Abstract:
Abstract The Caribbean spiny lobster, Panulirus argus (Latreille, 1804), is an important resource in the Western Atlantic Ocean, Caribbean Sea, and Gulf of Mexico. A nemertean egg predator of P. argus, Carcinonemertes conanobrieniSimpson, Ambrosio & Baeza, 2017, was recorded in Florida in 2017 and Colombia in 2018. This study sought to investigate the presence of C. conanobrieni in P. argus in Saint Kitts and Nevis, eastern Caribbean Sea. Nemerteans were observed in the brood masses of P. argus. Molecular techniques confirmed its presence in the P. argus population on Saint Kitts. Egg, juvenile, and adult stages of C. conanobrieni and of an undescribed Carcinonemertes sp. were found in the brood masses of 27 of 31 (87%) ovigerous P. argus. Differences in the shape of the nemertean egg masses (string versus spherical arrangement) were confirmed as two distinct species of Carcinonemertes Coe, 1902 using molecular analyses. The juvenile stage of the undescribed Carcinonemertes sp. was detected in the gills of 72 of 320 (23%) of the P. argus individuals from Saint Kitts. The prevalence of branchial nemertean infestation was not significantly different among male and female P. argus. Female lobsters with ovaries in early stages of gonadal maturation were more likely to have branchial nemerteans relative to those with ovaries in late stages of maturation, suggestive of nemertean migration from gills to egg mass once the host oviposits. This is the first report of C. conanobrieni in the Caribbean spiny lobster from the eastern Caribbean, which is approximately 1,300 miles from Florida and 1,000 miles from Colombia. This is also the first report of an undescribed nemertean worm in P. argus. The high prevalence of Carcinonemertes spp. in P. argus raises concern for the extent to which these egg predators may be impacting P. argus in Saint Kitts, an important fishery on the island.

Abstract:
The land snail species Drymaeus virgulatus (Férussac, 1821) is recorded in St. Kitts in the form of 21 complete or fragmented shells collected on the island in November 2019. The complete shells were collected on the backshores of South Frigate Bay and Majors Bay, while shell fragments come from naturally exposed sections of the North Frigate Bay sand ridge, where they date to the pre-Columbian period. This evidence therefore attests to the past presence of this species on the island. The absence of this taxon from recent malacological surveys points to it having been extirpated from St. Kitts. Apertural fragments of a large bulimulidae collected in the caves of Tintamarre Island, near St. Martin, are also related to this taxon, adding a second West Indian island from which this species apparently disappeared in recent centuries.

Abstract:
Leptospirosis is an important bacterial zoonotic disease that affects humans and many animal species. Knowledge of prevalence of Leptospira in a given geographic region supports the implementation of effective control/prevention programmes and thus reduces the transmission risks. This study provides Leptospira seroprevalence and serovar distribution in dogs and cats on the Caribbean island of Saint Kitts. Convenient serum samples from domestic dogs (n=101) and cats (n=50) were tested by the microscopic agglutination test (MAT) using 21 Leptospira serovars belonging to 17 serogroups. Seroprevalence was recorded at 73.2% in dogs (95% confidence interval CI: 62.5–80.1%). Agglutinating antibodies to Leptospira were present against 13 of the 21 serovars tested by MAT. The highest seroprevalence was observed for serovar Autumnalis (56.4%) followed by Icterohaemorrhagiae (27.7%), Canicola (17.8%), Djasiman (14.9%), Bratislava (11.9%), Pyrogenes (11.9%), and Pomona (7.9%). A very low seroprevalence (4%, 95% CI: 0.5–14%) was observed in cats. This data confirms that dogs in Saint Kitts have a high-level exposure to a diverse set of Leptospira serovars.

Abstract:
Dermatophilosis is a form of dermatitis caused by the bacterium Dermatophilus congolensis. The disease usually presents as localized purulent dermatitis, crusty hair masses or widespread matting of the hair. This condition is most common in domestic ruminants; but it can also affect other wild animals and humans. Antimicrobial therapy is used in many regions to treat clinical dermatophilosis with varying results. In this study, we aimed to assess the antimicrobial susceptibility of D. congolensis isolates. Fifty-two isolates were obtained from animals showing clinical signs of the disease at farms in St. Kitts. The isolates were then confirmed as D. congolensis by phenotypic tests, PCR and MALDI-TOF Mass Spectrometry. Furthermore, minimum inhibitory concentrations (MIC) of 16 antimicrobial agents were determined, using the broth microdilution method. Although most antimicrobials showed MICs in line with published values, the tetracycline results displayed a clear bimodal distribution over the tested range, with most isolates showing low MICs and 6 isolates much higher values (+/− 100-fold increase). These results indicate the presence of acquired tetracycline resistance in D. congolensis on the island of St. Kitts. Whether the current observation has implications for efficacy of treating the disease must be confirmed in further research.

A new species of deep-water brachyuran crab, Euphrosynoplax dincao, from southeastern Brazil is described and illustrated. The genus Euphrosynoplax currently consists of two species, E. clausa Guinot, 1969, and E. campechiensis Vázquez-Bader & Gracia, 1991, both only known from the Gulf of Mexico. The new species can be easily separated from its northern counterparts by a suite of carapace and appendage characters. Euphrosynoplax campechiensis is recorded for the first time from the Caribbenan Sea (Guadeloupe and between Saint Kitts and Nevis).

Chiroderma improvisum is a rare bat previously known only on the Caribbean Islands of Guadeloupe and Montserrat. We report the first recorded capture of C. improvisum on the island of Saint Kitts, 80 km northwest of Montserrat. Cytochrome b (cytB) gene analysis of the single captured specimen confirmed the identity of the bat as C. improvisum; however, there is enough difference to indicate some population divergence, and possibly differentiation at the subspecific level among islands. We also report the first records of an ectoparasite, Periglischrus iheringi (Acarina: Spinturnicidae), from this bat.

Antimicrobial resistance has been described in all ecosystems, including wildlife. Here we investigated the presence of methicillin-resistant and susceptible staphylococci in both colony-born and wild vervet monkeys (Chlorocebus sabaeus). Through selective isolation, PCR, MALDI-TOF, and whole-genome sequencing, methicillin-resistant and susceptible Staphylococcus spp. isolated from vervet monkeys were characterized. We obtained putatively methicillin-resistant staphylococci from 29 of the 34 nasal samples collected. Strains were identified by MALDI-TOF analysis. Staphylococcus cohnii (n = 15) was the most commonly isolated species, while nine other species were isolated one or two times. PCR analysis indicated that eight [28%] strains were mecA positive. The whole-genome sequencing [WGS] included eight methicillin-resistant strains (S. epidermidis (n = 2), S. cohnii (n = 3), S. arlettae (n = 2) and S. hominis (n = 1)), nine additional S. cohnii strains and two strains that could not be identified by MALDI-TOF, but genetically characterized as one S. cohnii and one S. warneri. Different resistance genes carried by different mobile genetic elements, mainly blaZ (n = 10) and tet(K) (n = 5) were found, while msr(A), cat, fosB, dfrG, erm(C), mph(C) and str were identified in one to three strains. Phylogenetic analysis of the S. cohnii strains based on SNPs indicated four clusters associated with colony born or wild. In addition, one singleton S. cohnii isolated did not form a separate group and clustered within other S. cohnii strains submitted to the NCBI. In this study, we demonstrated the presence of AMR and mobile genetic elements to both colony-born and wild vervet monkeys. We also identified a previously undescribed prevalence of S. cohnii in the nasal flora of these monkeys, which merits further investigation.

Introduction: After a large outbreak of dengue virus (DENV) serotype-3 in Saint Kitts and Nevis (SKN) in 2008, we performed a cross-sectional study to determine the prevalence of anti-DENV immunoglobulin G (IgG) antibodies in expatriate and local persons affiliated with an American veterinary school there. Methodology: This campus community comprised mostly expatriate students and faculty and Kittitian administrative staff. In 2009, a stratified random sample of students, faculty and staff was invited to complete an electronic survey to assess risk factors for DENV and provide blood for testing for anti-DENV IgG antibodies by an enzyme-linked immunosorbent assay. IgG-positive specimens were also tested by a 90% plaque reduction neutralization test (PRNT90) to determine immunoreactivity to DENV (1-4) serotypes and West Nile virus. Risk factors for anti-DENV IgG seropositivity were determined using simple and adjusted logistic regression. Results: Of the 118 participants, the overall prevalence of DENV IgG antibodies was 44.1% (95% confidence interval [CI]: 35.1-53.0%), ranging from 30.1% in students, 100.0% in staff and 57.9% in faculty (p < 0.001). Duration of residence in St. Kitts was the only variable significantly associated with seropositivity on multiple logistic regression (adjusted odds ratio [95% CI]: 1.21 [1.07-1.37]). The serotype of DENV was determined in 11 persons: DENV-1 (n = 4), DENV-2 (n = 3), and DENV-3 (n = 4). Conclusions: Expatriate students and faculty moving to St. Kitts from non-endemic areas were at high risk of DENV infection. There is a need for increased emphasis on pre-travel mosquito-borne virus prevention education for persons moving to St. Kitts to study and work.

In this study, we detected and characterized Leptospira infection and exposure in rats on the Caribbean island of Saint Kitts for the first time. We detected Leptospira infection in 17/29 (59%), 14/29 (48)%, and 11/29 (38)% of rats by RT-PCR, culture, and immunofluorescence assay, respectively. Whole genome sequencing (WGS) and analysis and serogrouping of 17 Leptospira strains isolated from rats revealed their close relationship with L. interrogans serogroup Icterohaemorrhagiae (n = 10) and L. borgpetersenii serogroup Ballum (n = 7). WGS, serogrouping, and additional PCR tests on rat kidneys confirmed mixed infections with L. interrogans and L. borgpetersenii in the kidneys of three rats. Microscopic agglutination test (MAT) was positive for 25/29 (87%) of the rats tested, and the response was restricted to serovars Icterohaemorrhagiae {24/29(83%)}, Mankarso {4/29(14%)}, Copenhageni {4/29(14%)}, Grippotyphosa {2/29(7%)}, and Wolffi {1/29(3%)}. Interestingly, there was no agglutinating antibody response to serovar Ballum. We observed a similar pattern in the serologic response using Leptospira isolates obtained from this study with each of the rat sera, with strong response to L. interrogans isolates but minimal reactivity to L. borgpetersenii isolates. Our findings suggest the use of multiple complementary diagnostic tests for Leptospira surveillance and diagnosis to improve the accuracy of the data.

Schistosomiasis elimination status in the Caribbean is reviewed with information on historical disease background, attempts to control it and current situation for each locality in the region where transmission has been eliminated (Sint Maarten, Saint Kitts, Vieques), eliminated but not yet verified (Puerto Rico, Dominican Republic, Antigua, Montserrat, Guadeloupe, Martinique) and still ongoing (Saint Lucia, Suriname). Integrated control initiatives based on selective and mass treatment and snail control using environmental, chemical and biological methods along with public service improvements (housing, safe water, sanitation) and changes in demography (urbanization) and economy (change from sugarcane and banana production to tourism) have resulted in reduction in the burden of schistosomiasis over the past century. Introduction of Biomphalaria-competitor snails into the region as a cost-effective, low maintenance control method appears to have had the most sustainable impact on transmission reduction. A regional inventory of B. glabrata, other Biomphalaria species and Biomphalaria-competitor snails as well as investigation of possible animal reservoir hosts in persisting endemic areas would be helpful for control. Elimination of schistosomiasis appears achievable in the Caribbean. However, a regional surveillance and monitoring program is needed to verify elimination in the various localities and identify and monitor areas still endemic or at risk.

The spider genus Oxyopes Latreille occurs throughout the world, but is insufficiently known in South America and the Caribbean. In this study, three apparently related species of this genus are reported from the region. Oxyopes salticus Hentz, 1845 is redescribed and illustrated, and extreme variation in colour pattern and in female genitalia is reported within this species. Oxyopes m-fasciatus Piza, 1938 and Oxyopes nigrolineatus Mello-Leitão, 1941 are both considered junior synonyms of O. salticus. The material examined in this study, together with information from the literature, show this species is widely distributed throughout the continental Americas and the Caribbean. Oxyopes crewi Bryant, 1948 is redescribed and illustrated, and its geographic distribution is extended to Bahamas, Cuba, Dominican Republic, Jamaica, and Saint Kitts. A new species, Oxyopes incantatus sp. nov., is described and illustrated for the first time based on specimens from the Galápagos Islands.

AbstractThis article presents a political economic analysis of exit from federations. After the federation has formed, members’ benefits from it may be different than expected. If a member ends up not benefitting, it may wish to secede i.e. exit the federation. Based on formal models, we show how state-contingent exit penalties can induce socially efficient exit decisions: they force the secessionist member to take into account the lost benefits of the federation for the other. Even if ex-ante specified exit penalties cannot be made state-contingent, they may still enhance social welfare by preventing forceful exit. Empirical evidence concerning Montenegro, Saint Kitts and Nevis, and the EU is compatible with these claims. In spite of the simplifications inherent to any modeling exercise, we hope that our results stimulate more research into exit clauses as a means to mitigate the problem of violent secessionism.

L'efficacité du Lamstreptocide A et B a été étudiée sur 9 cas naturels de dermatophilose bovine et caprine dans 8 fermes différentes de Saint Kitts, par des méthodes histopathologiques et bactériologiques classiques. Les lésions de 5 des ruminants traités ont séché et les croûtes d'un animal gravement atteint se sont décollées considérablement, laissant apparaître un tissu érythémateux, 3 semaines après l'application du produit. En dehors de 3 cas sans gravité qui n'ont pu être suivis et qui seraient guéris, aucun des 5 animaux n'était véritablement guéri 3 semaines après le traitement, ni même après une deuxième application du produit. Un test in vitro sur la sensibilité au produit appliqué en surface de gélose, à des concentrations de plus de 1 p. 100, a révélé un ralentissement de la croissance de Dermatophilus congolensis. Néanmoins, il n'y avait pas d'inhibition de la croissance de la bactérie par des disques de papier filtre imprégné.

The Revised Treaty of Chaguaramas (‘the RTC’) is an attempt on the part of a group of Caribbean States to respond in a collective manner to the pressing challenges posed by the forces of globalization and liberalization. The RTC seeks, inter alia, to deepen regional economic integration through the establishment of a Caribbean Community (‘CARICOM’) including a CARICOM Single Market and Economy (‘CSME’). The States in question—Antigua & Barbuda, The Bahamas, Barbados, Belize, Dominica, Grenada, Guyana, Haiti, Jamaica, Montserrat, St Kitts and Nevis, Saint Lucia, St Vincent and the Grenadines, Suriname and Trinidad & Tobago—are for the most part former British colonies that gained their independence in the 1960s and 1970s. The RTC signals yet another important step in the tortuous path taken by these Anglophone Caribbean States ‘to avoid the looming threat of marginalization’1 following the failure in 1962 of the West Indies Federation.2 Significantly, this latest step is being taken side by side with the non English speaking civil law States of Haiti and Suriname thereby adding a new and interesting dimension to the integration process.

This study aimed to molecularly survey and evaluate the genetic diversity of Bartonella spp. in mongooses and their fleas from St. Kitts. Spleen (n = 54), blood (n = 71), and pooled flea samples, all identified as Ctenocephalides felis (n = 53), were submitted to TaqMan real-time quantitative PCR (qPCR) targeting Bartonella-nuoG fragment (84 bp). Positive samples underwent further conventional PCR assays targeting five loci (gltA, rpoB, fstZ, nuoG, and ITS), subsequent sequencing, and phylogenetic and haplotype analyses. The overall occurrence of Bartonella spp. in mongooses and fleas was 51.2% (64/125 [95% CI (42.1–60.2%)]) and 62.3% (33/53) [95% CI (47.9–75.2%)]), respectively. From samples sequenced across the five loci, 50.8% (33/65) were identified as Bartonella henselae, 26.2% (17/65) were 96.74–99.01% similar by BLAST analysis to an unidentified Bartonella sp. previously reported in Japanese badgers (Meles anakuma), and 23.1% (15/65) were co-infected with both species. Nucleotide polymorphism analysis showed low diversity amongst haplotypes but did concur with phylogenetic analysis, placing the unidentified species in a separate clade from B. henselae by multiple mutational events. Our data confirms that mongooses and Ctenocephalides felis fleas collected from them are not only potential reservoirs for B. henselae but also a novel Bartonella sp. which we propose be called ‘Candidatus Bartonella kittensis’.

This study was performed to investigate the potential asymptomatic Leptospira reservoir status among African green monkeys (AGMs) in the Caribbean island of Saint Kitts, and whether there is any renal pathology associated with Leptospira exposure. Forty-eight percent of AGMs tested were positive for Leptospira antibodies by the microscopic agglutination test. Leptospira DNA was detected in 4% of kidney samples tested using a lipl32 gene based PCR. We observed minimal to severe microscopic renal lesions in 85% of the AGM kidneys evaluated. The majority of the AGMs (n = 26) had only minimal to mild interstitial nephritis and a few (n = 3) had moderate to severe lesions. The presence of interstitial nephritis was not significantly associated with Leptospira exposure. The presence of infected AGMs in a small surface limited geographic region may pose zoonotic threat to humans and animals. The impact of Leptospira infection in renal pathology in AGMs warrants further investigation. AGMs residing in a natural setting in an insular, surface limited Leptospira endemic geographic region may offer opportunities for comparative studies to advance the field of leptospirosis. Due to their similarity to humans, such studies in AGMs may also provide translational opportunities to advance Leptospira research.

Anaplasma platys and Ehrlichia canis are obligate intracellular, tick-borne rickettsial pathogens of dogs that may cause life-threatening diseases. In this study, we assessed the usefulness of PCR and a widely used commercial antibody-based point-of-care (POC) test to diagnose A. platys and E. canis infection and updated the prevalence of these pathogens in dogs inhabiting the Caribbean island of Saint Kitts. We detected A. platys in 62/227 (27%), E. canis in 84/227 (37%), and the presence of both in 43/227 (19%) of the dogs using PCR. POC testing was positive for A. platys in 53/187 (28%), E. canis in 112/187 (60%), and for both in 42/187 (22%) of the samples tested. There was only a slight agreement between A. platys PCR and POC test results and a fair agreement for E. canis PCR and POC test results. Our study suggests that PCR testing may be particularly useful in the early stage of infection when antibody levels are low or undetectable, whereas, POC test is useful when false-negative PCR results occur due to low bacteremia. A combination of PCR and POC tests may increase the ability to diagnose A. platys and E. canis infection and consequently will improve patient management.

Small Indian mongooses (Urva auropunctata) are among the most pervasive predators to disrupt the native ecology on Caribbean islands and are strongly entrenched in their areas of introduction. Few studies, however, have considered the microbial ecology of such biological invasions. In this study, we investigated the gut microbiota of invasive small Indian mongooses in terms of taxonomic diversity and functional potential. To this end, we collected fecal samples from 60 free-roaming mongooses trapped in different vegetation zones on the island Saint Kitts. The core gut microbiome, assessed by 16S rRNA amplicon gene sequencing on the Ion S5TM XL platform, reflects a carnivore-like signature with a dominant abundance of Firmicutes (54.96%), followed by Proteobacteria (13.98%) and Fusobacteria (12.39%), and a relatively minor contribution of Actinobacteria (10.4%) and Bacteroidetes (6.40%). Mongooses trapped at coastal sites exhibited a higher relative abundance of Fusobacterium spp. whereas those trapped in scrubland areas were enriched in Bacteroidetes, but there was no site-specific difference in predicted metabolic properties. Between males and females, beta-diversity was not significantly different and no sex-specific strategies for energy production were observed. However, the relative abundance of Gammaproteobacteria, and more specifically, Enterobacteriaceae, was significantly higher in males. This first description of the microbial profile of small Indian mongooses provides new insights into their bioecology and can serve as a springboard to further elucidating this invasive predator’s impact throughout the Caribbean.

The Memorandum of Understanding (MOU) on Port State Control in the Caribbean Region is the fourth in a series commencing in 1982 with the Paris Memorandum (Europe). The other two cover the Latin American (1992) and Asia Pacific (1993) regions, and further regional developments are in the offing. These documents represent an innovative exercise by port states at the regional level to guard against accidents and pollution caused by vessels that do not conform with generally accepted international rules and standards. Following the practice of the Paris MOU, most also ensure conformity with international standards for seafarers to ensure acceptable living and working conditions on board ships. Each MOU identifies the relevant rules by reference to specific conventions adopted under the auspices of the International Maritime Organization (IMO) and the International Labor Organization (ILO).

Studies investigating perceived stress and mindfulness awareness support mind-body therapy (MBT) effectiveness in reducing stress and anxiety and, thus, has potential to decrease work-related stress. A pre/postexperimental design involved 30 faculty and staff working at Ross University School of Veterinary Medicine, Saint Kitts and Nevis, who experienced a two-day MBT intervention programme. An additional 16 faculty and staff not involved in MBT who went about their daily work schedules served as contemporary controls. Demographics, Perceived Stress Scale 10 (PSS-10), Mindful Attention Awareness Scale (MAAS), 16 Personality Factor (16PF) Openness to Change subscale and saliva cortisol concentrations were analysed. Control participants reported significantly perceived less stress (PSS-10: M=13; sd=1.4) than intervention participants (M=20; sd=6.6) during pretest. However, at post-test the intervention group reported a significant decrease in perceived stress (M=11; sd=6.0). MAAS pretest results indicated the intervention group displayed a lower average score (M=54; sd=15.3) than control participants (M=68; sd=2.0). Post-MAAS intervention scores showed improvements in mindfulness (M=63; sd=15.3). Correlations between the 16PF Openness to Change subscale and MAAS were r=0.03 and r=−0.17 for the intervention and control groups, respectively. Mean concentrations of saliva cortisol indicated a larger and significant decline in cortisol for the intervention group both during day 1 (P=0.0001) and day 2 (P=0.0008). In conclusion, these preliminary results provide support that MBTs in veterinary academia can improve psychological and physiological aspects of personal wellbeing.

The Caribbean spiny lobster Panulirus argus, a long-lived decapod crustacean, is fished throughout its geographical range and generates close to 1 billion US dollars annually. Fisheries across this range are fully- or over-exploited, spurring management strategies such as closed seasons and minimum size limitations. The objective of this study was to determine the minimum legal size and appropriate closed season for the lobster population of Saint Kitts and Nevis by assessing and comparing the functional, physiological and morphological determinants of sexual maturity. Such a comparison has never been attempted for this species. Females did not reach functional maturity until 87.8 mm carapace length (CL) based on the presence of a spermatophore and eggs, but reached morphological maturity based on the presence of ovigerous setae at 72.5 mm CL. Physiological maturity was intermediate at 82.8 mm CL. Determinants of functional maturity overestimate the size at sexual maturity, while the presence of ovigerous setae underestimates it. The gonadosomatic index, a reliable method of assessing physiological maturity in female P. argus, can be used to monitor changes in reproductive activity in both males and females. Gross assessment of the ovary may be used to determine size at physiological maturity, and functional maturity can be used to calculate peak periods of reproduction. The peak physiological and functional maturity for P. argus occurs from March to May. As a result, we propose a closed season from March to June and suggest assessment of reproductive dynamics in order to determine minimum legal size for harvesting P. argus.

During April of 2002, symptoms of stunting and chlorotic curled leaves of reduced size, similar to those caused by Tomato yellow leaf curl virus (TYLCV), were observed for the first time in commercial tomato (Solanum lycopersicum) in the northwest region of Martinique. Six months later, many tomato fields had more than 80% of plants expressing these symptoms and yield was drastically reduced. Samples from two symptomatic plants were collected and analyzed by PCR. Primers PC1 (5′-TGACTATGTCGAAGCGACCAGG-3′) and PC2 (5′-CGACATTACAGCCTCAGACTGG-3′) were used to amplify a 950-bp fragment within the coat protein gene (CP) of TYLCV species (1). Primer pair MP16-MP82 (2) amplified a 550-bp fragment from the conserved nonanucleotide sequence (TAATATTAC) to the 5′ end of the CP gene. Products of expected sizes were obtained with both pairs of primers from symptomatic samples but not from uninfected ones. The two overlapping PCR products were cloned into a pGEM-T Easy Vector (Promega, Madison, WI) and sequenced. A BLAST analysis was conducted with begomovirus sequences available in the GenBank database at the NCBI, and DNAMAN software (Lynnon Corporation, Quebec, Canada) was used for further comparisons. The 1275-bp sequence (GenBank Accession No. EF490995) shared 99% nucleotide identity with the partial sequences of TYLCV from Antigua and Barbuda (GenBank Accession No. EF028240), Saint Kitts and Nevis (GenBank Accession No. EF028239), and the two overlapping sequences from Guadeloupe (GenBank Accessions No. AY319645 and AY319646). It was at least 98% identical to TYLCV isolates from Florida (GenBank Accession No AY530931), Dominican Republic (GenBank Accession No. AF024715), and Cuba (GenBank Accession No. AJ223505). These results confirm the introduction of TYLCV into Martinique, possibly from a nearby Caribbean country, and reveal its southward spread in the Lesser Antilles. The nearness of the islands in the Lesser Antilles (20 to 100 km distant) probably permitted the rapid spread of TYLCV through the movement of plant material or wind transport of viruliferous whiteflies from one island to the next. Monitoring the spread of TYLCV in this Caribbean archipelago is important for regional virus management and in forecasting the spread of TYLCV to nearby countries in South America. References: (1) Y. In December 1995, leaf scald symptoms were observed in sugarcane (Saccharum sp.) cultivar B64277 in French Guyana. Symptomatic plants occurred both in a sugarcane germplasm collection near the road between Sinnamary and Saint-Elie and in a nursery near Sinnamary. Sugarcane imported from Martinique had been used to establish the germplasm collection that in turn had been used to establish the nursery. Ten-month-old mature plants in the germplasm collection had abnormal side shoots on the lower part of the stalks and suckers (nonmillable stalks) with white scalded areas on leaves. Leaves on 1-month-old shoots in the nursery exhibited chlorosis and white, pencil-line streaks. Samples prepared from symptomatic stalks from the two locations were plated on a selective medium (1), and two isolates of Xanthomonas albilineans were recovered. Both of these isolates caused leaf scald symptoms on leaves of sugarcane cultivar B69566 inoculated by a decapitation technique, and belong to serovar 3 previously reported in the Caribbean from Guadeloupe, Martinique, and St. Kitts. The RFLP (restriction fragment length polymorphism) pattern of these two isolates was different from the 54 patterns among 218 other strains collected throughout the world (2), but similar to the pattern of a strain of serovar 3 from Martinique. This indicated that the pathogen might have been introduced with cuttings imported from Martinique. Three stalks of mature cane from varieties B5992, B64277, and R570 from the germplasm collection were tested for the presence of Clavibacter xyli subsp. xyli, causal agent of ratoon stunting disease. Immunofluorescence tests on sap (3) revealed the presence of the pathogen in the three stalks of B64277. All sugarcane plants in the nursery and the germplasm collection were destroyed by the use of glyphosate sprays in January 1996 in an attempt to arrest the spread of the two bacterial pathogens. In order to obtain healthy seed cane for future planting, a new germplasm collection of 0.6 ha and consisting of 11 cultivars was planted in January 1996 with disease-free, tissue-cultured plants provided by the CIRAD sugarcane breeding station in Guadeloupe. The present paper discusses the results of comprehensive historical-and-archive, cartographic, bibliographic and morphological study on the development stage of Orthodox temple construction in Saint Petersburg and surrounding areas at the beginning of 1917. The integral spatial area of citizens’ seasonal life was identified, within the boundaries of which the common trend of Orthodox temple placing was formed. This area included Saint Petersburg, its suburban police districts, the nearest territories, called uyezds, and the following towns under royal administration: Gatchina, Krasnoye Selo, Pavlovsk, Petergof, Oranienbaum, Strelna, Tsarskoye Selo. This integral metropolitan functional area had in total 987 cathedrals, churches, chapels and kiots; particularly in Saint Petersburg there were 676 temples, which meant that approximately each twelve developed land plots had one temple. Such an extraordinary territorial density of placing temples formed a special multilayer system of visual and skyline composition of town planning, which completely differs from a modern one that appeared after mass destructions of 1920 – 1960.

Penelitian ini merupakan penelitian pengembangan (research and development) model Borg & Gall dengan melaksanakan: studi pendahuluan, desain produk, penyusunan draft penelitian, validasi instrumen, uji coba dan revisi produk, dan penyempurnaan produk akhir. Subjek coba dalam penelitian ini adalah siswa SMA Negeri 1 Pakem kelas X MIA 2 dan X MIA 3. Data yang diperoleh adalah data kualitatif dan kuantitatif. Hasil penelitian ini menunjukkan bahwa: (1) perangkat pembelajaran dengan kegiatan ecotourism yang dikembangkan, berdasarkan hasil validasi secara keseluruhan memiliki kualitas baik, (2) perkebunan buah naga Sabila Farm mempunyai potensi biologis untuk menunjang pembelajaran biologi kelas X SMA pada materi keanekaragaman hayati, (3) perangkat pembelajaran yang dikembangkan dengan kegiatan ecotourism dapat mengukur keterampilan proses sains dan sikap peduli lingkungan peserta didik. Abstract:
ABSTRACTA study comparing the ICT (immunochromatography technology)ToxoplasmaIgG and IgM rapid diagnostic test (LDBio Diagnostics, France) with a fully automated system, Architect, was performed on samples from university hospitals of Marseille and Saint-Etienne. A total of 767 prospective sera and 235 selected sera were collected. The panels were selected to test various IgG and IgM parameters. The reference technique,ToxoplasmaIgGII Western blot analysis (LDBio Diagnostics), was used to confirm the IgG results, and commercial kits Platelia Toxo IgM (Bio-Rad) and Toxo-ISAgA (bioMérieux) were used in Saint-Etienne and Marseille, respectively, as the IgM reference techniques. Sensitivity and specificity of the ICT and the Architect IgG assays were compared using a prospective panel. Sensitivity was 100% for the ICT test and 92.1% for Architect (cutoff at 1.6 IU/ml). The low-IgG-titer serum results confirmed that ICT sensitivity was superior to that of Architect. Specificity was 98.7% (ICT) and 99.8% (Architect IgG). The ICT test is also useful for detecting IgM without IgG and is both sensitive (100%) and specific (100%), as it can distinguish nonspecific IgM from specificToxoplasmaIgM. In comparison, IgM sensitivity and specificity on Architect are 96.1% and 99.6%, respectively (cutoff at 0.5 arbitrary units [AU]/ml). To conclude, this new test overcomes the limitations of automated screening techniques, which are not sensitive enough for IgG and lack specificity for IgM (rare IgM false-positive cases).

Abstract:
ABSTRACTSerodiagnosis of amoebiasis remains the preferred method for diagnosis of amoebic liver abscess (ALA). However, the commercially available kits are problematic in areas of endemicity due to the persistently high background antibody titers. Human serum samples (n= 38) from patients with ALA who live in areas of endemicity were collected from Hospital Universiti Sains Malaysia during the period of 2008 to 2010. Western blots using excretory-secretory antigen (ESA) collected from axenically grownEntamoeba histolyticawere probed with the above serum samples. Seven antigenic proteins of ESA with various reactivities were identified, i.e., 152 kDa, 131 kDa, 123 kDa, 110 kDa, 100 kDa, 82 kDa, and 76 kDa. However, only the 152-kDa and 110-kDa proteins showed sensitivities above 80% in the Western blot analysis. All the antigenic proteins showed undetectable cross-reactivity when probed with healthy human serum samples (n= 30) and serum samples from other infections (n= 33). From the matrix-assisted laser desorption ionization–two-stage time of flight (MALDI-TOF/TOF) analysis, the proteins were identified as heavy subunits ofE. histolyticalectin andE. histolyticapyruvate phosphate dikinase, respectively. Use of theE. histolyticalectin for diagnosis of ALA has been well reported by researchers and is being used in commercialized kits. However, this is the first report on the potential use of pyruvate phosphate dikinase for diagnosis of ALA; thus, this molecule merits further evaluation on its diagnostic value using a larger panel of serum samples.

ABSTRAKKegiatan pengabdian kepada masyarakat ini dilakukan guna meningkatkan kemampuan penggunaan dan pemanfaatan set eksperimen Komponen Instrumen Terpadu (KIT) IPA untuk penggalian konsep-konsep dasar Fisika siswa melalui pembelajaran berbasis eksperimen. Kegiatan pengabdian dilaksanakan di SMPN 2 Sekotong Kabupaten Lombok Barat, dengan peserta guru-guru Fisika dan siswa-siswa kelas 8. Metode yang digunakan dalam kegiatan ini adalah direct instruction, cooperative learning dan diskusi. Adapun set eksperimen yang digunakan adalah KIT IPA untuk materi Mekanika, Panas dan Hidrostatistika, Optik, dan Listrik Magnet. Pada kegiatan ini peserta memperoleh pengetahuan tentang metode pembelajaran berbasis eksperimen dan keterampilan merakit serta memodifikasi KIT IPA yang dapat digunakan untuk mendukung pembelajaran Sains. Dengan demikian, pembelajaran Sains akan menjadi pembelajaran yang menyenangkan dan diharapkan meningkatkan minat dan prestasi belajar siswa. ABSTRACTThis community service activity is carried out to improve the ability to use the IPA Integrated Instrument Components (IIC) experimental set to explore the student basic concepts of physics through experiment-based learning. This activity was carried out at SMPN 2 Sekotong West Lombok Regency with participants from physics teachers and 8th-grade students. The method that used is direct instruction, cooperative learning and discussion. The experimental set used is the IPA IIC for the materials of Mechanics, Heat and Hydrostatic, Optics, and Magnetic Electricity. In this activity, participants gain knowledge about experimental based learning methods and skills of assembling and modifying science KITs that can be used to support science learning. Thus, science learning will be fun learning and it is expected to increase student interest and learning achievement. Keywords: IIC; physics concept; experiment; skills; learning interest.

Abstract:
Hereditary neuromuscular diseases are a group of genetic diseases characterized by an onset of the disease in most cases in childhood, having a steadily progressive course of the pathological process, leading to more rapid disability of patients and having a high mortality rate at the age of 18–20 years.Objective. To study a condition of the intra-organ structure in patients with hereditary muscular atrophy and muscular dystrophy using testing of nerve tissue in the organotypic environment in order to justify the prescription of symptomatic neuroprotective therapy.Materials and methods. Ninety patients with hereditary neuromuscular diseases (spinal muscular atrophy types 1, 2 and 3 [n = 30], Duchenne muscular dystrophy [n = 60]) were examined; the control group consisted of 30 healthy people. In vitro – explants of sensory ganglia of 10–12-day-old chicken embryos. A comprehensive clinical, laboratory and experimental study was conducted. Concentrations of neurotrophic factors (Brain Growth Factor, Nerve Growth Factor, Ciliary Neurotrophic Factor) were determined by the enzyme immunoassay method in blood plasma samples using RayBiotech kits and in accordance with the manufacturer's instructions. The experimental study included 300 explants of sensory ganglia of 10–12-day chicken embryos cultured in Petri dishes on collagen substrates in a CO2 incubator (Sanyo, Japan) for 3 days at 36.5 °C and 5% CO2. In order to clarify the biochemical mechanisms involved in pathological cascades in patients with hereditary neuromuscular diseases, a test system was developed that included a sequential analysis of the patient's blood plasma in an organotypic tissue culture at a dilution of 1: 70, followed by an addition of reagents to the medium: synthetic nerve growth factor (NGF) (100 pg/ml). Explant cultivation was carried out according to the method developed at Institute for Physiology n.a. I.P. Pavlov (Saint Petersburg, Russia). Visualization of the objects was made using Axiostar Plus microscope (Carl Zeiss, Germany). The resulting images were analyzed with the help of ImageJ software. A morphometric method was used to quantify the growth of explants. The area index (AI) was calculated as the ratio of the area of the explant growth zone to the initial area. AI reference value was 100%.Results. Brain Growth Factor concentration was at the highest level in patients with progressive amyotrophy, while in patients with progressive myodystrophy, the blood concentra tion of this factor was at a level comparable to the control data, and in some patients the concentration of Brain Growth Factor was lower than normal. NGF concentration showed the highest values in the group of patients with progressive amyotrophies. Blood plasma of patients with progressive amyotrophy dose dependently inhibits the growth of neurites of the spinal ganglia, and blood plasma of patients with myodystrophy has a neurite-weakening effect on the growth of neurites. Introduction of synthetic NGF (100 pg/ml) to organotypic tissue culture containing blood plasma of patients with myodystrophy increased the area index value of 114.0 [111.0; 116.0]%; in explants containing blood plasma of patients with progressive amyotrophy, increased growth of neurites was not observed AI = 80.0 [74.5; 83.0]%.Conclusion. The data obtained are indicative of features of neurotrophic regulation in patients with hereditary muscular atrophy and muscular dystrophy, which should be taken into account when conducting symptomatic treatment aimed at stimulating reparative processes in the nervous tissue. We recommend patients with the neurite-weakening effect of blood plasma to have neuroprotective drugs therapy, and in case of patients with neuritis-inhibiting effect on neurites in organotypic culture of nervous tissue we recommend choosing a drug in vitro individually using pharmacological analysis.

Abstract:
The aim of the study was to evaluated the effect of giving water hyacinth and fish oil in quail diet on the performance, egg cholesterol and egg fatty acid profile. This study used 200 unsex day old quail divided into 5 treatments and 4 replications that were reared 5 weeks and then selected male or female. The results of sexing quail at week 5 were 95 females and 86 males. Growth period diets (0-5 weeks) contains 24% crude protein and metabolic energy of 2900 kcal kg-1 and production period diets (5-12 weeks) contains 22% crude protein and 2900 kcal kg-1. The experimental design used a completely randomized design with the treatment of using water hyacinth that was 0%,1%, 2%, 3 % and 4% with fish oil which was 0%, 4%, 5%, 6% and 7% in quail diet. The variables measured were quail performance (0-5 week age and 5-12 week age), egg cholesterol, and egg fatty acid profile. The results showed that using of water hyacinth meal 1% -4% and fish oil 4%-5% in the diet of the growth period resulted in higher feed consumption and body weight gain than diet without water hyacinth and fish oil but feed conversion rasio were same. Quail performance of the production period was not affected by all treatments. Unsaturated fatty acids tend to increased in quail eggs. The lowest cholesterol level of quail eggs was by giving 2% of water hyacinth meal and 5% of fish oil in the diet. It was concluded that water hyacinth can be used as local feed ingredients in quail diet until 4%.
