Abstract:
In the domain of cyber security, the defence mechanisms of networks has traditionally been placed in a reactionary role. Cyber security professionals are therefore disadvantaged in a cyber-attack situation due to the fact that it is vital that they maneuver such attacks before the network is totally compromised. In this paper, we utilize the Betweenness Centrality network measure (social property) to discover possible cyber-attack paths and then employ computation of similar personality of nodes/users to generate predictions about possible attacks within the network. Our method proposes a social recommender algorithm called socially-aware recommendation of cyber-attack paths (SARCP), as an attack predictor in the cyber security defence domain. In a social network, SARCP exploits and delivers all possible paths which can result in cyber-attacks. Using a real-world dataset and relevant evaluation metrics, experimental results in the paper show that our proposed method is favorable and effective.

Abstract:
The comparative study results of pollen morphology of the mutational witches’ brooms and the normal part of the tree crown in Scots pine are presented. There is a decrease of pollen grains size, especially the sacci, in witches’ brooms. The witches’ brooms with more intensive branching have more expressive changes. Also, the witches’ brooms have more abnormal pollen grains. The data on the occurrence about 10 anomalous morphotypes of pollen grains are reported, most of that are related to the abnormal development of succi: different size of sacci, deformed sacci, reduced sacci, fused sacci, additional sacci, compress sacci, lack of one or both sacci. A high percentage of anomalies in the sacci development may be associated with less developed reticular sculpture of ectexine in witches’ brooms pollen.

Abstract:
Sarco(endo)plasmic reticulum calcium adenosine triphosphatase is responsible for transporting cytosolic calcium into the sarcoplasmic reticulum and endoplasmic reticulum to maintain calcium homeostasis. Sarco(endo)plasmic reticulum calcium adenosine triphosphatase is the dominant isoform expressed in cardiac tissue, which is regulated by endogenous protein inhibitors, post-translational modifications, hormones as well as microRNAs. Dysfunction of sarco(endo)plasmic reticulum calcium adenosine triphosphatase is associated with heart failure, which makes sarco(endo)plasmic reticulum calcium adenosine triphosphatase a promising target for heart failure therapy. This review summarizes current approaches to ameliorate sarco(endo)plasmic reticulum calcium adenosine triphosphatase function and focuses on phospholamban, an endogenous inhibitor of sarco(endo)plasmic reticulum calcium adenosine triphosphatase, pharmacological tools and gene therapies.

Abstract:
The alpha-sarcin cytotoxin is an extracellular fungal protein that inhibits protein biosynthesis by specifically cleaving one phosphodiester bond of the 28 S rRNA. The His137 residue of alpha-sarcin is suggested to be involved in the catalytic activity of this protein, based on the observed sequence similarity with some fungal ribonucleases. Replacement of this residue by Gln (H137Q mutant variant of alpha-sarcin) abolishes the ribonuclease activity of the protein. This has been demonstrated for an homogeneous preparation of the H137Q alpha-sarcin by measuring its effect against both intact rabbit ribosomes and the homopolymer poly(A). The conformation of H137Q alpha-sarcin is highly similar to that of the wild-type protein, which has been analysed by CD and fluorescence spectroscopy. Both H137Q and wild-type alpha-sarcin exhibit identical CD spectra in the peptide-bond region, indicating that no changes at the level of the secondary structure are produced upon mutation. Only minor differences are observed in both near-UV CD and fluorescence emission spectra in comparison to those of the wild-type protein. Moreover, H137Q alpha-sarcin interacts with phospholipid vesicles, promoting the same effects as the native cytotoxin. Therefore, we propose that His137 is part of the ribonucleolytic active site of the cytotoxin alpha-sarcin.

Abstract:
alpha-Sarcin is a cytotoxic protein produced by the mould Aspergillus giganteus. Insertion of alpha-sarcin into asolectin membranes has been demonstrated by protein labelling with photoreactive phospholipids. alpha-Sarcin added externally to tRNA-containing asolectin liposomes degrades the entrapped tRNA. Trypsin-containing asolectin liposomes were also prepared. Encapsulated trypsin degrades alpha-sarcin, even in the presence of a large excess of external hen egg-white trypsin inhibitor to prevent any alpha-sarin degradation outside the vesicles. These processes occur only with acidic phospholipids and were not observed when phosphatidylcholine vesicles were used. These results indicate that alpha-sarcin penetrates the lipid bilayer and becomes exposed to the lumen of negatively charged liposomes.

Abstract:
Abstract The ribotoxin α-sarcin belongs to a family of ribonucleases that cleave the sarcin/ricin loop (SRL), a critical functional rRNA element within the large ribosomal subunit (60S), thereby abolishing translation. Whether α-sarcin targets the SRL only in mature 60S subunits remains unresolved. Here, we show that, in yeast, α-sarcin can cleave SRLs within late 60S pre-ribosomes containing mature 25S rRNA but not nucleolar/nuclear 60S pre-ribosomes containing 27S pre-rRNA in vivo. Conditional expression of α-sarcin is lethal, but does not impede early pre-rRNA processing, nuclear export and the cytoplasmic maturation of 60S pre-ribosomes. Thus, SRL-cleaved containing late 60S pre-ribosomes seem to escape cytoplasmic proofreading steps. Polysome analyses revealed that SRL-cleaved 60S ribosomal subunits form 80S initiation complexes, but fail to progress to the step of translation elongation. We suggest that the functional integrity of a α-sarcin cleaved SRL might be assessed only during translation.

Abstract:
The aim was to apply AWGS criteria to estimate the prevalence of sarco-osteoporosis and investigate its relationship with frailty, in a sample of 316 community-dwelling Chinese older people. Regression analysis was performed using frailty as the dependent variable. The results showed that the prevalence rate of sarco-osteoporosis was 10.4% in older men and 15.1% in older women. ≧80 years old (OR 4.8; 95% CI, 3.05–10.76;P=0.027), women (OR 2.6; 95% CI, 1.18–2.76;P=0.036), and higher level of comorbidity (OR 3.71; 95% CI, 1.61–10.43;P=0.021) were independently associated with the likelihood of being sarco-osteoporosis. In the frail group, sarco-osteoporosis occurred in 26.3% of men, in 38.5% of women, and in lower proportion in the prefrail (13.6% of men; 16.2% of women) and nonfrail group (1.6% of men; 1.9% of women) (P<0.05, resp.). Furthermore, the likelihood of being frail/prefrail was substantially higher in the presence of sarco-osteoporosis (OR 4.16; 95% CI, 2.17–17.65;P=0.019in men; and OR 4.67; 95% CI, 2.42–18.86;P=0.007in women). The results indicate that patients with sarco-osteoporosis are more likely to be ≧80 yrs with higher burden of comorbidities and to have frailty/prefrailty, especially for women.

Abstract:
alpha-Sarcin from Aspergillus giganteus and the ribosome-inactivating proteins (RIPs) from higher plants inactivate the 60 S ribosomal subunit. The former is an RNAase, whereas RIPs are N-glycosidases. The site of cleavage of RNA and that of N-glycosidic depurinization are at one nucleotide distance in 28 S rRNA [Endo & Tsurugi (1987) J. Biol. Chem. 262, 8128-8130]. The effect of alpha-sarcin and that of RIPs on the interaction of elongation factors with Artemia salina (brine shrimp) ribosomes have been investigated. alpha-Sarcin inhibits both the EF1 (elongation factor 1)-dependent binding of aminoacyl-tRNA and the GTP-dependent binding of EF2 (elongation factor 2) to ribosomes, whereas two of the RIPs tested, ricin from Ricinus communis (castor bean) and volkensin from Adenia volkensii (kilyambiti), inhibit only the latter reaction. EF2 protects ribosomes from inactivation by both alpha-sarcin and ricin. The EF1-binding site is affected only by alpha-sarcin. The sensitivity of this site to alpha-sarcin is increased by pretreatment of ribosomes with ricin. A. salina ribosomes were highly resistant to the third RIP tested, namely gelonin from Gelonium multiflorum. All four proteins tested have, however, a comparable activity on the rabbit reticulocyte-lysate system.

Abstract:
Postmenopausal osteoporosis (PMOP) and sarcopenia are common diseases that predominantly affect postmenopausal women. In the occurrence and development of these two diseases, they are potentially pathologically connected with each other at various molecular levels. However, the application of metabolomics in sarco-osteoporosis and the metabolic rewiring happening throughout the estrogen loss-replenish process have not been reported. To investigate the metabolic alteration of sarco-osteoporosis and the possible therapeutical effects of estradiol, 24 mice were randomly divided into sham surgery, ovariectomy (OVX), and estradiol-treated groups. Three-dimensional reconstructions and histopathology examination showed significant bone loss after ovariectomy. Estrogen can well protect against OVX-induced bone loss deterioration. UHPLC-Q-TOF/MS was preformed to profile semi- polar metabolites of skeletal muscle samples from all groups. Metabolomics analysis revealed metabolic rewiring occurred in OVX group, most of which can be reversed by estrogen supplementation. In total, 65 differential metabolites were identified, and pathway analysis revealed that sarco-osteoporosis was related to the alterations in purine metabolism, glycerophospholipid metabolism, arginine biosynthesis, tryptophan metabolism, histidine metabolism, oxidative phosphorylation, and thermogenesis, which provided possible explanations for the metabolic mechanism of sarco-osteoporosis. This study indicates that an UHPLC-Q-TOF/MS-based metabolomics approach can elucidate the metabolic reprogramming mechanisms of sarco-osteoporosis and provide biological evidence of the therapeutical effects of estrogen on sarco-osteoporosis.

Abstract:

Full text
Abstract:
The anti-tumour protein alpha-sarcin causes fusion of bilayers of phospholipid vesicles at neutral pH. This is demonstrated by measuring the decrease in the efficiency of the fluorescence energy transfer between N-(7-nitro-2-1,3-benzoxadiazol-4-yl)-dimyristoylphosphatidylethano lamine (NDB-PE) (donor) and N-(lissamine rhodamine B sulphonyl)-diacylphosphatidylethanolamine (Rh-PE) (acceptor) incorporated in dimyristoylphosphatidylcholine (DMPG) vesicles. The effect of alpha-sarcin is a maximum at 0.15 M ionic strength and is abolished at basic pH. alpha-Sarcin promotes fusion between 1,6-diphenylhexa-1,3,5-triene (DPH)-labelled DMPG and dipalmitoyl-PG (DPPG) vesicles, resulting in a single thermotropic transition for the population of fused phospholipid vesicles. Bilayers composed of DMPC and DMPG, at different molar ratios in the range 1:1 to 1:10 PC/PG, are also fused by alpha-sarcin. Freeze-fracture electron micrographs corroborate the occurrence of fusion induced by the protein. alpha-Sarcin also modifies the permeability of the bilayers, causing the leakage of calcein in dye-trapped PG vesicles. All of the observed effects reach saturation at a 50:1 phospholipid/protein molar ratio, which is coincident with the binding stoichiometry previously described.

Abstract:
AbstractThis article explores the way assumptions about gender prevalent in twenty-first century readers impact our understanding of Sarai. It interrogates the way a mere glimpse allows us to instantaneously assign a person gender, something trans theorist Julia Serano calls gendering. Through this article, we see how the third-party accounts of Sarai in Genesis 11:19–12:20 parallel the experience of that introductory glance today. By undertaking a close reading, different themes emerge that work to both confirm and challenge her fit within dominant gender norms. Indeed, Sarai cannot easily be subsumed into cisnormative gender expectations that privilege consistent coherence between the sex assigned at birth and gender identity and expression. Genesis 20:1–20 is then placed in discussion with the earlier portion of Sarai’s story. It provides the opportunity to revisit how observers within the text see and understand her. In turn new details emerge that seek to confirm Sarai’s fit within cisnormativity; but in doing so, they end up disrupting our perception that Sarai is a cisnormative woman. Ultimately, this reading establishes that Sarai does not neatly fit our preconceptions, which opens up the potential to consider her a transgender or a gender-diverse figure.

"Stability of Sources of Resistance to Cowpea Aphid (Aphis craccivora Koch, Hemiptera: Aphididae) across Major Cowpea Production Zones in Ghana." Abstract:
Aphids (Aphis craccivora Koch) are an important vegetative stage pest of cowpea in Africa. The use of resistant cultivars is among the best management option for this pest, but the success of this strategy is influenced by the stability of the resistant genotype to the cowpea aphid biotypes present in the major cowpea growing areas in a country. This work, therefore, aimed at identifying cultivars/genotypes with stable resistance to aphid infestation across different cowpea growing ecologies in Ghana and estimating yield loss due to aphid infestation at the seedling stage. To ascertain the stability of aphid-resistant cultivars/genotypes, four cultivars/genotypes (SARC1-57-2, SARC1-91-1, IT97K-499-35, and Zaayura) and a susceptible check (Apagbaala) were tested across 18 locations in Ghana. An on-station experiment was used to quantify yield losses due to aphid attack at the seedling stage in the five cultivars/genotypes mentioned above together with 5 additional cultivars/genotypes [i.e., IT99K-573-3-2-1, IT99K-573-1-1, Padituya, Resistant BC4F3 (Zaayura//(Zaayura × SARC1-57-2)), and Susceptible BC4F3 (Zaayura//(Zaayura × SARC1-57-2))]. The results showed that SARC1-57-2 was stable in all ecologies, in terms of its resistance to aphids; it had high vigour score (3.8 ± 0.03) and low plant mortality (3.7 ± 0.22%) compared to the susceptible genotypes. The number of days to flowering and maturity were significantly higher in aphid-infested plants than in the uninfested ones. Grain yield loss was estimated to range between 3.8 and 32.8%. Except for SARC1-57-2, Resistant BC4F3, and Padituya, the remaining cultivars/genotypes sustained significant yield losses under aphid infestation. Thus, the aphid-resistance gene in SARC1-57-2 is stable against aphids. This resistance genotype can be incorporated into cowpea improvement programmes to breed for aphid-resistant cultivars. Also, the cultivation of such improved cultivars will reduce pesticide usage in cowpea production.

Abstract:
The representative of the genus Jaera Leach inhabiting continental waters throughout Europe has a rather complex taxonomic history. After discovering it in the River Tisza, Hungary, Dudich (1930) identified it as Jaera nordmanni (Rathke). This emerged to be a mistake, as the species was found to be identical with the subsequently described Jaera sarsi Valkanov (Kesselyák 1938). On the contrary, Veuille (1979) argued that the freshwater form is different from the brackish water species J. sarsi, and described it as a new species, J. istri Veuille, 1979. The new terminology has become widely used; however, recently the concept received criticism, implying that the characteristics distinguishing J. istri from J. sarsi fall within the confines of intraspecific variability (Tobias et al. 2005). Here I tentatively accept the latter point of view, and will refer to the species as J. sarsi in the following, but I also wish to point out that this matter could be settled using molecular methods.

Abstract:
Analyzing numerous samples from Brazil, we found only three previously described species of Ilyocryptus (Cladocera: Anomopoda: Ilyocryptidae): I. paranaensis paranaensis Paggi, 1989, I. sarsi Stingelin, 1913 and I. spinifer Herrick, 1882. Two new taxa, being Neotropical subspecies of the species previously described from Palaearctic, I. cuneatus brasiliensis subsp. nov. and I. silvaeducensis paraensis subsp. nov., are described. Maps of Brazilian records are provided for each species using both original and literature data. Most part of records of Brazilian ilyocryptids concerns I. spinifer, which is among the most common cladocerans in the Neotropics, while other taxa occur significantly rare. I. spinifer has a wide distributional range. “I. sarsi” seems to be a group of close topical congeners, quite possible that I. sarsi s. str. is an exclusively Neotropical taxon. Three other recorded species are Neotropical endemics. We believe that previous records of I. sordidus (Lévin, 1848) from Brazil were misidentifications, probably of two newly described taxa, or I. sarsi.

Abstract:
Variant histologies of bladder cancer (BC) often present with advanced tumor stage and the status of perioperative therapy is unclear. Thereby, squamous cell carcinoma (SCC), adenocarcinoma (ADENO), and sarcomatoid urothelial carcinoma (SARCO) are the most frequent variants. Nectin-4 has emerged as a highly interesting target in BC and might guide therapeutic application of antibody–drug conjugates (ADC). We therefore aimed to investigate expression patterns and prognostic value of Nectin-4 in variant histologies of BC. A single-center retrospective analysis was conducted of patients who underwent radical cystectomy (RC) for BC and revealed variant histologies of BC in the final specimens. Immunohistochemical staining for Nectin-4 was performed on tissue microarrays with 59 SCC, 22 ADENO, and 24 SARCO, and Nectin-4 expression was scored using the histochemical scoring system (H-score). Overall survival (OS) and progression-free survival (PFS) was calculated by Kaplan–Meier method. Median expression of Nectin-4 was 150 (range 0–250) in SCC, 140.5 (range 30–275) in ADENO, and 10 (0–185) in SARCO, with significantly lower levels for SARCO compared to SCC or ADENO (p < 0.001). For SCC, ADENO or SARCO no differences regarding OS or PFS were observed based on Nectin-4 expression levels (p > 0.05). Multivariate analysis revealed nodal stage as an independent prognostic factor for OS and PFS and metastases for PFS but not Nectin-4 expression. In conclusion, Nectin-4 was not prognostic in histological subtypes of BC in our study cohort. However, the high expression of Nectin-4 in SCC and ADENO might guide future treatment with novel Nectin-4-directed ADCs and provide this high-risk patient collective with a new promising therapeutic option. Testing Nectin-4 expression as a biomarker should be considered in trials with SARCO, where low Nectin-4 expression has been observed.
