Abstract:

Abstract:
Abstract Extrachromosomal DNA circularization is a common event in cancer cells and frequently serves as a vehicle for cancer oncogene amplification. Random segregation of oncogene-containing extrachromosomal circular DNA promotes rapid intercellular heterogeneity, conferring tumors the ability to rapidly evolve and escape therapy. Smaller, copy-number neutral extrachromosomal circular DNAs are also abundantly identified in both healthy and malignant tissues, but their function in cancer is still unknown. Understanding how extrachromosomal circular DNAs contribute to intercellular heterogeneity in cancer cells remains crucial, however methods for an unbiased characterization of extrachromosomal circular DNAs in single cells are lacking. We introduce scEC&T-seq (single cell extrachromosomal circular DNA and transcriptomic sequencing), a method for parallel detection of extrachromosomal circular DNAs and full-length mRNA in single cells. We demonstrate the ability of our method to isolate and detect extrachromosomal circular DNAs genome-wide from all range of sizes in single cells. We observed that whereas large oncogene-containing circular DNAs are clonally present in most cancer cells, only a very small fraction of small circular DNAs are recurrently identified in single cells, indicating yet unknown prerequisites for maintenance and propagation. Our method was able to capture and recapitulate the structural complexity of oncogene-containing extrachromosomal circular DNAs in single cells, and the matching transcriptomic data allowed us to identify fusion transcripts resulting from the rearranged extrachromosomal structures. In addition, we observed that whereas the main structure of extrachromosomal circular DNAs is mostly stable in single cells, intercellular differences in extrachromosomal circular DNAs’ content can drive differences in oncogene transcription levels in single cells. We envision that by integrating extrachromosomal circular DNA and mRNA sequencing, our method will not only be useful to investigate the impact of intercellular heterogeneity in extrachromosomal circular DNA in tumor evolution, but also to interrogate its function in other biological and pathological processes. Abstract:

Abstract:

Abstract:

Williams, Nicholas Mark. "A study on performing the Hungarian Rhapsodies in the Liszt tradition." Thesis, Edith Cowan University, Research Online, Perth, Western Australia, 2020. https://ro.ecu.edu.au/theses/2360.

Abstract:
Franz Liszt’s Hungarian Rhapsodies (1851, 1853) have long been among the most popular collections of piano music. They have also long garnered a reputation for “superficial brilliance and effect” which seems to have influenced the way that famous pianists play the works in public. But would a performer immersed in the Liszt tradition have approached them differently? This dissertation aims to promote a re-evaluation of the Hungarian Rhapsodies from this perspective: considering Liszt’s own ideas on music and performance, the writings and recordings of his pupils, and Liszt’s book Des Bohémiens et de leur musique en Hongrie (1859).

Abstract:
Will Automated Vehicles be Safer than Conventional Vehicles? One of the critically important questions that has emerged about advanced technologies in transportation is how to test the actual effects of these advanced systems on safety, particularly how to evaluate the safety of highly automated driving systems. Richard Young's Critical Analysis of Prototype Autonomous Vehicle Crash Rates does a deep dive into these questions by reviewing and then critically analyzing the first six scientific studies of AV crash rates.

Abstract:
In representations of the First World War (1914–1918), the figure of the sacrificial offering appears and reappears to consecrate an otherwise uncertain purpose—to make holy, sacer- facere. On the military side, in Britain especially, the war witnessed a shift from a professional army to a civilian recruit, u a conscript mass, whose individual deaths required justification. The figure of the sacrificial offering recurs with a frequency commensurate with the scale of this first mass war. This quantity of death, however, ultimately overwhelms the logic of sacrifice, which turns on the quality, the specialness, the worthiness, of the individual victim. Drawing on the philosophy of Giorgio Agamben, this chapter argues that the proportionate economy of individual sacrifice is eventually disrupted by indiscriminate slaughter, and the failure of the old model is recorded in a range of British fiction, including works by Richard Aldington, Ford Madox Ford, and David Jones.

Abstract:
This introductory chapter provides an overview of federalism. It first defines the key institutional features of federal governance and presents evidence as to the relative importance of federal states among the nations of the world. The chapter then offers an empirical evaluation of the ability of Democratic Federalism to advance the three objectives embraced by its supporters: economic efficiency, political participation and democratic stability, and the protection of individual rights and liberties. Causation is always a question in such exercises, but the correlations are clear. Societies governed by the principles of Democratic Federalism are richer, safer for personal rights and liberties, and democratically more engaged.

Full text
Abstract:
Katherine Mansfield’s claim that she and Elizabeth von Arnim are ‘worms of the same family’ would seem a curious analogy. But this deprecatory trope aligning the writer’s perspective with an insect’s, nonetheless, uses commonly held assumptions to interrogate gendered subjectivity and class dichotomies in the postwar world. In ‘The Fly,’ Mansfield depicts a businessman distracted by the seemingly harmless entertainment of dousing a housefly with ink; his god-like play with the fly evokes an attempt to master trauma – the loss of his son in the war, his consequent lack of purpose, and his complicity. Likewise, Mabel Waring in Virginia Woolf’s ‘The New Dress’ compulsively repeats an image she’s conjured of a fly crossing a saucer as a bulwark against the sense of irrelevance she feels in upper-class society. In Kangaroo, D.H. Lawrence’s writer Richard Somers faces a dark night of the soul trying on and rejecting different ideologies in order to secure a sense of purpose in his work; he sees himself as a fly harrowingly climbing up and continually falling back into a pot of ointment. Each protagonist considers or takes on the point of view of a housefly, attempting to master a trauma that, seen fully, would threaten his or her identity. These scenes should be an affective climax, however, as the deprecating choice of a housefly suggests, they are instead moments not of epiphany or emotional release but of existential impasse.

Abstract:
A number of recent events inside and outside of the heritage sector have triggered a lively and largely constructive debate about the excavation, display, and conservation of human remains in the UK (see Jenkins 2008, 2010; Moshenska 2009; Sayer 2009, 2010a; Parker Pearson et al. 2011; Giesen 2013). Two events have been of particular significance: the reburial of human remains prompted by requests to museums from the Pagan community, and independently of these requests the Ministry of Justice decided to revisit its conditions for the excavation of human remains (Parker Pearson et al. 2013). In the short term, these issues seem to have been resolved through open consultation and campaigning by archaeologists. British archaeologists consider that they have public support; public-facing archaeology develops strong links within local communities, the Portable Antiquities Scheme engages members of the public in the discovery of metal objects on a national scale, and TV and Radio programmes regularly include archaeology or excavation as their central theme. There are various ways to engage with archaeology outside of a traditional museum environment: people can shift soil or sit back and read about it in numerous academic and popular books, in magazines, and digitally on the internet. This chapter discusses this new digital environment by describing and analysing three events in British burial archaeology which deliberately sought coverage online and within global media. These are: 1) the burial campaign which was instrumental in raising the profile of the reburial problem in England; 2) the discovery of a cow and woman buried in the same grave in a fifth- and sixth-century cemetery at Oakington, Cambridgeshire; 3) the investigation of King Richard III’s final resting place in the city of Leicester. One of us was instrumental in publicizing the first two events; neither of us was involved in the third. We will refer also to a recent case in East Anglia where negative media publicity came unsought by the archaeologists concerned. In the mid-twentieth century, archaeology found a place in mass broadcasting and early shows like Animal, Vegetable and Mineral or Chronicle captured the public imagination (Bailey 2010).
