este trabajo es evaluar el efecto de ácido hidroxámico suberoilanilida (SAHA), un iHDAC, en células tumorales orales, tanto por sí solo como en combinación con carboplatino, este último debido a que presenta menos efectos adversos que cisplatino. Lo que se propone es que existe un efecto antitumoral citotóxico sinérgico de SAHA y Carboplatino in vitro en líneas tumorales de COCE. Para llevar esto a cabo se utilizó una línea tumoral oral, CAL27. Para medir citotoxicidad se utilizó el ensayo colorimétrico 3-(4,5dimethythiazol-. 2-yl)-2,5-diphenyl bromuro de tretazolio (MTT) y para evaluar el efecto en la migración celular se desarrolló el ensayo de herida. Se analizaron los datos usando t-test Student u ANOVA. Como resultados se observó que SAHA presenta un efecto dosis y tiempo dependiente, además en combinación con carboplatino presenta efecto citotóxico estadísticamente significativo con respecto al control y a cada tratamiento por separado, observándose entre ellos un efecto sinérgico. Además SAHA es un potente inhibidor de la migración celular en comparación con carboplatino. Esto sugiere que se requiere una mayor evaluación de los efectos de SAHA para considerarlos como un coadyuvante a la quimioterapia antineoplásica.

Full textA contaminação do vinho que produz cheiro a “mofo” é um problema que afeta a indústria corticeira. O principal agente contaminante é 2,4,6-TCA que, em solução alcoólica, o limiar de deteção humana é da ordem de 5 ng/L. As características da rolha de cortiça tornam difícil a sua descontaminação e os métodos de deteção disponíveis atualmente não permitem a aplicação na indústria. Neste trabalho, é explorada a deteção de gases halogenados pela formação de uma dupla camada na superfície de um emissor de iões alcalinos. Os iões alcalinos são emitidos por ionização superficial. A corrente iónica alcalina em vácuo segue, nas condições experimentais, a lei de Child-Langmuir e a função de trabalho é calculada num determinado intervalo de temperatura pela equação de Saha-Langmuir. Embora os resultados tenham tido um sucesso limitado, verificou-se que a introdução de clorofórmio (como exemplo de gás halogenado) produziu um aumento da corrente iónica que depende da temperatura e da pressão. A recuperação da corrente foi feita ao aquecer o emissor até cerca de 1200K durante alguns minutos. O campo eléctrico de extração que foi aplicado variou entre 30V/cm e 135V/cm em regime contínuo e a temperatura variou entre 800K e 1200K.
